Objective: This study aimed to investigate the usefulness of endoscopic ultrasound-guided tissue acquisition (EUS-TA) for diagnosing focal liver lesions in patients with a history of multiple primary malignant neoplasms. Methods: Among patients who underwent EUS-TA for focal liver lesions between 2016 and 2022, those with a history of multiple malignant neoplasms were included. A histologically confirmed malignant tumor within the past 5 years before EUS-TA was defined as a history of malignant neoplasm. The primary outcomes were diagnostic ability and adverse events of EUS-TA. Results: This study included 16 patients (median age, 73 [33-90] years), the median tumor size was 32 (6-51) mm, 14 had a history of double malignant neoplasms, whereas two had triple malignant neoplasms. Malignant neoplasms were detected histologically or cytologically in all cases. Immunohistochemistry was performed in 75% (12/16), and the final diagnosis of EUS-TA was metastatic liver tumor in 12 patients, and primary malignant liver tumor in four patients. The primary site could be identified in 11 of 12 metastatic tumor cases. The diagnostic yield of EUS-TA was 100% (16/16) for differentiating benign and malignant tumors and 94% (15/16) for confirming the histological type including the primary site of metastatic lesions. No adverse events were associated with the procedure. Conclusion: EUS-TA is a useful diagnostic modality for focal liver lesions in patients with a history of multiple malignant neoplasms, allowing for the differential diagnosis of primary and metastatic tumors and identification of the primary site of metastatic lesions.

High rates of sexually transmitted infections and unplanned pregnancy continue to plague young adults in the USA with low condom use a contributing factor. To better understand condom acquisition, errors, and breakage among US cisgender college students, a survey was conducted across six structurally diverse institutions of higher education in 2019-2020 prior to the COVID-19 pandemic. Students who had used external condoms in the last year (N = 1584) were asked about specific on- and off-campus locations of condom acquisition and practices related to condom use. Findings indicate that students most frequently acquired condoms off-campus with location differences between genders and relationship status. Condom errors were common, with no consistent patterns related to gender, but unpartnered students were more likely than those in relationships to experience condom errors. Multivariate logistic regression indicated that relationship status, applying condom on wrong side, adding condom after sex started, removing condom during sex, condom slipping off, and problems with fit were predictors for condom breakage. The study results provide guidance for healthcare and sexuality education professionals working with college students to better address the differing needs of college students regarding condom acquisition and correct condom use.

Causation is a core feature of human cognition and language. How children learn about intricate causal meanings is yet unresolved. Here, we focus on how children learn verbs that express causation. Such verbs, known as lexical causatives (e.g., break and raise), lack explicit morphosyntactic markers indicating causation, thus requiring that the child generalizes the causal meaning from the context. The language addressed to children presumably plays a crucial role in this learning process. Hence, we tested whether adults adapt their use of lexical causatives to children when talking to them in day-to-day interactions. We analyzed naturalistic longitudinal data from 12 children in the Manchester corpus (spanning from 20 to 36 months of age). To detect semantic generalization, we employed a network approach with semantics learned from cross-situational contexts. Our results show an increasing trend in the expansion of causative semantics, observable in both child speech and child-directed speech. Adults consistently maintain somewhat more intricate causative semantic networks compared to children. However, both groups display evolving patterns. Around 28-30 months of age, children undergo a reduction in the degree of causative generalization, followed by a slightly time-lagged adjustment by adults in their speech directed to children. These findings substantiate adults' adaptation in child-directed speech, extending to semantics. They highlight child-directed speech as a highly adaptive and subconscious teaching tool that facilitates the dynamic processes of language acquisition.

Accurate segmentation of the pancreas in computed tomography (CT) holds paramount importance in diagnostics, surgical planning, and interventions. Recent studies have proposed supervised deep-learning models for segmentation, but their efficacy relies on the quality and quantity of the training data. Most of such works employed small-scale public datasets, without proving the efficacy of generalization to external datasets. This study explored the optimization of pancreas segmentation accuracy by pinpointing the ideal dataset size, understanding resource implications, examining manual refinement impact, and assessing the influence of anatomical subregions. We present the AIMS-1300 dataset encompassing 1,300 CT scans. Its manual annotation by medical experts required 938 h. A 2.5D UNet was implemented to assess the impact of training sample size on segmentation accuracy by partitioning the original AIMS-1300 dataset into 11 smaller subsets of progressively increasing numerosity. The findings revealed that training sets exceeding 440 CTs did not lead to better segmentation performance. In contrast, nnU-Net and UNet with Attention Gate reached a plateau for 585 CTs. Tests on generalization on the publicly available AMOS-CT dataset confirmed this outcome. As the size of the partition of the AIMS-1300 training set increases, the number of error slices decreases, reaching a minimum with 730 and 440 CTs, for AIMS-1300 and AMOS-CT datasets, respectively. Segmentation metrics on the AIMS-1300 and AMOS-CT datasets improved more on the head than the body and tail of the pancreas as the dataset size increased. By carefully considering the task and the characteristics of the available data, researchers can develop deep learning models without sacrificing performance even with limited data. This could accelerate developing and deploying artificial intelligence tools for pancreas surgery and other surgical data science applications.

Salmonella enterica is a globally disseminated pathogen that is the cause of over 100 million infections per year. The resulting diseases are dependent upon host susceptibility and the infecting serovar. As S. enterica serovar Typhimurium induces a typhoid-like disease in mice, this model has been used extensively to illuminate various aspects of Salmonella infection and host responses. Due to the severity of infection in this model, researchers often use strains of mice resistant to infection or attenuated Salmonella. Despite decades of research, many aspects of Salmonella infection and fundamental biology remain poorly understood. Here, we use a transposon insertion sequencing technique to interrogate the essential genomes of widely used isogenic wild-type and attenuated S. Typhimurium strains. We reveal differential essential pathways between strains in vitro and provide a direct link between iron starvation, DNA synthesis, and bacterial membrane integrity.IMPORTANCESalmonella enterica is an important clinical pathogen that causes a high number of deaths and is increasingly resistant to antibiotics. Importantly, S. enterica is used widely as a model to understand host responses to infection. Understanding how Salmonella survives in vivo is important for the design of new vaccines to combat this pathogen. Live attenuated vaccines have been used clinically for decades. A widely used mutation, aroA, is thought to attenuate Salmonella by restricting the ability of the bacterium to access particular amino acids. Here we show that this mutation limits the ability of Salmonella to acquire iron. These observations have implications for the interpretation of many previous studies and for the use of aroA in vaccine development.

The ecological environment of wetlands in semi-arid regions has deteriorated, and vegetation succession has accelerated due to climate warming-induced aridification and human interference. The nutrient acquisition strategies and biomass allocation patterns reflect plant growth strategies in response to environmental changes. However, the impact of nutrient acquisition strategies on biomass allocation in successional vegetation remains unclear. We investigated 87 plant communities from 13 wetland sites in the semi-arid upper Yellow River basin. These communities were divided into three successional sequences: the herbaceous community (HC), the herbaceous-shrub mixed community (HSC), and the shrub community (SC). The nutrient composition of stems and leaves, as well as the biomass distribution above and belowground, were investigated. Results revealed that aboveground biomass increased with succession while belowground biomass decreased. Specifically, SC exhibited the highest stem biomass of 1,194.53 g m-2, while HC had the highest belowground biomass of 2,054.37 g m-2. Additionally, significant positive correlations were observed between leaf and stem biomasses in both HC and SC. The nitrogen (N) and phosphorus (P) contents within aboveground parts displayed an evident upward trend along the succession sequence. The highest N and P contents were found in SC, followed by HSC, and the lowest in HC. Stem N was negatively correlated with stem, leaf, and belowground biomass but positively correlated with root-shoot ratio. Leaf P displayed positive correlations with aboveground biomass while showing negative correlations with belowground biomass and root-shoot ratio. The ratios of C:N, C:P, and N:P in stem and leaf exhibited positive correlations with belowground biomass. The random forest model further demonstrated that stem N and leaf P exerted significant effects on aboveground biomass, while leaf P, stem N and P, and leaf C:P ratio had significant effects on belowground components. Additionally, the root-shoot ratio was significantly influenced by leaf P, leaf C:P ratio, and stem N, P, and C:P ratio. Therefore, the aboveground and belowground biomasses exhibited asynchronism across successional sequences, while plant nutrient acquisition strategies, involving nutrient levels and stoichiometric ratios, determined the biomass allocation pattern. This study offers valuable insights for assessing vegetation adaptability and formulating restoration plans in the semi-arid upper Yellow River basin.

BACKGROUND AND AIMS: Understanding how maize roots proliferate in phosphorus (P)-rich soil patches is critical for improving nutrient acquisition and crop productivity. This study explores the mechanisms of root adaptation to heterogeneous P availability, focusing on sucrose metabolism and the role of local P signals. METHODS: A split-root system with chambers of differing Pi concentrations (0 and 500 µM) was used to examine maize root responses. Various physiological and biochemical parameters, including root growth, sucrose partitioning, enzyme activities, and gene expression, were measured to elucidate the underlying mechanisms. KEY RESULTS: Root proliferation, particularly of second-order lateral roots, was markedly enhanced in P-rich patches. Sucrose was preferentially allocated to the Pi-supplied side, as confirmed by Fourier-transform infrared (FTIR) microscopy. Sucrose content in these roots decreased, indicating active metabolism. Higher activities of cell-wall invertase and sucrose synthase were observed in the Pi-supplied roots, supporting enhanced carbohydrate utilization. CONCLUSIONS: Local P availability triggers significant adjustments in sucrose metabolism and allocation, enhancing the sink capacity of maize roots in P-rich patches. These changes facilitate efficient lateral root proliferation and Pi utilization, highlighting the critical role of local P signals in nutrient acquisition strategies. This research provides deeper insights into the adaptive responses of maize to heterogeneous P environments, offering potential strategies for improving crop nutrient efficiency.

The current study aimed to investigate whether the use of formal language (Modern Standard Arabic [MSA]) by young children in diglossic Arab communities offers diagnostic insights, especially for verbal autistic children and to further explore this phenomenon. We used a cohort study design, with 4-6-year-old fluent first language Arabic-speaking children attending Arabic Kindergartens in two representative Kuwait governates. Reported cases for MSA use were assessed via a computer-based structured language test and corroborated cases were further assessed for exposure to sources of MSA, verbal IQ, temperamental characteristics, and autism spectrum disorder (ASD). Four children from the same class without developmental difficulties were selected for each MSA user as control group. The frequency of MSA use among verbal pre-schoolers was 0.46%. Use of MSA did not correlate with parents' education, amount of exposure to MSA, verbal IQ, but with severity of ASD. Predicted probability of ASD in the presence of MSA was 0.86. Executive functions of ASD-MSA users were similar to those of the control group and significantly higher than unselected autistic peers in the literature. The use of MSA has the potential to serve as a strong sign for the diagnosis of verbal autistic children, often missed or delayed in being diagnosed. We also discuss strategies via which language is acquired in ASD.

We examined neural mechanisms associated with the learning of novel morphologically derived words in native Hebrew speakers within the Complementary Learning Systems (CLS) framework. Across four sessions, 28 participants were trained on an artificial language, which included two types of morphologically complex words: linear (root + suffix) with a salient structure, and non-linear (root interleaved with template), with a prominent derivational structure in participants' first language (L1). A third simple monomorphemic condition, which served as baseline, was also included. On the first and fourth sessions, training was followed by testing in an fMRI scanner. Our behavioural results showed decomposition of both types of complex words, with the linear structure more easily learned than the non-linear structure. Our fMRI results showed involvement of frontal areas, associated with decomposition, only for the non-linear condition, after just the first session. We also observed training-related increases in activation in temporal areas specifically for the non-linear condition, which was correlated with participants' L1 morphological awareness. These results demonstrate that morphological decomposition of derived words occurs in the very early stages of word learning, is influenced by L1 experience, and can facilitate word learning. However, in contrast to the CLS framework, we found no support for a shift from reliance on hippocampus to reliance on cortical areas in any of our conditions. Instead, our findings align more closely with recent theories showing a positive correlation between changes in hippocampus and cortical areas, suggesting that these representations co-exist and continue to interact with one another beyond initial learning.

Antiviral drugs have classically been developed by directly disrupting the functions of viral proteins. However, this strategy has been largely unsuccessful due to the rapid generation of viral escape mutants. It has been well established that as compared to the virus-centric approach, the strategy of developing antiviral drugs by targeting host-dependency factors (HDFs) minimizes drug resistance. However, recent reports have indicated that drug resistance against some of the host-targeting antiviral agents can in fact occur under some circumstances. Long-term selection pressure of a host-targeting antiviral agent may induce the virus to use an alternate cellular factor or alters its affinity towards the target that confers resistance. Alternatively, virus may synchronize its life cycle with the patterns of drug therapy. In addition, virus may subvert host's immune system to perpetuate under the limiting conditions of the targeted cellular factor. This review describes novel potential mechanisms that may account for the acquiring resistance against agents that target HDFs.

Fusing three-dimensional (3D) and multispectral (MS) imaging data holds promise for high-throughput, comprehensive plant phenotyping to decipher genome-to-phenome knowledge. The acquisition of high-quality 3D multispectral point clouds (3DMPCs) of plants remains challenging because of poor 3D data quality and limited radiometric calibration methods for plants with complex canopy structure. We proposed a novel 3D spatial-spectral data fusion approach to collect high-quality 3DMPCs of plants by integrating the next-best-view (NBV) planning for adaptive data acquisition and Neural REference Field (NeREF) for radiometric calibration. Our approach was used to acquire 3DMPCs of perilla, tomato and rapeseed plants with diverse plant architecture and leaf morphological features and evaluated by the accuracy of chlorophyll content and equivalent water thickness (EWT) estimation. Results showed that the completeness of plant point clouds collected by our approach was improved by an average of 23.6% compared with the fixed viewpoints alone. The NeREF-based radiometric calibration with the hemispherical reference outperformed the conventional calibration method by reducing the root mean square error (RMSE) of 58.93% for extracted reflectance spectra. The RMSE for chlorophyll content and EWT predictions decreased by 21.25% and 14.13% using the partial least squares regression (PLSR) with the generated 3DMPCs. Our study provided an effective and efficient way to collect high-quality 3DMPCs of plants under the natural light condition, which improves the accuracy and comprehensiveness of phenotyping plant morphological and physiological traits and facilitates plant biology and genetic studies and breeding programs.

BACKGROUND AND AIM: Although rapid on-site cytological evaluation (ROSE) for endoscopic ultrasound (EUS)-guided tissue acquisition (EUS-TA) may increase diagnostic yield, it is not widely available. Macroscopic on-site evaluation (MOSE) is an alternative modality although it is not standardized for EUS-guided fine-needle biopsy (FNB). We evaluated diagnostic performance of MOSE compared with conventional technique of EUS-TA using core biopsy needle. METHODS: Consecutive patients undergoing EUS-FNA for solid lesions were randomized to MOSE or conventional arms. The primary and secondary outcome measures were diagnostic accuracy, diagnostic yield, sensitivity, specificity, positive and negative predictive values, and the number of passes, respectively. The optimum parameters for macroscopic visible core (MVC, i.e., length, number) by MOSE to achieve accurate diagnosis were evaluated. RESULTS: Ninety-six patients (48 conventional and 48 MOSE) were enrolled. Mean lesion size was larger in MOSE arm (32.67 ± 7.22 vs 29.31 ± 6.98 mm, P = 0.023). Diagnostic accuracy (95.8% vs 91.6%), diagnostic yield (97.9% vs 95.8%), procedure duration, and adverse events of the two methods were similar. Median number of passes with MOSE was less (2 vs 3 P = 0.000). Area under the receiver operating characteristic curve showed that with MOSE, obtaining a total MVC length of 11.5 mm had 93.3% sensitivity, and 2.5 MVC cores (each 4 mm) had 86.7% sensitivity for malignancy diagnosis. CONCLUSIONS: EUS-FNB with MOSE, a simple reliable technique, can achieve a high and comparable diagnostic accuracy with lesser number of passes. Obtaining longer length and greater number of MVC increase the sensitivity to diagnose malignancy with MOSE.

OBJECTIVE: To evaluate the clinical effectiveness of surface electromyography combined with a spine 3D data system. METHODS: 10 idiopathic scoliosis patients (age: 12.90±3.67 years, height: 155.90±20.07 cm, weight: 48.00±12.86 kg, 6 major thoracic lateral bends and 4 lumbar bends) who were selected to attend the outpatient clinic of Ruijin Rehabilitation Hospital, First diagnosed as scoliosis by imaging examination (EOS system), evaluation by using the surface electromyography monitoring system, the radiation-free 3D spine data acquisition and analysis system, the surface electromyography of the paravertebral muscles (root mean square value (RMS) of the resting standing and action position of the spine) and the Cobb angle of scoliosis were recorded. RESULTS: The RMS of the convex side was larger than that of the concave side in the two postures (P<0.05), and The RMS value of bilateral action position was significantly higher than that of rest position (P<0.05). There was no correlation between RMS ratio and Cobb angle in two positions of thoracic scoliosis (P>0.05). There was a significant correlation between the RMS ratio at rest position of lumbar scoliosis and Cobb angle. At rest, the RMS value of convex side was significantly correlated with the Cobb angle measured by the EOS system and the 3D data system (P<0.05, of which P<0.01). The Cobb angle measured by 3D data system and the EOS system was significantly correlated (P<0.01). CONCLUSION: Surface EMG can be used to evaluate the motor function of paravertebral muscles on both sides of scoliosis patients. The new 3D data system has high accuracy in evaluating scoliosis. The combination of the two can dynamically evaluate scoliosis, which is more clinically effective.

Climbing plants are important components of tropical and many temperate forest ecosystems. Current studies regard climbing plants as a single ecological plant type and ignore the ecological differences resulting from their climbing mechanisms, which may lead to a misrepresentation of the role of climbing plants in forest dynamics. Based on behavioral traits and economic traits of climbing plants, we test the hypothesis that tendril climbers and stem twiners are characterized by different resource acquisition strategies. We quantified and compared 4 behavioral traits and 7 economic traits of four stem twining vines and four tendril vines in a temperate oak forest and further tested their differences in resource acquisition strategy. Our study found that tendril vines were scattered in a group distinct from stem twining vines along the first axes of the principal component analysis using four behavioral traits and seven economic traits, being located at the more acquisitive end with more hosts, a larger distance to length ratio of stem, higher leaf and root nitrogen concentrations, and lower leaf carbon content, while stem twining vines showed the opposite trends. These results indicate that tendril vines have a more acquisitive strategy than stem twining vines. The findings suggest a functional variability among the different climbing mechanisms, and which should be accounted for in future studies.

This study aimed to investigate how ecological constraints influence individual player skill demands in community-level junior Australian Football (AF). Footage from 68 junior AF games, incorporating 1874 unique players, were analysed for individual skill demands such as disposals, kicks, handballs, tackles, marks, and shots at goal per player per game. Multiple linear mixed effect models were created for each skill, using player as a random effect. The impact of age, relative area per player (RAPP), number of players on the team, quarters in midfield, and weather on skill demands was assessed in U9-U14 boys. Finally, the influence of sex on the same skills was explored in U11-U14 boys and girls. Younger players were found to have fewer disposals and handballs, but more tackles, per player than older players (all p < 0.05). Further, more quarters in the midfield increased all skill demands (all p < 0.001). Finally, girls had fewer disposals and marks, but more tackles, per player than boys (all p < 0.05). Overall, age, sex, and position significantly impacted players' exposure to skills in junior AF. Game designers should consider methods to improve skill involvement and reduce tackling exposure early in the junior pathway, with further emphasis on the girls' game.

Hyperbaric oxygen (HBO) exposure has recently been reported to be effective in spatial learning and memory. Additionally, HBO exposure considerably improves performance on motor tasks. These findings suggest that HBO exposure may facilitate motor learning. However, the specific effects of HBO exposure on motor learning remain largely unexplored. The present study aimed to investigate the effects of HBO exposure on motor learning tasks. In the experimental animal models (control n = 8, HBO n = 8), the HBO environment was exposed to 100 % oxygen with the chamber at 2.0 atmosphere absolute (ATA) for 90 min/day for 20 days. The motor learning task was an accelerated rotating bar task (bar width, 3 and 6 cm; rotation speed, 4-40 rpm; acceleration, 0.4, 0.6, and 0.8 rpm/s). The learning task was performed for 3 consecutive days. The HBO group showed a main effect of the day factor on the bar with a width of 6 cm, and significant differences were observed for each day comparison. However, no main effect of the day factor was observed in the control group. Additionally, significant differences were found in the bar with a width of 3 cm for both groups between days 1 and 2 and between days 1 and 3. In conclusion, these findings suggest that HBO exposure has a positive effect on more challenging motor learning tasks.

Chemistry assigns phosphorus and its most oxidized form, inorganic phosphate, unique roles for propelling bioenergetics and metabolism in all domains of life, possibly since its very origin on prebiotic Earth. For plants, access to the vital mineral nutrient profoundly affects growth, development and vigour, thus constraining net primary productivity in natural ecosystems and crop production in modern agriculture. Unlike other major biogenic elements, the low abundance and uneven distribution of phosphate in Earth's crust result from the peculiarities of phosphorus cosmochemistry and geochemistry. Here, we trace the chemical evolution of the element, the geochemical phosphorus cycle and its acceleration during Earth's history until the present (Anthropocene) as well as during the evolution and rise of terrestrial plants. We highlight the chemical and biological processes of phosphate mobilization and acquisition, first evolved in bacteria, refined in fungi and algae and expanded into powerful phosphate-prospecting strategies during land plant colonization. Furthermore, we review the evolution of the genetic and molecular networks from bacteria to terrestrial plants, which monitor intracellular and extracellular phosphate availabilities and coordinate the appropriate responses and adjustments to fluctuating phosphate supply. Lastly, we discuss the modern global phosphorus cycle deranged by human activity and the challenges imposed ahead. BACKGROUND: Endobronchial Ultrasound-Guided Transbronchial Aspiration (EBUS-TBNA) has become the standard for initial lung cancer diagnosis and staging. Previous guidelines have generally focused on the "when" and "how" of EBUS-TBNA; however, little guidance is available on handling and processing specimens during and after acquisition to help optimize both diagnostic yield and tissue integrity for ancillary studies. This document examines the available literature on EBUS-TBNA specimen processing and handling. METHODS: Rigorous methodology was applied to provide a trustworthy evidence-based guideline and expert panel report. Panelists developed key clinical questions utilizing the PICO (population, intervention, comparator, and outcome) format, addressing specific topics in EBUS-TBNA specimen processing. MEDLINE (via PubMed) and the Cochrane Library were systematically searched to identify relevant literature, supplemented by manual searches. References were screened for inclusion with document evaluation tools to assess the quality of included studies, extract meaningful data, and grade the level of evidence to support each recommendation or suggestion. RESULTS: Our systematic review and critical analysis of the literature of the 9 PICO questions related to handling and processing EBUS-TBNA specimens resulted in nine evidence-based statements. CONCLUSIONS: Evidence of the handling and processing of EBUS-TBNA specimens varies in strength but is satisfactory in some areas to guide clinicians in certain aspects of specimen handling. Additional research in many aspects of specimen handling and processing is needed to help improve our knowledge base.

Selecting a dog that is incompatible with the owner's expectations can negatively impact both parties. Previous studies on dog acquisition have primarily focused on shelter environments, using closed-ended questions to assess hypothetical preferences. In contrast, our study employed open-ended questions with a convenience sample of Austrian dog owners (N = 1077) to retrospectively explore why the owners chose their dogs. We also examined consistency in owners' responses and the influence of owner characteristics (age, education, household composition, previous dog experience, purpose of acquisition) on their reasons. Content analysis revealed 24 codes; the frequency of codes was 2.4/response. The most frequent codes were breed-based choice (29%), choosing on a whim, without careful consideration (24%), work/sport skills (22%), and rescuing a dog (17%). The least frequent were the age (1%), health (1%), sex (1%), and guarding skills (0.6%) of the dog. Twelve codes were consistent over time, and ten were consistent across dogs, indicating that the owners showed a consistent preference for certain traits. Except for the owner's education level, all characteristics affected the likelihood of mentioning at least one code. Most associations were found with the presence of children in the household: owners with children preferred friendly, easily manageable, and easy-to-train dogs and were less likely to adopt or rescue compared to owners living without children. Our findings also highlight discrepancies between spontaneous (free-text) reports and responses to closed-ended questions, underscoring the importance of qualitative data in better understanding the motivations behind and the factors influencing dog acquisition.

Ceresini treehoppers are present in northern California vineyard ecosystems, including the closely related Spissistilus and Tortistilus (Hemiptera: Membracidae). These membracids are not direct pests of wine grapes, but S. festinus is a vector of grapevine red blotch virus (GRBV). No information is available on the ability of Tortistilus spp. to transmit GRBV. In this study, Tortistilus were collected on yellow panel cards across 102 vineyard sites and surrounding areas in Napa Valley, California, USA in 2021-2023. Specimens were morphotyped, sexed and tested for GRBV ingestion and acquisition by multiplex PCR or qPCR. Phylogenetic analysis of the partial sequence of mt-COI and ITS gene fragments of a subset of 40 Tortistilus specimens revealed clustering in a monophyletic clade with T. wickhami with the former barcode sequence. Only 6% (48/758) of the T. wickhami tested positive for GRBV, but none of the heads with salivary glands (0%, 0/50) of the dissected specimens tested positive for GRBV, indicating no virus acquisition. In contrast, half of the dissected heads with salivary glands of S. festinus (52%, 12/23), from the same collection vineyard sites, tested positive for GRBV. Together, our findings confirmed the presence of T. wickhami in northern California vineyards and suggested a dubious role of this treehopper as a vector of GRBV.
