RESUMO

The ranaviruses are an emerging group of viruses that infect amphibians, fish, and reptiles. Although ranaviruses have not been linked to extinctions, emergence in amphibian communities has resulted in population declines for some species. We present the results of ranavirus-associated mortality in a Dutch national park in the aftermath of an outbreak associated with a common midwife toad virus (CMTV)-like ranavirus. We monitored five bodies of water across Dwingelderveld National Park, the Netherlands, in 2011-13. Dead and live amphibians were counted weekly July-September and every 2 wk in June and October. Dead amphibians were collected and tested for ranavirus infection. In addition, we measured biologic, chemical, and physical site characteristics to test for a correlation with ranavirus-associated mortality. Ranavirus infection was widespread in our study area and we observed nearly continuous presence of dead, ranavirus-infected amphibians in the presence of asymptomatic, live amphibians throughout our study. Fatalities occurred in larval, subadult, and adult amphibians. Ranavirus infection prevalence (based on fatal cases) was significantly associated with increasing fractions of adults and subadults compared to juveniles and larvae in the population, but was unrelated to any other measured site characteristics. Our findings showed that a CMTV-like ranavirus can persist long term in an ecosystem, affecting a diversity of amphibian species and life stages for a prolonged period. This study illustrates the importance of monitoring the modes of spread for ranaviruses and their impact on amphibian populations.

The relative contributions of the flagellum and the flagella-associated bacterial motility in the invasion of Caco-2 cells by Salmonella serotype Enteritidis were investigated using an fliC mutant defective in flagellin production and a motA mutant that carries flagella but is non-motile. Infection assays demonstrated that, at 1 h of infection, both the fliC and the motA mutants were severely impaired in bacterial invasion compared to the parental strain. Infection assays at 3 h infection demonstrated virtually equal invasion levels for both non-motile mutants and the parental strain. Together these data suggest that flagella-mediated bacterial motility accelerates the invasion of Salmonella but is not required for the invasion event per se.

Feline amyloidosis is an uncommon disorder caused by deposits of amyloid in a variety of organs. Most frequently encountered types are amyloid derived of pancreatic islet amyloid polypeptide (AIAPP) in older cats and of the apolipoprotein, apo-serum amyloid A (AA) in Abyssinian/Somali (Aby) and Siamese/Oriental (Siam) cats occurring at a relatively young age. For the AA protein of the Aby, Siam and domestic shorthair cat (DSH) breed different amino acid sequences have been described. It is not yet clear, however, whether the tendency to develop AA amyloidosis in Aby and Siam is associated with specific apoSAA protein sequences and whether this is breed specific. In this study, DNA from one Siam and two DSH cats revealed on Southern blot three bands suggesting at least three genes or gene clusters. The SAA cDNAs of hepatic mRNA from three Abys, five Siams and five DSHs were amplified by RT-PCR, cloned and sequenced. Siams and Abys had limited SAA sequence variability. All five Siams, three of which were positive for amyloid, had the amyloidogenic Siam SAA and the amyloidogenic Aby SAA sequence. Two of the Abys, both with amyloid, had the amyloidogenic Aby SAA sequence. The third Aby, without amyloid, missed its amyloidogenic sequence. The SAA sequences of the DSHs found in the present preliminary survey, suggested a possible tendency for more variability, whereas the amyloidogenic Siam as well as the amyloidogenic Aby sequence were found once. Up to five different sequences were found in a single animal. All five DSHs, moreover, had a specific sequence lacking in the Siams and Abys. The present results, especially those of the Siams, favor that in addition to the occurrence of amyloid associated SAA genes other factors such as infections and inflammatory processes are involved in the development of phenotypical amyloidosis.

The enterocytes of the small intestine are occasionally exposed to pathogenic bacteria, such as Salmonella enteritidis 857, an etiologic agent of intestinal infections in humans. The expression of the heat shock response by enterocytes may be part of a protective mechanism developed against pathogenic bacteria in the intestinal lumen. We aimed at investigating whether S. enteritidis 857 is able to induce a heat shock response in crypt- and villus-like Caco-2 cells and at establishing the extent of the induction. To establish whether S. enteritidis 857 interfered with the integrity of the cell monolayer, the transepithelial electrical resistance (TEER) of filter-grown, differentiated (villus-like) Caco-2 cells was measured. We clearly observed damage to the integrity of the cell monolayer by measuring the TEER. The stress response was screened in both crypt- and villus-like Caco-2 cells exposed to heat (40-43 degrees C) or to graded numbers (10(1)-10(8)) of bacteria and in villus-like cells exposed to S. enteritidis 857 endotoxin. Expression of the heat shock proteins Hsp70 and Hsp90 was analyzed by polyacrylamide gel electrophoresis and immunoblotting with monoclonal antibodies. Exposure to heat or Salmonella resulted in increased levels of Hsp70 and Hsp90 in a temperature-effect or Salmonella-dose relationship, respectively. Incubation of Caco-2 cells with S. enteritidis 857 endotoxin did not induce heat shock gene expression. We conclude that S. enteritidis 857 significantly increases the levels of stress proteins in enterocyte-like Caco-2 cells. However, our data on TEER clearly indicate that this increase is insufficient to protect the cells.
