ABSTRACT

Embryo implantation is one of the most critical steps in the reproductive process. The failure of embryo implantation to continue development is one of the important reasons leading to infertility. The success of embryo implantation depends on the high receptivity of endometrium and the embryo with implantation ability. Autophagy is a process in which cytoplasm, organelles, and inclusions are absorbed by double-membrane vesicles and transported to lysosomes for degradation and recycling, which is a way to maintain the homeostasis. A large amount of evidence have shown that autophagy plays an important role in all aspects of embryo implantation. Based on this, this paper explored the relationship between autophagy and endometrial receptivity and embryo implantation ability. According to the latest research progress, this paper combed 5 mechanisms (promotion of decidualization of endometrial stromal cells, promotion of apoptosis, regulation of hormone levels, coordination of inflammation, and promotion of ovulation) of 14 kinds of Chinese medicine monomers, including emodin, catalpol, paeoniflorin, resveratrol, folic acid, zearalenone, curcumin, wogonin, quercetin, chrysin, berberine, apigenin, phisetine, and kaempferol, in regulating different links of autophagy intervention in embryo implantation. This paper is expected to provide references and ideas for future Chinese medicine monomers to improve the success rate of embryo implantation.

ABSTRACT

【Objective】 To analyze the researches on blood donation service during 2001 to 2020, explore the development of blood donation service in China and discover the hot spots and weaknesses in current research, aimed to provide reference for future research. 【Methods】 The research team, institutions and hotspots of related literature from China National Knowledge Infrastructure (CNKI) were analyzed using CiteSpace. 【Results】 After analyzing the selected 969 relevant literature, it is concluded that there is few communication between the core author teams. The inter-agency research network is not mature enough, and the cooperation and communication between institutions need to be further strengthened. The high-frequency keywords were volunteer donors, satisfaction, blood donation response and humanized service. Quality control in the blood donation service process, maintenance and recruitment of regular blood donors, and the establishment and improvement of voluntary blood donation teams were research hot spots. 【Conclusion】 China has made fruitful achievements in the research of blood donation service after 20 years of development. The key point of future research is to strengthen the cooperation between different research teams, integrate the first-line practice of blood collection and supply, as well as explore the individualized and localized theory of blood donation service.

ABSTRACT

Objective To observe the effects of total glucosides of white paeony(GWP) on psychological ethology and neuroendocrine system of stress rats. Methods Ten male Sprague-Dawley rats were treated as normal control group,the others (n=40) were constructed by isolated custody and chronic unpredictable stimuli to build the model of stress rats.Stress rats were randomly divided into 4 groups:model control group,GWP high dose group and low dose(66,33 mg·kg-1) group,fluoxetine(5. 0 mg·kg-1) group,each group had 10 rats.The changes of rat behavior were monitored by locomotor activity system.The change of emotion was observed by tail suspension test.The pathology change of adrenal was observed by HE staining.The levels of CorT and ACTH in serum were measured by radioimmunoassay.The expression of CRH mRNA of hypothalamic were detected by RT-PCR. Results The activity of the level scoring and vertical scoring of GWP high dose group was obviously improved as compared to that in model control group,while static time was significantly shortened,Adrenal index of GWP groups were increased significantly(P<0.05),thymus index of the treatment group was declined(P<0.05),CorT and ACTH content were decreased (P<0.05) and hypothalamus CRH mRNA of GWP group was decreased(P<0.05). Conclusion The improvement of activity and related hormone anomalous changes were observed in rats treated with GWP.The mechanism of its actions might be involved adjusting the endocrine function,decreasing CorT and ACTH content and the expression of hypothalamic CRH mRNA of stress rats.

ABSTRACT

Objective To study the association of early prognosis and risk factors about early neurological deterioration (END) in aged patients with cerebral hemorrhage in basal ganglia. Methods One hundred and thirty-nine aged patients with cerebral hemorrhage in basal ganglia were selected. The patients were divided into END group (59 cases) and non-END group (80 cases), the clinical data were retrospectively analyzed. The patients were followed up 3 months after discharge, the prognosis was evaluated by modified Rankin scale. Logistic regression analysis was used to determine the independent risk factors of END. Results The incidence of END was 42.4%(59/139). The incidence of unfavourable prognosis in END group was significantly higher than that in non-END group:78.0%(46/59) vs. 30.0%(24/80), and there was statistical difference (P0.05). The Logistic regression analysis results showed that brain ventricle hemorrhage and white blood cell count at admission were independent risks factor for END in aged patients with cerebral hemorrhage in basal ganglia(P < 0.05 or < 0.01). Conclusions The aged patients with cerebral hemorrhage in basal ganglia will be prone to END, the brain ventricle hemorrhage and white blood cell count at admission are independent risks factor for END, and the early prognosis is poor.

ABSTRACT

Objective To evaluate the differentiation of mouse tracheal epithelial cells ( MTEC) at an air-liquid interface and to investigate the influences of influenza virus on the cystic fibrosis transmem-brane conductance regulator ( CFTR) in primary cultured MTEC for further elucidating the possible mecha-nism of imbalanced fluid and salt transportation in respiratory system caused by influenza virus infection . Methods The morphology of primarily cultured MTEC was observed under inverted microscope .Trans epi-thelial electrical resistance ( TEER) was measured by a resistance meter to evaluate the integrity of cultured MTEC.An Ussing chamber apparatus was used to record the short-circuit current of primary cultured MTEC . Results The primarily cultured MTEC clustered together and had a tight pavement-like appearance under light microscope .The TEER was greater than 1000 Ωafter 6 days of culture .Influenza virus could reduce the short-circuit current of CFTR to (52.77±10.30)%in intact cell membrane and to (41.50±1.09)%in monolayer MTEC after increasing the permeability of basement membrane .It had been proved that CFTR was essential to maintaining the balance of fluid and salt transportation in respiratory system.Conclusion Mouse MTEC are efficiently cultured at a air-liquid interface and the primarily cultured cells are highly simi-lar to those in a normal physiologic state .Influenza virus may block the secretion of anions through inhibiting the function of CFTR , which may induce the development of chronic obstructive pulmonary disease and the incidence of asthma .

ABSTRACT

Objective To construct MIA3 psicheck2 wild-type and mutant vectors targeting miR-374b,and provide the previous guarantee for the dual luciferase reporter assay.Methods The amplification primer was firstly designed according to mice MIA3-3'UTR sequence information,mice whole blood genomic DNA was taken as the template for PCR amplification of MIA3-3'UTR sequence,and the PCR product was cloned into psicheck2 dual luciferase reporter vector.Then,mutant primer was designed to mutate the MiR-374b seed sequence target TATTATA into AAATTAT so as to construct mutant vector.At last,the vector enzyme digestion evaluation and sequencing method was used to evaluate the constructed vectors.Results It could be seen from the analysis of agarose electrophoresis that the PCR amplification size of vector was consistent with the theoretical size.DNA sequencing evaluation showed that the MIA3-3'UTR-WT vector had been constructed successfully.The construction of mutant vector has successfully mutated the MiR-374b seed sequence target TATTATA into AAATTAT.Conclusion The successful construction of the vector will lay a foundation for the further evaluation on whether there is an actual binding site between the miR-374b and the chondrogenic differentiation-related target gene MIA3.
