RESUMO

Sibipiruna (Caesalpinia peltophoroides Benth) is a tree of the Brazilian Atlantic Forest. It is a flowering ornamental tree widely planted throughout Brazil and indicated for restoration of degraded areas. We examined protein profile changes in leaves of seedlings of C. peltophoroides grown in nutrient solution under greenhouse conditions, after exposure to cadmium (Cd; 32 mg/L). A two-dimensional gel was used to analyze proteins expressed in response to stress 24 and 72 h after initiation of treatment with Cd. Various protein bands were identified that were related to stress response and/or metabolic adjustments, including proteins involved with resistance to stress, including detoxification, degradation, antioxidant, transport, signal transduction, photosynthesis, electron transport, biosynthesis reactions, and transcription regulation. After 24 h of Cd exposure, the genes of most of these proteins were upregulated. These putative proteins were associated with resistance to stress, including heat shock proteins, heat stress transcriptional factor and other transcriptional factors, aquaporins, glutathione transferase and choline monooxygenase. Most of the putative proteins observed after 72 h of exposure to Cd were downregulated. They were mainly photosynthetic process proteins, such as NAD(P)H-quinone oxidoreductase, photosystem I assembly, and photosystem II CP47 chlorophyll apoprotein. There were also proteins involved with degradation, biosynthesis and antioxidant activity, such as ATP-dependent Clp protease, methylthioribose-1-phosphate and glutathione peroxidase 2. Based on preliminary proteomic analysis, we conclude that proteins related to photosynthetic activity are inhibited, decreasing plant performance under stress conditions and that several proteins related to defense mechanisms are activated, inducing the plant defense response.

An in vitro regeneration protocol has been standardized via direct and indirect methods from excised root explants of C. bonduc, a threatened woody legume used for the treatment of contagious diseases, inflammation, leprosy, antiperiodic, febrifuge, anthelmenthic, urinary disorders, leucorrhoea, piles and to heal wounds. MS medium supplemented with 17.75 micromol BAP and 2.46 micromol IBA, induced a mean of 3.40 +/- 1.07 shoots directly from the surface of excised root explant. Subsequently, the shoots rooted readily on MS half strength medium with out growth regulators. In indirect organogenesis, callogenic frequency was optimized (96.66%) at the concentration of 9.04 micromol 2, 4-D and 0.88 micromol BAP. An average, 15.30 +/- 5.25 shoots were differentiated from the root callus at the concentration of 17.57 micromol BAP and 2.85 micromol IAA. Shoots regenerated through callus were rooted well on MS half strength medium with growth regulators at 2.95 micromol IBA. Rooted plantlets were transferred to the pots containing sterilized soil and were successfully hardened at greenhouse condition for three weeks then exposed to the natural environment. Survival rate was more (95%) in plantlets derived through direct organogenesis than (60%) the plantlets regenerated through root calli.

We intended to establish how efficient the leaf antioxidant responses of C. echinata are against oxidative environmental conditions observed in an urban environment and their relations to growth and biomass parameters. Plants were grown for 15 months in four sites: Congonhas and Pinheiros, affected by pollutants from vehicular emissions; Ibirapuera, affected by high O(3) concentrations; and a greenhouse with filtered air. Fifteen plants were quarterly removed from each site for analysis of antioxidants, growth and biomass. Plants growing in polluted sites showed alterations in their antioxidants. They were shorter, had thicker stems and produced less leaf biomass than plants maintained under filtered air. The fluctuations in the levels of antioxidants were significantly influenced by combined effects of climatic and pollution variables. The higher were the antioxidant responses and the concentrations of pollutant markers of air contamination in each site the slower were the growth and biomass production.

The effects induced by long-term (30 day) and short-term (6h) exposures to ozone on the physiological parameters in young plants of Caesalpinia echinata Lam., a Brazilian tree species, were determined. Potted plants were maintained in open-top chambers in Valencia, Spain, under charcoal filtered air (mean O3 level: 29 microg m3), nonfiltered air (NF; 43 microg m3), and nonfiltered air plus O3 (NF + O3; 68 microg m3), simulating prevailing concentrations observed in the city of São Paulo, Brazil, during spring months (50 microg m3 in 2002). In the plants kept in NF + O3 for 30 days, although no foliar visible injuries were observed, the net carbon assimilation rate was reduced to 50%, stomatal conductance 42%, and transpiration 40%, when compared to the results for the NF plants. No changes in antioxidants, in leaf, stem, and root biomass, and in the root/shoot ratio were observed. Significant reductions were observed in gas exchange and in PSII photochemical efficiency (Fv/Fm) after 6 h of exposure to an O3 peak. The species was shown to be sensitive to ambient O3 concentrations measured in São Paulo.

It has previously been demonstrated that cork tissue increases the efficiency of the production of lipophilic secondary metabolites in diverse plant cell suspension cultures. In the present study, three new homoisoflavonoids--named dihydrobonducellin, 2'-methoxydihydrobonducellin, and 2'-methoxybonducellin--and bonducellin and isobonducellin were isolated from Caesalpinia pulcherrima cultured cells coincubated with cork tissue. Cork tissue increased the production of 2'-methoxybonducellin by about 7-fold relative to control cells, and more than 80% of the product was recoverable from the cork tissue. When cork tissue and methyl jasmonate or yeast extract were added simultaneously to the medium, the amount of 2'-methoxybonducellin produced increased further. The production of the other four homoisoflavonoids was enhanced by variable amounts. Our results indicate that the addition of cork tissue would be an effective technique for investigating formation of secondary metabolites that usually accumulate only in trace amounts.
