ABSTRACT In this study, we investigated the chromosomes of three species of Sicarius spiders from the Brazilian Caatinga, using classical and molecular cytogenetic techniques. Based on the phylogenetic approach, we also discussed about the variation of diploid number, types of sex chromosome system and changes in the localization of ribosomal genes of Scytodoidea. Sicarius are Synspermiata spiders that together with the genera Loxosceles and Hexophthalma constitute the family Sicariidae. In this group, the available cytogenetic data showed a low diploid number range (2n♂=18 to 2n♂=23) and the presence of only multiple sex chromosome systems (X1X2Y and X1X20). Mitotic metaphase cells exhibited 2n♂=16+X1X2Y for Sicarius cariri and S. ornatus, and 2n♂=18+XY for S. tropicus. In these species, silver impregnation revealed nucleolar organizer region (Ag-NOR) on the terminal region of pair 1. In S. ornatus and S. tropicus, the results obtained with fluorescent in situ hybridization (FISH) using 18S rDNA probe were similar to Ag-NOR, however in S. cariri, the ribosomal sites were localized in the terminal region of the X1 sex chromosome. In this work, we presented the first description of a simple sex chromosome system for Sicariidae, helping to understand how the XY sex chromosome system evolved from the X1X2Y system. Additionally, FISH data incongruous with Ag-NOR indicate that the cytogenetic studies in Sicariidae allow investigating the relation between the karyotype evolution and the distribution and the activity of rDNA genes.


RESUMEN En este estudio, investigamos los cromosomas de tres especies de arañas Sicarius de la Caatinga brasileña, utilizando técnicas de citogenética clásica y molecular. Usando un enfoque filogenético, también discutimos la variación del número diploide, los tipos de sistema cromosómico sexual y los cambios en la localización de los genes ribosómicos en Scytodoidea. Los Sicarius son arañas Synspermiata que, junto con los géneros Loxosceles y Hexophthalma, constituyen a la familia Sicariidae. En este grupo, los datos citogenéticos disponibles mostraron un rango de número diploide bajo (2n♂=18 a 2n♂=23) y únicamente la presencia de sistemas de cromosomas sexuales múltiples (X1X2Y y X1X20). Las células mitóticas en metafase mostraron 2n♂=16+X1X2Y para Sicarius cariri y S. ornatus, y 2n♂=18+XY para S. tropicus. En estas especies, la impregnación de plata reveló la región organizadora nucleolar (Ag-NOR) en la región terminal del par 1. En S. ornatus y S. tropicus, los resultados obtenidos con la hibridación in situ fluorescente (FISH) utilizando la sonda de ADNr 18S fueron similares a los de Ag-NOR, sin embargo, en S. cariri los sitios ribosomales se localizaron en la región terminal del cromosoma sexual X1. En este trabajo, presentamos la primera descripción de un sistema cromosómico sexual simple para Sicariidae, ayudando a entender cómo el sistema cromosómico sexual XY evolucionó a partir del sistema X1X2Y. Además, los datos de FISH incongruentes con Ag-NOR indican que los estudios citogenéticos en Sicariidae permiten investigar la relación entre la evolución del cariotipo y la distribución y la actividad de los genes de ADNr.

Abstract Species of Trichogramma Westwood, 1833 (Hymenoptera: Trichogrammtidae) are frequently used as biological control agents against Lepidoptera, but practical application of these egg endoparasitoids are complicated because of their complex taxonomy. This study aimed to compare sequences of internal transcribed spacer regions of ribosomal DNA (ITS2-rDNA) of Trichogramma accessions with those deposited in GenBank in order to access the reliability of the ITS2 as a barcode for discriminating species and evaluating the genetic diversity. ITS2-rDNA sequences obtained from seventeen specimens of Trichogramma confirmed previous identifications based on morphological characteristics. Multiple sequence alignment revealed the existence of highly conserved regions in ITS2 sequences while the neighbour-joining dendrogram indicated that the specimens formed three clusters comprising T. manicobai and T. marandobai (group I), T. galloi (group II) and T. pretiosum (group III). The ITS2 marker was shown to be a powerful DNA barcode for discriminating Trichogramma species and could be used to complement the morphological approach.


Resumo Espécies de Trichogramma Westwood, 1833 (Hymenoptera: Trichogrammatidae) são freqüentemente usadas como agentes de controle biológico contra Lepidoptera, esses endoparasitóides de ovos apresentam taxonomia complexa, o que dificulta sua aplicação prática. Este estudo teve como objetivo comparar seqüências de regiões espaçadoras internas transcritas de DNA ribossômico (ITS2-rDNA) de acessos de Trichogramma com aquelas depositadas no GenBank, a fim de avaliar a confiabilidade do ITS2 barcode para discriminar espécies e avaliar a diversidade genética. As seqüências de ITS2-rDNA obtidas de dezessete espécimes de Trichogramma confirmaram identidades anteriores com base em características morfológicas. O alinhamento de múltiplas sequências revelou a existência de regiões altamente conservadas nas sequências ITS2, enquanto o dendrograma indicou que os espécimes formavam três grupos compreendendo T. manicobai e T. marandobai (grupo I), T. galloi (grupo II) e T. pretiosum (grupo III). O marcador ITS2 mostrou ser um poderoso DNA barcode para discriminar espécies de Trichogramma podendo ser usado como complemento da abordagem morfológica.

ABSTRACT

The objective of this study was to investigate the impact of chronic obstructive pulmonary disease (COPD)-heart failure (HF) coexistence on linear and nonlinear dynamics of heart rate variability (HRV). Forty-one patients (14 with COPD-HF and 27 HF) were enrolled and underwent pulmonary function and echocardiography evaluation to confirm the clinical diagnosis. Heart rate (HR) and R-R intervals (iRR) were collected during active postural maneuver (APM) [supine (10 min) to orthostasis (10 min)], respiratory sinus arrhythmia maneuver (RSA-M) (4 min), and analysis of frequency domain, time domain, and nonlinear HRV. We found expected autonomic response during orthostatic changes with reduction of mean iRR, root mean square of successive differences between heart beats (RMSSD), RR tri index, and high-frequency [HF (nu)] and an increased mean HR, low-frequency [LF (nu)], and LF/HF (nu) compared with supine only in HF patients (P<0.05). Patients with COPD-HF coexistence did not respond to postural change. In addition, in the orthostatic position, higher HF nu and lower LF nu and LF/HF (nu) were observed in COPD-HF compared with HF patients. HF patients showed an opposite response during RSA-M, with increased sympathetic modulation (LF nu) and reduced parasympathetic modulation (HF nu) (P<0.05) compared with COPD-HF patients. COPD-HF directly influenced cardiac autonomic modulation during active postural change and controlled breathing, demonstrating an autonomic imbalance during sympathetic and parasympathetic maneuvers compared with isolated HF.

ABSTRACT

@#We report on a 12 years old female patient who had been diagnosed with patellofemoral instability – recurrent dislocation and anterior knee pain. Radiologic evaluation further revealed osteochondritis dissecans (OCD) of the lateral femoral trochlea. She underwent surgical procedure with isolated medial patellofemoral ligament reconstruction, no surgical procedure was done to the OCD lesion. Postoperatively, there was clinical improvement of patellofemoral instability, with radiological evidence of healing of the OCD lesion. Isolated realignment procedures such as medial patellofemoral ligament reconstruction may be associated with spontaneous healing of osteochondritis dissecans of the lateral femoral trochlea.

ABSTRACT

Oral mucositis (OM) is a common and dose-limiting side effect of cancer treatment, including 5-fluorouracil (5-FU) and radiotherapy. The efficacy of the therapeutic measures to prevent OM is limited and disease prevention is not fully observable. Amifostine is a cytoprotective agent with a described anti-inflammatory potential. It is clinically used to reduce radiotherapy and chemotherapy-associated xerostomia. This study investigated the protective effect of amifostine on an experimental model of OM. Hamsters were divided into six groups: saline control group (5 mL/kg), mechanical trauma (scratches) of the right cheek pouch; 5-FU (60 and 40 mg/kg, ip, respectively, administered on days 1 and 2); amifostine (12.5, 25, or 50 mg/kg) + 5-FU + scratches. Salivation rate was assessed and the animals were euthanized on day 10 for the analysis of macroscopic and microscopic injury by scores. Tissue samples were harvested for the measurement of neutrophil infiltration and detection of inflammatory markers by ELISA and immunohistochemistry. 5-FU induced pronounced hyposalivation, which was prevented by amifostine (P<0.05). In addition, 5-FU injection caused pronounced tissue injury accompanied by increased neutrophil accumulation, tumor necrosis factor-alpha (TNF-α), and interleukin-1 beta (IL-1β) tissue levels, and positive immunostaining for TNF-α, IL-1β, and inducible nitric oxide synthase (iNOS). Interestingly, amifostine prevented the inflammatory reaction and consequently improved macroscopic and microscopic damage (P<0.05 vs 5-FU group). Amifostine reduced inflammation and protected against 5-FU-associated oral mucositis and hyposalivation.

ABSTRACT

O objetivo deste trabalho foi verificar os efeitos da solução poli-iônica intravenosa contendo 84mEq/L de lactato (L84) sobre os equilíbrios hidroeletrolítico e ácido-base de equinos, quando administrada de forma rápida ou lenta. Cinco equinos sadios adultos receberam a infusão contínua intravenosa da L84, em volume correspondente a 10% do peso corporal, em duas ocasiões: a) infusão rápida (16,66mL/kg/h) durante seis horas; b) infusão lenta (8,33mL/kg/h) durante 12 horas. Amostras de sangue venoso foram colhidas ao início da infusão (hora zero) e três, seis, nove, 12 e 24 horas após, e amostras de urina nas horas zero, seis, 12 e 24. Determinaram-se pH (sanguíneo e urinário), pCO2, HCO3 -, BE, PPT, lactato L, Na+, K+, Cl-, AG, SID, Atot, VVP, densidade urinária e excreções fracionadas urinárias de lactato L, Na+, K+ e Cl-. A L84 provoca efeito alcalinizante iatrogênico de menor magnitude quando administrada de forma lenta, porque os mecanismos renais, atuantes durante o período de infusão, promovem a correção gradativa do desequilíbrio. Pode-se concluir que a infusão de forma lenta da solução L84 em equinos é recomendável nos casos em que se suspeite de acidose metabólica e não seja possível quantificar o grau do desequilíbrio.(AU)


The aim of this study was to investigate the effects of an intravenous polyionic solution containing 84mEq/L of lactate (L84) on the hydroelectrolyte and acid-base balances when administered quickly or slowly in horses. Five healthy adult horses received the L84 solution, in a volume corresponding to 10% of BW, by continuous intravenous infusion, in two instants: a) rapid infusion (16.66mL/kg/h) during 6 hours; b) slow infusion (8.33mL/kg/h) during 12 hours. Venous blood samples were taken at the beginning of the infusion (hour 0) and 3, 6, 9, 12, and 24 hours after. Urine samples were taken at 0, 6, 12, and 24h. pH (blood and urine), pCO2, HCO3-, BE, TPP, L-lactate, Na+, K+, Cl-, AG, SID, Atot, PVV, urine specific gravity, and L-lactate, Na+, K+, and Cl- renal fractional clearance were determined. The L84 solution causes lower magnitude alkalizing effect when administered slowly, due to the gradual correction of the iatrogenic imbalance by the kidneys during the infusion period. The L84 solution infused at a low rate in horses could be recommended in cases where metabolic acidosis is suspected and it is not possible to quantify the imbalance degree.(AU)

Early dietary treatment of phenylketonuria (PKU), an inborn error of phenylalanine (Phe) metabolism, results in normal cognitive development. Although health-related quality of life (HRQoL) of PKU patients has been reported as unaffected in high-income countries, there are scarce data concerning HRQoL and adherence to treatment of PKU children and adolescents from Brazil. The present study compared HRQoL scores in core dimensions of Brazilian early-treated PKU pediatric patients with those of a reference population, and explored possible relationships between adherence to treatment and HRQoL. Early-treated PKU pediatric patient HRQoL was evaluated by self- and parent-proxy reports of the Pediatric Quality of Life Inventory (PedsQL) core scales. Adherence to treatment was evaluated by median Phe levels and percentage of results within the therapeutic target range in two periods. Means for total and core scales scores of PedsQL self- and parent proxy-reports of PKU patients were significantly lower than their respective means for controls. Adequacy of median Phe concentrations and the mean percentage of values in the target range fell substantially from the first year of life to the last year of this study. There was no significant difference in mean total and core scale scores for self- and parent proxy-reports between patients with adequate and those with inadequate median Phe concentrations. The harmful consequences for intellectual capacity caused by poor adherence to dietary treatment could explain the observed decrease in all HRQoL scales, especially in school functioning. Healthcare system and financial difficulties may also have influenced negatively all HRQoL dimensions.

Aparelhos de automação, cujo uso está estabelecido na hematologia dos animais domésticos, ainda não são empregados rotineiramente em felídeos selvagens. Esse estudo objetivou avaliar a técnica automatizada na contagem de células sanguíneas de jaguatiricas (Leopardus pardalis), comparando-a com a técnica manual. Foram coletadas amostras sanguíneas de oito jaguatiricas, que se submeteram à contagem de eritrócitos, leucócitos, plaquetas e à determinação do volume globular pelo método manual padrão e por meio do aparelho BC - 2800 VET® com a configuração para gatos domésticos. Os resultados foram avaliados por meio do teste t para dados pareados, e as técnicas submetidas à análise de correlação de Pearson. A técnica automatizada demonstrou resultados estatisticamente semelhantes para eritrócitos e leucócitos. Apenas para volume globular houve diferença significativa entre as técnicas manual e automatizada, mas a correlação foi alta. Apesar de não haver diferença significativa entre as técnicas para plaquetas, a correlação foi baixa. Conclui-se que o aparelho BC - 2800 VET® com a configuração para gatos domésticos é uma técnica confiável na realização do eritrograma e do leucograma para jaguatiricas. Para a determinação do parâmetro volume globular, o aparelho pode ser utilizado, desde que se faça a correção. Para a contagem de plaquetas, a técnica manual é recomendada.(AU)


Automated equipment, whose use is established in hematology of domestic animals, is not yet routinely used in blood cells count of wildlife, due to lack of studies that validate its use. The purpose of this study was to evaluate the automated technique for blood cells count of ocelots (Leopardus pardalis), comparing it with the manual technique. Blood samples were collected from eight ocelots, which were submitted to counting of erythrocytes, leukocytes, platelets and packed cell volume by the standard manual method and by the device VET ® 2800 BC with the configuration for domestic cats. The results were evaluated using the t test for paired data and the techniques submitted to Pearson correlation. The automated technique showed statistically similar results to erythrocytes and leukocytes. Significant difference was found only for packed cell volume between the manual and automated techniques, but the correlation was high. Although there was no significant difference between the techniques for platelets, the correlation was low. We concluded that the 2800 BC VET ® device with the configuration for domestic cats is a reliable technique in performing the erythrocyte and leukocyte counts for ocelots. The device may be used to determine packed cell volume, provided the correction is made. The manual technique is recommended for the platelet count.(AU)

ABSTRACT

The aim of this study was to evaluate the use of polyamide 12 intramedullary rods in osteotomized humerus in cockerels (White Plymouth Rock) and analyze, by radiography and histopathology, bone consolidation and the formation of bone callus. Ten cockerels were subjected to transverse osteotomy of the right humeral diaphysis followed by the insertion of polyamide 12 locked rods, with two nails in each bone fragment. Radiographies obtained at the immediate pre and post-operative period, and every 7 days for 3 months revealed no perioperatory complications. Radiography revealed a radiopaque bone callus between the 4th and 5th week post-surgery. Histopathology detected an amphophylic material (polyamide 12 rod) in the intramedullary region and the proliferation of highly vascularized connective tissue between the rod and the cortical bone. This connective tissue was highly cellular with differentiation into osteoblasts. The collagen fibers varied from loose to dense and a differentiated bone matrix, containing osteocytes in gaps and the development of bone marrow, was also observed; indicating the formation of a bone callus without signs of implant rejection. The polyamide 12 intramedullary rod was effective in the stabilization of the fractures used in this experimental model, with no rejection reaction for at least 90 days.(AU)


O presente estudo teve como objetivo avaliar a implantação de hastes intramedulares de poliamida 12, em úmeros osteotomizados de galos (Plymouth Rock Branca), bem como avaliar a consolidação óssea, por meio da verificação da formação de calo ósseo, analisando exames radiográficos e histopatológicos. Os galos foram submetidos à osteotomia transversa da diáfise do úmero direito, sendo implantadas posteriormente as hastes de poliamida 12 bloqueada com dois parafusos em cada fragmento ósseo. Após o procedimento cirúrgico, foram realizadas radiografias nos momentos pré e pós-operatório imediato e, posteriormente, a cada sete dias, durante três meses, até a realização da eutanásia. Não houve complicações perioperatórias. Ao exame radiográfico, os animais apresentaram formação de calo ósseo radiopaco entre a quarta e quinta semanas de pós-operatório. Ao exame histopatológico, foi possível notar, em todos os animais, presença de material anfofílico (haste de poliamida 12) ocupando a região intramedular óssea, e entre a haste e as corticais ósseas, proliferação de tecido conjuntivo altamente vascularizado. Tal tecido apresentou-se altamente celular com diferenciação em osteoblastos. Notaram-se também fibras colágenas, variando de aspecto frouxo a denso, e observou-se formação de matriz óssea diferenciada com osteócitos em lacunas e formações de medula óssea, denotando formação de calo ósseo sem sinais de rejeição do implante. A haste intramedular de poliamida 12 foi eficaz para estabilização das fraturas utilizando-se esse modelo experimental, sem reação de rejeição pelo período de 90 dias.(AU)

ABSTRACT

The cryosurgery is a very useful therapy for the treatment of a variety of neoplastic and non-neoplastic processes. Nevertheless, it is still poorly described as an option for the treatment of specific cutaneous neoplasms, such as squamous cell carcinoma. The purpose of the present study was to analyze the clinical response of cryosurgery for the treatment of squamous cell carcinoma in cats. For this 13 squamous cell carcinoma lesions were selected in 11 cats, diagnosed through citopathological and/or histopathological examinations. The lesions were frozen using liquid nitrogen spray, and the evaluations were performed in the moment of freeze and approximately every 15 days until the wound was completely healed. The response of cryosurgery was considered complete with tumoral remission on 38.5% of the cats, and partial on 46.1%. The main complications included crusting and nostril stenosis. The presented results suggested that cryosurgery is effective and may be a viable option for the treatment of squamous cell carcinoma in cats. The effectiveness of the therapy; however, depends on the correct selection of the candidates for cryosurgery based on the lesion size, and the attendance to some criteria, such as the freezing time and post-operative care.(AU)


A criocirurgia tem se mostrado uma terapia de grande auxílio no tratamento de uma variedade de processos neoplásicos e não neoplásicos. Apesar disso, ainda é pouco descrita a utilização da criocirurgia como opção de tratamento de neoplasias cutâneas específicas, como o carcinoma de células escamosas. Objetivou-se com o presente estudo analisar a resposta clínica da criocirurgia para o tratamento do carcinoma de células escamosas em gatos. Para isso, foram selecionadas 13 lesões do tipo carcinoma de células escamosas em 11 gatos, diagnosticadas por meio de análise citopatológica e/ou histopatológica. As lesões foram congeladas por meio de nitrogênio líquido na forma de spray, e as avaliações realizadas no momento do congelamento e aproximadamente a cada 15 dias, até a completa cicatrização da ferida. A resposta da criocirurgia foi considerada completa, com remissão tumoral em 38,5% dos gatos e parcial em 46,1% destes. As principais complicações verificadas incluíram a formação de crostas e estenose de narina. Os resultados apresentados no presente estudo sugerem que a criocirurgia é efetiva e pode ser uma opção viável para o tratamento do carcinoma de células escamosas em gatos. O sucesso do tratamento, entretanto, depende da correta seleção dos candidatos à técnica quanto ao tamanho da lesão, e do atendimento a alguns critérios, como o tempo de congelamento e os cuidados pós-operatórios.(AU)

ABSTRACT

La duplicación del sistema colector renal es la variación más frecuente del sistema urinário y se puede presentar como duplicación completa o incompleta, así como la inserción ectópica en todas las partes del sistema urinario a partir de la parte distal hacia la vejiga. Este artículo presenta uma duplicación completa no ectópica del ureter en el riñón izquierdo de un cadáver de un individuo brasileño, de sexo masculino. Ambos uréteres tenían origen en el hilio renal y continuaban hasta la vejiga urinaria separadamente, desembocando en ostios diferentes, en el área del trígono vesical, donde el ostio perteneciente al uréter que drenaba el polo superior presentó posición distal y lateral en relación al ostio del uréter que drenaba el polo inferior. Esta disposición es uma excepción a la regla de Weigert-Meyer, que indica que el uréter del polo superior, por el hecho de permanecer fijo por más tiempo al conducto mesonéfrico, presenta mayor migración, terminando medial e inferiormente al uréter que drena el polo inferior en 97 % de los casos.


Duplication of the urinary tract is the most frequent variation of this system and may present as complete or incomplete duplication, as well as ectopic insertion throughout all parts of the urinary system from distal to the bladder. This article presents a complete non-ectopic duplication of the ureter in the left kidney of a cadaver of one Brazilian individual of male sex. In this study, both ureters originated in the renal hilum and continued to the urinary bladder separately, opening into different ostia, in the area of the bladder trigone, where the ostium belonging to the ureter that draining the upper pole presented a distal and lateral position in relation to the ostium of the ureter draining the lower pole. This arrangement is an exception to the Weigert-Meyer rule, which indicates that the ureter of the upper pole, due to its longer fixation to the mesonephric duct, presents a greater migration, ending medial and inferior to the ureter draining the inferior pole in 97 % of cases.

ABSTRACT

Objetivou-se avaliar os efeitos do extrato de própolis (LLOSC2) sobre o comportamento ingestivo e os parâmetros sanguíneos em cordeiros alimentados com dieta concentrada. Foram utilizados quatro cordeiros machos, castrados, sem raça definida e com peso médio inicial de 32±1,92kg, mantidos em gaiolas metabólicas individuais durante todo o período experimental. O delineamento experimental foi em quadrado latino 4x4, com quatro animais, quatro dietas e quatro períodos experimentais. As dietas diferiram em relação à adição ou não de extratos de própolis à ração. As dietas testadas diferiam na adição de zero (controle), 1(88,16mg/g de flavonoides), duas (176,32mg/g de flavonoides) ou três (264,48mg/g de flavonoides) doses de aditivo à base de extrato de própolis LLOSC2. A dieta tinha relação volumoso:concentrado de 40:60 e foi formulada para ganhos de 250g. A ração concentrada era composta de milho e farelo de soja. O comportamento ingestivo dos animais foi observado por 24 horas, divididas em quatro períodos de seis horas. As coletas de sangue eram realizadas no último dia do período. Não houve efeito das doses LLOSC2 sobre o tempo despendido em alimentação, ruminação, ócio e ingestão de água. A duração do ciclo ruminativo foi maior nos animais que receberam duas doses de LLOSC2, e o número de movimentos mastigatórios também tendeu a aumentar no período que compreendia das 13 às 19h. Já a dieta contendo uma dose de LLOSC2 aumentou a duração da ruminação do bolo bem como o seu número total de mastigações no período de uma a sete horas. Houve uma diminuição nos níveis séricos de cortisol com a adição de três doses de LLOSC2. O hemograma, o leucograma, o perfil bioquímico e as imunoglobulinas não foram afetados pela inclusão das doses de LLSOC2. A administração de extrato de própolis LLOSC2 à dieta de ovinos não afetou seu comportamento ingestivo, os parâmetros hematológicos, bioquímicos e imunológicos estudados.(AU)


The objective of this study was to evaluate the effects of propolis extract (LLOS 2) on feeding behavior and blood parameters in lambs fed concentrate diet. Four male lambs were castrated, mongrel and average weight of 32±1.92kg, kept in individual metabolic cages throughout the trial period. The experimental design was a Latin square 4 x 4 with four animals, four treatments and four experimental periods. The diets differed according to addition or not of propolis extracts to feed. The experimental diets differed in addition to zero (control), 1 (88.16mg/g flavonoids) 2 (176.32mg/g flavonoids) or 3 (264.48mg/g flavonoids) Additive doses based on LLOSC2 propolis extract. The diet had a forage:concentrate ratio of 40:60 and was formulated to 250g gains. The concentrate ration was composed based on corn and soybean meal. The feeding behavior of the animals were observed for 24 hours, divided into four periods of 6 hours. Blood collections were made on the last day of the period. There was no effect of LLOSC2 doses on time spent eating, ruminating, leisure and water intake. The duration of the ruminative cycle was higher in animals that received two doses of LLOSC2 and the number of chewing movements also tended to increase in the period comprised from 13 to 19h. Since the diet containing a dose of LLOSC2, increased duration of rumination cake as well as its total number of chewing in the period of 1 to 7 hours. There was a decrease in serum cortisol with the addition of three doses of LLOSC2. The blood count, white blood cell count, biochemical profile and immunoglobulins were not affected by the inclusion of doses of LLSOC2. Administration of LLOSC2 propolis extract the sheep diet did not affect their feeding behavior, hematological, biochemical and immunological studies.(AU)

ABSTRACT

The aim of this study was to evaluate the effect of two different extenders (Skimmed Milk Glucose - SMG or Lactose - Egg Yolk - LEY) on physical characteristics and fertility of fractionated donkey semen cooled at 5°C. For this, four Pêga donkeys were used as semen donors. The sperm rich fraction of the ejaculate was diluted preparing insemination doses containing 400 x 106 motile spermatozoa in a volume of 22 mL, cooled to 5°C and stored up to 48 hours in a container proposed by Palhares (1997). Sperm motility and vigor were assessed in fresh semen, after first semen dilution, before insemination, at 24 and 48 hours after storage. For the fertility evaluation, 44 mares were inseminated with semen stored for a period between 12 and 24 hours. The mares were inseminated on fixed days (Mondays, Wednesdays, and Fridays) after the detection of a follicle greater than a 30mm diameter in one of the ovaries through ovulation. Pregnancy diagnosis was performed on day 12 post-ovulation, using transrectal ultrasonography. Semen diluted in SMG showed superior sperm motility than LEY, at the Pre-AI evaluation (P<0.05). At 48 hours of storage, all donkeys had motility values between 45 and 53% for semen diluted in SMG, while only one donkey showed motility greater than 30% in the LEY treatment. The pregnancy rate/cycle for mares inseminated with semen diluted in SMG was superior than that obtained using LEY (56.52% vs 4.76%, respectively).(AU)


Objetivou-se com o presente experimento avaliar o efeito de dois diferentes diluidores (leite em pó desnatado glicose - SMG ou lactose gema de ovo - LEY) sobre as características físicas e a fertilidade do sêmen asinino coletado de forma fracionada e resfriado a 5ºC. Para isso, quatro jumentos da raça Pêga foram utilizados como doadores de sêmen. A fração espermática rica do ejaculado foi diluída preparando-se doses inseminantes contendo 400 x 106 espermatozoides móveis em um volume de 22 mL, resfriadas a 5ºC e armazenadas por até 48 horas em contêiner proposto por Palhares (1997). A motilidade e o vigor espermáticos foram avaliados no sêmen fresco, após a pré-diluição, antes das inseminações, às 24 e 48 horas de armazenamento. Para avaliação de fertilidade, 44 éguas foram inseminadas com sêmen armazenado por um período entre 12 e 24 horas, em dias fixos (segundas, quartas e sextas-feiras), após a detecção de um folículo de diâmetro maior ou igual a 30mm em um dos ovários, até a ovulação. O diagnóstico de gestação foi realizado a partir de 12 dias após a ovulação, por meio de ultrassonografia transretal. O sêmen diluído em SMG apresentou motilidade espermática superior à do LEY, já a partir do tempo pré-IA. Às 48 horas de armazenamento, todos os jumentos apresentaram valores de motilidade entre 45% e 53%, quando o sêmen foi diluído em SMG, enquanto apenas um jumento apresentou motilidade superior a 30% no tratamento utilizando LEY. A taxa de concepção/ciclo das éguas inseminadas também foi superior para o sêmen diluído em SMG em relação ao diluído em LEY (56,52% versus 4,76%, respectivamente).(AU)

Abstract Species of Trichogramma Westwood, 1833 (Hymenoptera: Trichogrammtidae) are frequently used as biological control agents against Lepidoptera, but practical application of these egg endoparasitoids are complicated because of their complex taxonomy. This study aimed to compare sequences of internal transcribed spacer regions of ribosomal DNA (ITS2-rDNA) of Trichogramma accessions with those deposited in GenBank in order to access the reliability of the ITS2 as a barcode for discriminating species and evaluating the genetic diversity. ITS2-rDNA sequences obtained from seventeen specimens of Trichogramma confirmed previous identifications based on morphological characteristics. Multiple sequence alignment revealed the existence of highly conserved regions in ITS2 sequences while the neighbour-joining dendrogram indicated that the specimens formed three clusters comprising T. manicobai and T. marandobai (group I), T. galloi (group II) and T. pretiosum (group III). The ITS2 marker was shown to be a powerful DNA barcode for discriminating Trichogramma species and could be used to complement the morphological approach.


Resumo Espécies de Trichogramma Westwood, 1833 (Hymenoptera: Trichogrammatidae) são freqüentemente usadas como agentes de controle biológico contra Lepidoptera, esses endoparasitóides de ovos apresentam taxonomia complexa, o que dificulta sua aplicação prática. Este estudo teve como objetivo comparar seqüências de regiões espaçadoras internas transcritas de DNA ribossômico (ITS2-rDNA) de acessos de Trichogramma com aquelas depositadas no GenBank, a fim de avaliar a confiabilidade do ITS2 barcode para discriminar espécies e avaliar a diversidade genética. As seqüências de ITS2-rDNA obtidas de dezessete espécimes de Trichogramma confirmaram identidades anteriores com base em características morfológicas. O alinhamento de múltiplas sequências revelou a existência de regiões altamente conservadas nas sequências ITS2, enquanto o dendrograma indicou que os espécimes formavam três grupos compreendendo T. manicobai e T. marandobai (grupo I), T. galloi (grupo II) e T. pretiosum (grupo III). O marcador ITS2 mostrou ser um poderoso DNA barcode para discriminar espécies de Trichogramma podendo ser usado como complemento da abordagem morfológica.

ABSTRACT

Until recently, it was believed that only two lymnaeid species (i.e. Galba viatrix and Pseudosuccinea columella) occurred in Uruguay. However, based on a molecular approach, an additional species Galba cubensis, was recently discovered. The aim of this study was to molecularly characterize different lymnaeid populations from the northern region of Uruguay. The lymnaeids collections were carried out in two farms of the departments of Paysandú and Tacuarembó. The collected lymnaeids were divided in two fractions, one fraction was used for conchological analyses and detection of trematode larval stages, while the other fraction was used for molecular studies. Three PCRs targeting the 16S, ITS-2 and COI DNA regions were performed and the amplicons obtained were direct sequenced. The sequences were used for homology search and construction of phylogenetic trees by the maximum-likelihood method. The sequencing results revealed that both isolates corresponded to Galba neotropica. The phylogenetic analyses placed our isolates among the G. neotropica monophyletic group, closely related to other isolates of this species found in several South American countries. To our knowledge, this is the first record of G. neotropica in Uruguay and the confirmation as competent intermediate host of Fasciola hepatica. Further studies are needed to define the distribution and the role of each lymnaeid species in the transmission of F. hepatica in Uruguay.


Tradicionalmente se indicaba que existían dos especies de limneidos en Uruguay: Galba viatrix y Pseudosuccinea columella. Sin embargo, en los últimos años se identificó por medio de técnicas moleculares una tercera especie, Galba cubensis. El objetivo de los autores fue muestrear e identificar por medios moleculares poblaciones de limneidos del norte del país. Las colectas fueron realizadas en establecimientos rurales de los departamentos de Tacuarembó y Paysandú. Los caracoles colectados fueron divididos en dos fracciones, una de ellas fue destinada para el estudio morfológico de las conchillas y búsqueda de larvas de trematodos. La otra fracción se usó para la caracterización molecular. Tres genes fueron amplificados (ITS2, COI y 16S) utilizando protocolos de PCRs previamente descriptos. Las secuencias obtenidas se utilizaron para estudios de homología y construcción de árboles filogenéticos por medio del método de máxima verosimilitud. Por medio de la secuenciación se pudo confirmar que los dos aislamientos corresponden a Galba neotropica. Los estudios filogenéticos colocan ambos aislamientos dentro del grupo monofilético de G. neotropica junto a otros encontrados en distintas regiones de Sudamérica. Hasta lo que sabemos, el presente, es el primer registro de G. neotropica en Uruguay, además de comprobarse su capacidad para actuar como hospedero intermediario de Fasciola hepatica en condiciones de campo. Se sugieren futuros estudios para determinar la distribución y el rol de cada especie de limneido en la transmisión de F. hepatica.

ABSTRACT

Papain is a proteolytic enzyme removed from the leaves of green papaya and/or latex. This enzyme is widely known as a medicinal fruit used extensively in human medicine for the treatment of wounds of various etiologies. However, studies and reports in veterinary medicine are scarce. Another herbal drug widely used in wound healing is Sunflower oil (Helianthus annus). It has inflammatory and antimicrobial properties which stimulate the local neovascularization, promoting tissue granulation, cell migration, fibroblast proliferation, and differentiation. Three dogs were treated with infected necrosis wounds, considered large, extent, and severe, with varied etiology. All cases were treated with papain gel with the exception of one dog, which was given the sunflower oil at the end of the treatment. Papain gel shows effectiveness in the treatment of wounds especially with wound debridement and removal of necrotic tissue. In addition, the healing time was shorter when compared to the treatment with sunflower oil. Finally, the herbal drugs have a low cost and high accessibility. This study contributes to create a new research regarding the use of this drug in animal wound healing.(AU)


A papaína é uma enzima proteolítica retirada do mamão-papaia verde e/ou do látex das folhas do mamoeiro, tendo propriedades medicinais conhecidas na medicina humana para tratamento de feridas de diversas etiologias. Estudos e/ou relatos na medicina veterinária são escassos. Nesse sentido, objetivou-se descrever três casos de cães, que foram atendidos no Hospital Veterinário da Universidade Federal de Lavras, Brasil, todos com feridas infectadas, necrosadas, com etiologia variada e consideradas de grande extensão e gravidade, sendo tratados com gel de papaína. O tratamento demonstrou efetividade na recuperação das feridas, com a formação de grande quantidade de tecido de granulação, em um período menor que os tratamentos convencionais. Por se tratar de um fitoterápico, de baixo custo e alta acessibilidade, o gel de papaína pode ser utilizado no tratamento de feridas de grande extensão em cães e, assim como em humanos, parece ter um efeito benéfico no processo de cicatrização. Pesquisas devem ser conduzidas para elucidar a ação desse produto nos tecidos, bem como as diferentes concentrações a serem administradas.(AU)

The aim of this study was to evaluate the physical and morphological characteristics of the sperm-rich fraction of jackass semen. To this end, 130 ejaculates from five Pêga jackasses were collected using an open model artificial vagina. The sperm-rich fraction was collected using the split-ejaculate method and assessed for the number of mounts/ejaculate, for physical and morphological characteristics of the semen and number of doses produced/ejaculate. It was observed that all characteristics evaluated differed among the five jackasses, except for the head defect rates. The mean values obtained for the jackass sperm-rich fraction collected were: number of mounts/ejaculate - 1.27; semen volume - 20.21mL; motility - 84.53%; vigor - 4.46; motility after dilution - 80.10%; sperm concentration/mL - 894.38 x 10 6; total sperm/ejaculate - 16.14 x 10 9; number of insemination doses/ejaculate (400x10 6 motile sptz) - 33.39; number of insemination doses/ejaculate (800 x 10 6 motile sptz) - 16.69; and percentage of normal sperm - 90.46%. Thus, in the present experiment the split-ejaculate method using an open artificial vagina worked well with the jackasses, and the sperm-rich fraction of the ejaculate of Pêga jackasses had high quality and sperm concentration, allowing its use for semen processing without reducing the number of insemination doses produced per ejaculate.(AU)


Foram coletados 130 ejaculados de cinco jumentos da raça Pêga, utilizando-se vagina artificial modelo aberta, com o objetivo de se avaliar as características físicas e morfológicas da fração rica em espermatozoides do ejaculado de asininos. Para tal, a fração rica em espermatozoides, composta pelos três primeiros jatos ejaculados, foi coletada e avaliada quanto ao número de montas/ejaculado, quanto às características físicas e morfológicas do sêmen, bem como quanto ao número de doses inseminantes produzidas/ejaculado. Observou-se que todas as características avaliadas diferiram entre os cinco reprodutores avaliados, com exceção do percentual de defeitos de cabeça. Os valores médios obtidos da coleta da fração rica do ejaculado de jumentos foram: número de montas/ejaculado - 1,27; volume de sêmen - 20,21mL; motilidade - 84,53%; vigor - 4,46; motilidade pós-diluição - 80,10%; espermatozoides/mL - 894,38 x 10 6; espermatozoides/ejaculado - 16,14 x 10 9; número de doses inseminantes/ejaculado (400x10 6 sptz móveis) - 33,39; doses inseminantes/ejaculado (800 x 10 6 sptz/móveis) - 16,69; e percentual de espermatozoides normais - 90,46%. Assim, no presente experimento, observou-se boa aceitação dos reprodutores à coleta fracionada utilizando-se a vagina artificial aberta, sendo a fração rica do ejaculado de jumentos da raça Pêga caracterizada por alta qualidade e concentração espermática, o que viabilizou sua utilização para o processamento do sêmen, sem prejuízos quanto ao número de doses inseminantes produzidas/ejaculado.(AU)

The first three jets of the sperm-rich fraction of Pêga jackasses were collected and assessed separately. Five fertile Pêga jackasses were used as semen donors and underwent fractionated semen collection, using an open model artificial vagina. The first three jets of the semen were collected separately and assessed for volume, sperm motility, vigor, concentration/mL of semen, and sperm morphology. These characteristics were compared between first, second and third jets and between jackasses. It was observed that the jet volume differed (P<0.05) between jackasses, although it was similar (P>0.05) between first, second and third jets. Sperm motility did not differ (P>0.05) between jets and jackasses. Vigor was similar (P>0.05) between jets of the same jackass, and only the first jet differed (P<0.05) between jackasses. The first, second and third jets of the sperm-rich fraction had decreased sperm concentrations (P<0.05) of 955.56, 725.56 and 280.56x 106 sperm/mL of semen, respectively. Sperm morphology differed between the first three jets only for the incidence of mid-piece defect, higher in the third one (4.26%), compared to the first (3.36%) and second (3.38%) ones. When comparing the morphological characteristics of the sperm-rich fraction between five jackasses, regardless of the jet, there were differences in the percentage of normal sperm, proximal cytoplasmic droplet, mid-piece and head defects.(AU)


Objetivou-se, no presente experimento, caracterizar os três primeiros jatos da fração rica do ejaculado de jumentos da raça Pêga. Para tal, cinco jumentos foram submetidos à coleta fracionada do sêmen, utilizando-se vagina artificial modelo aberta. Os três primeiros jatos da fração rica do ejaculado foram coletados separadamente e avaliados quanto ao volume, à motilidade e ao vigor espermáticos, à concentração espermática/mL de sêmen e à morfologia espermática. Comparações foram realizadas entre jatos e entre jumentos. Observou-se que o volume do jato diferiu (P<0,05) entre os jumentos, embora fosse similar (P>0,05) entre os jatos. A motilidade espermática não diferiu (P>0,05) entre jatos nem entre jumentos. O vigor espermático foi similar (P>0,05) entre os jatos de um mesmo jumento, e apenas o vigor do jato 1 diferiu (P<0,05) entre os jumentos. Independentemente do jumento, a fração rica do ejaculado foi composta por três jatos apresentando concentrações espermáticas decrescentes (P<0,05), com 955,56; 725,56 e 280,56 x 106 espermatozoides/mL de sêmen. A morfologia espermática diferiu entre os três jatos avaliados apenas para a incidência de defeitos de peça intermediária, sendo maior no jato três (4,26%), em relação aos jatos um (3,36%) e dois (3,38%). Comparando-se as características morfológicas do sêmen entre os cinco jumentos avaliados, independentemente do jato, observaram-se diferenças entre os reprodutores quanto ao percentual de espermatozoides normais, com gota citoplasmática proximal, com defeitos de peça intermediária e de cabeça.(AU)

Semen from the first 15mL of the ejaculate (P1) obtained from two boars (30mL) was diluted in glycine-egg yolk extender, cooled at 5°C in a special container and rediluted in standard doses of 3x109 mobile spermatozoa after 12h of storage. Semen was also stored up to 24h after redilution. The physical characteristics of the semen were evaluated at different storage periods (fresh, 0h, 12h, rediluted, 24h, and 36h). The reproductive performance of the boars and their fertility regarding the insemination of primiparous sows were also determined. Two treatments were used: T1-15B sows inseminated with semen originated from hyperconcentrated heterospermic doses (15x109 mobile spermatozoa per dose), rediluted after 12h of storage at 5°C for standard doses of 3x109 mobile spermatozoa per dose and stored at 5°C up to 24h after redilution (n=10); T2-3B sows inseminated with standard heterospermic doses (3x109 mobile spermatozoa per dose), stored at 5°C up to 36h after semen collection (n=10). There was no effect (P>0.05) of treatments on the spermatic motility, even though a pronounced decrease (P>0.05) of their values at 12h of storage was recorded. However, they remained higher than 70% until 36h. There was effect of treatments on spermatic vigour at 0h (P<0.05), when T1-15B vigour was higher. There was also effect of the storage period for both treatments with a progressive decrease throughout 36h of storage, although the differences were not always significant. Pregnancy rates (90%) and the number of total farrowed piglets (15, 11-T1-15B; 13, 44- T2-3B) did not differ (P>0.05) between the treatments. It was concluded that the semen hyperconcentration of 15 billion of mobile spermatozoa per dose, stored at 5°C for 12h, did not result in drawbacks considering the physical characteristics of the semen, maintaining the pregnancy rates and prolificacy of the inseminated sows.


Os primeiros 15mL do ejaculado (P1) de dois varrões foram coletados (30mL) e diluídos em diluidor glicina-gema de ovo, resfriados a 5°C em contêiner especial e rediluídos para doses padrão de 3x109 espermatozoides (sptz) móveis, após 12 horas de armazenamento. Além disso, foram armazenados por até 24 horas após a rediluição, sendo as características físicas avaliadas em diferentes períodos de estocagem (fresco, zero hora, 12h, Red12h, 24h e 36h) e a fertilidade avaliada por meio de fêmeas primíparas inseminadas. Foram realizados dois tratamentos: T1-15B: porcas inseminadas com sêmen de doses heterospérmicas hiperconcentradas (15x109 sptz móveis/dose), rediluídas após 12 horas de armazenamento a 5°C para doses padrão de 3x109 sptz móveis/dose, e armazenadas a 5°C por até 24 horas após a rediluição (n=10); T2-3B: porcas inseminadas com doses heterospérmicas padrão (3x109 sptz móveis/dose), armazenadas a 5°C por até 36 horas após coleta. Não houve efeito (P>0.05) dos tratamentos sobre a motilidade espermática e, embora tenha ocorrido queda (P<0.05) às 12 horas, a motilidade foi superior a 70% durante as 36 horas de armazenamento. Houve efeito (P<0.05) dos tratamentos no tempo zero hora quanto ao vigor espermático, sendo E1T1-15B superior. Além disso, houve efeito do período de estocagem para os dois tratamentos, com queda progressiva do vigor ao longo das 36 horas, embora nem sempre as diferenças tenham sido significativas. As taxas de gestação (90%) e o número total de leitões nascidos (15, 11 - T1-15B; 13, 44 - T2-3B) não diferiram (P>0.05) entre os tratamentos. Concluiu-se que a hiperconcentração do sêmen para 15x109 sptz móveis/dose, armazenado a 5°C por 12 horas não resultou em prejuízos quanto à manutenção das características físicas do sêmen e ao desempenho reprodutivo dos varrões, sendo capaz de manter a taxa de gestação e a prolificidade das fêmeas inseminadas.
