PURPOSE: To investigate the settling characteristics of a scleral lens in Chinese adults with refractive error and to provide guidance for its clinical application. METHODS: A total of 21 healthy Chinese adults (27.2 ± 4.1 years) with refractive error were enrolled in this study. The average spherical equivalent was -5.50 ± 2.92 D. Subjects were fitted with 15.6 mm diameter scleral lenses. The central post-lens tear thickness (PoLTT) was measured immediately after lens placement, 30, 60, 120, and 240 min after lens insertion at the dispensing visit and immediately after lens placement and 240 min after three months through optical coherence tomography. Statistical analyses were conducted using repeated measures analysis of variance and paired-t test. RESULTS: At the dispensing visit, the amount of settling after 240 min of lens wear was 126 ± 33 µm. After three months, the amount of settling was 98 ± 55 µm after 240 min. No significant difference was detected in the PoLTT immediately after lens placement between the dispensing visit and after three months (t = -0.246, p = 0.807), while a significant difference was noted at 240 min after lens insertion (t = -6.575, p < 0.001). The amount of settling was higher at the dispensing visit than that after three months (average difference = 28 ± 63 µm, t = 2.733, p = 0.01). The prediction model of PoLTT over time was y = 26.263-0.690 × t + 0.001 × t2 + 0.926 × y30 (R2 = 0.939), where y denotes the predicted PoLTT at t min after lens insertion, y30 denotes the PoLTT at 30 min after lens insertion. CONCLUSION: For the investigated small-diameter scleral lens (material: Boston XO, diameter: 15.6 mm, four-zone and periphery toric design), the PoLTT decreased over time after lens insertion in Chinese adults with refractive error, and the amount of settling varied among individuals (range: 71-204 µm). The amount of settling did not increase further after three months, indicating the long-term fitting stability of the scleral lens. Practitioners could estimate the PoLTT using the prediction model based on the PoLTT at 30 min after wearing lenses.

Purpose: To clarify the effects of acute hyperglycemia on the responses of choroidal structural components and vascularity index during light modulation in healthy participants using techniques including image binarization and artificial intelligence (AI) segmentation based on swept-source optical coherence tomography (SS-OCT). Methods: Twenty-four eyes of 24 healthy participants were imaged at different stages after ambient light, 40 min of dark adaptation, and 5 min of light adaptation in two imaging sessions: control and after receiving 75 g of oral glucose solution. The choroidal structural parameters, including luminal volume (LV), stromal volume (SV), total choroidal volume (TCV), and choroidal vascularity index (CVI) within a 6 mm area were determined using a custom algorithm based on image binarization and AI segmentation of SS-OCT. These measurements were compared among the conditions after adjusting for axial length, age to identify the differences. Results: In the dark, CVI decreased (-0.36 ± 0.09%) significantly in acute hyperglycemia compared to the control condition. During the transition to ambient light, there was an increasing trend in the choroidal parameters compared with the control experiment. However, only TCV (0.38 ± 0.17 mm3) and LV (0.27 ± 0.10 mm3) showed a significant increase at the time point of 5 min after ambient light. Conclusion: Analysis of choroidal structural parameters and CVI based on SS-OCT images is a potentially powerful method to objectively reflect subtle changes in neurovascular coupling between the choroid and photoreceptor during dark adaptation.

Viruses are the most abundant microorganisms on the earth, their existence in contaminated waters possesses a significant threat to humans. Waterborne viral infections could be fatal to sensitive population including young child, the elderly, and the immune-compromised. It is imperative to remove viruses during water treatment to better protect public health, especially in the light of evidence of detection of coronaviruses genetic fragments in raw sewage. We reported bench-scale experiments evaluating the extent and mechanisms of removal of a model virus (spring viremia of carp virus, SVCV) in water by adsorption. Microspheres made by boronic acid-modified bacterial cellulose with excellent mechanical strength were successfully fabricated as packing materials for the column to remove glycoproteins and enveloped viruses from water. The synthesized adsorbent was characterized by attenuated total reflectance Fourier transform infrared (ATR-FTIR) spectroscopy, Atomic Force Microscopy (AFM), Scanning Electron Microscopy (SEM), and Brunauer Emmett Teller (BET) measurement. The adsorption efficiency of glycoproteins was investigated by SDS-PAGE and the Broadford protein assay, while the binding capacity with the virus (spring viremia of carp virus) was monitored by cell culture to calculate the viral cytopathic effect and viral titer caused by the virus. The data obtained from the above experiments showed that ∼3-log removal of SVCV in 3 h, which significantly reduced the virus concentration from microspheres packed column. The present study provides substantial evidence to prove beyond doubt that material based on bacterial cellulose seems to have the potential for virus removal from water which can be extended to systems of significant importance.

PURPOSE: To investigate the impact of penetration and image analysis in different optical coherence tomography (OCT) instruments on the measurement of choroidal vascularity parameters. METHODS: Twenty-three healthy volunteers were imaged using two swept-source OCTs and one spectral-domain OCT. A fully automatic segmentation method based on ResNet-UNet and Niblack local threshold binarization was performed to quantify the relevant choroidal vascular parameters, including choroidal vascularity index, total choroidal volume, and luminal volume. The intraclass correlation coefficient (ICC) and coefficient of repeatability (COR) were used to analyze the repeatability and consistency of automatic and manual segmentation, respectively. RESULTS: Both swept-source OCT devices showed good consistency of luminal volume and total choroidal volume measurements (all ICC value >0.98 with COR% < 8.53%) based on manual segmentation, whereas the consistency of the spectral-domain OCT was lower (ICC value <0.60 with COR% > 40%), which was greatly improved after using the automatic algorithm (ICC value >0.99 with COR% < 4%). The repeatability of choroidal vascularity index obtained from different OCT images using manual or automatic segmentation showed good agreement (all ICC values >0.85), whereas the choroidal vascularity index measurement from the spectral-domain OCT was larger than the other two swept-source OCT devices (ICC value <0.65). CONCLUSION: For healthy youngsters, the penetration of OCT plays a role in the measurement precision for choroidal vascularity parameters, and automatic segmentation can improve the ability of choroidal boundary identification with deficient penetration, suggesting these factors need to be considered in clinical work.

RESUMO

Herein we report a nickel-catalyzed cross-electrophile ring-opening reaction of vinyl epoxides wherein aryl iodides, alkyl iodides, and benzyl chlorides can all serve as the electrophilic coupling partners, providing a new approach to preparing multisubstituted allylic alcohols. This new method features broad substrate scope (76 examples), good step-economy, and high L/B- and E/Z selectivity as well as mild reaction conditions.

Significant concerns have been raised regarding to the pollution of antibiotics in recent years due to the abuse of antibiotics and their high detection rate in water. Herein, a novel super adsorbent, boronic acid-modified bacterial cellulose microspheres with a size of 415 µm in diameter was prepared through a facile water-in-oil emulsion method. The adsorbent was characterized by atomic force microscopy, scanning electron microscopy, and fourier transform infrared spectroscopy analyses to confirm its properties. The microspheres were applied as packing materials for the adsorption of tetracycline (TC) from an aqueous solution and hoggery sewer via the reversible covalent interaction between cis-diol groups in TC molecules and the boronic acid ligand. TC adsorption performance had been systemically investigated under various conditions, including the pH, temperature, TC concentration, contact time, and ionic strength. Results showed that the adsorption met pseudo-second-order, Elovich kinetic model and Sips, Redlich-Peterson isothermal models. And the adsorption process was spontaneous and endothermic, with the maximum TC adsorption capacity of 614.2 mg/g. After 18 adsorption-desorption cycles, the adsorption capacity remained as high as 84.5% compared with their original adsorption capacity. Compared with other reported adsorption materials, the microspheres had high adsorption capacity, a simple preparation process, and excellent recovery performance, demonstrating great potential in application on TC removal for water purification and providing new insights into the antibiotic's adsorption behavior of bacterial cellulose-based microspheres.

In this protocol, we demonstrate our discovery that iron is able to efficiently catalyze the reductive allylic defluorinative ketyl olefin coupling reaction between α-trifluoromethyl alkenes and unactivated ketones. This operationally simple cross-electrophile reaction circumvents the use of pre-generated organometallics and allows for the synthesis of diverse functional-group-rich tertiary gem-difluorohomoallylic alcohols through a polarity-reversed strategy. Preliminary mechanistic studies support a mechanism that proceeds through a ketyl formation/olefin insertion/ß-fluoro elimination sequence.

In this work, we theoretically study the generation of circularly polarized XUV vortices from high harmonic generation driven by bicircular Laguerre-Gaussian (LG) fields with different frequency ratios, by using the strong-field approximation theory. Our simulation shows that the amplitude of the generated vortices from the ω-3ω bicircular LG field is about one order of magnitude larger than that from the ω-2ω field, irrespective of the harmonic order and the orbital angular momentum of the bicircular driving fields. Our analysis shows that the great increase of the vortex amplitude for the ω-3ω field originates from the harmonic enhancement of a single atom. Furthermore, in terms of quantum-orbit theory, the underlying physics of the harmonic enhancement of the single atom for the ω-3ω field is revealed. Our work provides a simple and robust method to increase the amplitude of the circularly polarized XUV vortices.

Herein we demonstrate the first successful application of cycloalkyl silyl peroxides (CSP) as an electrophilic coupling partner in the cross-electrophile coupling reaction. Diverse CSP are efficiently cross-coupled with an array of α-trifluoromethyl alkenes under the catalysis of nickel with the assistance of zinc as the reducing agent. This method allows the use of unstrained CSP as the carbonyl-containing alkyl source in the allylic defluorinative reaction, to access a variety of gem-difluoroalkenes bearing a pendent ketone moiety with high functionality tolerance.

PURPOSE: We investigated the effects of retinal ischemia, neurodegeneration, and subclinical edema on best-corrected visual acuity (BCVA) in the early stages of diabetic retinopathy (DR). METHODS: Ischemia was evaluated by the microvascular parameters measured by optical coherence tomography angiography. Neurodegeneration and subclinical edema were identified by the intraretinal layer thickness obtained by optical coherence tomography. Eyes with nonproliferative diabetic retinopathy (n = 132) from 89 patients were analyzed. Eyes were classified as having normal BCVA (n = 88 [66.7%], Snellen equivalent ≥ 20/20) or decreased BCVA (n = 44 [33.3%], Snellen equivalent < 20/20). The prevalence of ischemia, neurodegeneration, and subclinical edema was explored in patients with and without decreased BCVA, and correlations between BCVA and these pathological pathways were determined. RESULTS: Vessel density in the deep retinal capillary plexus (DRCP) and thickness of ganglion cell layer plus inner plexiform layer (GCL-IPL) were significantly lower in eyes with decreased BCVA compared with eyes with normal BCVA (both P < 0.05). In the final multiple regression predictive model, age, DRCP vessel density, and GCL-IPL thickness (all P ≤ 0.044) were predictors of BCVA. DRCP vessel density and GCL-IPL thickness have an interactive effect on visual acuity. The proportions of ischemia and neurodegeneration were significantly higher in eyes with decreased BCVA than in eyes with normal BCVA (P = 0.001 and P = 0.004, respectively). CONCLUSION: During the natural course of the early stages of DR, ischemia and neurodegeneration were the main disease pathways associated with visual acuity, and the mechanisms varied among patients.

White spot syndrome virus (WSSV) is a fatal pathogen threatening global crustacean industry with no commercially available drugs to control WSSV. To address the urgent need for finding effective antiviral agents against WSSV, we examined the anti-WSSV activities of 11 common antiviral agents in crayfish Procambarus clarkia. The results showed that acyclovir displayed the highest inhibition on WSSV replication in vivo (92.59%, 50 mg/kg). Acyclovir repressed WSSV proliferation followed a dose-dependent fashion and pre- or post-treatment of acyclovir exerted strong inhibition on the viral loads. Further, we observed a markedly reduced expression levels of WSSV genes (immediate-early IE gene ie1, DNA polymerase gene DNApol and envelope protein gene Vp28) that are crucial in viral life cycle with the acyclovir treatment during the early infection. Meantime, we also found a significantly increased expressions of anti-oxidative as well as apoptosis related genes, suggesting that acyclovir could effectively suppress WSSV replication in vivo. Finally, acyclovir treatment could significantly improve the survival rate of WSSV-challenged crayfish by 56%. Taken together, acyclovir has the potential to be developed as a promising preventive or therapeutic agent against WSSV infection, and this finding may provide a reference for rapid discovery anti-WSSV agent in crustacean aquaculture.

Self-healing concrete designs can protect against deterioration and improve durability. However, there is no unified conclusion regarding the effective preparation and mechanical properties of self-healing concrete. In this paper, microcapsules are used in cement-based materials, the reasonable dosage of microcapsules is determined, and the self-healing performance of the microcapsule self-healing system under different curing agents is explored. The microcapsules and curing agent are shown to enhance the flexural and compressive strength of mortar specimens at relatively low contents. The optimal microcapsule content in terms of compressive strength is 1-3%. When the content of the microcapsule reaches 7%, the strength of the specimen decreases by approximately 30%. Sodium fluorosilicate is better-suited to the microcapsule self-healing cement-based system than the other two fluorosilicates, potassium fluorosilicate and magnesium, which have similarly poor healing performance as curing agents. Healing time also appears to significantly influence the microcapsule self-healing system; mortar specimens that healed for 28 days are significantly higher than those that healed for 7 days. This work may provide a valuable reference for the design and preparation of self-healing cementitious composite structures.

The amounts of the components in a microcapsule self-healing system significantly impact the basic performance and self-healing performance of concrete. In this paper, an orthogonal experimental design is used to investigate the healing performance of microcapsule self-healing concrete under different pre-damage loads. The strength recovery performance and sound speed recovery performance under extensive damage are analyzed. The optimum factor combination of the microcapsule self-healing concrete is obtained. Scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS) are carried out on the concrete samples before and after healing to determine the healing mechanism. The results show that the healing effect of self-healing concrete decreases with an increase in the pre-damage load, and the sound speed recovery rate increases with an increase in the damage degree. The influence of the sodium silicate content on the compressive strength and compressive strength recovery rate of the self-healing concrete increases, followed by a decrease. The optimum combination of factors of the microcapsule self-healing system is 3% microcapsules, 30% sodium silicate, and 15% sodium fluosilicate. The results can be used for the design and preparation of self-healing concrete.

Herein we report a chromium-catalyzed allylic defluorinative ketyl olefin coupling between aldehydes and α-trifluoromethyl alkenes, which provides novel and efficient access to diverse gem-difluorohomoallylic alcohols. Remarkably, the high chemoselectivity of this reaction enables the conversion of the formyl moiety in the presence of various easily reducible functionalities including ketone, organohalides, aziridine, sulfone, alkyne, and unactivated alkene. The utility of this method is demonstrated by various simple derivatizations of the attached hydroxyl group of the coupling products. The preliminary mechanistic investigations suggest a reaction pathway with a rate-limiting C-C forming step followed by facile ß-fluoro elimination.

Herein we report a nickel-catalyzed asymmetric reductive aryl-allylation of aryl iodide-tethered unactivated alkenes, wherein both acyclic allyl carbonates and cyclic vinyl ethylene carbonates can serve as the coupling partners. Furthermore, the direct use of allylic alcohols as the electrophilic allyl source in this reaction is also viable in the presence of BOC anhydride. Remarkably, this reaction proceeds with high linear/branched-, E/Z- and enantio-selectivity, allowing the synthesis of various chiral indanes and dihydrobenzofurans (50 examples) containing a homoallyl-substituted quaternary stereocenter with high optical purity (90-98% ee). In this reductive reaction, the use of pregenerated organometallics can be circumvented, giving this process good functionality tolerance and high step-economy.

BACKGROUND: The aim of this study is to evaluate the level and role of serum irisin in elderly patients with type 2 diabetes mellitus (T2DM) using case-control study. METHODS: A total of 71 patients with T2DM were selected as the case group according to the inclusion criteria and exclusion criteria; and the ratio of 1:1 was calculated according to the inclusion rate of the residents. The cohort established in Guankou Town, Jimei District, Xiamen City, Fujian Province, China and the residents of this cohort were selected at the age of 60 and above. A total of 71 healthy subjects were included as the control group with the same gender and the age with a difference of ± 5 years old. The clinical data of the subjects were collected to determine their previous history, blood pressure, body mass index (BMI), hemoglobin (HB), liver function test, renal function test, fasting blood glucose and serum lipid. The irisin level in serum was measured by enzyme-linked immunosorbent assay (ELISA). The data were analyzed by using SPSS17.0 software. Single factor analysis using Chi-square test or t-test was performed to compare the differences between T2DM patients with the control group of the general data, clinical indicators and irisin level in serum. Logistic regression was used to analyze the protective factors and risk factors of diabetes mellitus. RESULTS: The results of single factor analysis showed that the level of irisin in T2DM group was significantly lower than that in the control group (703.37 ± 241.51 ng/mL and 800.22 ± 275.59 ng/mL, respectively). The levels of BMI, systolic blood pressure (SBP), diastolic blood pressure (DBP), triglyceride (TG) and fasting plasma glucose (FPG) in T2DM group were higher than those in control group, and differences were statistically significant. Logistic regression analysis indicated that irisin may be a protective factor for type 2 diabetes (odds ratio (OR) = 0.997, 95% confidence interval (CI): 0.994 - 0.999). CONCLUSIONS: The serum irisin level in T2DM group was significantly lower than that in control group, suggesting that irisin may be a protective factor for type 2 diabetes.

Herein we successfully applied Katritzky salts as the alkyl electrophile in the reductive allylic defluorinative cross-coupling reaction with α-trifluoromethyl alkenes. The merger of C-F and C-N bond cleavage via the synergistic effect of nickel and zinc allows the efficient synthesis of diverse gem-difluoroalkenes with a good tolerance for various functional groups.

Herein, we report a method for efficient synthesis of gem-difluorobishomoallylic alcohols starting from trifluoromethyl-substituted alkenes and epoxides via a titanocene-catalyzed reductive domino reaction, which consists of a Ti(III)-mediated radical-type ring opening and the following allylic defluorinative cross-coupling reaction via sequential radical addition and ß-F elimination. Notably, complete regioselectivity and high tolerance of functionalities can be achieved in this reaction. Furthermore, diverse 6-fluoro-3,4-dihydro-2H-pyrans have been prepared through derivatization of the cross-coupling products in one single step.

Unactivated alkyl chlorides are abundant building blocks in organic synthesis, but they have been rarely engaged in cross-electrophile coupling. Herein, we report a Ni/Ti-cocatalyzed reductive allylic defluorinative cross-coupling between trifluoromethyl alkenes and unactivated alkyl chlorides and bromides, enabling the efficient preparation of diverse functional-group-rich gem-difluoroalkenes. Notably, synthesis of the gem-difluoroalkene analogues of azaperone, haloperidol, and benperidol was also accomplished using our method as a key step.

Hydrofunctionalization of trifluoromethyl alkenes is highly challenging, because the nucleophilic addition is oftentimes followed by ß-F elimination in this case. By the use of tetrabutylammonium decatungstate (TBADT) as a hydrogen-atom-transfer (HAT) photocatalyst for acyl C-H activation, we successfully avoid the ß-F elimination in the hydroacylation of trifluoromethyl alkenes with aldehydes. This light (390 nm) promoted reaction provides a facile and efficient access to various ß-CF3 ketones in complete regiocontrol with high functionality tolerance and 100% atom economy.
