Phosphorus (P) is essential for biological systems, playing a pivotal role in energy metabolism and forming crucial structural components of DNA and RNA. Yet its bioavailable forms are scarce. Phytate, a major form of stored phosphorus in cereals and soils, is poorly bioavailable due to its complex structure. Phytases, enzymes that hydrolyze phytate to release useable phosphorus, are vital in overcoming this limitation and have significant biotechnological applications. This study employed novel method to isolate and characterize bacterial strains capable of metabolizing phytate as the sole carbon and phosphorus source from the Andes mountains soils. Ten strains from the genera Klebsiella and Chryseobacterium were isolated, with Chryseobacterium sp. CP-77 and Klebsiella pneumoniae CP-84 showing specific activities of 3.5 ± 0.4 nkat/mg and 40.8 ± 5 nkat/mg, respectively. Genomic sequencing revealed significant genetic diversity, suggesting CP-77 may represent a novel Chryseobacterium species. A fosmid library screening identified several phytase genes, including a 3-phytase in CP-77 and a glucose 1-phosphatase and 3-phytase in CP-84. Phylogenetic analysis confirmed the novelty of these enzymes. These findings highlight the potential of phytase-producing bacteria in sustainable agriculture by enhancing phosphorus bioavailability, reducing reliance on synthetic fertilizers, and contributing to environmental management. This study expands our biotechnological toolkit for microbial phosphorus management and underscores the importance of exploring poorly characterized environments for novel microbial functions. The integration of direct cultivation with metagenomic screening offers robust approaches for discovering microbial biocatalysts, promoting sustainable agricultural practices, and advancing environmental conservation.

Zinc is a vital trace element, yet its deficiency is common in various populations. This study addresses the gap in understanding zinc intake and its relationship with key nutritional parameters in a Colombian population. We analyzed data from 12,987 individuals, focusing on the daily intake of zinc, phytate, protein, and calcium, and used the phytate/zinc molar ratio as an input parameter in the Miller et al. (2013) model. This model was employed to estimate the total absorbed zinc (TAZ) and the fractional absorption of zinc (FAZ). Our findings highlight a general trend towards insufficient intake compared to the standards of the Institute of Medicine (IOM) and Colombia, with a significant percentage of the population falling below the estimated average requirement (EAR) and recommended daily allowance (RDA) for zinc, underscoring the need for targeted nutritional strategies. Our study contributes to a broader understanding of zinc nutrition and public health implications in Colombia, providing a basis for future dietary guidelines and health interventions.

The objective of this study was to evaluate the effect of a novel consensus bacterial 6-phytase variant (PhyG) on egg productivity, eggshell quality, and body composition of laying hens fed inorganic phosphate-free diets with reduced energy and nutrients from 23 to 72 wk of age. Five treatments were randomly assigned, performing 28 replicates per treatment with 4 hens each, totaling 560 Hy-Line W80 birds. A positive control (PC) feed was formulated to contain adequate levels of energy and nutrients. A negative control (NC) feed was formulated without added inorganic phosphate (0.12% nonphytic phosphorus [nPP]) and reduced in Ca, Na, dig AA, and metabolizable energy in comparison with PC feed. Phytase was supplemented in the NC feed at 0, 300, 600, and 900 FTU/kg of feed. The responses evaluated were performance, egg quality, economic analysis, body composition, and tibia composition. Data were analyzed by a 2-factor (diet and age) repeated measure analysis. Overall, the feed intake, hen-day egg production, egg mass, and egg revenue were reduced by the complete removal of dicalcium phosphate (DCP) (P < 0.05). Supplement phytase in the NC diet elicits a positive response on each one of those variables. Laying hens consuming the NC feed with 900 FTU/kg of phytase produced more eggs per hen-housed compared with the phytase dosages of 300 and 600 FTU/kg. Body composition was not affected by dietary nPP, Ca, Na, dig AA, and energy reductions (P > 0.05). At 72-wk-old, tibia ash was reduced in hens consuming the NC diet vs. PC (P < 0.05) and no difference was observed between hens supplemented with phytase and the PC feed. Margin over feeding cost increased in a dose-dependent manner with phytase supplementation. Supplementation with 900 FTU/kg of phytase is recommended to improve the number of eggs produced per hen-housed and the number of marketable eggs produced through 23 to 72 wk of age, under this dietary setting.

The study was carried out with the objective of evaluating the effects of using phytase levels at different energy densities in the diet of broilers, from 28 to 35 days of age. The experimental diets contained increasing levels of apparent metabolizable energy corrected for nitrogen balance and different levels of phytase. Growth performance variables, nutrient digestibility, as well as variables related to bone integrity were examined. Diets containing 500 FTU/kg in combination with 3150kcal.kg-1 of AMEn resulted in better growth performance. Supplementation with 500 and 1000 FTU/kg in the diets provide better coefficients of apparent and ileal digestibility of calcium and phosphorus, but negatively influence the deposition of calcium, phosphorus, dry matter, and ash in the tibia, in addition to adversely affecting the breakage and area of these bones in broilers. Bone length and the levels of magnesium and zinc present in the tibias were not influenced by the energy and phytase levels of the diets. There is no interaction between the different energetic densities and the phytase doses in the variables analyzed in the studied period. Increasing the energy density of diets resulted in improved apparent and ileal digestibility for most nutrients.


O estudo foi realizado com o objetivo de avaliar os efeitos da utilização de níveis de fitase em diferentes densidades energéticas na dieta de frangos de corte, no período de 28 a 35 dias de idade. As dietas experimentais contiveram níveis crescentes de energia metabolizável aparente corrigida para balanço de nitrogênio e diferentes níveis de fitase. Foram avaliadas variáveis de desempenho zootécnico, digestibilidade de nutrientes e variáveis relacionadas à integridade óssea. Dietas contendo 500 FTU/kg de fitase em combinação com 3150kcal.kg-1 de EMAn resultam em melhor desempenho zootécnico. A suplementação com 500 e 1000 FTU/kg nas dietas propicia melhores coeficientes de digestibilidade aparente e ileal do cálcio e do fósforo, mas influencia negativamente na deposição de cálcio, fósforo, matéria seca e cinzas das tíbias, além de afetar adversamente a quebra e a área desses ossos em frangos de corte. O comprimento do osso e os níveis de magnésio e zinco presentes nas tíbias não são influenciados pelos níveis energéticos e de fitase das dietas. Não há interação entre as diferentes densidades energéticas e as doses de fitase nas variáveis analisadas no período estudado. O aumento da densidade energética das dietas resulta em melhora na digestibilidade aparente e ileal para a maioria dos nutrientes.

Neuropathic pain is a condition with varying origins, including reduced dietary micronutrient intake. Phytate is a polyphosphate found in seeds and grains that can act as an antinutrient due to the ability of sequester essential divalent metals. Here we tested whether moderate dietary phytate intake could alter nociceptive pain. We subjected weaning mice to a chow supplemented with 1% phytate for eight weeks. Body weight gain, glycemic responses, food ingestion, water ingestion, and liver and adipose tissue weights were not altered compared to controls. We observed a decreased mechanical allodynia threshold in the intervention group, although there were no changes in heat- or cold-induced pain. Animals consuming phytate showed reduced spinal cord tumor necrosis factor (TNF), indicating altered inflammatory process. These data provide evidence for a subclinical induction of mechanical allodynia that is independent of phytate consumption in animals with otherwise normal phenotypic pattern.

The effects of increasing doses of bacterial phytase (Citrobacter braakii) on performance and carcass characteristics of growing-finishing pigs was evaluated. A total of 120 barrows weighing 25.16 ± 2.80 kg and 68 days old were submitted to five treatments: positive control diet (PC) containing inorganic phosphorus (P) and limestone (Ca); negative control (NC) with reductions in P (by 0.13%) and Ca (by 0.11%); and three NC diets supplemented with 1500 (NC15), 3000 (NC30) and 4500 (NC45) units of phytase (FYT)/kg. The daily weight gain (DWG) in growth phase I (68-91 days) was higher in the PC, NC15 and NC30, compared to the NC (1.06, 1.06, 1.06 vs. 0.95, respectively). The DWG in finishing phase II (141-156 days) was higher in the NC15 (1.20 kg) and NC30 (1.14 kg) than in the NC45 (0.94 kg). The final weights (LW156) in the NC15 (122.95 kg LW) were higher than NC (116.47 kg LW) and NC45 (114.43 kg LW). Over the entire period, a quadratic effect (2012 FYT) was observed for the DWG. The increasing levels of phytase in corn and soybean meal-based diets improved the DWG and carcass traits; however, the addition of more than 3000 FYT/kg of feed should be carefully studied to determine enzyme viability.

Brazil is the world's third largest common bean (Phaseolus vulgaris L.) producer, and 60% of its population consumes this legume. Although organic farming is a sustainable alternative to nonorganic agriculture, its effect on chemical composition is still controversial. Therefore, the aim of this study was to investigate differences in the nutritional and phenolic compounds profiles between organically and nonorganically produced Brazilian black beans. Samples were obtained from the same harvest periods and from near geographical locations at metropolitan and coastal regions of Rio de Janeiro state, Brazil. No residues of 294 evaluated pesticides were detected in the samples. In both regions, organic beans had 17% fewer lipids, 10% less phytate and 20% more proteins when compared to nonorganic ones. Sixteen different phenolic compounds were identified as soluble and insoluble forms in black beans, with anthocyanins being the most abundant (on average, 66%). In both regions, soluble and total phenolic compounds contents in organic beans were consistently higher (on average, 25% and 28%, respectively) than in nonorganic ones. Our results show that organic farming improves the nutritional profile and increases the phenolic compounds content of black beans.

Dietary zinc (Zn) deficiency is widespread globally, and is particularly prevalent in low- and middle-income countries (LMICs). Cowpea (Vigna unguiculata (L.) Walp) is consumed widely in LMICs due to its high protein content, and has potential for use in agronomic biofortification strategies using Zn. This study aimed to evaluate the effect of Zn biofortification on grain nutritional quality of 29 cowpea genotypes. Zn application did not increase cowpea yield. In 11 genotypes sucrose concentration, in 12 genotypes total sugar concentration, and in 27 genotypes storage protein concentration increased in response to Zn supply. Fifteen genotypes had lower concentrations of amino acids under Zn application, which are likely to have been converted into storage proteins, mostly comprised of albumin. Phytic acid (PA) concentration and PA/Zn molar ratio were decreased under Zn application. Six genotypes increased shoot ureides concentration in response to Zn fertilization, indicating potential improvements to biological nitrogen fixation. This study provides valuable information on the potential for Zn application to increase cowpea grain nutritional quality by increasing Zn and soluble storage protein and decreasing PA concentration. These results might be useful for future breeding programs aiming to increase cowpea grain Zn concentrations through biofortification.

Phosphorus is one of the most abundant chemical elements but has a low bioavailability index. Therefore, microrganisms play a fundamental role in providing insoluble phosphorus to plants. The objective of this study was to evaluate the capacity of bacteria to solubilize inorganic phosphates in soils with different fertilization histories. Soil and rhizosphere samples were collected from a Red Distroferric Latosol, including a control without mineral or organic fertilizer (C), treatment with mineral fertilizer (MF) according to the needs of each crop, and treatment with organic fertilizer [300 m3 ha-1 of swine wastewater (SW)]. The medium containing calcium phytate presented more colony-forming units (CFU) for all fertilization histories, and growth in treatments C and MF was 50% higher than treatment with SW. CFU values in soils treated with SW were lower than those in the other treatments, and the diversity of insoluble phosphate-solubilizing bacteria (PSB) was higher in treatment C. These results indicate a negative relationship between phosphorus concentrations and the number of PSB.


O fósforo é um dos elementos químicos mais abundantes, mas tem um baixo índice de biodisponibilidade. Portanto, micro-organismos desempenham um papel fundamental no fornecimento de fósforo insolúvel para as plantas. O objetivo deste trabalho é avaliar a capacidade das bactérias em solubilizar fosfatos inorgânicos em solos com diferentes históricos de fertilização. Amostras de solo e rizosfera foram coletadas de um Latossolo Vermelho Distroférrico, incluindo controle sem adubação mineral ou orgânica (C), tratamento com adubação mineral (MF) de acordo com as necessidades de cada cultura e tratamento com adubação orgânica [300 m3 ha-1 de águas residuárias da suinocultura (SW)]. O meio contendo fitato de cálcio apresentou mais unidades formadoras de colônias (UFC) para todas as histórias de fertilização, e o crescimento nos tratamentos C e MF foi 50% maior que o tratamento com SW. Os valores de CFU nos solos tratados com SW foram menores que nos demais tratamentos, e a diversidade de bactérias insolúveis solubilizadoras de fosfato (PSB) foi maior no tratamento C. Esses resultados indicam uma relação negativa entre as concentrações de fósforo e o número de PSB.

Cereals and pseudocereals are a rich source of nutrients and trace elements, but their dietary bioavailability is low due to the presence of phytate (IP6), an antinutritional compound with the ability to chelate cations and proteins. Phytase is an enzyme that catalyzes the hydrolysis of IP6 and it is used as an additive improving the nutritional quality of grain-based foods. The aim of this study was to select lactic acid bacteria (LAB) isolated from pseudocereals with phytase activity, characterize their production and activity, and purify the enzyme. LAB strains isolated from grains and spontaneous sourdough of quinoa and amaranth were grown in the Man Rogosa and Sharpe medium where the inorganic phosphate (Pi) was replaced by 1% of IP6. Phytase activity was determined by measuring the Pi released from IP6. Phytase of Lactobacillus (L.) plantarum CRL1964 (PhyLP) showed the highest specific activity from 73 LAB evaluated. IP6 induces PhyLP production, which is at its maximum at the end of the exponential phase. PhyLP was thermostable and maintained its activity under acidic conditions. The enzymatic activity is stimulated by ethylenediaminetetraacetic acid, Co2+, and ascorbic acid. PhyLP was partially purified and showed a molecular mass of 55 kDa. L. plantarum CRL1964 and/or PhyLP have the potential to be included in the processing of cereal/pseudocereals based products for animal feed and/or the food industry improving its nutritional value.

The aim of this study was to evaluate the effect of two types of phytases (the 1st produced from Escherichia coli and the 2nd from Aspergillus oryzae) with different dosages (300 and 900 Unit Phytase or FTU) on performance, organ biometry and bone quality of replacement pullets in the period of 8 to 17 weeks of age. A total of 288 Hy-Line White laying hens were used at 8 weeks of age. A completely randomized design was adopted in a 2 x 2 factorial scheme, totaling 4 treatments with 8 replicates of 9 birds each. Performance, biometric variables and bone characteristics were evaluated. There was no significant interaction between the factors for any of the evaluated variables. The conversion and the accumulated consumption were influenced (p 0.05) by the phytase dosages, with the highest results being observed for the consumption obtained with 300 FTU, and the best results for the feed conversion obtained with the dosage of 900 FTU. The relative weight of the liver was influenced by the treatments (p 0.05), presenting higher values with bacterial phytase and a dosage of 900 FTU. The sternum length and tibial deformity were influenced by fungal phytase (p 0.05). It is concluded that the use of 900 FTU superdosing and fungal phytase improves the performance and bone characteristics of light replacement pullets.(AU)

There is a scarcity of information on mineral and phytate content in plant-based foods in Bolivia. This study aimed to analyze iron, zinc, calcium, and phytate content and estimate the mineral bioavailability of foods consumed in Chapare, Bolivia. Minerals and phytate were analyzed, and bioavailability was estimated in 17 food samples. Leafy vegetables and green legumes had the highest mineral content, followed by pseudocereals. Estimated mineral bioavailability was low for cereals, dry legumes, pseudocereals, and flaxseeds foods mainly due to phytate content. But estimated zinc bioavailability for black cornmeal, yellow corn, and dry peas was moderate. Strong correlations (p < 0.01) were found between the three minerals, while phytate correlated negatively to iron, zinc, and calcium. To get an overview of the estimated mineral bioavailability of plant-based diets, we have included foods, from the same area, analyzed in a previous study where the evaluated diet covers 80% of RNI for iron and zinc, but <40% of calcium. In conclusion, leafy vegetables and green legumes had the highest contents of minerals and the lowest phytate content of the foods analyzed in the study. The usage of processing strategies and dietary diversification to reduce phytate content would significantly improve estimated mineral bioavailability in plant-based diets.

Although aquaculture is among the fastest growing food production sectors in the world, one of the bottlenecks for the continuity of its expansion is the dependence of animal protein on commercial feed formulations. Vegetable proteins are an alternative due to the low cost and high availability. However, this protein source is accompanied by a series of antinutritional and pro-inflammatory compounds, including phytate. Phytases can be added in feed for phytate degradation and increase nutrient availability. However, the use of purified phytases significantly increases the production costs. An interesting alternative is to use probiotics genetically modified as bioreactors for phytase production. In the present study, a strain of Bacillus subtilis secreting a fungal phytase was used to evaluate the effect of a feed with high content of soybean meal on zebrafish (Danio rerio). We analysed the condition factor (K) of fish, and the expression of genes related to the immune system, inflammatory response and oxidative. stress. The results obtained demonstrate that the transgenic probiotic was efficient in improving the fish condition factor, stimulating the immune system, reducing the inflammatory response and oxidative stress. Thus, probiotics acting as phytase bioreactors can be considered an interesting tool for the adaptation of commercial species to feed of lower cost.

One of the greatest challenges is to reduce malnutrition worldwide while promoting sustainable agricultural and food systems. This is a daunting task due to the constant growth of the population and the increasing demands by consumers for functional foods with higher nutritional values. Cereal grains are the most important dietary energy source globally; wheat, rice, and maize currently provide about half of the dietary energy source of humankind. In addition, the increase of celiac patients worldwide has motivated the development of gluten-free foods using alternative flour types to wheat such as rice, corn, cassava, soybean, and pseudocereals (amaranth, quinoa, and buckwheat). Amaranth and quinoa have been cultivated since ancient times and were two of the major crops of the Pre-Colombian cultures in Latin- America. In recent years and due to their well-known high nutritional value and potential health benefits, these pseudocereals have received much attention as ideal candidates for gluten-free products. The importance of exploiting these grains for the elaboration of healthy and nutritious foods has forced food producers to develop novel adequate strategies for their processing. Fermentation is one of the most antique and economical methods of producing and preserving foods and can be easily employed for cereal processing. The nutritional and functional quality of pseudocereals can be improved by fermentation using Lactic Acid Bacteria (LAB). This review provides an overview on pseudocereal fermentation by LAB emphasizing the capacity of these bacteria to decrease antinutritional factors such as phytic acid, increase the functional value of phytochemicals such as phenolic compounds, and produce nutritional ingredients such as B-group vitamins. The numerous beneficial effects of lactic fermentation of pseudocereals can be exploited to design novel and healthier foods or grain ingredients destined to general population and especially to patients with coeliac disease.

Phytate (L12- ) is a relevant natural product. It interacts strongly with biologically relevant cations, due to the high negative charge exhibited in a wide pH range. The synthesis and crystal structures of the mixed-ligand Cu(II) polynuclear complexes K(H2 tptz)0.5 [Cu(H8 L)(tptz)] ⋅ 3.6H2 O (1), K(H2 O)3 {[Cu(H2 O)(bpca)]3 (H8 L)} ⋅ 1.75H2 O (2), and K1.5 (H2 O)2 [Cu(bpca)](H9.5 L) ⋅ 8H2 O (3) (tptz=2,4,6-tri(pyridin-2-yl)-1,3,5-triazine; Hbpca=bis(2-pyridylcarbonyl) amine) are reported herein. They were obtained by the use of an aromatic rigid amine, which satisfies some of the metal coordination sites and promotes the hierarchical assembly of 2D polymeric structures. Speciation of phytate-Cu(II)-Hbpca system and determination of complex stability constants were performed by means of potentiometric titrations, in 0.15 M NMe4 Cl at 37.0 °C, showing that, even in solution, this system is able to produce highly aggregated complexes, such as [Cu3 (bpca)3 (H7 L)]2- . Furthermore, the Cu(II)-mediated tptz hydrolysis was studied by UV-vis spectroscopy at room temperature in 0.15 M NMe4 Cl. Based on the equilibrium results and with the aid of molecular modelling tools, a plausible self-assembly process for 2 and 3 could be proposed.

Phytases are enzymes involved in organic phosphorus cycling in nature and widely used as feed additives in animal diets. Thermal tolerance is a desired property of phytases. The objectives of this study were to screen and characterize bacterial phytases from Chilean hydrothermal environments. In this study, 60% (30 of 63) of screened thermophilic (60 °C) isolates showed phytase activity in crude protein extracts. The characterization of phytase from two selected isolates (9B and 15C) revealed that both isolates produce phytases with a pH optimum at 5.0. The temperature optimum for phytate dephosphorylation was determined to be 60 and 50 °C for the phytases from the isolates 9B and 15C, respectively. Interestingly, the phytase from the isolate 15C showed a residual activity of 46% after incubation at 90 °C for 20 min. The stepwise dephosphorylation of phytate by protein extracts of the isolates 9B and 15C was verified by HLPC analysis. Finally, the isolates 9B and 15C were identified by partial sequencing of the 16S rRNA gene as members of the genera Bacillus and Geobacillus, respectively.

Background: Estimated physiologic requirements (PRs) for zinc increase in late pregnancy and early lactation, but the effect on dietary zinc requirements is uncertain.Objective: The aim of this study was to determine changes in daily fractional absorbed zinc and total absorbed zinc (TAZ) from ad libitum diets of differing phytate contents in relation to physiologic zinc requirements during pregnancy and lactation.Methods: This was a prospective observational study of zinc absorption at 8 (phase 1) and 34 (phase 2) wk of gestation and 2 (phase 3) and 6 (phase 4) mo of lactation. Participants were indigenous Guatemalan women of childbearing age whose major food staple was maize and who had been randomly assigned in a larger study to either of 2 ad libitum feeding groups: low-phytate maize (LP; 1.6 mg/g; n = 14) or control maize (C; 7.1 mg/g; n = 8). Total dietary zinc (milligrams per day, TDZ) and phytate (milligrams per day) were determined from duplicate diets and fractional absorption (FAZ) by dual isotope ratio technique (TAZ = TDZ × FAZ). All variables were examined longitudinally and by group and compared with PRs. TAZ values at later phases were compared with phase 1. Measured TAZ was compared with predicted TAZ for nonpregnant, nonlactating (NPNL) women.Results: TAZ was greater in the LP group than in the C group at all phases. All variables increased from phase 1 to phases 2 and 3 and declined at phase 4. TAZ increased by 1.25 mg/d (P = 0.045) in the C group and by 0.81 mg/d (P = 0.058) in the LP group at phase 2. At phase 3, the increases were 2.66 mg/d (P = 0.002) in the C group and 2.28 mg/d (P = 0.0004) in the LP group, compared with a 1.37-mg/d increase in PR. Measured TAZ was greater than predicted values in phases 2-4.Conclusions: Upregulation of zinc absorption in late pregnancy and early lactation matches increases in PRs of pregnant and lactating women, regardless of dietary phytate, which has implications for dietary zinc requirements of pregnant and lactating women.

myo-Inositol phosphates are an important group of biomolecules that are present in all eukaryotic cells. The most abundant member of this family in nature is InsP6 (phytate, L12- in its fully deprotonated form). Phytate interacts strongly with inorganic and organic cations, and this interaction is essential for determining the possible functions of this biomolecule. Herein, the chemical, thermodynamic, and structural characterization of phytate-MnII species is presented in a study aimed at understanding how the interaction of the two components modulates their biological roles and their bioavailability. Polynuclear complexes Mn5 (H2 L)⋅16 H2 O (1) and (H2 terpy)2 [Mn(H6 L)(terpy)(H2 O)]⋅17 H2 O (terpy=terpyridine) (2) were prepared and characterized by different techniques. The isolation of 1 and the determination of its solubility, together with potentiometric titrations of the MnII -phytate system, allow the full description of this binary system. The preparation and crystal structure of 2 show a novel coordination mode of phytate, that is, the formation of infinite polymeric chains through equatorial phosphate groups.
