Purpose: Low-intensity pulsed ultrasound (LIPUS) has been used to stimulate the healing of the fresh fracture, delayed union, and non-union in both animal and clinical studies. Besides, biphasic calcium phosphate ceramic (BCP) is a promising biomaterial for bone repair as it shows favorable biocompatibility, osteoinduction, and osteoconduction. However, scarcity is known about the combined effect of LIPUS and BCP on bone formation. Methods: The combined effect of LIPUS and BCP was studied in a beagle model. Twelve dogs were used. BCP granules without any additions were implanted into bilateral erector spinae muscles. One side is the BCP group, while the counterlateral side is LIPUS + BCP group. Histological and histomorphometric analyses, and quantitative real-time polymerase chain reaction were evaluated. Results: Compared with BCP alone, the LIPUS + BCP showed no advantages in early bone formation. Furthermore, the Notch signaling pathway-related mRNA has no significant difference between the two groups. Conclusions: The preliminary results showed that the BCP, which has intrinsic osteoinduction nature, was an effective and promising material. However, LIPUS has no enhanced effect in BCP induced ectopic bone formation. Furthermore, LIPUS has no effect on the Notch signaling pathway. Whether costly LIPUS could be used in combination with BCP should be a rethink.

Abstract Autosomal recessive primary microcephaly (MCPH) is a neurodevelopmental disorder characterized by a congenitally reduced head circumference (-3 to -5 SD) and non-progressive intellectual disability. The objective of the study was to evaluate pathogenic mutations in the ASPM gene to understand etiology and molecular mechanism of primary microcephaly. Blood samples were collected from various families across different remote areas of Pakistan from February 2017 to May 2019 who were identified to be affected with primary microcephaly. DNA extraction was performed using the salting-out method; the quality and quantity of DNA were evaluated using spectrophotometry and 1% agarose gel electrophoresis, respectively in University of the Punjab. Mutation analysis was performed by whole exome sequencing from the Cologne Center for Genomics, University of Cologne. Sanger sequencing was done in University of the Punjab to confirm the pathogenic nature of mutation. A novel 4-bp deletion mutation c.3877_3880delGAGA was detected in exon 17 of the ASPM gene in two primary microcephaly affected families (A and B), which resulted in a frame shift mutation in the gene followed by truncated protein synthesis (p.Glu1293Lysfs*10), as well as the loss of the calmodulin-binding IQ domain and the Armadillo-like domain in the ASPM protein. Using the in-silico tools Mutation Taster, PROVEAN, and PolyPhen, the pathogenic effect of this novel mutation was tested; it was predicted to be "disease causing," with high pathogenicity scores. One previously reported mutation in exon 24 (c.9730C>T) of the ASPM gene resulting in protein truncation (p.Arg3244*) was also observed in family C. Mutations in the ASPM gene are the most common cause of MCPH in most cases. Therefore, enrolling additional affected families from remote areas of Pakistan would help in identifying or mapping novel mutations in the ASPM gene of primary microcephaly.

Background: Captive tigers can live a long life, around 26 years. Among the diseases described some of non-infectious origin are quite common, such as chronic kidney disease, spondylosis, and biliary cysts or tumors. On the other hand, pyometra has been frequently reported in lions, who have a higher risk of developing the disease than tigers and leopards. Pyometra is a disease with few descriptions in tigers and it may be related to the physiological features of the species. The animal is listed as Endangered on the International Union for the Conservation of Nature Red List of Threatened. The present report aims to describe the diagnosis and treatment of pyometra in a captive tigress. Case: A 7-year-old entire female tiger (Panthera tigris) weighing 140 kg was presented with a 3-day history of anorexia and prostration. For clinical examinations, collection of laboratory and imaging tests, the patient initially underwent dissociative anesthesia to allow catheterization of the cephalic vein and intravenous general anesthesia for orotracheal intubation followed by anesthetic maintenance in isoflurane. On general physical examination, the animal had normal colored mucosa, vital parameters within normal limits, and a body condition score of 6 on a scale of 9. There was no presence of vulvar secretion. The blood count and the biochemical exams showed values within the normal range for the species. The chest X-ray in the right and left views did not demonstrate pulmonary abnormalities. Ultrasonographic examination of the abdomen showed distension of the uterine body and horns, which have intraluminal hyperechoic fluid content without flocculation. Based on the imaging exam, the diagnosis was suggestive of pyometra. Exploratory celiotomy was performed via ventral midline, confirming the condition, which was treated by ovariohysterectomy. The surgical technique was performed as described for therapeutic ovariohysterectomy in dogs and cats. Culture of uterine content identified Escherichia coli. The histological analysis identified diffuse endometritis associated with follicular cysts. The tiger had complete recovery without any complications. The patient was releasing 13 days after the surgical procedure and in the last contact four months after the surgery, it was in perfect health conditions. Discussion: Pyometra in large exotic felids has been occasionally reported, mainly in animals more than 10 years of age. Although the tigress in the report is estimated to be seven years old. The patient in question started with anorexia and prostration and as there was already a history of cystic endometrial hyperplasia, a possible pyometra was suspected, despite being uncommon in the species. There was not vaginal discharge. The definitive diagnosis was by means of ultrasound examination and ovariohysterectomy was performed. Abdominal surgery for these large felids is complex, due to the intra-abdominal volume the flank approach or by laparoscopic is suggested, however in this case a ventral midline incision was performed without intercurrences and complications in the post-operative period. The surgical technique like that used in small animals was effective for the treatment of pyometra in the tigress with the use of ovariohysterectomy. Culture of uterine content identified Escherichia coli, which has been the most commonly isolated pathogen in pyometra of large felids. It was concluded that, as in bitches with pyometra, early diagnosis and surgical treatment is ideal for the patient's recovery.

Biological Resource Banks (BRB) or Genetic Resource Banks (GRB) are critical tools for the conservation of animal biodiversity. According to the International Union for Conservation of Nature, more than 38,500 species are threatened with extinction, out of a total of 138,300 surveyed species. These banks are repositories of biological samples and data recovered and preserved for the long term by zoos, universities, research centers and other conservation organizations. In recent years, BRB have increasingly included ovarian and testicular tissues as additional options to rescue and propagate wild species, especially those at risk of extinction. After in vitro culture or grafting, gonadal tissues are potential sources of matured gametes that can be used for Assisted Reproduction Technologies while informing about gametogenesis or mechanisms involved in infertility. It therefore is crucial to properly recover, cryopreserve, and culture these tissues using species-specific protocols. Developing BRBs is currently one of the strategies to preserve species from the Caatinga biome - an exclusively Brazilian biome with a rich wild fauna that suffers from anthropogenic activities. Among wild species from this biome, studies have been primarily conducted in collared peccaries, agoutis, cavies, and armadillos to preserve their ovarian and testicular tissues. Additionally, domestic species such as the domestic cat and donkeys have been proposed as models for wild species that are phylogenetically close. This review addresses the main technical aspects involved in obtaining BRB derived from gonadal tissues in some wild species of the Caatinga biome. It reports recent advances and perspectives to use these biological materials for wildlife conservation.(AU)

Non-native predators are known to cause ecological impacts through heightened consumption of resources and decimation of native species. One such species is Micropterus salmoides. Often introduced for sport fishing worldwide; it has been listed by International Union for Conservation of Nature and Natural Resources as one of the 100 of the world's worst invasive species. Whilst impacts conferred towards native prey are well known, its relationship with other non-native species has received much less attention. Therefore, the aim of this study was to experimentally investigate the feeding preference of M. salmoides towards native prey (Geophagus iporangensis) compared to non-native prey (Oreochromis niloticus and Coptodon rendalli) using comparative consumption and the Manly-Chesson selectivity index. We found a higher consumption by M. salmoides towards the non-native prey and a tendency for this non-native preference to increase when prey availability increased. Our results are in line with the hypothesis that the invasion of the non-native predator can be facilitated by the high abundance and reproductive rate of the non-native prey. This is relevant considering the interaction of multiple invaders in an ecosystem, in particular environments modified by humans. Interspecific relationships are complex and their understanding is necessary for environmental management decision-making.(AU)

The concern about pain management in exotic mammals is increasing. However, only a few studies describe locoregional anesthesia techniques in rabbits. Thus, this report aims to describe the lumbar paravertebral block, guided by peripheral nerve stimulation, in a rabbit undergoing mid-femoral amputation. We received a one-year-old rabbit weighing 1.27kg with a history of non-union of the right femur and osteomyelitis. The animal came to us for high pelvic limb amputation. After premedication with ketamine, midazolam, and morphine, the patient was maintained under general anesthesia in a non-rebreathing circuit under spontaneous ventilation with isoflurane vaporized in 0.6 oxygen. Using a peripheral nerve stimulator, we performed the lumbar paravertebral block, positioning the needle between the L5 and L6 vertebrae and injecting 0.2mL/kg bupivacaine. The patient remained stable throughout the procedure, without the need for analgesic rescue with opioids during surgery and postoperatively. The discharge occurred after 24 hours. The lumbar paravertebral block proved efficient for trans and postoperative analgesia for high pelvic limbic amputation in rabbits.

Background: Pelvic fractures are commonly ecountered in cats. The ilium is included in approximately 50% of pelvic fractures in cats. Although conservative management of the pelvic fracture is known to have good prognosis, surgical fixation is recommended for the pelvic fracture with ilial fractures. 3D printing is an accurate digital modeling method by converting two-dimensional (2D) images into 3D model. 3D printing in veterinary medicine has been described in the various orthopedic diseases such angular limb deformities or fractures. The use of 3D model for presurgical planning is helpful to improve the results of complex surgical interventions. The objective of this study is to identify the usefulness of fabricating 3D models based on allogenic 2D data with aid of 3D printer program. Case: A 4-year-old spayed female domestic shorthair cat presented with hindlimb lameness. Radiography revealed left ilial body fracture with multiple fractures including left tuber ischium, pubic symphysis, and bilateral cranial ramuses of pubis. Digital imaging and communication in medicine (DICOM) files of the complete ilium were extracted from a computed tomography (CT) scan of a healthy and castrated male cat of same breed. Volume rendering images were prepared by Slicer program based on DICOM files. After acquired 3D data were converted to STL (Standard Triangulation Language) file for the 3D printing machine, non-fracture involved area was removed except the left ilium using the Meshmixer® software and then data was stored as STL files again. The length of 3D model was adjusted to actual size using 3D printer firmware based on actual size of the ilial length. Subsequently, 3D modelling was performed using 3D printer. The plate was precontoured to conform to the lateral surface of the 3D model. Internal fixation using precontoured plate resulted in accurate bone alignment. The alignment of the ilium remained appropriate throughtout the follow-up period. Complete union was identified 6 months after surgery. Discussion: Although the most common method for the ilial fractures is the application of bone plates to the lateral side of the ilium, bone plate fixation is more challenging in cats owing to the small bone size and small surface area of the fracture sites compared to dogs. Therefore, it is important to make accurate presurgical planning for successful result of a surgery. 3D printing based on actual patient have several limitations including time for complete printing process and additional anesthetic episode to acquire CT images. However, in simple cases, a presurgical CT scan may not be necessary if the plate can be precisely precontured based on the data from the other patient. In this way, the additional financial burden for CT examination also might be reduced. On the other hand, technical difficulty of utilizing hardware and software, and financial availability were another drawback. However, in this current study, most of limitations regarding conventional 3D printing based on actual patient were overcome by the application of the fused deposition modeling (FDM) technology with allogenic data. In the current study, total time for fabrication of 3D model based on FDM technology, was less than 3 h, which are much shorter than the time for conventional 3D printing. The difference of the ilial length between the actual patient and the 3D model was adjusted through the firmware program. Thus, differences of the conformation or size between the 3D model and the ilium of the patient was negligible during the surgery, although 2D images were not acquired from actual patient. Intraoperative contouring of the plate was not required. In conclusion, through presurgical plate contouring by the 3D bone template based on non-patient specific data, successful treatment of the ilial fracture was possible.

The current global situation requires urgent decision-making to reverse processes of mass extinction of thousands of species. As a way of showing the importance of joint actions in this process, we aim to present the concept of One Conservation as a new proposal for the integration of sustainability, in situ and ex situ conservation for the restoration of ecosystems. According to the United Nations, we are beginning the decade of ecosystem restoration and in association with the International Union for Conservation of Nature guidelines, we can join efforts in the conservation of the planet. The survival of many species of wild animals depends on the management of populations currently maintained in ex situ conditions (under human care). To facilitate the exchange of genetic material between in situ and ex situ populations, reproductive biotechniques have become a great tool, making it possible to restore species in their natural environments. For effective conservation to occur, there must be an integrated view of the problem as a whole, and action for solutions must take place jointly by different spheres of society. Even more, conservation must be carried out by the public sector, the private sector, the third sector, and not less importantly, the agricultural sector. Therefore, One Conservation is defined as an interconnection between ex situ and in situ conservation plans, anthropic actions in the environment, and research in different areas that encompass conservation.(AU)

Background: The spectacled bear (Tremarctos ornatus) is the only bear species inhabiting South America and is classified as vulnerable according to the International Union for Conservation of Nature (IUCN) Red List of Threatened Species. Among the few publications on the use of general anesthesia and advanced monitoring of ursids in veterinary hospital settings, little is described regarding chemical restraint, general anesthesia and monitoring of spectacled bears. This case series describes the use of a dexmedetomidine-tiletamine-zolazepam chemical restraint combination and its effects on cardiorespiratory variables and arterial blood gases observed in two spectacled bears undergoing isoflurane anesthesia for imaging and/or surgical procedures. Cases: Two female, one adult and one senile, all-term captive spectacled bears were referred to the Veterinary Teaching Hospital at the Universidade Estadual Paulista - Unesp, Botucatu campus, both with a presumable history of recent trauma. After immobilization with an intramuscular (IM) administration of tiletamine-zolazepam (3.8 - 4.3 mg/kg) and dexmedetomidine (6.4 - 7.6 µg/kg), induction of anesthesia was achieved by means of intravenous (IV) propofol (1 - 2 mg/kg). After orotracheal intubation animals underwent isoflurane anesthesia under mechanical ventilation through the remainder of the procedures. Initial settings of inspiratory flow rate were adjusted to obtain peak airway pressure (Ppeak ) of 10 cmH2 O and tidal volumes (Vt) of 10 mL/kg, as well as respiratory rates (ƒR) and inspiration-to-expiration (I:E) ratio of 10 breaths/min and 1:2, respectively, and were then adjusted throughout anesthesia to maintain normocapnia (end-tidal carbon dioxide concentrations between 35 and 45 mmHg). One of the individuals was chemically restrained (6.4 mg/kg of tiletaminezolazepam and 7.7 µg/kg of dexmedetomidine) on a second anesthetic event for imaging...

Anlurfa is the province with the third-largest agricultural area in Turkey. However, it has only 0.62% of the total number of cooperatives in the country. This study aimed to determine the perceptionsof farmers in anlurfa regarding agricultural cooperatives and the factors affecting them. The main research material was obtained through face-to-face surveys involving farmers in anlurfa selected by a simple random sampling method. The sampling volume was determined with a 95% confidence limit and a 5% error margin. Multiple regression analysis was used in Stata software. According to the results of the research, 39.7% of the participants were cooperative members and more than half of them were not active members. Of the participants, 30.3% had a positive perception of cooperatives, 36.82% were undecided, and 32.88% had negative opinions. Age, marital status, non-agricultural income, experience, social security, farmer registration system, cooperative membership, and land variables positively affected farmers perceptions. However, the variables of education, income, and union membership had negative effects on farmers perceptions. No statistically significant relationship was reported between a farmers perception and any of the following variables: the number of people that make up the farmers household, the number of individuals engaged in agriculture and non-agricultural work in the family, and the status of the farmers lease. Conceptual, local, cultural, educational, and structural problems related to cooperatives exist in anlurfa. These issues need to be given more attention by the public. This research is the first study on this subject to be conducted in anlurfa.(AU)

This review aimed to describe aspects of bird fractures, some immobilization techniques and the complications of fractures. Although the principles of fracture repair in birds are similar to those of small animals, there are differences that should be considered during the application of the immobilization method. Fractures in birds can be immobilized by external coaptation (splints and bandages), internal fixation (pins, cerclage wires, bone plates) and external fixation, among others, each one with its own advantages and disadvantages. The severity of the fracture interferes in the rate and course of bone healing; however, clinical stability may precede radiographic evidence, which is visible in 3-6 weeks. Avian fractures tend to be open and comminuted, because of the scarce cover of soft tissues. Open fractures have high risk of developing osteomyelitis and, consequently, may develop delayed union or non-union.

Open fractures have high risk of developing osteomyelitis and, consequently, may develop delayed union or non-union.(AU)

This study aimed to characterize a preclinical model of critical size defect (CSD) in the radius of rabbits.Twenty adults (> 7 months), female, New Zealand Rabbits, weighing between 3,5 to 5 Kg were used. They underwent a 1.5 cm long ostectomy of the diaphysis of the radius, starting 2 cm from the carpus joint. Radiographic analyses were performed at 15, 30, 60, and 90 days postoperatively, in order to evaluated bone callus formation, periosteal reaction, and bone bridge formation. The methodology allowed to precisely create bone defects of 1.5cm. Over time, no bone deposition was found at 15 days, but mild bone callus formation was observed in three animals after 60 days postoperative. At 90 days postoperatively only one rabbit presented bone consolidation radiographically, the others presented non-union. The critical bone defect proposed in this study was satisfactory, feasible with very low risk of complications.

In this case, a Brazilian row dog was submitted to grafting with pedicle flap of greater omentum, after resistant osteomyelitis present in open fracture osteosynthesis of open tibia grade II. For 16 days the communication of the flap was maintained, but at the risk of peritonitis, the pedicle was sectioned. In a sequence of surgical interventions, after 89 days, there was bone healing and remission of osteomyelitis, even in the presence of multi-resistant bacteria. In this report, the omentum was effective as adjuvant therapy in the treatment of osteomyelitis and guaranteed the return of limb function.(AU)

Background: Phoenicopterus chilensis is a South American wild bird classified as a near threatened specie in the List ofEndangered Species of the International Union for Conservation of Nature. With the increase of the contact between human population and wild animals, this species’ habitat is becoming increasingly vulnerable, with a declining population.Due to the importance of its conservation, the knowledge of abnormalities that affect this species becomes essential. Thisreport aims to describe the first diagnosed case of unilateral renal agenesis in chilean-flamingo.Case: A captive male chilean-flamingo (Phoenicopterus chilensis) was found dead in its enclosure and then was referredat necropsy. The cause of death was the presence of tracheal fungal granuloma, caused by Aspergillus sp. Macroscopically, during the examination of the body cavity, the absence of the right kidney was noted, also evidencing the absence of thecaudal renal vein. Emerging from the caudal division of the kidney was noted a blind-end renal vessel. For the histopathological examination, the remaining kidney was fixed in 10% formalin, processed by paraffin embedding technique andstained with Hematoxylin and Eosin (HE). When analyzing the left kidney parenchyma, preserved histological architecturewas noted, without any changes in the tissue structures of the organ. Thus, no hypertrophy by compensatory mechanismsof the remaining kidney was observed.Discussion: Unilateral renal agenesis is a very rare congenital defect in dogs, cats and also in birds. It occurs in the complete absence of one of the kidneys, a situation with which the animal can live satisfactorily if there is a normal kidney toassume the functions. In association, ipsilateral ureteral agenesis and compensatory...

The present study was carried out to evaluate the prevalence and hematological effects of Toxoplasma gondii in sheep and goat in district Dera Ghazi Khan. Blood samples (n=204) were collected comprise goats (n=101) and sheep (n=103) alongwith age, gender and breeds of animals. Samples were collected randomly from 25 flocks of 7 different union council Viz. Vehova, Tibbi Qaisrani, Lakhani, Kohar, Tuman Qaisrani, Nutkani and Kot Qaisrani of Tehsil Taunsa Sharif at least 4 animals from each flock. All ruminants divide into three groups based on age, breed and gender. The prevalence was detected through two different kits Viz. LAT and ELISA kit. The overall prevalence suspected in goats through LAT and ELISA kit was (35.64%), (32.67%) and in sheep was (25.24%), (23.30%) respectively. The Toxoplasma gondii had a significant effect on goats in age groups and non-significant all other groups of goats and sheep. Toxoplasma gondii had a significant effect on all hematological parameters like Hemoglobin, total leukocyte cells, granulocytes, lymphocytes, platelets, and red blood cells, except monocytes. In conclusion of the current study, toxoplasmosis is prevalent among ruminants, reveals the possibility of transmission to humans on the use of host animals as protein source.(AU)

The collage on pieces of wood using adhesives has been performed since the Pharaoh era. Over the years, technologies have been developed especially for adhesives and different species of wood, adapting to the desired diverse uses. In this context we can also infer the methods of analysis and its efficiency. The aim of this study was to evaluate the resistance in the glue-line by using adhesives in Paulownia tomentosa wood (Japanese kiri). The bonded joints were prepared with resorcinol formaldehyde and melamine urea formaldehyde adhesives. The wood-adhesive interface was evaluated by means of the thickness of the main and secondary glue-lines using photomicrographs, shear strength, and percentage of wood imperfections. The adhesives presented non-uniform behavior in the glue-line. It was possible to observe that the adhesives penetrated in the wood anatomical structures (elements of vessels and rays). The resorcinol formaldehyde adhesive had higher penetration values. The kiri wood imperfection percentage was greater than 92%, independent of the adhesive. The high efficiency observed in bonding with both adhesives allows the use of wood of Paulownia tomentosa in Brazil for furniture and civil construction.
