RESUMO

Neospora caninum is an apicomplexan parasite with worldwide distribution, which primarily causes disease in dogs and cattle. It has been recognized as an important cause of epidemic and endemic abortion in cattle around the world. There is no study to demonstrate the country level picture of the disease in Iran and estimate the extent of the problem, quantitatively. Therefore, the aim of the present study was to systematically review the observational studies on bovine neosporosis in Iran and estimate an overall serologic prevalence of the infection in the cattle population. Search for all published reports on N. caninum in Iran was conducted from inception until September 2018. Major English (PubMed, Scopus and Science Direct) and Persian (IranDoc, SID, Iranmedex, and Magiran) electronic database sources were used for this study. After removing duplicated articles, 128 documents remained. Unrelated articles (89) were excluded after primary screening and 39 articles were evaluated for eligibility. Finally, 37 studies from 33 articles were recognized as eligible and were included in the present work. The author screened the studies and extracted the data twice, carefully. Based on the meta-analysis, the overall serologic prevalence of N. caninum in 10,967 cattle with 2619 positive ones was 23.6 % with 95 % CI: 19.8-27.9 %. The random effect model was used for meta-analysis because the Cochran Q value (795.10, df = 36, P < 0.001) and Higgins index (I2 = 95.47 %) showed statistically significant heterogeneity. In subgroup meta-analysis, abortion status of the animal and geographical location of the study showed significant association with serologic prevalence, while age and breed did not have significant associations. No publication bias was detected, and sensitivity analysis showed stability of the overall estimate after excluding any individual study. It could be concluded that N. caninum is widespread in cattle population in Iran and the time is right to implement national level educational and control programs for it.

Infectious animal diseases caused by pathogenic microorganisms such as bacteria and viruses threaten the health and well-being of wildlife, livestock, and human populations, limit productivity and increase significantly economic losses to each sector. The pathogen detection is an important step for the diagnostics, successful treatment of animal infection diseases and control management in farms and field conditions. Current techniques employed to diagnose pathogens in livestock and poultry include classical plate-based methods and conventional biochemical methods as enzyme-linked immunosorbent assays (ELISA). These methods are time-consuming and frequently incapable to distinguish between low and highly pathogenic strains. Molecular techniques such as polymerase chain reaction (PCR) and real time PCR (RT-PCR) have also been proposed to be used to diagnose and identify relevant infectious disease in animals. However these DNA-based methodologies need isolated genetic materials and sophisticated instruments, being not suitable for in field analysis. Consequently, there is strong interest for developing new swift point-of-care biosensing systems for early detection of animal diseases with high sensitivity and specificity. In this review, we provide an overview of the innovative biosensing systems that can be applied for livestock pathogen detection. Different sensing strategies based on DNA receptors, glycan, aptamers and antibodies are presented. Besides devices still at development level some are validated according to standards of the World Organization for Animal Health and are commercially available. Especially, paper-based platforms proposed as an affordable, rapid and easy to perform sensing systems for implementation in field condition are included in this review.

A 2-month-old juvenile central bearded dragon was presented for anorexia and cachexia. Another specimen from the same cage had died suddenly 2 weeks prior. Fecal analysis revealed a high quantity of Isospora amphiboluri and a few pinworm eggs. Other examinations were not performed and the animal died a few days later despite supportive care. A third individual from the same cage presented with anorexia and a distended cœlom and was euthanized. In this third dragon, histological examination revealed intestinal coccidiosis, basophilic intranuclear inclusions compatible with adenovirus infection, acute hepatic necrosis with intrahepatocytic and intraenteritic organisms typical of microsporidia and renal gout. A PCR confirmed the diagnosis of adenovirosis. Sequencing showed that the PCR product was 100% identical to the corresponding portion of the agamid adenovirus 1 genome. A PCR for the detection of Encephalitozoon (E.) cuniculi was positive. Partial sequencing revealed 100% identity to an E. cuniculi-like organism previously found in bearded dragons. In cases where environmental factors such as poor hygiene or stress can be excluded, the presence of opportunistic pathogens in high numbers can be due to a systemic (viral) infection with temporary immunosuppression.

BACKGROUND: Bovine besnoitiosis, caused by the apicomplexan Besnoitia besnoiti, is a chronic and debilitating disease considered as emerging in Europe. In Spain, Portugal and France it is endemic and foci of infection were recorded in Germany, Switzerland, Hungary, Greece and Italy. In Italy, cases of bovine besnoitiosis were registered both in imported and autochthonous cattle, and mostly in central regions; high seroprevalence was also revealed by an epidemiological survey performed in the southern part of the country. Aiming to update information on the disease in northwestern and insular areas of Italy, where data on bovine besnoitiosis were missing, a serosurvey was designed for the present study. METHODS: Three thousand one hundred and forty bovine blood samples from both dairy and beef farms (n = 126) were collected in northwestern regions (Lombardy, Piedmont and Liguria) and in the island of Sardinia. Samples were analyzed by a standardized in-house ELISA and those resulted positive were re-tested by Western Blot (WB) for confirmation. On results obtained by both ELISA and WB, apparent (AP) and true prevalence (TP) were calculated at individual and herd levels. Further, a panel of sera resulted positive to ELISA was analyzed by IFAT. RESULTS: A total of 712 animals (AP = 22.7%; TP = 18.8%) and 109 farms (AP = 86.5%; TP = 88.2%) showed a positive reaction in ELISA. Only ten (AP = 0.3%; TP = 0%) specimens proceeding from five farms (AP = 3.9%; TP = 1.7%) from Lombardy were confirmed positive to the WB, corresponding to two Holstein Friesian cows and eight beef cattle. IFAT showed a low sensitivity (44.4%) scoring positive in only four samples out of 9 positive to WB. CONCLUSIONS: The survey demonstrated that bovine besnoitiosis cannot still be considered endemic in whole Italy. In fact, independent foci of infection were registered only in Lombardy region. Therefore, a sanitary strategy aimed to increase control measures and to organize monitoring plans, by adequate diagnostic tools is necessary to avoid overestimation of B. besnoiti in Italy.

A crude resinous extract from Commiphora swynnertonii was tested against an experimental coccidial infection in local chickens. A total of 80 growing chickens were randomly assigned into five groups, which received different treatments. Chickens in G1 were not infected with coccidian oocysts and therefore served as a negative control. All chickens in G2, G3, G4 and G5 were infected through oral administration of coccidian oocysts suspension at a dosed rate of 1.5 × 10(4) Eimeria spp. oocysts per bird. Starting from day 3 post-infection (p.i), chickens in different groups were treated for 7 consecutive days as follows: G1 and G2 (positive control) received 5 ml of normal saline as placebo, G3 and G4 were given the extract at 400 and 800 mg/kg bodyweight whereas G5 received anticoccidial drug. Clinical signs, bodyweights, oocysts counts and mortality rates were observed regularly. Results showed that oral administration of the resinous extract to chickens with coccidiosis significantly reduced mortality rate from 94 to 25 % and oocysts counts from 1.03 × 10(5) to 6.55 × 10(3) oocysts/g faeces (p < 0.05). Also a body condition score chart indicated less severe clinical signs of the disease in the groups which received the extract. Mean daily body weights were slightly reduced by the administration of the extract but this effect disappeared by day 7 p.i. These findings clearly indicate that resinous extract from C. swynnertonii has significant anticoccidial effect against experimental Eimeria spp. infection in chickens. A larger field trial to validate the use of the extract in chickens naturally infected with Eimeria spp. is required.

Neospora caninum is a canine parasite which is considered a significant cause of bovine abortion. Two cattle herd groups were serologically studied with the objective of studying the prevalence of infection by N. caninum associated with BHV1 and BVDV infections. In group I, 15 dairy herds (476 samples) naturally infected by the three infectious agents were analyzed,. In group II, three dairy herds (100 samples) of cows vaccinated for two viruses were analyzed, in order to determine the infection prevalence by N. caninum. In the first group, an infection prevalence of 12.61, 34 and 28.3% was determined for N. caninum BHV1 and BVDV, respectively. In the second group, a seropositive prevalence of 46, 85 and 76%, respectively, was determined for N. caninum, BVH1 and BVDV. In the first group, the virus and N. caninum had shown in the first group 4.41% positive samples in association with BVH1, 3.15% with BVDV, and 8.41% with BVH1 and BVDV.
