ABSTRACT

To systematically evaluate the effect of traditional Chinese medicine(TCM) on the expression of inflammatory factors in peripheral blood of patients with coronary heart disease complicated with anxiety and depression,and explore its efficacy and safety in treatment of anxiety and depression. In this study,CNKI,VIP database,WanFang database,PubMed and Cochrane Library were searched to collect randomized controlled trials(RCTs) of TCM in the treatment of coronary heart disease complicated with anxiety and depression,and 2 researchers independently screened the literatures and extracted the data. The quality of the included literatures was evaluated with Cochrance bias risk evaluation tool and Meta analysis was conducted by Cochrane Revman 5.3 software. A total of 21 research articles were included,with a total sample size of 2 342 cases,1 175 cases in the treatment group and 1 167 cases in the control group. Meta analysis results showed that the treatment group reduced the hypersensitive C-reactive protein(hs-CRP)[standard mean difference(SMD)=-1.61,95% confidence interval(CI)(-2.14,-1.09),P<0.01],interleukin(IL)-8[mean difference(MD)=-5.03,95% CI(-8.37,-1.70),P=0.003],IL-17[MD=-33.27,95% CI(-40.15,-26.39),P<0.01],tumor necrosis factor(TNF)-α[SMD=-1.18,95% CI(-1.98,-0.38),P<0.01],and homocysteine(Hcy)[MD=-3.45,95% CI(-4.85,-2.04),P<0.01]. The treatment group was better than the control group in terms of relieving anxiety and depression,i.e. scores of Hamilton anxiety scale(HAMA) [SMD=-1.97,95% CI(-2.48,-1.46),P<0.01],Hamilton depression scale(HAMD) [SMD=-1.94,95% CI(-2.50,-1.38),P<0.01],and self-rating depression scale(SDS)[SMD=-0.72,95% CI(-0.90,-0.54),P<0.01],so in terms of ,with statistically significant difference. 4 articles mentioned that no obvious adverse reactions occurred,4 articles mentioned that the treatment group had drowsiness,dry mouth and bitter mouth,gastrointestinal reactions,but the incidence rates were significantly lower than those of the control group. The other 13 articles did not mention the occurrence of adverse reactions.

OBJECTIVE@#To evaluate and analyze the accuracy of three-dimensional camera system acquisition based on stereophotography for the photographic acquisition of images of facial deformities.@*METHODS@#3D digital models of 45 waxen facial models of patients with facial deformities were obtained via a 3D camera system based on stereophotography. A total of 19 feature lines were measured on each 3D model by using the software. The measurements taken by the coordinate-measuring machine were accepted as standard values. Statistical analysis was performed to evaluate the accuracy of the system and the corresponding factors.@*RESULTS@#Statistical analysis results showed that the measured values of the characteristic distance obtained by 3D camera system were statistically different from the standard values (P0.05). The facial regions significantly influenced measuring error (P0.05). The middle facial regions had significant difference with bilateral facial regions on the percentage of measurement values (P0.05).@*CONCLUSIONS@#The accuracy of 3D camera system based on stereophotography for the photographic acquisition of facial deformities are influenced by the morphology of the facial deformities and facial regions. Moreover, the measuring error is acceptable in clinical settings.

ABSTRACT

OBJECTIVE To explore the protective effects and mechanisms of Ginsenoside Rg1 (Rg1) on H2O2-induced hippocampal neurons aging in vitro. METHODS The primary culture hippo-campal neurons(7 d)were randomly placed into six groups:normal control group,H2O2(200 μM)treat-ment group,and H2O2+Rg1(1,5 and 10μM)groups.The neurons were with Rg1(1,5 and 10 μmol·L-1) for 6h. H2O2(200 μmol·L-1) was added to the medium and incubate for 18 h. The Dihydroethidium (DHE) staining was performed for ROS production assessment. The LDH release and Hoechst 33258 were performed to examine the neuronal damage and apoptosis. The immunoblot was used to deter-mine the expression of β-Gal,NOX2,p22phox,p47phox,NLRP-1,ASC and Caspase-1 in hippocampal neurons.The ELISA was performed to detect the levels of IL-1β and IL-18 released in the supernatant in hippocampal neurons.RESULTS Rg1(5 and 10 μmol·L-1)significantly reduced the ROS production, attenuated H2O2-induced neuronal damage and apoptosis (P<0.05, P<0.01). The immunoblot results showed that Rg1(5 and 10 μmol·L-1)treatment significantly decreased the expression of β-Gal,NOX2, p22phox,p47phox,NLRP-1,ASC and Caspase-1 in hippocampal neurons(P<0.05,P<0.01).Additionally, Rg1(5 and 10 μmol·L-1)treatment significantly decreased IL-1β and IL-18 release in the supernatant. CONCLUSION The protective effect of Rg1 in H2O2-induced hippocampal neurons aging may be due to inhibit NOX2-NLRP1 activation.

ABSTRACT

Helicobacter pylori (Hp),a proven pathogen of digestive disease,is closely associated with gastrointestinal diseases including peptic ulcer,chronic active gastritis,stomach cancer,and stomach-associated lymphoid tissue lymphoma. Thus,precise and timely diagnosis of Hp is of great significance. In addition to C or C breath test,rapid urease test,and other commonly used Hp diagnosis methods,some new endoscopic techniques such as magnifying endoscopy,narrow-band imaging,confocal laser endomicroscopy,Fuji intelligent chromoendoscopy,and I scanning have been used for the direct observation of the fine-structure of stomach with Hp infection or for the identification of living Hp. This article reviews the application of digestive endoscopy in the diagnosis of Hp infection.

ABSTRACT

Objective To construct plasmids for knock-out of protein arginine methyltransferase 3 (prmt3) gene using CRISPR/Cas9 gene editing method and examine the effect of prmt3 knockout on the proliferation of human non-small cell lung cancer(NSCLC)A549 cells.Methods Synthesized sgRNA oligos targeting prmt3 gene were cloned into LentiCRISPR vector and positive constructs confirmed by sequencing later .After infection with the packaged virus , A549 cells were screened with puromycin , and then the single clones were isolated .The protein level of PRMT3 in individual cell clones was analyzed with Western blot . Biological assay of clone formation , wound healing , flow cytometry assay and mass spectrometry ( MS) analysis were used to compare cellular proliferation behavior changes between control cells and cells with prmt3 gene knockout .Results The LentiCRISPR plasmids targeting prmt3 gene were confirmed by sequencing , and the PRMT3 protein level was significantly decreased in PRMT 3 KO cells compared with control cells .Depletion of PRMT3 promoted cell proliferation and led to cell cycle arrest at G 2/M phase, but had no influence on cell migration .Besides, some PRMT3 substrate candidates were identified with mass spectrum assays .Conclusion A549 cells with prmt3 gene knockout based on CRISPR/Cas9 are successfully established .PRMT3 can regulate cell cycle and proliferation .

ABSTRACT

The study was aimed to investigate the correlation between the immunocompetence and the fingerprints of supernatant extracts from Radix Astragali by multi-index integrated evaluation, and reveal the material basis of Radix Astragali improving the immunological function. After oral administration 10 batch of supernatant extracts from Radix Astragali on immunosuppressed mice, phagocytic index, delayed type hypersensitivity (DTH) degree, organs index and the level of cytokines IFN-γ, IL-4 (ELISAs) were measured, and each common peak from HPLC-DAD/HPLC-ELSD fingerprints were correlated with the above data. A number of components in supernatant extracts of Radix Astragali display a substantial correlation with the immunocompetence. Supernatant extracts of Radix Astragali can significantly enhance immunological function on mice, which is related to various components in Radix Astragali.

ABSTRACT

An HPLC method was established for the determination of adenosine, γ-aminobutyric acid (GABA) and six flavonoids (calycosin-7-glucoside, ononin, calycosin, isoliquiritigenin, formononetin and medicarpin) in Radix Hedysari. The samples were extracted with methanol by refluxing for 4 h. The HPLCDAD was performed on a Diamonsil C18 column (250 mm×4.6 mm, 5 μm) with acetonitrile-water as the mobile phase. The column temperature was at 40℃ and the flow rate was 1.0 mL·min-1, while the temperature of drift tube was 110.5℃ and the nebulizing gas flow was 3.1 L·min-1 for the ELSD system. The results showed all the eight components had good linear relationships (r2=0.992 8-1.000 0) in the range of the test concentration. The RSD of precision, stability and repeatability were less than 2%. The average recovery rates were 96.78%-103.45%, and RSD were 0.29%-1.61%. The index component contents of Radix Hedysari form 24 different origins were determined and used as variable factors in clustering analysis. The results were classified into 2 groups basically in accordance with the regional cluster. And the consequence was in consistent with the results of principal component analysis. This HPLC method is simple, shows good sensitive and accurate, and provides the experimental basis for multi-index control of Radix Hedysari. Clustering analysis for Radix Hedysari quality control has a certain reliability and objectivity.

ABSTRACT

<p><b>OBJECTIVE</b>To assess the role of p38 mitogen-activated protein kinases (p38MAPK) in the protective effect of remifentanil preconditioning (RPC) on hepatic ischemia-reperfusion injury in rats.</p><p><b>METHODS</b>Ninety-six male SD rats were randomly assigned into sham-operated group, ischemia-reperfusion group (I/R group), RPC group, and SB (an inhibitor of p38 MAPK) +RPC group. The rats were sacrificed at the end of reperfusion, and serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), tumor necrosis factor-α (TNF-α), and interleukin-1β (IL-1β) were measured. HE staining was used to observe the hepatic histopathological changes, and Western blotting was employed to examine p38MAPK and pp38MAPK protein expression. TUNEL staining was used to examine cell apoptosis in the liver tissues.</p><p><b>RESULTS</b>Compared with sham-operated group, I/R group showed significantly increased serum levels of AST, ALT, TNF-α and IL-1β with obvious histopathological changes and cell apoptosis in the liver. RPC significantly decresed the elevated serum levels of AST, ALT, TNF-α and IL-1β and lessened hepatic histopathological changes, and caused reduced p38MAPK phosphorylation and hepatic cell apoptosis index. The protective effects of RPC were abolished by SB 203580 pretreatment.</p><p><b>CONCLUSION</b>RPC attenuates the production of inflammatory factors by activating p38MAPK signal pathway to improve hepatic ischemia-reperfusion injury, and these effects can be blocked by SB203580, a p38MAPK inhibitor.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of endostar in controlling ascites tumor formation in mice.</p><p><b>METHODS</b>Mouse models bearing ascites tumors were established via intraperitoneal injection of H22 and S180 cell lines. Eighty-eight ICR mice were randomly assigned into 4 groups, namely the control group (0.9% normal saline) and 3 endostar groups with 8 mg/kg endostar administration daily, every other day or every two days. The peritoneal membrane permeability of the mice was assessed using Evan blue staining. The body weight curve of mice was drawn, and the cumulative ascites volume and number of red cells and tumor cells in the malignant ascites were determined. The survival of the mice was evaluated to assess the therapeutic effect of endostar.</p><p><b>RESULTS</b>Compared with the control group, the mice receiving daily endostar injection showed obviously lower ascites accumulation and peritoneal capillary permeability (P<0.05) with reduced count of ascites tumor cells and red cells and tumor burden of the abdominal cavity. The mice with daily endostar injection also showed longer survival than the control group.</p><p><b>CONCLUSIONS</b>Continuous intraperitoneal injection can be the optimal means for endostar administration for treatment of malignant ascites.</p>

ABSTRACT

Recent work indicates that an Aurora kinase inhibitor MK-0457 (VX-680), a small-molecule inhibitor of Aurora kinases A, B, C and BCR-ABL, FLT-3, JAK-2, can block the progression of cell growth cycle, causing apoptosis in a range of human tumors. MK-0457 has the activity against expressions of wild-type and mutated bcr-abl gene, including the T315I mutant, and can inhibit the activity of FLT-3, JAK-2 and their mutated types as well. Clinical applications suggest that the MK-0457 has therapeutic effect on the highly refractory CML and CML with poor prognosis, Ph(+) ALL with T315I mutant, relapse refractory AML and JAK-2 positive myeloproliferative diseases (MPD). The intensive preclinical studies and the on-going phase II clinical trials will open up a new vista of therapy for some hematological malignancies. This review focuses on the pharmacologic action of MK-0457 and its clinical trial as well as combined application.

ABSTRACT

<p><b>OBJECTIVE</b>To study the plasma homocysteine (Hcy) distributive characteristics in different sex and age groups of the community residents in Guangdong, and to analyze its associated factors as well as its effects on cerebral artery stenosis.</p><p><b>METHODS</b>In this study, we presented data concerning 2500 residents who had undergone a systematic health checkup,including clinical and biologic examinations. Plasma was saved from 1020 residents who were asymptomatic but having TCD examination. Hcy concentration was measured by fluorescence ration biochemical assay kit.</p><p><b>RESULTS</b>The average plasma Hcy levels of males (15.0 micromol/L+/-5.8 micromol/L) were significantly higher than those of females (12.4 micromol/L-I 3.9 micromol/L, P< 0.05). Hcy level increased parallel with age. The prevalence rate of hyperhomocysteinemia in males (47.2%) was higher than in females (23.2%). Hcy levels in MCA stenosis group (18.3 micromol/L+ 5.3 micromol/L) were significantly higher than that in control group (12.9 micromol/L+/- 4.6 micromol/L, P< 0.05). Based on logistic regression analysis, Hcy was only associated with sex and age, but not with blood lipids etc.</p><p><b>CONCLUSION</b>Hcy levels were different in sex and age groups which emerged as independent predictors of hyperhomocysteinemia but had no association with blood lipids in the community residents, suggesting that hyperhomocysteinemia might worsen cerebral artery stenosis.</p>

ABSTRACT

@#ObjectiveTo analyze the clinical characteristics and treatment of severe acute respiratory syndrome (SARS).MethodsDuring epidemic period of SARS,all suspected patients were isolated and treated at very early stage. Clinical symptoms, signs and changes of the chest film were observed carefully to make early diagnosis and begin early intervention. 14 SARS cases with definite diagnosis were treated by Western Medicine combined with Chinese Traditional Medicine.3 cases of them with severe type were treated by timely technical ventilation,which was nasotracheal intubition guided with fibronchoscope,with the principle of low tide volume and high PEEP to improve oxygen combination.ResultsAll patients were cured to discharge. Only two cases of mild pulmonary fibrosis were found after 2 months' follow-up. None of the medical workers taking part in rescuing and treating SARS patients and none of the family members of the patients were infected.ConclusionDuring SARS epidemic period,early isolation,early diagnosis,early combined treatment of Western Medicine and Chinese Traditional Medicine,and prompt technical ventilation intervention on severe cases are important factors to successfully rescue and treat SARS patients.
