RESUMO

Eleven alkaloids (1-11) including seven new ones, 1-7, were isolated from the solid fermentation of Aspergillus fumigatus VDL36, an endophytic fungus isolated from the leaves of Vaccinium dunalianum Wight (Ericaceae), a perennial evergreen shrub distributed across the Southwest regions of China, Myanmar, and Vietnam. Their structures were elucidated on the basis of extensive spectroscopic methods. The isolates were evaluated for in vitro antifungal activities against five phytopathogenic fungi (Fusarium oxysporum, Coriolus versicolor, Fusarium solani, Botrytis cinerea, Fusarium graminearum). As a result, the new compounds fumigaclavine I (1), 13-ethoxycyclotryprostatin A (5), 13-dehydroxycyclotryprostatin A (6), and 12ß-hydroxy-13-oxofumitremorgin C (7) exhibited antifungal activities with MIC values of 7.8-62.5 µg/mL which were comparable to the two positive controls ketoconazole (MIC = 7.8-31.25 µg/mL) and carbendazim (MIC = 1.95-7.8 µg/mL). Furthermore, compounds 1 and 5 demonstrated potent protective and curative effects against the tomato gray mold in vivo. Preliminary structure-activity relationships of the tested indole diketopiperazine alkaloids indicate that the introduction of a substituent group at position C-13 enhances their biological activities.

Alongside fermentable sugars, weak acids, and furan derivatives, lignocellulosic hydrolysates contain non-negligible amounts of lignin-derived aromatic compounds. The biological funnel of lignin offers a new strategy for the "natural" production of protocatechuic acid (PCA). Herein, Pseudomonas putida KT2440 was engineered to produce PCA from lignin-derived monomers in hydrolysates by knocking out protocatechuate 3,4-dioxygenase and overexpressing vanillate-O-demethylase endogenously, while acetic acid was used for cell growth. The sugar catabolism was further blocked to prevent the loss of fermentable sugar. Using the engineered strain, a total of 253.88 mg/L of PCA was obtained with a yield of 70.85% from corncob hydrolysate 1. The highest titer of 433.72 mg/L of PCA was achieved using corncob hydrolysate 2 without any additional nutrients. This study highlights the potential ability of engineered strains to address the challenges of PCA production from lignocellulosic hydrolysate, providing novel insights into the utilization of hydrolysates.

Composites with excellent thermomechanical and thermochemical properties are urgently needed in the aerospace field, especially for structural applications under high-temperature conditions. Carbon fiber-reinforced Si-based composites are considered the most promising potential high-temperature materials due to their excellent oxidation resistance and ablative behaviors, good structural designability, and excellent mechanical properties. The reinforcement of the relevant composites mainly involves carbon fiber, which possesses good mechanical and temperature resistance abilities. In this paper, the ablation behaviors and mechanisms of related composites are reviewed. For carbon fiber-reinforced pure Si-based composites (C/SiM composites), the anti-ablation mechanism is mainly attributed to the continuous glassy SiO2, which inhibits the damage of the substrate. For C/SiM composite doping with refractory metal compounds, the oxides of Si and refractory metal together protect the main substrate from ablation and oxidation. Moreover, in addition to thermochemical damage, thermophysical and thermomechanical behavior severely destroy the surface coating of the substrate.

High-performance electromagnetic interference (EMI) shielding materials with ultralow density and environment-friendly properties are greatly demanded to address electromagnetic radiation pollution. Herein, carbon nanotube/polylactic acid (CNT/PLA) materials with different CNT contents, which exhibit characteristics of light weight, environmental protection and good chemical stability, are fabricated using 3D printing technology, where CNTs are evenly distributed and bind well with PLA. The performances of 3D-printed CNT/PLA composites are improved compared to pure 3D-printed PLA composites, which include mechanical properties, conductive behaviors and electromagnetic interference (EMI) shielding. The EMI shielding effectiveness (SE) of CNT/PLA composites could be improved when the content of CNTs increase. When it reaches 15 wt%, the EMI SE of 3D-printed CNT/PLA composites could get up to 47.1 dB, which shields 99.998% of electromagnetic energy. Meanwhile, the EMI shielding mechanism of 3D-printed CNT/PLA composites is mainly of absorption loss, and it generally accounts for more than 80% of the total shielding loss. These excellent comprehensive performances endow a 3D-printed CNT/PLA composite with great potential for use in industrial and aerospace areas.

Factor XI (FXI) deficiency is a rare bleeding disorder of unpredictable severity that correlates poorly with FXI coagulation activity and that poses great challenges for perioperative hemostatic management and the dialysis methods potentially available to new end-stage renal disease (ESRD) patients. We describe an individual with both ESRD and severe FXI deficiency, who successfully underwent peritoneal dialysis (PD) after emergency abdominal surgery. In the traditional concept, recent abdominal surgery is a contraindication to PD, especially for patients with bleeding risk. However, this case report highlights that PD can still be an possible option for patients with FXI deficiency who have just undergone abdominal surgery; laparoscopic PD catheter placement offers a chance to establish PD access in patients traditionally viewed as noncandidates for this method. Careful perioperative management and fresh frozen plasma transfusion ensure successful surgery. This case should be of help to clinicians and patients considering PD in similar situations.

Electronic textiles (e-textiles) hold great promise for serving as next-generation wearable electronics owing to their inherent flexible, air-permeable, and lightweight characteristics. However, these e-textiles are of limited performance mainly because of lacking powerful materials combination. Herein, a versatile e-textile through a simple, high-efficiency mixed-dimensional assembly of 2D MXene nanosheets and 1D silver nanowires (AgNWs) are presented. The effective complementary actions of MXene and AgNWs endow the e-textiles with superior integrated performances including self-powered pressure sensing, ultrafast joule heating, and highly efficient electromagnetic interference (EMI) shielding. The textile-based self-powered smart sensor systems obtained through the screen-printed assembly of MXene-based supercapacitor and pressure sensor are flexible and lightweight, showing ultrahigh specific capacitance (2390 mF cm-2 ), robust areal energy density (119.5 µWh cm-2 ), excellent sensitivity (474.8 kPa-1 ), and low detection limit (1 Pa). Furthermore, the interconnected conductive MXene/AgNWs network enables the e-textile with ultrafast temperature response (10.4 °C s-1 ) and outstanding EMI shielding effectiveness of ≈66.4 dB. Therefore, the proposed mixed-dimensional assembly design creates a multifunctional e-textile that offers a practical paradigm for next-generation smart flexible electronics.

BACKGROUND: Compared with sun-exposed melanomas, less is known regarding the pathogenesis of sun-protected melanomas. Sun-protected melanomas share many epidemiologic factors, but their genetic heterogeneity is not well studied. OBJECTIVE: We investigated the genomic profile of acral, mucosal, and vulvovaginal melanomas. We hypothesize that mucosal melanomas, recognized for their uniquely aggressive clinical behavior, have distinct genomic features. METHODS: We performed whole transcriptome messenger RNA and DNA (1711 genes) sequencing, messenger RNA expression profiling, tumor mutational burden, ultraviolet signature, and copy number variants analysis on 29 volar/digital acral, 7 mucosal, and 6 vulvovaginal melanomas. RESULTS: There was significant genetic heterogeneity, particularly in acral melanomas, with 36% having BRAF alterations, whereas other melanomas had none (P = .0159). Nonzero ultraviolet signatures were more frequent in acral melanomas, suggesting greater ultraviolet involvement. Mucosal melanomas formed a distinct group with increased expression of cell cycle and proliferation genes. Various targetable aberrations were identified, such as AURKA and ERBB2, in mucosal and acral melanomas, respectively. LIMITATIONS: The sample size was a small. CONCLUSION: There is significant genetic heterogeneity among sun-protected melanomas. Mucosal melanomas have upregulation in cell cycle and proliferation genes, which may explain their aggressive behavior. Ultraviolet radiation plays some role in a subset of acral but not other melanomas.

Cerebrocardiac syndrome (CCS) is one of the secondary myocardial injuries after stroke. Cerebrocardiac syndrome may result in a poor prognosis with high mortality. Understanding the mechanism of the brain-heart interaction may be crucial for clinical treatment of pathological changes in CCS. Accumulating evidence suggests that the inflammatory response is involved in the brain-heart interaction after stroke. Systemic inflammatory response syndrome (SIRS) evoked by stroke may injure myocardial cells directly, in which the interplay between inflammatory response, oxidative stress, cardiac sympathetic/parasympathetic dysfunction, and splenic immunoregulation may be also the key pathophysiology factor. This review article summarizes the current understanding of inflammatory response and immune regulation in brain-heart interaction after stroke.

BACKGROUND: The conversion of lignin-derived aromatic monomers into valuable chemicals has promising potential to improve the economic competitiveness of biomass biorefineries. Pinosylvin is an attractive pharmaceutical with multiple promising biological activities. RESULTS: Herein, Escherichia coli was engineered to convert the lignin-derived standard model monomer cinnamic acid into pinosylvin by introducing two novel enzymes from the wood plant: stilbene synthase from Pinus pinea (PpSTS) and 4-Coumarate-CoA ligase from Populus trichocarpa (Ptr4CL4). The expression of Ptr4CL4 drastically improved the production of pinosylvin (42.5 ± 1.1 mg/L), achieving values 15.7-fold higher than that of Ptr4CL5 (another 4-Coumarate-CoA ligase from Populus trichocarpa) in the absence of cerulenin. By adjusting the expression strategy, the optimized engineered strain produced pinosylvin at 153.7 ± 2.2 mg/L with an extremely high yield of 1.20 ± 0.02 mg/mg cinnamic acid in the presence of cerulenin, which is 83.9% ± 1.17 of the theoretical yield. This is the highest reported pinosylvin yield directly from cinnamic acid to date. CONCLUSION: Our work highlights the feasibility of microbial production of pinosylvin from cinnamic acid and paves the way for converting lignin-related aromatics to valuable chemicals.

As a dehydration product of pentoses in hemicellulose sugar streams derived from lignocellulosic biomass, furfural is a prevalent inhibitor in the efficient microbial conversion process. To solve this obstacle, exploiting a biorefinery strain with remarkable furfural tolerance capability is essential. Pseudomonas putida KT2440 (P. putida) has served as a valuable bacterial chassis for biomass biorefinery. Here, a high-concentration furfural-tolerant P. putida strain was developed via adaptive laboratory evolution (ALE). The ALE resulted in a previously engineered P. putida strain with substantially increased furfural tolerance as compared to wild-type. Whole-genome sequencing of the adapted strains and reverse engineering validation of key targets revealed for the first time that several genes and their mutations, especially for PP_RS19785 and PP_RS18130 [encoding ATP-binding cassette (ABC) transporters] as well as PP_RS20740 (encoding a hypothetical protein), play pivotal roles in the furfural tolerance and conversion of this bacterium. Finally, strains overexpressing these three striking mutations grew well in highly toxic lignocellulosic hydrolysate, with cell biomass around 9-, 3.6-, and two-fold improvement over the control strain, respectively. To our knowledge, this study first unravels the furan aldehydes tolerance mechanism of industrial workhorse P. putida, which provides a new foundation for engineering strains to enhance furfural tolerance and further facilitate the valorization of lignocellulosic biomass.

This paper provides a comprehensive understanding of the amplitude- and frequency-dependent characteristics of rubber springs. The dynamic nonlinear inelasticity of rubber is a key academic problem for continuum mechanics and a bottleneck problem for the practical use of rubber structures. Despite intensive efforts witnessed in industrial applications, it still demands an unambiguous constitutive model for dynamic nonlinear inelasticity, which is known as the Payne effect. To this end, three types of rubber springs (shear-type (ST), compression-type (CT) and shear-compression-combination-type (SCCT)) were tested with amplitude and frequency sweeps in different conditions. We investigated and present changes in dynamic stiffness and loss factor with amplitude, frequency and the hysteresis loops of different rubber springs. We also propose a hypothesis and research strategy to study a constitutive model involving multiple factors of hyperelasticity, the Mullins effect, viscoelasticity and the Payne effect, which we hope will provide new ideas for the establishment of a constitutive equation.

Assisted reproductive technology has continued to develop in recent years, the technology has become more and more mature, and it has improved the total gestational age of the population. Assisted reproductive technology has improved twin pregnancy rates with the use of ovulation-inducing drugs. The risk factor of twins is much higher than singletons, and adverse pregnancy outcomes such as stillbirth and premature birth are more likely to occur than singletons, especially the special complications of monochorionic twins (MC), and the neonatal mortality and morbidity are also increased. Prenatal diagnosis and prognosis of twin pregnancy, as well as therapeutic interventions, are of current concern. Ultrasound can realize the understanding of intrauterine growth and development of twin pregnancy fetuses during pregnancy, can more accurately determine gestational age, organ function, and maturity, and timely detect fetal growth and development abnormalities in twin pregnancy, while the prognosis and treatment still need further improvement. The purpose of this study was to investigate the clinical value and treatment progress of prenatal ultrasound monitoring in twin pregnancy.

Flexible, lightweight, and durable electromagnetic interference (EMI) shielding materials are urgently required to solve the increasingly serious electromagnetic radiation pollution. Transition metal carbides/nitrides (MXenes) are promising candidates for EMI shielding materials because of their excellent metallic electrical conductivity. However, MXenes are highly susceptible to oxidization when exposed to wet environments, leading to the loss of their functional properties and degradation of reliability and stability. Herein, an interfused core-shell heterogeneous reduced graphene oxide (rGO)/MXene aerogel (GMA) is designed for the first time via coaxial wet spinning and freeze-drying. The fabricated GMAs exhibit excellent EMI shielding performance, and the EMI shielding effectiveness (SE) and specific EMI SE can be up to 83.3 dB and 3119 dB·cm3/g, respectively, which is higher than most carbon-based and MXene-based aerogels and foams. More importantly, GMAs have only a 17.4 % degradation in EMI shielding performance after 120 days due to the protection of hydrophobic graphene sheath, exhibiting superior EMI shielding durability to its MXene film counterpart. Moreover, the hydrophobic GMAs exhibit good oil/water separation and thermal insulation performance. The interfused core-shell GMAs are highly promising for applications in durable EMI shielding, thermal insulation, oil/water separation and sensors, etc.

Aberrant activity of the H3K27 modifiers EZH2 and BRD4 is an important oncogenic driver for atypical teratoid/rhabdoid tumor (AT/RT), and each is potentially a possible therapeutic target for treating AT/RT. We, therefore, determined whether targeting distinct histone modifier activities was an effective approach for treating AT/RT. The effects of EZH2 and BRD4 inhibition on histone modification, cell proliferation, and cell invasion were analyzed by immunoblotting, MTS assay, colony formation assay, and cell invasion assay. RNA- and chromatin immunoprecipitation-sequencing were used to determine transcriptional and epigenetic changes in AT/RT cells treated with EZH2 and BRD4 inhibitors. We treated mice bearing human AT/RT xenografts with EZH2 and BRD4 inhibitors. Intracranial tumor growth was monitored by bioluminescence imaging, and the therapeutic response was evaluated by animal survival. AT/RT cells showed elevated levels of H3K27 trimethylation (H3K27me3) and H3K27 acetylation (H3K27ac), with expression of EZH2 and BRD4, and lack of SMARCB1 proteins. Targeted inhibition of EZH2 and BRD4 activities reduced cell proliferation and invasiveness of AT/RT in association with decreasing H3K27me3 and H3K27ac. Differential genomic occupancy of H3K27me3 and H3K27ac regulated specific gene expression in response to EZH2 and BRD4 inhibitions. A combination of EZH2 and BRD4 inhibition increased the therapeutic benefit in vitro and in vivo, outperforming either monotherapy. Overall, histones H3K27me3 and H3K27ac were elevated in AT/RT cells and distributed in distinct chromatin regions to regulate specific gene expression and to promote AT/RT growth. Targeting EZH2 and BRD4 activity is, therefore, a potential combination therapy for AT/RT.

1,2-Propanediol is an important building block as a component used in the manufacture of unsaturated polyester resin, antifreeze, biofuel, nonionic detergent, etc. Commercial production of 1,2-propanediol through microbial biosynthesis is limited by low efficiency, and chemical production of 1,2-propanediol requires petrochemically derived routes involving wasteful power consumption and high pollution emissions. With the development of various strategies based on metabolic engineering, a series of obstacles are expected to be overcome. This review provides an extensive overview of the progress in the microbial production of 1,2-propanediol, particularly the different micro-organisms used for 1,2-propanediol biosynthesis and microbial production pathways. In addition, outstanding challenges associated with microbial biosynthesis and feasible metabolic engineering strategies, as well as perspectives on the future microbial production of 1,2-propanediol, are discussed.

BACKGROUND: Lignocellulosic biomass is an attractive and sustainable alternative to petroleum-based feedstock for the production of a range of biochemicals, and pretreatment is generally regarded as indispensable for its biorefinery. However, various inhibitors that severely hinder the growth and fermentation of microorganisms are inevitably produced during the pretreatment of lignocellulose. Presently, there are few reports on a single microorganism that can detoxify or tolerate toxic mixtures of pretreated lignocellulose hydrolysate while effectively transforming sugar components into valuable compounds. Alternatively, microbial coculture provides a simpler and more efficacious way to realize this goal by distributing metabolic functions among different specialized strains. RESULTS: In this study, a novel synthetic microbial consortium, which is composed of a responsible for detoxification bacterium engineered Pseudomonas putida KT2440 and a lactic acid production specialist Bacillus coagulans NL01, was developed to directly produce lactic acid from highly toxic lignocellulosic hydrolysate. The engineered P. putida with deletion of the sugar metabolism pathway was unable to consume the major fermentable sugars of lignocellulosic hydrolysate but exhibited great tolerance to 10 g/L sodium acetate, 5 g/L levulinic acid, 10 mM furfural and HMF as well as 2 g/L monophenol compound. In addition, the engineered strain rapidly removed diverse inhibitors of real hydrolysate. The degradation rate of organic acids (acetate, levulinic acid) and the conversion rate of furan aldehyde were both 100%, and the removal rate of most monoaromatic compounds remained at approximately 90%. With detoxification using engineered P. putida for 24 h, the 30% (v/v) hydrolysate was fermented to 35.8 g/L lactic acid by B. coagulans with a lactic acid yield of 0.8 g/g total sugars. Compared with that of the single culture of B. coagulans without lactic acid production, the fermentation performance of microbial coculture was significantly improved. CONCLUSIONS: The microbial coculture system constructed in this study demonstrated the strong potential of the process for the biosynthesis of valuable products from lignocellulosic hydrolysates containing high concentrations of complex inhibitors by specifically recruiting consortia of robust microorganisms with desirable characteristics and also provided a feasible and attractive method for the bioconversion of lignocellulosic biomass to other value-added biochemicals.

Durable electromagnetic interference (EMI) shielding is highly desired, as electromagnetic pollution is a great concern for electronics' stable performance and human health. Although a superhydrophobic surface can extend the service lifespan of EMI shielding materials, degradation of its protection capability and insufficient self-healing are troublesome issues due to unavoidable physical/chemical damages under long-term application conditions. Here, we report, for the first time, an instantaneously self-healing approach via microwave heating to achieve durable shielding performance. First, a hydrophobic 1H,1H,2H,2H-perfluorooctyltriethoxysilane (POTS) layer was coated on a polypyrrole (PPy)-modified fabric (PPy@POTS), enabling protection against the invasion of water, salt solution, and corrosive acidic and basic solutions. Moreover, after being damaged, the POTS layer can, for the first time, be instantaneously self-healed via microwave heating for a very short time, i.e., 4 s, benefiting from the intense thermal energy generated by PPy under electromagnetic wave radiation. This self-healing ability is also repeatable even after intentionally severe plasma etching, which highlights the great potential to achieve robust and durable EMI shielding applications. Significantly, this approach can be extended to other EMI shielding materials where heat is a triggering stimulus for healing thin protection layers. We envision that this work could provide insights into fabricating EMI shielding materials with durable performance for portable and wearable devices, as well as for human health care.

Molecular imprinting represents one of the most promising strategies to design artificial enzyme inhibitors. However, the study of molecularly imprinted enzyme inhibitors (MIEIs) remains at a primary stage. Advanced applications of MIEIs for cell regulation have rarely been explored. Using a solid-phase oriented imprinting strategy so as to leave the active site of the enzymes accessible, we synthesized two MIEIs that exhibit high specificity and potent inhibitory effects (inhibition constant at low nM range) towards trypsin and angiogenin. The trypsin MIEI inhibits trypsin activity, tryptic digestion-induced extracellular matrix lysis and cell membrane destruction, indicating its utility in the treatment of active trypsin-dependent cell injury. The angiogenin MIEI blocks cancer cell proliferation by suppressing the ribonuclease activity of angiogenin and decreasing the angiogenin level inside and outside HeLa cells. Our work demonstrates the versatility of MIEIs for both enzyme inhibition and cell fate manipulation, showing their great potential as therapeutic drugs in biomedicine.

In this work, two typical fluorescent sensors were generated by exploiting molecularly imprinted polymeric hydrogels (MIPGs) for zearalenone (ZON) and glucuronic acid (GA) detection, via the analyte's self-fluorescence property and receptor's fluorescence effect, respectively. Though significant advances have been achieved on MIPG-fluorescent sensors endowed with superior stability over natural receptor-sensors, there is an increasing demand for developing sensing devices with cost-effective, easy-to-use, portable advantages in terms of commercialization. Zooming in on the commercial potential of MIPG-fluorescent sensors, the MIPG_ZON is synthesized using zearalanone (an analogue of ZON) as template, which exhibits good detection performance even in corn samples with a limit of detection of 1.6 µM. In parallel, fluorescein-incorporated MIPG_GA is obtained and directly used for cancer cell imaging, with significant specificity and selectivity. Last but not least, our consolidated application results unfold new opportunities for MIPG-fluorescent sensors for environmentally and medicinally important analytes detection.

A molecularly imprinted polymer (MIP), which is synthesized by a nanomolding process around a template, has emerged as a promising analytical tool for environmental quality monitoring and food safety test. In this work, a fluoroprobe with Ag-doped MIP nanolayer (16 nm thickness) is successfully prepared for the highly selective detection of oxytetracycline (OTC) in real samples (i.e. Yangtze River water, swine urine). In the MIP nanolayer, two functional monomers (i.e. 4-(2-acrylamidoethylcarbamoyl)-3-fluorophenylboronic acid, methacrylic acid) synergistically constitute the specific recognition sites. Meanwhile, the doped Ag enhances the detection sensitivity (with a detection limit of 5.38 nM) and accelerates the detection rate (within 2.5 min) even in real samples. Therefore, the present study paves the way for the preparation of MIP-based fluoroprobes, showing great prospects in environmental quality and food safety tests.
