A 4 yr old male Maltese dog presented with a 1 wk history of intermittent neck pain and progressive difficulty walking. Neurologic evaluation was consistent with a left-sided brainstem lesion. On oral examination, left lingual hemiatrophy was evident suggesting hypoglossal nerve involvement. A dumbbell-shaped extra-axial mass in the left side of the caudal fossa extending extracranially through the hypoglossal canal was detected by MRI. At postmortem histologic examination, the hypoglossal nerve was diffusely infiltrated by fusiform neoplastic cells arranged in Antoni A and Antoni B patterns. This is the first description of a malignant nerve sheath tumor selectively involving the hypoglossal nerve in a dog.

Leiomyosarcomas arising from paratesticular structures are rare. We report a case of epididymal sarcoma in an aged male Rottweiler that had histological features suggestive of neural origin but with muscular histogenesis confirmed by immunohistochemistry. Histologically, the lesion comprised a hypercellular neoplasm that had interlacing bundles of spindloid cells, prominent nuclear palisading and many Verocay bodies. Immunohistochemically, the neoplastic cells were strongly positive for smooth muscle markers but negative for neural markers, leading to the diagnosis of leiomyosarcoma. While the presence of nuclear palisading and Verocay bodies has been regarded as strong evidence for the diagnosis of schwannomas, the present report documents the presence of true Verocay bodies in a non-neuronal tumour. It also indicates that immunophenotyping is required for an accurate diagnosis when Verocay bodies are detected in sarcomatous or spindle cell tumours and provides prognostic information on this case. To the best of our knowledge, this is the first report of the presence of true Verocay bodies in a neoplasm other than a nerve sheath tumour in animals.

RESUMO

The present study supports the usefulness of ancillary techniques, such as immunohistochemistry, as a valid diagnostic tool in the field of fish oncology. The immunohistochemical patterns observed in four neoplasms on four individual teleosts belonging to different species are described. Cytokeratin, vimentin, actin, S100, calretinin, and Melan-A antibodies were used. Diagnoses of papilloma in a Bream Abramis brama, fibroma in a Sand Steenbras Lithognathus mormyrus, schwannoma in a Crucian Carp Carassius carassius, and melanoma in a spontaneously inbred Xiphophorus hybrid were made. Diagnosis of tumors in fish is not always easy to carry out, and the tool provided by antibodies used on mammalian tissue is essential for obtaining definitive, unambiguous, and inexpensive identification.

An eleven-year-old, female spayed Boxer dog was diagnosed with a uveal schwannoma (formerly known as the spindle cell tumor of the blue-eyed dog or SCTBED) despite having a uniformly brown iris. The patient presented to emergency for ocular discomfort, and the right globe was subsequently enucleated due to glaucoma and submitted for histopathology. Upon histopathologic evaluation, a uveal schwannoma was diagnosed and confirmed with immunohistochemical staining. Complete metastatic evaluation 1 and 6 months after initial presentation did not reveal evidence of metastasis, and the dog remains systemically healthy. This case represents a unique variant of uveal schwannoma and is relevant because although the vast majority of these tumors occur in blue-eyed dogs, clinicians should not completely rule out this tumor as a differential based on the iris color.

Peripheral nerve sheath tumours (PNSTs) are neoplastic growths derived from Schwann cells, perineural cells or both. Malignant PNSTs (MPNSTs) are uncommon in domestic animals. This report describes the concomitant occurrence of PNSTs in a 10-year-old female cocker spaniel with a clinical history of respiratory impairment. Grossly, there was a large infiltrative mass in the caudal lobe of the right lung; smaller nodules were observed in the other lobes of the right lung. Furthermore, a small encapsulated cutaneous nodule was observed on the left hindlimb. Histopathology of the pulmonary tumours revealed the proliferation of pleomorphic spindle-shaped cells with moderate mitotic index arranged in interwoven bundles and concentric Antoni A and Antoni B patterns; invasion of the adjacent pulmonary tissue was observed. The cutaneous nodule consisted of neoplastic mesenchymal cells in interwoven bundles with concentric whorls, but without the marked anisokaryosis, binucleation and infiltrative growth seen in the pulmonary tumour. Immunohistochemistry revealed that both tumours were immunoreactive for vimentin, glial fibrillary acidic protein and S100 protein, but were negative for factor VIII. These findings are indicative of a MPNST in the lung with a concomitant benign PNST in the limb. This case represents the first report of a primary MPNST in the lung of a dog. This neoplastic growth should be included in the differential diagnosis of primary malignant pulmonary tumours of dogs.

Pigmented tumors have been reported infrequently in captive deer. We document herein the clinical progression and gross and histopathologic features of pigmented tumors diagnosed as melanoma and pigmented schwannoma in 11 white fallow deer ( Dama dama). Affected animals were part of a captive herd maintained at a drive-through park in southern Oregon and were 5-17 y of age during the study period (2004-2013). Primary lesion locations included periocular, perineal, and neck tissues, with cutaneous and internal metastases later identified at autopsy in some cases of malignant melanoma. Diagnoses included 7 malignant melanomas, 2 benign melanomas, and 2 pigmented schwannomas. Diagnosis of melanoma was based on typical histomorphologic features, and final diagnosis of pigmented schwannomas was based on histomorphologic features with negative staining for melan A and positive staining for laminin. Metastasis was found in 3 of 7 cases diagnosed as malignant melanoma; 2 had extensive pulmonary involvement and resulted in euthanasia of the animal; 1 animal developed eyelid and ear lesions that also resulted in euthanasia.

A 14-year-old Maltese dog presented with progressive exophthalmos and external deviation of the right eye. Ultrasonography revealed the presence of a retrobulbar mass and fine-needle aspiration cytology was performed, which detected a malignant mass. There was no evidence of metastasis on thoracic and abdominal radiography. Computed tomography showed no invasion into the bony orbit and no metastasis to the lung or lymph nodes. Exenteration was performed to remove the mass completely. Malignant peripheral nerve sheath tumor was confirmed by histopathological examination.

This case report describes the diagnosis of a peripheral nerve sheath tumor of the deep branch of the radial nerve distal to the elbow in a dog. The lesion was identified using computed tomography and ultrasonography and confirmed as sarcoma on histopathological analysis of incisional biopsies. Clinical signs dramatically improved following surgical biopsy before recurring three months later. Repeat epineurotomy of the deep branch of the radial nerve resulted in clinical improvement for a further month before signs once again returned. Epineurotomy as a palliative treatment for peripheral nerve sheath tumors has not been previously described, but may have a place in palliation of clinical signs in specific cases of peripheral nerve sheath tumors in which limb amputation is not an option.

BACKGROUND: Stereotactic radiotherapy (SRT) is an emerging technique for treating tumors in animals. OBJECTIVES: To assess the outcome of dogs with suspected intracranial trigeminal nerve peripheral nerve sheath tumors (PNST) treated with SRT. ANIMALS: Eight dogs with presumptive PNST. METHODS: This was a retrospective study of dogs identified by searching UC Davis Veterinary Medical Teaching Hospital medical records for dogs treated with SRT for a presumed PNST. Presumptive diagnosis was based on magnetic resonance imaging. SRT was delivered in 3 dose fractions of 8 Gray (Gy) on consecutive days or every other day to a total dose of 24 Gy. RESULTS: Median disease-specific survival was 745 days (range: 99-1375 days, n = 6). No signs of acute adverse effects of radiation treatment were recorded. Late radiation effects versus tumor progression could not be confirmed histopathologically because of few animals undergoing necropsy. CONCLUSIONS AND CLINICAL IMPORTANCE: This study provides preliminary evidence that dogs with PNST benefit from SRT in terms of long-term survival. The treatment appears to be well tolerated and requires fewer anesthetic events for animals compared to full-course radiation.

A 9-year-old spayed/female Chinese Pug dog presented for evaluation of a mass located on the rostral aspect of the tongue. An incisional biopsy was acquired, submitted, and interpreted as a possible granular cell tumor based on hematoxylin and eosin, and periodic acid Schiff histopathologic staining characteristics. The diagnosis was supported by immunohistochemical evaluation that was positive for S-100, vimentin, and neuron-specific enolase. Based on the absence of mitotic figures in the incisional biopsy, a partial glossectomy was performed with gross margins of at least 1-cm. The excisional biopsy revealed significant features of malignancy, with neoplastic cells in close association with peripheral nerves, consistent with malignant peripheral nerve sheath tumor. Tumor-free margins were obtained, and the glossectomy had expected healing with no recurrence apparent 6-months following surgery.

In this retrospective study was determined the frequency of canine skin peripheral nerve sheath tumors (PNST) in cases diagnosed by the Setor de Patologia Veterinária of the Universidade Federal do Rio Grande do Sul (SPV-UFRGS), Brazil, between the years 2000 and 2012. The canine profiles, as well as histological, immunohistochemical and prognostic aspects of the tumors were based on 70 samples, comprising 40 females, 29 males and one unspecified sample. Between 2000 and 2012, 2,984 skin tumors of dogs were diagnosed in the SPV-UFRGS, totaling 2.34% of skin neoplasms in dogs. Animals that comprised the largest amount of samples (43%) were those with no breed (SRD), followed by German Shepherds (10%). Females were more affected than males (40/70 - 57% and 29/70 - 41% respectively). Skin PNST of this research showed predominant localization on the limbs (40% in the forelimbs and 29% in the hindlimbs); affecting adult dogs, mostly aged between 8 and 11 years (54%). The samples were routinely processed for hematoxylin and eosin, and were also evaluated by toluidine blue and Masson's trichrome staining, and immunohistochemistry (IHC) anti-vimentin, -S-100, -GFAP, -actin, von Willebrand factor and neurofilament. Anisocytosis and anisokaryosis, mitotic index, intratumoral necrosis, invasion of adjacent tissues, tumor location, local recurrence and metastasis were related to the diagnosis of benign (49/70) or malignant tumor (21/70). The Antoni A histological pattern was observed more frequently in benign tumors. The immunohistochemistry helped to diagnose PNST, and anti-vimentin and anti-protein S-100 showed the highest rates of immunostaining. Throughout statistical analysis of animals with tumor recurrence, it was found that the chance of an animal with a malignant peripheral nerve sheath tumor to develop recurrence is 4.61 times higher than in an animal that had a benign tumor...


Neste estudo retrospectivo foi determinada a frequência dos tumores de bainha de nervo periférico (TBNP) na pele de cães diagnosticados pelo Setor de Patologia Veterinária da Universidade Federal do Rio Grande do Sul (SPV-UFRGS), Brasil, no período de 2000 a 2012. O histórico dos cães, assim como a histologia, a imuno-histoquímica e o prognóstico em relação aos tumores foram baseados em 70 amostras histológicas, correspondendo a 40 fêmeas (57%), 29 machos (41%) e uma amostra sem a informação do sexo. De 2000 a 2012, foram diagnosticados 2984 tumores de pele de cães pelo SPV-UFRGS, totalizando 2,34% das neoplasias em cães. A maior parte das amostras (43%) foi proveniente de animais sem raça definida (SRD), seguidos pelos da raça Pastor Alemão (10%). A localização predominante dos TBNP desta pesquisa ocorreu em membros (40% em membro tóracico e 29% em membro pélvico), afetando principalmente cães de oito a 11 anos idade (54%). As amostras foram processadas rotineiramente para coloração de Hematoxilina e eosina, e foram realizadas as colorações de azul de toluidina e tricrômio de Masson, assim como realizada imuno-histoquímica (IHQ) anti-vimentina, -S-100, -GFAP, -actina, von Willbrand e neurofilamento. Os fatores anisocitose e anisocariose, índice mitótico, necrose intratumoral, invasão tumoral em tecidos adjacentes, localização tumoral, local de recorrência e metástase foram relacionados com o diagnóstico de benignidade (49/70) ou malignidade (21/70). O padrão histológico de Antoni A foi observado com mais frequência em tumores benignos. A IHQ auxiliou no diagnóstico dos TBNP, com maiores índices de imunomarcação anti-vimentina e S-100. A análise estatística das amostras de animais que apresentaram recidiva tumoral evidenciou que a chance de um animal com tumor de bainha de nervo periférico maligno desenvolver recidiva é 4,61 vezes maior do que em animais que apresentam tumor benigno...

Nodular lung lesions in swine are frequently due to abscesses or granulomatous pneumonia. Although tumours are rarely reported in modern pig farming, they should be considered as a differential diagnosis when nodular lung lesions are found. A first-parity sow exhibiting respiratory signs was euthanized. Several whitish firm nodules, not encapsulated, ranging in diameter from 0.5 to 5 cm were present in all lung lobes. Microscopically, the nodules were composed of dense neoplastic cells, mainly in Antoni types A and B patterns, infiltrative and with development of emboli. All neoplastic cells stained positively by immunohistochemistry for vimentin and S-100 protein, with variable immunostaining for glial fibrillary acidic protein and stained negative for cytokeratin. Based on the gross, histological and immunohistochemical features, the tumor was diagnosed as malignant peripheral nerve sheath tumour.

An adult Goldfish (Carassius auratus) developed an exophitic, multinodular, mass in the dorsal region involving the fin. The clinical and pathologic approach included ultrasonography, cytology, histopathology, immunohistochemistry, and ultrastructural evaluation. B-mode and color Doppler ultrasonographic findings showed an oval, well-defined mass with slightly inhomogeneous parenchymatous echotexture associated with an intense intralesional vascularization. Cytology and histology revealed neoplastic pleomorphic spindle cells arranged in a storiform pattern or in palisades typical of Antoni A pattern schwannoma. Moderate anisocytosis and anisokaryosis and occasional binucleation were also present. The neoplastic tissue deeply infiltrated the skeletal muscle of the dorsal region. Immunohistochemistry showed a diffuse cytoplasmic immunoreactivity of neoplastic cells to S100 protein and proliferating cell nuclear antigen (PCNA); glial fibrillary acidic protein was negative. The PCNA proliferation rate was 23.5% (calculated as the mean of 10 fields). Ultrastructurally, neoplastic cells were juxtaposed with parallel nuclei forming the typical palisade pattern and sharing cytoplasmic and nuclear features with human schwannoma. This is the first cytologic description supplied with echographic investigation of a malignant schwannoma in a fish species.

A 4-year-old castrated labrador retriever presented for cardiac evaluation to determine the etiology of cardiogenic pulmonary edema diagnosed 1 month prior. A large pedunculated mass involving the ventral aspect of the mural mitral valve leaflet and the endocardial surface of the left ventricular free wall, resulting in severe mitral regurgitation, was identified on echocardiogram. Histopathology and immunohistochemistry of this mass and other endocardial masses identified at necropsy for S-100 protein were consistent with a diagnosis of schwannoma. To the authors' knowledge, this is the first case of a benign intracardiac schwannoma described in the left heart of a dog.

The formalin-fixed, amber-colored right globe from a 12-week-old female silver Labrador Retriever dog was submitted to the Comparative Ocular Pathology Laboratory of Wisconsin for light microscopic evaluation. The clinical history described a collapsed anterior chamber and multifocal nodular lesions in the peripheral iris. Histologically, immunohistochemically, and ultrastructurally, the uveal mass was consistent with a malignant schwannoma; there was extension along peripheral nerves within the sclera. The signalment and behavior of the neoplasm distinguish it from the uveal schwannoma of blue-eyed dogs and bear some resemblance to the ocular lesions in human neurofibromatosis. The dilute color mutation may contribute to the cause. Six weeks later, the dog did not develop any additional masses.

A malignant epithelioid schwannoma of the oral cavity was diagnosed in an 8-year-old domestic short-hair cat. The mass was located in the gingiva of the upper left premolar to molar region and showed multinodular growth patterns. The mass comprised epithelioid cells arranged in densely packed sheets. Tumor cells had large, round to oval nuclei with prominent nucleoli and an abundant eosinophilic cytoplasm. Immunohistochemically, most of the tumor cells were positive for S-100 protein, glial fibrillary acidic protein and vimentin, but all lacked melanoma-associated antigen and muscle and neuroendocrine markers. Stains for type IV collagen showed linear immunoreactivity around single cells and groups of cells. Ultrastructurally, tumor cells were separated by a well-defined basement membrane, and interdigitating cell processes were observed. To our knowledge, this is the first report of feline malignant epithelioid schwannoma.

Schwannomas arising in the parotid gland are rare in man and are not documented in domestic animals. This report describes the clinical, morphological and immunohistochemical findings of a benign schwannoma in the parotid gland of a 12-year-old warmblood gelding. A slow-growing mass was surgically excised from the parotid gland and did not recur within the following 6 months. The tumour was well circumscribed and was composed of densely packed spindle cells partly arranged in an Antoni A pattern and intermixed with hypocellular areas resembling the Antoni B pattern. Tumour cells expressed vimentin, S100 protein, glial fibrillary acidic protein and variably neuron-specific enolase. Less than 5% of the neoplastic cells were immunolabelled with the proliferation marker Ki67. Although malignant melanomas frequently involve the parotid gland in horses, schwannomas should be included in the differential diagnosis of parotid masses.

Canine malignant peripheral nerve sheath tumors (MPNSTs) occur not only in the peripheral nervous system (PNS) but also in soft tissue and various organs (non-PNS). The most important diagnostic criterion is proof of peripheral nerve sheath origin. This is difficult in non-PNS MPNSTs, and its differential diagnosis is challenging. Canine perivascular wall tumors (PWTs) also commonly arise in soft tissue. Their histopathological features are quite similar to those of canine MPNSTs, making their differential diagnosis challenging. To elucidate whether the morphological features are applicable to diagnose non-PNS MPNSTs and to demonstrate useful markers for distinction between canine MPNSTs and PWTs, the authors examined 30 canine MPNSTs and 31 PWTs immunohistochemically for S100, nestin, NGFR, Olig2, claudin-1, CD57, PRX, α-SMA, desmin, and calponin. Among canine MPNSTs, the PNS tumors displayed significantly higher S100 and Olig2 expression than the non-PNS tumors. The expression levels of the other markers did not differ significantly, suggesting that the same morphological diagnostic criteria are applicable regardless of their location. The PWT cells displayed significantly weaker immunoreactivity than MPNSTs to markers used except α-SMA and desmin. Cluster analysis sorted most canine MPNSTs and PWTs into 2 distinctly different clusters, whereas 3 MPNSTs and 6 PWTs were assigned to the opposing cluster. These 3 MPNSTs were negative for almost all markers, while these 6 PWTs were positive for only neuronal markers. In particular, NGFR and Olig2 were almost negative in the rest of PWT cases. These findings suggest that NGFR and Olig2 are useful to distinguish these 2 tumors.

Reports of primary nervous system tumors in wild raccoons are extremely rare. Olfactory tumors were diagnosed postmortem in 9 free-ranging raccoons from 4 contiguous counties in California and 1 raccoon from Oregon within a 26-month period between 2010 and 2012. We describe the geographic and temporal features of these 10 cases, including the laboratory diagnostic investigations and the neuropathologic, immunohistochemical, and ultrastructural characteristics of these tumors in the affected animals. All 9 raccoons from California were found within a localized geographic region of the San Francisco Bay Area (within a 44.13-km radius). The tight temporal and geographic clustering and consistent anatomic location in the olfactory system of tumor types not previously described in raccoons (malignant peripheral nerve sheath tumors and undifferentiated sarcomas) strongly suggest either a common cause or a precipitating factor leading to induction or potentiation of neuro-oncogenesis and so prompted an extensive diagnostic investigation to explore possible oncogenic infectious and/or toxic causes. By a consensus polymerase chain reaction strategy, a novel, recently reported polyomavirus called raccoon polyomavirus was identified in all 10 tumors but not in the normal brain tissue from the affected animals, suggesting that the virus might play a role in neuro-oncogenesis. In addition, expression of the viral protein T antigen was detected in all tumors containing the viral sequences. We discuss the potential role of raccoon polyomavirus as an oncogenic virus.
