RESUMO

Preserved feed from meadows contaminated with ragwort (Jacobaea vulgaris, Gaertn.) may expose livestock to pyrrolizidine alkaloids (PA). Dairy cows are considered to be very susceptible animals and a PA ingestion can lead to liver and further organ damages and even death. Due to the lack of data, the present study aimed to evaluate critical PA doses based on organ effects, with a special focus on liver lesions and on indicators of energy metabolism. Therefore, 16 dairy cows (n = 4 per group) were exposed to increasing PA doses (group: CONMolasses: <0.001 mg PA/kg body weight (BW)/day (d); PA1: 0.47 mg PA/kg BW/d; PA2: 0.95 mg PA/kg BW/d; PA3: 1.91 mg PA/kg BW/d) for 28 days. Constant dosing was ensured by a defined PA extract administered orally once daily. Histological examinations of the livers showed infiltration by immune cells, higher proportions of apoptotic cells and enlargement of hepatocyte nuclei in the highest exposed group. In addition, bile volume increased with PA dose, which may indicate a cholestasis. Despite the signs of incipient liver damage, liver lipid content and clinical chemical parameters related to energy metabolism, such as glucose, non-esterified fatty acids and ßhydroxybutyrate, remained unaffected. Fat depot masses were also not significantly altered over time, suggesting that PA exposure did not induce a wasting syndrome. The liver showed slight microscopic changes already at a dosage of 0.95 mg PA/kg BW/d. However, the short-term metabolic indicators of energy status, lipolysis and ketogenesis, glucose, NEFA and BHB, as well as changes in fat depot, which serves as a longer-term indicator of lipolysis, remained unaffected in all treatment groups in the chosen scenario. These findings suggest that despite histopathological and clinical-chemical evidence of PA-associated hepatocellular lesions, liver function was not compromised.

The aim of the study was to investigate the effect of N-arachidonoylethanolamide (AEA), an endocannabinoid with orexigenic characteristics, on plasma endocannabinoid concentrations, feed intake, energy balance, lipomobilisation, and hepatic lipid metabolism of early-lactating dairy cows. The experiment involved 10 pairs of Holstein half-sibling cows (end of 2nd-3rd pregnancy). Half-sibs of each pair were randomly assigned to either AEA (n = 10) or control (CON) group (n = 10). From day 1 to 30 postpartum, the AEA group received 5 intraperitoneal injections per week of 3 µg/kg body weight AEA and the CON group 0.9% NaCl. In week 1-3 postpartum, AEA administration had no effect on dry matter intake, body weight, or lipomobilisation, but increased plasma triglyceride concentration on d 21 p.p. and mRNA abundances of genes related to hepatic triglyceride synthesis. In week 4 postpartum, the AEA group showed reduced feed intake and whole-body carbohydrate oxidation, but increased whole-body fat oxidation and hepatic lipid accumulation, likely as a result of a counter-regulatory leptin increase. In conclusion, the present study shows a tissue-specific AEA insensitivity and may point to a leptin-controlled regulation of the ECS in early-lactation.

Maternal exposure to various stimuli can influence pre- and postnatal development of the offspring. This potential has been discussed for glyphosate (GLY), active substance in some non-selective herbicides. Accordingly, present study investigated putative effects of GLY residues in rations on cows and their offspring. Dams received either GLY-contaminated (GLY groups) or control (CON groups) rations combined with low (LC groups) or high (HC groups) concentrate feed proportions (CFP) for 16 weeks during mid- and late lactation and early gestation (59±4 days at beginning of GLY exposure; mean±SE). During this feeding trial, average daily GLY exposures of dams were 1.2 (CONLC), 1.1 (CONHC), 112.5 (GLYLC) and 130.3 (GLYHC) µg/kg body weight/d. After a depletion period (107±4 days; mean±SE) and calving, blood samples of dams and their calves were collected (5-345 min after birth) before calves were fed colostrum and analyzed for hematological and clinical-chemical traits, redox parameters, functional properties of leukocytes and DNA damage in leukocytes. No evidence for malformations of newborn calves could be collected. At parturition, most analyzed blood parameters were not affected by dietary treatment of dams during gestation. Significant GLY effects were observed for some traits, e.g. blood non-esterified fatty acids (NEFA) in calves. These deviations of GLY groups from CON groups likely resulted from strong time-dependent responses of NEFA levels within the first 105 minutes after birth and before colostrum intake (Spearman´s rank correlation R = 0.76, p<0.001). Additionally, significant GLY effects did not result in differences in measures that were beyond normally observed ranges questioning a pathological relevance. In summary, no evidence for teratogenic or other clear effects of GLY or CFP on analyzed parameters of dams and their newborn calves could be collected under applied conditions. However, detailed studies including GLY exposure during late and complete gestation period would be needed to rule out teratogenic effects.

Glyphosate (GLY), the active substance in non-selective herbicides, is often found in ruminant feed. The present feeding study aimed to investigate the effects of GLY-contaminated rations and different concentrate feed proportions (CFP) on the health of fattening German Holstein bulls. Bulls were grouped by low (LC) or high (HC) CFP with (GLYLC, GLYHC) or without GLY-contaminations (CONLC, CONHC) in their rations. Intakes (dry matter, water) and body weight were documented continuously lasting over an average range from 392.2 ± 60.4 kg to 541.2 ± 67.4 kg (mean ± SD). Blood samples collected at the trial's beginning, and after 7 and 15 weeks, were analyzed for hematological and clinical-chemical traits, functional properties of leukocytes, redox parameters and DNA damage. The average GLY exposures of 128.6 (GLYHC), 213.7 (GLYLC), 1.3 (CONHC) and 2.0 µg/kg body weight/d (CONLC) did not lead to GLY effects for most of the assessed parameters relating to animal health and performance. CFP and time displayed marked influences on most of the experimental parameters such as higher dry matter intake and average daily gain in HC compared with the LC groups. GLY effects were rather weak. However, the observed interactive effects between GLY and CFP and/or time occurring in an inconsistent manner are likely not reproducible. Finally, all animals remained clinically inconspicuous, which brings into question the physiological relevance of putative GLY effects.

The use of the herbicide glyphosate and its formulations on protein-rich feedstuff for cattle leads to a considerable intake of glyphosate into the rumen of the animals, where glyphosate may potentially impair the 5-enolpyruvylshikimate-3-phosphate pathway of the commensal microbiota, which could cause dysbiosis or proliferation of pathogenic microorganisms. Here, we evaluated the effects of pure glyphosate and the formulations Durano TF and Roundup® LB plus in different concentrations on the fermentation pattern, community composition and metabolic activity of the rumen microbiota using the Rumen Simulation Technique (RUSITEC). Application of the compounds in three concentrations (0.1 mg/l, 1.0 mg/l or 10 mg/l, n = 4 each) for 9 days did not affect fermentation parameters such as pH, redox potential, NH3-N concentration and production of short-chain fatty acids compared to a control group. Microbial protein synthesis and the degradation of different feed fractions did not vary among the treatments. None of the used compounds or concentrations did affect the microbial diversity or abundance of microbial taxa. Metaproteomics revealed that the present metabolic pathways including the shikimate pathway were not affected by addition of glyphosate, Durano TF or Roundup® LB plus. In conclusion, neither pure glyphosate, nor its formulations Durano TF and Roundup® LB plus did affect the bacterial communities of the rumen.

The objective was to validate the efficacy of Moocall® comparing it to a routine clinical examination. Altogether 38 Holstein cows were enrolled in this study (Moocall® group: 16 heifers and 8 cows; control group: 9 heifers and 5 cows). Clinical examinations were performed every 6 h over the 7 days period before the predicted calving date. The examined traits were changes in pelvic ligament relaxation, edema of the vulva, teat filling, vaginal secretion, tail tip flexibility, tail raising and behavior. There were no significant differences in Moocall® alerts between heifers and cows. The time lag between the first warning of Moocall® and the onset of labor was 21.2 ± 20.2 h (max: 95.4 h; min: 0.1 h; p = 0.87) for heifers and 29.6 ± 29.6 h (max: 177.8 h; min: 0 h; p = 0.97) for cows. Linear models including Moocall® alerts showed a significantly better fit to the time until calving than models without Moocall® information (without variable selection: p = 0.030, with variable selection: p < 0.01). In the best-fitting model, class 2 alerts (enhanced tail activity over 2 h) contributed with a higher significance (p < 0.01). Vice versa, models including additional traits were outperformed the use of Moocall® alerts alone. In the best fitting model, class 2 alerts (enhanced tail activity during 2 h) contributed with a higher significance (p < 0.01) than any of the best clinical predictive parameters, such as pelvic ligament relaxation (p = 0.01), tail tip flexibility (p = 0.01) or behavior (p = 0.01).

Methane (CH4) from ruminal feed degradation is a major pollutant from ruminant livestock, which calls for mitigation strategies. The purpose of the present 4 × 2 factorial arrangement was to investigate the dose-response relationships between four doses of the CH4 inhibitor 3-nitrooxypropanol (3-NOP) and potential synergistic effects with low (LC) or high (HC) concentrate feed proportions (CFP) on CH4 reduction as both mitigation approaches differ in their mode of action (direct 3-NOP vs. indirect CFP effects). Diet substrates and 3-NOP were incubated in a rumen simulation technique to measure the concentration and production of volatile fatty acids (VFA), fermentation gases as well as substrate disappearance. Negative side effects on fermentation regarding total VFA and gas production as well as nutrient degradability were observed for neither CFP nor 3-NOP. CH4 production decreased from 10% up to 97% in a dose-dependent manner with increasing 3-NOP inclusion rate (dose: p < 0.001) but irrespective of CFP (CFP × dose: p = 0.094). Hydrogen gas accumulated correspondingly with increased 3-NOP dose (dose: p < 0.001). In vitro pH (p = 0.019) and redox potential (p = 0.066) varied by CFP, whereas the latter fluctuated with 3-NOP dose (p = 0.01). Acetate and iso-butyrate (mol %) decreased with 3-NOP dose, whereas iso-valerate increased (dose: p < 0.001). Propionate and valerate varied inconsistently due to 3-NOP supplementation. The feed additive 3-NOP was proven to be a dose-dependent yet effective CH4 inhibitor under conditions in vitro. The observed lack of additivity of increased CFP on the CH4 inhibition potential of 3-NOP needs to be verified in future research testing further diet types both in vitro and in vivo.

Glyphosate-based herbicides are among the most used non-selective herbicides worldwide and inhibit synthesis of aromatic amino acids in plants, bacteria, and fungi. Given the broad usage, controversies concerning potential effects of glyphosate on health and especially on gut microbiomes arose. For cattle, it has been proposed based on in vitro data that glyphosate has detrimental effects on the ruminal microbiome, which manifest as a specific inhibition of bacteria involved in fiber degradation and as an enrichment of specific pathogens. In the present study, glyphosate effects on the ruminal microbiome were analyzed in vivo using glyphosate contaminated feedstuffs with strong differences in dietary fiber and dietary energy content in order to reproduce the proposed detrimental glyphosate effects on the rumen microbiome. While significant impact of dietary factors on the ruminal microbiome and its products are pointed out, no adverse glyphosate effects on ruminal microbiome composition, diversity, and microbial metabolites are observed.

The climate-relevant enteric methane (CH4) formation represents a loss of feed energy that is potentially meaningful for energetically undersupplied peripartal dairy cows. Higher concentrate feed proportions (CFP) are known to reduce CH4 emissions in cows. The same applies to the feed additive 3-nitrooxypropanol (3-NOP), albeit through different mechanisms. It was hypothesised that the hydrogen not utilised for CH4 formation through the inhibition by 3-NOP would be sequestered by propionate formation triggered by higher CFP so that it could thereby give rise to a synergistically reduced CH4 emission. In a 2 × 2-factorial design, low (LC) or high (HC) CFP were either tested without supplements (CONLC, CONHC) or combined with 3-NOP (NOPLC, 48.4 mg/kg dry matter (DM); NOPHC, 51.2 mg 3-NOP/kg DM). These four rations were fed to a total of 55 Holstein cows from d 28 ante partum until d 120 post partum. DM intake (DMI) was not affected by 3-NOP but increased with CFP (CFP; p < 0.001). CH4/DMI and CH4/energy-corrected milk (ECM) were mitigated by 3-NOP (23% NOPLC, 33% NOPHC) (p < 0.001) and high CFP (12% CON, 22% 3-NOP groups) (CFP × TIME p < 0.001). Under the conditions of the present experiment, the CH4 emissions of NOPLC increased to the level of the CON groups from week 8 until the end of trial (3-NOP × CFP × TIME; p < 0.01). CO2 yield decreased by 3-NOP and high CFP (3-NOP × CFP; p < 0.001). The reduced body weight loss and feed efficiency in HC groups paralleled a more positive energy balance being most obvious in NOPHC (3-NOP × CFP; p < 0.001). ECM was lower for NOPHC compared to CONHC (3-NOP × CFP; p < 0.05), whereas LC groups did not differ. A decreased fat to protein ratio was observed in HC groups and, until week 6 post partum, in NOPLC. Milk lactose and urea increased by 3-NOP (3-NOP; p < 0.05). 3-NOP and high CFP changed rumen fermentation to a more propionic-metabolic profile (3-NOP; CFP; p < 0.01) but did not affect rumen pH. In conclusion, CH4 emission was synergistically reduced when high CFP was combined with 3-NOP while the CH4 mitigating 3-NOP effect decreased with progressing time when the supplement was added to the high-forage ration. The nature of these interactions needs to be clarified.

Glyphosate (GLY) is worldwide one of the most used active substances in non-selective herbicides. Although livestock might be orally exposed via GLY-contaminated feedstuffs, not much is known about possible hepatotoxic effects of GLY. As hepatic xenobiotic and nutrient metabolism are interlinked, toxic effects of GLY residues might be influenced by hepatic nutrient supply. Therefore, a feeding trial with lactating dairy cows was conducted to investigate effects of GLY-contaminated feedstuffs and different concentrate feed proportions (CFP) in the diets as tool for varying nutrient supply to the liver. For this, 61 German Holstein cows (207 ± 49 days in milk; mean ± standard deviation) were either fed a GLY-contaminated total mixed ration (TMR, GLY groups, mean GLY intake 122.7 µg/kg body weight/day) or control TMR (CON groups, mean GLY intake 1.2 µg/kg body weight/day) for 16 weeks. Additionally, both groups were further split into subgroups fed a lower (LC, 30% on dry matter basis) or higher (HC, 60% on dry matter basis) CFP resulting in groups CONHC (n = 16), CONLC (n = 16), GLYHC (n = 15), GLYLC (n = 14). Blood parameters aspartate aminotransferase, γ-glutamyltransferase, glutamate dehydrogenase, cholesterol, triglyceride, total protein, calcium, phosphorus, acetic acid and urea and histopathological evaluation were not influenced by GLY, whereas all mentioned parameters were at least affected by time, CFP or an interactive manner between time and CFP. Total bilirubin blood concentration was significantly influenced by an interaction between GLY and CFP with temporarily elevated concentrations in GLYHC, whereas the biological relevance remained unclear. Gene expression analysis indicated 167 CFP-responsive genes, while seven genes showed altered expression in GLY groups compared to CON groups. Since expression changes of GLY-responsive genes were low and liver-related blood parameters changed either not at all or only slightly, the tested GLY formulation was considered to have no toxic effects on the liver of dairy cows.

l-carnitine plays an important role in energy metabolism through supporting the transport of activated fatty acids to the subcellular site of ß-oxidation. An acute phase reaction (APR) is known as an energy consuming process. Lipopolysaccharides (LPS) are often used in animal models to study intervention measures during innate immune responses such as APR. Thus, the aim of the study was to investigate the effects of dietary l-carnitine supplementation during an LPS-induced APR in mid-lactating German Holstein cows. Animals were assigned to a control (CON, n = 26) or l-carnitine group (CAR, n = 27, 25 g rumen-protected l-carnitine/cow/d) and received an intravenous injection of LPS (0.5 µg/kg body weight) at day 111 post-partum. Blood samples were collected from day 1 pre-injection until day 14 post-injection (pi). From 0.5 h pi until 72 h pi blood samplings and clinical examinations were performed in short intervals. Clinical signs of the APR were not altered in group CAR except rumen motility which increased at a lower level compared to the CON group after a period of atonia. Group CAR maintained a higher insulin level compared to group CON even up to 72 h pi which might support glucose utilization following an APR.

Seaweeds contain a myriad of nutrients and bioactives including proteins, carbohydrates and to a lesser extent lipids as well as small molecules including peptides, saponins, alkaloids and pigments. The bioactive bromoform found in the red seaweed Asparagopsis taxiformis has been identified as an agent that can reduce enteric CH4 production from livestock significantly. However, sustainable supply of this seaweed is a problem and there are some concerns over its sustainable production and potential negative environmental impacts on the ozone layer and the health impacts of bromoform. This review collates information on seaweeds and seaweed bioactives and the documented impact on CH4 emissions in vitro and in vivo as well as associated environmental, economic and health impacts.

Dairy cows are metabolically challenged during the transition period. Furthermore, the process of parturition represents an energy-consuming process. The degree of negative energy balance and recovery from calving also depends on the efficiency of mitochondrial energy generation. At this point, L-carnitine plays an important role for the transfer of fatty acids to the site of their mitochondrial utilisation. A control (n = 30) and an L-carnitine group (n = 29, 25 g rumen-protected L-carnitine per cow and day) were created and blood samples were taken from day 42 ante partum (ap) until day 110 post-partum (pp) to clarify the impact of L-carnitine supplementation on dairy cows, especially during the transition period and early puerperium. Blood and clinical parameters were recorded in high resolution from 0.5 h to 72 h pp. L-carnitine-supplemented cows had higher amounts of milk fat in early lactation and higher triacylglyceride concentrations in plasma ap, indicating increased efficiency of fat oxidation. However, neither recovery from calving nor energy balance and lipomobilisation were influenced by L-carnitine.

The objective of the present study was to validate the accuracy of algorithms, implemented in the currently available RumiWatch Converter (RWC) version V0.7.4.5 of the RumiWatch System (RWS), for the classification of behavioural characteristics from jaw and head movements which are monitored by a noseband halter comprising a pressure sensor and a triaxial accelerometer. The accurate classification of behavioural characteristics in different time resolutions is critical for the usage of the RWS for scientific and practical purposes as chewing behaviour provides essential indicators for the assessment of diet adequacy in dairy cows. To validate the RWC V0.7.4.5 classification accuracy for behavioural characteristics of rumination, eating, drinking, other activity and ruminating chews per bolus by direct observation as reference method, 14 dairy cows participated in the trial. Concordance between the consolidated 1-min and 1-h classification results was assessed. The RWC V0.7.4.5 classified only rumination and ruminating chews per bolus precisely, whereas an algorithm optimisation for the classification of eating, drinking and other activity is required. Additionally, classification results from the 1-min and 1-h time summaries were not in agreement with each other except for rumination.

The aim of this study was to evaluate energy metabolism and lipid mobilization via ultrasonic measurements (USM), considering inner fat depots, in lactating dairy cows differing in body condition score (BCS) and fed rations with low (35% at dry matter basis; C35) or high (60% at dry matter basis; C60) concentrate feed proportions postpartum. Sixty pluriparous German Holstein cows were arranged in a 2 × 2 factorial design from d 42 antepartum (relative to calculated calving) until d 120 postpartum. Animals were divided into a group with a lower (initial BCS = 3.1 ± 0.38 SD; BCSL) and a group with a higher (initial BCS = 3.83 ± 0.41 SD; BCSH) BCS. Due to higher dry matter intake C60 groups reached the positive energy balance earlier, whereas C35 groups had a more pronounced negative energy balance. Although this would suggest a more pronounced mobilization of C35 groups the USM revealed no differences between feeding groups. Differences in BCS between both BCS groups remained almost the same over the trial. This was not reflected in ultrasonic data, as lipid mobilization was higher in higher conditioned cows. These findings demonstrate the extended possibilities of USM to depict metabolic processes.

In spring, transition from a total mixed ration (TMR) to pasture requires rumen adaptions for the cow. It had been shown that transition period does not necessarily mean an increased risk for subacute ruminal acidosis (SARA). After adaption to pasture, however, supplying low amounts of concentrate did indicate increased risk, but caused no adverse effects on rumen morphology and absorption capacity. The present study aimed to investigate the effect of transition, and how a supply of 4.5 kg dry matter concentrate·cow-1 · day-1 during fulltime grazing influenced different rumen parameters. During a 12-week trial eleven rumen-cannulated dairy cows were observed during transition from confinement to pasture (PG; n = 6) and compared to cows fed TMR indoors (CG; n = 5). The CG stayed on a TMR based ration (35% corn silage, 35% grass silage, 30% concentrate; dry matter basis), whereas the PG slowly switched to a pasture-based ration (week 0 and 1 = TMR, week 2 = TMR and 3 h pasture·day-1, week 3 and 4 = TMR and 12 h pasture·day-1, and week 5 to 11 = pasture combined with 4.5 kg DM concentrate · cow-1·day-1). Papillae surface area decreased during transition and increased again during fulltime grazing, while the fractional absorption rate of volatile fatty acids (VFA) was not influenced. This suggests only a limited effect of papillae surface area on VFA absorption rate. Feeding changes resulted in different fermentation profiles of VFA. Changing ratio of starch to sugar during transition to fulltime grazing plus concentrate supply did not lead to lower rumen pH. In conclusion, the concentrate supply combined with high fermentable grass during fulltime grazing increased papillae surface area but did not affect absorption rate or rumen pH, so that risk for SARA was not increased.

It had been shown that supply of low amounts of concentrate after transition to full-time grazing caused energy deficits, resulting in a lower milking performance and changes in a variety of variables indicative for energy metabolism. The present study aimed to investigate how a moderate concentrate supply (4.5 kg dry matter cow/day) after transition to pasture influences health and production indicators. Over a 12-week trial period dairy cows were observed during transition from confinement to pasture (pasture group: PG) and compared to cows fed TMR indoors (confinement group: CG). On average, the PG consumed less feed and energy than the CG and mobilized body reserves, which is mirrored in a decrease of body condition and various fat depots. These effects were paralleled by elevated serum concentrations of non-esterified fatty acids and ketone bodies as well as an increase in liver fat content. The physical activity (elevated walking, eating, decreasing rumination time) of the PG was significantly higher than that of the CG, which intensified the energy deficiency and resulted in a lower milk yield. In conclusion, the moderate concentrate supply was insufficient to counterbalance the lower energy intake from pasture during transition.

The aim of this study was to assess differences of ruminal pH and rumination activity between the prepartal and postpartal period of cows and to investigate changes of feeding behaviour and ruminal fermentation depending on postpartal rations with slight differences in roughage energy and fibre concentration. Therefore, 11 rumen-cannulated German Holstein cows were used from 3 weeks antepartum until 16 weeks postpartum. During the dry period, all cows received the same diet. After calving cows were assigned to two groups receiving partial mixed rations with different energy concentrations (6.1 MJ NEL/kg DM [moderate], with 18% straw on dry matter [DM] basis, group MR) or 6.5 MJ NEL/kg DM [high], with 6% straw on DM basis, group HR). In both groups, concentrates were supplied on average at 45% of total DM intake. Circadian measurements of ruminal pH, ruminal temperature and rumination activity were conducted weekly during the transition period and bi-weekly after week 4 postpartum. Throughout the postpartal period, feed intake behaviour was measured, and twice during the experiment, rumen fluid composition and net-acid-base excretion in urine were determined. Mean ruminal pH, feeding behaviour, rumination activity and rumen fluid composition did not differ between the groups, whereas group HR exhibited higher diurnal variations of pH and had a lower rumen temperature than group MR postpartum. However, these differences also occurred during the prepartal period when groups received equal rations. It was observed that ruminal pH values and the risk for subacute ruminal acidosis (SARA) were increased during postpartal period, whereby the circadian pH values showed high cow-individual variations. Overall, differences of energy and fibre concentrations in roughage did not affect the observed variables, whereby it should be noted that the number of cows was quite low in this study. Our results suggest that the ruminal pH characteristics are rather cow individual, indicating that cows presumably differ in their susceptibility to develop SARA.

Vitamin E (Vit. E) is discussed to influence ruminal biohydrogenation. The objective of this study was to investigate the influence of a Vit. E supplementation on rumen fermentation characteristics, ruminal microbial protein synthesis as well as ruminal organic matter fermentation. Furthermore, we aimed to investigate the influence of Vit. E supplementation on short-chain fatty acids (SCFA) and protozoa concentrations in the rumen and, in addition, on transfer rates of middle-chain and long-chain fatty acids into the duodenum in lactating dairy cows. Eight rumen and duodenum fistulated German Holstein cows were assigned to either a group receiving 2,327 IU/d Vit. E (138.6 IU/kg DM DL-α-tocopherylacetate; n = 4) or a control group (23.1 IU/kg DM; n = 4). Neither ruminal protein synthesis nor organic matter fermentation was influenced by treatment. Vit. E did not act on the concentrations of short-chain fatty acids and protozoa in rumen fluid. Duodenal flow of C13:0 (1.3 versus 0.2 g/d, p = 0.014) and iso-C14:0 (1.0 versus 0.5 g/d, p = 0.050) was higher in the Vit. E group. We observed a trend for higher duodenal flows for C12:0 (1.6 versus 0.9 g/d, p = 0.095) and anteiso-C15:0 (12.2 versus 8.9 g/d, p = 0.084). Transfer rate of C12:0 tended to be higher in the Vit. E group (125.61 versus 73.96, p = 0.082). No other transfer rates were affected by treatment. Further studies are necessary to investigate the influence of Vit. E on rumen microbiota and their fatty acid production as well as on the impact of different doses of Vit. E supplementation on variables of protein synthesis efficiency.

The aim of this study was to investigate the effects of different energy supplies from roughage and concentrates on performance, health and energy efficiency during early lactation. For this purpose an experiment was conducted containing 64 pluriparous German Holstein cows from 3 weeks prepartum until 16 weeks postpartum. During dry period all cows received an equal dry cow ration. After calving, cows were assigned in a 2 × 2 factorial arrangement to one of four groups, receiving either a moderate (MR, 6.0 MJ NEL) or a high (HR, 6.4 MJ NEL) energy concentration in roughage and furthermore moderate (MC, 150 g/kg energy-corrected milk (ECM)) or high amounts of concentrates (HC, 250 g/kg ECM) on dry matter (DM) basis, which were allocated from an automatic feeding system. Higher allocation of concentrates resulted in an increase of DM intake at expense of roughage intake. HC cows had a higher milk yield than MC cows, whereas ECM was higher in HR cows due to a decrease of milk fat yield in MR groups. Energy balance and body condition score were elevated in HC cows, but no differences occurred in development of subclinical ketosis. Furthermore, energy efficiency variables were lower in HC groups because the greater energy intake was not associated with a considerable elevation of milk yield. Consistency of faeces did not indicate digestive disorders in any of the treatment groups although the faecal manure score was significantly lower in HR groups. Our results underline the importance of a high energy uptake from roughage, which can contribute to an adequate performance and beneficial efficiency, especially at lower amounts of concentrates in ration. Feeding concentrates on an average amount of 9.4 kg/d compared to 6.4 kg/d on DM basis improved the energy balance in our trial, but without consequences for metabolic blood variables and general health of the cows.
