RESUMO

A new polymer bearing aldehyde groups was designed and synthesized by grafting 4-pyridinecarboxaldehyde onto poly(epichlorohydrin). Antibodies can be directly immobilized on the surface of the polymer film through the covalent bonding of aldehyde groups of the film with amino groups of antibodies. In this study, human immunoglobulin G (IgG) was used as a model analyte for the fabrication of an electrochemical impedance immunosensor. Using the proposed immunosensor, IgG in the range from 0.1 to 80 ng ml(-1) was detected with a detection limit of 0.07 ng ml(-1) (signal/noise [S/N]=3). In addition, the electrochemical impedance immunosensor displays good stability and reproducibility.

RESUMO

Highly sensitive and label free detection of prostate specific antigen (PSA) still remains a challenge in prostate cancer diagnosis. In this paper, we propose a sensitive electrochemical immunosensor based on electrocatalytic platinum nanoparticles conjugated to a recombinant scFv antibody. Gold disc electrodes functionalised with a l-Cysteine (Cys) self-assembled monolayer (SAM) were used to covalently bind PSA specific monoclonal antibody (anti-PSA) using N-ethyl-N'-(3-dimethylaminopropyl) carbodiimide and N-hydroxysuccinimide (EDC/NHS) chemistry. Immunosensing was completed using sandwich-type immunoreaction of the PSA-antigen (1-30 ng/mL) between anti-PSA immobilized on the l-Cys modified electrode using label free electrochemical impedance (EIS) technique. Furthermore, highly specific in-house generated scFv fragments as receptor proteins were utilised for one step site-directed immobilisation on the surface of platinum nanoparticles (PtNPs). To improve the sensitivity of the immunoassay, these scFV labelled electrocatalytic PtNPs were then used for covalent hybridisation to the PSA modified electrode and then applied in a hybridisation assay to determine the concentration of the PSA by measuring the faradaic current associated with reduction of peroxide in solution. Semi-log plots of the PSA concentration vs. faradaic current are linear from 1 to 30 ng/mL and pM concentrations can be detected without the need for molecular, e.g., PCR or NASBA, amplification.

Bovine interferon gamma (BoIFN-γ) released by T cells plays very important roles in early diagnosis of Mycobacterium tuberculosis (MTB) infections and control of bovine tuberculosis. In this work, a label-free electrochemical impedance immunosensor is for the first time developed for highly sensitive determination of BoIFN-γ. Cylinder-shaped TiO2 nanorods are synthesized by a facile hydrothermal method, and show high surface area and good hydrophicility. The immunosensor is fabricated by the immobilization of BoIFN-γ monoclonal antibody on the TiO2 nanorods modified glassy carbon electrode. The prepared TiO2 and immunosensor are characterized using transmission electron microscopy, scanning electron microscopy, X-ray diffraction, contact angle measurement, cyclic voltammetry, and electrochemical impedance spectra. The BoIFN-γ concentration is measured through the relative increase of impedance values in corresponding specific binding of BoIFN-γ antigen and BoIFN-γ antibody. The relative increased impedance values are proportional to the logarithmic value of BoIFN-γ concentrations in a wide range of 0.0001 to 0.1 ng/mL with a low detection limit of 0.1 pg/mL. The developed BoIFN-γ immunosensor shows a 249-fold decrease in detection limit in comparison with current enzyme-linked immunosorbent assay. This study provides a new, simple, and highly sensitive approach for very potential application in early diagnosis of MTB infections and control of bovine tuberculosis.

A sensitive and stable label-free electrochemical impedance immunosensor for the detection of Salmonella typhimurium was developed by immobilising anti-Salmonella antibodies onto the gold nanoparticles and poly(amidoamine)-multiwalled carbon nanotubes-chitosan nanocomposite film modified glassy carbon electrode (AuNPs/PAMAM-MWCNT-Chi/GCE). Electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV) were used to verify the stepwise assembly of the immunosensor. Co-addition of MWCNT, PAMAM and AuNPs greatly enhanced the sensitivity of the immunosensor. The immobilisation of antibodies and the binding of Salmonella cells to the modified electrode increased the electron-transfer resistance (Ret), which was directly measured with EIS using [Fe(CN)6](3-/4-) as a redox probe. A linear relationship of Ret and Salmonella concentration was obtained in the Salmonella concentration range of 1.0×10(3) to 1.0×10(7) CFU mL(-1) with a detection limit of 5.0×10(2) CFU mL(-1). Additionally, the proposed method was successfully applied to determine S. typhimurium content in milk samples with satisfactory results.
