RESUMO

The rapid global emergence of antibiotic resistance genes (ARGs) is a substantial public health concern. Livestock manure serves as a key reservoir for tetracycline resistance genes (TRGs), serving as a means of their transmission to soil and vegetables upon utilization as a fertilizer, consequently posing a risk to human health. The dynamics and transfer of TRGs among microorganisms in vegetables and fauna are being investigated. However, the impact of different vegetable species on acquisition of TRGs from various manure sources remains unclear. This study investigated the rhizospheres of three vegetables (carrots, tomatoes, and cucumbers) grown with chicken, sheep, and pig manure to assess TRGs and bacterial community compositions via qPCR and high-throughput sequencing techniques. Our findings revealed that tomatoes exhibited the highest accumulation of TRGs, followed by cucumbers and carrots. Pig manure resulted in the highest TRG levels, compared to chicken and sheep manure, in that order. Bacterial community analyses revealed distinct effects of manure sources and the selective behavior of individual vegetable species in shaping bacterial communities, explaining 12.2% of TRG variation. Firmicutes had a positive correlation with most TRGs and the intl1 gene among the dominant phyla. Notably, both the types of vegetables and manures significantly influenced the abundance of the intl1 gene and soil properties, exhibiting strong correlations with TRGs and elucidating 30% and 17.7% of TRG variance, respectively. Our study delineated vegetables accumulating TRGs from manure-amended soils, resulting in significant risk to human health. Moreover, we elucidated the pivotal roles of bacterial communities, soil characteristics, and the intl1 gene in TRG fate and dissemination. These insights emphasize the need for integrated strategies to reduce selection pressure and disrupt TRG transmission routes, ultimately curbing the transmission of tetracycline resistance genes to vegetables.

As today the 137Cs fallout peak, in sediment cores, corresponds only to 25% of its initial concentration, alternatives to the use of 137Cs as a210Pb sediment dating validation tool are proposed. In highly industrialized bays, such as Guanabara Bay in the Rio de Janeiro metropolitan region, several chemical/compounds from the surrounding industry releases may be applied as validation tools. Chromium and copper profiles in a sediment core adequately fit the expected pattern due to the implementation of a chemical plant in 1958, reaching their maximum discharge in 1982 and subsequently decreasing due to the operation of a new wastewater treatment plant. A diffuse source such, as the PCB-based mixture Askarel, was also applied as an alternative validation tool, and the observed concentration profile reproduced the expected behavior, with increasing concentration after the Second World War and a decrease after the ban of this product in 1981. The observed Aroclor 1254 and 1260 mixture chlorination rates fit the most widely distributed PCB-based products in the country.

The objectives of this research were to assess ingested plastics and accumulated heavy metals in four urban gull species. Additionally, the relationships between ingested plastics and selected demographic and health metrics were assessed. Between 2020-2021 during the non-breeding seasons, 105 gulls (46 American herring gulls (HERG, Larus argentatus smithsonianus), 39 great black-backed gulls (GBBG, Larus marinus), 16 Iceland gulls (Larus glaucoides), 4 glaucous gulls (Larus hyperboreus)) were killed at a landfill in coastal Newfoundland and Labrador, Canada, as part of separate, permitted kill-to-scare operations related to aircraft safety. Birds were necropsied, the upper gastrointestinal tract contents were processed using standard techniques, and livers were analyzed for accumulated As, Cd, Hg, and Pb. The relationships between ingested plastics, demographics, and health metrics were assessed in HERG and GBBG. Across all four species, 85 % of birds had ingested at least one piece of anthropogenic debris, with 79 % ingesting at least one piece of plastic. We detected interspecific differences in plastic ingestion and hepatic trace metals, with increased ingested plastics detected in GBBG compared with HERG. For GBBG, levels of ingested plastic were relatively greater for birds with higher scaled mass index, while HERG with more ingested plastic had higher liver lead concentrations.

The human silencing hub (HUSH) preserves genome integrity through the epigenetic repression of invasive genetic elements. However, despite our understanding of HUSH as an obligate complex of three subunits, only loss of MPP8 or Periphilin, but not TASOR, triggers interferon signaling following derepression of endogenous retroelements. Here, we resolve this paradox by characterizing a second HUSH complex that shares MPP8 and Periphilin but assembles around TASOR2, an uncharacterized paralog of TASOR. Whereas HUSH represses LINE-1 retroelements marked by the repressive histone modification H3K9me3, HUSH2 is recruited by the transcription factor IRF2 to repress interferon-stimulated genes. Mechanistically, HUSH-mediated retroelement silencing sequesters the limited pool of the shared subunits MPP8 and Periphilin, preventing TASOR2 from forming HUSH2 complexes and hence relieving the HUSH2-mediated repression of interferon-stimulated genes. Thus, competition between two HUSH complexes intertwines retroelement silencing with the induction of an immune response, coupling epigenetic and immune aspects of genome defense.

RESUMO

Chemicals that modulate phytohormones serve as a research tool in plant science and as products to improve crop productivity. Subtype selectivity refers to a ligand to selectively bind to specific subtypes of a receptor rather than binding to all possible subtypes indiscriminately. It allows for precise and specific control of cellular functions and is widely used in medicine. However, subtype selectivity is rarely mentioned in the realm of plant science, and it requires integrated knowledge from chemistry and biology, including structural features of small molecules as ligands, the redundancy of target proteins, and the response of signaling factors. Here, we present a comprehensive review and evaluation of phytohormone receptor subtype selectivity, leveraging the chemical characteristics of phytohormones and their analogues as clues. This work endeavors to provide a valuable research strategy that integrates knowledge from chemistry and biology to advance research efforts geared toward enhancing crop productivity.

The PM2.5 and PM10 particles were characterized in terms of morphology (size and shape) and surface elemental composition at two different (traffic and industrial) locations in urban region of India and further linked to different morphological defining parameters. The overall PM2.5 and PM10 showed significant daily variability indicating higher PM10 as compared to PM2.5. PM2.5/PM10 ratio was found to be 0.58 ± 0.10 indicating the abundance of PM2.5. Soot aggregates, aluminosilicates, and brochosomes particles were classified based on morphology, aspect ratio (AR), and surface elemental composition of single particles. The linear regression analysis indicates the significant correlation between area equivalent (Daeq) and feret diameter (Dfd) (R2 0.86-0.98). Higher aspect ratio (1.48 ± 0.87-1.43 ± 0.50) was noted at traffic site as compared to industrial site (1.33 ± 0.58-1.29 ± 0.30), while circularity showed the opposite trend. Fractal dimension (Df) of soot aggregates estimated by the soot parameters method (SPM) were found to be 1.70, 1.72, and 1.88, mainly attributed to vehicular emissions, biomass, and industrial emission/coal burning, respectively. This further inferred that freshly emitted soot particles exhibited lacey in nature with spherical shape (Df 1.70) at traffic site, while at industrial location, they were different with compact shapes (Df 1.88) due to particle aging processes. This study inferred the synoptic changes in mass, chemical characteristics, and morphology of aerosol particles which provide the new insights into individual atmospheric particle and their dynamic nature.

Pharmaceuticals are widespread in aquatic environments and might contribute to the prevalence of antibiotic resistance. However, the co-effect of antibiotics and non-antibiotic pharmaceuticals on the gut microbiome of fish is poorly understood. In this study, we characterized the variation of the zebrafish gut microbiome and resistome after exposure to sulfamethoxazole (SMX) and aspirin under different treatments. SMX contributed to the significant increase in the antibiotic resistance genes (ARGs) richness and abundance with 46 unique ARGs and five mobile genetic elements (MGEs) detected. Combined exposure to SMX and aspirin enriched total ARGs abundance and rearranged microbiota under short-term exposure. Exposure time was more responsible for resistome and the gut microbiome than exposure concentrations. Perturbation of the gut microbiome contributed to the functional variation related to RNA processing and modification, cell motility, signal transduction mechanisms, and defense mechanisms. A strong significant positive correlation (R=0.8955, p<0.001) was observed between total ARGs and MGEs regardless of different treatments revealing the key role of MGEs in ARGs transmission. Network analysis indicated most of the potential ARGs host bacteria belonged to Proteobacteria. Our study suggested that co-occurrence of non-antibiotics and antibiotics could accelerate the spread of ARGs in gut microbial communities and MGEs played a key role.

Pamphagidae is a family of Acridoidea that inhabits the desert steppes of Eurasia and Africa. This study employed flow cytometry to estimate the genome size of eight species in the Pamphagidae. The results indicate that the genome size of the eight species ranged from 13.88 pg to 14.66 pg, with an average of 14.26 pg. This is the largest average genome size recorded for the Orthoptera families, as well as for the entire Insecta. Furthermore, the study explored the role of repetitive sequences in the genome, including their evolutionary dynamics and activity, using low-coverage next-generation sequencing data. The genome is composed of 14 different types of repetitive sequences, which collectively make up between 59.9% and 68.17% of the total genome. The Pamphagidae family displays high levels of transposable element (TE) activity, with the number of TEs increasing and accumulating since the family's emergence. The study found that the types of repetitive sequences contributing to the TE outburst events are similar across species. Additionally, the study identified unique repetitive elements for each species. The differences in repetitive sequences among the eight Pamphagidae species correspond to their phylogenetic relationships. The study sheds new light on genome gigantism in the Pamphagidae and provides insight into the correlation between genome size and repetitive sequences within the family.

Rare earth elements (REEs) are a group consisting of the following 17 metals: Ce, Dy, Er, Eu, Gd, Ho, La, Lu, Nd, Pr, Pm, Sc, Sm, Tb, Tm, Y and Yb. In the current century, the number of applications of REEs has significantly increased. They are being used as components in high technology devices of great importance industrial/economic. However, information on the risk of human exposure to REEs, as well as the potential toxic effects of these elements is still limited. In general terms, dietary intake is the main route of exposure to metals for non-occupationally exposed individuals, which should be also expected for REEs. The current paper aimed at reviewing the studies -conducted over the world- that focused on determining the levels of REEs in foods, as well as the dietary intake of these elements. Most studies do not suggest potential health risk for consumers of freshwater and marine species of higher consumption, or derived from the intake of a number of vegetables, fruits, mushrooms, as well as other various foodstuffs (honey, tea, rice, etc.). The current estimated daily intake (EDI) of REEs does not seem to be of concern. However, considering the expected wide use of these elements in the next years, it seems to be clearly recommendable to assess periodically the potential health risk of the dietary exposure to REEs. This is already being done with well-known toxic elements such as As, Cd, Pb and Hg, among other potentially toxic metals.

Potentially toxic elements (PTEs) in seawater and sediments may be amplified along the aquatic food chain, posing a health threat to humans. This study comprehensively analyzed the concentrations, distribution, potential sources, and health risk of 7 PTEs in multimedia (seawater, sediment and organism) in typical subtropical bays in southern China. The results indicated that Zn was the most abundant element in seawater, and the average concentration of Cd in sediment was 3.93 times higher than the background value. Except for As, the seasonal differences in surface seawater were not significant. The content of Zn in fishes, crustacea, and shellfish was the highest, while the contents of Hg and Cd were relatively low. Bioaccumulation factor indicated that Zn was a strongly bioaccumulated element in seawater, while Cd was more highly enriched by aquatic organisms in sediment. According to principal component analysis (PCA), and positive matrix factorization (PMF), the main sources of PTEs in Quanzhou Bay were of natural derivation, industrial sewage discharge, and agricultural inputs, each contributing 40.4 %, 24.2 %, and 35.4 %, respectively. This study provides fundamental and significant information for the prevention of PTEs contamination in subtropical bays, the promotion of ecological safety, and the assessment of human health risk from PTEs in seafood.

AIM: To analyse postnatal characteristics, clinical and laboratory findings, results of investigations in the newborn (25 gestational weeks; Apgar score: 6/9 points; born per caesarean section; birth weight: 600 g; birth length: 31 cm; head circumference: 21 cm) from the first high-risk pregnancy with acquired form of acrodermatitis enteropathica. RESULTS: After summarizing the clinical picture with laboratory findings, we analysed the components of parenteral nutrition with regard to the deficiency of trace elements and vitamins. The zinc depletion dominated. CONCLUSION: The diagnosis is clinical, based on the presence of a typical clinical picture together with a low serum zinc concentration. Standard preparations with elementary elements do not sufficiently cover the daily needs of children, other possibilities of supplementation in intravenous form are not available. It is necessary to supplement zinc in premature children, in children with high losses of zinc (with diarrhoea, in patients with a stoma, in patients with severe skin disease) (Fig. 4, Ref. 15).

A significant concern in our society is the potential impact on both health and the environment of air pollutants released during the incineration of waste. Therefore, it is crucial to conduct thorough control and monitoring measures. In this context, the objective of this research was to study the evolution of particulate matter (PM2.5) and associated trace elements during the period before and after the installation of an Energy Recovery Plant (ERP). For that, a descriptive and temporal analysis of PM2.5 concentration and composition were performed on two similar areas (impact/control) using the Before-After/Control-Impact (BACI) design and two periods (before from January 01, 2018 to February 06, 2020 and after from December 10, 2020 to September 30, 2022). Results showed a decrease in the levels of PM2.5 and associated trace elements is observed in the impact zone (IZ) and in the control zone (CZ) throughout the study period. In the case of PM2.5, the most notable decrease occurred in the period of the start-up of the ERP, a period that coincides with the confinement and restrictions of COVID, with a subsequent increase in both zones, without reaching the levels observed in the period prior to the start-up of the ERP. Selenium is the only trace element that increases significantly in the IZ. In conclusion, a decrease is observed for all pollutants except selenium in both zones, although less pronounced in the IZ. Since selenium already showed an upward trend in the phase prior to the start of the ERP, it is necessary to investigate its evolution and find out the possible cause.

Ophiuroids, as an important group of echinoderms, are widely distributed in marine benthic habitats. Previous studies have identified two primary feeding types of ophiuroids in the Yellow Sea, including carnivorous (Ophiura sarsii vadicola and Stegophiura sladeni) and suspension feeders (Ophiopholis mirabilis). Despite their ecological role in the benthic food webs, little is known about their accumulation of trace metal elements (TMEs). In this study, the content of TMEs (Pb, As, Cd, Hg, Cr, Cu, Zn), methylmercury (MeHg) and δ15N value of three ophiuroids species from the North Yellow Sea were determined. Our results showed that the contents of some TMEs (As, Cd, Cr, Cu and Zn) and MeHg were significantly different in three species of ophiuroid (p < 0.05). There were significant correlations between the accumulations of trace metal elements (Pb, Cd and Zn) and the δ15N value of the ophiuroids (p < 0.05). Additionally, As and Zn exhibited opposite correlations in ophiuroid with two feeding types, which may be related to their host species and different feeding habits. This study provided fundamental data for understanding the distribution of trace metal elements in echinoderms.

To identify a novel optimized strategy for preventing fraudulent substitutions of squid species and origins, forty European squids (Loligo vulgaris) and forty flying squids (Todarodes sagittatus) from the Mediterranean Sea and Atlantic Ocean were analyzed for δ13C, δ15N, La, Ce, Pr, Nd, Sm, Eu, Gd, Tb, Dy, Ho, Er, Yb, and Lu using isotope ratio mass spectrometry and inductively coupled plasma-mass spectrometry. While δ13C and δ15N variations were mainly species-related, they alone could not reliably distinguish samples. To address this issue, decision rules were developed using Classification and Regression Tree analysis. Threshold values for δ13C (-19.91‰), δ15N (14.87‰), and Pr (0.49 µg kg-1) enabled successful discrimination among Mediterranean European squids, Atlantic European squids, Mediterranean flying squids, and Atlantic flying squids, achieving over 90% accuracy, 81% precision, 80% sensitivity, and 93% specificity. This method holds promise for enhancing traceability and safety in the seafood industry, ensuring product integrity and consumer trust.

Patients with malabsorptive conditions can develop micronutrient deficiencies, even if they receive vitamins, minerals, or trace elements through their enteral or parenteral nutrition. Additionally, clinicians have faced challenges with micronutrient supplementation during parenteral product shortages and when transitioning patients from parenteral to enteral/oral nutrition. Evaluating micronutrient deficiencies through laboratory markers has various limitations, including that many are acute phase reactants, may not reflect storage status, or may not be readily available in clinical practice. Furthermore, clinicians can become overwhelmed with the variety of vitamin and mineral products available, the differences in dosages and ingredients in these products, and lastly, the inherent challenges associated with an impaired gastrointestinal tract. The current review will discuss some challenges clinicians may encounter in clinical practice during the evaluation, assessment, and prescription of micronutrient supplementation in patients with malabsorptive conditions.

Buckwheat starch has attracted worldwide attention in the food industry as a valuable raw material or food additive. Nitrogen (N) and sulfur (S) are two nutrients essential to ensure grain quality. This study investigated the combined application of N fertilizer (0, 45 and 90 kg N ha-1) and S fertilizer (0 and 45 kg SO3 ha-1) on the chemical composition, structure and physicochemical properties of buckwheat starch. The results showed that increasing the fertilizer application decreased amylose content and starch granule size but increased light transmittance, water solubility and swelling power. The stability of the absorption peak positions and the decrease in short-range order degree suggested that fertilization influenced the molecular structure of buckwheat starch. In addition, increases in viscosity and gelatinization enthalpy as well as decreases in gelatinization temperatures and dynamic rheological properties indicated changes in the processing characteristics and product quality of buckwheat-based foods.

All-RNA-mediated targeted gene integration methods, rendering reduced immunogenicity, effective deliverability with non-viral vehicles, and a low risk of random mutagenesis, are urgently needed for next-generation gene addition technologies. Naturally occurring R2 retrotransposons hold promise in this context due to their site-specific integration profile. Here, we systematically analyzed the biodiversity of R2 elements and screened several R2 orthologs capable of full-length gene insertion in mammalian cells. Robust R2 system gene integration efficiency was attained using combined donor RNA and protein engineering. Importantly, the all-RNA-delivered engineered R2 system showed effective integration activity, with efficiency over 60% in mouse embryos. Unbiased high-throughput sequencing demonstrated that the engineered R2 system exhibited high on-target integration specificity (99%). In conclusion, our study provides engineered R2 tools for applications based on hit-and-run targeted DNA integration and insights for further optimization of retrotransposon systems.

Recent advances in DNA sequencing technology have enabled the precise decoding of genomes in non-model organisms, providing a basis for unraveling the patterns and mechanisms of sex chromosome evolution. Studies of different species have yielded conflicting results regarding the traditional theory that sex chromosomes evolve from autosomes via the accumulation of deleterious mutations and degeneration of the Y (or W) chromosome. The concept of the 'sex chromosome cycle,' emerging from this context, posits that at any stage of the cycle (i.e., differentiation, degeneration, or loss), sex chromosome turnover can occur while maintaining stable sex determination. Thus, understanding the mechanisms that drive both the persistence and turnover of sex chromosomes at each stage of the cycle is crucial. In this review, we integrate recent findings on the mechanisms underlying maintenance and turnover, with a special focus on several organisms having unique sex chromosomes. Our review suggests that the diversity of sex chromosomes in the maintenance of stable sex determination is underappreciated and emphasizes the need for more research on the sex chromosome cycle.

This study assessed the effectiveness of nano zero-valent iron loaded on biochar (BC-nZVI) during swine manure composting. BC-nZVI significantly reduced the abundance of antibiotic resistance genes (ARGs), metal resistance genes (MRGs), and mobile genetic elements (MGEs). BC-nZVI modified the preference of MGEs to carry ARGs and MRGs, and the corrosion products of BC-nZVI could destroy cell structure, hinder electron transfer between cells, and weaken the association between ARGs, MRGs, and host bacteria. Functional genes analysis revealed that BC-nZVI down-regulated the abundance of genes affecting the transmission and metabolism of ARGs and MRGs, including type IV secretion systems, transporter systems, two-component systems, and multidrug efflux pumps. Furthermore, the BC-nZVI decreased genes related to flagella and pili production and cell membrane permeability, thereby hindering the transfer of ARGs, MRGs, and MGEs in the environment. Redundancy analysis demonstrated that changes in the microbial community induced by BC-nZVI were pivotal factors impacting the abundance of ARGs, MRGs, and MGEs. Overall, this study confirmed the efficacy of BC-nZVI in reducing resistance genes during swine manure composting, offering a promising environmental strategy to mitigate the dissemination of these contaminants.
