RESUMO

Escherichia coli and Salmonella Typhimurium are the main pathogens of diarrhea in weaned piglets. The prevention of bacterial diarrhea in weaned piglets by phage is rarely reported. We conducted this study to evaluate the preventive effect of phages on mixed Escherichia coli and Salmonella Typhimurium infections in weaned piglets. A novel phage named NJ12 was isolated by using Salmonella Typhimurium SM022 as host bacteria and characterized by electron microscopy, genomic analysis and in vitro bacteriostatic activity. Phage NJ12 and a previously reported phage EP01 were microencapsulated with sodium alginate to make phage cocktail. Microencapsulated phage cocktail and PBS (Phosphate buffer solution) were used to piglets the phage and phage-free group through oral administration before bacterial infection 2 h, respectively. Piglets of the phage and phage-free group were consumed with feed contaminated with 6 mL (108CFU/mL) Escherichia coli O157:H7 GN07 (GXEC-N07) and 6 mL (108CFU/mL) SM022 every day for seven consecutive days. The results showed that piglets in the phage-free group had more severe diarrhea, larger decreased average weight gain and higher levels of neutrophils compared with piglets in phage group. Meanwhile, piglets in the phage-free group had higher load of SM022 and GN07 in jejunal tissue and more severe intestinal damage compared with piglets in group phage in vivo. In addition, oral administration phage can significant decreased the relative abundance of Enterobacteriaceae but hardly repaired the changes of diversity and composition of gut microbiota caused by the mixed infection of SM022 and GN07. This implies that phage used as a feed additive have a marvelous preventive effect on bacterial diarrhea during weaning of piglets.

Swine dysentery (SD) caused by pathogenic Brachyspira spp. is an economic challenge for the swine industry. In research settings, experimental reproduction of swine dysentery typically relies on intragastric inoculation which has shown variable success. This project aimed to improve the consistency of the experimental inoculation protocol used for swine dysentery in our laboratory. Over six experiments, we evaluated the influence of group housing in inoculated pigs using a frozen-thawed broth culture of strongly hemolytic B. hyodysenteriae strain D19 (Trial A), compared the relative virulence of B. hyodysenteriae strains D19 and G44 (Trial B), compared inoculum volumes (50 mL vs 100 mL) for G44 and B. hampsonii 30446 (Trial C), and performed three independent trials evaluating intragastric inoculation using different oral inoculation methods: oral feed balls (Trial D), and oral syringe bolus of 100 mL (Trial E) or 300 mL (Trial F). Intragastric inoculation with a fresh broth culture of B. hyodysenteriae strain G44 resulted in a shorter incubation period and a higher proportionate duration of mucohemorrhagic diarrhea (MMHD) compared to D19. Intragastric inoculation with either 50 or 100 mL of B. hampsonii 30446 or B. hyodysenteriae (G44) were statistically equivalent. Oral inoculation with 100 mL or 300 mL also yielded similar results to intragastric inoculation but was more expensive due to the additional work and supplies associated with syringe training. Our future research will use intragastric inoculation with 100 mL of a fresh broth culture containing B. hyodysenteriae strain G44 as it yields a high incidence of mucohaemorrhagic diarrhea with a reasonable cost.

Brachyspira hyodysenteriae, an etiologic agent of swine dysentery (SD), is known for causing colitis. Although some aspects of colonic defenses during infection have been described previously, a more comprehensive picture of the host and microbiota interaction in clinically affected animals is required. This study aimed to characterize multiple aspects of colonic innate defenses and microbiome factors in B. hyodysenteriae-infected pigs that accompany clinical presentation of hemorrhagic diarrhea. We examined colonic mucus barrier modifications, leukocyte infiltration, cathelicidin expression, as well as microbiome composition. We showed that B. hyodysenteriae infection caused microscopic hemorrhagic colitis with abundant neutrophil infiltration in the colonic lamina propria and lumen, with minor macrophage infiltration. Mucus hypersecretion with abundant sialylated mucus in the colon, as well as mucosal colonization by [Acetivibrio] ethanolgignens, Lachnospiraceae, and Campylobacter were pathognomonic of B. hyodysenteriae infection. These findings demonstrate that B. hyodysenteriae produces clinical disease through multiple effects on host defenses, involving alterations of mucosal innate immunity and microbiota. Given that B. hyodysenteriae is increasingly resistant to antimicrobials, this understanding of SD pathogenesis may lead to future development of non-antibiotic and anti-inflammatory alternative therapeutics.

Swine dysentery is causally associated with Brachyspira hampsonii and B. hyodysenteriae infection. Given the importance of transmission models in understanding re-emergent diseases and developing control strategies such as vaccines, the objective of this experiment was to evaluate two experimental natural transmission (seeder pig) models in grower pigs, each with 24 animals. Seeder pigs were intragastrically inoculated using broth cultures of either B. hampsonii strain 30446 (genomovar II) or B. hyodysenteriae strain G44. In trial 1, three seeder pigs were placed into two pens containing nine susceptible contact pigs creating a 1:3 seeder:contact ratio. This was sufficient to achieve natural B. hampsonii infection of 13/18 (72%) contact pigs, however, the incidence of mucoid or mucohemorrhagic diarrhea (MMHD) in contact pigs differed significantly between pens (4/9 versus 9/9; P = 0.03). In trial 2, eight seeder pigs inoculated intragastrically with B. hampsonii did not develop MMHD but when re-inoculated with B. hyodysenteriae 14 days later, all developed mucohemorrhagic diarrhea within 13 days of re-inoculation. Two seeder pigs were placed into each of 4 contact pens each containing 4 pigs. This 1:2 seeder:contact ratio resulted in natural infection of 14/16 (87%) contact pigs with incubation period ranging from 9-15 days. There were no significant differences among pens in incubation period, duration, clinical period or severity of diarrhea. These trials demonstrated that a 1:2 seeder:contact ratio with groups of six grower pigs per pen sustained natural transmission of B. hyodysenteriae G44 with greater consistency in the incidence of MMHD among pens compared to a B. hampsonii 30446 transmission model using 1:3 seeder:contact ratio in pens of 12. Understanding why B. hampsonii intragastric inoculation failed in one experiment warrants additional research.

Brachyspira hyodysenteriae is one of the agents of swine dysentery (SD) and its eradication is an effective, but costly control measure. Being a voluntary measure, knowledge about drivers of motivation and satisfaction regarding the eradication of SD would help to convince farmers to eradicate. We aimed to describe eradications performed in Switzerland and to analyse factors influencing the pig owners' perception (motivation and satisfaction) of SD eradications to provide a basis to formulate recommendations and guidelines. Pig farmers (n = 68) having conducted an SD eradication and being interested in the study were interviewed using a standardised digital questionnaire. We assessed their motivation as moderately or highly motivated. Based on the farmers' evaluation of nine aspects of the eradication, satisfaction was considered to be moderate (<7/9 aspects positively evaluated) or high (≥7/9). Farms with fattening pigs and farms with breeding stock were analysed separately in subsets. First, multivariable factor analysis for mixed data (FAMD) were performed to describe the main patterns of variation. Then, risk factors for motivation and satisfaction were quantified by means of logistic regression models. Mainly total depopulations (73.5%) had been performed. Of the 36 farmers with breeding pigs, 24 were highly motivated, and 20 highly satisfied. Of the 61 farmers with fattening pigs, 45 were highly motivated and 42 highly satisfied. The FAMD revealed that the two main components explained only 17.0% and 11.0% (breeding stock) and 13.0% and 11.0% (fattening pigs) of the total variation, respectively. For farmers with breeding stock no significant factors for motivation were detected, but they were more satisfied (OR 25.0) when they had a batch farrowing of 3 weeks. Farmers with fattening pigs were more likely to be more motivated when providing access to outdoor areas (OR 3.3) and when it was their own initiative (OR 5.5). Farmers were more likely to be satisfied when they had only fattening pigs (OR 5.7), when the eradication was their own initiative (OR 5.5) and when they did not disinfect the barns during the eradication (OR 15.6). Farmers deciding themselves to eradicate are presumably more likely convinced of the benefits of the eradication. Satisfaction associated with a 3-weeks batch farrowing might be related to an easier to organise eradication and no disinfection to reduced labour and costs. In summary, the majority of the farmers were satisfied with the eradication. Education could promote self-motivation of farmers, and subsidies might support the implementation of SD eradications.

Research on the effects of different fiber types and levels on infection with Brachyspira hyodysenteriae on growth performance and nutrients digestibility in pigs is scarce. The objective of the current study was to investigate the effects of infection with B. hyodysenteriae when feeding diets varying in soluble and insoluble dietary fiber (DF) on the expression of swine dysentery, growth performance, and digestibility of organic matter (OM) nutrients. A total of 96 growing pigs (26.9 ± 2.5 kg) were used for the experiment and divided into six blocks. The growing pigs were fed one of four diets for 12 wk: low fiber (LF), high fiber (HF), high soluble fiber (HS), and high insoluble fiber (HI). After 2 wk, half of the pigs were inoculated with B. hyodysenteriae. Half of the pigs in each group were euthanized at week 6 for the measurement of the apparent digestibility at the ileum, cecum, colon, and total tract. The remaining pigs were maintained to observe and analyze the clinical expression of fecal score and excretion of B. hyodysenteriae, growth performance, and total tract digestibility up to 12 wk. In the current study, the experimental diets did not influence the expression of infection in the pigs. The body weight and average daily gain (ADG) were in line with the results of clinical expression from week 4 to 6. However, the ADG of the infected pigs started to recover from week 6 (P < 0.05) and then recovered from week 8 to 12 (P < 0.05). The infection with B. hyodysenteriae did not impair apparent ileal digestibility (AID; P > 0.05), whereas the apparent digestibility of OM, total non-starch polysaccharide, non-cellulosic polysaccharide, and cellulose in the cecum of the infected pigs was higher than non-infected pigs (P < 0.05). The apparent colonic digestibility of ash and nitrogen was higher in non-infected pigs than in infected pigs (P < 0.05). The pigs fed the LF diet had a higher digestibility in all segments of the intestinal tract, whereas the HS diet had the lowest AID but higher or similar to the LF diet in the cecum, colon, and the total tract (P < 0.05). The pigs fed the HF and HI diets, with a high proportion of insoluble fiber, had a lower digestibility in the hindgut than the other two diets (P < 0.05). In conclusion, the infection with B. hyodysenteriae negatively influenced clinical signs of swine dysentery and growth performance but did not impair AID, and neither soluble nor insoluble DF influenced the expression of the infection.


Swine dysentery is a severe disease that can cause increased mortality and poor feed efficiency with bloody diarrhea. This disease can be treated with antibiotics, but there is a limitation of using antibiotics due to governmental policy, thereby the incidence of swine dysentery has been increased. We, therefore, try to find alternatives with diverse fiber sources and understand the mechanism of swine dysentery in growing pigs. In this study, infection of Brachyspira hyodysenteriae showed a negative influence on growth performance, but compensatory growth and recovery were observed in pigs after 6 wk of the infection. The apparent ileal digestibility was not affected by the infection, and the digestibility of non-starch polysaccharides in the cecum was rather increased than decreased probably because of interaction between B. hyodysenteriae and specific bacteria, which can stimulate fiber degradation in the cecum. However, fiber type and level did not influence the prevention and alleviation of the infection.

The aim of this study was to evaluate genetic diversity, distribution, evolution and population structure of Brazilian Brachyspira hyodysenteriae strains isolated from swine. Multilocus Sequence Typing (MLST) analysis using seven housekeeping genes was applied to 46 isolates obtained from outbreaks of swine dysentery that occurred between 2011 and 2015 in the states of Minas Gerais, São Paulo, Mato Grosso, Rio Grande do Sul and Santa Catarina. Historical isolates from Rio Grande do Sul obtained in 1998 were also included in the study. An independent international profile of the global sequences deposited in the B. hyodysenteriae database was used for comparisons with the Brazilian strains. All isolates from 2011 to 2015 were classified into nine sequence type (STs) and divided into four clonal complexes. These findings indicated genetic relationships among the B. hyodysenteriae from different Brazilian states, among historical strains isolated in 1998 and from recent outbreaks, and relatedness with global isolates. Seven STs were unique and, to date, only reported in Brazil.

Bovine coronavirus (BCoV) is associated with three distinct clinical syndromes in cattle that is, neonatal diarrhoea, haemorrhagic diarrhoea in adults (the so-called winter dysentery syndrome, WD) and respiratory infections in cattle of different ages. In addition, bovine-like CoVs have been detected in various species including domestic and wild ruminants. However, bovine-like CoVs have not been reported so far in odd-toed ungulates. We describe an outbreak of WD associated with a bovine-like CoV affecting several captive wild ungulates, including Indonesian tapirs (Acrocodia indica) an odd-toed ungulate species (Perissodactyla) which, with even-toed ungulates species (Artiodactyla) form the clade Euungulata. Genomic characterization of the CoV revealed that it was closely related to BCoVs previously reported in America. This case illustrates the adaptability of bovine-like CoVs to new species and the necessity of continued surveillance of bovine-like CoVs in various species.

Porcine circovirus-like virus (PCLV) is a type of circular Rep-encoding single-stranded DNA virus and may be associated with the development of diarrheal symptoms in pigs. In this study, we retrospectively analyzed three years of past cases in Anhui, China, and reported a case of hemorrhagic enteritis and death in a pregnant sow possibly caused by PCLV. In addition, we analyzed the evolutionary characteristics of PCLV and found that mutation, recombination and selective pressure all played an important role in the evolution of PCLV. We identified N15D and T17S as well as L56T, T58R, K59Q, M62R, L75I and R190K mutations in two different branches, and we noted recombination events in the Rep of a group of Chinese strains. Analysis of selection pressure revealed that PCLV gained more positive selection, indicating that the virus is in a continuous evolutionary state. The PR2 plot, ENC-plot and neutrality analysis showed a greater role of natural selection than that of mutational pressure in the formation of codon usage patterns. This study is the first to identify PCLV in sows with hemorrhagic dysentery and death, and it provides new epidemiological information on PCLV infection in pigs in China.

Bovine coronavirus (BCoV) is the causative agent of winter dysentery (WD). In adult dairy cattle, WD is characterized by hemorrhagic diarrhea and a reduction in milk production. Therefore, WD leads to significant economic losses in dairy farms. In this study, we aimed to isolate and characterize local BCoV strains. BCoV positive samples, collected during 2017-2021, were used to amplify and sequence the S1 domain of S glycoprotein and the full hemagglutinin esterase gene. Based on our molecular analysis, local strains belong to different genetic variants circulating in dairy farms in Israel. Phylogenetic analysis revealed that all local strains clustered together and in proximity to other BCoV circulating in the area. Additionally, we found that local strains are genetically distant from the reference enteric strain Mebus. To our knowledge, this is the first report providing molecular data on BCoV circulating in Israel.

Swine dysentery (SD) is an enteric disease associated with strongly ß-hemolytic Brachyspira spp. that cause mucohemorrhagic diarrhea primarily in grower-finisher pigs. We characterized alteration of colonic mucin composition and local cytokine expression in the colon of pigs with acute SD after B. hyodysenteriae (Bhyo) infection and fed either a diet containing 30% distillers dried grains with solubles (DDGS) or a control diet. Colonic tissue samples from 9 noninoculated pigs (Control, N = 4; DDGS, N = 5) and 10 inoculated pigs experiencing acute SD (Bhyo, N = 4; Bhyo-DDGS, N = 6) were evaluated. At the apex of the spiral colon, histochemical staining with high-iron diamine-Alcian blue revealed increased sialomucin (P = .008) and decreased sulfomucin (P = .027) in Bhyo pigs relative to controls, with a dietary effect for sulfomucin. Noninoculated pigs fed DDGS had greater expression of sulfomucin (P = .002) compared to pigs fed the control diet. Immunohistochemically, there was de novo expression of mucin 5AC (MUC5AC) in the Bhyo group while mucin 2 (MUC2) expression was not significantly different between groups. RNA in situ hybridization to detect the pro-inflammatory cytokine IL-1ß often showed increased expression in the Bhyo group although without statistical significance, and this was not correlated with MUC5AC or MUC2 expression, suggesting IL-1ß is not a major regulator of their secretion in acute SD. Expression of the anti-inflammatory cytokine TGF-ß1 was significantly suppressed in the Bhyo group compared to controls (P = .005). This study reveals mucin and cytokine alterations in the colon of pigs with experimentally induced SD and related dietary effects of DDGS.

As a multifactorial cause, gastric ulceration-mediated diarrhea is widely prevalent in the weaned piglets, impairing pig health and economic benefits. With full implementation of antibiotic stewardship programs in China, Bacillus cereus (B. cereus) and Aspergillus fumigatus (A. fumigatus) were identified frequently in porcine feedstuffs and feeds of the animal industry. Association between feed-borne B. cereus and frequent diarrhea remains unclear. In the present study, we conducted a survey of B. cereus and A. fumigatus from feeds and feedstuffs in pig farms during hot season. Interestingly, B. cereus, B. subtilis, B. licheniformis and B. thuringinesis were isolated and identified from piglets' starter meals to sow feeds, accounting for 56.1%, 23.7%, 13.7% and 6.5%, respectively. Obviously, both B. cereus and B. subtili were dominant contaminants in the survey. In an in vitro study, Deoxynivalenol (DON) contents were determined in a dose-dependent manner post fermentation with B. cereus (405 and DawuC). Subsequently, 36 weaned piglets were randomly assigned to four groups and the piglets simultaneously received the combination of virulent B. cereus (Dawu C) and A. fumigatus while animals were inoculated with B. cereus (Dawu C), A. fumigatus or PBS as the control group. Clinically, piglets developed yellow diarrhea on day 5 and significant reductions of relative body weight were observed in the B. cereus group, and co-infection group. More importantly, IgG titers against Classical swine fever virus (CSFV) and Porcine epidemic diarrhea (PED) were reduced dramatically during 14-day observation in co-infection group, the B. cereus (Dawu C) group or the A. fumigatus group. However, lower Foot and mouth disease (FMD) -specific antibodies were reduced on day 7 compared to those of the control group. Additionally, lower lymphocyte proliferations were found in the B. cereus group and the co-infection group compared to the control group. Postmortem, higher lesions of gastric ulceration were observed in the B. cereus group and the co-infection group from day 7 to day 14 compared with those of the A. fumigatus group and the control group. Compared to the A. fumigatus group, higher DON contents were detected in the stomach inoculated with B. cereus and the co-infection with A. fumigatus. In conclusion, our data support the hypothesis that B. cereus might be associated with severe diarrhea by inducing gastric ulcerations and A. fumigatus might aggravate immune suppression, threating a sustainable swine industry. Swine dysentery (SD) is a global, production-limiting disease of pigs in commercial farms. It is associated with infection by Brachyspira hyodysenteriae and B. hampsonii, and characterized by mucohaemorrhagic diarrhea and colitis, SD prevention, treatment or control relies heavily on antimicrobials as no commercial vaccines are available. This is linked to our poor understanding of the disease pathogenesis. Our goal was to characterize the host-pathogen interactions during the early stage of infection. We employed dual RNA-seq to profile mRNA and miRNA following 1-h incubation of colonic explants with a pathogenic or a non-pathogenic B. hampsonii strain. Our results suggest that the pathogenic strain more efficiently interfered with the host's ability to activate and build a humoral response (through IL-4/CCR6/KLHL6 interactions), epithelial wound repair mechanisms (associated with LSECtin impairment of macrophages), induced mitochondrial dysfunction (linked to MDR1), and loss of microbiome homeostasis. The pathogenic strain also up-regulated the expression of stress-associated genes, when compared to the non-pathogenic strain. These results shed a light on the pathophysiological mechanisms that lead to SD and will contribute to the development of novel disease control tools.

Bovine coronavirus (BCoV) is a recognized cause of severe neonatal calf diarrhea, with a negative impact on animal welfare, leading to economic losses to the livestock industry. Cattle production is one of the most important economic sectors in Uruguay. The aim of this study was to determine the frequency of BCoV infections and their genetic diversity in Uruguayan calves and to describe the evolutionary history of the virus in South America. The overall detection rate of BCoV in Uruguay was 7.8% (64/824): 7.7% (60/782) in dairy cattle and 9.5% (4/42) in beef cattle. The detection rate of BCoV in samples from deceased and live calves was 10.0% (6/60) and 7.6% (58/763), respectively. Interestingly, there was a lower frequency of BCoV detection in calves born to vaccinated dams (3.3%, 8/240) than in calves born to unvaccinated dams (12.2%, 32/263) (OR: 4.02, 95%CI: 1.81-8.90; p = 0.00026). The frequency of BCoV detection was higher in colder months (11.8%, 44/373) than in warmer months (1.5%, 3/206) (OR: 9.05, 95%CI: 2.77-29.53, p = 0.000013). Uruguayan strains grouped together in two different lineages: one with Argentinean strains and the other with Brazilian strains. Both BCoV lineages were estimated to have entered Uruguay in 2013: one of them from Brazil (95%HPD interval: 2011-2014) and the other from Argentina (95%HPD interval: 2010-2014). The lineages differed by four amino acid changes, and both were divergent from the Mebus reference strain. Surveillance should be maintained to detect possible emerging strains that can clearly diverge at the antigenic level from vaccine strains.

The anaerobic intestinal spirochaete Brachyspira hyodysenteriae colonises the large intestine of pigs and causes swine dysentery (SD), a severe mucohaemorrhagic colitis. SD occurs worldwide, and control is hampered by a lack of vaccines and increasing antimicrobial resistance. B. hyodysenteriae strains typically produce strong beta-haemolysis on blood agar, and the haemolytic activity is thought to contribute to the pathogenesis of SD. Recently, weakly haemolytic variants of B. hyodysenteriae have been identified in Europe and Australia, and weakly haemolytic strain D28 from Belgium failed to cause disease when used experimentally to infect pigs. Moreover, pigs colonised with D28 and then challenged with virulent strongly haemolytic strain B204 showed a delay of 2-4 days in developing SD compared to pigs not exposed to D28. The current study aimed to determine whether Australian weakly haemolytic B. hyodysenteriae strain MU1, which is genetically distinct from D28, could cause disease and whether exposure to it protected pigs from subsequent challenge with strongly haemolytic virulent strains. Three experimental infection studies were undertaken in which no diseases occurred in 34 pigs inoculated with MU1, although mild superficial lesions were found in the colon in 2 pigs in one experiment. In two experiments, significantly fewer pigs exposed to MU1 and then challenged with strongly haemolytic virulent strains of B. hyodysenteriae developed SD compared to control pigs not previously exposed to MU1 (p = 0.009 and p = 0.0006). These data indicate that MU1 lacks virulence and has potential to be used to help protect pigs from SD.

INTRODUCTION: The eradication of Swine Dysentery (infection with Brachyspira hyodysenteriae) within farm populations is extremely complex and its success is significantly linked to the compliance of pig farmers performing the onsite eradications, emphasizing the importance of their motivation. The present study focuses on the motivation and satisfaction of Swiss pig farmers concerning eradication of Swine Dysentery on their farms and has the intention of generating data to establish a basis for decision-making regarding future eradications. A structured survey was conducted on 68 Swiss pig farms where an eradication of Swine Dysentery had already taken place. The data collected were then statistically analyzed. Of the study's population, 50 (73.5%) farms underwent eradication using the total depopulation method and 18 (26.5%) farms conducted an eradication process using the partial depopulation method; both in accordance to the guidelines of Switzerland's Pig Health Service (SUISAG Schweinegesundheitsdienst; SGD). When asked about the driving factors behind their decision to perform the eradications, owners cited their own interest (57.4%; 39/68) (primarily due to financial aspects (38.5%; 15/39)) or external pressure (42.7%; 29/68) (predominantly through marketers as well as through the health status assigned by the SGD (each 11/29)). The duration of the eradication on farms differed significantly and costs varied considerably within the different production types as well as the type of eradication performed. Utilized means and processes for eradication were the same (drugs, disinfectant for manure) or much alike (e.g. emptying of the pigpens) however stock-specific and individual variations were exhibited. The majority of the interviewed livestock owners were motivated to perform the eradication (86.6%; 58/67), satisfied with having performed the eradication and satisfied with the outcome of the eradication (89.7%; 61/68). The cost efficiency of the farms (54.4%; 37/68) and the health status of the herds (63.2%; 43/68) were perceived as having improved and the usage of drugs having diminished (59.7%; 40/67) post eradication. The effort of an eradication was described as acceptable to too great. The study showed herd eradication of Swine Dysentery to be perceived as mainly positive. However, effort as well as costs are felt to be considerable and could, therefore, provide the most important starting point to increasing the willingness of pig owners to perform eradications.


INTRODUCTION: L'assainissement d'exploitations avec de problèmes de dysenterie porcine (infection à Brachyspira hyodysenteriae) peut prendre beaucoup de temps et son succès est étroitement lié à l'observance des personnes qui l'exécutent. C'est pourquoi les éleveurs de porcs doivent être motivés pour effectuer cette lutte. La présente étude a notamment porté sur la motivation et la satisfaction des détenteurs suisses de porcs en ce qui concerne l'assainissement de la dysenterie porcine afin de générer des données et, partant, des aides à la décision pour de futures mesures de lutte. Pour cela, des données ont été collectées au moyen d'un entretien structuré et analysées statistiquement dans 68 exploitations suisses après un assainissement complet de la dysenterie. Dans ces exploitations, 50 assainissements totaux (73.5%) et 18 assainissements partiels (26,5%) ont été effectués conformément aux directives du Service sanitaire porcin suisse (SUISAG SGD). L'intérêt propre (57,4%, 39/68) (principalement dû aux aspects financiers (38,5%; 15/39)) et la pression externe (42,7%; 29/68) (principalement par statut SGD ou par les négociants (chacun pour 11/29)) étaient les raisons de l'assainissement. La durée de l'assainissement et les coûts variaient considérablement selon les types de production de même que selon les types d'assainissement. Dans le cadre de l'assainissement, les moyens utilisés et les processus étaient les mêmes (médicaments, désinfectant de fumier) respectivement étaient similaires (par exemple, la vidange des écuries) mais présentait des variations individuelles spécifiques à l'exploitation. La majorité des propriétaires d'animaux interrogés étaient motivés à un assainissement (86,6%, 58/67) et étaient également satisfaits de l'avoir fait (97,1%, 66/68) et du résultat (89,7%, 61/68). Après l'assainissement, la rentabilité (54,4%; 37/68) et la santé du cheptel (63,2%; 43/68) ont été jugées subjectivement meilleures et la consommation de médicaments (59,7%; 40/67) inférieure. Le coût de l'assainissement était, selon le type de production, considéré comme justifié jusqu'à élevé. On a pu montrer que l'assainissement de la dysenterie porcine est généralement évalué positivement. Cependant, les efforts et les coûts sont considérables et représentent des points de départ possibles pour optimiser la volonté des éleveurs de porcs.

OBJECTIVES: Multidrug-resistant Escherichia coli isolates recovered from food-producing animals are a global public-health concern, especially those with transferable mechanisms of antimicrobial resistance such as extended-spectrum ß-lactamase (ESBL) and mcr-1 genes. Here we report for the first time E. coli recovered from diarrhoeic and healthy pigs harbouring blaCTX-M and/or mcr-1 from Argentinean farms. METHODS: During 2017, a total of 34 E. coli were recovered from 31 faecal samples from diarrhoeic piglets and healthy fattening pigs from five pig farms in three Argentinean provinces. Antimicrobial susceptibility was evaluated by agar diffusion and resistance genes were identified by PCR. Multiplex PCR was applied to screen for ST69, ST73, ST95 and ST131 clones. Genetic relationships were evaluated by XbaI-PFGE. RESULTS: A high diversity of resistance profiles was observed (20 profiles among 34 isolates), and 71% of isolates were multidrug-resistant. Resistance to third-generation cephalosporins (3GCs) was observed in 28 isolates and was associated with blaCTX-M (24), blaCMY (3) and blaPER-2 (1) genes. blaCTX-M alleles were grouped by specific PCR as follow: 17 blaCTX-M-8/25; 4 blaCTX-M-1/15; 2 blaCTX-M-2; and 1 blaCTX-M-9/14. Twelve isolates were positive for mcr-1, of which six were also resistant to 3GCs and were positive for blaCTX-M-8/25 (4), blaCTX-M-1/15 (1) or blaCMY (1). High genetic diversity was observed, discriminating 29 profiles. One ST131 and two ST95 human-associated clones were detected. CONCLUSION: Here we describe E. coli isolates recovered from diarrhoeic piglets and healthy fattening pigs harbouring ESBL and/or mcr-1 genes. 3GC resistance was mainly associated with CTX-M, in particular with blaCTX-M-8/25 alleles.

An acute epidemic of porcine epidemic diarrhea (PED) has affected the USA since 2013 and spread all around the world. In France, the immune status of the pig population against PED virus (PEDV) was expected to be low due to the absence of circulation of the virus since the 80's and a compulsory notification of PED was set up in 2014. Here, we reported the first case of a PED outbreak in December 2014 in the North of France after a long absence of the disease, the monitoring of the excretion and the control measure implementation. The isolated strain in France in December 2014 was a PEDV "S-InDel" strain which was close to the "S-InDel" German PEDV strain isolated in May 2014. The individual shedding duration of PEDV in feces was estimated around 20 days for pigs of different ages. Biosecurity measures implemented allowed the limitation of PEDV spread to fattening and farrowing rooms without dissemination to the nursery block. Using strict biosecurity measures, direct shipment of infected fatteners to the slaughterhouse, strict decontamination protocols with a quarantine of 6 weeks for replacement gilts without voluntary contamination helped PEDV fade out within the herd and avoided the spread to other herds. PEDV presence in manure was investigated as well as the inactivation treatment of the virus present in the liquid manure. An increase to a pH 12 of liquid manure by liming led to the absence of PEDV detection by RT-PCR after seven days.
