ABSTRACT

Antimicrobial resistance of microorganisms and the unwanted side effects of chemoradiation therapy in cancer are major issues in healthcare. In recent times, protein-based drugs have emerged as promising candidates due to their high specificity, less side effects, etc. In this context, the rhizome of Trillium govanianum was first explored for biologically active proteins/peptides. For this, three protein fractions namely Aqueous protein fraction (APF), Hexane-Methanol-treated aqueous protein fraction (HMAPF), and Methanol-treated aqueous protein fraction (MAPF) were prepared and evaluated for antimicrobial and antiproliferative activities. In antifungal activity, HMAPF showed the lowest MIC90 values of 1.56 µg/ml against Candida parapsilosis and Candida glabrata and 3.12 µg/ml against Candida albicans and Candida auris. The antifungal activity was further confirmed by a chitinase assay, a growth kinetics and a proteinase inhibitory assay. Surprisingly, none of the three protein fractions exhibited antibacterial activity against Escherichia coli and Staphylococcus aureus. Moreover, APF exhibited potent antiproliferative and antioxidant activities with IC50 values of 18 µg/ml and 227 µg /ml, respectively. For HMAPF, an IC50 value of 70 µg/ml against the MDA-MB-231 cell line was observed. The present results demonstrate that the protein fractions, particularly HMAPF and APF, might serve as potential sources of a dual antifungal and antiproliferative protein-based drug.

ABSTRACT

Repeated exposure to pathogens leads to evolutionary selection of adaptive traits. Many species transfer immunological memory to their offspring to counteract future immune challenges. Transfer factors such as those found in the colostrum are among the many mechanisms where transfer of immunologic memory from one generation to the next can be achieved for an enhanced immune response. Here, a library of 100 plants with high protein contents was screened to find plant-based proteins that behave like a transfer factor moiety to boost human immunity. Aqueous extracts from candidate plants were tested in a human peripheral blood mononuclear cell (PBMC) cytotoxicity assay using human cancerous lymphoblast cells-with K562 cells as a target and natural killer cells as an effector. Plant extracts that caused PBMCs to exhibit enhanced killing beyond the capability of the colostrum-based transfer factor were considered hits. Primary screening yielded an 11% hit rate. The protein contents of these hits were tested via a Bradford assay and Coomassie-stained SDS-PAGE, where three extracts were confirmed to have high protein contents. Plants with high protein contents underwent C18 column fractionation using methanol gradients followed by membrane ultrafiltration to isolate protein fractions with molecular weights of <3 kDa, 3-30 kDa, and >30 kDa. It was found that the 3-30 kDa and >30 kDa fractions had high activity in the PBMC cytotoxicity assay. The 3-30 kDa ultrafiltrates from the top two hits, seeds from Raphanus sativus and Brassica juncea, were then selected for protein identification by mass spectrometry. The majority of the proteins in the fractions were found to be seed storage proteins, with a low abundance of proteins involved in plant defense and stress response. These findings suggest that Raphanus sativus or Brassica juncea extracts could be considered for further characterization and immune functional exploration with a possibility of supplemental use to bolster recipients' immune response.

ABSTRACT

Pereskia aculeata Miller is an edible plant species belonging to the Cactaceae family. It has the potential to be used in the food and pharmaceutical industries due to its nutritional characteristics, bioactive compounds, and mucilage content. Pereskia aculeata Miller is native to the Neotropical region, where it is traditionally employed as food in rural communities, being popularly known as 'ora-pro-nobis' (OPN) or the Barbados gooseberry. The leaves of OPN are distinguished by their nontoxicity and nutritional richness, including, on a dry basis, 23% proteins, 31% carbohydrates, 14% minerals, 8% lipids, and 4% soluble dietary fibers, besides vitamins A, C, and E, and phenolic, carotenoid, and flavonoid compounds. The OPN leaves and fruits also contain mucilage composed of arabinogalactan biopolymer that presents technofunctional properties such as thickener, gelling, and emulsifier agent. Moreover, OPN is generally used for pharmacological purposes in Brazilian folk medicine, which has been attributed to its bioactive molecules with metabolic, anti-inflammatory, antioxidant, and antimicrobial properties. Therefore, in the face of the growing research and industrial interests in OPN as a novel food source, the present work reviews its botanical, nutritional, bioactive, and technofunctional properties, which are relevant for the development of healthy and innovative food products and ingredients.

ABSTRACT

OBJECTIVES: The 2020 Kidney Disease Outcome Quality Initiative guidelines recommend adjusting phosphorus intake to achieve and maintain normal serum phosphorus levels for adults living with chronic kidney disease. These guidelines also recommend considering the dietary source of phosphorus as different sources have different bioavailability; however, phosphorus food lists are not provided. Therefore, the aim of this study is to investigate the current teaching materials in Canada regarding low phosphorus diet. DESIGN AND METHODS: Using a geographical approach, websites from each province and territories' government, health, and renal programs (where applicable) were reviewed for resources on dietary phosphorus restriction in chronic kidney disease. All publicly available handouts/booklets/printable webpages were obtained and reviewed for recommendations on how to implement a low phosphorus diet. RESULTS: Sixty-one resources in total met inclusion criteria (52 handouts from health agencies in 6 provinces and 9 handouts from the Kidney Foundation of Canada). Items with minimal nutrition value, such as cola, beer and cocoa, chocolate, and baking powder, were the most commonly restricted with 84% (51/61) resources making this recommendation. Plant proteins and minimally processed dairy were restricted in 80% (49/61) of resources. Processed animal meat was recommended to be restricted in 70% (43/61) of resources and whole grains in 65% (40/61). Sixty-three percent of the handouts (39/61) discuss avoiding phosphorus additives. CONCLUSIONS: Many resources restrict items with minimal nutrition value to lower phosphorus intake; however, plant foods, including plant proteins and whole grains, continue to be restricted in the majority of resources, despite having lower bioavailability. The 2020 Kidney Disease Outcome Quality Initiative guidelines recommend considering bioavailability of phosphorus source when implementing low phosphorus diets; current handouts in Canada would likely benefit from review.

ABSTRACT

To clarify the mechanism of the response of sugar beet (Beta vulgaris L.) to cadmium (Cd) stress, this study investigated changes in the phenotype, physiological indexes, and subcellular structure of B. vulgaris under Cd treatment and the transcriptional pattern of the BvHIPP24 gene (a heavy metal-associated isoprenylated plant protein involved in heavy metal detoxification). The plant height and shoot and root growth of B. vulgaris seedlings were inhibited to some extent under 0.5 and 1 mM Cd, with gradually wilting and yellowing of leaves and dark brown roots. When the Cd concentration was increased, malondialdehyde content and the activities of peroxidase, superoxide dismutase, and glutathione S-transferase increased differentially. qPCR indicated that the expression of BvHIPP24 was induced by different concentrations of Cd. Although transmission electron microscopy revealed damage to nuclei, mitochondria, and chloroplasts, B. vulgaris exhibited strong adaptability to 0.5 mM Cd according to a comprehensive analysis using the membership function. The results showed that B. vulgaris may reduce cell damage and improve its Cd tolerance by regulating functional gene expression and antioxidant enzymes. This study increases our understanding of the Cd-tolerance mechanism of B. vulgaris and provides insights into the use of B. vulgaris in Cd bioremediation.


Sugar beet is a novel energy crop with superior characteristics for both heavy metal phytoremediation and biomass energy development. This work is the first to investigate both the morphological, physiological, and ultrastructural response of sugar beet to cadmium stress and the induction of a functional metallochaperone gene by cadmium. This study explains the cadmium tolerance mechanism of sugar beet based on a comprehensive evaluation and provides an important theoretical basis for further application of beet in heavy metal bioremediation.

ABSTRACT

Ground ivy (Glechoma hederacea L.) has been known as a medicinal plant in folk medicine for generations and, as a member of the Lamiaceae family, is characterized with a high content of rosmarinic acid. The aim of the present study was to formulate delivery systems containing bioactive compounds from ground ivy in encapsulated form and incorporated into candies. Liposomes were examined as the encapsulation systems that were additionally coated with an alginate-plant protein gel to reduce leakage of the incorporated material. Bioactive characterization of the ground ivy extract showed a high content of total phenolics (1186.20 mg GAE/L) and rosmarinic acid (46.04 mg/L). The formulation of liposomes with the high encapsulation efficiency of rosmarinic acid (97.64%), with at least a double bilayer and with polydisperse particle size distribution was achieved. Alginate microparticles reinforced with rice proteins provided the highest encapsulation efficiency for rosmarinic acid (78.16%) and were therefore used for the successful coating of liposomes, as confirmed by FT-IR analysis. Coating liposomes with alginate-rice protein gel provided prolonged controlled release of rosmarinic acid during simulated gastro-intestinal digestion, and the same was noted when they were incorporated into candies.

The Expert Panel for Cosmetic Ingredient Safety (Panel) reviewed the safety of 19 plant-derived proteins and peptides, which function mainly as skin and/or hair conditioning agents in personal care products. The Panel concluded that 18 plant-derived proteins and peptides are safe as used in the present practices of use and concentration as described in this safety assessment, while the data on Hydrolyzed Maple Sycamore Protein are insufficient to determine safety.

BACKGROUND: Orange pomace polyphenols have potential for use as nutraceutical ingredients in functional foods and beverages. However, owing to their low water solubility and bioaccessibility, they are not being utilized to their full potential. The goal of this research is to assess the impact of encapsulation on hesperetin (HT - a model orange polyphenol) water solubility, antioxidant activity, and in vitro bioaccessibility. RESULTS: In this study, a citrus flavonoid aglycone, HT, was encapsulated within water-dispersible colloidal complexes (d = 350 ± 8 nm) formed by electrostatic attraction of pea protein isolate and high-methoxyl pectin at a mixing ratio of 1:1 (v/v) and pH 4. The maximum amount of HT that could be dispersed in water was much higher for the encapsulated form (99 ± 7 µg mL-1 ) than the non-encapsulated form (<10 µg mL-1 ). The radical scavenging activity of the encapsulated HT (>90%, pH 4) was much higher than the non-encapsulated form (<15% at pH 4 or 7). The in vitro bioaccessibility of encapsulated HT (27 ± 7%) was also much higher than the non-encapsulated form (<7%). CONCLUSION: These results suggest that a well-designed, biopolymer-based delivery system may improve the effective incorporation of HT, and potentially other orange pomace polyphenols, into food and beverage products. This could provide an additional high-value use for orange juicing by-products while introducing a new nutraceutical product to the food and beverage industry. © 2022 Society of Chemical Industry.

Plant-based proteins are attractive components which may serve as sustainable alternatives to current petrochemical products. Both soy protein and major corn protein, zein, are of interest in food packaging applications due to their sustainability, biodegradation properties, and inherent physicochemical properties. This study discusses the development of bioplastic materials, where it explores the effects of combining zein, soy protein, and plasticizing latexes derived from plant oil-based monomers (POBMs) on properties of resulting bioplastic films. By looking for synergistic effects of soy protein's inherent film formation ability and zein's higher strength, we prepare strong yet flexible soy-zein films as materials, called proteoposites. Incorporation of natural additive POBM-latexes helps to plasticize and hydrophobize the bioplastic films and thus to improve mechanical and barrier properties. Variation of the POBM-latexes' particle size further aims to enhance the performance of resulting bioplastic films. As a result, modified soy-zein proteoposite films with improved moisture resistance, enhanced mechanical behavior, and greater barrier properties were developed. Machine learning-based computational models were utilized in order to find main structural factors affecting the bioplastic's properties and develop a quantitative structure-property relationship model between the physicochemical properties of the film components and the resulted bioplastics' properties and performance. The developed model effectively predicts experimental outcomes with >85% (R2: 0.85) accuracy. The newly synthesized proteoposites confirmed the machine learning model predictions. As a result, proteoposite films made of two plant proteins and modified with POBM-latexes can be considered as an attractive and viable replacement for petrochemical food packaging products.

BACKGROUND: The lectin from Cratylia argentea (CFL) is able to modulate the immune system response and is thus a potential phytotherapeutic substance. HYPOTHESIS/PURPOSE: In this study, we investigated the role of CFL on control of bacterial infection caused by Listeria monocytogenes, the causative agent of human listeriosis. STUDY DESIGN: Swiss mice were infected with L. monocytogenes and then treated with CFL. METHODS: Adult Swiss mice weighing with 30-40 g were infected intraperitoneally with a bacterial suspension (0.2 ml; 1 × 107 CFU/ml). After 30 min, the mice were treated with CFL intravenously at concentrations of 0.1 or 10 mg/kg. Control mice received phosphate-buffered saline (PBS). The animals were euthanized 24 h after infection. RESULTS: We observed that i.v. administration of CFL to Swiss mice did not cause acute toxicity, and reduced the leukocyte counts in the bloodstream 24 h after infection with virulent L. monocytogenes. There was a reduction in the bacterial burden within peritoneal macrophages after infection in CFL-treated mice. Accordingly, the bacterial counts in the bloodstream, spleen and liver also decreased in comparison with the PBS group. Histological damage in the spleen and liver was lower in mice that received CFL treatment. In vitro antimicrobial assays demonstrated that CFL does not inhibit the growth of L. monocytogenes. The mRNA expression of the anti-inflammatory cytokine IL-10 was enhanced with CFL treatment after infection. CONCLUSION: The lectin from C. argentea (CFL) has immunomodulatory and anti-infective properties of pharmacological interest for control of infectious diseases.

ABSTRACT

The increasing interest of the biopharmaceutical industry to exploit plants as a commercially viable production system is demanding the development of new strategies to maximize product recovery. Aqueous two-phase systems (ATPSs) are a primary recovery technique that has shown great potential for the efficient extraction and purification of biological products, from organelles to proteins and low-molecular-weight compounds. The evaluation of different system parameters upon the partitioning behavior can provide the conditions that favor the concentration of contaminants and the desired target protein in opposite phases. The protocols described here provide the basic strategy to explore the use of ATPSs for the isolation and partial purification of native and recombinant proteins from plants and plant-derived extracts.

The Bokashi leachate (BL) is a by-product from the anaerobic digestion of food waste, following the Bokashi composting method. Bokashi leachate is acidic and it contains effective microorganisms hence it has potential to be a functional feed additive to the plant proteins based diets for fish farming. This study evaluated the growth performance and feed utilization of the red tilapia (Oreochromis sp.) fingerlings fed with the BL supplemented soybean meal (SBM) based diets. After an 8-week feeding trial, fish fed with the 5% BL supplemented SBM diet attained the highest weight gain. This result was significantly higher (p < 0.05) than those fed with the 0% BL supplemented SBM diet, and comparable (p > 0.05) to those fed with the control full fish meal (FM) diet. Generally, dietary inclusion of BL enhanced the fish feed intake on the SBM diet but it did not show clear sign of improvement in their feed utilization. In addition, no significant difference was found across the hepatosomatic index and viscerosomatic index from all dietary treatments. These outcomes concluded that dietary inclusion of BL can enhance the feed intake and growth performance of the red tilapia fingerlings fed with the SBM based diet without compromising their health, and the optimum BL inclusion level was 5%. Nevertheless, further study on the properties and substances content of the BL produced from different types and ratios of food waste is strongly recommended. In this study, BL was also discovered to be capable of reducing the crude fiber content in the SBM diets. Such observation deserves a further exploitation on the application of BL to manipulate the crude fiber content in the plant proteins based diets in fish farming.

Peanut proteins are mostly composed of arachins and conarachins, globular proteins that can form gels under thermal denaturation or enzymatic treatment. We explored here how ionic strength (0.5 M or 0.8 M) and gelation process (a thermal treatment preceded or not by an enzymatic pre-treatment) could affect peanut protein gel properties. Gel formation and final properties were characterized by rheology, and gel structure was observed by confocal microscopy. We found that the ionic strength imposed during protein extraction determines the arachins/conarachins ratio, and that conarachins-rich samples give stronger gels, which is attributed to their higher content in free thiol groups and lysine residues. The gel storage modulus exhibited a power-law dependence with the protein concentration, which exponent depended on the gelation process. Rheological results, together with confocal microscopy imaging, showed that an enzymatic pre-treatment resulted in denser structures than when a simple thermal treatment was applied.

This study aimed to assess the astaxanthin (Ax) accumulation in hepatocytes isolated from farmed Atlantic salmon fed different diets (rich marine, poor, poor with marine phospholipids (MPL) and poor with docosahexaenoic acid (DHA)). Nuclear magnetic resonance (NMR) spectroscopy was used for the Ax detection and quantification. The use of the 13C-enriched Ax allowed the assessment of short-time Ax metabolism. The substitution of fish oil and meal in fish feed on plant analogs and the addition of MPL caused further catabolism and decrease of Ax accumulation in hepatocytes from 17 to about 6 mg/kg or to almost zero in the case of DHA addition. Signals assignment of the native and 13C-enriched astaxanthin in acetone were performed using 1D and 2D NMR spectra.

ABSTRACT

The structure of the MP-4 protein was previously determined at a resolution of 2.8 Å. Owing to the unavailability of gene-sequence information at the time, the side-chain assignment was carried out on the basis of a partial sequence available through Edman degradation, sequence homology to orthologs and electron density. The structure of MP-4 has now been determined at a higher resolution (2.22 Å) in another space group and all of the structural inferences that were presented in the previous report of the structure were validated. In addition, the present data allowed an improved assignment of side chains and enabled further analysis of the MP-4 structure, and the accuracy of the assignment was confirmed by the recently available gene sequence. The study reinforces the traditional concept that conservative interpretations of relatively low-resolution structures remain correct even with the availability of high-resolution data.

ABSTRACT

Bone fractures often result in complications that require surgical intervention to promote fracture healing. Tissue engineering seeks to alleviate the need for autologous bone grafting by utilizing scaffolds that can promote bone fracture healing. Plant-derived materials are desirable biomaterials because of their biodegradability, availability, and low immunogenicity. Among various plant-derived proteins, zein, which is a corn protein, has shown promise for bone repair. However, when processed, zein is often blended with synthetic materials to improve mechanical properties and overall hydrolytic stability. In this study, pure zein was electrospun to create fibrous scaffolds and cross-linked with trimethylolpropane triglycidyl ether to improve hydrolytic stability. Scaffolds were characterized and evaluated in vitro for promoting the osteogenic differentiation of MC3T3-E1 cells, which are bone progenitor cells. Cross-linked zein scaffolds retained their uniform fiber morphologies after hydration. MC3T3-E1 cells grew and differentiated on the zein scaffolds even in the absence of induction factors, as demonstrated by increased alkaline phosphatase activity, mineralization, and early upregulation of Runx2 gene expression, a transcription factor associated with osteoblast differentiation. These studies demonstrate that stable, zein fibrous scaffolds could have potential for use in bone repair applications.

Previously we showed that, for optimum growth, micronutrient levels should be supplemented above current National Research Council (2011) recommendations for Atlantic salmon when they are fed diets formulated with low levels of marine ingredients. In the present study, the impact of graded levels (100, 200, 400%) of a micronutrient package (NP) on vertebral deformities and bone gene expression were determined in diploid and triploid salmon parr fed low marine diets. The prevalence of radiologically detectable spinal deformities decreased with increasing micronutrient supplementation in both ploidy. On average, triploids had a higher incidence of spinal deformity than diploids within a given diet. Micronutrient supplementation particularly reduced prevalence of fusion deformities in diploids and compression and reduced spacing deformities in triploids. Prevalence of affected vertebrae within each spinal region (cranial, caudal, tail and tail fin) varied significantly between diet and ploidy, and there was interaction. Prevalence of deformities was greatest in the caudal region of triploids and the impact of graded micronutrient supplementation in reducing deformities also greatest in triploids. Diet affected vertebral morphology with length:height (L:H) ratio generally increasing with level of micronutrient supplementation in both ploidy with no difference between ploidy. Increased dietary micronutrients level in diploid salmon increased the vertebral expression of several bone biomarker genes including bone morphogenetic protein 2 (bmp2), osteocalcin (ostcn), alkaline phosphatase (alp), matrix metallopeptidase 13 (mmp13), osteopontin (opn) and insulin-like growth factor 1 receptor (igf1r). In contrast, although some genes showed similar trends in triploids, vertebral gene expression was not significantly affected by dietary micronutrients level. The study confirmed earlier indications that dietary micronutrient levels should be increased in salmon fed diets with low marine ingredients and that there are differences in nutritional requirements between ploidies.

ABSTRACT

Fining treatments involve the addition of a substance or a mixture to wine, and are generally carried out in order to clarify, stabilize or modify the wine's organoleptic characteristics. Usually these fining agents will bind the target compound(s) to form insoluble aggregates that are subsequently removed from the wine. The main reasons to perform wine fining treatments are to carry out wine clarification, stabilization and to remove phenolic compounds imparting unwanted sensory characteristics on the wine, which is an operation that often relies on the use of animal proteins, such as casein, gelatin, egg and fish proteins. However, due to the allergenic potential of these animal proteins, there is an increasing interest in developing alternative solutions including the use of fining proteins extracted from plants (e.g., proteins from cereals, grape seeds, potatoes, legumes, etc.), and non-proteinaceous plant-based substances (e.g., cell wall polysaccharides and pomace materials). In this article, the state of the art alternative fining agents of plant origins are reviewed for the first time, including considerations of their organoleptic and technological effects on wine, and of the allergenic risks that they can pose for consumers.

ABSTRACT

To determine the effects of replacing fish oil (FO) with a mixture of vegetable oils (VO) in plant protein-rich diets on reproductive performance, the fatty acid profile of embryos as well as health indices of female rainbow trout (Oncorhynchus mykiss) brooders (initial mean body weight, 1.8 ± 0.1 kg), a 3-month feeding trial was conducted. Four isoproteic (ca. 42%) and isoenergetic (ca. 20 MJ/kg) diets were formulated in which 50% (FO50/VO50), 75% (FO25/VO75), and 100% (VO100) of FO were replaced by a mixture of VO, whereas the control diet (FO100) was prepared with FO as the major source of lipid. Fish fed the VO100 had the lowest fertilization (73.0 ± 2.5%), survival at eyed-embryo stage (62.5 ± 5.0%), and hatching rate (56.0 ± 4.7%) rates. Brood fish fed the FO50/VO50, FO25/VO50, and VO100 diets had higher levels of saturated and monounsaturated fatty acids in embryos in comparison with fish fed FO100 diet. The levels of docosahexaenoic acid of embryos gradually decreased during embryogenesis in all treatments, whereas the concentrations of eicosapentaenoic acid was greatly increased at hatching day (35 days after spawning). Regarding serum biochemical parameters, fish fed the VO100 diet had the highest serum glucose, cholesterol, and low-density lipoprotein levels. The results of the current study revealed that replacement of dietary FO with a mixture of VO up to 75% did not have any adverse effects on reproductive performance and health indices of O. mykiss females.

ABSTRACT

Metabolic syndrome (MS) is characterized with high prevalence, constant increase of people suffering from this disease and high rate of cardiovascular complications. The key factors, leading to the development of metabolism disorders during MS, are visceral fat mass growth and decrease in sensitivity of peripheral tissues to insulin, which are associated with disorders of carbohydrate, lipid, purine metabolism and arterial hypertension. The main results of in vivo studies of hypolipidemic properties of soy protein, rice bran protein and their enzymatic hydrolysates using laboratory rats and mice with experimentally induced or genetically associated dyslipidemia are presented in this review. The analysis of reviewed publications shows that consumption of soy protein provides body weight loss, normalizes lipid metabolism, reduces insulin resistance. The consumption of rice protein by laboratory animals, as well as soy protein, leads to decrease of serum cholesterol level and also provides steroid excretion, such as cholesterol and bile acids, with feces. Enzymatic hydrolysis of food proteins allows obtaining peptide mixtures with high biological value and improved functional properties, especially water solubility and intestinal absorption. In their turn, hypolipidemic peptides of hydrolysates can play a key role in endogenous cholesterol homeostasis by means of disturbing its micellar solubility, intestinal absorption, changing bile acids entherohepatic circulation, and also lowering the expression of some genes of proteins - mediators of lipid transport. It has been concluded, that hypolipidemic properties of obtained enzymatic hydrolysates of food proteins determine the prospects of their use in specialized food products for prevention of metabolic disorders.
