RESUMO

CmNACP1 mRNA has been shown to move long distance through the phloem in Cucurbita maxima (pumpkin) and through a graft junction. Whereas the phloem transport of several different mRNAs has been documented in other systems as well, its function remains, for most of these RNAs, largely unknown. To gain insight into the possible role of these RNAs, we searched for the closest homologs of CmNACP1 in Arabidopsis, a model plant much more amenable for analysis. A phylogenetic approach using the predicted NAC domain indicated that ANAC059, ANAC092, ANAC079, ANAC100, ANAC046, and ANAC087 form a single clade with CmNACP1. In the present work, we analyzed the possible function of the ANAC087 gene in more detail. The promoter region of this gene directed expression in the vasculature, and also in trichomes, stem, apexes, and developing flowers which supports the notion that ANAC087 and CmNACP1 are orthologs. Overexpression of the ANAC087 gene induced increased branching in inflorescence stem, and also development of ectopic or aerial rosettes in T1 and T2 plants. Furthermore, overexpression of ANAC087 leads to accelerated leaf senescence in 44 days post-germination (dpg). Interestingly, a similar phenotype was observed in plants expressing the ANAC087 gene upstream region, also showing an increase in ANAC087 transcript levels. Finally, the results shown in this work indicate a role for ANAC087 in leaf senescence and also in rosette development.

RESUMO

Rosette-forming glioneuronal tumor (RGNT) is a rare and distinct primary nervous system tumor. The literature on this novel neoplasm is sparse and limited to mostly case reports. Reviews on the characteristics of this tumor are fewer and far between with the latest up to a decade old. We thus provide a comprehensive review of recent literature to characterize presenting symptoms, radiological evidence, treatment options, and prognosis of this novel neoplasm. A Medline search for case reports detailing primary rosette-forming glioneuronal tumors was performed. RGNTs are a benign tumor of indolent course with mixed glial and neurocytic components. There is a slight female predominance with mean age of presentation at 23.57 years. Headaches, visual disturbances, and nausea and vomiting are the most common symptoms. Most RGNTs have solid and cystic components, arising most frequently in the fourth ventricle or cerebellum. Management is usually through surgery with gross total resection (GTR) providing better prognosis.

RESUMO

Guzmania monostachia is an epiphytic heteroblastic bromeliad that exhibits rosette leaves forming water-holding tanks at maturity. Different portions along its leaf blades can display variable degrees of crassulacean acid metabolism (CAM) up-regulation under drought. Since abscisic acid (ABA) can act as an important long-distance signal, we conducted a joint investigation of ontogenetic and drought impacts on CAM intensity and ABA levels in different leaf groups within the G. monostachia rosette. For this, three groups of leaves were analysed according to their position within the mature-tank rosette (i.e., younger, intermediate, and older leaves) to characterize the general growth patterns and magnitude of drought-modulated CAM expression. CAM activity was evaluated by analysing key molecules in the biochemical machinery of this photosynthetic pathway, while endogenous ABA content was comparatively measured in different portions of each leaf group after seven days under well-watered (control) or drought treatment. The results revealed that G. monostachia shows more uniform morphological characteristics along the leaves when in the atmospheric stage. The drought treatment of mature-tank rosettes generally induced in older leaves a more severe water loss, followed by the lowest CAM activity and a higher increase in ABA levels, while younger leaves showed an opposite response. Therefore, leaf groups at distinct ontogenetic stages within the tank rosette of G. monostachia responded to drought with variable degrees of water loss and CAM expression. ABA seems to participate in this tissue-compartmented response as a long-distance signalling molecule, transmitting the drought-induced signals originated in older leaves towards the younger ones.

RESUMO

The melamine (M)/cyanuric acid (CA) supramolecular system is perhaps one of the most exploited in the field of self-assembly because of the high complementarity of the components. However, it is necessary to investigate further the factors involved in the assembly process. In this study, we analyzed a set of 13 M n /CA m clusters (with n , m = 1, 2, 3), taken from crystallographic data, to characterize the nature of the hydrogen bonds involved in the self-assembly of these components as well as to provide greater understanding of the phenomenon. The calculations were performed at the B3LYP/6-311++G(d,p) and ω-B97XD (single point) levels of theory, and the interactions were analyzed within the framework of the quantum theory of atoms in molecules and by means of molecular electrostatic potential maps. Our results show that the stablest structure is the rosette-type motif and the aggregation mechanism is governed by a combination of cooperative and anticooperative effects. Our topological results explain the polymorphism in the self-assembly of coadsorbed monolayers of M and CA. Graphical abstract The aggregation steps of the melamine-cyanuric co-crystal is driven by a hydrogen-bonded network which is governed by a complex combination of cooperative and anticooperative effects.

Introduction: Processing and presentation of antigens is involved in the phenomenon of multiple immunological synapses of human Macrophage-Lymphocyte Rosette (MLR) between monocyte-derived macrophages and CD4 + T cells of total leukocyte cultures autologous extracted from blood; here autologous apoptotic neutrophil antigens are presented by the endocytic pathway or via Class II. Class II Compartment (MIIC) has been characterized by B cells and dendritic cells in murine models. Objective: To study the ultrastructural evolution in the spatial organization MIIC in macrophages in the phenomenon of human MLR. Methods: healthy human blood samples were used, anticoagulated with heparin (n = 10) donated by Blood Bank, UNC, anonymous. Autologous leukocyte cultures in TC199 medium (SIGMA, St. Louis, MO). Cell culture samples were taken at 1, 2, 3, 20, 48 and 96 h. MLR technique was applied. The citopreparations underwent processing techniques for ultrastructural study with MET: Zeiss LEO-906E. Results: We observed multivesicular, multilamellar, and tubular bodies in the spatial organization of MIIC throughout the culture time. Tubular structures appeared at 48 h of culture. We conclude that spatial organization of MIIC takes various aspects coinciding with the occurrence of macrophage transformation in culture and its role as antigen presenting cell (APC) in MLRs. Given the autologous antigens presented we postulate that the profile of macrophages in MLRs could correspond to alternative or M2 (AU) .

RESUMO

Relocation of Espeletia grandiflora (Asteraceae) plants as a strategy for enrichment of disturbed paramo areas (PNN Chingaza, Colombia). Ecological restoration of the Andean paramos faces several ecological barriers mainly at the phase of dispersal and establishment of native species. With the aim to contribute to the enrichment of degraded areas, different strategies have to be developed to overcome those barriers. In this work we studied the response of individuals of Espeletia grandiflora (Asteraceae) to the relocation as a strategy for ecological restoration programs. We also evaluated the effect of size of relocated individuals on their survival and development. The work was carried out in an experimental plot at 3 424m altitude in the sector Lagunas de Siecha of Chingaza National Park, Colombia. We relocated 200 plants that belonged to three different size classes: 5, 10 and 15cm of initial height. The following variables were registered: survival, plant height, number of living leaves and stem diameter of each individual. We also evaluated the differences between individuals in survival and development. In terms of survival the most efficient size classes corresponded to 15cm high; the survival was 85% after two years. The relative growth rates for height and stem diameter decreases with the increase in size, but the absolute increase in height did not show significant differences between the three sizes tested. Since the stem diameter was found the strongest survival predictor after two years of relocation activities, we suggest its use as a criterion for selection of relocation individuals. The relocation of individuals of E. grandiflora had a positive side effect, carrying other species that may contribute to the enrichment and restoration of degraded areas. Among these, we found species of the genus Hypericum, as well as Arcytophyllum nitidum and Calamagrostis effusa, which should be evaluated in terms of survival and development for the subsequent implementation of the relocation strategy. In this study we verified the successful relocation of individuals of E. grandiflora as a strategy for enrichment of paramos, and provided values of survival and growth, which should be useful for planning and predicting with greater certainty the success of restoration programs in the paramo.
