ABSTRACT

This study analyzed the quality markers(Q-markers) of Yuquan Capsules(YQC) based on serum pharmacochemistry of Chinese medicine and detected the components and metabolites of YQC absorbed into the blood by UPLC-Q-TOF-MS and UNIFI systems. As a result, 32 components of YQC were detected, including 17 prototype components and 15 metabolized components. Among them, 12 prototype components(ginsenoside Rh_2, genistein, formononetin, puerarin, daidzein, schizandrin A, schizandrin B, schizandrin C, schizandrol A, schizandrol B, gomisin D, and ononin) and 12 metabolized components(ginsenoside Rg_1, ginsenoside Rg_2, ginsenoside Rg_3, ginsenoside Ro, 3'-methoxypuerarin, daidzin, astragaloside Ⅱ, astragaloside Ⅳ, glycyrrhizic acid, liquiritigenin, isoliquiritin, and verbascoside) showed inhibitory effects and pharmacological activities against diabetes, and these 24 blood-entering components against diabetes were identified as Q-markers of YQC.

ABSTRACT

To investigate the effects of Kluyveromyces marxianus M3 isolated from Tibetan mushrooms on diet-induced hypercholesterolemia in rats, female Wistar rats were fed a high-cholesterol diet (HCD) for 28 d to generate hyperlipidemic models. Hyperlipidemic rats were assigned to four groups, which were individually treated with three different dosages of K. marxianus M3+HCD or physiological saline+HCD via oral gavage for 28 d. The total cholesterol (TC), triglycerides (TG), high-density lipoprotein cholesterol (HDL-C), and low-density lipoprotein cholesterol (LDL-C) levels in the serum and liver of the rats were measured using commercially available enzyme kits. In addition, the liver morphology was also examined using hematoxylin and eosin staining and optical microscopy. According to our results, the serum and liver TC, TG, LDL-C levels and atherogenic index (AI) were significantly decreased in rats orally administered K. marxianus M3 (p <0.01), and the HDL-C levels and anti atherogenic index (AAI) were significantly increased (p <0.01) compared to the control group. Moreover, K. marxianus M3 treatment also reduced the build-up of lipid droplets in the liver and exhibited normal hepatocytes, suggesting a protective effect of K. marxianus M3 in hyperlipidemic rats.

FUNDAMENTO: una de las vías fundamentales para garantizar la calidad en los laboratorios clínicos es mediante el uso del suero control o controlador. El alto costo de estos productos en el mercado nacional es una problemática hoy en día. OBJETIVO: evaluar la estabilidad de un suero bovino adulto para proteínas totales como controlador bioquímico en los laboratorios clínicos. MÉTODO: se realizó un estudio cuasiexperimental, mediante muestreo probabilístico durante tres años (enero de 2010-diciembre de 2012). Se evaluó la estabilidad a tiempo real del suero enriquecido con proteínas totales durante 12 meses a dos temperaturas (refrigeración y congelación). El estudio quedó diseñado para tres lotes por mes y tres determinaciones por lote, para un número total de muestra del estudio (N=63). RESULTADOS: se obtuvo un suero control líquido para proteínas totales. La concentración de proteínas totales, en el producto obtenido en condiciones de refrigeración y congelación se mantuvieron estables por un tiempo de 12 meses. El producto obtenido al compararse con el suero control de la firma OLYMPUS, mostró fluctuaciones similares con respecto a la concentración de proteínas totales. CONCLUSIONES: se logró un material de referencia como controlador estable por un período de 12 meses.


BACKGROUND: one of the main ways to guarantee the quality in clinical labs is by using a control serum or controller. A problem we face is the lack of these products in the domestic market due to their high cost. OBJECTIVE: to evaluate the stability of an adult bovine serum for total proteins as a biochemical controller in clinical labs. METHOD: a quasiexperiment was conducted during three years (January 2010- December 2012) through a probabilistic sampling. The stability in real time of the serum enriched with total proteins was evaluated during 12 months under two temperatures (refrigeration and freezing). The study was designed for three batches per month and three determinations per batch, for a total number of sample of the study (N=63). RESULTS: a liquid control serum for total proteins was obtained. The concentration of total proteins in the product obtained under refrigeration and freezing conditions kept stable for 12 months. The obtained product, when compared to the control serum from the company OLYMPUS, showed similar fluctuations regarding the concentration of total proteins. CONCLUSIONS: a reference material was obtained as a stable controller for a period of 12 months..

ABSTRACT

Background: Aberrant glycosylation is the universal feature of cancer and components of various glycoconjugates, such as sialic acid is found to rise in various malignancies. The objective of this study was to evaluate the serum and salivary sialic acid in oral potentially malignant disorders (OPMD) and oral cancer (OC) to investigate the possibility of using this as a diagnostic marker. Materials and Methods: The study included 85 subjects, who were grouped as control (30), OPMD patients (25), and oral cancer patients (30). Serum and unstimulated whole saliva was collected from subjects of all groups and sialic acid estimation was done using spectrophotometry. The results were tabulated and analyzed statistically. Results: The mean serum sialic acid levels in normal, OPMD, and oral cancer group were 7.515, 19.620, and 55.235 mg/dL, respectively, whereas the levels of salivary sialic acid were 1.5113, 2.3302, and 9.0304 mg/dL, respectively. A very highly significant rise (P < 0.005) in serum and salivary sialic acid was observed in the study subjects compared with that of the control. Conclusions: The present study showed a significant and gradual increase in serum and salivary sialic acid from control to oral potentially malignant disorders to oral cancer. From this study we can suggest that sialic acid can be used as a reliable biomarker. As this monosaccharide is observed in saliva in detectable quantity, saliva can be used as a diagnostic medium for screening and early detection of oral cancer.

ABSTRACT

This study aimed to measure the levels of interferon-gamma (IFN-gamma), tumor necrosis factor-alpha (TNF-alpha), interleukin 1 (IL-1), interleukin 6 (IL-6), and nitrite/nitrate (NOx) in serum of dogs experimentally infected with Rangelia vitalii. Twelve female mongrel dogs were divided into 2 groups; group A (uninfected controls) composed by healthy dogs (n=5) and group B consisting of dogs inoculated with R. vitalii (n=7). Animals were monitored by blood smear examinations, which showed intraerythrocytic forms of the parasite on day 5 post-infection (PI). Blood samples were collected through the jugular vein on days 0, 10, and 20 PI to determine the serum levels of IFN-gamma, TNF-alpha, IL-1, IL-6, and NOx. Cytokines were assessed by ELISA quantitative sandwich technique, and NOx was measured by the modified Griess method. Cytokine levels (IFN-gamma, TNF-alpha, IL-1, and IL-6) were increased (P<0.01) in serum of infected animals. Serum levels of NOx were also increased on days 10 PI (P<0.01) and 20 PI (P<0.05) in infected animals. Therefore, the infection with R. vitalii causes an increase in proinflammatory cytokines and nitric oxide content. These alterations may be associated with host immune protection against the parasite.

ABSTRACT

A EPO é um fator de crescimento glicoprotéico sintetizado pelas células adjacentes aos túbulos proximais renais regulada via mecanismo de "feed back" envolvendo a tensão de oxigênio tissular. Na baixa tensão de oxigênio arterial, a produção de EPO aumenta causando uma maior produção de eritrócitos na medula óssea. Devido ao pouco conhecimento da concentração de EPO sérica em equinos e a ausência de trabalhos sobre o efeito da idade e sexo sobre a sua concentração o trabalho teve como objetivo comparar a concentração sérica de eritropoietina em equinos da raça Árabe de sexos e idades diferentes. Foram utilizados 31 equinos da raça Árabe, com idades de seis a 12 meses (jovens) e acima de 24 meses (adultos), sendo 13 machos (seis jovens e sete adultos) e 18 fêmeas (oito jovens e 10 adultas), clinicamente sadios. As amostras de sangue foram colhidas por venipunção jugular e o soro armazenado até o momento do processamento. A concentração sérica de eritropoetina foi determinada pelo método de radioimunoensaio (RIA) utilizando kit comercial (EPO Trac TM125I RIA, Diagnostic Systems Laboratories, Webster, Texas, USA). Para análise estatística dos dados utilizou-se o Teste t de Student ao nível de 5% de significância (P<0,05). Não foram observadas diferenças significativas (P<0,05) quando se compararam os animais divididos entre sexos e as idades de seis a 12 meses (jovens) e acima de 24 meses (adultos). Conclui-se que a concentração de eritropoietina em equinos da raça Árabe independe do sexo e idade, e pode ser utilizada como valores de referência, porém ressalta-se a necessidade da obtenção de valores de referência para cada laboratório.(AU)


The erythropoietin (EPO) is a glicoproteic grow factor synthesized by the adjacent cells in the proximal renal tubes and controlled by feedback mechanism that involve the tension of tissular oxygen. In the presence of low tension of arterial oxygen, the production of EPO increases and it causes a major production of erythrocytes in the marrowbone. Due to poor knowledge of the serum concentration of erythropoietin in equines and the absence of information about the differences between age and gender, the aim of this study was to compare the serum concentration of erythropoietin in Arabian horses, clinically healthy with different age and gender. A total of 31 horses were used from 6 to 12 months old (young) and over 24 months old (adult), 13 males (six young and seven adult) and 18 females (eight young and ten adult) clinically healthy. Blood samples were collected from jugular vein using a vacuntainer with blood clot gel activator and the serum was separated by centrifugation and stored until the time of processing. The EPO serum concentration was measured by radioimmunoassay method (RIA) using a commercial kit (EPO Trac TM125I RIA, Diagnostic Systems Laboratories, Webster, Texas, USA). For statistical analysis of data were used the Student t test at 5% level of significance (P<0.05). It was not observed any statistic differences (P<0.05) between the sex and age of the animals from 6 to 12 months old and over of 24 months. We concluded that the concentration of the EPO in Arabian equines is independent of the sex and age, and can be used as reference values however it is very important and necessary to obtain references values for each laboratory.(AU)

Purpose : The hallmark of chronic hepatitis B (CHB) infection is the presence of hepatitis B surface antigen (HBsAg) positivity for at least 6 months. Recently, serum levels of HBsAg have been compared with serum HBV DNA as a surrogate marker to monitor CHB patients. However, data correlating these two markers are scarce. Hence, the present study was done to correlate HBV DNA with HBsAg in CHB patients. Materials and Methods: Consecutive patients of CHB were included. HBV DNA was measured by real-time polymerase chain reaction (PCR). Serum HBsAg was measured by Architect HBsAg. Results: Of the 198 patients enrolled, 166 fulfilled the inclusion criteria (mean age 43 ± 14 years, 87% males) and the median HBV DNA was 1.7 × 10 3 (range 6.0-1.1 × 10 8 ) IU/ml. Median HBsAg was 8.7 × 10 3 (range 5.0-3.2 × 10 5) IU/ml. Overall correlation between HBV DNA and HBsAg was weak but significant (Spearman ρ = 0.443, P < 0.01). Correlation in HBe antigen-positive group was better (ρ = 0.402, P < 0.01) in comparison to HBe antigen-negative group (ρ = 0.193 P = 0.05). Good correlation existed in treatment-naïve group (ρ = 0.538, P < 0.01) .Correlation was regardless of normal or raised alanine transaminase (ALT). Eighty (48%) patients had high HBV DNA (≥2000 IU/ml). Correlation in high DNA group was significant (P < 0.01). The best cut-off of HBsAg for diagnosing high DNA is 3.36 ×10 3 IU/ml. Conclusions: Serum HBsAg correlates with HBV DNA in CHB patients, especially in high serum HBV DNA, HBe antigen-positive and treatment-naïve group. HBsAg levels can be used for predicting high serum HBV DNA levels.

ABSTRACT

Struma ovarii are specialized form of mature ovarian teratoma comprised predominantly of thyroid tissue (>50%). Most of the struma ovarii are benign; rarely can they undergo malignant transformation. Elevated CA-125 levels with benign struma ovarii have been seen in only 5 cases in literature. The association of malignant struma ovarii and high CA-125 levels with pseudo-Meig syndrome has been reported in only 2 cases in English literature. We describe a case of a 46-year-old multigravida who presented with an abdominal mass and raised CA-125 levels. Radiological investigations revealed bilateral cystic adnexal masses with ossified elements on left side suggesting a teratoma. Intraoperative frozen section and final pathology revealed bilateral teratoma with follicular variant of papillary thyroid carcinoma arising in the left ovary. To the best of our knowledge, this is the first case of malignant struma ovarii in combination with bilateral teratoma. The dilemmas related to preoperative diagnoses with elevated CA-125 levels, mimicking an epithelial ovarian neoplasm; intraoperative frozen section consultation; management and follow-up issues in this rare malignancy are discussed.

Background: Hepsin (HPN), a type II trans-membrane serine protease, has been reported to be one of the most up regulated genes in prostate cancer. The aim of the present study was to find out immuno-histochemistry based phenotypic expression of HPN in formalin fixed paraffin embedded sections of prostate cancer compared with that in benign prostatic hyperplasia, in a prospective clinical setting, to know the differential status of HPN expression in benign and malignant prostatic disease. Materials and Methods: Tissue biopsies of histologically proven cases of prostatic cancers (48), benign prostatic hyperplasia (42), benign prostatic hyperplasia with prostatic intraepithelial neoplasia (7) and 4 cases of benign prostatic hyperplasia with prostatitis, were subjected to immunohistochemical staining with HPN antibody by strepavidin biotin method. Results: Hepsin expression was 100% in prostate carcinoma, 11.9% (5/42) in benign prostatic hyperplasia, 57.14% (4/7) in benign prostatic hyperplasia with prostatic intraepithelial neoplasia (PIN), and none in benign prostatic hyperplasia with prostatitis. Hepsin staining showe higher expression in high grade tumor in comparison to low grade tumor. Conclusions: Positive immunohistochemical expression of hepsin in cent percent cases of prostate cancer cases is intriguing, underscoring the significance of hepsin gene expression in prostate cancer.

ABSTRACT

Backgrounds: Helicobacter pylori infect more than half of the global population. It is suggested to be related with gastritis, peptic ulcer disease (PUD), and gastric cancer. Aims: The aim of this present study was to evaluate proinflammatory cytokines including interleukin 1, 6, 8, 10, and thrombomodulin in H. pylori-infected patients with PUD and gastric cancer. Patients: This cross-sectional study was conducted in Taleghani Hospital on 111 patients with H. pylori infection. Materials and Methods: Patients were divided into three groups of PUD, cancer, and control (normal on endoscopy), according to the results of endoscopy. The serum levels of interleukins 1, 6, 8, and 10 and thrombomodulin was determined using enzyme-linked immunosorbent assay (ELISA) technique. H. pylori infection was diagnosed by histological examination of the endoscopic biopsy. Results: One hundred eleven patients were included in the study; 30 as PUD group, 30 as gastric cancer group, and 51 as controls. There was no significant difference between the means of IL-1 and IL-10 levels among the three groups (P = 0.744 and 0.383, respectively). IL-6, IL-8, and thrombomodulin levels were found to be statically different among the three groups (P < 0.05). The level of IL-6, IL-8, and thrombomodulin in cancer group was significantly higher than PUD and control groups (P < 0.05). Conclusion: There is a significant association between H. pylori infection and serum IL-6, IL-8, and thrombomodulin but such relation is not present between H. pylori and IL-1 and IL-10. Immunity response (IL-6, IL-8 and thrombomodulin) is more severe in cancer patient than PUD.

ABSTRACT

Sparganosis is a tissue invading helminthiasis infecting intermediate hosts, including humans. Strong immune responses are expected to occur in early phases of infection. Thus, we investigated cytokine expressions in splenic dendritic cells and in sera after experimental infection of mice. In splenic dendritic cells, TNF-alpha and IL-1beta expression peaked at week 1 and week 3 post-infection (PI), respectively, and also early phase (week 2 PI) depressed cytokine expression was noticed. Serum IL-1beta concentration increased significantly at week 2 PI and peaked at week 6 PI, and that of TNF-alpha peaked at week 6 PI. These results showed that pro-inflammatory cytokines, TNF-alpha and IL-1beta, are chronologically regulated in mouse sparganosis.

ABSTRACT

La Toxoplasmosis es una de las protozoonosis más frecuentes del ser humano. El objetivo de este estudio fue determinar la seroprevalencia de la toxoplasmosis en una población infantil del Municipio Mara, estado Zulia. Se estudiaron 119 muestras de sueros de ambos sexos (53 masculinos y 66 femeninos) con edades comprendidas entre 4-13 años. Los anticuerpos toxoplásmicos se determinaron por de la técnica de hemaglutinación indirecta. El análisis estadístico se realizó a través de la prueba de Chi cuadrado y la correlación de Spearman. La prevalencia general de infección fue de 15,9 por ciento. La frecuencia de sueros positivos fue mayor en el rango de edad de 10-13 años (8/25) con un 32,0 por ciento. Se observó que el número de casos positivos aumentó con relación al grupo de edad. No se observaron diferencias estadísticamente significativas con relación al sexo, sin embargo se apreció una mayor tendencia de positividad para el sexo femenino en el grupo de 10-13 años. La media geométrica fue de 9.479. El rango de títulos positivos fue de 1:64 a 1:2.097.512, el título más bajo fue de 1:64 con 5,26 por ciento (1/19) y el más alto 1:2.097.512 con 5,26 por ciento (1/19). Los resultados obtenidos indican que esta enfermedad es frecuente en esta población infantil.


Toxoplasmosis is one of the most frequent protozoonosis in the human being. The objective of this study was to determine toxoplasmosis seroprevalence in an infantile population from the Mara Municipality, State of Zulia. 119 serum samples from both sexes (53 male and 66 female) with ages between 4-13 years were studied. The toxoplasmic antibodies were determined by the indirect hemaglutination technique. Statistical analysis was done using the Chi square test and Spearman’s correlation. General prevalence of the infection was 15.9 percent. The positive serum frequency was greater in the 10-13 year age range (8/25) with 32.0 percent. It was observed that the number of positive cases increased in relation to the age group. No statistically significant differences related to gender were observed; nevertheless, a greater positive tendency was observed for females in the 10-13 year group. The geometric average was 9.479. The range of positive titles was from 1:64 to 1:2.097.512; the lowest title was 1:64 with 5.26 percent (1/19) and the highest was 1:2.097.512 with 5.26 percent (1/19). Results indicate that this disease is frequent in this infantile population.

ABSTRACT

Autologous serum has been used to treat dry eye syndrome for many years. It contains several growth factors, vitamins, fibronectin and other components that have been considered important for corneal and conjunctival integrity. Serum eye drops are usually prepared as an unpreserved blood solution. The serum is by nature well tolerated and its biochemical properties are somewhat similar to natural tears. Autologous serum eye drops have been reported to be effective for the treatment of severe dry eye-related ocular surface disorders (Sjõgren's syndrome), and also other entities such as superior limbic keratoconjunctivitis, graft-versus-host disease, Stevens-Johnson syndrome, ocular cicatricial pemphigoid, recurrent or persistent corneal erosions, neurotrophic keratopathy, Mooren's ulcer, aniridic keratopathy, filtering blebs after trabeculectomy, and post-keratorefractive surgery. The purpose of this study is to review the recently published literature on ocular surface diseases treated with human autologous serum eye drops.


O soro autólogo tem sido adotado como uma nova abordagem para tratar síndrome do olho seco porque contém vitaminas, alguns fatores de crecimento e fibronectina que são considerados importantes contribuintes para integridade corneana e conjuntival. Colírio de soro autólogo é produzido sem preservativo. O soro é não-alérgico e suas propriedades bioquímicas são similares à lágrima. O soro autólogo tópico tem sido relatado efetivo para o tratamento de olho seco grave relacionado a distúrbios da superfície ocular como na síndrome de Sjõgren, ceratoconjuntivite límbica superior, doença do enxerto versus hospedeiro, síndrome de Stevens-Johnson, procedimentos cerato-refrativos, erosão corneana persistente ou recorrente, ceratopatia neurotrófica, úlcera de Mooren, ceratopatia associada à aniridia, e bolhas filtrantes após trabeculectomia. O objetivo do presente estudo é revisar a literatura recentemente publicada sobre doenças da superficie ocular tratadas com soro autológo tópico.

ABSTRACT

PURPOSE: This study was conducted to investigate the presence of bcl-2 protein in the serum of patients with viral hepatitis and to find out if there is any correlation between bcl-2 protein levels and cellular oxidative stress in the pathogenesis of viral hepatitis. METHODS: This study was carried out on 130 patients with viral hepatitis, 70 with chronic hepatitis, 30 with liver cirrhosis and 30 with hepatocellular carcinoma (HCC) in addition to 20 healthy persons as the control. Serum bcl-2 protein was estimated by enzyme-linked immunosorbent assay, serum malondialdehyde (MDA), nitric oxide (NO) and antioxidant enzymes (GSH, GSH-px, GR and SOD) were measured using spectrophotometric analysis. RESULTS: bcl-2 protein level was significantly elevated in the serum of HCC, cirrhosis and chronic hepatitis groups as compared to control group. There were significant positive correlations between higher bcl-2 protein level and viral hepatitis markers (HBsAg, anti-HCV antibodies) in HCC and cirrhotic patients as compared to chronic hepatitis group. An increase in oxidative stress markers (MDA, NO) and a decrease in antioxidant enzyme activities (SOD, GSH and GSH-px) were observed. However, there was a negative correlation between bcl-2 levels and GR in all studied patient groups. CONCLUSIONS: The release of oxidative free radicals, deficiency in antioxidant enzymes and the expression of bcl-2 protein might play a role in the pathogenesis of viral hepatitis. The ability to measure bcl-2 protein in the serum could be useful as a prognostic marker of cancer patients.

ABSTRACT

To determine concentrations of certain electrolytes in cerebrospinal fluid [CSF] and serum of Khuzestan native buffaloes, samples of CSF from lumbosacral cistern and blood from jugular vein were obtained from 70 clinically-healthy male [n = 30] and female [n = 40], mature [n = 34] and immature [n = 36] animals. Samples were analyzed for sodium and potassium by flame photometry, magnesium by atomic absorption spectrophotometry, calcium by ortocresol phetalein method and phosphorous by phosphomolibdate method using an automated biochemical analyzer. Statistical analyses showed that regardless of age and sex, the mean values of sodium [139.0 +/- 0.70 mmol/1] and magnesium [1.50 +/- 0.24 mmol/1] in CSF were significantly [P<0.001] higher than their mean values in serum; 134.5 +/- 6.3 mmol/1 and 0.72 +/- 0.08 mmol/1, respectively. However, the mean values of potassium [3.47 +/- 0.67 mmol/1], calcium [2.14 +/- 0.25 mmol/1] and phosphorus [1.22 +/- 0.17 mmol/1] in CSF were significantly [P<0.001] lower than their mean values in serum; 5,34 +/- 0.92 mmol/1, 2.33 +/- 0.17 mmol/1 and 1.44 +/- 0.08 mmol/1, respectively. Moreover, in males, females, matures and immature animals, the mean values of sodium and magnesium in CSF were significantly higher than those in the serum and the mean values of potassium, calcium and phosphorus in serum were significantly higher than those in CSF [P<0.01]. Age of animals did not affect any of these electrolytes. The mean value of serum phosphorus in females was significantly higher than that in males [P<0.001]. However, the mean values for potassium, calcium and phosphorus in CSF were significantly higher in males than in females [P<0.05]

ABSTRACT

A study was carried out to assess the effect of 60 ppm and 120 ppm exposure of sulphur dioxide gas for one hour daily for 2 and 4 week on serum sodium(Na+) and potassium (K+) ion concentration in albino rat, Rattus norvegicus (Berkenhout). The present findings reveal that serum sodium (Na+) ion concentration increased significantly after 4 week exposure to 120 ppm of sulphur dioxide, while serum potassium ion (K+) concentration decreased significantly after 4 week exposure to 120 ppm of sulphur dioxide.

ABSTRACT

To prepare low-cost quality controls [QC] human serum and scientifically evaluate its advantages/disadvantages when compared with commercially available sera. The home made QC serum was prepared as per WHO recommended protocol from four healthy volunteers. It was screened for HIV, HCV and HBV, pooled together and stabilized with ethylene glycol. The initial 40 values were used for calculation of means, SDs and CVs for seventeen routinely measured analytes and results were compared with those of commercially available lyophilized human sera. The average concentrations of seventeen commonly analyzed constituents were found to be near the middle of the physiological range of healthy subjects and the home made serum could be a good substitute for the commercial serum of normal range. The narrower CVs of the analytes imply a lesser vial to vial variation in the home made sera. Additional advantages include easy preparation, no need for reconstitution and lower cost. Home made serum is a good substitute for the commercial serum of the normal range especially in developing countries like Pakistan
