ABSTRACT

Objective The purpose of this work was to compare the efficacy of narrow band imaging-assisted transurethral resection of bladder tumour (TURB) with conventional white light imagingassisted TURB in primary non-muscle-invasive bladder cancer.Methods Several databases were searched,including Cochrance Libarary,PubMed,Embase,Wanfang,VIP,CNKI and CBM.The endpoints were biochemical failure in 3 months,1 year and 2 years.We performed a meta-analysis of the published data.The results are expressed as risk ratio (RR),with the corresponding 95% confidence interval (CI).Results The final analysis included seven trials comprising 1889 patients.Biochemical failure in 3months (RR0.73,95% CI 0.55-0.96;P =0.02),1 year(RR 0.69,95% CI 0.58-0.80;P < 0.01) and 2 years (RR 0.58,95% CI 0.41-0.82;P =0.002) were fewer in patients who received narrow band imagingassisted TURB.The recurrence rates of 3 months,1 year and 2 years were 14.44%,29.35% and 30% in white light imaging group.The recurrence rates of 3 months,1 year and 2 years were 3.61%,9.35% and 12.59% lower in narrow band imaging group compared with white light imaging group.Conclusions Narrow band imaging-assisted TURB was superior to conventional white light imaging-assisted TURB in primary non-muscle-invasive bladder cancer in 2 years.

ABSTRACT

BACKGROUND:Previous experiments have shown that modified platelet-rich plasma activated by liquid nitrogen freezing and thawing can promote the proliferation of human bone marrow mesenchymal stem cells and dental pulp stem cells in a concentration-dependent manner. OBJECTIVE:To investigate the effect of modified platelet-rich plasma at different concentrations on the proliferation of dental pulp stem cells from human exfoliated deciduous teeth. METHODS:Platelets were selected and harvested by automatic blood cellanalyzer, and then activated by liquid nitrogen freezing and thawing.α-MEM served as basal medium. Different concentrations of modified platelet-rich plasma (2%, 5%, 10%, 20%) or 10%fetal bovine serum were added, respectively. The difference in cellproliferation was observed. RESULTS AND CONCLUSION:Modified platelet-rich plasma at different concentrations could promote the proliferation of dental pulp stem cells from deciduous teeth. The effects of 2%modified platelet-rich plasma and 10%fetal bovine serum on promoting the proliferation of dental pulp stem cells from deciduous teeth were similar. These results indicated that 2%modified platelet-rich plasma could promote the proliferation of dental pulp stem cells from deciduous teeth, and substitute for fetal bovine serum in the amplification of dental pulp stem cells from deciduous teeth in vitro.

ABSTRACT

Objective:To investigate the effect of modified plateletrich plasma(mPRP)on the osteogenic differentiation of stemcells from human exfoliated deciduous teeth(SHED).Methods:mPRP at 1%,2%,5%,10% and FBS at 10% were added to thecultured SHED of passage 4,respectively.The influence of mPRP on alkaline phosphatase(ALP)activity was evaluated using ALPkit.RUNX2 and osteocalcin mRNA expression in the treated cells were examined by realtime PCR.Results:mPRP enhanced ALPactivity in the SHED,and the effect of mPRP was more obvious at 2%.Treatment of the cells with 2% mPRP upregulated the mRNAexpressions of RUNX2 and osteocalcin.Conclusion:mPRP can promote the osteogenic differentiation of SHED.

ABSTRACT

Objective To observe the effect of exposure of emphysema with intermittent hypoxia on oxidative stress injury of myocardial cells in rats. Methods Sixty male Wistar rats were divided randomly into four experimental groups(each n=15). The normal control group was bred normally. The emphysema group was exposed to cigarette smoke twice a day(once 30 minutes). The intermittent hypoxia(IH)group was exposed to intermittent hypoxia circumstance 8 hours/day,and the emphysema with IH group was exposed to cigarette smoke twice a day (once 30 minutes)and intermittent hypoxia circumstance 8 hours/day. Each group was exposed for 8 weeks. At the beginning of 9 weeks,the blood gas analysis was performed in 5 rats selected randomly from each group,and the rest rats were sacrificed and their hearts and lungs were taken. Under light microscope,the lung tissues stained with hematoxylin-eosin(HE)were examined. The lung pathology and the results of blood gas analysis showed that the emphysema with IH rat model was established successfully. The levels of malonaldehyde(MDA)and superoxide dismutase(SOD)in rat myocardium were measured by enzyme-linked immunosorbent assay(ELISA),and the subunit p22phox mRNA expressions of nicotinamide adenine dinucleotide phosphate(NADPH)-oxidase were detected by real-time reverse transcription-polymerase chain reaction(RT-PCR). Results Compared with the normal group, the MDA levels and p22phox mRNA expressions were increased obviously in emphysema group, IH group and emphysema with IH group〔MDA(μmol/g):2.93±0.54, 3.58±0.63, 4.51±0.72 vs. 1.75±0.56, p22phox mRNA:0.043±0.004,0.067±0.015,0.123±0.016 vs. 0.018±0.002,all P<0.05〕,but the activities of SOD were decreased significantly(U/mg:36.07±4.79,33.51±7.12,24.29±5.36 vs. 46.08±5.12,all P<0.05). In emphysema with IH group,the increase of MDA levels and p22phox mRNA expressions and decrease of SOD levels were more remarkable compared with those in emphysema group and IH group(all P<0.05). The expression of p22phox mRNA was positively correlated with MDA level(r=0.734,P<0.001). Conclusion The myocardial tissue oxidative stress injury in rats induced by emphysema with intermittent hypoxia exposure is more serious than that induced by exposure of either emphysema or intermittent hypoxia alone,NADPH oxidase possibly being the important medium of myocardial cell response to oxidative stress.

ABSTRACT

BACKGROUND:Previous experiments have shown that improved platelet-rich plasma can promote the proliferation of human dental pulp cells in a concentration-dependent manner, particularly when the concentration is 10%. OBJECTIVE:To investigate the effect of platelet-rich plasma at different concentrations on the proliferation and immunogenicity of human bone marrow mesenchymal stem cells. METHODS:Human bone marrow mesenchymal stem cells from healthy donors were cultured and passaged for 3-4 passages, identified by flow cytometry and differentiation inductions. Platelet-rich plasma samples which were manufactured from the venous blood of the same donor were used for culturing human bone marrow mesenchymal stem cells. The proliferation of human bone marrow mesenchymal stem cells was measured by cellcounting kit-8 method and the growth curves were drawn. The most suitable concentration of platelet-rich plasma was selected to culture human bone marrow mesenchymal stem cells for three generations and the Stro-1 expression rate on the surface of human bone marrow mesenchymal stem cells was determined through flow cytometry. RESULTS AND CONCLUSION:Platelet-rich plasma at the concentration of 5%-10%evidently promoted the proliferation of human bone marrow mesenchymal stem cells on the 6th and 8th days. The most effective concentration to promote the proliferation was 10%. Platelet-rich plasma at the concentration of 10%stil promoted the proliferation of human bone marrow mesenchymal stem cells on the 10th day, and maintained a better immunogenicity of human bone marrow mesenchymal stem cells compared to the control group. These findings indicate that platelet-rich plasma can promote the proliferation of human bone marrow mesenchymal stem cells in a concentration-dependent manner, and 10%platelet-rich plasma is better to maintain the immunogenicity of human bone marrow mesenchymal stem cells.

ABSTRACT

Objective To investigate the methylation status of the promoter of RASSF1A gene in the tissues of bladder cancer and renal carcinoma and its role in tumorigenesis. Methods Methylation specific PCR (MSP) was used to examine the methylation status of RASSF1A in bladder cancer and renal carcinoma.The specimen included 45 cases of bladder cancer tissues and paired adjacent normal tissues from cystectomy,9 cases of bladder cancer tissues from microresection,3 cases of normal bladder tissues from people without tumor,12 cases of renal carcinoma tissues and paired adjacent normal tissues. Results 30 of 54(55.6%)bladder cancer tissues showed RASSF1A gene aberrant methylation.Of which,26 of 45 ( 57.8 %) bladder cancer tissues from cystectomy showed aberrant methylation,while one of the paired normal tissues showed gene methylation,4 of 9 (44.4%) bladder cancer tissues from microresection showed gene methylation.Eight of 12(66.7%)renal carcinoma tissues showed aberrant methylation,while none of the paired adjacent normal tissues showed gene methylation.None of the three normal bladder tissues showed aberrant methylation.There was no significant correlation between the methylation status and the clinical pathological parameters in bladder cancer tissues from cystectomy,though there was a trend that more frequent methylation was detected in higher stage bladder cancer. Conclusions Hypermethylation of RASSF1A gene in bladder cancer and renal carcinoma suggeste its possible role in the tumorigenesis of both tumors.

ABSTRACT

Objective To analyze quantitatively the histological components, and to observe the morphological changes of the transition zone in benign hyperplastic prostates (BPH) and adult normal prostates. Methods The quantitative morphometry of 40 BPH specimens obtained from suprapubic transvesical prostatectomy and 10 normal prostatic specimens (controls) were performed with computer-assisted image analysis system,and all the samples were stained with hematoxylin and eosin (HE) and the Masson trichrome.The area percentages of stromal (smooth muscle and connective tissue),glandular epithelial and luminal components,the average areas of acini,lumens and epithelia,and the heights of epithelial cells were quantitated,respectively. ABSTRACT

Objective To evaluate intravesical instillation of epirubicin plus BCG to prevent recurrence of transitional cell cancer of bladder after surgical management. Methods A total of 44 cases of bladder cancer who underwent TURBT or partial cystectomy were divided into 2 groups.①Group 1 received intravesical instillation of epirubicin plus BCG (24 cases).Within 1 week following operation single dose of intravesical epirubicin instillation was given and from the second week regular intravesical BCG was instilled afterwards.②Group 2 received BCG alone (20 cases).Within 1 week after surgical management regular intravesical BCG instillation was given. All the patients were followed up for 24 months. ABSTRACT

Objective:To observe the effects of toadpoison and arsenic on the ultrastructure of dental pulp,Methods :Dental pulp of 36 teeth in 3 dogs was exposed by operation and then direct dressing with toadpoison paste or arsenic paste was performed. 1,2,6 and 24 hours after operation ,the pulps were processed and examinedunder a trasmission electron microscope (JEM-2000EX). Results:In toadpoison treated group ,the deformityof the basernembrane of pulp nerve ,swelling of metochondrion,degeneration of nerve axite were observed 1 h after dressing,and were more obvious in 2, 6 and 24 h. Damage of capillary and pulp cells were alsoobserved.In arsenic treated group ,swelling and break of capillaries ,deformity and degeneration of mitochondrion and endoplasmic reticulum in endoepithelial cells were observed,and were more obvious following time lasting. Damages of pulp nerve and pulp cells were also observed. Conclusion:Toadpoison and arsenic are toxic to pulp tissues. But the angiotoxicity of arsenic is stronger than that of toadpoison,the neurotoxicity and protoplasmic toxicity of toadpoison are stronger than those of arsenic.

ABSTRACT

Objective To evaluate the expression of endothelial Per-ARNT-Sim domain protein-1(EPAS-1)/hypoxia-inducible factor-2?(HIF-2?) and its relationship with vascular endothelial growth factor (VEGF) in bladder transitional cell carcinoma (BTCC). Methods The expression of EPAS-1/HIF-2? and VEGF by immunohistochemical staining was examined in 60 cases of BTCC (Grade Ⅰ in 28 cases,Grade Ⅱ in 12,Grade Ⅲ in 20) and 8 subjects with normal urothelium. Of the 60 cases 29 had superficial (≤pT 1) bladder cancer and 31 had invasive (≥pT 2 bladder cancer).?2 test was used to assess the relationship between EPAS-1/HIF-2? and VEGF expression versus tumor grade and stage. Results The expressions of EPAS-1/HIF-2? and VEGF were negative in all normal bladder tissue but high in BTCC.Of the 60 cases,34 (56.6%) were positive and 26 ( 43.4%) were negative for EPAS1/HIF-2?.Four cases (14.3%) of Grade Ⅰ,11 (91.7%) of Grade Ⅱ and 19 (95.0%) of Grade Ⅲ were positive for EPAS-1/HIF-2?. Positive staining was seen in 5 (17.2%) of the superficial and 29 (93.5%) of the invasive cancer cases.EPAS-1/HIF-2? positivity was correlated with high histological grade (r=0.862,P

ABSTRACT

Objective To retrospectively analyze the indication of endosseous dental implant,evaluate the effectiveness of clinical application of endosseous dental implant for tooth loss,and study the possible factors which may affect the success rate of implant denture.Methods One hundred and fifty-seven endosseous dental implants in 94 patients were followed up for 4 months to 8 years.The indications of endosseous dental implant were studied based on the changes in the distribution of dental implants in jawbone.The success rate of osseointegration and restoration of the endosseous dental implant were evaluated by checking the soft tissue around the implants,the stability of the implants,the bone resorption around the implants,and the degree of stability against rotation of the implants.Results The rate of performing endosseous dental implant for restoration of full-mouth anodontia was 7.6%,the rate of endosseous dental implant used in the free end anodontia was 25.5%,and it was 66.9% in non-free end anodontia.The success rate of osseointegration in the endosseous dental implant was 98.1%,and 5 years after prosthesis the success ratio of the endosseous dental implant was 93.8%.The success ratio of the endosseous dental implants which the primary stability(anti-rotation torsion)

ABSTRACT

Objective To investigate the promoter methylation status and the transcription level of BLU in bladder carcinoma and their significance.Methods The methylation status of promoter and the transcription level of BLU were examined by methylation specific PCR(MSP) and RT-PCR in bladder cancer tissues from 54 patients,adjacent normal tissues from 45 cases and normal bladder tissues from 3 noncancerous patients.Results ① 31.5%(17/54) bladder cancer tissues showed BLU hypermethylation;neither the paired adjacent normal tissues nor the three normal bladder tissues showed BLU hypermethylation.② There was no significant correlation between the methylation status and the clinical pathological parameters in bladder cancer.③ The expression of BLU was absent in 42.2%(19/45) of bladder carcinoma from cystectomy.④ Loss of BLU expression was associated with the stage and grade of the tumor(P
