AIMS: The study aimed to assess the antihyperglycemic activity of Pulicaria mauritanica. BACKGROUND: Pulicaria mauritanica is a medicinal and aromatic plant used for the treatment of many diseases such as inflammation, diabetes, and intestinal disorders. OBJECTIVE: The main goals of this present paper were to confirm the antihyperglycemic capacity of aqueous extract from Pulicaria mauritanica in normoglycemic and diabetic rats over a period of time (7 days of treatment). METHODS: The effect of the aqueous extract of Pulicaria mauritanica from aerial parts (AEPM) on glucose and lipid metabolism was tested using an acute test (single dose during 6 hours) and subchronic assay (repeated oral administration for seven days) at a dose of 60 mg/kg and the serum glucose levels were measured in normoglycemic and streptozotocin(STZ)-induced diabetic rats. In addition, the glycogen content in the liver, extensor digitorum longus (EDL), and soleus was evaluated. The antioxidant activity, phytochemical screening, and quantification of some secondary metabolites of this extract were also performed. RESULTS: AEPM at a dose of 60 mg/kg reduced the plasma glucose concentrations significantly in STZ-induced diabetic rats after a single oral administration (p<0.05). This lowering effect became more significant during the repeated oral administration in hyperglycemic rats (p<0.0001). Also, the findings showed that this plant exhibited a significant increase in liver and skeletal soleus muscle glycogen content in diabetic rats. AEPM revealed a remarkable antioxidant activity in addition to the presence of polyphenol compounds such as flavonoids, tannins, saponins, sterols, glucides, terpenoids, quinones, anthraquinones, and mucilage. CONCLUSION: The study shows that AEPM exhibits antihyperglycemic activity in diabetic rats, and it increases liver and muscle glycogen content.

BACKGROUND AND OBJECTIVE: The use of herbicides leads to severe damage, especially to the environment. The aim of this investigation was operated to study the allelopathic effect of Calotropis procera, Hyoscyamus muticus and Pulicaria undulata aqueous extracts on the seed germination of Portulaca oleracea and Chenopodium murale. MATERIALS AND METHODS: The aerial parts of the plants (Calotropis procera, Hyoscyamus muticus and Pulicaria undulata) washed well in distilled water and were dried in room temperature then ground to fine powder. Aqueous extract was prepared using distilled water. RESULTS: In this study, different concentrations of plant extracts (1, 3 and 5%) were used to illustrate which of these concentrations have the potential to inhibit seed growth of Portulaca oleracea and Chenopodium murale. Concentration of 5% for all plant extracts used to inhibited the growth of all seeds of Chenopodium murale while, concentration of 5% for Pulicaria undulata extract only inhibited the growth of all seeds of Portulaca oleracea. CONCLUSION: This work dealt with the use of allelopathy strategy in preventing the growth of some weeds to be one of the safest alternative ways of using herbicides. The results showed the effective effect of plant extracts for both Calotropis procera, Hyoscyamus muticus and Pulicaria undulata on the seed germination of Portulaca oleracea and Chenopodium murale.

BACKGROUND: Pulicaria crispa (P. crispa) is a plant from the Compositae family that exhibits antioxidant, anti-inflammatory, antibacterial, and cytotoxic activities. OBJECTIVE: The current study aimed at investigating the immunomodulatory effects of P. crispa extract in lipopolysaccharide- (LPS-) stimulated human monocytic THP-1 cells. METHODS: To induce macrophage differentiation, THP-1 cell lines were treated with phorbol-12-myristate 13-acetate, followed by exposure to LPS with or without 50 or 100 µg/ml of P. crispa extract. The following tests were employed to test the immunomodulatory effects of the extract: MTT assay, ELISA, Western blotting analysis, cell migration and phagocytosis assays, and Annexin V staining method. RESULTS: Exposure to 100 µg/ml P. crispa extract significantly reduced THP-1 cell proliferation, migration, and phagocytosis (in LPS-stimulated cells, but not in unstimulated cells). Moreover, the extract alone significantly reduced the rate of THP-1 cell apoptosis, while it increased the rate of late apoptosis. Molecular investigations showed that treatment with P. crispa extract significantly upregulated the expression of ERK1, p-MAPK, P-P38, and Bcl2, while it significantly reduced the expression of ERK5, Bax, NF-κB, P-NF-κB, CCL1, CCL2, CCL5, CCL22, CXCL1, and CXCL10. CONCLUSION: Pulicaria crispa extract exhibited anti-inflammatory, antiproliferative, antimigratory, and antiphagocytic effects in LPS-stimulated THP-1 cells. Future studies should investigate these mechanisms in animal models with chronic inflammatory diseases.

The lack of an effective chemotherapy for treatment of protozoan disease urges a wide investigation for active compounds, and plant-derived compounds continue to provide key leads for therapeutic agents. The current study reports the in vitro antiprotozoal evaluation of the Algerian medicinal plant Pulicaria inuloides against Leishmania amazonensis, Trypanosoma cruzi, and Acanthamoeba castellanii str. Neff. All the extracts from the aerial part showed to be present a higher leishmanicidal activity than anti-Acanthamoeba or Trypanosoma. Therefore, bioguided fractionation of the active CHCl3 extract led to the isolation and characterization of the flavonol, quercetagetin-3,5,7,3'-tetramethyl ether (1) as the main component. The structure of compound 1 was established by extensive 1D and 2D NMR spectroscopic analysis (COSY, HSQC, HMBC, and ROESY experiments), chemical transformation (derivatives 2 and 3), and comparison with data in the literature. Compound 1 and derivatives 2 and 3 were further evaluated against the promastigote and amastigote stage of L. amazonensis. Compounds 1-3 exhibited moderate leishmanicidal activity with IC50 values ranging from 0.234 to 0.484 mM and from 0.006 to 0.017 mM for the promastigote and amastigote forms, respectively, as well as low toxicity levels on macrophages (CC50 ranging from 0.365 to 0.664 mM). This study represents the first report of the antiprotozoal evaluation of Pulicaria inuloides, and the results highlight this species as a promising source of leishmanicidal agents.

In recent years, antimicrobial activities of essential oils have been intensively explored, mainly in researching and developing new antimicrobial agents to overcome microbial resistance. The present study investigates the chemical composition and antimicrobial activities of essential oils obtained from two Asteraceae: Asteriscus graveolens (Forssk.) Less. and Pulicaria incisa (Lam.) DC. Chemical analysis was performed using a combination of capillary GC-FID and GC/MS analytical techniques. The major component of Asteriscus graveolens were cis-chrysanthenyl acetate (31.1%), myrtenyl acetate (15.1%), and kessane (11.5%), while for Pulicaria incisa the main components were chrysanthenone (45.3%) and 2,6-dimethylphenol (12.6%). The oils obtained from the aerial parts were tested against sixteen microbial strains by agar well diffusion technique and dilution methods and showed minimum inhibitory concentrations (MIC) in the range of 19 - 1250 µg/ml. A good antibacterial activity against a common nosocomial pathogen, Acinetobacter baumanniiATCC 19606 was observed, especially from Pulicaria incisa essential oil, with a MIC value up to 19 µg/ml. These results give significant information about the pharmacological activity of these essential oils, which suggest their benefits to human health, having the potential to be used for medical purposes.

Levels of obesity in Middle Eastern countries are increasing. Phytochemicals have anti-obesogenic properties as evidenced by prevention of adipocyte differentiation and blocking triacylglyceride (TG) accumulation. In Yemen, Pulicaria jaubertii E. Gamal-Eldin (PJ) is a food additive and a traditional medicine. We tested the hypothesis that phytochemicals present in PJ inhibit adipocytic responses during differentiation of 3T3-L1 preadipocytes to adipocytes. Methanolic extracts of PJ did not block expression of fatty acid binding protein 4 (FABP4) a marker of differentiation but did inhibit TG accumulation. Treatment of 3T3-L1 preadipocytes increased NADPH:quinone oxidoreductase 1 (NQO1), a suppressor of TG accumulation. Further fractionation of the methanolic PJ extract with hexane and dichloromethane (DCM) demonstrated that bioactivity towards TG reduction and elevated expression of NQO1 and other antioxidant genes (glutamate cysteine ligase catalytic unit, glutathione disulfide reductase, glutathione peroxidase (GPx) 4 resided in the DCM fraction. Activity towards depleting GSH and elevating the expression of catalase and GPx3 were found in the DCM and hexane fractions. Analysis by gas chromatography and liquid chromatography coupled with mass spectrometry demonstrated the presence of catechin-like moieties in the DCM and methanolic fractions and suggest that these components were partially responsible for the bioactivity of these fractions. In summary, our data indicate that fractions derived PJ exhibit anti-adipogenic properties in part through the presence of catechin-like compounds.

A green, one-step approach for the preparation of graphene/Ag nanocomposites (PE-HRG-Ag) via simultaneous reduction of both graphene oxide (GRO) and silver ions using Pulicaria glutinosa plant extract (PE) as reducing agent is reported. The plant extract functionalizes the surfaces of highly reduced graphene oxide (HRG) which helps in conjugating the Ag NPs to HRG. Increasing amounts of Ag precursor enhanced the density of Ag nanoparticles (NPs) on HRG. The preparation of PE-HRG-Ag nanocomposite is monitored by using ultraviolet-visible (UV-Vis) spectroscopy, powder X-ray diffraction (XRD), and energy dispersive X-ray (EDX). The as-prepared PE-HRG-Ag nanocomposities display excellent surface-enhanced Raman scattering (SERS) activity, and significantly increased the intensities of the Raman signal of graphene.

The green synthesis of metallic nanoparticles (NPs) has attracted tremendous attention in recent years because these protocols are low cost and more environmentally friendly than standard methods of synthesis. In this article, we report a simple and eco-friendly method for the synthesis of silver NPs using an aqueous solution of Pulicaria glutinosa plant extract as a bioreductant. The as-prepared silver NPs were characterized using ultraviolet-visible spectroscopy, powder X-ray diffraction, transmission electron microscopy, energy-dispersive X-ray spectroscopy, and Fourier-transform infrared spectroscopy. Moreover, the effects of the concentration of the reductant (plant extract) and precursor solution (silver nitrate), the temperature on the morphology, and the kinetics of reaction were investigated. The results indicate that the size of the silver NPs varied as the plant extract concentration increased. The as-synthesized silver NPs were phase pure and well crystalline with a face-centered cubic structure. Further, Fourier-transform infrared spectroscopy analysis confirmed that the plant extract not only acted as a bioreductant but also functionalized the NPs' surfaces to act as a capping ligand to stabilize them in the solvent. The developed eco-friendly method for the synthesis of NPs could prove a better substitute for the physical and chemical methods currently used to prepare metallic NPs commonly used in cosmetics, foods, and medicines.

Two new compounds, the sesquiterpene (1E,5E)-8ß-acetoxy-4α-hydroxy-7ßH-germacra-1(10),5-dien-14-oic acid (2), and a nor-sesquiterpene, (5E)-8ß-acetoxy-4α-hydroxy-7ßH-germacr-5-en-10-one (3), were isolated from Pulicaria canariensis ssp. lanata, along with ten known compounds, including the flavonoid 5,3'-dihydroxy-3,7,4'-trimethoxyflavone (4). From Pulicaria burchardii, we isolated seven known compounds; the physical and spectroscopic data of the triterpenoid 3ß-hydroxytaraxaster-20-en-30-al (1) are reported. The structures of compounds 1-3 were determined on the basis of HR-MS, and 1D- and 2D-NMR studies. The structure of 2 was corroborated by X-ray crystal diffraction. Cell viability experiments revealed that the semisynthetic flavonoid 4b was the most cytotoxic compound against human leukemia cells, and the cytotoxicity was caused by induction of apoptosis, as determined by microscopy of nuclear changes.
