RESUMO

Exposure to carbamates has been linked with adverse health effects on developmental period. This study aimed to monitor exposure to carbofuranphenol of pregnant women from Sheyang Birth Cohort and investigate associations between prenatal exposure to carbofuranphenol and birth outcomes. During June 2009 to January 2010, 1100 pregnant women living in Sheyang County participated in our study and donated urine sample. Urinary carbofuranphenol concentration was measured by gas chromatography-tandem mass spectrometry. Associations between urinary carbofuranphenol levels and infant birth outcomes were assessed by generalized linear models. Urinary carbofuranphenol concentrations varied from 0.01 to 395.40µg/L (0.01-303.93µg/g for creatinine adjusted), the geometric mean, median and inter quartile range are 0.81µg/L (1.28µg/g cr), 0.80µg/L (1.23µg/g cr) and 0.27-2.20µg/L (0.47-3.11µg/g cr), respectively. No statistically significant association between maternal urinary carbofuranphenol levels and birth outcomes was found in total infants and female infants. In male neonates, carbofuranphenol level was significantly associated with head circumference (b=-0.226; 95% confidence interval: -0.411, -0.041; P=0.01) and ponderal index (b=0.043, 95% CI: 0.004, 0.083; P=0.03). These findings suggested that the pregnant women were generally exposed to carbofuranphenol and prenatal exposure to carbofuranphenol might have adverse effects on fetal development.

A new gas chromatographic-tandem mass spectrometric method has been developed and validated for the determination of two N-methylcarbamates, carbofuran and carbaryl and their metabolites in applicators' urine specimens. Mild conditions were used for sample preparation based on enzymic hydrolysis and solid-phase extraction using Oasis HLB sorbent cartridges. Amides, phenols and ketones were first converted to volatile derivatives of trifluoroacetic acid anhydride (TFAA) and afterwards were quantitated using tandem mass spectrometry. Linear calibration equations (1-200 ng mL(-1) urine) were obtained from fortified urine samples for all eight compounds, carbaryl, 1-naphthol, 2-naphthol, and carbofuran, 3-hydroxycarbofuran, 7-phenol, carbofuran-3-keto, 3- hydroxycarbofuranphenol. For all compounds, the limit of detection was lower than 0.1 ng mL(-1). Precision for all compounds, at the concentrations of 1, 10 and 100 ng mL(-1) (n = 5) in-fortified urine samples ranged from 0.7% to 18%. Accuracy was calculated at two concentrations 8 and 80 ng mL(-1) (n = 5) and ranged from -8.4% to 8.2%. Relative recoveries at concentrations of 1, 10 and 100 ng mL(-1), ranged from 71% to 116%. The method was successfully applied to five male applicators and 10 non-applicators (including both smokers and non-smokers).

RESUMO

Optimal conditions have been determined for furadan isolation from biological fluids by means of the mixture of solvents ethyl acetate-acetone in volume 1:1. Possible purification of the compound from coextractive substances of biomaterial on the column with silica gel L 100/160 mcm is demonstrated. IR-spectrophotometric methods and high performace liquid chromatography with a detector of photodiode matrix are proposed for identification and quantitation of furadan in blood and urine specimens.

The pharmacokinetics of furathiocarb were studied in vivo in male Sprague-Dawley rats following dermal treatment. HPLC and post-column derivatization were used for the analysis of furathiocarb and its metabolites (carbofuran, 3-hydroxycarbofuran and 3-ketocarbofuran). Carbofuran and 3-hydroxycarbofuran were detected in plasma and urine rather than furathiocarb. 3-Ketocarbofuran, another potential metabolite, was not observed in any sample. The concentration of carbofuran was higher than that of 3-hydroxycarbofuran in plasma, but the reverse was the case in urine. The corresponding area under the plasma concentration-time curve, Tmax, and Cmax values of carbofuran and 3-hydroxycarbofuran for 1500 mg kg-1 doses were 2.4-8.0 mg equiv hml-1, 12 h and 0.1-0.4 mg equiv ml-1, respectively. T1/2 was calculated only for 3-hydroxycarbofuran (28 h). Two metabolites were excreted in a dose-dependent manner without saturation.

This paper investigates the acute effects of carbofuran in workers of two pesticide-formulating plants. Mean airborne carbofuran concentrations ranged from 0.025 to 1.115 mg/m3 in plant A and from 0.018 to 0.067 mg/m3 in plant B, respectively. In workers of plant A the post-shift blood cholinesterase activity was significantly reduced, compared to pre-shift values. No difference in blood cholinesterase activity was found between pre- and post-shift values in workers of plant B. During the investigation, 25 cases of acute carbofuran poisoning were diagnosed by their clinical picture and depressed cholinesterase activity in blood. Usual symptoms included dizziness, weakness, blurred vision, nausea and sweating. Pallor, epigastric pain, vomiting and chest tightness occurred only in a few cases. Myosis was recorded in 24 cases. Fasciculation of muscle gastrocnemius induced by percussion was found in 6 cases, and four of them had also fasciculation of muscle orbicularis oculi. Inhibition of cholinesterase activity in the blood was related with the clinical features; however, the inhibition was rapidly reversible. In most cases, recovery was complete within 2-3 hours, with or without atropine treatment, after the subjects were removed from exposure. Rapid onset, mild illness and quick recovery are typical characteristics of occupational acute carbofuran poisoning.

We measured 12 analytes in urine of 1000 adults living in the United States to establish reference range concentrations for pesticide residues. We frequently found six of these analytes: 2,5-dichlorophenol (in 98% of adults); 2,4-dichlorophenol (in 64%); 1-naphthol (in 86%); 2-naphthol (in 81%); 3,5,6- trichloro-2-pyridinol (in 82%); and pentachlorophenol (in 64%). The 95th percentile concentration (95th PC) for 2,5-dichlorophenol (indicative of p-dichlorobenzene exposure) was 790 micrograms/liter; concentrations ranged up to 8700 micrograms/liter. 2,4-Dichlorophenol concentrations ranged up to 450 micrograms/ liter, and the 95thPC was 64 micrograms/liter. 1-Naphthol and 2-naphthol (indicative of naphthalene exposure) had 95thPCs of 43 and 30 micrograms/liter, respectively; concentrations of 1-naphthol ranged up to 2500 micrograms/liter. Chlorpyrifos exposure was indicated by 3,5,6-tricholoro-2-pyridinol concentrations of 13 (95thPC) and 77 micrograms/liter (maximum observed). Pentachlorophenol had a 95thPC of 8.2 micrograms/liter. Other analytes measured included 4-nitrophenol (in 41%); 2,4,5-trichlorophenol (in 20%); 2,4,6-trichlorophenol (in 9.5%); 2,4-dichlorophenoxyacetic acid (in 12%); 2-isopropoxyphenol (in 6.8%); and 7-carbofuranphenol (in 1.5%). The 95thPCs of these analytes were < 6 micrograms/liter. p-Dichlorobenzene exposure is ubiquitous; naphthalene and chlorpyrifos are also major sources of pesticide exposure. Exposure to chlorpyrifos appears to be increasing. Although pentachlorophenol exposure is frequent, exposure appears to be decreasing. These reference range concentrations provide information about pesticide exposure and serve as a basis against which to compare concentrations in subjects who may have been exposed to pesticides.

Six prairie grain farmers were monitored for pesticide exposure and related adverse effects while they mixed and/or sprayed carbofuran (Furadan 480F) with ground rig application equipment to control grasshoppers in southern Alberta, Canada. Dermal exposure was estimated with Tegaderm patches placed at seventeen locations on the skin beneath the work clothes. Hand and wrist exposure was determined by the amount of chemical found in hand rinses and on wrist patches. Potential inhalation exposure was measured with an air sampler using polyurethane foam as the adsorbent. Urine samples were collected at 24-hr intervals after exposure and monitored for carbofuran. Blood samples were analyzed for acetylcholinesterase (AChE), pseudocholinesterase (ChE) and several other blood parameters. The results indicated that during the mixing and/or spraying operation, a farmer could potentially be exposed to a total of 1,264 micrograms carbofuran per kg of active ingredient (a.i.) used. Of this amount, 1,262 micrograms/kg (or 99.8%) was dermal and 2 micrograms/kg (or 0.2%) could be through the inhalation route. Hand and wrist exposure was about 1,100 micrograms/kg a.i. (or 87% of total exposure). Excretion of the chemical in the urine amounted to 28 micrograms/kg a.i. No ChE inhibition was observed. Other blood measurements were within normal ranges. The farmers showed no acute adverse effects during exposure and for four days after exposure. These results are discussed in relation to the mammalian toxicity of carbofuran.

On-column transesterification with methanol was applied for the gas chromatographic determination of N-methylcarbamates extracted from human urine. Transesterification conversion efficiencies of N-methylcarbamates dioxacarb, carbofuran and OMS-22, calculated from the amount of the on-column produced O-methyl-N-methylcarbamate (DMC), were 96, 77 and 76% with detection limits of 8, 10 and 10 ng, respectively. In the investigated concentration range of 0.2-3 micrograms/ml of urine the extraction efficiencies with methylene chloride were independent of the initial concentration of N-methylcarbamate added to urine samples of non-exposed persons. The recoveries and rel. S.D. were 74 +/- 11, 64 +/- 8 and 79 +/- 12% for dioxacarb, carbofuran and OMS-22, respectively. The procedure was applied for the gas chromatographic determination of carbofuran and its metabolites containing the N-methylcarbamic group extracted from urine samples of occupationally exposed persons in a pesticide formulating plant. The level of extracted N-methylcarbamates and the concentration of degradation products of organophosphorus pesticides detected in the urine of the same persons were correlated with the blood and plasma cholinesterase activities. Although the determination of DMC includes only a smaller part of the excreted N-methylcarbamate, a simultaneous determination of both carbamates and organophosphorus residues made it possible to distinguish the cause of depression in cholinesterase activity, indicating early and specifically the exposure to a particular group of agents hazardous to health.
