RESUMO

Sugarcane straw burning generates particulate matter with complex composition resulting in atmosphere pollution. Sugarcane straw sugarcane burning particles (PSSB) contain several chemical compounds that were previously identified to be associated with carcinogenic and mutagenic processes. The aim of the present study was to extract PSSB under lab conditions and subsequently determine phyto- and cytogenotoxicity of these particles using Lactuca sativa L. and Allium cepa L. bioassays. Seeds of lettuce var. Cinderela and onion cv. Vale-Ouro IPA-11 were germinated in Petri dishes containing different concentrations of PSSB at 25, 50, 100, 200 or 300 mg/ml as well as control for 72 hr. Seed germination of lettuce was inhibited by PSSB, in a concentration-dependent manner, accompanied by decreased root growth, suggesting phytotoxic effects. Further, reduction of mitotic index and high number of chromosomal alterations in onion of meristematics cells indicated a cytogenotoxic action attributed to PSSB. Although the chemical composition of PSSB in question has not been determined, the phyto- and cytogenotoxic effects may be linked to the possible presence of polycyclic aromatic hydrocarbons (PAHs), which are were identified as the main constituents of particulate matter resulting from burning of sugarcane straw, in addition to exerting adverse biological effects that might result in mutations and cancer. Data demonstrated that the use of plants bioassays might be an important tool for biomonitoring air quality.

Sewage sludges are a rich underused source of phosphorus (P) which contributes to environmental degradation, yet if recaptured, could return significant amounts of P to agricultural systems. Hydrothermal carbonisation (HTC) can efficiently recover P, with the added ability to transform P species into potentially more desirable forms for direct application to crops. P dynamics in hydrochars have primarily examined P speciation and chemical extractability as indicators of P bioavailability, but few studies directly evaluate the agronomic effectiveness of hydrochars as P fertilisers. As such, there is a clear need to assess the suitability of hydrochar as a source of bioavailable P in plant systems and the influence of HTC synthesis conditions. Response Surface Modelling of HTC synthesis conditions (pH, temperature and time), revealed initial pH significantly influence P distribution. Mild conditions of 180 °C for 30 min at pH 8.0 maximised P recovery (99%) along with carbon (62%) and nitrogen (43%) in hydrochars. Systematic characterisation of hydrochar P by chemical extraction and P L2,3-edge X-ray absorption near edge spectroscopy revealed H2O, NaHCO3 and NaOH- P fractions were significantly (p < 0.05) reduced in all hydrochars, while HCl-P fraction increased with HTC temperatures at pH 7. In contrast, P L2,3-edge XANES spectra were remarkably similar in raw sludges and corresponding hydrochars, regardless of HTC temperature or pH, revealing P was predominantly present as ferric phosphate with some hydroxyapatite. Multiple linear regression modelling suggested a significant relationship between chemical extractability and P bioavailability to wheat present in the raw sludges and hydrochars. This research provides further insight into the potential to use hydrothermal treatment for recovery and agricultural reuse of P, the importance of operational conditions on P transformation and the relationship between P speciation and bioavailability. The value of sewage sludge in a more sustainable global P cycle is also highlighted.

In this work the toxicity caused by explosive industries effluent (yellow water) at different levels of toxicity (genetic, cellular and organismal level) was evaluated by the Allium cepa test and the Sorghum sudanense germination. The results showed that the effluent paralyze the mitotic process, keeping the cells in the interphase, decreasing the mitotic index in A. cepa. Chromosomal abnormalities such as c-metaphases, adhesions, breaks, early ascending chromosomes and irregular nucleus were observed for this receptor species. The germination of S. sudanense was reduced, and the development of the radicles were affected, showing reduced tolerance index at the highest concentrations of the effluent. Thus, it is concluded that the effluent from the explosive industry is extremely toxic to the tested organisms, both in cellular and chromosomal level and also for seed germination.

The carbamazepine (CBZ) is one of the most frequently detected anticonvulsant drugs in water bodies. Although there are reports of its ecotoxicological effects in the scientific literature, toxicity studies have not focused on establishing the mechanism by which CBZ produces its effect at environmentally relevant concentrations. The objective of this work was to evaluate cyto-genotoxicity and its relationship with oxidative stress produced by carbamazepine in the Allium cepa model. The cytotoxicity and genotoxicity, as well as the biomarkers of oxidative stress were analyzed in the roots of A. cepa, exposed to 1 and 31.36 µg L-1 after 2, 6, 12, 24, 48 and 72 h. The results show that genotoxic capacity of this drug in the roots of A. cepa is related to the generation of oxidative stress, in particular with production of hydroperoxides and oxidized proteins. Also, the cytotoxic effect has a high correlation with DNA damage. The results of the present study clearly indicate that bioassays with sensitive plants such as A. cepa are useful and complementary tools to evaluate the environmental impact of emerging contaminants.

The operation of the Mining-and-Chemical Combine (MCC), the largest producer of weapons-grade plutonium in Russia, has resulted in radioactive contamination of the Yenisei River floodplain. Investigations carried out in Novosibirsk and Krasnoyarsk institutes have shown that the floodplain of the Yenisei downstream of the MCC is contaminated by radioactive particles (RP) of various types and activities. Analytical characterization of the RP showed that most of them were fuel particles, which were carried into the Yenisei after incidents at the MCC reactors. The plutonium and caesium isotope ratios (238Pu/239,240Pu; 137Cs/134Cs) vary substantially between the particles, indicating different source terms and time intervals when the RP were formed. In addition to fuel RP, there were particles that contained activation radionuclides. The experiment on dissolution of RP using the model solution (the simulated stomach fluid) showed different cumulative extractions of radionuclides from the particles: 60Co and 137Cs extractions were the lowest, the extracted fractions of europium and americium isotopes were the largest, and plutonium occupied an intermediate position. High concentrations of radionuclides in RP are sources of exposure of organisms in aquatic and terrestrial ecosystems to low radiation doses. The plant bioassays of the effects of γ-radiation from RP showed the effect of low doses of γ-radiation on growth parameters of aquatic plant Elodea canadensis growing in the Yenisei River floodplain. The presence of RP from different sources in the Yenisei River floodplain makes this region a unique site for studying environmental effects of the particles.

Stigmasterol, bergapten, and α-tomatine were isolated from tomato roots. The preliminary phytotoxic activities of stigmasterol and α-tomatine were evaluated in a wheat-coleoptile bioassay, and α-tomatine was more active than stigmasterol. To confirm its phytotoxic activity, α-tomatine was tested on Lactuca sativa and two weeds ( Lolium perenne and Echinochloa crus-galli), and it was active in all cases. The stimulatory activities of α-tomatine and stigmasterol on parasitic-plant germination were also evaluated, and α-tomatine was found to be active on Phelipanche ramosa, a parasitic plant of tomato. α-Tomatine was identified in root exudates by LC-MS/MS. This confirms that α-tomatine is exuded by roots into the environment, where it could act as both an allelochemical and a stimulator of P. ramosa, a parasitic plant of tomato.

The aim of this study was to assess the toxicological potential of binary drug mixtures and individual drugs under different pH conditions with different inorganic ion additions on the germination of Sorghum bicolor (sorgo) seeds. To assess whether the given drug mixtures were more phytotoxic than the individual compounds, concentration addition (CA) and independent action (IA) approaches were applied to estimate the predicted phytotoxicity values, followed by calculations of the MDR (model deviation factor) to determine the deviation from the predicted values. Binary mixtures of chloramphenicol with ketoprofen, diclofenac sodium, and oxytetracycline hydrochloride proved to be synergistic. The pH changes had the largest impact on the phytotoxicity of the diclofenac sodium and ketoprofen solutions, elevating their toxicity toward S. bicolor, and the co-presence of inorganic ions was shown to have an impact on ketoprofen, chloramphenicol, and oxytetracycline hydrochloride. Most of the interactions between sorgo plants and pharmaceuticals with added ions were antagonistic in nature, particularly those calculated using the IA model, with a few cases (one case for ketoprofen and chloramphenicol, two cases for oxytetracycline hydrochloride, and four cases for diclofenac sodium) of overestimation.

A series of phenyl analogues of brassinosteroids was prepared via alkene cross-metathesis using commercially available styrenes and 24-nor-5α-chola-2,22-dien-6-one. All derivatives were successfully docked into the active site of BRI1 using AutoDock Vina. Plant growth promoting activity was measured using the pea inhibition biotest and Arabidopsis root sensitivity assay and then was compared with naturally occuring brassinosteroids. Differences in the production of plant hormone ethylene were also observed in etiolated pea seedlings after treatment with the new and also five known brassinosteroid phenyl analogues. Antiproliferative activity was also studied using normal human fibroblast and human cancer cell lines.

AbstractLepidaploa rufogrisea is a promising candidate in the search of natural herbicides, with compounds (sesquiterpene lactones-glaucolide B) which have been reported to present phytotoxic potential against weeds. Thus, this work aimed to accomplish the phytochemical investigation of L. rufogrisea as well as the evaluation of its biological effects on the plant model Lactuca sativa. To this end, different extracts (aqueous, hydroalcoholic and hexane) of L. rufogrisea at different concentrations [0.25, 0.5, 1.0, 2.0 and 4.0 % (m/v)] were evaluated from macroscopic (percentage of germinated seeds, germination speed index and root length) and microscopic analysis (mitotic index and cell cycle alterations) in plantlets (150 per treatment) and meristematic cells (10 000 per treatment) of L. sativa var. Cristina. The extracts decreased the germination speed index and also the root growth of L. sativa plantlets. Cell proliferation was not significantly affected by the extracts. However, some chromosome and nuclear alterations in the mitotic spindle were observed, due to the interference of allelochemicals present in L. rufogrisea. Moreover, the plant model L. sativa demonstrated to be efficient for this kind of studies.


ResumenLepidaploa rufogrisea es una candidata prometedora en la búsqueda de herbicidas naturales, con compuestos (lactano sesquiterpeno- glaucolíde B) los cuales han sido reportados por presentar un potencial fitotóxico contra las malas hierbas. Por lo tanto, este trabajo tuvo como objetivo lograr la investigación fitoquímica de L. rufogrisea, así como también la evaluación de sus efectos biológicos sobre el modelo de planta Lactuca sativa L. Para esto, diferentes extractos (acuosos, hidroalcohólicos y hexanos) de L. rufogrisea fueron evaluados. El porcentaje de semillas germinadas, el índice de velocidad de germinación y la longitud de raíz de plántulas de L. sativa var. Cristina fueron evaluadas, así como también la citotoxicidad de los extractos en las células meristemáticas. Los extractos disminuyeron el índice de velocidad de germinación y también el crecimiento de las raíces de las plántulas de L. sativa. La proliferación celular no se vio significativamente afectada por los extractos. Sin embargo, se observaron alteraciones cromosómicas y nucleares, debido a la interferencia de los aleloquímicos presentes en L. rufogrisea en el huso mitótico. Además, el modelo de planta L. sativa demostró ser eficiente para tales estudios.

In the present study, leachate toxicity of a municipal solid waste landfill located in the Sinos River Valley region (southern Brazil) was evaluated using plant bioassays. Leachate toxicity was assessed by analysis of seed germination and root elongation of lettuce (Lactuca sativa L.) and rocket plant (Eruca sativa Mill.) and root elongation of onions (Allium cepa L.). Bioassays were performed by exposing the seeds of L. sativa and E. sativa and the roots of A. cepa to raw leachate, treated leachate (biological treatment) and negative control (tap water). The levels of metals detected in both samples of leachate were low, and raw leachate showed high values for ammoniacal nitrogen and total Kjeldahl nitrogen. There is a reduction in the values of several physicochemical parameters, which demonstrates the efficiency of the treatment. Both L. sativa and A. cepashowed a phytotoxic response to landfill leachate, showing reduced root elongation. However, the responses of these two plant species were different. Root elongation was significantly lower in A. cepa exposed to treated leachate, when compared to negative control, but did not show any difference when compared to raw leachate. In L. sativa, seeds exposed to the raw leachate showed significant reduction in root elongation, when compared to treated leachate and negative control. Seed germination showed no difference across the treatments. The results of the study show that plant species respond differently and that municipal solid waste landfill leachate show phytotoxicity, even after biological treatment.

No presente trabalho, a toxicidade do lixiviado (chorume) de um aterro sanitário municipal, localizado na região do Vale do Rio dos Sinos, foi avaliada utilizando bioensaios em plantas. A toxicidade do chorume foi analisada pelo teste de germinação e crescimento radicular de alface (Lactuca sativa L.) e rúcula (Eruca sativa Mill.) e pelo teste de crescimento radicular em cebola (Allium cepa L.). Os bioensaios foram realizados com a exposição de sementes de L. sativa e E. sativa e raízes de A. cepa a amostras de chorume bruto, tratado por processo biológico e controle negativo (água de abastecimento público). Os níveis de metais detectados nas amostras de chorume foram baixos, e o chorume bruto apresentou valores elevados de nitrogênio amoniacal e nitrogênio Kjeldahl total. Ocorreu redução nos valores de vários parâmetros físico-químicos, demonstrando a eficiência do tratamento biológico. Os resultados indicam fitotoxicidade do chorume em L. sativa e A. cepa, evidenciada pela redução do crescimento radicular. Entretanto, as respostas destas duas espécies diferiram. O crescimento radicular foi significativamente inferior em A. cepa exposta ao chorume tratado quando comparado ao controle negativo, mas não apresentou diferença quando comparado ao chorume bruto. Em L. sativa, a exposição das sementes ao chorume bruto causou redução no crescimento radicular quando comparado com o chorume tratado e controle negativo. Não foram observadas diferenças entre os tratamentos para o parâmetro germinação de sementes. Os resultados deste trabalho indicam que as espécies de plantas estudadas são diferentes em suas respostas e que o chorume apresenta toxicidade, mesmo após o tratamento biológico.

RESUMO

Thirteen monohydroxylated brassinosteroids analogues were synthesized and tested for their biological activity in plant and animal systems. The cytotoxic activity of the products was studied using human normal and cancer cell lines with 28-homocastasterone as positive control, their brassinolide type activity was established using the bean second-internode test with 24-epibrassinolide as standard.

The cytotoxicity effect of aqueous extracts of the lichens species Myelochroa lindmanii and Canoparmelia texana (Lecanorales, Parmeliaceae) were evaluated using meristematic cells of lettuce (Lactuca sativa) and maize (Zea mays). The seasonal effect was also evaluated. Extracts of M. lindmanii and C. texana inhibited root growth and/or percentage germination, possibly due to alterations in the cell cycle. The M. lindmanii extract showed anti-mitotic effects and blocked the cell cycle in metaphase so that c-mitosis and cells with chromosome duplication were produced. The C. texana extract appeared to hinder cell division, increasing the number of interphase cells. In addition, both extracts caused an increase in percentage of cell death. Clastogenic effects were also observed, such as sticky chromosomes, bridges, fragments and later segregation. Both lichen species are thus potential sources of biologically active substances with possible applications in biology, medicine and agronomy.
