RESUMO

Sexual selection is a field coloured by tension and contrasting views. One contested claim is the causal link from the definition of the sexes (anisogamy) to divergent selection on the sexes. Does theory truly engage with this claim? We survey the extent to which theory makes sex-specific assumptions and engages with anisogamy, and discuss these issues in a broader context. The majority of theory in sexual selection makes sex-specific assumptions and does not engage with the definition of the sexes. While this does not invalidate existing results, debates and criticisms regarding sexual selection force us to think deeper about its logical foundations. We discuss ways to strengthen the foundations of sexual selection theory by relaxing central assumptions.

BACKGROUND: Why the adaptive immune system turns against citrullinated antigens in rheumatoid arthritis (RA) and whether anti-citrullinated protein antibodies (ACPAs) contribute to pathogenesis are questions that have triggered intense research, but still are not fully answered. Neutrophils may be crucial in this context, both as sources of citrullinated antigens and also as targets of ACPAs. To better understand how ACPAs and neutrophils contribute to RA, we studied the reactivity of a broad spectrum of RA patient-derived ACPA clones to activated or resting neutrophils, and we also compared neutrophil binding using polyclonal ACPAs from different patients. METHODS: Neutrophils were activated by Ca2+ ionophore, PMA, nigericin, zymosan or IL-8, and ACPA binding was studied using flow cytometry and confocal microscopy. The roles of PAD2 and PAD4 were studied using PAD-deficient mice or the PAD4 inhibitor BMS-P5. RESULTS: ACPAs broadly targeted NET-like structures, but did not bind to intact cells or influence NETosis. We observed high clonal diversity in ACPA binding to neutrophil-derived antigens. PAD2 was dispensable, but most ACPA clones required PAD4 for neutrophil binding. Using ACPA preparations from different patients, we observed high patient-to-patient variability in targeting neutrophil-derived antigens and similarly in another cellular effect of ACPAs, the stimulation of osteoclast differentiation. CONCLUSIONS: Neutrophils can be important sources of citrullinated antigens under conditions that lead to PAD4 activation, NETosis and the extrusion of intracellular material. A substantial clonal diversity in targeting neutrophils and a high variability among individuals in neutrophil binding and osteoclast stimulation suggest that ACPAs may influence RA-related symptoms with high patient-to-patient variability.

AbstractWhen selection differs between the sexes, genes expressed by both males and females can experience sexually antagonistic (SA) selection, where beneficial alleles for one sex are deleterious for the other. Classic population genetics theory has been fundamental to understanding how and when SA genetic variation can be maintained by balancing selection, but these models have rarely considered the demographic consequences of coexisting alleles with deleterious fitness effects in each sex. In this article, we develop a stage-structured Mendelian matrix model and jointly analyze the evolutionary and demographic consequences of SA selection in obligately outcrossing (i.e., dioecious/gonochorous) and partially selfing hermaphrodite populations. We focus on identifying when SA polymorphisms are maintained by balancing selection and the population growth rate remains positive. Additionally, we analyze the effects of inbreeding depression manifesting at different life history stages and give an illustrative example of the potential for SA polymorphism in real populations using empirically estimated demographic rates for the hermaphroditic flowering plant Mimulus guttatus. Our results show that when population intrinsic growth rates approach 1, extinction occurs across large swathes of parameter space, favoring SA polymorphism or the fixation of male-beneficial alleles, and that inbreeding depression is a significant problem for maintaining SA polymorphism in partially selfing populations. Despite these demographic challenges, our example with M. guttatus appears to show that demographic rates observed in some real populations can sustain large regions of viable SA polymorphic space.

Based on the epidemiological link between periodontitis and rheumatoid arthritis (RA), and the unique feature of the periodontal bacterium Porphyromonas gingivalis to citrullinate proteins, it has been suggested that production of anti-citrullinated protein antibodies (ACPA), which are present in a majority of RA patients, may be triggered in the gum mucosa. To address this hypothesis, we investigated the antibody response to a citrullinated P. gingivalis peptide in relation to the autoimmune ACPA response in early RA, and examined citrulline-reactivity in monoclonal antibodies derived from human gingival B cells. Antibodies to a citrullinated peptide derived from P. gingivalis (denoted CPP3) and human citrullinated peptides were analyzed by multiplex array in 2,807 RA patients and 372 controls; associations with RA risk factors and clinical features were examined. B cells from inflamed gingival tissue were single-cell sorted, and immunoglobulin (Ig) genes were amplified, sequenced, cloned and expressed (n=63) as recombinant monoclonal antibodies, and assayed for citrulline-reactivities by enzyme-linked immunosorbent assay. Additionally, affinity-purified polyclonal anti-cyclic-citrullinated peptide (CCP2) IgG, and monoclonal antibodies derived from RA blood and synovial fluid B cells (n=175), were screened for CPP3-reactivity. Elevated anti-CPP3 antibody levels were detected in RA (11%), mainly CCP2+ RA, compared to controls (2%), p<0.0001, with a significant association to HLA-DRB1 shared epitope alleles, smoking and baseline pain, but with low correlation to autoimmune ACPA fine-specificities. Monoclonal antibodies derived from gingival B cells showed cross-reactivity between P. gingivalis CPP3 and human citrullinated peptides, and a CPP3+/CCP2+ clone, derived from an RA blood memory B cell, was identified. Our data support the possibility that immunity to P. gingivalis derived citrullinated antigens, triggered in the inflamed gum mucosa, may contribute to the presence of ACPA in RA patients, through mechanisms of molecular mimicry.

There is accumulating data suggesting that periodontitis is associated with increased risk of systemic and autoimmune diseases, including cardiovascular disease, rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE), and there is an unmet need to identify these individuals early. With the periodontal bacteria Porphyromonas gingivalis (Pg) as one of the key drivers of periodontitis, we set out to investigate whether antibodies to Pg virulence factor arginine gingipain (Rgp) could serve as a biomarker for periodontitis patients at increased risk of autoimmunity and systemic disease. We measured serum anti-Rgp IgG in three study populations: PAROKRANK (779 individuals with myocardial infarction (MI); 719 controls), where 557 had periodontitis, and 312 were positive for autoantibodies associated with RA/SLE; the PerioGene North pilot (41 periodontitis; 39 controls); and an SLE case/control study (101 SLE; 100 controls). Anti-Rgp IgG levels were increased in severe periodontitis compared to controls (p < 0.0001), in individuals positive for anti-citrullinated protein antibodies (p = 0.04) and anti-dsDNA antibodies (p = 0.035), compared to autoantibody-negative individuals; and in MI patients versus matched controls (p = 0.035). Our data support longitudinal studies addressing the role of anti-Rgp antibodies as biomarkers for periodontitis patients at increased risk of developing autoimmunity linked to RA and SLE, and mechanisms underpinning these associations.

The outcome of natural selection depends on the demographic processes of birth, death, and development. Here, we derive conditions for protected polymorphism in a population characterized by age- or stage-dependent demography with two sexes. We do so using a novel two-sex matrix population model including basic Mendelian genetics (one locus, two alleles, random mating). Selection may operate on survival, growth, or fertility, any or all of which may differ between the sexes. The model can therefore incorporate genes with arbitrary pleiotropic and sex-specific effects. Conditions for protected polymorphism are expressed in terms of the eigenvalues of the linearization of the model at the homozygote boundary equilibria. We show that in the absence of sexual dimorphism, polymorphism requires heterozygote superiority in the genotypic population growth rate. In the presence of sexual dimorphism, however, heterozygote superiority is not required; an inferior heterozygote may invade, reducing the population growth rate and even leading to extinction (so-called evolutionary suicide). Our model makes no assumptions about separation of time scales between ecological and evolutionary processes, and can thus be used to project sex×stage×genotype dynamics of eco-evolutionary processes. Empirical evidence that sexual dimorphism affects extinction risk is growing, yet sex differences are often ignored in evolutionary demography and in eco-evolutionary models. Our analysis highlights the importance of sexual dimorphism and suggests mechanisms by which an allele can be favored by selection, yet drive a population to extinction, as a result of the structure and interdependence of sex- and stage-specific processes.

Demographic processes and ecological interactions are central to understanding evolution and vice versa. We present a novel framework that combines basic Mendelian genetics with the powerful demographic approach of matrix population models. The ecological components of the model may be stage classified or age classified, linear or nonlinear, time invariant or time varying, and deterministic or stochastic. Genotypes may affect, in fully pleiotropic fashion, any mixture of demographic traits (viability, fertility, development) at any points in the life cycle. The dynamics of the stage × genotype structure of the population are given by a nonlinear population projection matrix. We show how to construct this matrix and use it to derive sufficient conditions for a protected genetic polymorphism for the case of linear, time-independent demography. These conditions demonstrate that genotype-specific population growth rates (λ) do not determine the outcome of selection. Except in restrictive special cases, heterozygote superiority in λ is neither necessary nor sufficient for a genetic polymorphism. As a consequence, the population growth rate does not always increase, and populations can be driven to extinction due to evolutionary suicide. We demonstrate the construction and analysis of the model using data on a color polymorphism in the common buzzard (Buteo buteo). The model exhibits a stable genetic polymorphism and declining growth rate, consistent with field data and previous models.

This paper presents a comprehensive theory for the demographic analysis of populations in which individuals are classified by both age and stage. The earliest demographic models were age classified. Ecologists adopted methods developed by human demographers and used life tables to quantify survivorship and fertility of cohorts and the growth rates and structures of populations. Later, motivated by studies of plants and insects, matrix population models structured by size or stage were developed. The theory of these models has been extended to cover all the aspects of age-classified demography and more. It is a natural development to consider populations classified by both age and stage. A steady trickle of results has appeared since the 1960s, analyzing one or another aspect of age × stage-classified populations, in both ecology and human demography. Here, we use the vec-permutation formulation of multistate matrix population models to incorporate age- and stage-specific vital rates into demographic analysis. We present cohort results for the life table functions (survivorship, mortality, and fertility), the dynamics of intra-cohort selection, the statistics of longevity, the joint distribution of age and stage at death, and the statistics of life disparity. Combining transitions and fertility yields a complete set of population dynamic results, including population growth rates and structures, net reproductive rate, the statistics of lifetime reproduction, and measures of generation time. We present a complete analysis of a hypothetical model species, inspired by poecilogonous marine invertebrates that produce two kinds of larval offspring. Given the joint effects of age and stage, many familiar demographic results become multidimensional, so calculations of marginal and mixture distributions are an important tool. From an age-classified point of view, stage structure is a form of unobserved heterogeneity. From a stage-classified point of view, age structure is unobserved heterogeneity. In an age × stage-classified model, variance in demographic outcomes can be partitioned into contributions from both sources. Because these models are formulated as matrices, they are amenable to a complete sensitivity analysis. As more detailed and longer longitudinal studies are developed, age × stage-classified demography will become more common and more important.

History matters when individual prior conditions contain important information about the fate of individuals. We present a general framework for demographic models which incorporates the effects of history on population dynamics. The framework incorporates prior condition into the i-state variable and includes an algorithm for constructing the population projection matrix from information on current state dynamics as a function of prior condition. Three biologically motivated classes of prior condition are included: prior stages, linear functions of current and prior stages, and equivalence classes of prior stages. Taking advantage of the matrix formulation of the model, we show how to calculate sensitivity and elasticity of any demographic outcome. Prior condition effects are a source of inter-individual variation in vital rates, i.e., individual heterogeneity. As an example, we construct and analyze a second-order model of Lathyrus vernus, a long-lived herb. We present population growth rate, the stable population distribution, the reproductive value vector, and the elasticity of λ to changes in the second-order transition rates. We quantify the contribution of prior conditions to the total heterogeneity in the stable population of Lathyrus using the entropy of the stable distribution.

INTRODUCTION AND HYPOTHESIS: Obstetric anal sphincter injuries (OASIS) are associated with an increased risk of faecal incontinence after vaginal delivery. The aim of this retrospective population-based cohort study was to assess whether mediolateral episiotomy is associated with a reduction in the rate of OASIS during operative vaginal delivery. METHODS: We used data from the Dutch Perinatal Registry (Perined) that includes records of almost all births between 2000 and 2010 in The Netherlands. In a cohort of 170,969 primiparous and multiparous women whose delivery was recorded, we estimated the association between mediolateral episiotomy and OASIS following both vacuum and forceps deliveries using univariate and multivariate logistic regression analysis. RESULTS: The incidences of OASIS following vacuum delivery in 130,157 primiparous women were 2.5% and 14% in those with and without a mediolateral episiotomy, respectively (adjusted OR 0.14, 95% CI 0.13-0.15), and in 29,183 multiparous women were 2.0% and 7.5%, respectively (adjusted OR 0.23, 95% CI 0.21-0.27). The incidences of OASIS following forceps delivery in 9,855 primiparous women were 3.4% and 26.7% in those with and without a mediolateral episiotomy, respectively (adjusted OR 0.09, 95% CI 0.07-0.11), and in 1,774 multiparous women were 2.6% and 14.2%, respectively (adjusted OR 0.13, 95% CI 0.08-0.22). CONCLUSIONS: The use of a mediolateral episiotomy during both vacuum delivery and forceps delivery is associated with a fivefold to tenfold reduction in the rate of OASIS in primiparous and multiparous women.

In the design of artefacts, tasks and environments for human use, the body dimensions of the target population are a critical element in spatial optimisation of the design. This study examines how the choices designers make affect the ability of different user groups to safely and effectively interact with a designed artefact. Due to the variability in body size and shape across different demographic groups, heterogeneous user populations are unlikely to experience uniform levels of performance. The associated variability in the rate of unacceptable user conditions is referred to here as disproportionate disaccommodation. This is both an ethical and a performance concern that can partially be addressed through improved design practice. Three methods for incorporating the consideration of user demographics and the corresponding variability in body size and shape are presented. They are compared with a baseline strategy in terms of accommodation and cost.

RESUMO

Our daily lives are a series of ordinary moments and unnoticed thresholds-times that define us in ways we often do not give much attention. While we consider childbirth to be one of life's extra-ordinary events, the hours of labor and birth need not be dramatic (or traumatic) ones. I describe a quiet, well-supported birth in the Netherlands that is cause for celebration of the beauty of an ordinary instant that can define and enrich the human experience.

Childbirth education was an important social movement in the 20th century but has lost its way in recent years. We describe the reasons for the dwindling importance of childbirth education and offer a proposal for reform that will align childbirth education with the needs of today's birthing mothers. Our plan will create "Centers for the Childbearing Year" (CCBYs) and a new model of childbirth educator, which we call the "birth coach." The CCBY is the place for women to go to for information and support related to fertility, pregnancy, childbirth, and newborn care; the birth coach combines the role of childbirth educator, doula, and postpartum caregiver. In creating a fresh model of childbirth education, we not only honor our pioneers but also rediscover the wisdom in community and relationship that childbirth offers us, and we learn in new ways to journey alongside each other to create new possibilities for birthing families.
