RESUMO

BACKGROUND: Non-invasive brain stimulation (NIBS), especially repetitive transcranial magnetic stimulation (rTMS), is a promising therapeutic tool for managing the psychiatric symptoms of Parkinson's disease (PD). This meta-analysis aims to assess the effect of NIBS on psychiatric symptoms in patients with PD. METHODS: Several electronic databases were systematically searched for relevant literature from inception to October 18, 2021. The therapeutic effects of NIBS were evaluated using a random-effects meta-analysis based on the standard mean difference (SMD) and multivariable-adjusted effect estimates and subgroup analyses. Publication bias and sensitivity analyses were used to explore the sources of heterogeneity. RESULTS: Finally, 57 and 15 studies with 4,010 participants were obtained from the meta-analysis and systematic review, respectively. Significant differences were found between NIBS and sham-stimulation/placebo participants for depression (SMD = -0.75, 95 % confidence interval [CI]: -0.99 to -0.50, I2 = 79 %), and rTMS combined with antidepressant therapy and antidepressant alone for depression (SMD = -0.98, 95 % CI: -1.37 to -0.59, I2 = 87 %) and anxiety (SMD = -1.46, 95 % CI: -2.44 to -0.48, I2 = 95 %). Meanwhile, lower risks were observed for post-treatment compared with pre-treatment for depression (relative risk [RR] = 0.66, 95 % CI: 0.57-0.76, I2 = 96 %) and anxiety (RR = 0.66, 95 % CI: 0.60-0.73, I2 = 67 %). CONCLUSIONS: NIBS can be considered an effective non-pharmacological option for treating PD depression; moreover, rTMS combined with antidepressants was demonstrated to be a useful tool against depression and anxiety in PD.

The cell-type-specific recording and manipulation is instrumental to disentangle causal neural mechanisms in physiology and behavior and increasingly requires intersectional control; however, current approaches are largely limited by the number of intersectional features, incompatibility of common effectors and insufficient gene expression. Here, we utilized the protein-splicing technique mediated by intervening sequences (intein) and devised an intein-based intersectional synthesis of transactivator (IBIST) to selectively control gene expression of common effectors in multiple-feature defined cell types in mice. We validated the specificity and sufficiency of IBIST to control fluorophores, optogenetic opsins and Ca2+ indicators in various intersectional conditions. The IBIST-based Ca2+ imaging showed that the IBIST can intersect five features and that hippocampal neurons tune differently to distinct emotional stimuli depending on the pattern of projection targets. Collectively, the IBIST multiplexes the capability to intersect cell-type features and controls common effectors to effectively regulate gene expression, monitor and manipulate neural activities.

RESUMO

A novel, easy-to-handle, and regioselective vicinal dioxidation of alkenes under transition metal and organic peroxide free conditions has been developed. This approach uses N-hydroxyphthalimide and its analogues as the transient nitroxyl-radical precursors and 2,2,6,6-tetramethylpiperidine-1-oxoammonium tetrafluoroborate (TEMPO+BF4-) as the oxidant as well as the source of persistent nitroxide. By employing this method, multifarious structurally important dioxidation products were efficiently synthesized from simple alkenes and complex bioactive molecule derivatives.

AIM: To explore an xeno-free and defined coating substrate suitable for the culture of H9 human embryonic stem cell-derived retinal pigment epithelial (hES-RPE) cells in vitro, and compare the behaviors and functions of hES-RPE cells on two culture substrates, laminin521 (LN-521) and truncated recombinant human vitronectin (VTN-N). METHODS: hES-RPE cells were used in the experiment. The abilities of LN-521 and VTN-N at different concentrations to adhere to hES-RPE cells were compared with a high-content imaging system. Quantitative real-time polymerase chain reaction was used to evaluate RPE-specific gene expression levels midway (day 10) and at the end (day 20) of the time course. Cell polarity was observed by immunofluorescent staining for apical and basal markers of the RPE. The phagocytic ability of hES-RPE cells was identified by flow cytometry and immunofluorescence. RESULTS: The cell adhesion assay showed that the ability of LN-521 to adhere to hES-RPE cells was dose-dependent. With increasing coating concentration, an increasing number of cells attached to the surface of LN-521-coated wells. In contrast, VTN-N presented a strong adhesive ability even at a low concentration. The optimal concentration of LN-521 and VTN-N required to coat and adhesion to hES-RPE cells were 2 and 0.25 µg/cm2, respectively. Furthermore, both LN-521 and VTN-N could facilitate adoption of the desired cobblestone cellular morphology with tight junction and showed polarity by the hES-RPE cells. However, hES-RPE cells cultivated in VTN-N had a greater phagocytic ability, and it took less time for these hES-RPE cells to mature. CONCLUSION: VTN-N is a more suitable coating substrate for cultivating hES-RPE cells.

Human albumin (HA) displays crucial roles in maintaining health and fighting diseases. Accurate determination of native HA in plasma or non-plasma samples are of immense significance in both basic research and clinical practice. Herein, a novel ratiometric two-photon fluorescent probe (N-butyl-4-(4-phenyl-benzoyloxy) 1,8-naphthalimide, BPBN) has been designed and developed for highly selective and sensitive sensing the enzymatic activities of HA, on the basis of its unique pseudo-esterase feature. BPBN exhibits excellent selectivity, high sensitivity and good reactivity under physiological conditions. As an enzymatic activity-based probe, BPBN can distinguish between native HA and denatured HA, while the currently used dye-binding method cannot. The probe has been successfully applied to measure native HA in plasma samples and the secreted HA in the hepatocyte culture supernatant. BPBN has also been used for two-photon imaging of HA reabsorption in living renal cells, and the results demonstrate that this probe exhibits good cell permeability, low cytotoxicity and high imaging resolution. All these findings suggest that BPBN can be reliably used for the highly selective and sensitive detection of native HA in complex biological samples, as well as for exploring HA-associated biological processes and the physiological functions of native HA in living cells.

RESUMO

A new phenyl ether derivative, 3-hydroxy-5-(3-hydroxy-5-methylphenoxy)-4-methoxybenzoic acid (1), along with two known analogues, 3,4-dihydroxy-5-(3-hydroxy-5-methylphenoxy)benzoic acid (2) and 3-hydroxy-5-(3-hydroxy-5-methylphenoxy)benzoic acid (3), were isolated from the fungus Aspergillus carneus collected from South China Sea. The structure elucidation of 1 was determined based on extensive NMR and MS spectroscopic analyses. Compound 2 showed a strong antioxidant activity with an IC50 value of 19.3 µM which was close to the positive control ascorbic acid (IC50 = 15.3 µM).
