RESUMO

Phytopathogenic bacteria not only affect crop yield and quality but also the environment. Understanding the mechanisms involved in their survival is essential to develop new strategies to control plant disease. One such mechanism is the formation of biofilms; i.e., microbial communities within a three-dimensional structure that offers adaptive advantages, such as protection against unfavorable environmental conditions. Biofilm-producing phytopathogenic bacteria are difficult to manage. They colonize the intercellular spaces and the vascular system of the host plants and cause a wide range of symptoms such as necrosis, wilting, leaf spots, blight, soft rot, and hyperplasia. This review summarizes up-to-date information about saline and drought stress in plants (abiotic stress) and then goes on to focus on the biotic stress produced by biofilm-forming phytopathogenic bacteria, which are responsible for serious disease in many crops. Their characteristics, pathogenesis, virulence factors, systems of cellular communication, and the molecules implicated in the regulation of these processes are all covered.

Copper-based compounds are widely used in agriculture as a chemical strategy to limit the spread of multiple plant diseases; however, the continuous use of this heavy metal has caused environmental damage as well as the development of copper-resistant strains. Thus, it is important to understand how the bacterial phytopathogens evolve to manage with this metal in the field. The MqsRA Toxin-Antitoxin system has been recently described for its function in biofilm formation and copper tolerance in Xylella fastidiosa, a plant-pathogen bacterium responsible for economic damage in several crops worldwide. Here we identified differentially regulated genes by X. fastidiosa MqsRA by assessing changes in global gene expression with and without copper. Results show that mqsR overexpression led to changes in the pattern of cell aggregation, culminating in a global phenotypic heterogeneity, indicative of persister cell formation. This phenotype was also observed in wild-type cells but only in the presence of copper. This suggests that MqsR regulates genes that alter cell behavior in order to prime them to respond to copper stress, which is supported by RNA-Seq analysis. To increase cellular tolerance, proteolysis and efflux pumps and regulator related to multidrug resistance are induced in the presence of copper, in an MqsR-independent response. In this study we show a network of genes modulated by MqsR that is associated with induction of persistence in X. fastidiosa. Persistence in plant-pathogenic bacteria is an important genetic tolerance mechanism still neglected for management of phytopathogens in agriculture, for which this work expands the current knowledge and opens new perspectives for studies aiming for a more efficient control in the field.

Resinous exudate obtained from the aerial parts of Adesmia boronioides Hook.f. were evaluated to determine anti-phytopathogenic effects. Briefly, resinous exudate was obtained by dipping fresh plant material in dichloromethane; chemical composition was determined by GC-MS; and minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were evaluated against four phytopathogenic bacteria. Resinous exudate yield was 8.5% (resin/fresh plant), of which esquel-6-en-9-one (14.25%), esquel-7-en-9-one (5.86%), and veratric acid (2.59%) were the effective antibacterial compounds. Tested against Pectobacterium carotovorum subsp. carotovora, Erwinia amylovora, Bacillus subtilis, and Pseudomonas syringae, MICs and MBCs ranged from 16 to 128 µg/mL and 32-256 µg/mL, respectively. These results provide initial evidence that resinous bush A. boronioides is a new and alternative source of substances with agricultural interest.

The antibiotic effect of aqueous extract of propolis was evaluated in vitro against five species of bacteria. The growth of Agrobacterium tumefaciens, Clavibacter michiganensis subsp. michiganensis and Xanthomonas axonopodis pv. phaseoli was completely inhibited in a medium containing 10% of propolis. Erwinia chrysanthemi was partially inhibited and Pseudomonas syringae pv. tabaci was not affected. Lower concentration of the aqueous extract was not effective to promote a desirable antibiotic action against all bacterial species tested. The active ingredient present in the propolis was thermostable. The potential use of the propolis to control phytopathogenic bacteria was demonstrated.


O efeito antibiótico de extrato aquoso de própolis, em várias concentrações, foi avaliado para cinco espécies de bactérias fitopatogênicas. Agrobacterium tumefaciens, Clavibacter michiganensis subsp. michiganensis e Xanthomonas axonopodis pv. phaseoli foram completamente inibidas em meio de cultura contendo 10% de extrato de própolis. Erwinia chrysanthemi foi parcialmente inibida, enquanto Pseudomonas syringae pv. tabaci se mostrou insensível ao extrato, desenvolvendo colônias idênticas àquelas observadas em meios livres de própolis. Concentrações menores não foram suficientes para exercer um efeito antimicrobiano desejável sobre as bactérias pertencentes a todas as espécies testadas. Resultados idênticos foram obtidos quando própolis foi incorporado ao meio antes ou após a autoclavagem, demonstrando que a substância ativa presente no extrato não é termosensível. Foi demonstrado, portanto, o uso potencial do própolis como antibiótico para o controle de bactérias fitopatogênicas.
