The severity of pulpitis is a crucial factor in determining the suitable treatment. There are no clear objective indicators to assess the stage of pulp inflammation that could be used in clinical practice. The change in blood pH of the pulp during the inflammatory phase could hypothetically serve as an indicator of the pulp inflammation severity. The aim of this study was to assess the pH values in the pulpal blood of mature teeth in patients with symptomatic pulpitis, in comparison with the healthy controls. The study included patients with symptoms of pulpitis in premolar or molar teeth (Test group; n = 24, mean age 36.04, SD 7.10), and healthy controls (Control group, n = 6, mean age 24.5, SD 5.94) referred for extraction of premolars or third molars, for orthodontic reasons. The pulpal blood was taken at the opening of the endodontic access cavity, and the pH value was measured in both groups. Statistical analysis was performed using the SPSS 27.0 program with a significance level of p ≤ 0.05. The Mann-Whitney test for dependent samples was performed to evaluate the statistical difference between the groups. The patients with symptomatic pulpitis had significantly decreased pulpal blood pH compared to the healthy controls (p ≤ 0.05). The mean pulpal blood pH was 7.27 (SD 0.06) and 7.40 (SD 0.02) in Test and Control groups, respectively. In the Test group, the pulpal blood pH values were significantly lower in the patients who had symptoms for 3 days or more (7.25, SD 0.05) than in those who had symptoms for up to 3 days (7.33, SD 0.03) (p < 0.001). The pH value of the pulpal blood gradually declines in cases of symptomatic pulpitis, along with increasing duration of the symptoms.

Background The purpose of this in vitro investigation was to evaluate the impact of five distinct commercial mouthwashes on the development of Candida albicans that had been adhered to heat-cured acrylic resin sheets. Methods This in vitro investigation was carried out at the MES Medical College's Microbiology Department in Perinthalmanna, Kerala, India. A total of 72 heat-cured acrylic resin sheets, size 10 × 10 × 2 mm, were fabricated. After disinfection, all 72 acrylic sheets were placed in a flask containing a suspension of the standard strain of Candida species (American Type Culture Collection) and incubated at 37ºC for 24 hours. Then, the acrylic sheets were randomly divided into six groups, with each group containing 12 acrylic sheets. Group 1 was the control group to which no mouthwash was added. In group 2, Colgate Plax was added. In group 3, Hiora Himalaya was added. In group 4, Oral B was added. In group 5, Listerine was added. In group 6, Pepsodent was added. Colony-forming units (CFUs) were assessed using a colony counter every six, 24, 48, and 120 hours. After obtaining the pH and CFU of all 72 specimens, software known as the Statistical Package for Social Sciences (SPSS) (IBM Corp., Armonk, NY) was used to analyze the data. Results Candida albicans adhered to heat-cured denture base acrylic resin sheets differed significantly in response to commercially available mouthwashes (Oral B, Colgate Plax, and Pepsodent) and non-commercial mouthwashes (Hiora Himalaya and Listerine) that contained cetylpyridinium chloride. Conclusions Compared to other mouthwashes that do not contain cetylpyridinium chloride (Listerine and Hiora Himalaya), mouthwashes with cetylpyridinium chloride as the active ingredient (Oral B, Pepsodent, and Colgate Plax) have shown good antifungal properties against the adhering Candida albicans on denture base resin.

Introducción y objetivos Las personas con cistinuria pueden experimentar eventos recurrentes de litiasis debido a la relativa insolubilidad de la cistina en el pH fisiológico de la orina, lo que resulta en deterioro de la función renal. El pHmetro Lit-Control® es un dispositivo médico que permite la automedición precisa del pH de la orina. El objetivo principal de este estudio fue comparar la usabilidad del pHmetro Lit-Control® con las tiras reactivas para la automonitorización domiciliaria del pH urinario por parte de pacientes con cistinuria, y su satisfacción general con cada herramienta.Pacientes y métodosSe incluyeron 28 pacientes (9 mujeres y 19 varones, de 19 a 76 años), que fueron asignados aleatoriamente a monitorizar su pH urinario con tiras reactivas (n=17) o el pHmetro Lit-Control® (n=11).ResultadosDespués de 6 meses de uso, la satisfacción con los 2 métodos fue similarmente alta, pero los pacientes calificaron (en una escala de 0 a 10) mejor el pHmetro en términos de facilidad de aprendizaje (media± DE, 8,11±0,60 vs. 7,06±1,18; p=0,038), facilidad de preparación (8,22±0,67 vs. 7,25±1,18; p=0,034) y facilidad de uso (8,22±0,67 vs. 7,25±1,39; p=0,062). En general, los pacientes no alcanzaron los objetivos de alcalinización (pH entre 7,0 y 8,0).ConclusionesEl pHmetro Lit-Control® demostró ser un dispositivo fácil de usar que puede facilitar el control del pH urinario en los pacientes con cistinuria. Queda justificado un estudio prospectivo para evaluar la correlación entre la monitorización del pH de la orina, una estrategia de tratamiento por objetivo y la recurrencia de los cálculos de cistina. Acid-base titration is a quantitative analysis that enables knowing the quantity of acidic or basic groups present in a solution sample. It consists in the addition of base or acid to the solution sample while monitoring the pH to reach a neutral pH. The titration can be automated and here we present a low cost Arduino based Open Source Pump (OSPump) modified to act as an automated titrator with an obsolete but reliable Metrohm 713 pH meter. Our device is 50 times less expensive than second hand units from the pH meter manufacturer and inherently open to customization. We present two validation cases of study, including the lipolysis of a vegetable olive oil in water emulsion, characterized by the OSPump Titrator.

There remains a critical need for the detection of bacterial contamination to ensure public health. In this work, we developed a pH meter-assisted biosensor based on glucose oxidase (GOx)-conjugated magnetic zeolitic imidazolate framework-8 (mZIF-8) for on-site evaluation of bacterial contamination. The mZIF-8/GOx conjugate was produced from the electrostatic interaction between GOx and mZIF-8 and was demonstrated to inhibit GOx activity without protein denaturation. However, the presence of bacteria can cause the release of GOx from the mZIF-8 surface through competitive binding with mZIF-8, resulting in the recovery of GOx activity, which converts glucose into gluconic acid and provides an amplified pH signal. This finding allows the mZIF-8/GOx conjugate to be a biosensor for on-site detection of bacterial contamination with a pH meter as a readout. Benefiting from the magnetic separation property of mZIF-8, greatly enhanced sensitivity and precision have been achieved with detection limits of 10 cfu/mL for Escherichia coli and 30 cfu/mL for Staphylococcus aureus. Meanwhile, the flexibility of this biosensor was validated by quantitative analysis of mixed bacteria including Gram-positive and Gram-negative bacteria with desired performances. The accurate bacterial determination in contaminated drinking water samples demonstrates the applicability of this biosensor for reliable home monitoring of water quality.

In the field of stimuli-responsive materials, introducing a pH-sensitive dyestuff onto textile fabrics is a promising approach for the development of wearable sensors. In this paper, the alizarin red S dyestuff bonded with a sol-gel precursor, namely trimethoxy-[3-(oxiran-2-ylmethoxy)propyl]silane, was used to functionalize polyethylene terephthalate fabrics, a semi-crystalline thermoplastic polyester largely used in the healthcare sector mainly due to its advantages, including mechanical strength, biocompatibility and resistance against abrasion and chemicals. The obtained hybrid halochromic silane-based coating on polyester fabrics was investigated with several chemical characterization techniques. Fourier transform infrared spectroscopy and X-ray Photoelectron Spectroscopy confirmed the immobilization of the dyestuff-based silane matrix onto polyethylene terephthalate samples through self-condensation of hydrolyzed silanols under the curing process. The reversibility and repeatability of pH-sensing properties of treated polyester fabrics in the pH range 2.0-8.0 were confirmed with diffuse reflectance and CIELAB color space characterizations. Polyester fabric functionalized with halochromic silane-based coating shows the durability of halochromic properties conversely to fabric treated with plain alizarin red S, thus highlighting the potentiality of the sol-gel approach in developing durable halochromic coating on synthetic substrates. The developed wearable pH-meter device could find applications as a non-invasive pH sensor for wellness and healthcare fields.

Aldehyde dehydrogenase (ALDH) was first developed as an enzymatic signaling system of a biosensor for sensitive point-of-care detection of pathogenic bacteria. ALDH and specific aptamers to Salmonella typhimurium (S. typhimurium), as organic components, were embedded in organic-inorganic nanocomposites as a biosensor signal label, integrating the functions of signal amplification and target recognition. The biosensing mechanism is based on the fact that ALDH can catalyze rapid oxidation of acetaldehyde into acetic acid, resulting in pH change with portable pH meter readout. The altered pH exhibited a linear relationship with the logarithm of S. typhimurium from 102 to 108 CFU/mL and detection limit of 46 CFU/mL. Thus, the proposed biosensor has potential application in the diagnosis of pathogenic bacteria.

In ionexchange chromatography, the pH gradient mode becomes more and more popular today for the analysis of therapeutic proteins as this mode can provide higher or alternative selectivity to the commonly used salt gradient mode. Ideally, a linear pH response is expected when performing linear gradients. However up to now, only a very few buffer systems have been developed and are commercially available which can perform nearly linear pH responses when flowing through a given column. It is also known that a selected buffer system (mobile phase) can work well on one column but can fail on other column. The goal of this study was to practically evaluate the effects that ionexchange columns (weak and strong exchangers) might have on effluent pH, when performing linear pH gradient separations of therapeutic monoclonal antibodies. To attain this objective, the pH was monitored on-line at the column outlet using a specific setup. To make comprehensive observations of the phenomenon, four different mobile phase conditions and five cation exchange columns (weak and strong exchangers) were employed. The obtained pH responses were systematically compared to responses measured in the absence of the columns. From this work, it has become clear that both the column and mobile phase can have significant effects on pH gradient chromatography and that their combination must be considered when developing a new method. Phase systems (column + mobile phase) providing linear pH responses are indeed the most suitable for separating mAbs with different isoelectric points and, with them, it is possible to elute mAbs across wide retention time ranges and with high selectivity.

Monitoring the spore life cycle is one of the main issues in several fields including environmental control, sustainable ecosystems, food security, and healthcare systems. In this framework, the study of the living organism resistance to extreme conditions like those mimicking space environments is particularly interesting. The assessment of the local change of the pH level can be extremely useful for this purpose. An optical physiometer method based on the Raman response of the graphene, which is able to locally sense pH of a fluid on a micrometric scale, has been recently proposed. Due to the presence of π -bonds at the surface, the electronic doping of graphene is determined by the external conditions and can be electrochemically controlled or altered by the contact with an acid or alkaline fluid. The doping level affects the vibrational energies of the graphene that can be monitored by conventional Raman spectroscopy. In addition, Surface-Enhanced Raman Spectroscopy (SERS) can give direct information on the biochemical changes occurring in spore components. In this work, we propose the joint use of Graphene-Based Raman Spectroscopy (GbRS) and SERS for the monitoring of the response of spores to exposure to low temperatures down to 100 K. The spores of the thermophilic bacterium Parageobacillus thermantarcticus isolated from an active volcano of Antarctica (Mt. Melbourne) were investigated. These spores are particularly resistant to several stressing stimuli and able to adapt to extreme conditions like low temperatures, UV irradiation, and γ -rays exposure. The results obtained showed that the joint use of GbRS and SERS represents a valuable tool for monitoring the physio-chemical response of bacterial spores upon exposure to stressing stimuli.

OBJECTIVES: Urinary pH is a decisive factor in several pathologies, there by an informative marker employed in treatment decisions. Although extensively used, the urinary pH dipstick test may not be sufficiently accurate or precise for clinical decisions and more robust methodologies need to be considered. In this study, we compare pH measurements when using aportable medical device and different dipstick tests. MATERIALS AND METHODS: Four pH dipstick brands and a Lit-Control® pH Meter were tested using commercial buffer solutions with seven distinct pH values representing the physiological range in urine (4.66; 5.0;5.5; 6.0; 7.0; 7.5; 8.0). A statistical analysis was performed to assess the correlation of measured versus real values, together with validity measures as resolution,precision and accuracy. RESULTS: Validity measures stated the superiority of the portable pH meter, with a reduced dispersion of data and more exact values. Additionally, correlation analysis demonstrate that the pH values obtained with the pH meter were the closest to the buffers' real pH values. CONCLUSION: The detailed comparative study presented here reveal the superiority of a portable pH meter to several of the most used dipstick brands in the clinic.Lit-Control® pH Meter represents a reliable alternative when a monitoring of urinary pH is needed, as may happen during the screening of diseases or treatment monitoringin the clinic, as well as during the self-monitoringby the patient under professional supervision at home.


OBJETIVOS: El pH urinario es un factor decisivo en varias patologías, en consecuencia, un marcador informativo empleado en las decisiones de tratamiento. Aunque ampliamente utilizada, la tira reactiva puede no ser lo suficientemente acurada o precisa para la toma de decisiones clínicas y otras metodologías más robustas deben ser consideradas. En este estudio,comparamos las mediciones de pH cuando usamos un dispositivo médico portátil y diferentes tiras reactivas. MATERIALES Y MÉTODOS: Se probaron cuatro marcas de tiras reactives de pH y un medidor de pH, Lit-Control® pH Meter, utilizando soluciones tampón comerciales con siete valores de pH distintos que representan el rango fisiológico de pH en orina (4.66; 5.0; 5.5; 6.0;7.0; 7.5; 8.0). Se realizó un análisis estadístico para evaluar la correlación de los valores medidos versus los reales, junto con las medidas de validez como resolución,precisión y exactitud. RESULTADOS: Las medidas de validez indicaron la superioridad del medidor de pH portátil, con una reducción de la dispersión de los datos y valores más exactos. Además, el análisis de correlación demuestra que los valores de pH obtenidos con el medidor de pH fueron los más cercanos a los valores de pH reales de los tampones.CONCLUSIÓN: El estudio comparativo detallado presentado aquí revela la superioridad de un medidor portátil de pH frente algunas de las marcas comerciales de tiras reactivas más ampliamente usadas en las práctica clínica. El Lit-Control® pH Meter representa una alternativa fiable cuando se necesita un monitoreo del pH urinario, como puede suceder durante la detección de enfermedades o la monitorización del tratamiento a nivel clínico, así como durante el autocontrol domiciliario del paciente, bajo supervisión profesional.

This study developed a portable and sensitive biosensor for the detection of telomerase activity extracted from HeLa cells by monitoring urease regulated pH change. In the presence of telomerase, the telomerase substrate (TS) primer was elongated by adding telomeric repeats (TTAGGG)n firstly. The TS primer was then immobilized onto streptavidin magnesphere paramagnetic particles (PMPs). The elongated repeat unit can hybridize with biotinylated complementary DNA (cDNA) to specifically capture biotinylated urease onto PMPs. After magnetic separation, the PMPs complex was mixed with urea solution. As urease can catalyze the hydrolysis of urea into ammonia and induce pH increasing of the solution, the activity of telomerase can be detected by pH-sensitive dyes or by a portable pH meter easily. The changed pH has a linear relationship with the logarithm of the number of HeLa cells from 50 to 10,000 and with a detection limit of ~20 HeLa cells, which indicated the proposed method have potential application in telomerase-related cancer diagnosis.

BACKGROUND: Urine pH is an integral part of a complete urinalysis, and is commonly measured in veterinary practice using semiquantitative reagent strips. OBJECTIVE: The aim of this study was to compare the urine pH of dogs and sheep, using visual interpretation of dipstick reactions, and using a pH-meter as the reference method. Agreement between the 2 methods was also assessed. An additional objective was to compare the urine pH before and after centrifugation. METHODS: A total of 50 voided urine samples from sheep and 52 from dogs were collected into sterile containers. For pH measurements, 2 methods were used, a pH-meter and urine dipstick reagent pads. Measurements were performed using urine samples before (whole urine) and after centrifugation (urine supernatant). For comparison of the 2 methods, Passing and Bablok regression analysis and Bland-Altman plots were used. RESULTS: The equation created to assess agreement between the 2 methods in dogs showed a constant bias at -0.14 and a positive proportional bias at 0.98. From a clinical standpoint, total bias was below and above the maximum acceptable bias in sheep and dogs, respectively. Clinically acceptable bias was also found using centrifuged urine samples in sheep, but the urine pH values before and after centrifugation were nearly identical in dogs. CONCLUSION: Urine dipstick reagent pads and pH-meters can be used interchangeably to determine urine pH in sheep without needing centrifugation. In contrast, pH-meters provide more accurate pH measurements than urine dipstick pads in canine urine, which is not improved by centrifugation.

Aflatoxin B1 (AFB1) can cause great threat to human health, so the development of convenient and portable device for sensitive detection of AFB1 is highly desired. The portable pH meter has the characters of facile operation, low cost, and easy availability. Therefore, in this study, we investigate the applicability of utilizing a pH meter as the readout to develop a portable sensor for AFB1. The specific detection of AFB1 is realized via the combination of AFB1-responsive aptamer-cross-linked hydrogel. Upon the addition of AFB1, AFB1 binds to its aptamer with high affinity in lieu of aptamer/DNA complex, causing the collapse of hydrogel network and results in the releasing of urease into the solution. The released urease can catalyse the hydrolysis of urea and result in the rise of pH value. The change of pH value has a direct relationship to the concentration of AFB1 in the range of 0.2-20µM with a detection limit of 0.1µM (S/N = 3). The proposed portable device is successfully applied to assay AFB1 in the food samples with satisfied results.

OBJECTIVE: Recurrent and chronic course of acne vulgaris, despite effect-proven therapies, point to an underfocused aspect in its pathogenesis and management. This study aims to assess in subjects with and without acne, the skin surface pH, a parameter that cumulatively represents functioning of various units of skin, including the barrier. METHODS: A total of 200 patients with acne and 200 age- and sex-matched controls were included. Under basal conditions, facial skin pH was derived from five sites using a skin pH-meter. The relation between skin pH and acne was evaluated according to sex. RESULTS: There were more subjects with normal skin pH in the control group compared to the case group, and the majority of acne occurrences in the case group were related to high skin pH (p=0.000). Mean pH among cases was higher than normal reference value (pH 4.5-5.5 for women, 4-5.5 for men) and that of controls p (<0.001). No significant association was observed between sex and skin pH in either cases or controls (p>0.05). CONCLUSION: Increased facial skin pH in patients with acne at basal conditions mirrors a chronic state of stratum corneum instability, which could be predisposing individuals to acne occurrence and/or recurrences. It could possibly be a common domain via which the classical pathomechanisms might be acting in acne. Integrating measures that maintain stratum corneum pH during therapy might prove worthwhile.

Vascular endothelial growth factor (VEGF) is a crucial signaling protein for the tumor growth and metastasis, which is also acted as the biomarkers for various diseases. In this research, we fabricate an aptamer-antibody sensor for point-of-care test of VEGF. Firstly, target VEGF is captured by antibody immobilized on the microplate, and then binds with aptamer to form the sandwich structure. Next, with the assist of glucose oxidase (GOx)-functionalized ssDNAs, hybridization chain reaction occurs using the aptamer as the primer. Thus, GOx are greatly gathered on the microplate, which catalyzes the oxidization of glucose, leading to the pH change. As a result, the detect limit at a signal-to-noise was estimated to be 0.5pg/mL of target by pH meter, and 1.6pg/mL of VEGF was able to be distinguished by naked eyes. Meanwhile, this method has been used assay VEGF in the serum with the satisfactory results.

BACKGROUND: The intensive care department of the institution use drug solutions within higher concentration to avoid fluid overload. The purpose of the study is to prove the physical stability of different injectable drugs within high concentration (amiodarone 25mg/mL, isosorbide 0.60mg/mL, lorazepam 0.16mg/mL, noradrenalin 0.120 and 0.240mg/mL, salbutamol 0.06mg/mL and sodium valproate 12mg/mL) to ensure the patients safety. METHODS: Five of 30 or 50mL polypropylene syringes were prepared for each solution under aseptic conditions and stored at room temperature. Immediately after the preparation (hour 0) and after 1, 4, 8, 24 and 48hours, 2mL of each solution were withdrawn from each syringe and placed in glass tubes to proceed to the stability test. All specimens were visually inspected in front of a black and of a white background and aliquots of each solution were centrifuged to proceed to microscopic inspection with a ten-fold magnification. The pH of each solution was measured with glass electrode pH-meter (Inolab level 1, WTW Weilhem, Germany with biotrode electrode, Hamilton, Bonaduz, Switzerland) and spectrophotometric measurements (Genesys 10 series, New-York, USA) were performed at three wavelengths (350, 410 and 550nm) to avoid the apparition of turbidity. RESULTS: For all the drugs included in the study, there was no significant change in pH, no color change, no turbidity or opacity and no precipitation observed in the solutions during the storage at room temperature for 48hours. No microaggregates were detected by microscope neither revealed by a change of absorbance. CONCLUSION: Within these limits, the preparations of amiodarone in 5% glucose polypropylene syringes and isosorbide, lorazepam, noradrenalin, salbutamol, valproate in 0.9% sodium chloride polypropylene syringes are physically stable at room temperature for 48hours. These results allow us to consider a study of chemical stability by high-performance liquid chromatography (HPLC).

AIM: If a relation exists between salivary IPHA, buffer capacity and caries experience, then this relationship could be used as screening chair side test for caries risk assessment. STUDY DESIGN: One hundred ninety seven children aged 4 to 6 years were examined. Data was collected by interview and clinical examination. They were divided into low, moderate and high caries experience group of 20 children each. Two ml of each sample was used to measure the pH value with pH meter. Regarding the buffering capacity, freshly prepared hydrochloric acid (HCl) was titrated into saliva and pH was recorded. The collected saliva samples were sent to Laboratory for measurement of calcium and phosphorus. IPHA was calculated and the negative logarithms of IPHA were used to determine the enamel solubility. The correlation between salivary IPHA, buffering capacity and caries experience were evaluated. RESULTS: There was a significant relation between pH, log IPHA and dental caries experience, it could be considered as a predictor of dental caries. CONCLUSION: pH measurement after HCl titration in saliva could be used as chair side screening test for the assessment of caries risk.

BACKGROUND: The clinical symptoms of facial and truncal acne differ. Skin surface acidity (pH), which is affected by sebum secretions, reflects the different clinical characteristics of the face and trunk. However, no studies have been conducted on truncal sebum production and skin pH. OBJECTIVE: We evaluated the differences and relationship between pH values of the face and trunk. We also evaluated the relationship between pH and the quantity of sebum produced in the trunk. METHODS: A total of 35 female patients clinically diagnosed with truncal acne were included. We measured pH on the face and truncal area using the Skin-pH-Meter PH 905®. We measured truncal sebum secretions using the Sebumeter SM 815®. Statistical analysis was performed to evaluate the correlations and differences between pH and sebum. RESULTS: Facial pH was significantly higher than chest and back pH values. The correlation between pH on the trunk and the face was significant. We used linear regression equations to estimate truncal pH using only measured pH from the chin. There was no significant relationship between truncal sebum secretion and pH. CONCLUSION: This was the first study that evaluated the differences and correlations between facial and truncal pH. We found that facial pH can predict truncal pH. In addition, we conclude that differences in pH and sebum secretion between the face and trunk are one of the reasons for differences in acne symptom at those sites.
