RESUMO

Through resources investigation and sample collection,a total number of 392 Dendrobium officinale from 38 different populations,9 provinces were processed for measuring and observing.Fourteen agronomy characterizations like stem height,stem diameter,number of node in stem were selected for further classification.The cluster analysis was performed using Ward and Euclidean method.The results showed that the threshold of genetic distance was 2.77.Thirtyeight populations were divided into 6 groups.The factor analysis showed that,the leaf shape,stem shape,pitch length and leaf color were very important factors for classification.This study establish the foundation for analyzing the genetic relationship of D.officinale from different populations.

Dendrobium is an important medicinal material in China. It has the effect of nourishing the stomach, nourishing yin and clearing heat. In China, there are many types of Dendrobium, and different Dendrobium species have different efficacy. The present study is aimed at distinguishing three major Dendrobium species from morphological and microscopic identification in Ta-pieh mountains area. In this article, the roots, stems and leaves of Dendrobium huoshanense, Dendrobium officinale, and Dendrobium moniliforme are used as materials to compare the differences of tissues of these three Dendrobium species by morphological indexes and microscopic identification of different Dendrobium. The stem morphology of these three Dendrobium species was significantly different except for stem internodes number and the middle part of the stem diameter by measuring the stems of Dendrobium. To ensure the safe use of Dendrobium, we built a fast and convenient method combining normal and fluorescence microscopy was applied in the present study to distinguish D. huoshanense, D. officinale, and D. moniliforme. The microscopic results show that different types of Dendrobium exhibit different states that can be distinguished under normal light normal and fluorescence microscopy. This comparative study of morphology and microscopy contributes to the development of identification and quality evaluation of Dendrobium.

Shihu is a famous Chinese Materia Medica derived from species of the Dendrobium. In order to differentiate 24 Dendrobium species, microscopic features of the 24 Dendrobium species were systemically observed, including the trend of vascular bundle number with different internodes and the transverse section characteristics of stem. The results indicate that the thickness of cuticle, the level of thickening of epidermal cells and cell types, the number of vascular bundles, the shape of vascular bundle sheath, the type and distribution of calcium oxalate crystals, the presence or absence of silica masses can be used to authenticate the 24 Dendrobium species. In addition, the number of vascular bundles in stems of 24 Dendrobium species also have changed at different internodes, and showed a trend of first increasing and then decreasing. For some specific species, the vascular bundle numbers had typical differences, which can provide some support for the identification of Dendrobium plants. Thus, using the multiple microscopic characteristics of stems can identify the Dendrobium plants effectively and quickly.

"Jinchai Shihu" were called Jinchai and recoded in "Taishang Zhouhou Yujingfang" of Tang Dynasty, which first clearly documented the name of Shihu in complex Dendrobium medicines and were condiered as superior medicinal articles. Morphological features are one of the naming principles for Chinese medicines. In this paper, botanical origin plants under the names of "Jinchai" and "Jinchai Shihu" were investigated. Based on documents from the local Chronicles and historical accounts, the Chinese characters of Jinchai have the distinctive features of gold color and two hair clasps. Moreover, the hair clasps are usually cylindrical in shape with uniform thickness in middle and upper part, and tapers off to the foot. And its bottom part style is simple and head part is complex. Thus we speculated the herbal "Jinchai" and "Jinchai Shihu" should have similar morphologic features as Chinese characters of Jinchai, including golden color and hairpin shape of stems without braches, short and solid sterm. After comparing the dried vegetative morphology of 10 common medicinal Dendrobium species, we suggested that of Dendrobium flexicaule matches well with the morphological features from historical herbal records.

The preliminary studies have shown that Dendrobium officinale possessed therapeutic effects on hypertension and atherosclerosis. Studies also reported that Dendrobium officinale polysaccharides showed antioxidant capabilities. However, little is known about its effects on myocardial cells under oxidative stress. The present study was designed to study the protective effect of Dendrobium officinale polysaccharides against H2O2-induced oxidative stress in H9c2 cells. MTT assay was carried out to determine the cell viability of H9c2 cells when pretreated with Dendrobium officinale polysaccharides. Fluorescent microscopy measurements were performed for evaluating the apoptosis in H9c2 cells. Furthermore, effects of Dendrobium officinale polysaccharides on the activities of antioxidative indicators (malondialdehyde, superoxide dismutase), reactive oxygen species (ROS) production and mitochondrial membrane potential (MMP) levels were analyzed. Dendrobium officinale polysaccharides attenuated H2O2-induced cell death, as determined by the MTT assay. Dendrobium officinale polysaccharides decreased malondialdehyde levels, increased superoxide dismutase activities, and inhibited the generation of intracellular ROS. Moreover, pretreatment with Dendrobium officinale polysaccharides also inhibited apoptosis and increased the MMP levels in H9c2 cells. These results suggested the protective effects of Dendrobium officinale polysaccharides against H2O2-induced injury in H9c2 cells. The results also indicated the anti-oxidative capability of Dendrobium officinale polysaccharides.

"Huoshan shihu," derived from Dendrobium huoshanense, is a well-known, valuable, and rare Daodi herb. Because of its higher price and scarce resources, its related species D. officinale and D. moniliforme are usually presented as "Huoshan shihu" for sale. Fengdou, the processed form of D. huoshanense, its identification is particularly important. To effectively identify Fengdou of D. huoshanense and protect the Daodi herb, the morphological and microscopic characteristics of the stems of three Dendrobium species were examined. The results showed that macroscopic and microscopic features helpful for identification of the three species were the total lengths and internode numbers of stems, diameter and length of each internode, number of vascular bundles, and presence or absence of silica masses. The key features useful for distinguishing between D. huoshanense and its related species, and also between their Fengdou, were the stem lengths and the trends of change in diameter in different internodes. The results of the study indicated that a combination of macroscopic and microscopic identification techniques might conveniently and effectively be applied for identification of Dendrobium species. "Longtou Fengwei" is the main feature of the Daodi herb "Huoshan shihu," (D. huoshanense Fengdou) and reflects the wisdom behind the protection of Daodi herbs in ancient times.

The Chinese Materia Medica, Tiepishihu, used as a tonic for over one thousand years, is a well-known precious medicine in China. According to the Chinese Pharmacopoeia, its source is the species Dendrobium officinale Kimura et Migo, which is distinguished from other species in Dendrobium genus. However, these species from the same genus are similar with Tiepishihu and caused confusion in the market. To find a quick and simple method to distinguish Tiepishihu from other similar species, histologic and microscopic methods were combined together to investigate the transverse section of stem of Tiepishihu and other similar species. Phloroglucinol test solution with hydrochloric acid was used to reveal the lignified tissue by staining the transverse section of Tiepishihu and similar species. Results revealed the unique identification characteristics to distinguish Tiepishihu from similar species, which were difficult to distinguish by other methods. The identification characteristics of Tiepishihu include the cells of vascular bundle sheath were stained red, parenchyma cells were not stained red. What's more, other species can be distinguished from each other with microscopic and histological characteristics. These characteristics proved stable and can be easily observed by normal light microscopic examination. This method is rapid, accurate, stable, and inexpensive.

The stems of Dendrobium officinale Kimura et Migo, named Tie-pi-shi-hu, is one of the most endangered and precious species in China. Because of its various pharmacodynamic effects, D. officinale is widely recognized as a high-quality health food in China and other countries in south and south-east Asia. With the rising interest of D. officinale, its products have a high price due to a limited supply. This high price has led to the proliferation of adulterants in the market. To ensure the safe use of D. officinale, a fast and convenient method combining normal and fluorescence microscopy was applied in the present study to distinguish D. officinale from three commonly used adulterants including Zi-pi-shi-hu (D. devonianum), Shui-cao-shi-hu (D. aphyllum), Guang-jie-shi-hu (D. gratiosissimum). The result demonstrated that D. officinale could be identified by the characteristic "two hat-shaped" vascular bundle sheath observed under the fluorescence microscopy and the distribution of raphides under normal light microscopy. The other three adulterants could be discriminated by the vascular bundle differences and the distribution of raphides under normal light microscopy. This work indicated that combination of normal light and fluorescence microscopy is a fast and efficient technique to scientifically distinguish D. officinale from the commonly confused species.

Dendrobium is one of the three largest genera and presents some of the most intricate taxonomic problems in the family Orchidaceae. Based on five DNA markers and a broad sampling of Dendrobium and its relatives from mainland Asia (109 species), our results indicate that mainland Asia Dendrobium is divided into eight clades (with two unplaced species) that form polytomies along the spine of the cladogram. Both Dendrobium and Epigeneium are well supported as monophyletic, whereas sect. Dendrobium, sect. Densiflora, sect. Breviflores, sect. Holochrysa, are paraphyletic/polyphyletic. Many ignored phylogenetic relationships, such as the one of major clades formed by D. jenkinsii and D. lindleyi (two members of sect. Densiflora), the Aphyllum group, the Devonianum group, the Catenatum group, the Crepidatum group, and the Dendrobium moniliforme complex are well supported by both molecular and morphological evidence. Based on our data, we propose to broaden sect. Dendrobium to include sect. Stuposa, sect. Breviflores, and sect. Holochrysa and to establish a new section to accommodate D. jenkinsii and D. lindleyi. Our results indicated that it is preferable to use a broad generic concept of Dendrobium and to pursue an improved infrageneric classification at sectional level, taking into account both morphology and current molecular findings.

OBJECTIVE: In order to provide theoretical and technological basis for the germplasm innovation and variety breeding in Dendrobium officinale, a study of the correlation between polysaccharide content and agronomic characters was conducted. METHODS: Based on the polysaccharide content determination and the agronomic characters investigation of 30 copies (110 individual plants) of Dendrobium officinale germplasm resources, the correlation between polysaccharide content and agronomic characters was analyzed via path and correlation analysis. RESULTS: Correlation analysis results showed that there was a significant negative correlation between average spacing and polysaccharide content, the correlation coefficient was -0.695. And the blade thickness was positively correlated with the polysaccharide content, but the correlation was not significant. The path analysis results showed that the stem length was the maximum influence factor to the polysaccharide, and it was positive effect, the direct path coefficient was 1.568. CONCLUSION: According to thess results, the polysaccharide content can be easily and intuitively estimated by the agronomic characters investigating data in the germpalsm resources screening and variety breeding. Therefore, it is a visual and practical technology guidance in quality variety breeding of Dendrobium officinale.

OBJECTIVE: To establish the seedling quality grading standard of Dendrobium nobile. METHODS: Height, stem thickness and stem nodes of seedlings (test-tube seedling and cutting seedling) from Chishui city Guizhou province were tested. Through statistical analysis, the key indicators for seedling quality grading were defined. RESULTS: Height and the number of cluster were the primary indicator of test-tube seedling; Height and the number of buds were the primary indicator of cutting seedling. CONCLUSION: The grade test-tube seedling quality of each grade should reach the following requirements: for the first grade seedlings height > or = 17cm, the numbers of cluster > or = 3; For the second seedling height 11 -17cm, the numbers of cluster > or =2. The grade cutting seedling requires the first grade seedlings height > or = 12cm, the numbers of buds > or = 3; The second grade seedling height 6 - 12cm, the numbers of cluster > or = 2. Seedlings those can hot reach the second grade requirements are defined as unqualified seedlings.

RESUMO

The water-soluble crude polysaccharide (DDP) obtained from the aqueous extracts of the stem of Dendrobium denneanum through hot water extraction followed by ethanol precipitation, was found to have an average molecular weight (Mw) of about 484.7 kDa. Monosaccharide analysis revealed that DDP was composed of arabinose, xylose, mannose, glucose and galactose in a molar ratio of 1.00:2.66:8.92:34.20:10.16. The investigation of antioxidant activity both in vitro and in vivo showed that DDP is a potential antioxidant.

To provide theoretical basis for breeding good variety of Dendrobium denneanum, agronomic and qualitative characters of 4 different lines and relationships among them were studied. The stem length, stem diameter, leaf length, leaf width, length/ width ratio and leaf area were measured. The single fresh and dry stem was weighed and drying rate was calculated. The contents of polysaccharides and total alkaloids were determined by sulfuric acid-phenol colorimetry and acid-dye colorimetry, respectively. The correlations between characters were analyzed. The results showed that differences in major agronomic characters between four lines were significant. The plant types of dq-1 and dq-2 were higher, dq-3 was medium and dq-4 was lower. The fresh weigh of stem and content of polysaccharides were the highest in dq-2, 7.81 g and 14.33%. While the highest content of total alkaloids and was 0. 486% in dq-3. There were significant correlations between agronomic characters, but these characters had low or non correlations with qualitative characters such as polysaccharides and total alkaloids. It was shown that the content of polysaccharides and total alkaloids were significantly different among 4 lines of D. denneanum, which could be selected for different uses.

Seed morphology of 17 Dendrobium species from Yunnan were observed under light microscope and environment scanning electron microscope and 16 of them were reported first time. All seeds were yellow and fusiform, but big differences existed in sizes of seeds and embryos, which were not significantly related to the division of sections under Dendrobium. Dendrobium seed consisted of embryo and testa with flocs on its cell walls. The density and length of flocs were different among the sections under Dendrobium.

The morphological characters and the polysaccharides content of Dendrobiam loddigesii from different habits were studied in this paper. The results showed that there were remarkable differences in morphology from five areas, such as stem height and diameter, laminae sizes and figures, amount of lateral bud, and texture of stem. The content of polysaccharides from sample of Xingyi and Anlong, Guizhou Province, respectively 22. 62% and 20. 96% , were higher than that of other areas. It can be preferentially choosed as good materal on commareial production.

OBJECTIVE: To compare the tissue formation and the content of polysaccharide between the wild Dendrobium candidum and the cultured ones and to find any existed differences. METHOD: Bare-handed microtomy and photomicrography; The content of polysaccharide is determined by phenol-sulphuric acid method. RESULT AND CONCLUSION: There are no marked noticeable differences between the wild D. candidum and the cultured ones in terms of the tissure formation and the content of polysaccharide.
