RESUMO

This study described the hepatobiliary anatomopathological lesions associated with trematode Platynosomum illiciens parasitism in Neotropical primates kept in captivity. In the evaluated organs, we observed portal fibrosis, biliary epithelial hyperplasia, and inflammatory reaction with a predominance of lymphocytes and plasmocytes, and in some cases infiltration of eosinophils and neutrophils.

The Dicrocoeliidae is a highly diverse and broadly distributed family of digeneans typically parasitic in the gall bladder and liver of their tetrapod hosts. So far, no study has reported dicrocoeliids, or any digeneans, from the ancient marsupial family Caenolestidae. Herein, we describe a new genus of dicrocoeliids (Otongatrema n. gen.) from Tate's shrew opossum Caenolestes fuliginosus and a new species of Metadelphis (Metadelphis cesartapiai n. sp.) from a phyllostomid bat Anoura peruana collected in Ecuador. Otongatrema can be readily distinguished from the morphologically closest dicrocoeliid genera Concinnum, Conspicuum and Canaania based on the position of the genital pore, distribution/position of the uterus and extent of vitellarium. Metadelphis cesartapiai can be easily differentiated from other Metadelphis spp. based on a combination of morphological characters including body shape and size, distribution of vitellarium, shape of the gonads as well as size of suckers and cirrus sac. In addition, we used newly generated partial sequences of the nuclear 28S rRNA gene and mitochondrial cox1 genes to examine phylogenetic affinities of the new taxa within the Dicrocoeliidae. Both the 28S and cox1 phylogenies confidently positioned Otongatrema as a sister/basal group to all other dicrocoeliids sequenced so far. The phylogenetic position of Otongatrema may be explained by a close co-evolutionary relationship with Caenolestidae, one of the most basal and most ancient groups of marsupials. In addition, our 28S phylogeny provides evidence that the complete or partial loss of intestinal structures has likely occurred independently at least 3 times in the evolutionary history of the Dicrocoeliidae.

RESUMO

Metadelphis tkachi n. sp. is described based on material from the gall-bladder of Molossus molossus (Pallas) (Chiroptera: Molossidae) in the Brazilian eastern Amazon. The main differentiating characters of the new species include lobed, tandem testes and elongated clusters of vitelline follicles commencing at the level of the seminal receptacle. The new species can be distinguished from Metadelphis lenti (Santos & Gibson, 1998) and Metadelphis apharyngotrema (Marshall & Miller, 1979) by the shape and position of the testes; from Metadelphis evandroi Travassos, 1944 by having tandem, lobed testes and shorter clusters of vitelline follicles; from Metadelphis compactus Travassos, 1955 by having tandem testes and more elongated clusters of vitelline follicles, and from Metadelphis alverangai Travassos, 1955 by having smaller testes and body, and vitellarium with large follicles. New morphological data are reported for M. lenti based on light and scanning electron microscopy; intraspecific variation was observed.

The enzyme adenosine deaminase (ADA) is critical for modulating the immune system, and in the presence of zinc, its activity is catalyzed. The aim of this study was to evaluate the ADA activity in pancreas of cattle naturally infected by Eurytrema coelomaticum in relation to the results of zinc levels, pathological findings and parasite load. For this study 51 slaughtered cattle were used. The animals were divided into two groups: Group A consisting of animals naturally infected by E. coelomaticum (n=33) and Group B of uninfected animals (n=18). Blood and pancreas were collected of each animal for analysis of zinc and ADA, respectively. Infected cattle showed a reduction on seric levels of zinc, and decreased ADA activity in the pancreas (P>0.05). A positive correlation between zinc levels and ADA activity was observed. Thus, high parasite load and severity of histopathologic lesions affect the ADA activity in pancreas, as well as the zinc levels in serum of infected animals (negative correlation between these variables). Therefore, we can conclude that cattle infected by E. coelomaticum have low ADA activity in pancreas, which can be directly related to zinc reduction, responsible for ADA activation and catalyzes.

The genus Brachylecithum was for the first time subject to molecular taxonomic phylogenetic analysis in order to ascertain relationships among its component taxa. We used two markers-the nuclear ribosomal 28S ribosomal DNA (rDNA) gene and the mitochondrial cox1 gene, for six species of the genus; 11 sequences of partial 28S rDNA and partial cox1 were obtained from adult B. capilliformis, B. glareoli, B. kakea, B. laniicola, B. lobatum, and B. strigis, and from larval stages obtained from snails of the genus Cepaea. We propose to synonymize B. strigis with B. lobatum, while the genetic differences in the 28S rDNA gene and mitochondrial cox1 gene confirm the species status of B. capilliformis and indicate a distinct group within Brachylecithum, including B. kakea and B. laniicola. Cercarial and metacercarial isolates from the snails showed 100 % similarity to B. lobatum; thus, it is the first record of Cepaea snails as intermediate hosts of this species and the first report on life cycle abbreviation within the Dicrocoeliidae.

This study investigated the occurrence of Eurytrema spp. in cattle by analysis of the partial 18S rRNA gene sequence. Trematodes from 44 bovine pancreas were collected and classified based on typical morphological features. PCR assay and sequence analyses of amplified products confirmed that the trematodes classified as Eurytrema coelomaticum were phylogenetically distinct from those identified as E. pancreaticum. The results of this study represent the first molecular characterization of E. coelomaticum within the Americas, and provide an efficient method to differentiate digenean trematodes of domestic animals.

The present work used Plackett-Burman experimental design to assess the influence of enzymes of nematophagous fungi versus Strongyloides westeri and trichostrongylides larvae and Platynosomum fastosum eggs. The variables studied in the Plackett-Burman design were the proteases and chitinases of AC001 or VC4 as destructive agents of S. westeri and trichostrongylides larvae, and P. fastosum eggs. All tested enzymes had a significant effect (P < 0.05) on the destruction of S. westeri larvae. Furthermore, only VC4 and AC001 proteases showed a significant effect (P < 0.05) on the destruction of trichostrongylides larvae. On the other hand, chitinases of VC4 showed the highest significance (P < 0.05) on the destruction of P. fastosum eggs. It is proposed that statistical planning for the use of enzymes derived from nematophagous fungi is a viable way to elucidate some questions about their mechanism of action.

Este estudo avaliou a ocorrência de Eurytrema spp. em bovinos por meio da análise da sequência parcial do gene 18S rRNA. Trematódeos procedentes de 44 pâncreas bovinos foram coletados e classificados com base em aspectos morfológicos típicos. A técnica de PCR e a análise da sequência de nucleotídeos dos produtos amplificados confirmaram que os trematódeos classificados como Eurytrema coelomaticum eram filogeneticamente distintos daqueles identificados como E. pancreaticum. Os resultados deste estudo representam a primeira caracterização molecular de E. coelomaticum nas Américas e disponibiliza um método eficiente para diferenciar trematódeos digenéticos em animais domésticos.

OBJECTIVE: The aim of this study was to investigate the prevalence of Dicrocoeliid larval stages in Helix lucorum Linnaeus, 1758 (Mollusca: Pulmonata) living in the vicinity of Kastamonu. METHODS: Snails were collected in the vicinity of Tasköprü, Kastamonu in March, April, May 2013. They were dissected while alive and their hepatopankreas were removed and placed on clean glass slide with a drop of 0.6% NaCl solution. The hepatopankreas were incised with a mounted needle and a thin film of the hepatopankreas fluid was drown out on a slide for examination of live parasites. After examination, larval stages fixed in %70 alcohol, formaldehyde-ethyl alcohol, 10% formalin and bouin. RESULTS: H. lucorum which is a land snail species has been reported for the first time being an intermediate host in the life cycle of Dicrocoeliid (Trematoda: Digenea) species with the prevalence of 27.6% from our country. Morphological and histological features of larval stages were determined. CONCLUSION: H. lucorum is an intermediate host in the life cycle of Dicrocoeliid species. The prevalence of infection was highest in April. This land snail species is important for the health of animal and human.

Platynosomum fastosum is a small hepatic trematode found in the biliary ducts and gall bladder of cats and other mammals. It is commonly found in tropical and subtropical regions. Some aspect of the life cycle of this parasite is not fully understood, however terrestrial snails, lizards and isopods are implicated as intermediate/paratenic hosts. The disease caused by P. fastosum is platynosomiais (named after the parasite) or 'lizard poisoning' since it is assumed that affected cats acquire the parasite by eating infected lizards. The clinical signs due to infection with P. fastosum may range from asymptomatic to progressive disease and at times death due to biliary tract obstruction and hepatic failure. Infection with this parasite should, therefore, be included in the differential diagnosis of cats with signs of hepatic diseases.

RESUMO

A review of the fauna of trematodes from the family Dicrocoeliidae parasitizing animals in Vietnam has been conducted. According to original and literature data, 57 trematode species from 15 genera were recorded. Data on hosts, localization, and distribution of all these species in the territory of Vietnam are given.

Durante o estudo dos metazoários parasitos de Paroaria dominicana Linnaeus, 1758 (Passeriformes), quatro infrapopulações de Platynosomum illiciens (Brown, 1901) (Trematoda) foram coletadas nos ductos hepáticos, com intensidades parasitárias de 10, 11, 18 e 36 espécimes respectivamente. Diferenças entre os valores morfométricos médios e o número total de ovos das infrapopulações foram estatisticamente testadas. O peso do fígado não se correlacionou ao peso das aves, ao comprimento do corpo das aves e à abundância parasitária. Foram detectadas correlações positivas entre o tamanho das infrapopulações e o número de ovos produzidos e entre o tamanho do corpo dos parasitos e o tamanho dos ovos. Correlações negativas foram observadas entre a intensidade parasitária e o peso do fígado; entre o tamanho das infrapopulações e o tamanho dos indivíduos dessas infrapopulações; entre o número e o tamanho dos ovos e entre o tamanho do corpo dos parasitos e o número de ovos. Os resultados indicaram que as infrapopulações maiores se estabeleceram em aves com fígados menores e que quanto maiores as infrapopulações foram observados menor tamanho corporal e menor tamanho dos ovos, os que se apresentaram em maior quantidade.

The efficacy and tolerance of the 80 microg/ml praziquantel (PZQ) and 40 microg/ml triclabendazole (TCZ) against adult stage Eurytrema pancreaticum in vitro were investigated at 3, 12, and 15 h incubation. Motility of the flukes and histopathological changes were studied. Sudden paralysis and death were observed after exposed to PZQ as early as 3h incubation. In contrast, the TCZ treated flukes showed active mobility at all intervals. By light microscopic examination, severe damages in various organs such as tegument, muscle, and testes were observed early at 12h incubation of these drugs. PZQ caused more severe damage to flukes than TCZ. There were vigorous contraction of musculature, progressive shrinkage of circular and longitudinal muscles, vacuolization and disintegration of the tegument disrupting the worms' outer surface including detachment of spines in the PZQ treatment. The cells in testes were slightly increased in size and followed by degeneration leaving several hollow spaces. The uterus and vitelline glands remained unaffected. The direct observation of the fluke motility and light microscopic study highly suggested that PZQ was more effective than TCZ treatment for the eurytremiasis infection.
