To find the appropriate range of information quantity, we studied how the information quantity of traffic engineering facilities (TEFs) on straight roads of grassland highways affects a driver's eye movements. We used a combination of survey, statistics, analysis of variance, and the establishment of virtual scene to do this research, and carried out simulated driving tests at six levels (Z0, Z1, Z2, Z3, Z4, and Z5) of TEF information quantity. The driver's fixation duration, visual search breadth, and glance speed were evaluated in a quantitative way. Results showed that the information quantity had a significant impact on eye movements. It is concluded that the information quantity from 0 to 10 bits/km may cause problems to drivers, whereas the information quantity of 40 bits/km serves as the limit. The information quantity from 30 to 40 bits/km is the appropriate one for TEF on grassland highways.

In order to study the change law of the fatigue degree of grassland expressway drivers over time, this paper takes the semidesert grassland landscape of Xilinhot city as the experimental environment and takes the provincial highway S101 (K278-K424) as an example to design an actual driving test. Taking Urumqi, Inner Mongolia Autonomous Region, as the experimental section, combined with the Biopac MP150 multichannel physiological instrument and its auxiliary knowledge software and mathematical statistics methods, the relationship between EEG and time was studied. The test results show that the primary fatigue factor F 1 and the secondary fatigue factor F 2 can summarize the fatigue law characterized by 96.42% of EEG information. During 130 minutes of driving on the prairie highway, the periods of high fatigue were 105 minutes and 125 minutes, respectively. Driving fatigue can be divided into three stages over time: 5-65 min fatigue-free stage, 70-85 min fatigue transition stage, and 90-130 min fatigue stage. Fatigue changes over time. The law follows the Gaussian function and the sine function.

BACKGROUND: Insects can be models for understanding human intestinal infection and pathology. Molting, a special period during which the old insect cuticle is shed and a new one is produced, is crucial for insect development. Holometabolous insects may experience several larva-to-larva moltings to become larger, a pupal molt and adult eclosion to become adults. During the larval molts, they stop feeding and become quiescent. Although the molting larvae become quiescent, it is not known if changes in microbiome, physiology, development and immunity of midguts occur. RESULTS: Transcriptome analysis indicated that functions such as metabolism, digestion, and transport may become reduced due to the downregulated expression of many associated genes. During the molting stage, midguts harbor less microflora and DNA synthesis decreases. Both ecdysone and juvenile hormone in the larval midgut likely degrade after entering the larva-to-larva molting stage. However, at 12 h after ecdysis, the feeding larvae of 5th instars that were injected with 20-hydroxyecdysone entered a molting-like stage, during which changes in midgut morphology, DNA synthesis, gene expression, and microflora exhibited the same patterns as observed in the actual molting state. CONCLUSION: This study is important for understanding insect midgut physiology, development and immunity during a special development stage when no food is ingested. Although the molting larva becomes immobile and quiescent, we demonstrate that numerous changes occur in midgut morphology, physiology, metabolism and microbiome during this period.

Plant phenolics are a group of important secondary metabolites that are toxic to many animals and insects if ingested at high concentrations. Because most insects consume plant phenolics daily, they have likely evolved the capacity to detoxify these compounds. Here, we used Drosophila melanogaster, Bombyx mori and Helicoverpa armigera as models to study the metabolism of plant phenolics by prophenoloxidases. We found that insect foreguts release prophenoloxidases into the lumen, and that the survival of prophenoloxidase-deletion mutants was impaired when fed several plant phenolics and tea extracts. Using l-DOPA as a model substrate, biochemical assays in large Lepidopteran insects demonstrated that low levels of l-DOPA are rapidly metabolized into intermediates by phenoloxidases. Feeding with excess l-DOPA showed that the metabolic intermediate 5,6-dihydroxyindole reached the hindgut either by passing directly through the midgut, or by transport through the hemolymph. In the hindgut, 5,6-dihydroxyindole was further oxidized by prophenoloxidases. Intermediates exerted no toxicity in the hemocoel or midgut. These results show that plant phenolics are not toxic to insects unless prophenoloxidase genes are lost or the levels of phenolics exceed the catalytic activity of the gut prophenoloxidases.

Bombyx mori Bidensovirus (BmBDV), a bipartite virus possesses two single-stranded linear DNAs (VD1 and VD2) and shows high pathogenic ability to Bombyx mori. Previous research found that the genes of nonstructural protein ns1 and ns2 were in the same transcript. To investigate the mechanism of transcriptional regulation of ns1 and ns2 genes, the 5'-flanking sequence (289 nt) of ns1 gene, encompasses the regions of the common terminal sequence (CTS) and the predicted P5 promoter from the 5'-terminus of the viral genome to the transcription initiation site of the ns1 gene was cloned and fused to the upstream of the luciferase reporter gene. The luciferase reporter assay showed that the 53 nt CTS of VD1 and VD2 can downregulate the activity of P5 by 13.3 %. The comparison in different cell lines showed that P5 possessed high promoter activity in BmN and Hi5 cell lines. Interestingly, P5 also had high activity in Hela cells, a kind of cancer cell of human. Subsequent truncated promoter analysis showed that the 31 nt (-236 to -206 nt) sequence is very important to P5 for the activity down to 36.5 % after deletion of it. While the activity also remained 26.5 % after the deletion of the TATA box, suggesting that the promoter is TATA independent. Moreover, in order to further understand the activity intensity of P5, a comparison with other three promoters, B. mori actin3 (Bm-actin3), B. mori nuclear polyhedrosis virus (BmNPV) immediate early 1 gene promoter (BmNPV-ie-1), and a synthetic promoter (3xP3) was carried out, the result indicated that the activity of P5 was weaker than that of anyone of them.
