RESUMO

Responsible use of natural resources and waste reduction are key concepts in bioeconomy. This study demonstrates that agro-food derived-biomasses from the Italian food industry, such as crude glycerol and cheese whey permeate (CWP), can be combined in a high-density fed-batch culture to produce a recombinant ß-galactosidase from Marinomonas sp. ef1 (M-ßGal). In a small-scale process (1.5 L) using 250 mL of crude glycerol and 300 mL of lactose-rich CWP, approximately 2000 kU of recombinant M-ßGal were successfully produced along with 30 g of galactose accumulated in the culture medium. The purified M-ßGal exhibited high hydrolysis efficiency in lactose-rich matrices, with hydrolysis yields of 82 % in skimmed milk at 4 °C and 94 % in CWP at 50 °C, highlighting its biotechnological potential. This approach demonstrates the effective use of crude glycerol and CWP in sustainable and cost-effective high-density Escherichia coli cultures, potentially applicable to recombinant production of various proteins.

Recognized for its bioactive compounds, açaí has become a functional food, but it has a low pulp yield, and the seeds are the main waste. This study investigates the potential of açaí seeds (Euterpe oleracea Mart.) to produce mannooligosaccharides (MOS) through enzymatic hydrolysis. Using response surface methodology (RSM), the research optimizes MOS extraction while minimizing mannose production and reducing processing time, achieving MOS production of about 10 g/L, a value within the range of similar investigations. The RSM quadratic models establish correlations between MOS production (M2-M5) and enzymatic hydrolysis conditions, with R2 values ranging from 0.6136 to 0.9031. These models are used to emphasize MOS performance (M2-M5) while reducing mannose production, which also promotes profitability by reducing time. Experimental validation agrees with model predictions, highlighting optimal conditions near 40 °C, intermediate enzyme loading, and basic pH that effectively promotes MOS generation on mannose within an accelerated processing time frame. With predictions of experimental results within a margin of error of < 9%, the validity of the models was acceptable. This research contributes to the advancement of the understanding of the enzymatic hydrolysis of açaí seeds, which is a step toward the sustainable use of resources with a focus on process engineering aspects.

The European Union (EU) is committed to transitioning toward a circular economy model, with food waste being one of the areas to be targeted. To close the loop of food waste generated during food processing and discarded at the retail or consumption phases, research and innovation parties proposed to valorize agro-food by-products to produce novel foods and food improvement agents (food additives, food enzymes, and food flavorings). In the EU, the authorization of such novel foods and food improvement agents is governed by different regulatory frameworks. A centralized safety assessment by the European Food Safety Authority (EFSA) is the prerequisite for their authorization through the so-called Union Lists. Up to December 2023, EFSA published 45 scientific opinions on the safety of novel foods, food enzymes, and food additives derived from by-products of plant and animal origin. The current study illustrates examples of these by-products for the production of novel foods or food improvement agents and the data requirements behind their respective safety assessments conducted by EFSA. In this review, applications on novel foods, food enzymes, and food additives received by EFSA were screened and analyzed to find the common scientific requirements and differences in terms of the safety evaluation of such products. Various by-products (i.e., corncobs, coffee husks, spent grains of barley and rice, grape pomace, pumpkin peels, bovine whey, eggshells, shrimp heads, and animal organs or tissues) were described in the applications as being processed (extraction, physical treatments, and chemical and enzymatic reactions) to obtain novel foods and food improvement agents. The heterogeneity and complexity of these products emphasize the challenge of their safety assessment, depending on the characteristics of each product. However, as this study shows, the scientific requirements underpinning their safety do not differ substantially in the different regulated product areas considered, with similar information needed to assess their safety in terms of identity, production process, compositional characterization, proposed/intended uses and exposure assessment, toxicological information, and allergenicity data. Additional nutritional information and data on the history of use are required in the case of novel foods.

INTRODUCTION: Commercial plastics are potentially hazardous and can be carcinogenic due to the incorporation of chemical additives along with other additional components utilized as brominated flame retardants and phthalate plasticizers during production that excessively produce large numbers of gases, litter, and toxic components resulting in environmental pollution. METHOD: Biodegradable plastic derived from natural renewable resources is the novel, alternative, and innovative approach considered to be potentially safe as a substitute for traditional synthetic plastic as they decompose easily without causing any harm to the ecosystem and natural habitat. The utilization of undervalued compounds, such as by-products of fruits and vegetables in the production of biodegradable packaging films, is currently a matter of interest because of their accessibility, affordability, ample supply, nontoxicity, physiochemical and nutritional properties. Industrial food waste was processed under controlled conditions with appropriate plasticizers to extract polymeric materials. Biodegradability, solubility, and air test analysis were performed to examine the physical properties of polymers prior to the characterization of the biofilm by Fourier-transformed infrared spectroscopy (FTIR) for the determination of polymeric characteristics. RESULT: The loss of mass examined in each bioplastic film was in the range of 0.01g to 0.20g. The dimension of each bioplastic was recorded in the range of 4.6 mm to 28.7 mm. The existence of -OH, C=C, C=O stretching, and other crucial functional groups that aid in the creation of a solid polymeric material are confirmed by FTIR analysis. This study provides an alternative approach for sustainable and commercially value-added production of polymeric-based biomaterials from agro-industrial waste as they are rich in starch, cellulose, and pectin for the development of bio-plastics. CONCLUSION: The rationale of this project is to achieve a straightforward, economical, and durable method for the production of bio-plastics through effective utilization of industrial and commercial fruit waste, ultimately aiding in revenue generation.

Natural toxins, such as mycotoxins and cyanotoxins, can contaminate food and feed, leading to toxicity in humans and animals. This study focused on using nine magnetic nanostructured agents to remove the main types of toxins. Initially, the efficacy of these materials was evaluated in water solutions, revealing that composites with sizes below 3 mm, containing magnetite, activated carbon, esterified pectin, and sodium alginate, removed up to 90% of mycotoxins and cyanotoxins with an adsorption of 873 ng/g. The application of the nanostructures was then assessed in beer, milk, Distillers Dried Grains with Solubles and water contaminated with cyanobacteria. The presence of matrix slightly decreases the adsorption capacity for some toxins. The maximum toxin removal capacity was calculated with cyanotoxins, composites achieved a removal of up to 0.12 mg/g, while nanocomposites (15 µm) reached 36.6 mg/g. Therefore, these findings point out the potential for using nanotechnology in addressing natural toxins contamination.

To align with the SDG 12.3 target and ensure global food security and sustainability, it is crucial to prioritize the reduction of food loss and waste. This paper aims to synthesize previous research on waste reduction tools like lean manufacturing in the agro-food processing industry and identify areas that require further investigation to assurance worldwide food security and promote sustainability. The study uses a systematic literature review that was proposed by Denyer and Tranfield. This research work provides a descriptive analysis of the evolution of lean manufacturing in agro-food processing and identifies research gaps. The review highlights the importance of demand forecasting, managing variable raw materials and products, increasing management involvement, promoting partnership among supply chain members, and addressing supply and demand seasonality and uncertainty to apply the approach to food waste reduction. Based on the findings, the paper suggests further research areas for future investigation that will help create a more sustainable and equitable food system. Reducing food loss and waste can ensure that everyone has access to safe, nutritious, and affordable food while protecting the planet's resources and reducing greenhouse gas emissions. This study may contribute to the theory of waste minimization, specifically in post-harvest food loss and waste minimization. The findings will help researchers conduct research work interested in minimizing food loss and waste to ensure global food security.

Salmonella is as an intracellular bacterium, causing many human fatalities when the host-specific serotypes reach the host gastrointestinal tract. Nontyphoidal Salmonella are responsible for numerous foodborne outbreaks and product recalls worldwide whereas typhoidal Salmonella are responsible for Typhoid fever cases in developing countries. Yet, Salmonella-related foodborne disease outbreaks through its food and water contaminations have urged the advancement of rapid and sensitive Salmonella-detecting methods for public health protection. While conventional detection methods are time-consuming and ineffective for monitoring foodstuffs with short shelf lives, advances in microbiology, molecular biology and biosensor methods have hastened the detection. Here, the review discusses Salmonella pathogenic mechanisms and its detection technology advancements (fundamental concepts, features, implementations, efficiency, benefits, limitations and prospects). The time-efficiency of each rapid test method is discussed in relation to their limit of detections (LODs) and time required from sample enrichment to final data analysis. Importantly, the matrix effects (LODs and sample enrichments) were compared within the methods to potentially speculate Salmonella detection from environmental, clinical or food matrices using certain techniques. Although biotechnological advancements have led to various time-efficient Salmonella-detecting techniques, one should consider the usage of sophisticated equipment to run the analysis by moderately to highly trained personnel. Ultimately, a fast, accurate Salmonella screening that is readily executed by untrained personnels from various matrices, is desired for public health procurement.

Agro-food waste is a rich source of biopolymers such as cellulose, chitin, and starch, which have been shown to possess excellent biocompatibility, biodegradability, and low toxicity. These properties make biopolymers from agro-food waste for its application in tissue engineering and regenerative medicine. Thus, this review highlighted the properties, processing methods, and applications of biopolymers derived from various agro-food waste sources. We also highlight recent advances in the development of biopolymers from agro-food waste and their potential for future tissue engineering and regenerative medicine applications, including drug delivery, wound healing, tissue engineering, biodegradable packaging, excipients, dental applications, diagnostic tools, and medical implants. Additionally, it explores the challenges, prospects, and future directions in this rapidly evolving field. The review showed the evolution of production techniques for transforming agro-food waste into valuable biopolymers. However, these biopolymers serving as the cornerstone in scaffold development and drug delivery systems. With their role in wound dressings, cell encapsulation, and regenerative therapies, biopolymers promote efficient wound healing, cell transplantation, and diverse regenerative treatments. Biopolymers support various regenerative treatments, including cartilage and bone regeneration, nerve repair, and organ transplantation. Overall, this review concluded the potential of biopolymers from agro-food waste as a sustainable and cost-effective solution in tissue engineering and regenerative medicine, offering innovative solutions for medical treatments and promoting the advancement of these fields.

The effect of operational conditions on the stability of acidogenic fermentation (AF) devoted to volatile fatty acids (VFAs) production still presents numerous gaps to achieve high yields and fully understand the responses of open microbiomes associated to this technology. To cope with that, this investigation was designed to assess the stability of VFAs production via AF of agro-food wastes at high hydraulic retention times (HRTs) (20 and 30 d) and pH oscillations (5.8-6.2). Similar bioconversion efficiencies (∼50 %) were reached regardless of the HRT, revealing that HRT of 20 d can be considered as a threshold from which, no further improvement was achieved. The combination of long HRTs, 25 °C and acid pHs promoted a robust microbiome that resulted in a stable outcome against pH variations, being Clostridiales order identified as key player of AF stability. These conditions mediated a high selectivity in the VFAs production profile, with acetic and butyric acids, prevailing in the VFAs pool (∼80 % of total VFAs) at HRT 20 d. The selection of appropriated conditions was shown to be critical to maximize the hydrolysis and acidogenesis of the substrate and attain a stable effluent against pH oscillations.

The present study investigated the combined production of reclaimed water for reuse purposes and polyhydroxyalkanoates (PHA) from an agro-food industrial wastewater. A pilot plant implementing a two-stage process for PHA production was studied. It consisted of a mainstream sequencing batch membrane bioreactor (SBMBR) in which selection of PHA-accumulating organisms and wastewater treatment were carried out in, and a side-stream fed-batch reactor (FBR) where the excess sludge from the SBMBR was used for PHA accumulation. The performance of the SBMBR was compared with that of a conventional sequencing batch reactor (SBR) treating the same wastewater under different food to microorganisms' ratios (F/M) ranging between 0.125 and 0.650 kgCOD kgTSS-3 d-1. The SBMBR enabled to obtain very high-quality effluent in compliance with the relevant national (Italy) and European regulations (Italian DM 185/03 and EU, 2020/741) in the field of wastewater reclamation, whereas the performances in the SBR collapsed at F/M higher than 0.50 kgCOD kgTSS-1d-1. A maximum intracellular storage of 45% (w/w) and a production yield of 0.63 gPHA L-1h-1 were achieved when the SBMBR system was operated with a F/M ratio close to 0.50 kgCOD kgTSS-1d-1. This resulted approximately 35% higher than those observed in the SBR, since the ultrafiltration membrane avoided the washout of dispersed and filamentous bacteria capable of storing PHA. Furthermore, while maximizing PHA productivity in conventional SBR systems led to process dysfunctions, in the SBMBR system it helped mitigate these issues by reducing membrane fouling behaviour. The results of this study supported the possibility to achieve combined recovery of reclaimed water and high-value added bioproducts using membrane technology, leading the way for agro-food industrial wastewater valorization in the frame of a circular economy model.

The agro-food surplus is considered the most abundant renewable resource, interesting for the economic and environmental impact, with a possible employment in different productive fields such as supplement in animal feed. For a safe and effective enhancement of agro-food wastes it is needed a toxicological analysis before of their employment, to avoid potential risks to animals, humans and environment. The purpose of this study was to evaluate the metals content (Cu, Fe, Zn, Mg, Na, K, Ca, Mn, Se, Zn, Co, Ni, V, Sb, Mo, Cr and Pb, Cd, As, Ba), in agro-food wastes through ICP-MS analysis. The results obtained showed the presence of metals analysed with a significant content of essential micro and macro-elements; with regard to toxic metals, only strawberries and fennel samples showed Pb levels above the MRL. These data confirm, from the toxicological point of view, a possible and safe reuse of the most of all agro-food wastes, as raw material at low cost and rich of essential nutrients and particularly as supplement in animal feed.

The purpose of this study was to scrutinize the interdependence among training quality, graduate demographic characteristics, and graduate competence and employability in the agro-food processing sub-sector of the TVET system in Ethiopia. To that effect, the commitment and competence of graduates and trainers, the quantitative and qualitative aspects of resource supplies, training and assessment practices including industry involvement or cooperative training, and the relationships with and influence of the human and material resources on graduate competence were examined. The descriptive survey design of the quantitative approach that involved 637 participants was employed. Data analyses through descriptive and inferential statistics revealed that colleges were challenged with trainer skill gaps, poor material resource supply, and poor cooperative training. It was also found that the human inputs (trainer competence, trainer commitment, and trainee characteristics) significantly predicted the core competence of graduates whereas material resources had a very weak predictive power. In addition, an inverse relationship between employment status and all forms of graduates' competence was found. Accordingly, it is plausible to conclude that poor institutional capacity that denied to pay necessary attention for the input and process of the training delivery has obstructed the competence of graduates.

Carotenoids are a large group of health-promoting compounds used in many industrial sectors, such as foods, feeds, pharmaceuticals, cosmetics, nutraceuticals, and colorants. Considering the global population growth and environmental challenges, it is essential to find new sustainable sources of carotenoids beyond those obtained from agriculture. This review focuses on the potential use of marine archaea, bacteria, algae, and yeast as biological factories of carotenoids. A wide variety of carotenoids, including novel ones, were identified in these organisms. The role of carotenoids in marine organisms and their potential health-promoting actions have also been discussed. Marine organisms have a great capacity to synthesize a wide variety of carotenoids, which can be obtained in a renewable manner without depleting natural resources. Thus, it is concluded that they represent a key sustainable source of carotenoids that could help Europe achieve its Green Deal and Recovery Plan. Additionally, the lack of standards, clinical studies, and toxicity analysis reduces the use of marine organisms as sources of traditional and novel carotenoids. Therefore, further research on the processing of marine organisms, the biosynthetic pathways, extraction procedures, and examination of their content is needed to increase carotenoid productivity, document their safety, and decrease costs for their industrial implementation.

Waste generated from the agro-food industry represents a concerning environmental, social and economic issue. The Food and Agriculture Organization of the United Nations defines food waste as all food that decreases in quantity or quality to the extent that it is thrown out by food service providers and consumers. The FAO reports that 17% of worldwide food production may be wasted. Food waste may include fresh products, food close to the expiration date discarded by retailers and food products from household kitchens and eating establishments. However, food waste offers different possibilities to extract functional ingredients from different sources, such as dairy, cereals, fruits, vegetables, fibers, oils, dye and bioactive compounds. The optimization of agro-food waste as an ingredient will help in the development and innovation of food products to generate functional food and beverages to prevent and treat several diseases in consumers.

Bioactive ingredients are part of the food chain and are responsible for numerous health benefits. Subcritical low temperature extraction has been employed to acquire bioactive ingredients because of its excellent properties, such as energy conservation, low temperature, elimination of residual solvent, and high extraction yield and quality. This review aims to provide a clear picture of the basics of subcritical-temperature extraction, its bioactive ingredient extraction efficiency, and possible applications in the agro-food industry. This review suggested that the extraction temperature, time, co-solvents, solid-fluid ratio, and pressure impacted the extraction efficiency of bioactive ingredients from foods and food by-products. Subcritical solvents are appropriate for extracting low polar ingredients, while the inclusion of co-solvents could extract medium and high polar substances. Bioactive ingredients from foods and food by-products can be used as antioxidants, colorants, and nutritional supplements. Additionally, this technology could remove pesticide residues in tea, concentrate edible proteins, and reduce cigarette tar. A new trend toward using subcritical low temperature extraction in extracting bioactive ingredients will acquire momentum.

Producing food by farming and subsequent food manufacturing are central to the world's food supply, accounting for more than half of all production. Production is, however, closely related to the creation of large amounts of organic wastes or byproducts (agro-food waste or wastewater) that negatively impact the environment and the climate. Global climate change mitigation is an urgent need that necessitates sustainable development. For that purpose, proper agro-food waste and wastewater management are essential, not only for waste reduction but also for resource optimization. To achieve sustainability in food production, biotechnology is considered as key factor since its continuous development and broad implementation will potentially benefit ecosystems by turning polluting waste into biodegradable materials; this will become more feasible and common as environmentally friendly industrial processes improve. Bioelectrochemical systems are a revitalized, promising biotechnology integrating microorganisms (or enzymes) with multifaceted applications. The technology can efficiently reduce waste and wastewater while recovering energy and chemicals, taking advantage of their biological elements' specific redox processes. In this review, a consolidated description of agro-food waste and wastewater and its remediation possibilities, using different bioelectrochemical-based systems is presented and discussed together with a critical view of the current and future potential applications.

Ammonia (NH3) volatilization, nitrous oxide (N2O) emissions, and nitrate (NO3-) leaching from agriculture cause severe environmental hazards. Research studies and mitigation strategies have mostly focused on one of these nitrogen (N) losses at a time, often without an integrated view of the agro-food system. Yet, at the regional scale, N2O, NH3, and NO3- loss patterns reflect the structure of the whole agro-food system. Here, we analyzed at the resolution of NUTS2 administrative European Union (EU) regions, N fluxes through the agro-food systems of a Temperate-Mediterranean gradient (France, Spain, and Portugal) experiencing contrasting climate and soil conditions. We assessed the atmospheric and hydrological N emissions from soils and livestock systems. Expressed per ha agricultural land, NH3 volatilization varied in the range 6.2-44.4 kg N ha-1 yr-1, N2O emission and NO3 leaching 0.3-4.9 kg N ha-1 yr-1 and 5.4-154 kg N ha-1 yr-1 respectively. Overall, lowest N2O emission was found in the Mediterranean regions, where NO3- leaching was greater. NH3 volatilization in both temperate and Mediterranean regions roughly follows the distribution of livestock density. We showed that these losses are also closely correlated with the level of fertilization intensity and agriculture system specialization into either stockless crop farming or intensive livestock farming in each region. Moreover, we explored two possible future scenarios at the 2050 horizon: (1) a scenario based on the prescriptions of the EU-Farm-to-Fork (F2F) strategy, with 25% of organic farming, 10% of land set aside for biodiversity, 20% reduction in N fertilizers, and no diet change; and (2) a hypothetical agro-ecological (AE) scenario with generalized organic farming, reconnection of crop and livestock farming, and a healthier human diet with an increase in the share of vegetal protein to 65% (i.e., the Mediterranean diet). Results showed that the AE scenario, owing to its profound reconfiguration of the entire agro-food system would have the potential for much greater reductions in NH3, N2O, and NO3- emissions, namely, 60-81% reduction, while the F2F scenario would only reach 24-35% reduction of N losses.

Patients with galactosemia accumulate galactose in their bodies, requiring a lifelong galactose-restricted diet. Therefore, accurate information on the galactose content in commercial agro-food resources is essential. The HPLC method generally used for sugar analysis has low separation and detection sensitivity. Here, we sought to establish an accurate analytical method for determining the galactose content in commercial agro-food resources. To that aim, we employed gas chromatography with flame-ionization detection to detect trimethylsilyl-oxime (TMSO) sugar derivatives (concentration: ≤0.1 mg/100 g). The galactose content in 107 Korean agro-food resources reflecting intake patterns was then analyzed. The galactose content in steamed barley rice was 5.6 mg/100 g, which was higher than that in steamed non-glutinous and glutinous rice. Moist-type and dry-type sweet potatoes, blanched zucchini, and steamed Kabocha squash had high galactose content (36.0, 12.8, 23.1, and 61.6 mg/100 g, respectively). Therefore, these foods are detrimental to patients with galactosemia. Among fruits, avocado, blueberry, kiwi, golden kiwifruit, and sweet persimmon had galactose contents of ≥10 mg/100 g. Dried persimmon had 132.1 mg/100 g and should therefore be avoided. Mushrooms, meat, and aquatic products showed low galactose content (≤10 mg/100 g), making them safe. These findings will help patients to manage their dietary galactose intake.

Increasing the overall use efficiency of nitrogen (N) and phosphorus (P) resources in food production while minimizing losses to the environment are required to meet the dual challenge of food security and sustainability. Yet studies quantifying the overall performance of different agro-system types and how these have changed over time remain rare, although they are essential to propose solution pathways. Here, we reconstructed fluxes of N and P within 78 watersheds of the St. Lawrence Basin (SLB) of eastern Canada between 1901 and 2011, using the Generalized Representation of Agro-Food System model (GRAFS). This analysis allowed us to classify different agro-food system types and to evaluate how agricultural specialization influenced nutrient efficiencies and potential losses to the environment over time. Using a cluster analysis, we identified four agro-food system types with different overall outcomes in efficiencies and losses. We show that agricultural practices in the SLB were similar until the 1950's and deemed unsustainable in several watersheds by depleting agricultural soils of their nutrients (particularly N). With the advent of manufactured fertilizers and the intensification of livestock farming, the SLB then rapidly shifted through the 1970s and 1980s to more intensified and highly unsustainable agro-food system types, where, in 2011, ~77 % of N and ~ 94 % of P inputs were lost to the environment. We also show that nutrient pollution continued to increase despite gains in the nutrient use efficiency of animal farming due to higher nutrient throughput from intensive production. The increased proportion of confined animals, disconnected from croplands, indeed resulted in inefficient nutrient recycling. While nutrient use efficiency may mitigate nutrient pollution, reducing the absolute nutrient flux through agro-food systems should be a priority, likely through a reconnection of crop and animal farming and an overall reduction of meat production, specifically from concentrated, intensive livestock systems.

Pancreatic ß-cells play a crucial role in maintaining glucose homeostasis, although they are susceptible to oxidative damage, which can ultimately impair their functionality. Thinned nectarines (TNs) have gained increasing interest due to their high polyphenol and abscisic acid (ABA) content, both of which possess antidiabetic properties. Nevertheless, the efficacy of these bioactive compounds may be compromised by limited stability and bioavailability in vivo. This study aimed to develop nanoformulations (NFs) containing pure ABA or a TN extract (TNE) at an equivalent ABA concentration. Subsequently, the insulinotropic and antioxidant potential of the NFs and their unformulated (free) forms were compared in MIN-6 pancreatic cells exposed to varying glucose (5.5 mM and 20 mM) and iron (100 µM) concentrations. NF-TNE treatment exhibited enhanced antioxidant activity compared to free TNE, while ABA-based groups showed no significant antioxidant activity. Moreover, MIN6 cells incubated with both high glucose and iron levels demonstrated significantly higher insulin AUC levels after treatment with all samples, with NF-TNE displaying the most pronounced effect. In conclusion, these results highlight the additional beneficial potential of TNE due to the synergistic combination of bioactive compounds and demonstrate the significant advantage of using a nanoformulation approach to further increase the benefits of this and similar phytobioactive molecules.
