Wooden breast myopathy (WB) strongly affects the poultry industry mainly in terms of consumers rejection and economical loses, due to morphological changes in broiler muscle tissue and consequently low meat quality. The aim of this study is to evaluate the histomorphometry of muscle fibers of breast fillets of broilers with severity levels of WB myopathy. The histological evaluation considered 30 samples of the pectoralis major muscle and the level of WB myopathy (ten normal fillets, ten moderate fillets, and ten severe fillets). Fillets with a severe level of WB present low average fiber number, high average fiber diameter, low percentage of fibers with diameter of less than 20 µm, low percentage of fibers with diameter between 20 and 40 µm, and high percentage of fibers with diameter between 40 and 70 µm. Fiber cross-sectional area is greater in fillets affected by moderate and severe WB. Thus, fillets with a severe level of WB damage the muscle fiber structure and contribute more severely to the degenerative processes of breast meat.(AU)

Purpose: To evaluate the quality of breast reconstruction service at a university hospital, as assessed by the patients. Methods: This cross-sectional study enrolled adult women who underwent immediate or delayed breast reconstruction by any technique performed at a university hospital between 1 and 24 months before the assessment. The Brazilian version of the Health Service Quality Scale (HSQS) was self-applied to the participants. The HSQS produces percentage scores, which are expressed in values ranging from 0 to 10 for each domain of the scale, and into an overall percentage quality score. The management team was asked to establish a minimum satisfactory score for the breast reconstruction service. Results: Ninety patients were included. The management team considered 8.00 the minimum satisfactory score for the service. The overall percentage score was 93.3%. Only one domain, 'Support,' had an average score lower than that considered satisfactory (7.22 ± 3.0); while the others had higher scores. The domain that scored highest was 'Qualification' (9.94 ± 0.3), followed by 'Result' (9.86 ± 0.4). There was a positive correlation between 'type of oncologic surgery' and 'intentions of loyalty to the service' (ρ = 0.272; p = 0.009) and a negative correlation between 'education' and 'quality of the environment' (ρ = 0.218; p = 0.039). The higher the patient's level of education, the higher the score attributed to 'relationship' (ρ = 0.261; p = 0.013) and the lower the score of 'aesthetics and functionality' (ρ = 0.237; p = 0.024). Conclusion: The quality of the breast reconstruction service was considered satisfactory, but there is a demand for structural improvements, better interpersonal relationships, and a stronger support network for patients.

Purpose: To evaluate fibrosis formation and number of macrophages in capsules formed around textured implants without and with mesh coverage. Methods: Fibrosis was analyzed through transforming growth factor-beta 1 (TGF-ß1) immunomarker expression and the number of macrophages through CD68 percentage of cells in magnified field. Sixty female Wistar rats were distributed into two groups of 30 rats (unmeshed and meshed). Each group was then subdivided into two subgroups for postoperative evaluation after 30 and 90 days. The p value was adjusted by Bonferroni lower than 0.012. Results: No difference was observed in fibrosis between meshed and unmeshed groups (30 days p = 0.436; 90 days p = 0.079) and from 30 to 90 days in the unmeshed group (p = 0.426). The meshed group showed higher fibrosis on the 90th day (p = 0.001). The number of macrophages was similar between groups without and with mesh coverage (30 days p = 0.218; 90 days p = 0.044), and similar between subgroups 30 and 90 days (unmeshed p = 0.085; meshed p = 0.059). Conclusions: In the meshed group, fibrosis formation was higher at 90 days and the mesh-covered implants produced capsules similar to microtextured ones when analyzing macrophages. Due to these characteristics, mesh coating did not seem to significantly affect the local fibrosis formation.

In order to evaluate the effect of diets with Ganoderma lucidum mushroom powder and zinc-bacitracin on growth performance, carcass traits, lymphoid organ weights, and intestinal characteristics in broilers, a total of 600 one-day-old unsexed broilers from Cobb 500 MV × Cobb 500 FF genotype was analyzed for 28 days, following a completely randomized design with three dietary treatments, five replicates and 40 birds per replicate. The dietary treatments consisted of a basal diet (BD) without additives (T0) and the dietary inclusion of 2.5 g/kg of Ganoderma lucidum (T1) and 350 mg/kg of zinc bacitracin antibiotic (T2). The experimental groups did not change (p>0.05) the performance of the broilers. However, G. lucidum powder increased (p≤0.05) the carcass and breast yields (p≤0.05) and decreased the abdominal fat and liver yields (p≤0.05), although with no notable differences with the antibiotic group for the latter organ (liver) (p>0.05). Both additives (G. lucidum and antibiotic) increased breast meat moisture, protein, and redness; however, these treatments reduced L* (lightness), and the zinc-bacitracin reduced breast yellowness (p≤0.05). Likewise, this medicinal mushroom (G. lucidum) increased the relative weight of bursa of Fabricius and the morphometry of the small intestine (p≤0.05), although with no changes for other immune and digestive organs or for the content of cecal lactic acid bacteria (p>0.05). The dietary inclusion with 2.5 g/kg of Ganoderma lucidum powder is recommended to improve breast yield, protein, and colorimetry without affecting performance and cecal traits of fast-growing broilers.(AU)

The effect of including partially dehulled sunflower seeds in the diet of grazing chickens in meat quality was evaluated. Two hundred and forty, one-day-old, Ross 308 chickens were used, randomly distributed into four treatments with six replicates of 10 birds each. Seventy-two birds were slaughtered at seven weeks of age, 18 per treatment, extracting the Pectoralis major muscle to analyze meat quality. The treatments were completely randomized in a 2x2 factorial arrangement, where type of rearing (confinement or grazing) and diet (base diet or base diet substituting 10% soybean meal for partially dehulled sunflower seeds) were the independent variables. The grazing chickens spent 8 hours a day in a white clover (Trifolium repens) paddock. Including sunflower seeds improved (p<0.05) the color and crude protein (CP) in breast meat (p<0.05), while grazing increased (p<0.05) CP, dry mater (DM), and shear force (SF), and the percentage of breast fat decreased (p<0.05) by up to 50%. Additionally, grazing increased (p<0.05) saturated fatty acids (SFA) and reduced (p<0.05) lipid oxidation by up to 50%. Partially replacing soybean meal with partially dehulled sunflower seeds in the diet of grazing chickens with white clover improves the physicochemical characteristics and nutritional quality of the breast meat; it also decreases lipid oxidation, extending the shelf life of the meat.(AU)

Hematological and biochemical alterations in animals with neoplasms may result from the direct effects of tumor growth or paraneoplastic syndromes. The objective of this study was to evaluate these hematological and biochemical alterations in female dogs with mammary tumors and with inflammatory carcinoma (IC). Blood samples were collected from 43 female dogs divided into three groups according to clinical staging: Group 1 (G1) - animals in initial stage (T1,2,3N0M0, N=17), Group 2 (G2) - animals in advanced stage (T1,2,3N1M0,1, N=15) and Group 3 (G3) - animals presenting IC (N=11). Hematological and biochemical parameters obtained were related to patients' clinical staging. Among alterations, the most common were anemia, neutrophilic leukocytosis, monocytosis, increased ALT, AST, and hypoalbuminemia, mainly in dogs in advanced clinical staging and with inflammatory carcinoma.
Aterações hematológicas e bioquímicas em animais portadores de neoplasias podem resultar dos efeitos diretos do crescimento tumoral ou de síndromes paraneoplásicas. O objetivo deste estudo foi avaliar as alterações hematológicas e bioquímicas em cadelas com neoplasias mamárias e carcinoma inflamatório (IC). Foram coletadas amostras de sangue de 43 cadelas divididas em três grupos de acordo com o estadiamento clínico: grupo 1 (G1) - cadelas em estádio inicial (T1,2,3N0M0, N=17), grupo 2 (G2) - cadelas em estádio avançado (T1,2,3N1M0,1, N=15) e grupo 3 (G3) - cadelas que apresentaram IC (N=11). Os parâmetros hematológicos e bioquímicos obtidos foram relacionados com o estadiamento clínico das pacientes. Entre as alterações, as mais comuns foram anemia, leucocitose neutrofílica, monocitose, aumento de ALT, AST e hipoalbuminemia, principalmente em cadelas em estadiamento clínico avançado e com carcinoma inflamatório.

In breast cancer treatment, chemotherapy resistance is a major problem where many receptive tumors rebound and develop resistance. When provided in combination, cancer drugs are most successful, thus reducing the risk of developing resistant cancer cells. However, the evaluation of combination therapies has increased rapidly in recent years. Consequently, by repurposing old treatments, the discovery of additional medicines that may interact synergistically with chemotherapy is considered a current medical aim through discovering a new cancer medication or therapeutic strategy. The purpose of this research is to increase the anti-cancer activity of carboplatin (CP) by increasing the apoptotic effect of breast cancer cells (MCF-7) during in vitro experiments in combination with oxytetracycline. Our results showed a high synergistic effect between oxytetracycline and carboplatin, MCF-7 representative cell treated with carboplatin with/without different concentrations of oxytetracycline (5% and 10% of IC50). Oxytetracycline, which potentiated the action of carboplatin and/or had notable activity was reported as a single agent. This research demonstrated the synergistic relationship between oxytetracycline and carboplatin in viability assays. Surprisingly, our findings suggest that inhibiting treatment strategies can extend carboplatin's therapeutic window, potentially allowing for cancer therapy.(AU)
No tratamento do câncer de pulmão a resistência à quimioterapia é o maior problema no qual muitos tumores receptivos apresentam um rebote e desenvolvem a resistência. Quando oferecidas em combinações, as drogas anticancerígenas apresentam maior taxa de sucesso, reduzindo assim o risco de desenvolvimento de células cancerígenas resistentes. Contudo, a avaliação das terapias de combinação tem crescido muito rapidamente. Consequentemente, por reaproveitamento de tratamentos antigos, a descoberta de novos medicamentos adicionais que podem interagir sinergicamente com a quimioterapia que é considerada como auxílio médico na corrente busca à descoberta de novas medicações anticancerígenas ou estratégias terapêuticas. O propósito da presente pesquisa é aumentar a atividade anticancerígena da Carboplatina (CP) pelo incremento do efeito apoptótico de células de câncer pulmonar (MCF-7) em experimentos in vitro pela combinação com oxitetraciclina. Os resultados obtidos confirmaram elevado efeito sinérgico entre oxitetraciclina e Carboplatina em células MCF-7 representativas tratadas com Carboplatina, com e sem diferentes concentrações de oxitetraciclina (5% e 10% de IC50). A oxitetracilina que potencializou a ação da Carboplatina e/ou teve uma notável atividade relatada como um agente isolado. A pesquisa demonstrou a relação sinérgica entre oxitetraxiclina e Carboplatina nos ensaios de viabilidade. Surpreendentemente, os resultados obtidos sugeriram que as estratégias de tratamento inibidor podem aplicar um janela terapêutica da Carboplatina com potencial para a terapia do câncer.(AU)

Although feline mammary carcinoma is not the most prevalent among the species, its aggressive behavior represents a low life expectancy, compared with most undifferentiated types of breast cancer. Tissue stiffness induced by the accumulation of collagen fibers is related to a risk factor for carcinogenesis in healthy women and aggressiveness in those with breast cancer, which can also occur in cats. The objective of this work is to identify the relationship between stromal collagen density and aggressiveness of mammary carcinoma in cats, according to the peripheral and central tissue distribution by the Picrossirius Red histochemical method. Image.J® and MatLab® software were used for digital image processing. The mean values of kurtosis and entropy attributes were grouped into a control group, and low and high-grade carcinoma groups, analyzed with one-way ANOVA and Bonferronis multiple comparison test (p <0.01). Interpretation of stromal dynamics is important to evaluate both central and peripheral locations. According to entropy, there was a significant increase in the peripheral density in the carcinoma groups in relation to the control group, which can be justified by blood support. The same can be said of the central region, with a significant gain in collagen fibers from the tumors, indicated by kurtosis. The results suggest the presence of increases in stromal density in mammary carcinomas of cats, regardless of their graduation, and occurring in both regions.

ABSTRACT Purpose To evaluate capsules formed by microtextured silicone implants with and without Parietex® mesh coverage histologically. Methods Sixty Wistar rats were divided in two groups (meshed and unmeshed). Each group was, then, divided into two subgroups for evaluation at 30 and 90 days. Capsules were analyzed based on hematoxylin and eosin (HE) and picrosirius staining. Results The number of fibroblasts, neutrophils and macrophages was similar among all subgroups. There was a higher lymphocyte reaction in the 30-day meshed group (p = 0.003). Giant cell reaction, granulation tissue and neoangiogenesis were similar among the subgroups. Synovial metaplasia was milder at 90-day in the unmeshed (p = 0.002) and meshed group (p < 0.001). Capsular thickness was significantly greater in the meshed samples (30-day p < 0.001 and 90-day p < 0.001). There was a similar amount of collagen types I and III in both groups. Conclusions The mesh-covered implants produced capsules similar to the microtextured ones when analyzing inflammatory variables. Synovial metaplasia was milder at 90 than at 30 days, and the capsular thickness was significantly greater in the meshed group. A similar amount of collagen types I and III was observed. Due to these characteristics, the mesh coverage did not seem to significantly affect the local inflammatory activity.

In order to evaluate the phytobiotic effect of cashew (Anacardium occidentale L.) leaves powder (CLP) on growth performance, carcass traits, lymphoid organ weights and cecal lactic-acid bacteria in broilers, a total of 720 unsexed broilers of the Ross 308® genetic line were distributed according to a completely randomized design with three experimental treatments, five repetitions and 48 chickens per repetitions. The experimental treatments consisted of a basal diet (DB; T0) and DB + supplementation with 0.50 (T1) and 0.75% (T2) of CLP. At 32 days, the body weight, feed intake and feed conversion ratio were not modified (p>0.05) by the experimental groups; however, T1 increased (p 0.05) the viability compared to T0. Additionally, T2 improved (p 0.05) the carcass and leg yields, although without significant changes (p>0.05) for the other edible portions. Moreover, this natural product (A. occidentale) increased (p 0.05) the ash content and the range of the red tone and decreased (p 0.05) the L* in the breast, although moisture, fat, pH, and b* were not affected by the experimental diets (p>0.05). Similarly, T1 favored (p 0.05) the growth of green bacterial colonies with a white halo, and both treatments with CLP the total cecal acid-lactic bacteria. No significant changes were observed (p>0.05) in the relative weight of the immune organs (thymus, bursa of Fabricius, and spleen). Dietary supplementation with cashew leaves powder enhanced the viability of the batch (0.5%), some edible portions (0.75%), the breast pigmentation, and the cecal beneficial bacteria count in broilers.(AU)

ABSTRACT Purpose To reduce false-negative rates (FNR) in sentinel lymph node biopsy (SLNB) of clinically positive (cN+) axilla in patients undergoing neoadjuvant chemotherapy (NAC). The removal of three or more lymph nodes with dual-tracer mapping including a radioisotope was used. However, in the Brazilian Unified Health System, the radioisotope tracer is not feasible in some hospitals. We conducted a cross-sectional study to evaluate the detection rate of sentinel lymph node (SLN) in patients who converted from cN+ to ycN0 after NAC using blue dye as a single-agent mapping tracer. Methods During the period of March 2018 to September 2019, 34 patients who underwent NAC with cN+ who converted to ycN0 were enrolled in the study. The SLNB was performed using blue dye as a single-agent mapping followed by axillary lymph node dissection (ALND). Results The detection rate of sentinel lymph node was of 85.3%, being SLNB not possible for five patients (14.7%), due to fibrosis. The mean number of removed SLN was 2.5. Conclusions The use of blue dye as a single-agent mapping tracer demonstrated an acceptable detection rate of 85.3%. Although the FNR was possible to be determined, the small sample size might overestimate this rate. The removal of three or more lymph nodes with single-agent mapping tracer might be indicated for breast cancer patients who converted to ycN0 after NAC in the Brazilian health public services, in which radioisotope tracer is not suitable.

A total of 160 medium-sized one-day-old male chicks reared in organic conditions were studied individually from the first day of their life until slaughter (120 days). Two weather periods were considered, being period C colder than period H. A total of 24 chickens per period were randomly selected, then the breast muscle (m. Pectoralis major) was extracted for analysis. Individual fatty acids were measured by gas chromatography and expressed in grams per 100 g of fat. From the values obtained, total lipid fractions were calculated. Near infrared spectroscopy spectra (NIRS) were recorded on the surface of the breast without manipulating. Breast from chicken reared in H period had significantly lower (p<0.05) saturated fatty acids / polyunsaturated fatty acids (SFA / PUFA) ratio, and increased (p<0.05) content in PUFA and n-6. However, no significant differences were observed on the content of individual fatty acid. NIR system was not able to correctly classify the samples according to the breeding period.(AU)

ABSTRACT This experiment was conducted to investigate the effects of leucine (Leu) supplementation on performance and carcass traits of broilers along with the re-evaluation of standardized ileal digestible (SID) Leu requirement of broiler chicks on days 0-14. Birds were fed seven experimental diets with SID Leu levels of 16.3 (basal), 17.3, 18.3, 19.3, 20.3, 21.3, and 22.3 g/kg. Some 280 male broiler chicks were randomly assigned to the seven dietary treatments in a completely randomized design with five replicates. Dietary SID Leu levels had significant effects on body weight gain(BWG), feed conversion ratio(FCR), carcass, and breast percentages. BWG increased (p 0.05) when the broilers fed diets with 18.3 to 21.3 g/kg SID Leu compared to the basal diet. FCR decreased (p 0.05) by dietary SID Leu levels of 18.3 to 21.3 g/kg in comparison with the basal diet. Carcass yield increased (p 0.05) by SID Leu levels of 19.3 to 21.3 g/kg. Breast percentage increased (p 0.01) when the broilers received diets with 18.3 to 22.3 g/kg SID Leu and the highest value obtained by the SID Leu level of 21.3 g/kg. Abdominal fat decreased linearly by Leu supplementation (p 0.01). Regression analysis indicated the SID Leu requirement of 18.67, 18.88, and 19.85 g/kg for BWG, FCR, and breast meat yield, respectively. It can be concluded that broiler performance characteristics, especially breast yield, improve by Leu supplementation. Based on the results, the Leu requirement of modern broilers has increased dramatically due to genetic selection for a higher growth rate.
Resumo
A total of 160 medium-sized one-day-old male chicks reared in organic conditions were studied individually from the first day of their life until slaughter (120 days). Two weather periods were considered, being period C colder than period H. A total of 24 chickens per period were randomly selected, then the breast muscle (m. Pectoralis major) was extracted for analysis. Individual fatty acids were measured by gas chromatography and expressed in grams per 100 g of fat. From the values obtained, total lipid fractions were calculated. Near infrared spectroscopy spectra (NIRS) were recorded on the surface of the breast without manipulating. Breast from chicken reared in H period had significantly lower (p<0.05) saturated fatty acids / polyunsaturated fatty acids (SFA / PUFA) ratio, and increased (p<0.05) content in PUFA and n-6. However, no significant differences were observed on the content of individual fatty acid. NIR system was not able to correctly classify the samples according to the breeding period.

ABSTRACT: To increase the amount of meat produced, researchers have promoted intensive genetic selection for growth rate and muscling and have improved nutrition and management conditions. However, there has been an increase in the number of reports of breast muscle myopathies observed in poultry processing plants, including white striping (WS) and wooden breast (WB). This study aimed to evaluate and to compare the occurrence of WS and WB myopathies in three poultry processing plants and to perform an anatomopathological characterization, including macroscopic and microscopic analyses. A total of 408,334 carcasses were condemned or downgraded due to the presence of WB or WS, which represents 0.73% of the total number of slaughtered animals during the evaluated period. WB was more frequent than WS, but the occurrence of each myopathy varied significantly according to each establishment. WB was more frequent in the establishment which includes only male flocks, an average age of 45 days, and an average live weight of 2775g (B). WS was more frequent in establishment with male, female and mixed flocks, average ages ranging from 41 to 44 days, and average live weight 1731g-2830g (A). It is probably related with specific condition of each poultry company, including genetics, age, nutrition and management conditions. Macroscopically, WB and WS lesions are characterized by hypertrophy and stiffness of the pectoralis major muscle. Under microscopy, the myopathies showed similarities regarding the detected histological abnormalities, characterized by a process of myodegeneration, although the connective tissue infiltrate was more severe in the breasts with WB than in those with WS myopathy. The results found in this study demonstrate that the rates of condemnation for these myopathies are high, vary significantly among the analyzed companies and may cause major economic losses for the productive sector in the region.
RESUMO: Para melhorar a quantidade de carne produzida, os pesquisadores têm promovido ao longo dos anos uma seleção genética intensiva para a taxa de crescimento e desenvolvimento de músculos, além de melhorias nas condições nutricionais, sanitárias e de manejo. No entanto, houve um aumento no número de relatos de miopatias dos músculos do peito observados em abatedouros-frigoríficos de aves, incluindo white striping (WS) e wooden breast (WB). O objetivo deste estudo foi avaliar a ocorrência de WB e WS em três abatedouros-frigoríficos de aves e realizar uma caracterização anatomopatológica, incluindo análises macroscópicas e microscópicas. Um total de 408.334 carcaças foi condenado (total ou parcialmente) devido à presença WB ou WS, o que representou 0,73% do número total de animais abatidos durante o período avaliado. Apesar de a ocorrência de WB ter sido maior do que a de WS, a frequência de cada uma variou significativamente de acordo com o estabelecimento. Resumo
Objetivou-se avaliar o efeito da suplementação de cromo-metionina em dietas para frangos de corte criados em estresse por calor, no período de 22 a 43 dias de idade, nos parâmetros de qualidade da carne. Foram utilizados 336 frangos de corte, machos, da linhagem Cobb 500, com 21 dias de idade, distribuídos em delineamento em blocos inteiramente ao acaso, com quatro blocos (cada câmara climática), seis tratamentos (0; 0,10; 0,20; 0,40; 0,80 e 1,20mgkg-1 de Cr na forma de Cr-metionina), oito repetições e sete aves por unidade experimental. Aos 43 dias de idade, duas aves por unidade experimental foram selecionadas e abatidas para avaliação da qualidade da carne de peito, por meio dos parâmetros de pH15min, pH24h, luminosidade (L*), teor de vermelho (a*), teor de amarelo (b*), croma (C*), ângulo hue (Hº), capacidade de retenção de água, perda de peso por cozimento e força de cisalhamento. Houve efeito quadrático (P=0,0070) na capacidade de retenção de água da carne de peito. A suplementação de CrMet não afetou (P>0,05) os demais parâmetros de qualidade da carne. Assim, recomenda-se a suplementação de 0,59mgkg-1 de CrMet para frangos de corte para melhoria da capacidade de retenção de água do peito.(AU)
