RESUMO

The induction of charge and spin in diluted magnetic semiconductor ZnO is explored for spintronic devices and its wide direct band gap (3.37 eV) and large exciton binding energy (60 meV) exhibit potential in UV photodetectors. We reported the ferromagnetic and optical properties of pure ZnO, Zn0.97Nd0.03O and Zn0.97Sm0.03O thin films. These thin films were synthesized by a metallo-organic decomposition method and annealed at 500 °C for 7 h. Rietveld refinement of the XRD data results in a wurtzite ZnO structure with Nd, Sm doping. The dopants and nanoparticle size are responsible for wurtzite structural deformation, inducing lattice strain effect, which may influence the band gap energy and high-TC ferromagnetism of ZnO. The average size of ZnO nanoparticles with Nd, Sm doping is 10 nm, confirmed with atomic force microscopy. The Raman spectra confirm the wurtzite structure of ZnO with crystalline quality and lattice defect formation with dopant Nd, Sm ions. A near-band-edge emission due to band gap energy is evaluated with photoluminescence spectra, which also involved multiple visible emissions due to oxygen vacancies. The oxygen vacancies-mediated magnetic interactions impart room temperature ferromagnetism in pure ZnO which is enhanced with Nd, Sm doping. The electron paramagnetic resonance spectra revealed the effects of defects and unpaired electrons responsible for observed room temperature ferromagnetism. The zero field cooling and field cooling magnetic measurements include antiferromagnetic interactions without any spin-glass formation. The observed ferromagnetism also correlates with first principle calculations reported for Nd, Sm-doped ZnO and suggests long-range ferromagnetic ordering attributed to defect carriers. The Nd, Sm doping into ZnO thin films significantly enhances absorption in the UV region and suggests its usability for UV detectors. Under UV irradiation (λ = 325 nm), the value of photocurrent in Nd, Sm:ZnO thin films is highly enhanced for possible use in UV sensors.

Buckwheat (Fagopyrum esculentum), one of the major traditional, underexploited crop having good nutritional value, can be grown in poor agroclimatic regions requires low inputs for its cultivation. Variability in biochemical parameters were observed in biotypes of buckwheat collected from different geographical regions. 1000 seed weight was varied from 9.48 to 15.22 g. Buckwheat biotypes contains high amount of protein (7.69-15.47%). Rutin, the most important ingredient of buckwheat was also varies (3.74-6.53%) in different biotypes. It was also found that many essential amino acids are also found in buckwheat. Variations among almost all estimated parameters were found to be highly significant.

BACKGROUND: Routine annual filarial surveys are conducted amongst various categories of military personnel and their families as per policies in vogue in the Armed Forces. The neglect and inattention faced by this disease needs to be addressed in terms of policy, provisioning and processes while dealing with filariasis in the Armed Forces. METHODS: Routine annual filarial survey was conducted in a garrison during the months of Nov and Dec in 2013 and 2014. Blood slides from 6305 and 10,162 persons were collected in 2013 and 2014 respectively. 546 (60.66%) civilian migratory labourers were also subjected to the filarial survey. RESULTS: Of the blood slides collected amongst service personnel, 41 were positive for mf in 2013 and 29 in 2014 (i.e. a slide positivity rate (SPR) of 0.65% and 0.28% respectively). Out of 546 blood slides of the migratory population, 10 were found mf positive (SPR 1.83%) and three males had lymphedema. CONCLUSION: It is recommended that routine annual filarial survey conducted in military garrisons should include all personnel belonging to known endemic states. Newer modalities of detection of infection may be considered to replace night blood surveys. An organization-specific surveillance programme on prevention and control of Lymphatic filariasis in the Armed Forces thus may need to be launched so that we can achieve elimination.

Water is the elixir of life. The requirement of water for very existence of life and preservation of health has driven man to devise methods for maintaining its purity and wholesomeness. The water can get contaminated, polluted and become a potential hazard to human health. Water in its purest form devoid of natural minerals can also be the other end of spectrum where health could be adversely affected. Limited availability of fresh water and increased requirements has led to an increased usage of personal, domestic and commercial methods of purification of water. Desalination of saline water where fresh water is in limited supply has led to development of the latest technology of reverse osmosis but is it going to be safe to use such demineralized water over a long duration needs to be debated and discussed.

Injury has been increasing exponentially, especially in children, and it has become a public health concern. The present study was conducted with the objective of determining the prevalence and profile of unintentional injury among children of 1-5-year age group in a rural community of India and also to find out the predictors. Primary caregiver was interviewed by using a structured interview schedule. Parent supervisory behaviour was assessed using Parent Supervision Attributes Profile Questionnaire (PSAPQ), and child injury risk-taking behaviour was assessed by using injury behaviour checklist (IBC). Children encountered any unintentional injury event during last three-month period were 261 (62.7%). PSAPQ score was significantly higher in those parents where children had no episode of injury compared to others. Among four components of the PSAPQ, protectiveness (p = 0.049) and risk tolerance (p = 0.001) score had significant positive association with the incidence of unintentional injury. Binary multivariable logistic regression technique had found that age of the child, gender, primary care giver, birth order of the baby, the number of siblings, social class and IBC score has significant association with history of unintentional injury. There is utmost need for the development of effective programmes and training strategies to prevent unintentional injury among under-five children in future.

Depleting reserves of fossil fuel and increasing effects of environmental pollution from petrochemicals demands eco-friendly alternative fuel sources. Jatropha curcas oil, an inedible vegetable oil, can be a substitute feedstock for traditional food crops in the production of environment-friendly and renewable fuel. Jatropha oil is looked up in terms of availability and cost and also has several applications and enormous economic benefits. The seed oils of various jatropha biotypes from hilly regions were screened out and evaluated for their physiochemical parameters, viz, seed index(520-600 g), oil content (15-42 %), biodiesel yield (71-98 %), moisture content (2.3-6.5 %), ash content (3.2-5.6 %), acid value (4.2-26), density (0.9172-0.9317 g/cm(3)), viscosity (5-37 mm(2)/s), saponification value (195.8-204.2 mg/g), iodine value (106.6-113.6 mg/g), flash point (162-235 °C), cetane value (46.70-50.06 °C), free fatty acid value (2.5-10.2 %), and refractive index (1.4600-1.4710). Fatty acid profiling of jatropha resembles as edible oilseeds. NAA with BAP was found to be superior for callus induction (up to 87 %), as well as for shoot regeneration (up to12 shoots). Root induction (90-100 %) was successfully obtained in MS medium with or without phytoregulators. Grown plantlets were successfully transferred from lab to field with a survival rate of 80 %.

An infectious bursal disease virus (IBDV), Hyd(C), from an outbreak was isolated and plaque-purified in BGM-70 cells. From the moderately virulent plaque-purified virus, two IBDVs with relatively high and low virulence were obtained by passaging the virus in specific pathogen free chickens and BGM-70 cells 10 and seven times, respectively. Comparison of amino acid sequences of the VP2 variable region of these viruses revealed that three amino acids at positions 279, 284 and 300 (Asp, Thr and Glu, respectively, in the plaque-purified virus) were changed. In in vitro- passaged virus, amino acid residues 279, 284 and 300 were Asn, Thr and Glu, whereas these were Asp, Ala and Gln in the in vivo -passaged virus. Change of residue 284 (Thr -->Ala) had a critical role in cell culture infectivity, whereas the change in residue 279 (Asp -->Asn) was associated with attenuation of the virus. No correlation could be observed between amino acid changes at position 300 and virulence or cell culture infectivity. Moreover, residue 330 (Arg) in heptapeptide motif SWSAR 330 GS was not found to be associated with the cell culture infectivity or virulence.

The polypeptides of three fowl adenovirus-4 (FAV-4) field isolates of hydropericardium syndrome from various geographical areas of the country and the standard FAV-1 (CELO virus) were subjected to sodium dodecyl sulfate polyacrylamide gel electrophoresis and analysed by protein immunoblotting with polyclonal antibodies to FAV-4 and FAV-1. Protein profile analysis of FAV-4 isolates revealed similarity of all the eight polypeptides with molecular weight ranging from 20 to 107 kDa but differed from CELO, particularly in their 24.2 kDa protein. Subsequent immunoblotting showed relatedness of at least five protein fractions of FAV-4 to CELO virus.

RESUMO

Antinuclear antibody (ANA) in chickens infected with reovirus was first detected at 3 weeks postinfection (PI). The antibody titer was greatest at 10 weeks PI (1:2560) and then declined. From 19 to 30 weeks PI, the birds were negative for ANA. The ANA was of both IgG and IgM types. The association between antinuclear factor and reoviral arthritis is discussed.

An isolate of egg drop syndrome-76 virus replicated best in primary chicken embryo liver cells and less well in duck embryo liver cells, duck embryo fibroblast cells and chicken embryo kidney cells. The cytopathic effect in chicken embryo liver cells was marked by the presence of round and refractile cells and detachment of cells from the glass surface. The intranuclear eosinophilic inclusion bodies were observed by 24 to 48 hours after infection. No virus multiplication was observed in primary quail embryo fibroblast cells, chicken embryo fibroblast cells or mammalian cells like Vero, BHK-21 and MDBK. Duck embryos supported the maximum growth of the virus, with allantoic fluid having the highest haemagglutinin titre, followed in order by chorioallantoic membrane, skin and internal organs. Chicken and quail embryos did not support the growth of the virus.

Cell mediated immunity was assayed in chickens on the basis of the dinitrochlorobenzene skin test, T lymphocyte numbers in peripheral blood and the graft-versus-host reaction. It was found that chickens that were to develop clinical Marek's disease following exposure to Marek's disease virus had significantly lower prechallenge levels of cell mediated immunity than those that were resistant to Marek's disease. The graft-versus-host reaction had a smaller coefficient of variation than the other two tests, and it was concluded that this test is likely to be the most reliable indicator of the outcome of infection with Marek's disease virus.

A sandwich ELISA was standardized to detect fowl adenovirus (FAV) group I antigen in various tissues, namely liver, spleen, bursa, thymus and kidneys of chicks experimentally infected with fowl adenovirus 4 (FAV-4) isolated from cases of inclusion body hepatitis-hydropericardium syndrome (IBH-HPS). The assay was found to be more sensitive and more specific in comparison to an agar gel immunodiffusion (AGID) test, as it could detect FAV antigen below the titer of 20,000 TCID50/ml and below 1.14 microg in 5% (w/v) suspensions of liver tissue. In 2-week-old experimentally infected chicks, the antigens were detectable by ELISA in liver from 3 to 15 days, in thymus from 3 to 7 days, and in kidneys, bursa and spleen from 3 to 10 days post infection (p.i.). Maximum antigen concentration in terms of ELISA absorbance values was detected in liver and kidneys, which could be used as tissues of choice for virus isolation or detection of viral antigens from IBH-HPS cases.

Light and electron microscopic evaluation of chick embryo fibroblast (CEF) cell culture inoculated with graded doses (0.25, 2.5 and 25 micrograms/ml medium) of aflatoxin B1 with and without infectious bursal disease virus (IBDV) was undertaken. The light microscopy revealed degeneration, detachment and necrosis of fibroblasts and multiple plaques formation in IBDV infected group without and with (0.25, 2.5 micrograms) aflatoxin B1. The cultures infected with virus, with or without 25 micrograms aflatoxin B1 showed complete detachment from glass surface. Electron microscopy of these cultures showed marked pyknotic or bizarre shaped nuclei, pronounced degenerative changes in the rough endoplasmic reticulum (RER), mitochondria and the presence of multiple vacuoles in the cytoplasm. The viruses were spherical, arrayed, complete, generally closer to nuclei and RER and indistinctly membrane bound. The viruses were either localised or scattered in the cytoplasm. Cultures containing 25 micrograms aflatoxin B1 without or infected with virus showed marked necrosis of cells. In latter group only a few viruses were seen either in infected cells or free in culture. Control cultures failed to show cytopathic changes as observed in the other three groups.

Two mouse monoclonal antibodies (MAbs), viz. 2B7 and 2 D10 raised against haemagglutinin-neuraminidase glycoprotein of Newcastle disease virus (NDV) were used to identify several other field isolates and vaccine strains of NDV. These MAbs reacted specifically with all the NDV strains/isolates in Dot-ELISA whereas, only MAb 2D10 reacted with all the NDV strains/isolates in agar gel precipitation test. These two tests employing the MAbs were standardised for rapid diagnosis and identification of NDV.

An Indian isolate of infectious bursal disease virus, i.e. IBDV-P/AD/81, was analysed for immunogenic activity of its structural polypeptides. Virus was purified from infected bursal homogenate by sucrose density gradient centrifugation. It showed five different structural polypeptides of 75.8, 45, 40.7, 33.1 and 27 kDa molecular weights in sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). Anti infectious bursal disease virus (IBDV) antibodies were tested using enzyme-linked immunosorbent assay (ELISA) and neutralization test (NT). Polypeptide 40.7 kDa (VP2) was known to have neutralizing epitopes. However, polyclonal anti VP2 failed to neutralize the virus. It was interpreted that VP2 had labile neutralizing epitopes which get altered confirmationally by SDS. Surprisingly, polyclonal anti 33.1 kDa (VP3) had mild neutralizing activity.

A rapid method of ultracentrifugation pelleting of avian adenovirus (AAV) from small volume of chloroform treated infected cell culture fluid or allantoic fluid was adapted for isolation of adenoviral DNA. The viral DNA extracted from semipurified viruses was found to be intact on agarose gel and pure enough (A260/280 = 1.85-1.92) for restriction enzyme analysis. Restriction endonuclease analysis of Indian strain of AAV serotype 1, AAV serotype 4 (group I AAVs) and egg drop syndrome-76 (EDS-76) virus genomes (group III AAV) with Hind III enzyme differentiated these viruses. The AAV serotype 1 and serotype 4 strain exhibited identical Hind III profile to European viral strains belonging to same serotypes however, the EDS-76 virus gave similar but not identical profile. The calculated genomic lengths for AAV serotype 1 and EDS-76 virus were approximately found to be 33.9 and 44.4 Kb, respectively.

In the present study characterisation has been done for six group I fowl adenoviruses (FAV) isolated from outbreaks of infectious hydropericardium (IHP) of chickens that occurred in different states/regions of India during the years 1994-98. These six viruses were identified as FAV serotype 4 by virus neutralisation and restriction endonuclease analyses. Antigenic analyses of the viruses revealed close relationship (R-values 0.93-0.96). Under the experimental conditions, we have been able to induce IHP using FAV serotype 4 isolate AD: 411 and were also able detect FAV antigens in myocardial tissues by immunofluorescence assay (a new observation), an indication that IHP causing FAV serotype 4 strain replicate in myocardial tissue. Restriction endonuclease analysis of the viral genomes (approximately 46 Kb), using Hind III, Sma I, Xba I, Bam HI, Pst I and Dra I produced identical genetic profiles. Pst I and Bam HI profiles for these six vitus isolates were identical to those published earlier for an IHP causing Pakistani FAV serotype 4 isolate KR31. The identical genetic profiles of viruses, chronology of the outbreaks of IHP in Pakistan during 1989 onward and later in Jammu and Kashmir, India (1994), suggest that FAV serotype 4 isolates involved in outbreaks of IHP in India had probably spread from Pakistan. In order to prevent further spread and economic losses due to IHP in India, based on the antigenic relatedness data in this paper, any one of the six studied FAV serotype 4 isolates can be used as a candidate for mass production of CEH culture based killed vaccine.
