RESUMO

Acadia National Park (ANP) is located on Mt. Desert Island, ME on the U.S. Atlantic coast. ANP is routinely a top-ten most popular National Park with over four million visits in 2022. The overall contribution and negative effects of long-range atmospheric transport and local sources of dioxin-like contaminants endangering natural and wildlife resources is unknown. Dioxin-like (DL) contaminants polychlorinated dibenzo-p-dioxins (∑PCDD) and polychlorinated dibenzofurans (∑PCDF), non-ortho coplanar PCBs (∑CP4), and polychlorinated naphthalenes (∑PCNs) were measured at the McFarland Hill air monitoring station (44.37°N, 68.26°W). On a mass/volume basis, total PCNs averaged 90.9 % (788 fg/m3) of DL contaminants measured annually, with 92.9 % of the collected total in the vapor-phase. Alternatively, total dioxin/furans (∑PCDD/Fs) represented 71.6 % of the total toxic equivalence (∑TEQ) (1.018 fg-TEQ/m3), with 69.7 % in the particulate-phase. Maximum concentrations measured for individual sampling events for ∑PCDD/F, ∑CP4, and ∑PCN were 159 (winter), 139 (summer), and 2100 (autumn), fg/m3 respectively. Whereas the maximum ∑TEQ concentrations for individual sampling events for ∑PCDD/F, ∑CP4, and ∑PCN were 2.8 (autumn), 0.38 (summer), and 0.71 (autumn), fg-TEQ/m3 respectively. Pearson correlations were calculated for ∑PCDD/Fs and ∑PCN particulate/vapor-phase air concentrations and PM2.5 wood smoke "indicator" species. The most significant correlations were observed in autumn for particulate-phase ∑PCDD/Fs suggesting a relationship between visitation-generated combustion sources (campfires and/or waste burning) or climate-change mediated forest fires. Significant Clausius-Clapeyron (C-C) correlations observed for particulate-phase ∑PCDDs (r2 = 0.567) as ambient temperatures decreased suggests a connection between localized domestic heating sources or visitor-based burning of wood/trash resources. Alternatively, highly significant C-C vapor-phase ∑CP4-PCBs correlations (r2 = 0.815) implies that the majority of ∑CP4-PCB loading to ANP is from long-range atmospheric transport processes. Based on these findings, Acadia National Park should be classified as a remote site with minor depositional impacts from ∑PCDD/Fs, ∑CP4-PCBs, and ∑PCN atmospheric transport or local diffuse sources.

Polycyclic aromatic hydrocarbons (PAHs) and dioxins are potential causes of multiple diseases by activating the aryl hydrocarbon receptor (AhR) pathway. Health risk assessment of chemicals primarily relies on the relative potency factor (RPF), although its accuracy may be limited when solely using EC50 values. The induction of cytochrome P4501A1 (CYP1A1) serves as a biomarker for AhR activation and is an integrator of dioxin-like toxicity. Here, we present a method for evaluating the risks associated with AhR activation using mathematical models of dose-CYP1A1 induction. The dose-effect curves for certain PAHs and dioxins, including Ant, BghiP, 1,2,3,4,7,8-HxCDD, and others, exhibited a non-classical S-shaped form. The toxic equivalent factor (TEF) profiles revealed a broad range of toxic equivalent factor values. The TEFs for PAHs ranged from approximately 0.01 to 6, with higher values being observed when the concentration was less than 10-10 M, with the exceptions of Ace, Phe, and BghiP. Most congeners of dioxins got the lowest TEF value at around 10-10 M, ranging from 0.04 to 1.00. The binding affinity of AhR to ligands did not display a strong correlation with the EC50 of CYP1A1 expression, suggesting that the AhR-mediated effects of PAHs and dioxins are not fixed but instead fluctuate with the dose. Air samples acquired from a parking area were used to compare the proficiency of RPF and our current approach. In the current method, naphthalene and chrysene were the primary contributors of PAHs to AhR-mediated risks in parking lots air samples, respectively. However, the contributions of naphthalene and chrysene could be disregarded in the RPF approach.

The Feng-Sang River is a metropolitan river in Kaohsiung City, Taiwan. In this study, Feng-Sang River sediments were analyzed to investigate the distributions and sources of polycyclic aromatic hydrocarbons (PAHs). The Sediment Quality Guidelines (SQGs), potentially carcinogenic PAHs (TEQcarc ), and toxic equivalence quotient (TEQ) were applied to evaluate influences of PAHs on ecosystems and microbial diversities. Results indicate that PAHs concentrations varied between seasons and locations. The concentrations of ∑16 PAHs ranged from 73.6 to 603.8 ng/kg in dry seasons and from 2.3 to 199.3 ng/kg in wet seasons. This could be because of the flushing effect during wet seasons, which caused the movement and dilution of the PAH-contaminated sediments. Diagnostic ratio analysis infers that high PAHs levels were generated by combustion processes and vehicle traffic, and results from multivariate descriptive statistical analysis also demonstrate that the vehicular traffic pollution could be the major emission source of PAHs contamination. Comparisons of PAHs with SQGs indicate that PAHs concentrations in sediment were below the effects range low (ERL) values, and thus, the immediate threat to organisms might not be significant. The diagnostic ratio analyses are effective methods for PAH source appointment. The metagenomic assay results imply that sediments contained essential microbial species with eminent diversity. The detected PAH-degrading bacteria (Desulfatiglans, Dechloromonas, Sphingomonas, Methylobacterium, Rhodobacter, Clostridium, and Exiguobacterium) played a key role in PAHs biotransformation, and Dechloromonas and Rhodobacter had a higher relative abundance. Results of microbial diversity analyses indicate that the contaminated environment induced the changes of governing microbial groups in sediments. PRACTITIONER POINTS: Diagnostic ratio analyses are effective methods for PAHs source appointment. Microbial composition in sediments are highly affected by anthropogenic pollution. Combustion and vehicle traffic contribute to urban river sediments pollution by PAHs. Dechloromonas and Rhodobacter are dominant PAHs-degrading bacteria in sediments.

Validated methodology for the simultaneous determination of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs), polychlorinated biphenyls (PCBs), polybrominated dibenzo-p-dioxins and dibenzofurans (PBDD/Fs) and polybrominated diphenyl ethers (PBDEs) in foods of animal origin is presented. Method performance indicators were equivalent or better than those required for the control of EU regulated (EU, 2017/644) PCDD/F and PCB congeners in these foods, and for risk assessment through dietary intake. The method uses a high (>90%) proportion of 13Carbon-labelled surrogates for internal standardisation combined with high resolution mass spectrometry that allow accurate quantitation, and this was confirmed by multiple successful participations in proficiency testing for PCDD/Fs, PCBs and PBDEs in food. The same validation and method performance requirements as used for PCDD/Fs were followed for PBDD/Fs. The analysis of a range of food samples (eggs, milk, fish, shellfish, pork, beef and poultry), showed the occurrence of all four classes of contaminants at varying concentration ranges. In general, PCBs were the most prominent contaminant, both, in terms of dioxin-like toxicity, as well as in the occurrence of non-dioxin-like congeners, an observation that concurs with those made in other studies on Italian foods. The levels of PCDD/F and PCB occurrence are consistent with a gradual decline in contamination as reported by some other similar studies. Although all the determined contaminants were detected in the sampled foods, there was poor correlation between the occurrences of the brominated and chlorinated contaminants, and between PBDEs and PBDD/Fs, but better associations were observed between the occurrences of the chlorinated contaminants.

Medium-flow atmospheric samplers were used to collect particulate (PM2.5) and gaseous samples from the indoor kitchen of each of 35 randomly selected rural houses in North China while a meal was being cooked. The concentrations of 16 polycyclic aromatic hydrocarbons (PAHs) and 9 nitro-PAHs (NPAHs) in the samples were quantified by high-performance liquid chromatography (HPLC). This study provided the real PAH and NPAH pollution characteristics and carcinogenic risk produced by cooking in rural indoor kitchens in North China. The mean PAH and NPAH concentrations in air in the indoor kitchens during cooking periods were 4049.1 and 1741.6 ng/m3, respectively. The PAH and NPAH concentrations were lower in the particulate phase than the gaseous phase. The mean PAH and NPAH concentrations were much higher for cooking using coal than for cooking using liquefied petroleum gas (LPG) or electricity. The PAH and NPAH benzo[a]pyrene toxic equivalent (TEQBaP) concentrations for cooking using coal were 1823.3 and 2760.9 ng/m3, respectively. Lower PAH and NPAH concentrations were found in kitchens with than without range hoods. Range hoods decreased the PAH and NPAH TEQBaP concentrations by 68.8% and 61.9%, respectively. Appropriate fuel and ventilation choice will improve air pollution in indoor kitchens during cooking. The results provide important evidence for changing cooking habits and developing policies for cooking in rural China.

Human exposure to polychlorinated dioxins and furans (PCDD/Fs) through the dietary pathway is widely recognised and regulations in some regions of the world help to limit food contamination. Similar information on the analogous polybrominated dioxins and furans (PBDD/Fs) is scarce, partly due to the higher threshold to analytical access and unavailability of some standard materials. The analytical methodology developed here determined twelve planar PBDD/F congeners using 13Carbon labelled PBDD/F surrogates and high resolution mass spectrometric detection, and was extensively validated prior to the analysis of a range of commonly consumed Italian foods. The methodology also allowed simultaneous determination of PCDD/Fs and polychlorinated biphenyls (PCBs). The results show that PBDD/Fs occurred in different foods over a range of concentrations from <0.001 pg/g to 4.58 pg/g in fish. The dietary exposure (upper bound) of different Italian population groups, resulting from these occurrence levels was estimated using the toxic equivalency (TEQ) approach that is commonly used for dioxin-like contaminants and ranged from 0.17 to 0.42 pg TEQ/kg bodyweight/day (lower bound - 0.01 pg TEQ/kg bodyweight/day) depending on the population subgroup. Although precautionary, upper bound values may provide a more realistic estimate of toxicity as not all congeners and foods were measured. As expected, children were more highly exposed than adults due to lower body weight. These exposure levels were between a quarter and a third of that arising from the sum of PCDD/Fs and PCBs (0.61 to 1.38 pg WHO-TEQ/kg bodyweight/day), but they contribute to dioxin-like toxicity. If this data is considered in view of the revised tolerable dioxin-like dietary intake published by EFSA in 2018, it is evident that the tolerable weekly intake of 2 pg/kg bodyweight/week would be exceeded by some of the assessed population sub-groups, or all sub-groups if the cumulative intake is considered.

Edible cod liver products including cod liver oil and canned cod liver, sampled over the last five decades from the North Atlantic region, including the Baltic Sea were analysed for a set of persistent and toxicologically significant polychlorinated naphthalene (PCN) congeners with some of the highest relative potencies (dioxin-like toxicity) among PCNs. The targeted congeners showed a near-universality of occurrence in all samples apart from the most recent sample of cod liver oil which was assumed to be highly purified, as cod livers from the same period and location showed appreciable amounts of PCNs. The majority of dominant congeners in legacy technical PCN mixtures were absent or occurred in low concentrations, raising the possibility that congeners arising from combustion related sources may be acquiring a greater significance following the decline and elimination of PCN production. The apparent appreciation in the relative amounts of PCN#70 in the last three to four decades may provide support for this view. The PCN contribution to dioxin-like toxic equivalence (TEQ) that was estimated for these samples (range 1.2-15.9 pg TEQ g-1) was significant in comparison to the EU regulated value of 1.75 pg TEQ g-1 for dioxins in fish oils. Most of the TEQ was associated with PCNs 66/67, 64/68, 69 and 73. Although metabolic processes are likely to influence this distribution, the profile is a little different to that observed in the tissues of higher order animals where PCNs #66/67 and #73 may contribute approximately 90% to the summed TEQ.

A retrospective analysis of a number of historical medicinal grade cod liver oil samples produced in Northern Europe revealed relatively high contamination levels of PCNs, PCDD/Fs and PCBs. The total toxic equivalence (TEQ) associated with PCDD/Fs, dl-PCBs and PCNs was in the range 95-427 pg g-1 for Baltic cod liver oils and from 70 to 148 pg g-1 for oils sourced from the North Atlantic. The corresponding range for canned cod liver products (Baltic Sea) sampled in 2017 ranged from 52 to 104 pg g-1 fat (33-34 pg g-1 ww). The contribution from PCBs to the overall TEQ toxicity was around 3-6-fold higher than from PCDD/Fs and ranged from 24 to 318 pg TEQ g-1ww. The estimated summed TEQ intakes of PCDD/Fs, dl-PCBs and dl-PCNs resulting from the consumption of the daily recommended doses was highest for the Baltic cod liver oils ranging from 16 to 293 pg kg-1 body mass (bm) day-1 for an adult, 20-183 pg kg-1 bm day-1 for a teenager and 15-131 pg kg-1 bm day-1 for a child. The contribution to daily adult TEQ intake from PCNs alone, although relatively small is estimated to contribute up to 5-fold above the recent EFSA proposed TWI of 2 pg kg-1 bm. The results indicate that although currently produced fish oils may undergo rigorous purification procedures and show low contaminant levels, cod livers sourced from the Baltic and consumed locally, continue to contribute substantially to the dietary intake of these contaminants.

The Love River and Ho-Jin River, two major urban rivers in Kaohsiung City, Taiwan, are moderately to heavily polluted because different types of improperly treated wastewaters are discharged into the rivers. In this study, sediment and river water samples were collected from two rivers to investigate the river water quality and accumulation of polycyclic aromatic hydrocarbons (PAHs) in sediments. The spatial distribution, composition, and source appointment of PAHs of the sediments were examined. The impacts of PAHs on ecological system were assessed using toxic equivalence quotient (TEQ) of potentially carcinogenic PAHs (TEQcarc) and sediment quality guidelines. The average PAHs concentrations ranged from 2161 ng/g in Love River sediment to 160 ng/g in Ho-Jin River sediment. This could be due to the fact that Love River Basin had much higher population density and pyrolytic activities. High-ring PAHs (4-6 rings) contributed to 59-90% of the total PAHs concentrations. Benzo(a)pyrene (BaP) had the highest toxic equivalence quotient (up to 188 ng TEQ/g). Moreover, the downstream sediments contained higher TEQ of total TPHs than midstream and upstream sediment samples. The PAHs were adsorbed onto the fine particles with high organic content. Results from diagnostic ratio analyses indicate that the PAHs in two urban river sediments might originate from oil/coal combustion, traffic-related emissions, and waste combustion (pyrogenic activities). Future pollution prevention and management should target the various industries, incinerators, and transportation emission in this region to reduce the PAHs pollution.

Polychlorinated naphthalenes (PCNs) have been found widely in the aquatic environment and can be transferred through food chains, which can magnify or dilute their toxic effects on humans. In this study, PCNs were analyzed in samples of 17 species of fish with different dietary habits collected in the Bohai coastal area in China. Dichloronaphthalenes, which have rarely been quantified in previous studies, were determined. The total PCN concentrations were from 7.3 to 214 pg/g wet weight, and the highest concentration was found in ditrema. The trichloronaphthalenes were the most abundant PCNs, followed by the dichloronaphthalenes and pentachloronaphthalenes. The relatively high contributions of the less-chlorinated homologs to the total PCN concentrations indicated that the main PCN sources around the Bohai were industrial thermal process emissions rather than technical PCN formulations. The trophic magnification factors of the PCN homologs were from 3.1 to 9.9, indicating that PCNs were biomagnified by fish. The trophic magnification factor of dichloronaphthalene and trichloronaphthalenes was 5.8 and 6.4, respectively, indicating for the first time that dichloronaphthalene and trichloronaphthalenes can undergo trophic magnification by fish. The two highest trophic magnification factors were for the pentachloronaphthalenes and hexachloronaphthalenes, probably because these PCNs having fewer vicinal carbon atoms without chlorine atoms attached are less easily biotransformed than the other homologs. The dioxin-like toxicities of the PCNs in the samples, expressed as potential toxic equivalences (TEQs), were assessed. The highest total TEQ was 0.0090 pg/g ww, in Pacific herring, and the hexachloronaphthalenes were the dominant contributors to the total TEQs in the fish samples. The PCN TEQs were much lower than the polychlorinated dibenzo-p-dioxin and dibenzofuran and dioxin-like polychlorinated biphenyl TEQs found in fish from the Bohai in previous studies, and made marginal contributions to overall human exposure to dioxin-like TEQs, suggesting that PCNs pose no toxicological concerns.

Polychlorinated naphthalenes (PCNs) are legacy contaminants that are listed by the Stockholm convention, initially for reduction of inadvertent production and ultimately, for elimination. They originate through releases from older electrical equipment, inadvertent contamination in industrial chemicals and from combustion processes such as incineration. Recent advances in measurement techniques have allowed a greater characterisation of PCN occurrence, yielding more specific data including individual PCN congener concentrations. Emerging data on food shows widespread occurrence in most commonly consumed foods from different parts of the world. Concurrently, toxicological studies have also allowed a greater insight into the potencies of some congeners, a number of which are known to elicit potent, aryl hydrocarbon receptor (AhR) mediated responses, often referred to as dioxin-like toxicity. The dietary pathway is widely recognised as the most likely route to non-occupational human exposure. This paper reviews some of the more recent findings on PCN occurrence in food, biota, and human tissues, and discusses the use of relative potencies to express PCN toxicity in foods.

All 209 congeners of polychlorinated biphenyls (PCBs) in edible fish tissue from the central Red Sea coast (Jeddah region) of Saudi Arabia were analyzed by isotope dilution high-resolution gas chromatography-mass spectrometry. The upper-bound total PCB (ΣPCB) levels in nine commonly consumed fish species from three areas were 0.2-82.5ng/g wet weight (17-8450ng/g lipid weight), which were at the lower end of reported global range and far below international tolerance limits (500-3000ng/g ww). Dioxin-like congeners contributed up to 12.8% (mean 6.5%) to ΣPCB in tissue samples, with the total PCB toxic equivalencies (TEQs) at a tolerable range (0.05-2.6pgTEQ/g ww or 2-238pgTEQ/g lw) for all species. PCB profiles were dominated by moderately chlorinated homologs, mainly hexachlorobiphenyls, but less chlorinated congeners were also consistently elevated, notably in Siganus rivulatus (Area III) and Mugil cephalus (Area I). It remains to be ascertained if the latter were breakdown products or due to fresh inputs. The top congeners based on dominance by both occurrence and abundance were identified as potential markers of ΣPCB in fish tissue, which can be used for future selective biomonitoring in case of reasonable constraints on full congener approach.

Toxic polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) may be formed during remediation of chlorinated phenols via Fenton oxidation. To highlight the need for monitoring the production of toxic byproducts in these reactions, this work assessed the influence of iron dose (0.09-0.36 mM) on the Fenton oxidation of 2-chlorophenol (2-CP, 15.56 mM), a potential precursor of PCDD/Fs, by quantifying 2-CP removal and mineralization rates as well as byproducts yields, including PCDD/Fs. Although the increase in the iron dose showed positive contribution to 2-CP oxidation, under the operating conditions of the current study (H2O2 at 20% of the stoichiometric dose and 20 °C), there was no effect on the mineralization rate, and TOC and chlorine balances were far to be closed, depicting the presence of chlorinated organic byproducts in the reaction medium. After 4 h of treatment, the total PCDD/Fs concentrations increased by 14.5-39 times related to the untreated sample when the iron doses tested decreased from 0.36 to 0.09 mM, with preferential formation of PCDFs over PCDDs and dominance of lower chlorinated congeners such as tetra and penta-PCDD/Fs. The treatment with the highest iron dose (0.36 mM) exhibited the lowest PCDD/Fs yields and was thus most successful at mitigating toxic byproducts of the Fenton oxidation, leading to lower sample toxic equivalence (TEQ) value.

This study reports the history of contamination of American eels (Anguilla rostrata) from eastern Lake Ontario (LO) by persistent organic pollutants (POPs). Three groups of 10 large female eels captured in eastern LO in each of 1988, 1998, and 2008 were analysed for polychlorinated dibenzo-p-dioxins and furans, polychlorinated biphenyls, several organochlorine pesticides, and polybrominated diphenyl ethers. Mean concentrations were up to 3-fold lower in 2008 compared to previous years. When combined with the results of previous studies, these data show that concentrations of POPs in American eels have declined exponentially since the early 1980s by an average of 9.1±1.9% per year. Toxic equivalent (TEQ) concentrations of dioxin-like compounds (DLCs) were calculated from fish toxic equivalency factors. Assuming an efficient transfer of DLCs to their eggs, egg TEQs prior to 2000 exceeded the threshold for chronic toxicity to embryos of European eels (Anguilla anguilla) and lake trout (Salvelinus namaycush) (4-5 pg/g ww of TEQ). These results suggest that embryotoxicity of maternally-derived DLCs from LO eels, historically a major contributor to the spawning stock of American eels, could have impaired the reproductive and recruitment success of the species.

Must molecular mechanisms be the same for concentration addition to predict the effect of a mixture or is it sufficient for compounds to affect similar pathways or common outcomes? Does concentration addition provide a closer approximation to observations than alternative models such as independent action? Suppose effects are mediated by hormone A through receptor R, synthesis of A is reduced by compound B, and that C is a competitive antagonist to A. Both B and C reduce levels of the receptor-ligand complex AR but via different specific mechanisms. Are combinations of B and C concentration additive? We used simple pharmacodynamic models, deriving mathematical models using equilibrium binding and mass balance. Assume A binds the receptor at one site of R with effects proportional to the concentration of AR. Let C act as a competitive antagonist via the Gaddum equation. Let B affect synthesis of A via a function g(B). We derive a model describing the joint response surface of B and C, and a function describing its isoboles. Under concentration addition, the isoboles must be negatively sloped straight lines. We show that linearity of the isoboles depends crucially on g(B). The mixture is concentration additive if g″(B), the second derivative of g(B) with respect to B, is always zero. Responses are greater than concentration additive if g″(B) is always positive and less than concentration additive if g″(B) is always negative. We describe functions g(B) that lead to all three cases as well as one that is greater than concentration additive in some regions and less than concentration additive in others. At least in this simple model, concentration addition cannot be assumed: mixtures of competitive antagonists and compounds that alter hormone synthesis can lead to results that are concentration additive, greater than concentration additive or less than concentration additive. Nevertheless, concentration addition appears to provide a closer approximation to the pharmacodynamic model examined here than independent action. Care needs to be taken in extrapolating to other situations, but analysis of simple pharmacodynamic models appears to be a useful strategy.
