Resumo

The purpose of this study was to evaluate the effects of salinity and plant-based diet or animal-plant combination diet on the performance and metabolic status of juvenile Nile tilapia (Oreochromis niloticus). The experimental design was completely randomized in a 4 × 2 factorial scheme with four replicates. The treatments were established by the combination of salinities of 0, 10, 20, and 30 g L-1 with an animal-plant combination diet (AP) or plant-based diet (P). The replicates were 60 L tanks with 12 fish per tank. Diets were provided for 32 days, and the fish were fed three times a day (8, 12, and 17 h) until apparent satiety. Daily feed intake (DFI) was measured, body weight (BW) was recorded at the beginning and end of the trial, and total length (TL) and standard length (SL) were measured at the end of the trial. Average daily gain (ADG), specific growth rate (SGR), feed conversion ratio (FCR), and survival rate were calculated. After the biometric measurements were made at the end of the trial, blood samples were collected to determine the plasma concentrations of total protein (TP), glucose, cholesterol, and triglycerides (TG). The fish were euthanized, and the hepatopancreas was collected and weighed; thereafter, the hepatosomatic index (HSI) was calculated. An interaction was detected between salinity and diet type for final BW, ADG, TL, and SL. These traits were not influenced by salinity when it was associated with the AP diet, but reduced linearly with salinity in the P diet. DFI and survival rate were independently affected by salinity: DFI reduced linearly with salinity levels and survival rate was higher at a salinity of 10 g L-1. HSI increased linearly with salinity levels and was lower in the P diet than in the AP diet. Salinity had a quadratic effect on plasma TP, and the maximum value for this metabolite (2.96 g dL-1) is attained at a salinity of 10.26 g L-1. There was an independent effect of diet on the plasma concentrations of cholesterol and TG, which were lower in the P diet than in the AP diet. The salinity of 10 g L-1 associated with diet composed of animal and plant ingredients led to a better performance, higher survival rate, and less stressful environmental conditions for juvenile Nile tilapia.(AU)

Lincomycin is a broad-spectrum antimicrobial acting against Gram-positive bacteria, widely used in veterinary medicine. In fish, there are only limited in vitro data, thus requiring the design of effective therapeutic protocols for their use in aquatic organisms. In this context, the objective was to evaluate the clinical safety of lincomycin treatment, administered orally in tilapia, through hematological, biochemical and somatic index evaluation. A total of 136 tilapia (+100g) were randomly distributed in 17 tanks (100L of water, n=8) to constitute the following treatments: TO (control group, not treated with lincomycin); T1, T2, T3 (treated with 10, 20 and 40mg/kg¹ of lincomycin b.w., respectively) and T4 physiological standard (reference values). Eight animals were sampled per treatment in 4 periods: 2, 4 and 8 days post-treatment (DPT), and a group that was treated for 8 days with the drug and then treated only with commercial feed until the 12th day (recovery period). Tilapia treated with lincomycin had no difference in the hematological and leukocyte evaluation, in the hepatic, renal and splenic somatic index. However, they presented a transient increase in the values of ALT, AST, cholesterol, triglycerides and creatinine, which returned to normal levels after the period of recovery (12DPT). Furthermore, an increase in total protein, albumin and globulin levels was observed in treated animals. It is concluded that although there were some transient changes during the experiment, lincomycin has a good clinical safety margin at doses of 10, 20 and 40mg/Kg¹ b.w. for Nile tilapia.

This study aimed to evaluate the clinical safety of doxycycline treatment (Sandoz Pharmacetical Industry from Brazil Ltd.), administered orally incorporated into the feed of Nile tilapia, Oreochromis niloticus. For this purpose, 75 Nile tilapia (±300g) from the same spawning were randomly distributed in 15 tanks (n=5), containing 100 L of water each, supplied with running water free from chlorine, to establish the following treatments: TO (Control - not treated with doxycycline); T1, T2, T3 and T4 (treated with 10, 20, 40 and 80mg of doxycycline/kg of body weight, respectively), in which 5 animals per treatment were sampled in 3 periods: 2, 4 and 8 days post-treatment (DPT). Blood samples were collected for hemogram determination and serum biochemical evaluation, as well as spleen, liver and kidneys (cranial and caudal) for somatic and histopathological evaluation. The results showed no significant difference among treatments in the serum values of creatinine, total protein, cholesterol, triglycerides, globulin, glycemia and alkaline phosphatase enzyme activity. However, serum levels of AST (aspartate aminotransferase) were significantly higher (P<0.05) in animals treated with 80 mg of doxycycline in the longest period of treatment (8 days). In the hematological evaluation of tilapia treated with doxycycline, no significant changes (P>0.05) were observed in erythrocyte counts, hematocrit, MCV, HCM and CHCM values. Doxycycline-treated tilapia did not present significant difference in the somatic analysis of spleen, liver and kidney when compared to control group. Therefore, the results demonstrated the clinical safety of oral treatment with doxycycline at doses of 10, 20, 40 and 80 mg/kg of b.w., although transient changes in liver functionality were observed after eight days of treatment with the dose of 80mg/kg.

Zileuton is an inhibitor of the 5-lipoxygenase enzyme that transforms essential fatty acid (EFA) substrates into leukotrienes (LTB4, LTC4, LTD4 and LTE4), but little is known about the use of this drug in teleost fish. Therefore, the objective of this study was to evaluate the clinical safety of treatments with 2,25 mg and 4,50 mg of zileuton/Kg-1 (bodyweight), administered orally in the diet, through biochemical and hematological analysis during the acute inflammatory reaction in Nile tilapia (Oreochromis niloticus), induced by Aeromonas hydrophila bacterins. The study used eighty tilapias, conditioned in 20 tanks (n=4), constituting the following treatments: T0 (control), T1 (2,25 mg zileuton) and T2 (4,50 mg zileuton), being sampled eight animals per treatment in three periods: 6, 24 and 48 hours post-inoculation, and a 10th group consisting of fish without any type of stimulus to obtain the reference values. In order to evaluate and determine the blood count and serum biochemical, it was necessary to collect blood samples. The hematology results of the tilapia treated with zileuton did not reveal alterations between tilapia subjected to different treatments and control fish (T0). The liver cytotoxicity analysis of tilapias treated with zileuton did not reveal significant (p≥0,05) alterations in AST and ALT serum enzymatic activity. The study of tilapia blood total protein showed decrease in the T1 group at 48 HPI. As the treatment time progressed, the results indicated decrease in the serum albumin levels for T2 group at 24 HPI. The determination of serum biochemichal of creatinine, cholesterol, triglycerides, and glucose did not differ statistically between treatments. The results observed in the hematological and biochemical analyzes allows to conclude that zileuton administered orally, at doses of 2,25 and 4,50 mg/Kg-1 (body weight) demonstrated to be clinically safe.(AU)

This study attempted to determine the associations between metabolic imbalances and lameness or hoof injuries in high-producing Holstein × Gir cows, and to determine whether the metabolic profile affects the occurrence of lameness. Eighty cows were followed from -60 to 60 days relative to calving and hoof injuries were reported on days -60, 7 and 60. Locomotion score (LS), body condition score (BCS), the concentrations of non-esterified fatty acids, ß-hydroxybutyrate, glucose, cholesterol, albumin, total protein, blood urea nitrogen (BUN), calcium, phosphorus, and magnesium, and the activity of aspartate aminotransferase were determined at days -42, -21, -7, 0, 7, 21 and 42. The McNemar and Chi-square tests were used to compare frequencies of lameness and hoof injuries over time and to verify the associations between lameness, BCS, hoof injuries, and metabolic disorders. Two-way repeated measures ANOVA was used considering groups (non-lame × lame cows) and variations of BCS and metabolites over time. Lameness and hoof injuries increased between days -60 (20% and 66.3%) and 60 (44.7% and 98.6%). Excessive postpartum loss of BCS (P=0.017) and subclinical hypocalcemia (P=0.012) were associated with lameness on day 60. In general, the metabolic profile did not differ between lame and non-lame cows but cholesterol, albumin, BUN and magnesium concentrations were higher in non-lame cows. The postpartum decrease in BCS can affect the occurrence of lameness, and the metabolic profile of lame cows shows little difference from that of non-lame cows.

The present study aimed to evaluate increasing levels of fish waste oil in diets for laying hens on serum biochemistry profile. 192 Hisex White laying hens at 29 weeks of age were used, with water and food ad libitum. The experimental design was completely randomized consisting of eight treatments corresponding to the inclusion levels of fish waste oil (0, 0.5, 1.0, 1.5, 2.0, 2.5, 3.0 and 3.5%) in the diets, with four replicates of six birds each. Data collected were subjected to polynomial regression at 5% of significance. Significant differences (P<0.05) were observed in triglycerides, glucose, total cholesterol, and uric acid. These parameters presented a decrease when hens fed diets with higher level of fish waste oil. The results of the present study indicated that the inclusion of fish waste oil caused a significant effect in the serum biochemical profile of laying hens, especially in glucose, triglycerides, total cholesterol, and uric acid concentrations. The inclusion level of 3.5% of fish waste oil caused larger disequilibrium in the serum biochemical profile of laying hens.(AU)

In the present study, seventy-two 30-week-old laying hens (Brown Hisex) were separated into two groups (control and treatment) and fed with a standard layer diet. In the treatment group the amount of effective microorganism (EM) added to the drinking water is equal to1000 ppm EM dose. Throughout the 8-week study period, 16 h thermoneutral (20-22 ºC) and 8 h hot (35-37 ºC) environmental temperature regime was applied daily. The results indicated that EM supplementation affected performance and some egg quality characteristics of layers exposed high environmental temperature. Feed intake and conversion ratio, yolk index, albumen index, yolk colour b, Haugh unit were decreased by supplemental EM. Serum glucose, cholesterol, triglycerides, calcium, total oxidant- antioxidant concentration were not affected by EM. It is concluded that EM supplementation in laying hens could have potential to improve feed intake and feed conversion ratio under high environmental temperature. However, supplemental EM with drinking water decreased some egg quality parameters.(AU)

Measuring metabolic parameters in the blood has been an indispensable tool for assessing the productive and health status of dairy cows for more than 100 years. The values of laboratory parameters depend on various preanalytical, analytical and postanalytical factors. The most important preanalytical factors are sample transport time and temperature, hemolysis, anticoagulant type, and sample volume. Preanalytical factors can lead to reduced stability of the analyte in the sample, which changes their concentration. Loss of stability changes the time of storage and manipulation of the sample, which determines the criteria for its acceptance or rejection. The two stability indicators are stability limit and maximum permissible instability. A stability limit (SL) is defined as the period of time in which a property variation does not exceed a maximum permissible instability (MPI). The aim of this study was to determine the SL and MPI for each analyte in the blood serum of cows and to determine whether SL differs in the function of the presence of preanalytical errors in the blood sample. Three hundred samples of dairy cow origin in different periods of lactation participated in this research. They were classified into 6 groups of 50 samples: according to the time from sampling to processing in the laboratory (0-4 h, 4-8 h and over 8 h; all transported on dry ice, protected from environmental factors, without preanalytical errors) and according to the presence of preanalytical errors (group with hemolysis, a group transported at ambient temperature and a group with a small sample volume). Each sample was aliquoted in two portions. One portion was left at +4°C and tested once a day for 6 days of sample storage, and the second portion, placed at -20°C, was tested once a month for 6 months. The MPI had a value ranging from 1.51 to 8.4. Metabolic profile analytes with lower MPI values (1.51-3.22) were albumin (ALB), total protein (TPROT), UREA, glucose (GLU), calcium (Ca), and phosphorus (P). Higher MPI values (5.1-8.4) were found for nonesterified fatty acids (NEFA), beta-hydroxybutirate (BHB), cholesterol (CHOL), triglycerides (TGC), total bilirubin (TBIL) and aspartat aminotransferase (AST). For most parameters, we can conclude that their PD% changed faster in storage conditions at +4°C compared to the regime of -20°C. The largest number of biochemical analytes in bovine blood serum shows preserved stability in the first 6 days at +4°C or 6 months at -20°C if transported to the laboratory within 8 h after sampling in ideal conditions and without the action of preanalytical errors. Prolonged transport under ideal conditions or the existence of preanalytical errors such as transport at room temperature, hemolysis or small sample volume shorten the stability of the ALB, NEFA, GLU, UREA and P. Concentration of all analytes decreases during the stability test except for UREA, NEFA, BHB and for CHOL and TGC in some groups. Variations in parameters such as BHB, NEFA, TBIL, AST, and Ca have shown potential clinical significance. At storage conditions at +4°C, clinically significant variations at at least one measurement point were found for AST (7.5% of samples), BHB (6.1% of samples), NEFA (9.9% of samples) and for TBIL (in 7% of samples). This study can help define acceptable delay times and storage conditions for bovine blood samples, which is of great importance because in working with farm animals it is often not possible to take samples in a short time and deliver them to the laboratory, and samples are often burdened with certain preanalytical errors with limited possibilities of re-sampling.(AU)

Background: Otitis is a frequent condition in dogs, with a multifactorial etiology covering primary causes, predisposing and perpetuating factors. It is characterized by an inflammatory process of structures that make up the auditory system, with prevalence data ranging from 15 to 20%, which can occur in any age group and without sex predisposition. According to the location, it can be classified as uni or bilateral, affecting the outer, middle and / or inner ear, the latter two being associated with neurological changes such as vestibulopathy, facial nerve paresis and Horner's syndrome. The morbidity and mortality of otitis media and internal may increase with delay in diagnosis, resulting in delay in the correct treatment, and the extension to brainstem is described in felines and considered rare in dogs. The present study aims to report an atypical case of otitis media and internal, with extension to the brain parenchyma in a brachycephalic dog. Case: A 3-year-old bitch French Bulldog was seen, weighing 12 kg, with a history of vomiting, apathy, nystagmus, ataxia and acute evolution hemiparesis. As a result of adoption, it was not known about the animal's morbid past. On special physical and clinical examination, depressed mental status, changes in postural reactions, nostril stenosis with unilateral mucopurulent nasal discharge, corneal ulcer, palpebral and labial ptosis, absence of eyelid reflex and spontaneous strabismus were observed, all of these findings observed on the left side of the face. No signs of otitis external to otoscopy were found. The clinical signs found characterize left facial nerve paresis and peripheral and central vestibulopathy. Otitis media / internal and meningoencephalitis were suspected. Hemogram, serum biochemistry (glucose, urea, creatinine, alanine aminotransferase, total proteins, albumin, globulin, alkaline phosphatase, gamma glutamyltransferase, phosphorus, total bilirubin, cholesterol and total calcium), urinalysis, rapid tests for erlichiosis, anaplasms , borreliosis, heartworm, distemper and leishmaniasis, CSF analysis, neurological panel performed by PCR (Polymerase Chain Reaction) that tested the animal for Bartonellosis, Lyme Disease (Borrelia burgdorferi), Blastomyces dermatitidis, Cryptococcus sp., Virus West Nile, Distemper, Toxoplasmosis, Neosporose and Coccidioides sp. using conjunctival swab, whole blood, urine and CSF samples. The samples were negative for all of these tests. As an image exam, brain magnetic resonance was performed. In the latter, otitis media and left internal was observed with extension of the lesion to the brain parenchyma. CSF analysis showed cytological changes compatible with a non-suppurative inflammatory process, mild pleocytosis with predominantly mononuclear leukocyte infiltrate. As a form of treatment, antibiotic therapy and total ablation of the auditory canal with lateral bulectomy were performed, showing good results with reduced clinical symptoms. Discussion: It is concluded that although the diagnosis of otitis media and internal with extension to brainstem is more common in cats, it should be included as a differential for central vestibular disorders in dogs, and that the diagnosis together with early and effective treatment are important, since that the evolution of the infection to the central nervous system represents a risk to the patient.

We aimed to evaluate the impact of milking management practices on milk quality though somatic cell count (SCC) and total bacterial count (TBC). By means of a survey that included farmers associated to two cooperatives, namely Cosulati in the state of Rio Grande do Sul and Castrolanda in the state of Paraná, the hypothesis that milking handling practices impact SCC and TBC levels negatively, regardless of the technological development level of the farm, was analyzed. For such, the quantile regression method was used to estimate models for the 10th, 50th, and 90th quantiles. The results confirmed that SCC and TBC levels decrease as the number of practices increases. However, only three practices impact SCC and TBC levels significantly, and their impact is not related to the farm technological level, according to tests performed by quantile regression. Finally, the study showed that some milk quality and management practices affect somatic cell count and total bacterial count more than others, which consists of using the correct active principle during dry cattle, post-dip, and the water temperature control periods.(AU)

During the periparturient dairy cows undergo a large energy demand, at the same time reducing their intake of dry matter. The negative energy balance resulting from this equation leads to severe metabolic disorders in production, and mainly in reproduction. The aim of this study was to evaluate the effect of protected choline on metabolic parameters, reproductive performance, and milk production in cows during the transition period. Fifty-four dairy cows were divided into three groups: control, supplementation with choline for 10 days prepartum (T10) and supplementation with choline for 20 days prepartum (T20). After delivery we measured fructosamine levels, cholesterol, non-esterified fatty acids (NEFA), beta-hydroxybutyrate (BHB), aspartate aminotransferase (AST), gamma glutamyltransferase (GGT), and total oxidant (TOS) on days 10, 20 and 30. We also evaluated milk production and interval between calving and conception. There was no effect of supplementation with choline on blood and production parameters. The interval between calving and conception was lower in the T20 group. Choline supplemented by 20 during the antepartum improved reproductive performance of dairy cows, although it did not change the metabolic profile.(AU)

This study aimed both to evaluate the metabolic profile of lambs fed with extruded forage and concentrate containing different roughageconcentrate ratios (R:C) and to evaluate in-vitro ruminal fermentation kinetics of these rations. An experiment was carried out with 20 weanling, female crossbred lambs (½Dorper x ½Santa Inês) aged 120 ± 8 days. The treatments were four R:C ratios of extruded forage (Foragge®) and extruded concentrate (Beef Total®) – 30%F:70%C, 40%F:60%C, 50%F:50%C, and 60%F:40%C – distributed randomly with five replicates. The analysis targeted energy metabolites (VLDL, triglycerides, and cholesterol), hepatic metabolites (alkaline phosphatase, gamma glutamyltransferase [GGT], and aspartate aminotransferase [AST]), and protein metabolites (total protein, uric acid, urea, creatinine, albumin, and globulin). In-vitro gas production was quantified by inoculating the extruded rations in bovine ruminal fluid with the following forage:concentrate proportions evaluated: 30%F:70%C, 40%F:60%C, 50%F:50%C, 60%F:40%C and 70%F:30%C. Fermentation kinetic values were compared using parallelism test and curve identity at the error probability level of 5%. A regression study was used to compare the means of the metabolites at the error probability level of 5%. Average dry matter intake data are provided descriptively. The extruded ration increased the hepatic metabolites (GGT and AST) without compromising the liver and kept the energy metabolites stable. Similarly, the serum concentration of protein metabolites remained at normal levels recommended to the ovine species. In-vitro fermentation pattern displayed high gas production associated with high ration digestibility. The extruded ration provides protein and energy supply to meet the animals’ nutritional requirements without compromising their health.

The effects of dietary camelina cake (CAMC) on broiler chicksgrowth performance, plasma lipid status and fatty acid profile (FA) of lymphoid organs were evaluated. Six hundred broilers (3-week-old, Cobb 500) were randomly allotted for 20 days in a feeding trials into 2 groups: control (C; corn-soybean meal-canola meal basal diet) and experimental (CAMC; 80 g CAMC/kg diet replaces canola meal from C diet). Blood samples (n=12/group) were collected on day 42 for plasma profile analysis (glucose, Glu; total cholesterol, TC; high-density lipoprotein cholesterol, HDL-C; low-density lipoprotein cholesterol, LDL-C; triglycerides) and immune organs (spleen, thymus, bursa of Fabricius) for FA analysis. The FA profile of lymphoid tissue was determined by gas chromatography. Feeding the CAMC diet did not influence broilers performance parameters or relative weights of lymphoid tissue, except the weight of bursa that decreased (p 0.05). Plasma lipids profile was affected by decreasing (p 0.05) the concentrations of Glu, TC, HDL-C and LDL-C in broilers fed the CAMC diet. In all lymphoid tissue, the total content of n-3 polyunsaturated fatty acids (PUFA) increased (p 0.001) and the total level of n-6 PUFA decreased (p 0.001) as the effect of the CAMC diet. The n-6:n-3 ratio decreased (p 0.001) up to 5:1 in all lymphoid tissue. The higher (p 0.05) concentration of arachidonic acid was found in the spleen, followed by the thymus and the bursa of the chicks fed the CAMC diet. Our results indicate that feeding 80 g/kg camelina cakes in broiler chicks finisher diet had no negative impact on productivity, beneficially alter the plasma lipid status and fatty acids profile of lymphoid tissue.(AU)

To investigate the effects of soy isoflavones (ISF) on the laying performance of Xueshan breeder hens at different phases of the egg production cycle, 32 weeks (wks) old local Chinese breeder hens were assigned to two groups with three replicates (750 hens/replicate) for control and treatment groups. Then every group was divided into three sub-groups following 4, 12, and 20 wks administration of control or ISF diet. General Linear Model procedure was used (2× 3), and treatments were considered of two factors (diets and time) with different levels (control and ISF diets; 4, 12, and 20 wks of diet supply time). Basal diet with 3 mg/kg ISF was used. Performance, egg quality, organs relative weight and cholesterol level were measured at three time-points of 36, 44, and 52-wks of age old. The results showed that both diets and time significantly affected the production performance and physiological parameters. In details, ISF inclusion significantly (p=0.02) decreased average egg weight and average feed intake compared to control at 52-wks of age old, however, feed conversation ratio was not affected by ISF diet. Moreover, ISF diet caused to an increase hatching rate (p=0.01) at 36-wks old hens, an abnormal egg rate decrease (p=0.02) at 44-wks old, an increase in yolk color (p=0.035) and Haugh unit (p=0.039), and a decrease in low-density lipoprotein cholesterol level (p=0.03) at 52-wks of age old. ISF diet also increased spleen relative weight (p=0.041) and total antioxidant capacity compared to the control group. The results revealed that ISF inclusion exerted some benefits, thus it could be a potential feed additive for the breeder hens.(AU)

This study investigated the existence of difference of fat deposition and lipid metabolism in horses with different races and skills that were used for the same kind of sport. 20 Purebred Arabian and 20 Thoroughbred horses trained for flat race were evaluated. The analyses performed were body condition score, weight and blood collected for determination of triglycerides, total cholesterol and non-esterified fatty acids. Ultrasonography of the thickness of the subcutaneous fat layer was performed on the Longissimus dorsi muscle between the 17th and 18th rib, the thickness of the subcutaneous fat layer on the Gluteus medius muscle using the acetabulum as reference, and the cross section of the same muscle. Race-trained Arabian horses showed greater fat layer deposition in the Gluteus medius and Longissimus dorsei muscles than Thoroughbred horses. These facts indicate that there is a metabolic difference, besides the phenotype, between the races. They also indicate the need to study specific physical conditioning programs for each kind of race.(AU)
