RESUMO

Our laboratory has purchased four LaCl (3)(Ce) scintillation crystals over the course of the last year for evaluation as part of an environmental management research and development project. In addition to the expected content of naturally radioactive (138)La, we have found that all four of these detectors are contaminated to various degrees with alpha particle emitting nuclides that we have determined to be (227)Ac and its daughters. The impact of these radionuclides on the background spectra and, thus, on the detection sensitivity of these detectors is presented and discussed.

RESUMO

We propose practices to optimize the quality of acquired gamma-ray spectra at a radioanalytical laboratory and to assist in the identification of extraneous spectral peaks including those from limitations in the physical setup, electronics, and counting conditions. This paper offers visible comparisons of high-quality spectra with those of inferior quality. We demonstrate the impacts on gamma-ray spectra taken with germanium (HPGe) detectors due to detector size, shape and energy resolution, background radiation, high-energy beta- or beta+ decay in the sample, beta(-)-, beta(+)- and gamma-ray backscatter, pole-zero cancellation, pulse pileup, coincidence summing, and unwanted sources of radiation.

A non-destructive technique to determine the time since last purification of weapons grade plutonium based on the 241Pu decay series has been developed and demonstrated to provide results that are consistent with our historical and process knowledge. This method is based on the use of the Bateman equations and the decay chain 241Pu-->237U & 241Am-->237Np-->233Pa. Secular equilibrium is assumed between the (2.1 x 10(6) year) 237Np and the (27 day) 233Pa. The proposed method is dependent upon the efficiency of the plutonium separation process but makes no assumption about the relative abundance of 241Pu in 239Pu. Even though the technique was developed for weapons grade plutonium, it is expected to apply to almost any material that contains plutonium.

Population of the 6+ 332 keV level in 232Th from the decay of 236U was observed for the first time. The 49, 112, and 171 keV gamma-ray energies and relative intensities from the decay of 236U were measured.

RESUMO

The combination of grazing incidence small angle x-ray scattering (GISAXS) and grazing incidence wide angle x-ray scattering (GIWAXS) with optical imaging ellipsometry is presented as an upgrade of the available measurement techniques at the wiggler beamline BW4 of the Hamburger Synchrotronstrahlungslabor. The instrument is introduced with the description of the alignment procedure to assure the measurement of imaging ellipsometry and GISAXS∕GIWAXS on the same sample spot. To demonstrate the possibilities of the new instrument examples of morphological investigation on films made of poly(3-hexylthiophene) and [6,6]-phenyl-C(61) butyric acid methyl ester as well as textured poly(9,9-dioctylfluorene-alt-benzo-thia-diazole) are shown.

RESUMO

We present a real-time study of the nanostructuring and cluster formation of gold nanoparticles deposited in aqueous solution on top of a pre-structured polystyrene colloidal thin film. Cluster formation takes place at different length scales, from the agglomerations of the gold nanoparticles to domains of polystyrene colloids. By combining in situ imaging ellipsometry and microbeam grazing incidence small-angle x-ray scattering, we are able to identify different stages of nanocomposite formation, namely diffusion, roughness increase, layer build-up and compaction. The findings can serve as a guideline for nanocomposite tailoring by solution casting.

We present an upgrade of the available measurement techniques at the wiggler beamline BW4 of the Hamburger Synchrotronstrahlungslabor (HASYLAB) to grazing incidence wide angle x-ray scattering (GIWAXS). GIWAXS refers to an x-ray diffraction method, which, based on the measurement geometry, is perfectly suited for the investigation of the material crystallinity of surfaces and thin films. It is shown that the overall experimental GIWAXS setup employing a movable CCD-detector provides the capability of reliable and reproducible diffraction measurements in grazing incidence geometry. Furthermore, the potential usage of an additional detector enables the simultaneous or successive measurement of GIWAXS and grazing incidence small angle x-ray scattering (GISAXS). The new capability is illustrated by the microbeam GIWAXS measurement of a thin film of the conjugated polymer poly(3-octylthiophene) (P3OT). The investigation reveals the semicrystalline nature of the P3OT film by a clear identification of the wide angle scattering reflexes up to the third order in the [100]-direction as well as the first order in the [010]-direction. The corresponding microbeam GISAXS measurement on the present morphology complements the characterization yielding the complete sample information from subnanometer up to micrometer length scales.

Casein films were successfully prepared with the spin-coating technique of aqueous casein solutions on base-treated glass surfaces. The film structure is investigated in real space with optical microscopy and atomic force microscopy and for the first time in reciprocal space with grazing incidence small-angle X-ray scattering (GISAXS). The size of the substructures detected in the film increases with pH from 170 nm (pH 5.1) up to 490 nm (pH 9.4). Dynamic light scattering experiments reveal that the average diameters of casein micelles in solution exhibit the same quantitative increase. This result suggests that the substructures detected in the bulklike films with GISAXS reflect intact casein micelles. However, with thin homogeneous casein films, the micelle size diminishes with decreasing film thickness. This indicates that the moderate pressures introduced by spin-coating force the micelles to rearrange into a more compact structure.

Self-supported particle-track-etched polycarbonate membranes with nearly perfect cylindrical pores are used for the preparation of similarly perfect cylindrical polypyrrole nanowires and nanotubes. A complete investigation of the structural properties that result at different stages of the preparation route of polypyrrole nanowires and nanotubes is based on a combination of real and reciprocal space techniques. Nanoporous membranes with nominal pore size ranging from 5 to 150 nm and pore density up to 10(9) pores/cm(2) made from 21-microm-thick polycarbonate films are used. Polypyrrole nanotubes or nanowires are synthesized inside the pores. A real-space picture of the nanomaterial results from scanning force microscopy (SFM) on ultrathin sections made in two directions to obtain structures in the sample surface as well as perpendicular to the surface. From a model-based fit to the small-angle and ultra-small-angle X-ray scattering (SAXS/USAXS) data, the geometric pore structure is obtained and compared to values determined with scanning electron microscopy (SEM). Nanopores, nanowires, and nanotubes are described by uniform solid cylinders or hollow tubes, which are oriented highly parallel to each other and exhibit a small size distribution. Below a critical pore diameter, solid nanowires are produced whereas above this limit hollow nanotubes result.

Cefotaxime, given intravenously, is currently used as a broad-spectrum antibiotic for prophylaxis of intra- and postoperative infections in ocular lens surgery. A proposed therapeutic and economic alternative is the use of orally active fluoroquinolone ofloxacine as prophylactic agent. A HPLC method was developed for determination of both antibiotics in ocular aqueous humor and plasma in order to optimize dosage for safe surpassing minimal inhibitory concentration in the humor compartment. For plasma determinations a solid-phase extraction procedure was used with ciprofloxacine as internal standard. Detection limits for direct HPLC-analysis of ocular aqueous humor was 0.08 microg/ml for all compounds, whereas in plasma 0.31 microg/ml could be determined after solid-phase extraction.

Isolated reactivity to hepatitis B virus (HBV) core antigen (anti-HBc) is observed relatively frequently in immunocompromised individuals, intravenous drug abusers (IVDA), and in the presence of HCV infection. The reason for the lack of HBsAg is not clear. The aim of the present study was to investigate which factors (genetic variability of S gene, low-level HBsAg, and immune complexes may be responsible for the failure of HBsAg detection with commercial HBsAg screening assays. Dilution series of two recombinant HBsAg escape mutants and dilutions of serum samples from chronic HBV carriers with multiple insertions in the a determinant and different HBsAg subtypes were tested with a highly sensitive assay that detects wild-type HBsAg (Elecsys HBsAg, Roche Diagnostics, Penzberg, Germany) and two assays that detect HBV wild-type and escape mutants (Murex HBsAg Version 3, Murex and Enzygnost HBsAg 5.0, Dade Behring, Marburg, Germany). Elecsys HBsAg showed in comparison to Murex HBsAg Version 3 and Enzygnost HBsAg 5.0 a reduced sensitivity for escape mutant detection. On the other hand, the best performance for HBsAg subtype detection was obtained with Elecsys HBsAg. In the second part of the study, a selected panel of isolated anti-HBc reactive (n = 104) serum samples (AxSYM Core) was submitted to testing by Elecsys HBsAg, Murex HBsAg Version 3, Enzygnost HBsAg 5.0, and HBsAg detection after immune complex dissociation (ICD) and anti-HBs determination with two different assays (AxSYM Ausab and Elecsys Anti-HBs). To assess the specificity of anti-HBc test results, all the samples were tested by a second anti-HBc assay (Elecsys Anti-HBc). Quantitative HBV DNA detection was undertaken with a commercially available HBV PCR assay (Amplicor HBV Monitor). HCV infection was present in 65.4% of anti-HBc only reactive individuals. Five AxSYM Core positive samples were negative by Elecsys Anti-HBc. Overall, 15 (14.4%) AxSYM Ausab negative samples gave positive results with Elecsys Anti-HBs (median value: 21 IU/ml). No low-level HBsAg carrier was detected among the isolated anti-HBc reactive individuals with Elecsys HBsAg. There was no evidence for the presence of immune complexes. Only one sample was repeatedly reactive by the Murex HBsAg, suggesting that the a mutant form of HBsAg was responsible for the isolated anti-HBc reactivity, however neutralisation assay was not interpretable and HBV DNA PCR was negative. Fifteen (14.4%) anti-HBc only positive individuals were HBV DNA carriers with concentrations ranging from 800 to more than >4,000,000 copies of viral DNA/ml. In conclusion, the most probable explanations for isolated anti-HBc reactivity in our study group are a possible interference of HBsAg synthesis by HCV infection (65.4%) and divergence of results of anti-HBs assays (14.4%). There is no evidence for the presence of low-level HBsAg carriers and immune complexes. HBsAg mutants cannot be excluded definitively by the test strategy used in the present evaluation.

The administration, in a double blind experiment, of piracetam to two homogeneous groups of 32 and 34 eight- to twelve-year-old brain-injured special education children lacking normal mental ability, where the daily doses administered for a period of thirteen weeks were 3 tablets or 1.2 g (administered to second and third graders) and 5 tablets or 2.0 g (administered to fourth graders), respectively, did not produce a lasting effect upon the capacity to acquire and apply knowledge, the concentration power, the proficiency level, and the behavior shown by pupils during and out of school hours. On the other hand, there was observed, in patients receiving placebos, a nonsignificant tendency toward improvement in physical well-being, so that the effect produced by the drug may here be considered to be a negative one.

Particle track etched polyimide membranes on silicon substrates covered with a native oxide layer are investigated. Preparation steps similar to the common classical particle track etched membrane production, giving rise to free-standing membranes, are successfully applied to the supported membranes. Polyimide films are used as a starting material for a template preparation based on high energy ion irradiation. The film/membrane structure is probed at different length scales by grazing incidence small-angle X-ray scattering at each individual preparation step. In addition, characterization with atomic force microscopy, variable-angle spectroscopic ellipsometry, Fourier transform infrared transmission, and attenuated total reflection spectroscopy is performed. An amount of 6 +/- 1 vol % pores inside the polyimide film is detected. The pores are oriented perpendicular to the substrate surface and have a conical shape, yielding a slightly reduced pore size at the substrate/film interface.

Antibody to the common "a" determinant of hepatitis B surface antigen (HBsAg) protects against infection with hepatitis B virus. A number of variant surface antigens with amino acid substitutions within the "a" determinant have been described in patients around the world. Both wild type and variant HBsAgs were expressed in the yeast Pichia pastoris and the antigens were semi-purified and quantitated. The effect on antigenicity of these changes was investigated in a quantitative fashion using four monoclonal antibodies known to bind to different epitopes within the common "a" determinant. The results suggest that amino acid substitution of T131I, K141E and G145R and insertion of 3 amino acids between residues 123 and 124 markedly affect the antigenic structure of HBsAg. These substitutions and insertions in the viral envelope may lead to evasion of the virus neutralizing antibody response and also to reduce efficiency of detection by immunoassays used for diagnosis and blood-bank screening.

The decay of 182Hf, now extinct, into stable 182W has developed into an important chronometer for studying early solar system processes such as the accretion and differentiation of planetesimals and the formation of the Earth and the Moon. The only 182Hf half-life measurements available were performed 40 years ago and resulted in an imprecise half-life of (9+/-2)x10(6) yr. We redetermined the half-life by measuring the specific activity of 182Hf based on two independent methods, resulting in a value of t(1/2)(182Hf)=(8.90+/-0.09)x10(6) yr, in good agreement with the previous value, but with a 20 times smaller uncertainty. The greatly improved precision of this half-life now permits very precise intercalibration of the 182Hf-182W isotopic system with other chronometers.
