O fígado desempenha uma função central no metabolismo devido à sua interposição entre o trato digestivo e a circulação geral do organismo. Ele é também o principal órgão envolvido na biotransformação de substâncias exógenas (xenobióticos), com capacidade de converter compostos hidrofóbicos em hidrossolúveis, mais facilmente eliminados pelo organismo. O gossipol é uma substância fenólica tóxica presente na semente de algodão (Gossypium sp). Com o objetivo de estudar os mecanismos envolvidos na hepatotoxicidade do gossipol avaliou-se os seus efeitos no sistema antioxidante do fígado de ratos no que diz respeito ao estresse oxidativo e aspectos histopatológicos. Foram utilizados ratos machos da linhagem Wistar, separados em dois grupos, sendo que um recebeu óleo de canola (veículo, grupo Controle) e o outro recebeu gossipol na dosagem de 40 mg/kg de peso vivo do animal por 15 dias (grupo Tratado). O tratamento com gossipol promoveu alterações na atividade sérica das enzimas marcadoras de dano hepático e um significativo estresse oxidativo caracterizado pela diminuição nos níveis da glutationa reduzida (GSH) e consequente aumento da glutationa oxidada (GSSG), incluindo, ainda, danos à membrana plasmática e de organelas demonstrados pela peroxidação lipídica. O resultado da avaliação histopatológica demonstrou degeneração dos hepatócitos.


The liver plays a central role in metabolism due to its interposition between the digestive tract and the general circulation of the organism. It is also the main organ involved in biotransformation of exogenous substances (xenobiotics), with ability to convert hydrophobic compounds in water-soluble, more easily eliminated by the body. Gossypol is a toxic phenolic substance present in cotton seed (Gossypium sp.). Aiming to study the mechanisms involved in the hepatotoxicity of gossypol we evaluate its effects on the antioxidant system of rat liver performing an experiment that investigated the oxidative stress and the histopathological alterations. In this study, we used Wistar rats, divided into two groups, one that received canola oil (vehicle, Control group) and another that received gossypol at a dose of 40mg/kg body weight of the animal for 15 days (Treated group). The treatment with gossypol caused alterations in the activity of seric enzymes that indicate hepatic injury and a significant oxidative stress characterized by a decrease of reduced glutathione (GSH) levels and a consequent increase in oxidized glutathione (GSSG), including further damage to the plasma membrane and organelles showed by lipid peroxidation. The result of histopathological evaluation showed degeneration of the hepatocytes.

BACKGROUND: Oilseed rape production is widespread in cereal growing areas. Many patients attending our clinics for seasonal allergy claim that they are allergic to it. AIM: To determine the prevalence of oilseed rape allergy in our population. POPULATION: General population within a mixed cereal farming area attending a rhinology and allergy clinic in a district general hospital, UK. METHODS & MATERIALS: Retrospective chart analysis. The results of 1475 consecutive patients who underwent skin prick allergy testing over a 2-year period (January 2003-December 2005) and of 640 consecutive patients over a 1-year period (September 2008-September 2009) were analysed and compared. RESULTS: Allergy to house dust mite was found to be most common (n = 526, 25%) followed by grass pollen (n = 519, 24%) and cereals (n = 429, 20%). Oilseed rape hypersensitivity was relatively uncommon, comprising only 2% of the population tested (n = 28). CONCLUSION: Oilseed rape does not cause significant allergy, even in areas of high production. It is likely that those patients exhibiting oilseed rape allergy may in fact be symptomatic due to the effect of other allergens, acting either synergistically with the oilseed rape allergen, or more likely, in spite of it.

AIM: Finnish children with atopic dermatitis (AD) are frequently sensitized and show positive food challenge to turnip rape. We examined whether French children are also allergic to this oilseed plant and whether mustard could be the cross-reacting allergen. METHODS: Turnip rape and mustard challenge was performed to 14 Finnish and 14 French children with atopic dermatitis and positive skin prick test to turnip rape. Specific IgE antibodies were measured by ImmunoCAP and enzyme-linked immunosorbent assay (ELISA). RESULTS: Open labial or oral challenge to turnip rape was positive in 14 (100%) Finnish and five (36%) French children and mustard challenge in five Finnish and five French children. IgE antibodies to oilseed rape and mustard were slightly more frequent in the Finnish (100% and 93%) than in the French (93% and 71%) children but rare (4%) in the 28 matched controls. The same findings were true for IgE antibodies to purified 2S albumin allergens, which showed similar cross-wise IgE inhibition patterns. CONCLUSION: French children with atopic dermatitis show IgE antibodies to turnip rape, oilseed rape and mustard similarly to the Finnish children. 2S albumin allergens in the seeds of these plants are highly cross-reactive and therefore, they all could be important sensitizers in children with atopic dermatitis.

Avaliaram-se os efeitos da inclusão de farelo de canola em dietas de juvenis de pacu (Piaractus mesopotamicus) sobre parâmetros de crescimento e composição corporal. Um total de 192 alevinos (9 a 15g) foram estocados em 24 tanques de cimento, de 100l de capacidade, durante 103 dias. O farelo de canola foi utilizado em quatro proporções: zero; 9,5 por cento; 19 por cento e 38 por cento da dieta, com ou sem farinha de peixe (12 por cento/dieta), totalizando oito tratamentos. A presença de farinha de peixe não afetou os parâmetros de crescimento avaliados. A inclusão de 38 por cento de farelo de canola na dieta diminuiu o ganho de peso dos peixes, valores médios de 28,74g a 50,70g, e piorou a conversão alimentar aparente, de 1,66 para 2,85. A taxa de eficiência protéica também foi menor nos peixes alimentados com 38 por cento de farelo de canola. As várias proporções de farelo de canola das dietas alteraram os teores de umidade, proteína bruta e lipídios dos peixes. A presença da farinha de peixe, nas dietas, somente influiu no teor de lipídios dos peixes alimentados com dietas contendo 9,5 por cento de farelo de canola. Conclui-se que até 19 por cento de farelo de canola pode ser adicionado às dietas de juvenis de pacu, sem que seu desenvolvimento seja prejudicado.


The effects of adding canola meal in the diet of pacu juveniles (Piaractus mesopotamicus) on growth parameters and body composition were evaluated. A total of 192 fingerlings (9 to 15g) were stocked in twenty-four, 100l cement tanks during 103 days. Canola meal was used at four levels (zero, 9.5 percent, 19 percent, and 38 percent of the diet) with (12 percent of the diet) or without fish meal, totalizing eight treatments. The fish meal did not affect the growth parameters. The inclusion of 38 percent canola meal decreased the body weigh gain (mean values of 28.74g to 50.70g) and negatively affected the apparent feed conversion (1.66 to 2.85). The protein efficiency ratio was lower in fish fed the highest level of canola meal. The several levels of canola meal in the diets changed the fish moisture and contents of crude protein and lipids. Fish meal in the diets only influenced the lipid content in fish fed 9.5 percent canola meal. The results suggest that the inclusion of up to 19 percent of canola meal in diets of pacu juveniles did not impair the fish growth.

Turnip rape and oilseed rape 2S albumins are new allergens in children with atopic dermatitis suspected for food allergy. We recently found that 11% (206/1887) of these children had a positive skin prick test to seeds of oilseed rape (Brassica napus) and/or turnip rape (Brassica rapa). In the present case-control study we examined how the children with atopic dermatitis sensitized to turnip rape and oilseed rape had been breast-fed and whether they had some common sensitization pattern to certain foods or pollens. A total of 64 children with atopic dermatitis and a positive skin prick test to turnip rape and/or oilseed rape (>or=5 mm) were examined. Sixty-four age- and sex-matched children with atopic dermatitis but negative skin prick tests to turnip rape and oilseed rape served as case controls. The turnip rape and/or oilseed rape sensitized children with atopic dermatitis had significantly more often positive skin prick tests reactions and IgE antibodies to various foods (cow's milk, egg, wheat, mustard; p < 0.01) and pollens (birch, timothy, mugwort; p < 0.01) than the control children. They had been exclusively breast-fed for a longer period (median 4 months; p < 0.05) and had more often associated asthma (36%) and allergic rhinitis (44%). Children with atopic dermatitis sensitized to oilseed rape and turnip rape had high frequency of associated sensitizations to all foods and pollens tested showing that oilseed plant sensitization affects especially atopic children who have been sensitized to multiple allergens.

A presente pesquisa teve como objetivo estudar o efeito de fontes deácidos graxos poliinsaturados- óleo de canola e/ou semente de linhaça com diferentes concentrações de vitamina E suplementar sobre o teor de produtos de oxidação de ácidos graxos na gema de ovos de galinhas. Utilizou-se 288 galinhas da linhagem Babcock que receberam dietas com 6% de óleo de canola, 20% de semente de linhaça moída ou 3% de óleo de canola e 10% de semente de linhaça moída com teores de 0, 100 e 200 UI/kg de a-tocoferol, para análise estatística dos resultados empregou-se arranjo fatorial 3X3 em blocos casualizados.Os grupos que receberam óleo de canola tiveram menores concentrações de malonaldeído (menor oxidação) na gema dos ovos que aqueles alimentados com linhaça, em ovos crus sem armazenamento e nos cozidos após armazenamento. Os ovos crustiveram as menores concentrações de produtos de oxidação dos ácidos graxos. Os grupos de ovos cozidos, que receberam 100 e 200UI/kgde vitamina E na dieta, tiveram menores teores de malonaldeído na gema dos ovos que os grupos não suplementados.


To investigate the effect of dietary sources of polyunsaturated fattyacids - canola oil and flaxseed – with different vitamin E supplementation upon the fatty acids oxidation produtcts deposition into the eggs. 288 Babcock laying hens were used. Birds were fed diets containing 6% of canola oil, 20% of flaxseed or a combination of 3% of canola oil and 10% of flaxseed, enriched with 0,100 or 200 Ul of dl-a-tocopheril acetate, hens were randomly allocated and the experimental design was a 3X3 factorial arrangement. The inclusion of flaxseed into the diet increased the yolk polyun saturated fatty acids oxidation products, in crude and cooked stored eggs. The concentration of fatty acids oxidation products de crease in crude eggs in all sources of polyunsaturated fatty acids. The cooked eggs without a-tocopherol supplementation in the diet increase the concentration of oxidation products when compared with cooked eggs from diets with 100 or 200UI a-tocopherol in the diet.

Estudou-se o efeito de diferentes fontes de ácidos graxos insaturados (óleo de canola e semente de linhaça), acrescidas de diferentes teores de alfa-tocoferol nas dietas de poedeiras, sobre a composição de ácidos graxos da gema do ovo foi estudado. Foram utilizadas 288 galinhas da linhagem Babcock que receberam dietas com 6 por cento de óleo de canola, 20 por cento de semente de linhaça moída ou 3 por cento de óleo de canola e 10 por cento de linhaça moída com teores de 0, 100 e 200UI/kg de alfa-tocoferol. As dietas com 20 por cento de semente de linhaça proporcionaram teores mais elevados de ácidos graxos poliinsaturados no ovo com aumento, em particular, dos teores de ácido alfa-linolênico e EPA (ácido eicosapentaenóico) e diminuição de ácido araquidônico na gema. Os teores de vitamina E contidos nas rações experimentais não determinaram alteração significativa na deposição dos diferentes ácidos graxos na gema dos ovos, exceto com relação aos ácidos graxos saturados.


The effect of dietary sources of polyunsaturated fatty acids - canola oil and flaxseed - with different vitamin E supplementation on the fatty acid deposition into the eggs of 288 Babcock laying hens was investigated. Birds were fed diets containing 6 percent of canola oil, 20 percent of flaxseed or a combination of 3 percent of canola oil and 10 percent of flaxseed, enriched with 0, 100 or 200Ul of dl-alpha-tocopheril acetate. The inclusion of flaxseed in the diets increased the yolk polyunsaturated fatty acids, mainly alpha-linolenic acid and EPA (eicosapentaenoic acid). The concentration of alpha-tocopherol in the diet did not change the egg yolk, fatty acids deposition but changed the saturated fatty acids deposition.

BACKGROUND: Children with IgE-mediated allergy to foods frequently react to seeds of oilseed rape (Brassica napus ssp. oleifera) and turnip rape (Brassica rapa ssp. oleifera) in skin prick tests (SPTs). Sensitization pathways are not known. OBJECTIVE: We identified possible major allergens in oilseed rape and turnip rape using sera from 72 atopic children (mean age, 3.3 years) with positive SPT responses to oilseed rape and turnip rape. METHODS: Allergens from oilseed rape and turnip rape seed extracts were purified by using gel filtration and cation exchange chromatography and characterized by means of reversed-phase chromatography, N-terminal amino acid sequencing, and matrix-assisted laser desorption ionization time-of-flight mass spectrometry. IgE binding of sera from 72 children with positive SPT reactions to oilseed rape and turnip rape and 72 age- and sex-matched atopic control subjects with negative SPT responses were analyzed by means of IgE ELISA and immunoblotting. In vivo reactivity of the purified allergens was tested with SPTs in 6 children. RESULTS: In IgE immunoblotting and IgE ELISA major reactivity was to a group of homologous, approximately 9.5- to 14.5-kd proteins. These allergens were identified as 2S albumins, also known as napins, by means of N-terminal amino acid sequencing. In ELISA approximately 80% of the patients had IgE to purified napins from both plants. In SPTs purified napins caused positive reactions in all 6 children tested. CONCLUSIONS: This study shows that 2S albumins in oilseed rape and turnip rape are new potential food allergens. Further studies are needed to clarify the routes of exposure and mechanisms of sensitization.

BACKGROUND: When skin prick testing (SPT) young children with atopic dermatitis (AD) for suspected food allergy, we frequently found positive reactions with turnip rape (Brassica rapa) and oilseed rape (Brassica napus). We performed food challenge to examine whether these children react clinically to turnip rape. METHODS: A total of 1887 children were screened with SPTs for sensitization to turnip rape and oilseed rape. Twenty-eight children with clearly positive SPT (> or =5 mm) were first subjected to labial challenge with turnip rape seeds followed, if negative, by open oral challenge for up to 7 days. Twenty-five children with AD but negative SPT to turnip rape and oilseed rape served as controls. RESULTS: Two-hundred and six (10.9%) children had positive SPT to turnip rape and/or oilseed rape. Twenty-five (89%) of 28 children showed a positive challenge reaction to turnip rape. Seventeen reacted with labial whealing, and eight in oral challenge with facial urticaria, flare-up of AD or abdominal symptoms. All 25 control children remained negative in the labial challenge. CONCLUSIONS: Turnip rape and oilseed rape seem to be new important food allergens in young children with AD. The modes of exposure to these allergens and the possible routes of sensitization remain to be established.
