RESUMO

This study utilizes digital image correlation (DIC) technology to measure the far-field displacements and strains of rock specimens during the entire loading and unloading. Through analyzing the distributions of strain, displacement and their variations per unit length at different stages, the variations of both length and migration velocity of the fracture process zone (FPZ) were studied, and the crack propagation was also investigated. In addition, the entire path of crack propagation was observed by scanning electron microscope (SEM). The results reveal that (1) the fractured ligament can be divided into three zones based on the displacement variation per unit length: intact zone, crack propagation zone, and FPZ. (2) The FPZ length reaches its maximum at the peak load and then decreases, and the minimum length even is only 1/3-1/2 of the maximum length. The FPZ migration velocity is - 48 to 1460 m/s. FPZ's microscale features are intergranular microcracks, transgranular microcracks, cleavage, and debris on fracture surface and around main crack propagation path. (3) The crack propagation length during peak load to peak-post 90% accounts for more than 1/3-1/4 of the entire post-peak length. Crack propagation is alternating fast and slow, i.e., the velocity of crack propagation varies regularly in the range of 24-700 m/s. The region of crack initial propagation is more severely damaged compared to other propagation regions.

RESUMO

The construction of the relation between the critical energy release rate, GIc, and the mode I fracture toughness, KIc, is of great significance for understanding the fracture mechanism and facilitating its application in engineering. In this study, fracture experiments using NSCB and CCCD specimens were conducted. The effects of specimen sizes, loading rate and lithology on the relation between GIc and KIc were studied. GIc was calculated by integrating the load-displacement curve according to Irwin's approach. Based on the measured KIc and GIc of the rock specimens, a relation between GIc and KIc was found to be different from the classical formula under linear elasticity. It was found that both specimen size and loading rate do not influence this relation.

RESUMO

This paper focuses on a new test method and theoretical model for measuring and evaluating the reopening pressure during hot dry rock hydraulic fracturing. Firstly, rock blocks of four lithologies were collected from the hot dry rock strata. Hydraulic fracturing tests at high temperatures in real-time were conducted using drilled cubic specimens and drilled cubic specimens with a pre-crack. Breakdown pressure, reopening pressure, and fracture toughness were measured, respectively. In addition, Brazilian splitting tests at high temperatures in real-time were performed using Brazilian disc specimens to measure tensile strength. Secondly, an empirical equation for evaluating the reopening pressure during hot dry rock secondary fracturing was developed based on fracture mechanics and hydraulic fracturing theory. Third, the values calculated by the new equation, considering breakdown pressure, fracture toughness, and tensile strength, were compared to the values determined by the classical equation and to measurement results. It was found that the new equation predicted closer reopening pressure to the measurement results, regardless of the lithology of the hot dry rock. Moreover, with increasing temperature in the specimens, the error between the value calculated by the new equation and the measurement value remained low. In contrast, the difference between the classical equation predictions and the measurement results was widened. In addition, the reopening pressure was positively correlated with tensile strength and fracture toughness. Variations in lithology and temperature affected tensile strength and fracture toughness, which then changed the hot dry rock reopening pressure.

RESUMO

Fish suffer from starvation due to environmental risks such as extreme weather in the wild and due to insufficient feedings in farms. Nutrient problems from short-term or long-term starvation conditions can result in stress-related health problems for fish. Yellowfin seabream (Acanthopagrus latus) is an important marine economic fish in China. Understanding the molecular responses to starvation stress is vital for propagation and culturing yellowfin seabream. In this study, the transcriptome and genome-wide DNA methylation levels in the livers of yellowfin seabream under 14-days starvation stress were analyzed. One hundred sixty differentially expressed genes (DEGs) by RNA-Seq analysis and 737 differentially methylated-related genes by whole genome bisulfite sequencing analysis were identified. GO and KEGG pathway enrichment analysis found that energy metabolism-related pathways such as glucose metabolism and lipid metabolism were in response to starvation. Using bisulfite sequencing PCR, we confirmed the presence of CpG methylation differences within the regulatory region of a DEG ppargc1a in response to 14-days starvation stress. This study revealed the molecular responses of livers in response to starvation stress at the transcriptomic and whole genome DNA methylation levels in yellowfin seabream.

RESUMO

Phyllite is widely distributed in nature, and it deserves to be studied considering rock engineering applications. In this study, uniaxial compression tests were conducted on foliated phyllite with different foliation angles under dry and water-saturated conditions. The impacts of water content and foliation angle on the stress-strain curves and basic mechanical properties of the Phyllite were analyzed. The experimental results indicate that the peak stress and peak strain decrease first and then increase with increasing foliation angle as a U-shape or V-shape, and the phyllite specimens are weakened significantly by the presence of water. Moreover, an approach with acoustic emission, digital image correlation, and scanning electron microscopic is employed to observe and analyze the macroscopic and mesoscopic failure process. The results show that tensile microcracks dominate during the progressive failure of phyllite, and their initiation, propagation, and coalescence are the main reasons for the failure of the phyllite specimens. The water acts on biotite and clay minerals that are main components of phyllite, and it contributes to the initiation, propagation, and coalescence of numerous microcracks. Finally, four failure modes are classified as followed: (a) for the specimens with small foliation angles α = 0° or 30° (Saturated), both shear sliding and tensile-split across the foliation planes; (b) for the specimens with low to medium foliation angles α = 30° (Dry) or 45°(Saturated), shear sliding dominates the foliation planes; (c) for the specimens with medium to high foliation angles α = 45° (Dry) or 60°, shear sliding dominates the foliation planes; (d) for the specimens with high foliation angles α = 90°, tensile-split dominates the foliation planes.

RESUMO

BACKGROUND: Colorectal cancer (CRC) is one of the most common and fatal cancers worldwide. Synaptophysin-like 2 (SYPL2) is a neuroendocrine-related protein highly expressed in skeletal muscle and the tongue. The involvement of SYPL2 in CRC, including its level of expression and function, has not been evaluated. AIM: To evaluate the correlations of SYPL2 expression with lymph node metastasis (LNM) and prognosis in patients with CRC. METHODS: The levels of expression of SYPL2 in CRC and normal colorectal tissues were analyzed in multiple public and online databases. The associations between clinical variables and SYPL2 expression were evaluated statistically, and the associations between SYPL2 expression and prognosis in patients with CRC were analyzed using the Kaplan-Meier method and univariate/multivariate Cox regression analyses. SYPL2 expression was assessed in 20 paired CRC tissue and adjacent normal colorectal tissue samples obtained from Fuyang People's Hospital, and the associations between SYPL2 expression and the clinical characteristics of these patients were investigated. Correlations between the levels of expression of SYPL2 and key targeted genes were determined by Pearson's correlation analysis. The distribution of immune cells in these samples was calculated using the CIBERSORT algorithm. Gene set enrichment analysis (GSEA) was performed to evaluate the biofunction and pathways of SYPL2 in CRC. RESULTS: SYPL2 expression was significantly lower in CRC tissue samples than in normal colorectal tissue samples (P < 0.05). High SYPL2 levels in CRC tissues correlated significantly with LNM (P < 0.05) and a poorer patient prognosis, including significantly shorter overall survival (OS) [hazard ratio (HR) = 1.9, P < 0.05] and disease-free survival (HR = 1.6, P < 0.05). High SYPL2 expression was an independent risk factor for OS in both univariate (HR = 2.078, P = 0.014) and multivariate (HR = 1.754, P = 0.018) Cox regression analyses. In addition, SYPL2 expression correlated significantly with the expression of KDR (P < 0.0001, r = 0.47) and the BRAF V600E mutation (P < 0.05). Higher SYPL2 expression was associated with the enrichment of CD8 T-cells and M0 macrophages in the tumor microenvironment. GSEA revealed that SYPL2 was associated with the regulation of epithelial cell migration, vasculature development, pathways in cancer, and several vital tumor-related pathways. CONCLUSION: SYPL2 expression was lower in CRC tissue than in normal colorectal tissue. Higher SYPL2 expression in CRC was significantly associated with LNM and poorer survival.

RESUMO

Computer visual systems can rapidly obtain a large amount of data and automatically process them with ease. These characteristics constitute advantages for the application of such systems in the automatic analysis of medical images, as well as in processing technology. The precision of image segmentation, which plays a critical role in computer visual systems, directly affects the quality of processing results. Coronary angiographs feature various background colors, complex patterns, and blurry edges. The image areas containing blood vessels cannot be precisely segmented through regular methods. Therefore, this study proposed an unsupervised learning algorithm that uses regional parameter expansion (RPE). This method was derived from the flood fill algorithm, which can effectively segment image areas containing blood vessels despite a complex background or uneven light and shadow. An optimal cover tree (OCT) algorithm was proposed for the establishment of coronary arteries and the estimation of vessel diameter. Through the region growing method, spanning trees were used to record the cover length of adjacent connections, thereby establishing vessel paths, and the length can be used to track changes in vessel diameter.

RESUMO

This study proposed a noninvasive blood glucose estimation system based on dual-wavelength photoplethysmography (PPG) and bioelectrical impedance measuring technology that can avoid the discomfort created by conventional invasive blood glucose measurement methods while accurately estimating blood glucose. The measured PPG signals are converted into mean, variance, skewness, kurtosis, standard deviation, and information entropy. The data obtained by bioelectrical impedance measuring consist of the real part, imaginary part, phase, and amplitude size of 11 types of frequencies, which are converted into features through principal component analyses. After combining the input of seven physiological features, the blood glucose value is finally obtained as the input of the back-propagation neural network (BPNN). To confirm the robustness of the system operation, this study collected data from 40 volunteers and established a database. From the experimental results, the system has a mean squared error of 40.736, a root mean squared error of 6.3824, a mean absolute error of 5.0896, a mean absolute relative difference of 4.4321%, and a coefficient of determination (R Squared, R2) of 0.997, all of which fall within the clinically accurate region A in the Clarke error grid analyses.

RESUMO

Red tilapia ( Oreochromis spp .) is one of the most popular fish in China due to its bright red appearance, fast growth rate, and strong adaptability. Understanding the sex determination mechanisms is of vital importance for the selection of all-male lines to increase aquacultural production of red tilapia. In this research, the genetic architecture for sex from four mapping populations ( n=1 090) of red tilapia was analyzed by quantitative trait loci (QTL)-seq, linkage-based QTL mapping, and linkage disequilibrium (LD)-based genome-wide association studies. Two genome-wide significant QTL intervals associated with sex were identified on ChrLG1 (22.4-23.9 Mb) and ChrLG23 (32.0-35.9 Mb), respectively. The QTL on ChrLG1 was detected in family 1 (FAM1), FAM2, and FAM4, and the other QTL on ChrLG23 was detected in FAM3 and FAM4. Four microsatellite markers located within the QTL were successfully developed for marker-assisted selection. Interestingly, three ( lpp, sox14, and amh) of the 12 candidate genes located near or on the two QTL intervals were abundantly expressed in males, while the remaining genes were more highly expressed in females. Seven genes ( scly, ube3a, lpp, gpr17, oca2, cog4, and atp10a) were significantly differentially expressed between the male and female groups. Furthermore, LD block analysis suggested that a cluster of genes on ChrLG23 may participate in regulating sex development in red tilapia. Our study provides important information on the genetic architecture of sex in red tilapia and should facilitate further exploration of sex determination mechanisms in this species.

RESUMO

As technology advanced, new e-health solutions are evolved to empower people to manage their care at home. This study explored the needs for disease management in activity tracking using photo diary through older adults' subjective perspective. It further aimed to suggest which lifestyle measures, symptoms and behaviours would be meaningful to include in such a digital diseases care management program for technology design. Both photo diary and focus group discussion were used, 11 older adults with multiple metabolism-related chronic diseases (Mean age, 72.5 ± 6.14 years) were recruited and asked to carry out the photo diary to trace their living situation and needs using a tablet camera. A focus group discussion was applied to identify the needs of chronic disease management, based on the results of living context tracing. Five themes, regular physical activity, smart management of healthy behaviors, healthy diet, regular daily routine and social connection, were identified by content analysis from photo diary and the focus group discussion. The results indicated that the photo diary program can raise awareness and promotes positive behavior changes. It is believed that the E-approach can be applied to the effectively enhance older adults' self-management by monitoring their health status and their daily routine activities.

RESUMO

Northwest China is burdened by declining soil fertility and poor capacity of water and nutrient retention. A pot experiment was conducted to examine the effects of organic acid conditio-ners (OASC) with four application rates (0, 20, 40, and 60 g·kg-1) on soil nutrients and crop growth. Maize and common vetch were the focus crops and loessial soil and irrigated desert soil were the soil types. The results showed that OASC application reduced water evaporation loss and significantly improved soil available nutrient content, with the most obvious effects on soil phosphorus. Available phosphorus content and proportion were increased by 256.5% and 227.4%, respectively, compared with no OASC treatment. The shoot dry weights of maize and common vetch on loessial soil were initially increased with the increasing OASC application rate and were highest at the application rate of 20 g·kg-1. The values progressively decreased with increasing OASC addition rates. Total salt content was significantly increased and the rate of emergence of common vetch decreased at OASC rate exceeding 20 g·kg-1. For irrigated desert soil, the OASC application rate did not affect total salt content when maize was planted. There was significant increase in soil total salt contents when common vetch was planted and at the OASC rate of 60 g·kg-1. The shoot dry weight of common vetch and maize was highest with the OASC application rates at 40 g·kg-1 and 60 g·kg-1, respectively. The optimal OASC rate for planting common vetch and maize on loess soil was 20 g·kg-1. The application rates of 40 g·kg-1 and 60 g·kg-1 were optimal for planting common vetch and maize on irrigated desert, respectively.

RESUMO

BACKGROUND: FGFR2 (fibroblast growth factor receptor 2) mutations are implicated in the etiopathogenesis of syndromic craniosynostosis, and C278F- or C342Y-FGFR2 mutations can lead to Crouzon syndrome. The dura mater exerts crucial effects in the regulation of cranial suture development. However, the underlying mechanisms of these biological processes are rarely studied. This research explored and analyzed the biological function of FGFR2 overexpressed by dura cells on cranial osteoblasts. METHODS: Dura cells and cranial osteoblasts from C57BL/6 mice aged 6 days were obtained and cultured respectively. Lentivirus-FGFR2 constructs were engineered with C278F- and C342Y-FGFR2 mutations. The dura cells were infected with the constructs and co-cultured with osteoblasts in a trans-well system. Four experimental groups were established, namely the Oste group, the Oste+Dura-vector group, the Oste+Dura-C278F group, and the Oste+Dura-C342Y group. FACS, CCK8, and EdU assays were used to evaluate the osteoblast proliferation levels. Western blot and RT-qPCR were used to measure the expressions of the factors related to proliferation, differentiation, and apoptosis. Furthermore, the expression levels of the key factors in the Hippo/YAP-PI3K-AKT proliferation pathway were measured and analyzed. Finally, rescue experiments were performed with an RNA interfering assay. RESULTS: The proliferation and differentiation levels of the osteoblasts in the Oste+Dura-C278F and Oste+Dura-C342Y groups were significantly up-regulated, but the apoptosis levels in the four groups were not significantly different. The YAP, TEADs1-4, p-PI3K, and p-AKT1 expressions in the mutant FGFR2 groups were higher than the corresponding expressions in the control groups, and the results of the rescue experiments showed a reverse expression tendency, which further confirmed the effects of the FGFR2 mutations in the dura cells on the proliferation of the osteoblasts and the underlying possible mechanisms. CONCLUSION: Our studies suggest that the Crouzon mutations (C278F- and C342Y-) of FGFR2 in dura cells can enhance osteoblast proliferation and differentiation and might influence the pathogenesis of craniosynostosis by affecting the Hippo/YAP-PI3K-AKT proliferation signaling pathway.

RESUMO

We examined the effects of long-term no-tillage sowing with crop stubbles on seedling emergence quality and yield of spring wheat under the three typical spring wheat planting modes of single wheat, wheat-corn intercropping and wheat-soybean intercropping in a long-term field experiment in Hexi oasis irrigated area, aiming to provide theoretical support for the efficient and sustai-nable production of spring wheat. The results showed that, compared with traditional tillage, no-tillage sowing with crop stubbles significantly decreased seedling emergence rate and emergence evenness of spring wheat in wheat-corn intercropping and wheat-soybean intercropping by 3.3%-8.6%, 9.6%-20.5%, 2.9%-8.8%, and 10.7%-61.7%, respectively. Emergence evenness was significantly increased by 14.9% in 2019, while seedling emergence rate was significantly decreased by 4.2% in 2020 under the mode of single wheat compared with traditional tillage. Seedling uniformity of spring wheat seedling stage were reduced under the three typical planting modes, including single wheat, wheat-corn intercropping and wheat-soybean intercropping. Spike number of spring wheat were equal to that of traditional tillage at harvest under the three planting modes of no-tillage sowing with crop stubbles, and the differences were not significant. Effects of seedling emergence rate of spring wheat on yield was weakened by increasing grain number per spike and 1000-grain weight of spring wheat under the three planting modes. Grain yield was significantly increased by 10.3%-12.9% (single wheat), 10.5%-11.9% (wheat-corn intercropping), and 10.3%-22.5% (wheat-soybean intercropping) at harvest, respectively. Our results indicated that no-tillage sowing with crop stubbles was the feasible tillage mode in the production process of spring wheat in Hexi oasis irrigation area with extremely serious farmland wind erosion degradation.

RESUMO

INTRODUCTION: The cementless Oxford partial knee arthroplasty is associated with low perioperative complications and good long-term survival rates. However, perioperative fractures remain a serious morbidity for patients. CASE REPORT: This case report describes an early post-operative tibial periprosthetic fracture through the keel slot, which we believe may be contributed by the deep implant keel design and the presence of a narrow metaphysis in the Asian knee. The patient subsequently underwent a revision total knee replacement and fixation of the periprosthetic fracture. CONCLUSION: This paper discusses the ways to identify patients at high risk of developing periprosthetic fractures and to minimize such occurrences, including adopting a modified tibial preparation, doing precise saw cuts, and considering a cemented tibial implant.

RESUMO

Haemosporidians infect a wide diversity of bat genera and species, yet little is known about their transmission cycles or epidemiology. Though several recent studies have focused on the genus Hepatocystis, an Old World parasite primarily infecting bats, monkeys, and squirrels, this group is still understudied with little known about its transmission and molecular ecology. These parasites lack an asexual erythrocytic stage, making them unique from the Plasmodium vertebrate life cycle. In this study, we detected a prevalence of 31% of Hepatocystis in short-nosed fruit bats (Cynopterus brachyotis) in Singapore. Phylogenetic reconstruction with a partial cytochrome b sequence revealed a monophyletic group of Hepatocystis from C. brachyotis in Malaysia, Singapore, and Thailand. There was no relationship with infection and bat age, sex, location, body condition or monsoon season. The absence of this parasite in the five other bat species sampled in Singapore indicates this Hepatocystis species may be host restricted.

After X-ray examination to exclude bone damage and deformity, 4 of them (3D guide plate group) randomly selected were for CT scan to obtain DICOM format data, and the data was imported into Mimics software for model, designed the ideal entry point and nail path for anterior cervicaltranspedicular screw (ATPS). After obtaining the personalized guide plate of the nail channel, it was exported as STL data, and the individual guide plate was printed by rapid prototyping and 3D printing technology. In turn, with the assistance of 3D guide plates, one-to-one personalized ATPS screws were placed on the four lower cervical cadaver specimens. Another 4 (free-hand group) lower cervical cadaver specimens were implanted with ATPS screws using free-hand technique. All specimens were performed CT thin-layer scanning and three-dimensional reconstruction after operation. The Tomasino method was used to evaluate the safety of the screws on the CT cross-sectional and sagittal images, to determine whether there was a cortical puncture of the lower and inner edges of the pedicle. According to the CT rating results, gradeⅠandⅡwere safe, and grade Ⅲ- Ⅴ were dangerous. And the accuracy of screws was recorded and analyzed between two groups. RESULTS: Two screws were inserted in each segment from C3 to C7 in 8 adult cadavers. A total of 80 screws were inserted, 40 in the 3D guide plate group, and 40 in the free-hand group. The Tomasino screw rating method was used to evaluate the safety of screw, 21 screws were gradeⅠ, 14 screws were gradeⅡ, 3 screws were grade Ⅲ, 1 screw was grade Ⅳ, 2 screws were grade Ⅴ in 3D guide plate group, while 14 screws were gradeⅠ, 8 screws were gradeⅡ, 8 screws were grade Ⅲ, 6 screws were grade Ⅳ, 2 screws were grade Ⅴ in free-hand group. The safety rate of 3D guide plate group was 87.5%, and 55.0% of the free hand group (χ2=8.7, P=0.003). CONCLUSION: The 3D printing rapid prototyping guide plate assisted insertion of the anterior cervical pedicle screw can significantly improve the accuracy and safety, and provide a theoretical basis for further clinical application.

RESUMO

In the central nervous system, hyperpolarization-activated cyclic nucleotide-gated (HCN) channels are essential to maintain normal neuronal function. Recent studies have shown that HCN channels may be involved in the pathological process of ischemic brain injury, but the mechanisms remain unclear. Autophagy is activated in cerebral ischemia, but its role in cell death/survival remains controversial. In this study, our results showed that the HCN channel blocker ZD7288 remarkably decreased the percentage of apoptotic neurons and corrected the excessive autophagy induced by oxygen-glucose deprivation followed by reperfusion (OGD/R) in hippocampal HT22 neurons. Furthermore, in the OGD/R group, p-mTOR, p-ULK1 (Ser757), and p62 were significantly decreased, while p-ULK1 (Ser317), atg5, and beclin1 were remarkably increased. ZD7288 did not change the expression of p-ULK1 (Ser757), ULK1 (Ser317), p62, Beclin1, and atg5, which are involved in regulating autophagosome formation. Besides, we found that OGD/R induced a significant increase in Cathepsin D expression, but not LAMP-1. Treatment with ZD7288 at 10 µmol/L in the OGD/R group did not change the expression of cathepsin D and LAMP-1. However, chloroquine (CQ), which decreases autophagosome-lysosome fusion, eliminated the correction of excessive autophagy and neuroprotection by ZD7288. Besides, shRNA knockdown of HCN2 channels significantly reduced the accumulation of LC3-II and increased neuron survival in the OGD/R and transient global cerebral ischemia (TGCI) models, and CQ also eliminated the effects of HCN2-shRNA. Furthermore, we found that the percentage of LC3-positive puncta that co-localized with LAMP-1-positive lysosomes decreased in Con-shRNA-transfected HT22 neurons exposed to OGD/R or CQ. In HCN2-shRNA-transfected HT22 neurons, the percentage of LC3-positive puncta that co-localized with LAMP-1-positive lysosomes increased under OGD/R; however, the percentage was significantly decreased by the addition of CQ to HCN2-shRNA-transfected HT22 neurons. The present results demonstrated that blockade of HCN2 channels provides neuroprotection against OGD/R and TGCI by accelerating autophagic degradation attributable to the promotion of autophagosome and lysosome fusion.

RESUMO

Bat flies are highly-specialized, hematophagous arthropods that are globally ubiquitous. There is little published research on bat flies (Diptera: Nycteribiidae) in Singapore and understanding the diversity of nycteribiids, host association and infestation rates can provide insight into this host-ectoparasite relationship. Nycteribiids were collected from bats trapped in Singapore (2011-2016) and identified using morphological keys. Host-ectoparasite relationships were investigated with logistic regression and Bayesian poisson regression. Nycteribiids were found to be monoxenously associated with their host bat species and host age, sex, species, and BBCI appear to contribute to differences in prevalence and intensity. Differences in host specificity between bat fly species in Singapore and their conspecifics in less disturbed habitats with higher bat biodiversity, such as Malaysia, Philippines and Thailand, suggest that the high host specificity in Singapore derives from the paucity of suitable hosts and abundance of single species roosts and not from their coevolved restrictions to them.

RESUMO

In deep underground engineering, in a large spatial, high-stress environment, rapid excavation is likely to affect the loading rate of the fault structure and to cause stick-slip. In this study, an experiment was conducted to explore the stick-slip characteristics at different loading rates. A double-sided shear experiment and the digital speckle correlation method were used to analyze the evolution of the displacement field, the slip displacement, and the slip rate of the fault's stick-slip activity at different loading rates as well as their correlation with the loading rate. The loading rate, moment magnitude, and stress drop of the fault's stick-slip and their corresponding relationships were studied. The results show that the occurrence of stick-slip is inversely proportional to the loading rate. The evolution of the fault-slip displacement field at different loading rates is similar. At a given loading rate, the magnitude is positively correlated with the stress drop. The magnitude and stress drop are inversely related to the loading rate.
