BACKGROUND: Yam (Dioscorea alata L.) is the staple food of many populations in the intertropical zone, where it is grown. The lack of phenotyping methods for tuber quality has hindered the adoption of new genotypes from breeding programs. Recently, near-infrared spectroscopy (NIRS) has been used as a reliable tool to characterize the chemical composition of the yam tuber. However, it failed to predict the amylose content, although this trait is strongly involved in the quality of the product. RESULTS: This study used NIRS to predict the amylose content from 186 yam flour samples. Two calibration methods were developed and validated on an independent dataset: partial least squares (PLS) and convolutional neural networks (CNN). To evaluate final model performances, the coefficient of determination (R2 ), the root mean square error (RMSE), and the ratio of performance to deviation (RPD) were calculated using predictions on an independent validation dataset. The tested models showed contrasting performances (i.e., R2 of 0.72 and 0.89, RMSE of 1.33 and 0.81, RPD of 2.13 and 3.49 respectively, for the PLS and the CNN model). CONCLUSION: According to the quality standard for NIRS model prediction used in food science, the PLS method proved unsuccessful (RPD < 3 and R2 < 0.8) for predicting amylose content from yam flour but the CNN model proved to be reliable and efficient method. With the application of deep learning methods, this study established the proof of concept that amylose content, a key driver of yam textural quality and acceptance, can be predicted accurately using NIRS as a high throughput phenotyping method. © 2023 The Authors. RESUMO

The beneficial effect of compost and compost tea on plant growth and protection is mainly associated with the microbial diversity and the presence of bacteria with plant growth-promoting effect. PGPR are considered as eco-friendly bio-fertilizers that may reduce the use of chemical pesticides and fertilizers. Three composts (AT, A10, and A30) were previously prepared from industrial wastes (olive mill wastewater, olive pomace, coffee ground, and phosphogypsum). In the present study, we isolated three bacterial strains from the compost teas. The phylogenetic identification of these bacterial strains (B.AT, B.A10, and B.A30) showed that they correspond to Serratia liquefaciens (B.AT and B.A10) and Achromobacter spanius (B.A30) species. A further characterization of the PGPR traits of these bacteria showed that they produce siderophore, exopolysaccharides, and IAA. Their effect on potato plant growth, yields, and tuber quality was performed under field culture conditions. Results showed that these strains can be characterized as PGPR, the best effect on potato plant growth was observed with Serratia liquefaciens (B.AT), the best yield and tuber quality was observed with Serratia liquefaciens (B.A10) while bacterial treatment with Achromobacter spanius (B.A30) is a Cd-tolerant PGPR.

The goal of the research is to determine the effect of pretreated poultry manure and irrigation on the yield and quality of potatoes (Solanum tuberosum) grown in fields located on the southern steppe of the Republic of Bashkortostan. Field experiments were repeated every three years. During vegetation, potato leaves and tubers were studied at the full blossom, leaves' decay, and potato harvest. The results showed that applying pretreated poultry manure at the rate of 120 t/ha favored higher tuber weight both on the rainfed and irrigated plots (from 0.23 to 0.82 kg/plant and from 0.24 to 1.02 kg/plant, respectively). On the rainfed and irrigated plots where poultry mature was not applied (control fields) the tubers' weight ranged from 0.08 to 0.31 kg/plant and from 0.16 to 0.50 kg/plant, respectively. Upon application of 40 t/ha of manure, under irrigation, the highest marketable value of tubers was 78%. On the rainfed plots the same value was 72% when applying poultry manure at a dose of 120 t/ha. Soil moisture monitoring showed that the potatoes did not get adequate water during the growing season. When the soil moisture on irrigated plots was 70% less than its minimum water capacity, potato plantings were watered.

Salinity is one of the harsh environmental stresses that destructively impact potato growth and production, particularly in arid regions. Exogenously applied safe-efficient materials is a vital approach for ameliorating plant growth, productivity, and quality under salinity stress. This study aimed at investigating the impact of foliar spray using folic acid (FA), ascorbic acid (AA), and salicylic acid (SA) at different concentrations (100, 150, or 200 mg/L) on plant growth, physiochemical ingredients, antioxidant defense system, tuber yield, and quality of potato (Solanum tuberosum L cv. Spunta) grown in salt-affected soil (EC = 7.14 dS/m) during two growing seasons. The exogenously applied antioxidant materials (FA, AA, and SA) significantly enhanced growth attributes (plant height, shoot fresh and dry weight, and leaves area), photosynthetic pigments (chlorophyll a and b and carotenoids), gas exchange (net photosynthetic rate, Pn; transpiration rate, Tr; and stomatal conductance, gs), nutrient content (N, P, and K), K+/ Na+ ratio, nonenzymatic antioxidant compounds (proline and soluble sugar content), enzymatic antioxidants (catalase (CAT), peroxidase (POX), superoxide dismutase (SOD), and ascorbate peroxidase (APX)) tuber yield traits, and tuber quality (dry matter, protein, starch percentage, total carbohydrates, and sugars percentage) compared with untreated plants in both seasons. Otherwise, exogenous application significantly decreased Na+ and Cl- compared to the untreated control under salt stress conditions. Among the assessed treatments, the applied foliar of AA at a rate of 200 mg/L was more effective in promoting salt tolerance, which can be employed in reducing the losses caused by salinity stress in potato grown in salt-affected soils.

Potato breeding relies heavily on visual phenotypic scoring for clonal selection. Obtaining robust phenotypic data can be labor intensive and expensive, especially in the early cycles of a potato breeding program where the number of genotypes is very large. We have investigated the power of genomic estimated breeding values (GEBVs) for selection from a limited population size in potato breeding. We collected genotypic data from 669 tetraploid potato clones from all cycles of a potato breeding program, as well as phenotypic data for eight important breeding traits. The genotypes were partitioned into a training and a test population distinguished by cycle of selection in the breeding program. GEBVs for seven traits were predicted for individuals from the first stage of the breeding program (T1) which had not undergone any selection, or individuals selected at least once in the field (T2). An additional approach in which GEBVs were predicted within and across full-sib families from unselected material (T1) was tested for four breeding traits. GEBVs were obtained by using a Bayesian Ridge Regression model estimating single marker effects and phenotypic data from individuals at later stages of selection of the breeding program. Our results suggest that, for most traits included in this study, information from individuals from later stages of selection cannot be utilized to make selections based on GEBVs in earlier clonal generations. Predictions of GEBVs across full-sib families yielded similarly low prediction accuracies as across generations. The most promising approach for selection using GEBVs was found to be making predictions within full-sib families.

The limited availability of phosphorus (P) in soils causes a major constraint in the productivity of potatoes, which requires increased knowledge of plant adaptation responses in this condition. In this study, six potato cultivars, namely, Agria, Lady Claire, Milva, Lilly, Sieglinde, and Verdi, were assessed for their responses on plant growth, leaf physiology, P use efficiency (PUE), and tuber quality with three P levels (Plow, Pmed, and Phigh). The results reveal a significant variation in the cultivars in response to different P availabilities. P-efficient cultivars, Agria, Milva, and Lilly, possessed substantial plant biomass, tuber yield, and high P uptake efficiency (PUpE) under low P supply conditions. The P-inefficient cultivars, Lady Claire, Sieglinde, and Verdi, could not produce tubers under P deprivation conditions, as well as the ability to efficiently uptake P under low-level conditions, but they were efficient in P uptake under high soil P conditions. Improved PUpE is important for plant tolerance with limited P availability, which results in the efficient use of the applied P. At the leaf level, increased accumulations of nitrate, sulfate, sucrose, and proline are necessary for a plant to acclimate to P deficiency-induced stress and to mobilize leaf inorganic phosphate to increase internal PUE and photosynthesis. The reduction in plant biomass and tuber yield under P-deficient conditions could be caused by reduced CO2 assimilation. Furthermore, P deficiency significantly reduced tuber yield, dry matter, and starch concentration in Agria, Milva, and Lilly. However, contents of tuber protein, sugars, and minerals, as well as antioxidant capacity, were enhanced under these conditions in these cultivars. These results highlight the important traits contributing to potato plant tolerance under P-deficient conditions and indicate an opportunity to improve the P efficiency and tuber quality of potatoes under deficient conditions using more efficient cultivars. Future research to evaluate molecular mechanisms related to P and sucrose translocation, and minimize tuber yield reduction under limited P availability conditions is necessary.

Yam (Dioscorea spp.) is a nutritional and medicinal staple tuber crop grown in the tropics and sub-tropics. Among the food yam species, water yam (Dioscorea alata L.) is the most widely distributed and cultivated species worldwide. Tuber dry matter content (DMC) and oxidative browning (OxB) are important quality attributes that determine cultivar acceptability in water yam. This study used a single nucleotide polymorphism (SNP) assay from a diversity arrays technology (DArT) platform for a genome-wide association study (GWAS) of the two quality traits in a panel of 100 water yam clones grown in three environments. The marker-trait association analysis identified significant SNPs associated with tuber DMC on chromosomes 6 and 19 and with OxB on chromosome 5. The significant SNPs cumulatively explained 45.87 and 12.74% of the total phenotypic variation for the tuber DMC and OxB, respectively. Gene annotation for the significant SNP loci identified important genes associated in the process of the proteolytic modification of carbohydrates in the dry matter accumulation pathway as well as fatty acid ß-oxidation in peroxisome for enzymatic oxidation. Additional putative genes were also identified in the peak SNP sites for both tuber dry matter and enzymatic oxidation with unknown functions. The results of this study provide valuable insight for further dissection of the genetic architecture of tuber dry matter and enzymatic oxidation in water yam. They also highlight SNP variants and genes useful for genomics-informed selection decisions in the breeding process for improving food quality traits in water yam.

A diploid, potato mapping population consisting of 149 individuals was assessed in three consecutive years for important agronomic and quality traits: tuber shape, regularity of tuber shape, eye depth, mean tuber weight, and tuber flesh color. Analysis of variance showed that the genotype had the largest influence on the phenotypic scores but effect of the genotype × year interactions was also strong. Using this data and an existing genetic map, a quantitative trait loci (QTL) analysis was conducted. From four to seven QTL were detected for each trait except tuber flesh color, which was determined by a major QTL on chromosome III explaining 76.8% of the trait variance. Additionally, a minor QTL for flesh color was localized on chromosome II. For the other traits, significant QTL were detected: for tuber shape on chromosome X, for regularity of tuber shape on chromosome III, for eye depth on chromosome IV, and for tuber weight on chromosome I. Some detected QTL confirmed previous studies, but new ones were also identified.

BACKGROUND: Chloride sensitivity of the potato (Solanum tuberosum L.) cultivars Marabel and Désirée was investigated in two pot experiments (soil/sand mixture and hydroponics). It was tested whether there are differential effects of KCl and K2 SO4 application on tuber yield and tuber quality, and whether both potato cultivars differ in their chloride sensitivity. RESULTS: Tuber yield, dry matter percentage of the tubers, starch concentration and starch yield were not significantly affected by potassium source (K2 SO4 or KCl). After exposure to salt stress in hydroponics (100 mmol L-1 NaCl, 50 mmol L-1 Na2 SO4 , 50 mmol L-1 CaCl2 ) for 5 days, 3-week-old potato plants had significantly reduced shoot dry mass after NaCl and Na2 SO4 application. However, CaCl2 treatment did not significantly affect shoot growth, although the chloride concentration reached 65 to 74 mg Cl- mg-1 dry matter, similar to the NaCl treatment. In contrast, growth reductions were closely related to sodium concentrations, thus plants suffered sodium toxicity and not chloride toxicity. CONCLUSION: Both potato cultivars are chloride-resistant and can be fertilised with KCl instead of K2 SO4 without the risk of depression in tuber yield or tuber quality. The statement that potatoes are chloride-sensitive and that chloride has negative effects on yield performance needs reconsideration. © 2017 Society of Chemical Industry.

The aim of the study involved evaluating the chemical composition of tubers of five potato cultivars that were grown under the same cultural practices in soils with low, medium, and high availability of phosphorus. The experimental designs corresponded to a randomized block with four replicates. Tuber samples were analyzed in terms of moisture, ash, protein, lipid, total sugar, fiber, starch, and phosphorus contents. The results suggested that increased availability of phosphorus in soil allowed the production of tubers with higher dry matter content, lower total sugar content, and a higher percentage of starch and protein. Hence, the aforementioned parameters constitute important factors corresponding to the nutritional and industrial quality of potatoes. Increased phosphorus availability in soil can promote significant changes in the composition of potato tubers, and thereby in potential uses of tubers.

Neste trabalho, objetivou-se avaliar o efeito de doses de nitrogênio (N) sobre os teores de carboidratos (açúcares solúveis totais, açúcares redutores e amido), matéria seca e sólidos solúveis totais nos tubérculos de quatro cultivares de batata. Os tubérculos originaram-se de quatro experimentos, realizados simultaneamente no campo, no outono/inverno, na Universidade Federal de Viçosa. Em todos os experimentos, foi utilizado delineamento em blocos casualizados, com quatro repetições. Os tratamentos foram compostos de cinco doses de N (0, 50, 100, 200 e 300 kg ha-1) na forma de sulfato de amônio, aplicadas no plantio das cultivares Ágata, Asterix, Atlantic e Monalisa. Foram analisados os teores de matéria seca, açúcares solúveis totais, açúcares redutores, amido e sólidos solúveis totais nos tubérculos, após 30 dias de armazenamento a 5ºC a 80-85 por cento de umidade relativa. Em todas as cultivares, as doses de N apresentaram efeito quadrático sobre o teor de sólidos solúveis totais e não houve efeito sobre os teores de matéria seca e de amido nos tubérculos de batata. Dependendo da cultivar, as doses de N influenciam positivamente, ou não influenciam os teores de açúcares redutores e açúcares solúveis totais nos tubérculos de batata.


This study aimed to evaluate the effect of nitrogen (N) doses on carbohydrate (total soluble sugars, reducing sugars and starch), dry matter and total soluble solids contents in tuber of four potato cultivars. The tubers originated from four experiments, simultaneously conducted in the field, during fall/winter, at the Federal University of Viçosa. These experiments were set in randomized complete block and four repetitions and we evaluated the effects of five N rates (0, 50, 100, 200, and 300 kg ha-1) on Agata, Asterix, Atlantic and Monalisa cultivars. Dry matter, total soluble sugars, reducing sugars, starch, and total soluble solids contents of the tubers were analyzed after a 30-day storage at 5ºC. N rates presented quadratic effects on the total soluble solids and presented no effects on dry matter and starch contents for all cultivars. Depending on the cultivar, N rates had positive effects or did not affect reducing sugars and total soluble sugars contents in the tubers.

Indian potato (Solanum tuberosum L.) processing industry has emerged fast due to economic liberalization coupled with growing urbanization, expanding market options and development of indegenous processing varieties. India's first potato processing varieties 'Kufri Chipsona-1' and 'Kufri Chipsona-2' were developed in 1998, followed by an improved processing variety 'Kufri Chipsona-3' in 2005 for the Indian plains and first chipping variety 'Kufri Himsona' for the hills. These varieties have >21% tuber dry matter content, contain low reducing sugars (<0.1% on fresh wt) and are most suitable for producing chips, French fries and dehydrated products. The availability of these varieties and standardization of storage techniques for processing potatoes at 10-12°C with sprout suppressant isopropyl N-(3-chlorophenyl) carbamate have revolutionized the processing scenario within a short span of 10 years. Currently about 4% of total potato produce is being processed in organized and unorganized sector. Potato processing industry mainly comprises 4 segments: potato chips, French fries, potato flakes/powder and other processed products. However, potato chips still continue to be the most popular processed product. The major challenge facing the industries lies in arranging round the year supply of processing varieties at reasonable price for their uninterrupted operation, besides several others which have been discussed at length and addressed with concrete solutions.

A seleção precoce de clones que possuam níveis elevados de matéria seca e baixos teores de açúcares redutores é uma necessidade nos programas de melhoramento para a qualidade de processamento da batata (Solanum tuberosum L.) na forma de palitos fritos ou chips. A seleção precoce tornou-se possível com a utilização de marcadores genéticos, visto que permitem a identificação precisa de indivíduos superiores. Assim, procura-se cada vez mais encontrar marcadores capazes de caracterizar tais indivíduos e utilizá-los via seleção assistida. O objetivo deste trabalho foi avaliar a eficiência da seleção assistida, utilizando os marcadores identificados por ANDREU (2004) que estariam associados ao teor de matéria seca e açúcares redutores em tubérculos de batata. Clones provenientes de 20 famílias foram avaliados nas gerações de plântula (P), primeira geração clonal (C1) e segunda geração clonal (C2). As estimativas das correlações simples para os caracteres entre gerações foram significativas, porém, baixas, confirmando a inviabilidade de se efetuar a seleção precoce nas primeiras gerações com base apenas em informações fenotípicas. Os marcadores utilizados forneceram um total de 16 marcas. Pela regressão múltipla stepwise, apenas sete dessas marcas tiveram associação com os caracteres estudados. Além disso, nenhuma marca associada ao teor de matéria seca de tubérculos na geração C1 teve associação significativa na geração C2. Isso também foi observado com o teor de açúcares redutores, o que é um indicativo da interação QTLs x ambientes. A seleção assistida não se mostrou eficiente em relação à fenotípica em nenhum dos casos avaliados, portanto, não sendo útil em uma possível seleção precoce. Esses resultados indicam que tais marcadores não estão próximos aos genes controladores dos caracteres desejados, sendo necessária a identificação de novos marcadores mais associados que possibilitem maior eficiência da seleção assistida.


Early generation selection for clones with high content of tuber dry matter and low levels of reducing sugars is required for potato (Solanum tuberosum L.) processing. Selection of superior clones at early generations became possible with the deployment of genetic markers, and can precisely identify the superior individuals. Therefore, it is necessary to identify genetic markers closely linked to genes of interest to do assisted selection. The aim of this research was to evaluate the efficiency of marker assisted selection with genetic markers previously identified by ANDREU (2004), which are assumed to be associated with dry matter and reducing sugars content in potato tubers. Clones from 20 families were evaluated during the seedling generation (S), first clonal generation (C1) and second clonal generation (C2). The estimated coefficients of correlation for all traits among generations were significant, even though of low magnitude, confirming that selection at early generation based only on phenotypic traits is inviable. A total of sixteen bands were amplified using these markers. However, by multiple stepwise regression, only seven of these bands showed association with the evaluated traits. Moreover, no markers associated with dry matter and reducing sugars content in the C1 were significantly associated with these traits in the C2, suggesting the existence of QTLs x environment interactions. The marker assisted selection resulted less efficient than the phenotypic selection in all cases studied, and thus is not recommended for early generation selection of clones for the processing industry. These results suggest that the markers used are not closely linked to the genes controlling the traits most important for processing. Therefore, it is important to identify new markers closely linked with such traits of interest that could improve the efficiency of marker assisted selection.

Os tubérculos de batata destinados ao processamento devem apresentar coloração clara após a fritura, o que é condicionado pelos baixos teores de açúcares redutores, glicose e frutose. Para auxiliar os programas de melhoramento na seleção de clones com baixos teores destes açúcares, o objetivo deste trabalho foi adaptar e validar uma metodologia simples, econômica e eficiente para quantificar o teor de glicose em tubérculos de batata utilizando um kit laboratorial enzimático desenvolvido para uso em amostras de sangue e urina. Os procedimentos de validação seguiram as recomendações do INMETRO dispostas no documento DOQ-CGCRE-008 de 2003. O método mostrou-se linear entre 1,25 e 40µg de glicose presente na amostra, sendo que o limite de quantificação foi 0,319mg g-1 de massa fresca de batata. A faixa linear de trabalho do método foi desde o limite de quantificação até 1,32mg g-1 de massa fresca de batata. A recuperação média da extração foi de 99,0 por cento. O método proposto foi aplicado para determinar o teor de glicose em amostras de batata "Macaca", que variou de 1,4 a 2,9mg g-1 massa fresca de batata.


The processing industry requires potato tubers with low levels of reduced sugars, glucose and fructose, which is responsible for the whiteness of the chip color. The objective of this research was to optimize and validate a simple, inexpensive, and precise enzymatic method to quantify glucose in potato tubers. This method will be useful in breeding programs to select clones for processing purposes with low levels of glucose. The validation procedure followed the recommendations described in INMETRO document DOQ-CGCRE-008 in 2003. The method was linear between 1.25 to 40µg of glucose in the sample. The quantification limit was 0.319mg g-1 of potato fresh weight. The linear working interval of the method started at the quantification limit and ended at 1.32mg g-1 of potato fresh weight. The average recovery in the extraction was 99.0 percent. The proposed method was used to determine glucose level in samples of ‘Macaca’ tubers, which ranged from 1.4 to 2.9mg g-1 potato fresh weight.
