The increased production and expanding demand for cellulosic products have required companies to provide solutions for the proper disposal of generated residues without compromising soil quality. In this context, the current research aimed at evaluating the ecotoxicological impacts of dregs application in subtropical soils. The experiments were carried out with the earthworm Eisenia andrei, the springtail Folsomia candida, and the potworm Enchytraeus crypticus in two subtropical soils (Neosol and Cambisol) collected in the southern region of Brazil. The considerable differences in texture and organic matter content motivated the choosing of these soils. The reproductive rate of all organisms was influenced, with varying EC50 values depending on the soil type. However, more pronounced effects were observed for Neosol. The springtail F. candida proved to be the most sensitive to contamination (EC50 = 5.8 g kg-1 for Neosol and 48.5 g kg-1 for Cambisol), followed by the earthworm E. andrei (EC50 = 62.4 g kg-1 for Neosol and 129.5 g kg-1 for Cambisol) and the potworm E. crypticus (EC50 = 67.2 g kg-1 for Neosol and 230.4 g kg-1 for Cambisol). As a result, while dregs can be used to correct soil acidity, they have been shown to have negative effects on important organisms responsible for functions critical to maintaining ecosystem quality.

The massive influx of Sargassum natans and S. fluitans to the shores of the Mexican Caribbean has raised concerns regarding their potential impact on soil quality and health in coastal and agroecosystems. The effects of Sargassum accumulation remain largely unexplored. This study aimed to assess the impact of Sargassum on soil ecosystems by examining the behavior and survival of the epigean earthworm Eisenia fetida. The earthworm was exposed to varying concentrations of Sargassum (0, 25, 50, 75, and 100%) in two toxicological tests. Results from the avoidance test demonstrated that E. fetida exhibited strong aversion (> 80%) to a diet containing 100% Sargassum. Conversely, the acute test revealed minimal mortality, but growth decreased with increasing Sargassum concentrations. These findings can serve as early warning bioindicators for assessing the environmental risk posed by Sargassum in soil ecosystems.

Soil contamination by antibiotics is a global issue of great concern that contributes to the rise of bacterial antibiotic resistance and can have toxic effects on non-target organisms. This study evaluated the variations of molecular, cellular, and histological parameters in Eisenia fetida earthworms exposed to sulfamethazine (SMZ) and tetracycline (TC), two antibiotics commonly found in agricultural soils. The earthworms were exposed for 14 days to a series of concentrations (0, 10, 100, and 1000 mg/kg) of both antibiotics. SMZ and TC did not affect the survival of E. fetida, however, other effects at different levels of biological complexity were detected. The two highest concentrations of SMZ reduced the viability of coelomocytes. At the highest TC concentration, there was a noticeable decline in cell viability, acetylcholinesterase activity (neurotoxicity), and the relative presence of mucopolysaccharides in the epidermis (mucous production). Glutathione S-transferase activity decreased in all TC treatments and at the highest SMZ concentration. However, levels of malondialdehyde and protein carbonyls did not change, suggesting an absence of oxidative stress. Tetracycline was neurotoxic to E. fetida and changed the integrity of the epidermis. Both antibiotics altered the intestinal microbiota of E. fetida, leading to a reduction in the relative abundance of bacteria from the phyla Proteobacteria and Bacteroidetes, while causing an increase in the phylum Actinobacteroidota. All observed changes indicate that both SMZ and TC can disrupt the earthworms' immune system and gut microbiome, while fostering the growth of bacteria that harbour antibiotic resistance genes. Finally, both antibiotics exerted additional metabolic and physiological effects that increased the vulnerability of E. fetida to pathogens.

Biobeds are presented as an alternative for good pesticide wastewater management on farms. This work proposes a new test for in-situ biomonitoring of pesticide detoxification in biobeds. It is based on the assessment of visually appreciable injuries to Eisenia fetida. The severity of the injury to each exposed individual is assessed from the morphological changes observed in comparison with the patterns established in seven categories and, an injury index is calculated. A linear relationship between the proposed injury index and the pesticide concentration was determined for each pesticide sprayed individually in the biomixture. The five pesticides used were atrazine, prometryn, clethodim, haloxyfop-P-methyl and dicamba. In addition, a multiple linear regression model (i.e., a multivariate response surface) was fitted, which showed a good generalization capacity. The sensitivity range of the injury test was tested from 0.01 to 630 mg kg-1 as the total pesticide concentration. This index is then used to monitor the detoxification of these pesticides in a biomixture (composed of wheat stubble, river waste, and soil, 50:25:25% by volume) over 210 days. The results are compared with standardized tests (Eisenia fetida avoidance test and Lactuca sativa seed germination test) carried out on the same biomixture. The results are also compared with data on the removal of pesticides. The injury test showed a better correlation with the removal of pesticides than the avoidance test and seed germination test. This simple and inexpensive test has proved to be useful for decontamination in-situ monitoring in biobeds.

Problem: Earthworm is a valuable source of biologically and pharmacologically active compounds, with applications in the treatment of various types of diseases; however, the main application they have been given is in the production of organic fertilizer. One of the alternatives for obtaining bioactive compounds is by means of enzymatic hydrolysis. Aim: This study proposes the optimization of the fractionation of the antioxidant enzymatic hydrolysate from Californian red worm (Eisenia fetida) protein. Methodology: For this purpose, the worms were separated and hydrolyzed using the enzyme Alcalase 2.4L for 4000s. The obtained hydrolysate was fractionated by means of a crossflow tangential ultrafiltration system, with a 3 kDa molecular weight cut-off ceramic membrane. A response surface design of the composite central factorial type was implemented to evaluate the effect of pH, transmembrane pressure, and flow factors on the response variables transmission, volume reduction factor (VRF) and permeate flow resistance. The transmissions focused on the antioxidant peptides, measured by three conventional methods such as TEAC, FRAP, ORAC, also known as TTEAC, TFRAP and TORAC, respectively. The evaluated resistances were the total resistance (Rtotal), fouling resistance (Rfouling), and gel resistance (Rgel). Result: The results showed that the three factors evaluated affect all the response variables either in their linear or quadratic terms or by some interaction. For each response variable, a mathematical model was obtained, with statistical significance and a non-significant lack of adjustment. The models obtained were used for a multi-objective optimization process in which transfers were maximized, and resistances were minimized. The efficiency of the optimum ultrafiltration process was 25 %. Conclusion: The neutral-alkaline pH is ideal for the ultrafiltration process of bioactive peptides, as it is where the highest transmissions of peptides with antioxidative capacity are found. Under optimal conditions, the 3 kDa membrane permeate was found to exhibit higher antioxidant capacity than the retentate and feed. Based on this, the fraction of less than 3 kDa emerges as a potential multifunctional ingredient, thanks to its antioxidant properties.

With the expansion of organic agriculture, research is needed to indicate economically and ecologically viable fertilizer options, especially in semiarid regions, with low soil organic matter and nitrogen content. In the Brazilian semiarid region, vermicomposts are widely used by farmers and are scientifically investigated; however, there is no information for millicompost, a new type of organic compound that has shown very promising results in other regions. Thus, this study aimed to analyze the decomposition rate, nutrient release, and microstructure evaluation of vermicomposts from different sources and of millicompost produced from plant residues, with the application of mineral nitrogen-urea and organo-mineral fertilizer in the Brazilian semiarid region. The experimental design was a randomized block in a 4 × 3 factorial scheme, with four replicates; four organic composts (millicompost, commercial vermicompost, vermicompost from bovine manure, vermicompost from goat manure); and three types of fertilization (without fertilizer, with mineral-urea and organo-mineral fertilizer). The organic composts were decomposed using litterbags at the soil surface. The variable's decomposition rate and the nutrient release were evaluated at six-time intervals (0, 30, 60, 90, 120, and 150 days), and microstructure was evaluated at the beginning and the end of the experiment, with scanning electron microscopy (SEM). The highest decomposition was verified for commercial vermicompost rich in macro and micronutrients and with lower P contents. The lignin:N ratio and the initial P content were more important in the permanence of the organic compost in the field than the C:N ratio. Regardless of the organic composts, the use of urea as a mineral fertilizer stimulated decomposition more than the organo-mineral fertilizer. The initial composition of the nutrients was decisive in the dynamics of nutrient release, mass loss, and decomposition of C. There was no pattern in the release order of macronutrients. However, for the micronutrients, the release order was Cu > Fe > Mn, in all treatments. Microstructure analysis is a visual analysis where differences are detected through microphotographs and the biggest difference occurred with millicompost, which showed elongated fibers and fiber bundles, forming a relatively open structure characteristic of the presence of fulvic acid. However, the addition of organo-mineral fertilizer formed agglomerates in compacted micro-portions, helping the mineralization of C and N.

Urban tropical lagoons are commonly impacted by silting, domestic sewage and industrial wastes and the dredging of their sediments is often required to minimize environmental impacts. However, the ecological implications of land disposal of dredged sediments are still poorly investigated in the tropics. Aiming to contribute to filling this gap, an ecotoxicological evaluation was conducted with dredged sediments from Tijuca Lagoon (Rio de Janeiro, Brazil) using different lines of evidence, including soil and sediment characterization, metal determination, and acute and avoidance bioassays with Eisenia andrei. Two different dredged sediment samples, a sandy sediment and another muddy one, were obtained in two distinct and spatially representative sectors of the Tijuca Lagoon. The sediments were mixed with an artificial soil, Ferralsol and Spodosol to obtain doses between 0 (pure soil) and 12%. The sediment dose that caused mortality (LC50) or avoidance responses (EC50) to 50% of the organisms was estimated through PriProbit analysis. Metal concentrations and toxicity levels were higher in the muddy sediment (artificial soil LC50 = 3.84%; Ferralsol LC50 = 4.58%; Spodosol LC50 = 2.85%) compared to the sandy one (artificial soil LC50 = 10.94%; Ferralsol LC50 = 14.36%; Spodosol LC50 = 10.38%), since fine grains tend to adsorb more organic matter and contaminants. Mortality and avoidance responses were the highest in Spodosol due to its extremely sandy texture (98% of sand). Metal concentrations in surviving earthworms were generally low, except sodium whose bioaccumulation was high. Finally, the toxicity is probably linked to marine salts, and the earthworms seem to accumulate water in excess to maintain osmotic equilibrium, increasing their biomass.

Chronic exposure to stress is a non-adaptive situation that is associated with mitochondrial dysfunction and the accumulation of reactive oxygen species (ROS), especially superoxide anion (SA). This accumulation of ROS produces damage-associated molecular patterns (DAMPs), which activate chronic inflammatory states and behavioral changes found in several mood disorders. In a previous study, we observed that an imbalance of SA triggered by rotenone (Ro) exposure caused evolutionarily conserved oxi-inflammatory disturbances and behavioral changes in Eisenia fetida earthworms. These results supported our hypothesis that SA imbalance triggered by Ro exposure could be attenuated by lithium carbonate (LC), which has anti-inflammatory properties. The initial protocol exposed earthworms to Ro (30 nM) and four different LC concentrations. LC at a concentration of 12.85 mg/L decreased SA and nitric oxide (NO) levels and was chosen to perform complementary assays: (1) neuromuscular damage evaluated by optical and scanning electron microscopy (SEM), (2) innate immune inefficiency by analysis of Eisenia spp. extracellular neutrophil traps (eNETs), and (3) behavioral changes. Gene expression was also evaluated involving mitochondrial (COII, ND1), inflammatory (EaTLR, AMP), and neuronal transmission (nAchR α5). LC attenuated the high melanized deposits in the circular musculature, fiber disarrangement, destruction of secretory glands, immune inefficiency, and impulsive behavior pattern triggered by Ro exposure. However, the effects of LC and Ro on gene expression were more heterogeneous. In summary, SA imbalance, potentially associated with mitochondrial dysfunction, appears to be an evolutionary component triggering oxidative, inflammatory, and behavioral changes observed in psychiatric disorders that are inhibited by LC exposure.

Hydrogels are polymers of great importance due to their multiple applications, which have led to an exponential increase in their production. However, once they have fulfilled their function, they become waste and their ecotoxicological effects are unknown. The aim of the present study was to evaluate the acute toxicity and total antioxidant capacity of the earthworm (Eisenia fetida) exposed to a terpolymeric hydrogel (acrylic acid, acrylamide, and 2-acrylamido-2-methyl-1-propane-sulfonic acid) crosslinked with modified kraft lignin. Four different amounts of hydrogel per unit area were evaluated (0.0924, 0.1848, 0.9242, and 1.848 mg hydrogel/cm2) plus a control, and three replicates were performed for each group. Starting from the amount of 0.1848 mg hydrogel/cm2, the earthworms showed physiological and behavioral alterations; at higher amounts, 0.9242 and 1.848 mg hydrogel/cm2, more acute signs were observed with mortality rates of 51.7% and 100%, respectively. On the other hand, the antioxidant activity assay showed that the higher the hydrogel exposure amount, the higher the oxidative stress, as evidenced by lower antioxidant activity (67.09% inhibition of the ABTSâ+ radical). Therefore, we concluded that the lignin-modified hydrogel generated oxidative stress and acute lethal toxic effects in Eisenia fetida.

Although the best-known use of the different species of earthworm is the production of organic fertilizer, they can also be considered a rich source of biologically and pharmacologically-active compounds, for use in the treatment of various diseases. In recent decades, with the development of biochemical technologies, research on the pharmaceutical effects of compounds extracted from different species of earthworms has begun. Enzymatic hydrolysis is the most common and widely used technique for producing bioactive hydrolysates, because it uses moderate operating conditions, with a certain specificity for the substrate. In the present study, the objective was to optimize and scale up the enzymatic hydrolysis of Eisenia foetida protein to obtain peptides with biological activity. The substrate characterization was carried out according to AOAC, a response surface design was performed for the optimization of the enzymatic hydrolysis and then the scaling was performed by means of dimensional analysis. The results show that the major component of the paste is protein, 65% of which is albumin, and the absence of pathogenic microorganisms was also found. Regarding optimization, it was found that the optimal hydrolysis conditions are achieved with pH = 8.5; temperature = 45 °C; amount of substrate = 125 g and volume of enzyme = 1245µL. For the scaling, 4 dimensionless pi-numbers were calculated which describe the process with no statistically significant differences between the model and the prototype; it can be concluded that the enzymatic hydrolysate of Eisenia foetida presents high values of antioxidant activity evaluated by different methodologies.

Vermicomposting is the bio-oxidation and stabilization of organic matter involving relationships between the action of earthworms and microorganisms and the activation and dynamics of several enzyme activities. Semi-arid farmers to make (extra) money and organic production, produce their vermicompost using plant residues and animal manure, but there is no information about the final product generated. Thus, this study aimed to analyze the potential of vermicomposting with mixtures of animal manure and vegetable leaves in the development of Eisenia foetida, microbial biomass, and enzymatic activity in the semi-arid region, Brazil. The experimental design applied was randomized block in a 6 × 4 factorial scheme with four replicates, with six treatments (mixtures of cattle manure, goat manure, cashew leaves, and catanduva leaves) and evaluated at four-time intervals (30, 60, 90, and 120 days of vermicomposting). The treatments were placed in polyethylene pots in the same site, environmental conditions, and residues proportions as used by farmers. The characteristics analyzed were the number of earthworms (NE), total earthworm biomass (TEB) and earthworm multiplication index (MI), microbial biomass carbon (MBC), and activities of enzymes ß-glucosidase, dehydrogenase, alkaline and acid phosphatases. The cattle manure vermicomposted shows the highest average values observed for NE, MI, TEB, MBC, and enzymatic activity, regardless of the plant leaves mix. In general, the enzymes activities were found in the descending order of ß-glucosidase > alkaline phosphatase > dehydrogenase > acid phosphatase. The maturation dynamics of vermicompost were characterized by a decline in the microbial population and number and biomass of earthworms in the substrate and consequently a decrease in new enzyme synthesis and degradation of the remaining enzyme pool. Microbial biomass and enzymatic activity were indicators for changes in the quality of vermicompost.

In this study, P. posthuma coelomic fluid (PCF) and body paste (PBP) was evaluated as antibacterial agent against ampicillin (AMP) resistant five Gram positive and four Gram negative clinical isolates. The antibacterial effect of different doses (i.e. 25-100 µg/ml) of PCF and PBP along with AMP and azithromycin (AZM) (negative and positive controls, respectively) were observed through disc diffusion and micro-dilution methods. All nine clinical isolates were noticed as AMP resistant and AZM sensitive. Antibacterial effects of PCF and PBP were dose dependent and zone of inhibitions (ZI) against all clinical isolates were between 23.4 ± 0.92 to 0 ± 00 mm. The sensitivity profile of PCF and PBP against clinical isolates was noticed as 44.44 and 55.56%, respectively. Both PCF and PBP showed bacteriostatic (BTS) action against S. aureus, S. pyogenes, K. pneumonia, N. gonorrhoeae. Moreover, the cumulative BTS potential of PCF and PBP against all isolates was 66.67 and 55.56%, respectively. The MICs of PCF and PBP were ranged from 50-200 µg/ml against selected isolates. The bacterial growth curves indicated that PCF and PBP inhibited the growth of all isolates at their specific MIC concentrations. However, PBP has better antibacterial potential compared to PCF against selected isolates. Therefore, it is concluded that both PCF and PBP of P. posthuma possess antibacterial and BTS potential against ampicillin resistant clinical isolates. This organism might be considered as a second choice of antibacterial agents and can further be utilized in pharmaceutical industries for novel drug manufacturing by prospecting bioactive potential agents.(AU)

Portanto, conclui-se que tanto o PCF quanto o PBP de P. posthuma possuem potencial antibacteriano e BTS contra isolados clínicos resistentes à ampicilina. Esse organismo pode ser considerado como uma segunda escolha de agentes antibacterianos e pode ainda ser utilizado nas indústrias farmacêuticas para a fabricação de novos medicamentos por meio da prospecção de agentes com potencial bioativo.(AU)

BACKGROUND: Some studies have suggested that mitochondrial dysfunction and a superoxide imbalance could increase susceptibility to chronic stressful events, contributing to the establishment of chronic inflammation and the development of mood disorders. The mitochondrial superoxide imbalance induced by some molecules, such as rotenone, could be evolutionarily conserved, causing behavioral, immune, and neurological alterations in animals with a primitive central nervous system. OBJECTIVE: Behavioral, immune, and histological markers were analyzed in Eisenia fetida earthworms chronically exposed to rotenone for 14 days. METHODS: Earthworms were placed in artificial soil containing 30 nM of rotenone distributed into a plastic cup that allowed the earthworms to leave and return freely into the ground. Since these organisms prefer to be buried, the model predicted that the earthworms would necessarily have to return to the rotenone-contaminated medium, creating a stressful condition. The effect on survival behavior in the immune and histological body wall and ventral nervous ganglia (VNG) structures, as well as gene expression related to inflammation and mitochondrial and neuromuscular changes. RESULTS: Rotenone-induced loss of earthworm escape behavior and immune alterations indicated a chronic inflammatory state. Some histological changes in the body wall and VNG indicated a possible earthworm reaction aimed at protecting against rotenone. Overexpression of the nicotinic acetylcholine receptor gene (nAChR α5) in neural tissues could also help earthworms reduce the degenerative effects of rotenone on dopaminergic neurons. CONCLUSION: These data suggest that mitochondrial dysfunction could be an evolutionarily conserved element that induces inflammatory and behavioral changes related to chronic stress.

Both earthworms and terrestrial isopods have been used to evaluate the quality of contaminated soil by NPs. However, most experiments have been conducted in the laboratory and under greenhouse conditions. Besides, little is known of Fe accumulation in earthworms from iron NPs (Fe NPs) under natural conditions. Therefore, the objective of this research was to evaluate the effect of manufactured NPs on the accumulation of Fe in macroinvertebrates from forest soil. Our results revealed that earthworms consume low amounts of Fe in a concentration of 1000 mg Fe NPs kg-1 of dry soil, with a behavior constant over time. Besides, we observed that earthworms could not detect Fe at low concentrations (1 or 10 mg Fe NPs kg-1), so they do not limit soil consumption, which translates into high amounts of Fe in their bodies. By contrast, the content of Fe in organisms is inversely proportional to increasing concentrations in the soil (R2 = -0.41, p < 0.05). Therefore, although studies are needed, in addition to considering environmental factors and the physicochemical properties of the soil, endogenous worms in the evaluated area could, under natural conditions, be useful to inform us of contamination of NP manufactured from Faith. Besides, for future research, a novel methodology should be considered to demonstrate more realistic avoidance behavior under field conditions.

Pontoscolex corethrurus (Müller, 1857) is an invasive tropical earthworm, globally distributed. It reproduces through parthenogenesis, which theoretically results in low genetic diversity. The analysis of the population structure of P. corethrurus using molecular markers may significantly contribute to understanding the ecology and reproductive system of this earthworm species. This work assessed the genetic diversity and population structure of P. corethrurus with 34 polymorphic inter simple sequence repeat markers, covering four populations in tropical and temperate pastures from Veracruz State. Nuclear markers distinguished two genetic clusters, probably corresponding to two distinct genetic lineages. The number of clones detected in the AC population was lower than expected for a parthenogenetic species. Also, the apparent lack of differences in population structures related to the geographic region among the populations studied may indicate that human-mediated transference is prevalent in these areas. Still, most individuals apparently belong to lineage A, and only a few individuals seem to belong to the lineage B. Thus, the admixture signatures found among the four populations of P. corethrurus may have facilitated a successful invasion by directly increasing fitness. In summary, addressing the genetic variation of P. corethrurus with ISSR markers was a suitable approach, as it evidenced the genetic diversity and relationships in the populations evaluated.

A rapid bioassay is presented for determining acute toxicity directly in soil. Modifying the Organisation for Economic Cooperation and Development (OECD) protocol 207, it uses a thin layer of moistened soil laid directly in the bottom of the bioassay jar into which the earthworms are placed and incubated. Examples are presented in comparisons between the soil contact bioassay vs. the filter paper bioassay run on Toxicity Characteristic Leaching Procedure (TCLP) extracts of pesticide contaminated soil and petroleum drilling cuttings. In 2,4-dichlorophenoxyacetic acid (2,4-D) contaminated soil (300mg/Kg), no mortality was found in soil extracts, but 100% mortality was found when exposed directly to soil. Treatment with the Daremend® product in five anaerobic/aerobic cycles slowly reduced the 24 h mortality (0%) but still showed 100% mortality at 48 h. However, severe sub-lethal effects (expulsion of celomic/bloody fluids) were reduced from 50% to 37%, and further treatment may reduce the toxicity to acceptable levels. The petroleum drilling cuttings treated by chemical oxidation (1.3% H2O2, w/w) and bioremediation (simulation of biopiles), showed a similar response, where 0% mortality in soil extracts was found, but 100% mortality with soil contact. Post-treatment with chemical oxidation resulted in a reduction in the soil contact bioassay to 3% and 13% mortality, within the accepted range (≤10%) of the OECD protocol. Observations are presented with respect to moisture control to prevent earthworm desiccation and recommendation for confirmation using the sub-chronic test in the OECD protocol but by testing the contaminated/treated soil itself rather than artificial soil.

The amount of nanoparticles (NP), such as TiO2, has increased substantially in the environment. It is still largely unknown, however, how NP might interact with earthworms and organic material and how this might affect the bacterial community structure and their functionality. Therefore, an arable soil was amended with TiO2 NP at 0, 150 or 300 mg kg-1 and subjected to different treatments. Treatments were soil amended with ten earthworms (Eisenia fetida (Savigny, 1826)) with fully developed clitellum and an average fresh mass of 0.5 to 500 g dry soil, 1.75 g tyndallized Quaker® oat seeds Avena sativa (L.) kg-1, or earthworms plus oat seeds, or left unamended. The bacterial community structure was monitored throughout the incubation period. The bacterial community in the unamended soil changed over time and application of oats, earthworm and a combination of both even further, with the largest change found in the latter. Application of NP to the unamended soil and the earthworm-amended soil altered the bacterial community, but combining it by adding oats negated that effect. It was found that the application of organic material, that is, oats, reduced the effect of the NP applied to soil. However, as the organic material applied was mineralized by the soil microorganisms, the effect of NP increased again over time.
