Flowers, leaves, fruits and buds of Tropaeolum majus are used for ornamental, medicinal and food purposes. However, salt stress limits the development and productivity of T. majus due to biochemical, physiological and anatomical disturbances. Polyamine application is an alternative for mitigating the harmful effects of salt stress. Thus, the objective of this work was to evaluate the effects of spermine application in T. majus grown under salt stress. The experiment was carried out in a completely randomized design, in a 3 × 2 factorial scheme, with 0, 40 mM (moderate salt stress) and 80 mM (severe salt stress) NaCl, and 0 and 1 mM spermine, and with five replicates. Growth (plant height, stem diameter, number of leaves, number of flowers, number of buds, leaf dry mass, stem dry mass and flower dry mass), gas exchange (gs, A, E, Ci and WUE), relative water content, contents of free amino acids, phenolic compounds, reducing and non-reducing sugars, lipid peroxidation and enzymatic activities (CAT, POD and APX) were evaluated. Spermine application decreased the harmful effects of salt stress on the growth and gas exchange and increased flowering in T. majus. Furthermore, the relative water content of T. majus increased under severe salt stress conditions. Spermine application reduced the contents of total phenolic compounds, free amino acids, reducing sugars and non-reducing sugars on leaves of T. majus. Spermine application increased CAT and POD activities in plants under severe salt stress and POD and APX in plants under moderate salt stress.

This study aimed to explore the influence of sound stimulation on heart rate and the potential coupling between cardiac and cerebral activities. Thirty-one participants underwent exposure to periods of silence and two distinct continuous, non-repetitive pure tone stimuli: low pitch (110 Hz) and high pitch (880 Hz). Electroencephalography (EEG) data from electrodes F3, F4, F7, F8, Fp1, Fp2, T3, T4, T5, and T6 were recorded, along with R-R interval data for heart rate. Heart-brain connectivity was assessed using wavelet coherence between heart rate variability (HRV) and EEG envelopes (EEGE). Heart rates were significantly lower during high and low-pitch sound periods than in silence (p < 0.002). HRV-EEGE coherence was significantly lower during high-pitch intervals than silence and low-pitch sound intervals (p < 0.048), specifically between the EEG Beta band and the low-frequency HRV range. These results imply a differential involvement of the frontal and temporal brain regions in response to varying auditory stimuli. Our findings highlight the essential nature of discerning the complex interrelations between sound frequencies and their implications for heart-brain connectivity. Such insights could have ramifications for conditions like seizures and sleep disturbances. A deeper exploration is warranted to decipher specific sound stimuli's potential advantages or drawbacks in diverse clinical scenarios.

AIM: To verify whether moderate physical training affects the muscle fibre composition of adult rats subjected to a low protein diet during the perinatal period. METHODS: Male Wistar rats were divided into two groups according to their mother's diet during gestation and lactation: control (17% casein, C) and low-protein (8% casein, LP). On postnatal day 60, half of each group was submitted to moderate physical training (8 weeks, 5 days/week(-1), 60 min/day(-1), at 70% of VO(2max), T) or not. After the physical training period, soleus and extensor digitorum longus (EDL) muscles were removed. Myofibrillar ATPase staining was used to classify muscle fibres as type I, IIa, IIb, and intermediate. RESULTS: In the EDL muscle, LP rats showed no changes in the fibre type proportion. Both the C + T and LP + T groups showed a higher percentage of fibres of type IIa, and a lower proportion of fibres of type IIb. In the soleus muscle, LP animals showed a reduction in the proportion of fibre types I and intermediate. C + T rats showed an increase in the fibre type I and IIa. In the LP + T rats, the proportions of the fibre types remained similar to control rats. CONCLUSIONS: Moderate physical training acts as a positive environmental stimulus that reverts the effects of a perinatal low-protein diet on the proportion of fibre types in skeletal muscle.

BACKGROUND: We aimed to investigate the effect of rest interval, between successive contractions, on muscular fatigue. METHODS: Eighteen subjects performed elbow flexion and extension (30 repetitions) on an isokinetic dynamometer with 80º of range of motion. The flexion velocity was 120º/s, while for elbow extension we used 5 different velocities (30, 75, 120, 240, 360º/s), producing 5 different rest intervals (2.89, 1.28, 0.85, 0.57 and 0.54 s). RESULTS: We observed that when the rest interval was 2.89 s there was a reduction in fatigue. On the other hand, when the rest interval was 0.54 s the fatigue was increased. CONCLUSIONS: When the resting time was lower (0.54 s) the decline of work in the flexor muscle group was higher compared with different rest interval duration.

The influence of creatine (Cr) supplementation on cortical and trabecular bone from ovariectomized rats was studied using FT-Raman spectroscopy. The intensity of organic-phase Raman bands was compared to mineral phase ones. Twenty-one female Wistar rats aged 3 months were divided into three groups (n = 7 per group): ovariectomized (OVX), ovariectomized treated with creatine (CRE) and sham-operated (SHAM) groups. Creatine supplementation (300 mg kg(-1) day(-1)) was provided for 8 weeks, starting 12 weeks after ovariectomy. FT-Raman spectroscopy was performed on the right medial femoral mid-shaft (cortical bone) and third lumbar vertebral body (trabecular bone). The integrated intensities of mineral phase (phosphate and carbonate bands at 959 and 1,071 cm(-1), respectively) and organic phase (amide I band at 1,665 cm(-1)) Raman bands were analyzed. The mineral-to-matrix (phosphate/amide I), carbonate-to-phosphate, and carbonate-to-amide I ratios were analyzed to assess bone quality. The phosphate content on trabecular bone was higher in the CRE group than the OVX group (p < 0.05). No significant changes in mineral or organic phases on cortical bone were observed. A radiographic assessment of bone density was encouraging as the same findings were showed by Raman intensity of phosphate from cortical (r(2) = 0.8037) and trabecular bones (r(2) = 0.915). Severe ovariectomy-induced bone loss was confirmed by FT-Raman spectroscopy. The results suggest that the chemical composition of trabecular bone tissue may be positively influenced by Cr supplementation after ovariectomy.

Joint hypermobility (JH) conditions suggest dysfunction in the autonomic nervous system (ANS) (dysautonomia), associated with multifactor non-articular local musculoskeletal pain, and remains a complex treatment. This study aims to determine the effects of musculoskeletal interfiber counterirritant stimulation (MICS) as an innovative treatment of myofascial trigger points (MTrPs) on the upper trapezius muscle in JH patients. We evaluate the ANS activity by wavelet transform spectral analysis of heart rate variability (HRV) in sixty women, equally divided: MTrP, MTrP + general joint hypermobility (GJH), and MTrP + joint hypermobility syndrome (JHS). The protocol phases were rest, stimulation, and recovery, with clinical and home treatment for three-days. All groups show a significantly decreased in pain perception during and post-treatment, and an increased parasympathetic ANS activity under MICS in the GJH and JHS groups. The variables low-frequency (LF) vs. high-frequency (HF) showed significant differences during the protocol phases, and the LF/HF ratio maintained a predominance of sympathetic activity (SA) throughout the protocol. The new MICS technique reduces the pain perception and modulates the ANS activity by an increase in vagal tone, and a decrease in sympathetic tone. This modulation was followed by an increase in the HRV in JH patients after treatment with MICS. Clinical Trials: RBR-88z25c5.

The solar dryer can reduce production costs, energy consumption, waste (use fruits outside the quality standard for fresh consumption) and is an alternative for small and medium producers. The solar dryer can reduce costs and is an alternative for small and medium producers worldwide. The consumption of fresh and processed tomatoes is high in the world, but post-harvest losses is also and drying is an alternative to reduce these losses. The temperature maintenance and drying time corresponds 30% of the costs. The objective was evaluated the tomato physicochemical characteristics after drying in handmade solar dryer. 'Carmen' tomato fruits were bleached in water, 2.5% NaCl solution, 2.5% NaCl + 0.5% CaCl2 solution and unbleached. Tomato slices were placed in a handmade solar dryer from 7:00 to 17:00. The solar dryer prototype was wood made, comprising a collector and a drying chamber. The average cost of the camera was US$ 13.08 (1 Brazilian Real = 0.26 United States Dollar). Water loss, drying kinetics, mathematical models and physicochemical characteristics of fresh and dried tomatoes were evaluated. The average length of solar drying for the four treatments was 30 h and the Midilli and Kucuk mathematical model was the most adjusted. The acidity, reducing sugars and soluble solids were concentrated by drying, while ascorbic acid was reduced. The pH did not change. Tomatoes 'Carmen' can be dried in a handmade solar dryer for 30 h while maintaining product quality.

High seed production makes Sporobolus indicus var. pyramidalis a difficult to control invasive grassland plant. The objective of the present study was to investigate the bioactivity of Cyperus rotundus, Phyllanthus tenellus and Ricinus communis green leaf extracts and of Carica papaya seeds on S. indicus germination without breaking dormancy, simulating the field conditions. The ethanolic extract bioactivity of C. rotundus, P. tenellus, R. communis green leaves and C. papaya seeds, at concentrations of 25, 50 and 75% in S. indicus germination was evaluated. Carotenoids, flavonoids, soluble phenolic compounds and total tannins were quantified in the extracts. The chemical component concentrations varied between alcoholic extracts. The P. tenellus extracts at all dilutions and those of R. communis and C. papaya at 75% completely suppressed S. indicus seed germination at five and ten days which can be attributed to their high tannin concentration, total phenolic compounds and flavonoids.

This study evaluated the effects of high-dose of short-term creatine supplementation (5g.kg(-1).day(-1) to 1 week) and long-term creatine supplementation (1g.kg(-1). day(-1) to 4-8 weeks) on kidney and liver structure and function of sedentary and exercised Wistar rats (Exercise sessions consisted of swimming at 80% of maximal work load supported during 5 days per week with daily sessions of 60 minutes throughout the duration of the supplementation). Seventy- two animals (245 ± 5g) were divided into four groups (n = 18): control diet Sedentary (SED), Creatine diet Sedentary (CRE), control diet Exercised (EXE), and Creatine diet Exercised (EXECRE). Histological and blood biochemical studies were performed after one, four, and eight weeks of creatine supplementation and exercise (n = 6). No differences were found when comparing SED, EXE and EXECRE groups for kidney and liver structure and function at one, four and eight weeks. However, the CRE group showed higher levels of creatinine (1.1 ± 0.2 vs. 0.4 ± 0.1 mg.dl(-1); p < 0.05), and urea (37 ± 3 vs. 19 ± 1 mg.dl(-1); p < 0.05) when compared with all others groups at four and eight weeks. At eight weeks, the CRE group presented increased levels of ALT (41 ± 7 vs. 23 ± 7 U.L(-1); p < 0.05), AST (89 ± 6 vs. 62 ± 5 U.L(-1); p < 0.05), GGT (8.0 ± 0.9 vs. 3.9 ± 1.0 U.L(-1); p < 0.05), and AP (125 ± 10 vs. 69 ± 9 U.L(-1); p < 0.05) also when compared with all others groups. Moreover, the CRE group demonstrated some structural alterations indicating renal and hepatic damage at four and eight weeks, respectively. These results suggest that long-term creatine supplementation (up to 4-8 weeks) may adversely affect kidney and liver structure and function of sedentary but not of exercised rats. Key pointsCreatine supplementation is an established ergogenic aid in sports and is now claimed to have therapeutical applications in a variety of diseases.Although acknowledged, this nutritional supplement is rarely monitored precisely about their possible side effects.Previous studies indicated that short-term creatine supplementation associate with the physical exercise may be safe, but the effect of long-term creatine supplementation is still unknown.There is a need for further research to elucidate the controversial points refers to renal and hepatic function after creatine supplementation.The results of the current study indicate that supraphysiological long-term creatine supplementation (up to 4-8 weeks) may adversely affect kidney and liver structure and function of sedentary but not of exercised rats.

BACKGROUND: The aim of this study is to assess the effect of pre-pregnancy physical training on metabolic responses and its effects on offspring. METHODS: Three groups of rats (n = 7 in each group): sedentary pregnant rats (PS), exercised during pregnancy (PE) and pregnant rats trained before and during pregnancy (PT) were compared. They were separated into three subgroups regarding water temperature: 28°C, 35°C or 39°C. Plasma triglycerides and glucose levels, weight gain during pregnancy and rectal temperature pre and post exercise (swim), as well as the offspring size and weight were analysed. RESULTS: Rectal temperature post exercise was lower than pre exercise at 28°C and 35°C, and higher at 39°C. Weight gain was lower at 39°C for the PT group and at 35°C for the PT and PE groups compared to the PS group. Plasma glucose, at 28°C and 39°C for PS and PE groups, was higher than those obtained at 35°C, while triglycerides were lower. For trained rats, plasma glucose and triglycerides were similar at all water temperatures. Trained rats presented lower triglyceride values at 35°C, and higher triglyceride values at 39°C compared to PS group. Glucose presented inverse results. None of the groups presented fetal reabsorption. However, in the PS group, the offspring presented lower weight gain at 28°C than at 35°C and 39°C. CONCLUSIONS: These results suggest that pre-pregnancy physical training induces steady values of triglycerides and glucose during exercise at all water temperatures.

Oenocarpus distichus presents economic, ecological, and dietary potential for pulp market processed in natura. Germplasm conservation and genetic improvement depend on genetic divergence studies. The objective was to quantify genetic divergence in a native population of O. distichus genotypes based on fruit biometrics. The fruit length and width, fruit mass, pulp and seed, pulp and almond thickness, and pulp yield per fruit were evaluated. All fruit biometric characteristics of O. distichus palms show genetic variability. Genetic variations among genotypes are essential for predicting heredity and heterosis, which are essential for improving O. distichus production. Pulp yield and seed mass were negatively correlated. Almond thickness and pulp, seed mass and transverse diameter were positively correlated. Genetic distances between pair of genotypes ranged from 0.07 to 48.10 with three genetically distinct groups. The seed mass, almond thickness and transverse diameter contributed to genetic divergence. Heritability estimates the genetic control that can be obtained from O. distichus germplasm. Correlations between the variable pair reduce the evaluation effort and the resources to measure the genotype allocations in heterogeneous groups presenting high genetic variability. This makes it possible to select individuals for hybridization programs with F1 generation gains. Correlation and relative contribution networks, based on relationships graphical between fruit biometric characteristics, allow the variables selection with less effort and fewer measurements. O. distichus fruit biometric characters are efficient to quantify genetic divergence between genotypes.

Bananas are usually ripened with calcium carbide (CaC2), a dangerous substance that can cause food poisoning. The objective was to test the empirical ripening banana method using Bowdichia virgilioides leaves compared to carbide. Ripening tests were carried out using 'Pacovan' banana fruits with B. virgilioides leaves and carbide following the empirical method used by Borborema farmers, Paraíba, Brazil. Bowdichia virgilioides leaves induced increased respiration and ascorbic acid production and reduced acidity, chlorophyll and pH in banana fruits like CaC2. Leaves of B. virgilioides induce ripening of 'Pacovan' banana with safer and same results than with CaC2.

Gladiolus grandiflorus L. is highly susceptible to Fusarium and losses caused by this disease varies from 60% to 100%. Injuries caused during harvest, transport and inadequate storage, facilitate infection. The dynamics of wound healing can reduce infection by Fusarium. The objective was to characterize the wound healing in corms of G. grandiflora stored under refrigeration and how it affects the entry and establishment of F. oxysporum f. sp. gladioli infection. Corms were wounded and stored at 12 ± 4°C and relative humidity of 90 ± 5%. Cell damage, fresh weight loss, respiration, phenolic compounds, tissue darkening, suberization, lignification and resistance to infection were evaluated. Wounds on corms caused transepidermal damage with collapse and cell death. Physiological (increased loss of mass and respiration) and biochemical changes (deposition of lignin and suberin, enzymatic activity) occurred in the cells neighboring those death by the injury. The injury caused gradual darkening of the tissue, injured and neighbor. Fusarium oxysporum infection decreased with wound healing. The healing of injured G. grandiflora corms stored at 12ºC occurs from the 3rd day after injury by the accumulation of suberin, lignin, and melanin, inhibiting F. oxysporum f. sp. gladioli infection.

This article reports the effect of low-level laser therapy (LLLT) on the edema formation and leukocyte influx caused by Bothrops jararacussu snake venom as an alternative treatment for Bothrops snakebites. The inflammatory reaction was induced by injection of 0.6 mg/kg of B. jararacussu venom, in gastrocnemius muscle. Cell influx and edema were evaluated at 3 or 24h after venom injection. Mice were irradiated at the site of injury by a low-level laser (685 nm) with a dose of 4.2J/cm(2). A therapy that combines LLLT and antivenom was also studied. B. jararacussu venom caused a significant edema formation 3 and 24h after its injection, and a prominent leukocyte infiltrate composed predominantly of neutrophils at 24h after venom inoculation. LLLT significantly reduced edema formation by 53% and 64% at 3 and 24h, respectively, and resulted in a reduction of neutrophils accumulation (P<0.05). The combined therapy showed to be more efficient than each therapy acting separately. In conclusion, LLLT significantly reduced the edema and leukocyte influx into the envenomed muscle, suggesting that LLLT should be considered as a potentially therapeutic approach for the treatment of the local effects of Bothrops species.

In the present work we investigated the effects of Crotalus durissus terrificus venom (CdtV) on the pulmonary mechanic events [static and dynamic elastance, resistive (DeltaP1) and viscoelastic pressures (DeltaP2)] and histology after intramuscular injection of saline solution (control) or venom (0.6 microg/g). The static and dynamic elastance values were increased significantly after 3 h of venom inoculation, but were reduced at control values in the other periods studied. The DeltaP1 values that correspond to the resistive properties of lung tissue presented a significant increase after 6h of CdtV injection, reducing to basal levels 12h after the venom injection. In DeltaP2 analysis, correspondent to viscoelastic components, an increase occurred 12 h after the venom injection, returning to control values at 24 h. CdtV also caused an increase of leukocytes recruitment (3-24 h) to the airways wall as well as to the lung parenchyma. In conclusion, C. durissus terrificus rattlesnake venom leads to lung injury which is reverted, after 24 h of inoculation.

Pasture implantation fragments and reduces the Amazonian forest area. The objective was to quantify landscape changes in 1985, 2000 and 2015 in northern Mato Grosso, Brazil. The study was carried out in three scenes obtained by the LANDSAT satellite of a microbasin (2742.33 ha) in the municipality of Alta Floresta. Forest, water bodies, pasture and exposed soil were the thematic classes determined to e mapping the land use evolution. The edge, density and shape indexes of the fragments were measured. Normalized vegetation difference (NDVI) values were high in 1985. Land use and occupation over 15 years (1985-2000) reduced forest cover by 69.8%, but it increased by 1.7% over the next 15 years (2000-2015). The number of exposed soil patches increased between the periods, but the total area and number of the patches of the forest fragments decreased. The high values of NDVI in 1985 showed vegetated areas with high density. Reducing forest cover decreases the size of the fragments, increases the isolation and the number of soil patches exposed. The mapping of land use showed a reduction of the Amazon forest in the microbasin in the north of Mato Grosso, in the years 2000 and 2015 compared to 1985.

The objective of this study was to analyze the behavior of autonomic modulation before, during and after the Modified Wingate Test (WanMT), through the analysis of Heart Rate Variability (HRV). Six volunteers between the ages of 40 and 70, post-revascularization procedures (angioplasty and/or surgery, mean duration 10 months), were submitted to supervised training for at least 10 to 14 months. The following protocol, divided into 5 phases, was used: 1) Rest Phase (RP): 180 seconds; 2) Submaximum Phase (SP): 30 seconds; 3) Maximum Phase (MP): 30 seconds; 4) Active Recuperation Phase (ARP); 120 seconds and; 5) Passive Recuperation Phase (PRP): 180 seconds. For the WanMT Test, we selected the load of 3.75% of corporal weight for all volunteers. To analyze the HRV, we used the following parameters: the interval RRr, MNN, SDNN, RMSSD and PNN50. We only observed results for the group according to RMSSD parameters during the rest phase of the test protocol in which the group remained in vagal presence and during all other phases in vagal depression. However, when we analyzed the PNN50, we observed that the group was in medium vagal presence during all of the phases of the test though there was no statistically significant difference (p> 0.05) between the phases. Therefore, we can say that all of the individuals had a similar profile in the autonomic response to the WanMT, confirmed by the parameters studied in the analysis of the HRV in the time domain.

Envenomation by the South American opisthoglyphous snake Philodryas olfersii causes local pain, edema, erythema and ecchymosis; systemic envenomation is rare. In this work, we examined the inflammatory activity of P. olfersii venom (10, 30 and 60 µg) in mouse gastrocnemius muscle 6 h after venom injection. Intramuscular injection of venom did not affect hematological parameters such as red cell count, hemoglobin, hematocrit, mean corpuscular volume, mean corpuscular hemoglobin and mean corpuscular hemoglobin concentration. The venom caused thrombocytopenia (at all three doses), leukopenia and lymphopenia (both at the two highest doses), as well as neutrophilia (30 µg), monocytosis (30 µg) and basophilia (10 µg). Of the cytokines that were screened [IL-1ß, IL-6, IL-10, IL-13, IL-17, TNF-α, IFN-γ, MIP-2 and KC] and IGF-1, only IGF-1 showed a significant increase in its circulating concentration, seen with 60 µg of venom; there were no significant changes in the cytokines compared to control mice. Histological analysis revealed the presence of edema, an inflammatory infiltrate and progressive myonecrosis. Edema and myonecrosis were greatest with 60 µg of venom, while the inflammatory infiltrate was greatest with 10 µg of venom. All venom doses caused the migration of polymorphonuclear and mononuclear leukocytes into muscle, but with no significant dose-dependence in the response. These findings show that, at the doses tested, P. olfersii venom does not cause hematological alterations and has limited effect on circulating cytokine concentrations. These data also confirm that the principal effects of the venom in mice are local edema, inflammatory cell infiltration and myonecrosis.

ABSTRACT Introduction: Currently there is a lack of clarity around the use of Fourier transform infrared (FT-IR) spectroscopy to analyze the effect of creatine (Cr) supplementation on the secondary structures of skeletal muscle tissue protein subjected to exercise. Objective: The objective of this study was to evaluate the spectral characteristics of the tibialis anterior muscle in rats subjected to exercise in a pool and to Cr supplementation. Methods: Experiment 1. First, an experiment was conducted to ensure that FT-IR would be able to detect change in the secondary structures of skeletal muscle tissue protein in the group of sedentary rats (SED) and in the group of rats that received creatine supplementation (CRE). Experiment 2. Next, the effect of physical exercise on the spectral characteristics of muscle tissue, especially when compared to the groups without exercise practice, was examined. Results: It was possible to verify that the peaks centered on 1658 cm-1 (amide I) and 1546 cm-1 (amide II) are characteristic spectra and indicated as markers of protein content. Conclusion: Thus, FT-IR spectroscopy proved to be able to monitor changes in secondary structures of skeletal muscle protein in both animals that received supplements and in those subjected to exercise and both cases reconciled. Furthermore, the FT-IR technique proved to be a viable method for the nondestructive evaluation of skeletal muscle protein structures. Level of evidence II, Investigation of treatment results.


RESUMEN Introducción: Actualmente, no hay claridad en lo que se refiere al uso de la técnica de espectroscopia de Infrarrojo con transformada de Fourier (FT-IR) para análisis del efecto de la suplementación de creatina (Cr) sobre las estructuras secundarias de la proteína del tejido muscular esquelético sometido a ejercicio. Objetivo: El objetivo de este estudio fue evaluar las características espectrales del músculo tibial anterior de ratones sometidos a ejercicio en piscina y a la suplementación con Cr. Métodos: Experimento 1. En primer lugar, fue realizada una experiencia para asegurar que la FT-IR sería capaz de detectar la variación en las estructuras secundarias de la proteína del tejido muscular esquelético en el grupo de ratones sedentarios (SED) y el grupo de ratones que sólo recibieron suplemento de creatina (CRE). Experimento 2. A continuación, fue examinado el efecto del ejercicio físico sobre las características espectrales del tejido muscular, especialmente cuando comparado con los grupos sin práctica de ejercicio. Resultados: Fue posible verificar que los picos centrados en 1658 cm−1 (amida I) y 1546 cm−1 (amida II) son espectros característicos e indicados como marcadores del tenor proteico. Conclusión: Siendo así, la técnica de espectroscopia de FT-IR mostró ser capaz de monitorizar las variaciones en las estructuras secundarias de la proteína del tejido muscular esquelético, tanto en animales que recibieron suplementos, como en los que fueron sometidos a ejercicio y ambos casos conciliados. Además, la técnica FT-IR probó ser un método viable para la evaluación no destructiva de las estructuras proteicas en el músculo esquelético. Storage temperature affects the rate and extent of wound-healing in a number of root and tuber crops. The effect of storage temperature on wound-healing in sugarbeet (Beta vulgaris L.) roots, however, is largely unknown. Wound-healing of sugarbeet roots was investigated using surface-abraded roots stored at 6 and 12°C for 28 days. Surface abrasions are common injuries of stored roots, and the storage temperatures used are typical of freshly harvested or rapidly cooled roots. Transpiration rate from the wounded surface and root weight loss were used to quantify wound healing. At 12°C, transpiration rate from the wounded surface declined within 14 days and wounded roots lost weight at a rate similar to unwounded controls. At 6°C, transpiration rate from the wounded surface did not decline in the 28 days after injury, and wounded roots lost 44% more weight than controls after 28 days storage. Melanin formation, lignification, and suberization occurred more rapidly at 12°C than at 6°C, and a continuous layer of lignified and suberized cells developed at 12°C, but not at 6°C. Examination of enzyme activities involved in melanin, lignin, and suberin formation indicated that differences in melanin formation at 6 and 12°C were related to differences in polyphenol oxidase activity, although no relationships between suberin or lignin formation and phenylalanine ammonia lyase or peroxidase activity were evident. Wound-induced respiration was initially greater at 12°C than at 6°C. However, with continued storage, respiration rate of wounded roots declined more rapidly at 12°C, and over 28 days, the increase in respiration due to injury was 52% greater in roots stored at 6°C than in roots stored at 12°C. The data indicate that storage at 6°C severely slowed and impaired wound-healing of surface-abraded sugarbeet roots relative to roots stored at 12°C and suggest that postharvest losses may be accelerated if freshly harvested roots are cooled too quickly.
