RESUMO

The Pooled Uranium Miners Analysis (PUMA) study is the largest uranium miners cohort with 119,709 miners, 4.3 million person-years at risk and 7754 lung cancer deaths. Excess relative rate (ERR) estimates for lung cancer mortality per unit of cumulative exposure to radon progeny in working level months (WLM) based on the PUMA study have been reported. The ERR/WLM was modified by attained age, time since exposure or age at exposure, and exposure rate. This pattern was found for the full PUMA cohort and the 1960 + sub-cohort, i.e., miners hired in 1960 or later with chronic low radon exposures and exposure rates. The aim of the present paper is to calculate the lifetime excess absolute risk (LEAR) of lung cancer mortality per WLM using the PUMA risk models, as well as risk models derived in previously published smaller uranium miner studies, some of which are included in PUMA. The same methods were applied for all risk models, i.e., relative risk projection up to <95 years of age, an exposure scenario of 2 WLM per year from age 18-64 years, and baseline mortality rates representing a mixed Euro-American-Asian population. Depending upon the choice of model, the estimated LEAR per WLM are 5.38 × 10-4 or 5.57 × 10-4 in the full PUMA cohort and 7.50 × 10-4 or 7.66 × 10-4 in the PUMA 1960 + sub-cohort, respectively. The LEAR per WLM estimates derived from risk models reported for previously published uranium miners studies range from 2.5 × 10-4 to 9.2 × 10-4. PUMA strengthens knowledge on the radon-related lung cancer LEAR, a useful way to translate models for policy purposes.

UNSCEAR recently recommended that future research on the lung cancer risk at low radon exposures or exposure rates should focus on more contemporary uranium miners. For this purpose, risk models in the German Wismut cohort of uranium miners were updated extending the follow-up period by 5 years to 1946-2018. The full cohort (n = 58,972) and specifically the 1960 + sub-cohort of miners first hired in 1960 or later (n = 26,764) were analyzed. The 1960 + sub-cohort is characterized by low protracted radon exposure of high quality of measurements. Internal Poisson regression was used to estimate the excess relative risk (ERR) for lung cancer per cumulative radon exposure in Working Level Months (WLM). Applying the BEIR VI exposure-age-concentration model, the ERR/100 WLM was 2.50 (95% confidence interval (CI) 0.81; 4.18) and 6.92 (95% CI < 0; 16.59) among miners with attained age < 55 years, time since exposure 5-14 years, and annual exposure rates < 0.5 WL in the full (n = 4329 lung cancer deaths) and in the 1960 + sub-cohort (n = 663 lung cancer deaths), respectively. Both ERR/WLM decreased with older attained ages, increasing time since exposure, and higher exposure rates. Findings of the 1960 + sub-cohort are in line with those from large pooled studies, and ERR/WLM are about two times higher than in the full Wismut cohort. Notably, 20-30 years after closure of the Wismut mines in 1990, the estimated fraction of lung cancer deaths attributable to occupational radon exposure is still 26% in the full Wismut cohort and 19% in the 1960 + sub-cohort, respectively. This demonstrates the need for radiation protection against radon.

This study aimed to quantify the effects of dietary inclusion of tannin-rich pomegranate peel (PP) on intake, methane and nitrogen (N) losses, and metabolic and health indicators in dairy cows. Four multiparous, late-lactating Brown Swiss dairy cows (796 kg body weight; 29 kg/d of energy corrected milk yield) were randomly allocated to 3 treatments in a randomized cyclic change-over design with 3 periods, each comprising 14 d of adaptation, 7 d of milk, urine, and feces collection, and 2 d of methane measurements. Treatments were formulated using PP that replaced on a dry matter (DM) basis 0% (control), 5%, and 10% of the basal mixed ration (BMR) consisting of corn and grass silage, alfalfa, and concentrate. Gaseous exchange of the cows was determined in open-circuit respiration chambers. Blood samples were collected on d 15 of each period. Individual feed intake as well as feces and urine excretion were quantified, and representative samples were collected for analyses of nutrients and phenol composition. Milk was analyzed for concentrations of fat, protein, lactose, milk urea N, and fatty acids. Total phenols and antioxidant capacity in milk and plasma were determined. In serum, the concentrations of urea and bilirubin as well as the activities of alanine aminotransferase (ALT), aspartate aminotransferase, glutamate dehydrogenase, alkaline phosphatase, and γ-glutamyl transferase were measured. The data were subjected to ANOVA with the Mixed procedure of SAS, with treatment and period as fixed and animal as random effects. The PP and BMR contained 218 and 3.5 g of total extractable tannins per kg DM, respectively, and thereof 203 and 3.3 g of hydrolyzable tannins. Total DM intake, energy corrected milk, and methane emission (total, yield, and intensity) were not affected by PP supplementation. The proportions of C18:2n-6 and C18:3n-3 in milk increased linearly as the amount of PP was increased in the diet. Milk urea N, blood urea N, and urinary N excretion decreased linearly with the increase in dietary PP content. Total phenols and antioxidant capacity in milk and plasma were not affected by the inclusion of PP. The activity of ALT increased in a linear manner with the inclusion of PP. In conclusion, replacing up to 10% of BMR with PP improved milk fatty acid composition and alleviated metabolic and environmental N load. However, the elevated serum ALT activity indicates an onset of liver stress even at 5% PP, requiring the development of adaptation protocols for safe inclusion of PP in ruminant diets.

Compared with maize silage- and concentrate-based diets, herbage-based diets were repeatedly shown to favourably influence the milk fatty acid (FA) profile. However, it is unclear how the herbage feeding mode (grazing vs indoor green-feeding) and conservation (fresh herbage vs hay vs silage) modify the milk FA profile. Therefore, the aim of the present experiment was to investigate the effect of different herbage utilisation methods (including herbage feeding mode and herbage conservation method) on the ruminal biohydrogenation of dietary FA and the consequences on the milk FA composition in cows of two breeds (Holstein and Montbéliarde). Concomitant effects of botanical composition and phenological stage of the herbage on milk FA profile were controlled for by harvesting barn-dried hay and silage simultaneously as first cut from the same ryegrass-dominated grassland in a semi-mountainous region. Seven weeks later, the first regrowth of the same plot was used as fresh herbage, either grazed or fed indoor (indoor green-feeding). Twenty-four Montbéliarde and 24 Holstein cows were randomly allocated to four groups of 12 cows balanced by breed, parity, and milk yield. In a free-stall barn, three groups were given ad libitum access to hay, silage, or fresh herbage, respectively. The fourth group was strip-grazing. All cows were supplemented with 3 kg DM/day of the same energy-rich concentrate. After 2 weeks of adaptation to the forage, samples of forage, concentrate, milk, blood, and rumen fluid were collected. Fatty acid composition of forages, rumen fluid, and milk was analysed by gas chromatography. Haymaking reduced total FA content of the herbage, in particular that of linoleic acid (LA) and α-linolenic acid (ALA). Still, rumen fluid lipids of hay-fed cows had the highest proportion of rumenic acid, LA, ALA, and total polyunsaturated fatty acids (PUFAs). Milk fat from hay-fed cows had the highest proportion of LA, and the apparent transfer rates from feed to milk of LA and ALA were higher in hay-fed cows than in silage-fed cows. The proportion of PUFAs was highest in milk fat from grazing and indoor green-fed Montbéliarde cows and lowest in silage-fed cows of both breeds. In conclusion, the herbage utilisation method affects the ruminal biohydrogenation of LA and ALA, whereby herbage drying particularly increases their transfer from herbage to milk.

1. Due to the increasing global demand for more sustainably produced animal protein, there is an intensive search for feeds to replace soybeans. Black soldier fly larvae (BSFL) appear to have great potential for replacing soybeans in poultry diets. The main objective of this study was to determine if the nutritional value of BSFL is superior to soybeans when feeding organic broilers, since smaller amounts of BSFL could replace the soybean content in the feed, thus saving even more resources.2. Eighty Hubbard S757, a slow growing organic broiler type, were fattened for 63 d, spending the last 49 d on one of five diets. Two soybean cake- and soybean oil-based diets (SS, SS-) were compared with three diets based on partially defatted BSFL meal and BSFL fat from two origins (AA-, AB-, BB-). Different from diet SS, diets SS-, AA-, AB-and BB- were designed with approximately 20% less lysine and methionine. Growth (n = 16), metabolisability, body nitrogen retention, carcase and meat quality (n = 8) were evaluated.3. Broilers of the insect-based feeding groups, AA- and AB-, grew similarly well compared to those of group SS. They also retained more nitrogen in the body than those fed BB- and SS-. Breast meat yield was higher with AA- and AB- than with BB- and SS-, but still lower than with SS. Dietary variations in physicochemical meat quality were of low practical relevance. Diet BB- resulted in a more yellow skin and meat. The fatty acid profile of the breast meat lipids reflected the high lauric acid proportion of the BSFL lipids, resulting in up to 80 times higher proportions than when feeding the soybean-based diets.4. The results indicate that high-quality BSFL, depending on their origin, may indeed be superior to soybean protein, but that the meat lipids from BSFL-fed broilers can contain significant amounts of lauric acid, which, from a human nutrition perspective, could have a negative impact on meat quality.

The concept of developmental programming suggests that maternal excessive energy intake during intrauterine development could permanently affect the offspring's performance. Spirulina might alleviate adverse programming effects, but currently has only been shown to affect productivity and product quality of livestock when fed directly. Therefore, we investigated effects of supplementing 20 g spirulina/day to 20 gestating and lactating sows fed either a commercial or a high energy diet (HED), on performance and meat quality of their piglets fattened for 4 months. Control and HED offspring did not differ in growth and slaughter performance. Maternal spirulina supplementation impaired growth performance in male but not in female offspring. Physicochemical meat quality was not affected by any treatment. Maternal spirulina intake tended to improve the polyunsaturated fatty acids (FA)/saturated FA ratio without affecting n-6/n-3 FA ratio in offspring meat. The present findings indicate a sex-specific programming effect for offspring growth in response to maternal spirulina but not high energy intake in pigs.

BACKGROUND: The Pooled Uranium Miners Analysis (PUMA) study draws together information from cohorts of uranium miners from Canada, the Czech Republic, France, Germany and the USA. METHODS: Vital status and cause of death were ascertained and compared with expectations based upon national mortality rates by computing standardized mortality ratios (SMRs) overall and by categories of time since first hire, calendar period of first employment and duration of employment as a miner. RESULTS: There were 51 787 deaths observed among 118 329 male miners [SMR = 1.05; 95% confidence interval (CI): 1.04, 1.06]. The SMR was elevated for all cancers (n = 16 633, SMR = 1.23; 95% CI: 1.21, 1.25), due primarily to excess mortality from cancers of the lung (n = 7756, SMR = 1.90; 95% CI: 1.86, 1.94), liver and gallbladder (n = 549, SMR = 1.15; 95% CI: 1.06, 1.25), larynx (n = 229, SMR = 1.10; 95% CI: 0.97, 1.26), stomach (n = 1058, SMR = 1.08; 95% CI: 1.02, 1.15) and pleura (n = 39, SMR = 1.06; 95% CI: 0.75, 1.44). Lung-cancer SMRs increased with duration of employment, decreased with calendar period and persisted with time since first hire. Among non-malignant causes, the SMR was elevated for external causes (n = 3362, SMR = 1.41; 95% CI: 1.36, 1.46) and respiratory diseases (n = 4508, SMR = 1.32; 95% CI: 1.28, 1.36), most notably silicosis (n = 814, SMR = 13.56; 95% CI: 12.64, 14.52), but not chronic obstructive pulmonary disease (n = 1729, SMR = 0.98; 95% CI: 0.93, 1.02). CONCLUSIONS: Whereas there are important obstacles to the ability to detect adverse effects of occupational exposures via SMR analyses, PUMA provides evidence of excess mortality among uranium miners due to a range of categories of cause of death. The persistent elevation of SMRs with time since first hire as a uranium miner underscores the importance of long-term follow-up of these workers.

The demand for protein sources alternative to soybean meal for supplementing forages low in metabolizable protein is large. The suitability of spirulina (Arthrospira platensis), a fast growing and resource-efficient blue-green microalga, as a source of metabolizable protein for dairy cows is known, but its effects on milk antioxidants and sensory properties were never investigated. Twelve cows were allocated to 2 groups and fed hay-based diets complemented with sugar beet pulp and wheat flakes in individual feeding troughs. The N content per kilogram of DM was equivalent between the 2 diets. Diet of 1 group was supplemented with 5% spirulina; the second group was supplemented with 6% soybean meal (control). After an adaptation period of 15 d, data were collected, and feed, milk, blood, and rumen fluid were sampled. Feeds were analyzed for proximate contents, and blood plasma was analyzed for total antioxidant capacity and antioxidant contents (tocopherol, phenols). Milk samples were analyzed for fatty acid profile, coagulation properties, color, and contents of fat, protein, lactose, total phenols, lipophilic vitamins, and provitamins (e.g., ß-carotene). Triangle tests were performed by a trained sensory panel on 6 homogenized and pasteurized bulk milk samples per treatment. The substitution of soybean meal by spirulina in the diet did not affect feed intake, milk yield, milk fat, protein, or lactose contents compared with the control group. However, the milk from the spirulina-fed cows had a higher content of ß-carotene (0.207 vs. 0.135 µg/mL) and was more yellow (b* index: 14.9 vs. 13.8). Similar to the spirulina lipids but far less pronounced, the milk fat from the spirulina-fed cows had a higher proportion of γ-linolenic acid (0.057 vs. 0.038% of fatty acid methyl esters) compared with milk fat from soybean meal-fed cows. Also trans-11 C18:1 (vaccenic acid) and other C18:1 trans isomers were elevated, but otherwise the fatty acid profile resembled that of cows fed the control diet. No sensory difference was found between milk from the 2 experimental groups. Furthermore, we observed no effects of substituting soybean meal by spirulina on total antioxidant capacity, α-tocopherol and total phenols in blood and milk. Effects on rumen fluid characteristics were minor. In conclusion, spirulina seems to be a promising protein source for dairy cows with certain improvements in nutritionally favorable constituents in milk and without side-effects on animal performance in the short term.

In the present study, the effects of feeding 450 g/day of rumen-protected fish oil (FO) compared to sunflower oil (SO) to Angus heifers (60 g/kg total intake) were quantified. Animal performance was not affected whereas the physico-chemical meat quality, assessed in three muscles, was slightly affected by diet. The oxidative shelf life of the perirenal fat declined with FO compared to SO. Despite the formerly shown increased n-3 fatty acid proportions of meat due to FO supplementation, a trained sensory panel identified an only slightly more intense fishy flavour in grilled steaks and beef patties from the FO compared to the SO group. In FO compared to SO patties, flavour intensity was more pronounced. The perception of off-flavours was negligible and differences between muscles were larger than between diets. In conclusion, supplementing ruminants with FO containing nutritionally beneficial n-3 fatty acids results in few side-effects on meat quality, restricted to quite faint off-flavours and a shorter fat shelf life.

Lightweight (LW) piglets from large litters display impaired growth performance compared with heavier littermates. This study investigated the growth performance and muscle development of early-weaned LW piglets (birthweight <1.2 kg) from large litters (17.3 ± 3.0 total born per litter), fed ad libitum a milk replacer supplemented with either l-carnitine (CAR) or l-arginine (ARG) from day 7 to day 28 of age. In total, 36 female and entire male Swiss Large White piglets, weaned on day 7 of age, were artificially reared in pairs in rescue decks. They were allocated to one of three dietary treatments: unsupplemented control (CON), 0.48 g l-carnitine·piglet-1 ·day-1 (CAR) or 1.20 g l-arginine·kg body weight-1 ·day-1 (ARG). Milk replacer was prepared daily in a 1:4 powder-to-water ratio and fed ad libitum. Piglets were weighed at birth and on days 7, 14, 21 and 28. Feed intake was assessed daily. Piglets were euthanized on day 28. The entire semitendinosus muscle (STM) was collected, and organs were weighed. Subsequently, the STM was divided into the light (STMl ) and dark (STMd ) portion, and contractile and metabolic traits were analysed by ATP histochemistry, enzyme activities and gene expression. No differences in growth performance, organ and STM weight and on contractile traits were found between groups. A tendency (p < .10) for an elevated lipid oxidation enzyme activity in the STMl and STMd and greater (p < .05) phosphorylation of the mammalian target of rapamycin pathway in the STMl of CAR compared with CON piglets was found. Despite these metabolic responses, the lack of effect of CAR and ARG supplementation on growth performance suggests that providing the milk replacer ad libitum in combination with added CAR and ARG is insufficient for eliciting faster growth of LW piglets.

It is still not fully understood whether and how factors such as time, age and smoking modify the relationship between lung cancer and radon at low exposures and exposure rates. Improved knowledge is necessary for the dose conversion of radon in working level month (WLM) into effective dose, as currently discussed by the International Commission on Radiological Protection (ICRP). An update of the German uranium miner cohort study (n = 58,974 men) with a 10-year extension of mortality follow-up (1946-2013) was used to further examine this issue. Internal Poisson regression was applied to estimate the excess relative risk (ERR) for lung cancer mortality per unit of cumulative radon exposure in WLM with exponential time-related effect modifiers. In the full cohort restricted to <100 WLM the estimated overall ERR/WLM was 0.006 [95% confidence interval (CI): 0.003; 0.010] based on 1,254 lung cancer deaths and 1,620,190 person-years at risk. Both age at and time since exposure turned out to be important modifiers of the ERR/WLM and were included in the final model. Here, the ERR/WLM centered on age at exposure of 30 years, and 20 years since exposure was 0.016 (95% CI: 0.008; 0.028). This value decreased statistically significantly by approximately 40% and 60% for each 10-year increase in age at exposure and time since exposure, respectively. The joint effect of smoking and radon exposure was investigated in the sub-cohort of miners hired in 1960 or later, which includes data on smoking status. The centered ERR/WLM was slightly higher for non/light smokers compared to moderate/heavy smokers (0.022 versus 0.013). The current findings provide evidence for an increased lung cancer risk at low radon exposures or exposure rates that is modified by age and time. The observed risk is lower, but statistically compatible to those of other miner studies at low exposures or exposure rates. These findings reject an additive- and support a sub- to (supra-) multiplicative interaction between smoking and radon.

Bioavailability of polyunsaturated fatty acids (PUFA) in ruminants is enhanced by their protection from ruminal biohydrogenation. Both n-3 and n-6 PUFA fulfil important physiological functions. We investigated potentially different incorporation patterns of these functional PUFA into three beef muscles with different activity characteristics. We supplemented 33 Angus heifers with rumen-protected oils characterized either by mainly C18:2 n-6 (linoleic acid (LA) in sunflower oil) or by C20:5 (eicosapentaenoic acid (EPA)) and C22:6 (docosahexaenoic acid (DHA)), both prevalent n-3 PUFA in fish oil. Contents and proportions of n-3 and n-6 PUFA of total fatty acids were elevated in the muscles of the respective diet group but they were partitioned differently into the muscles. For EPA and DHA, but not for LA, the diet effect was more distinct in the extensor carpi radialis compared to longissimus thoracis and biceps femoris. Partitioning of PUFA in metabolism could be related to muscle function. This has to be confirmed in other muscles, adipose tissues and organs.

As a result of the selection for genotypes with greater sow prolificacy, litter size increased and, concomitantly, average litter birth weight and early postnatal survival rates of low birth weight (L-BtW) offspring decreased. This study compared the impact of l-carnitine (CAR) and l-arginine (ARG) supplemented with a milk replacer and fed to L-BtW piglets born from large litters from days 7 to 28 of age on growth performance, carcass composition, organ and Semitendinosus muscle (STM) development. A total of 30 female and castrated Swiss Large White piglets weaned at 7 days of age were assigned to three milk replacer diets containing either no supplement (CON), CAR (0.40 g/piglet per day) or ARG (1.08 g/kg BW per day). Piglets were kept in pairs in rescue decks (0.54 m2). They were weighed daily and daily allowance of both, feed and ARG, was adjusted accordingly. Thus, feed allowance depended on growth. Each day, the milk replacer was prepared with water (1:4). Feed (allowance: 60 g dry matter/kg BW per day) was offered daily in six equal rations. Feed intake and feed efficiency was assessed for the pairs and apparent total tract-energy and -protein digestibility was determined from days 21 to 28 of age. On day 28, piglets were euthanized, blood samples were collected and the whole STM and organs were weighed. In STM, the size and metabolic properties of myofibers were determined. No difference in growth performance was found between dietary treatments, but piglets from the CAR group tended (P<0.10) to grow faster during the 1st experimental week and consume more feed from days 14 to 21 as compared with piglets of the CON group. A setback in growth in the last week in the CAR group coincided with the lower (P<0.05) energy and protein digestibility. Dietary treatments had no effect on STM and organ weight and myofiber size. Compared with the other groups, there were trends (P<0.10) for blood serum urea and glucose level to be greater in CAR and for non-esterified fatty acid level to be greater in ARG piglets. The greater (P<0.05) ratio of lactate dehydrogenase to either citrate synthase or ß-hydroxyacyl-CoA dehydrogenase indicated that the relative importance of the glycolytic compared with the oxidative pathway was greater in STM of CAR and ARG compared with CON piglets. These results suggest that ARG and CAR supplements were beneficial for muscle maturation whereas findings on phenotypic traits were rather unsystematic.

This article reviews opportunities for enriching the lipids of meat with n-3 fatty acids and conjugated linoleic acids (CLAs), both considered beneficial to human health. Special focus is put on feeds available and research carried out in Thailand. A differentiated consideration concerning the value of different n-3 fatty acids and isomers of CLAs is necessary. In ruminants, it is difficult to enrich the meat with n-3 fatty acids due to the extensive ruminal biohydrogenation of unsaturated fatty acids, but several possibilities to enhance the proportion of the most desired CLA isomer, rumenic acid, exist. By contrast, pork and poultry meat can be easily enriched with n-3 fatty acids. With purified CLA sources, CLAs also can be enhanced, but it is difficult to achieve this exclusively for rumenic acid. An interesting approach might consist in supplementing the CLA precursor vaccenic acid instead. Possible constraints for meat quality and in the fatty acid levels achieved are outlined.

Previous studies indicated that absolute CH4 emissions and CH4 yield might increase and that milk production efficiency might decrease with age in cattle. Both would make strategies to increase longevity in dairy cattle less attractive. These aspects were experimentally determined in Brown Swiss cattle distributed continuously across a large age range. Thirty lactating dairy cows (876-3,648 d of age) received diets consisting of hay, corn silage, and grass pellets supplemented with 0 or 5kg of concentrate per day. Twelve heifers (199-778 d of age) received hay only. Cows and heifers were members of herds subjected to the 2 different feeding regimens (with or without concentrate) for the past 10 yr. Methane emissions were measured individually for 2 d in open-circuit respiration chambers, followed by quantifying individual feed intake and milk yield over 8 d. Additional data on digestibility, rumination time, and passage time of feed of all experimental animals were available. Regression analyses were applied to evaluate effects of age and feeding regimen. Body weight, milk yield, and the hay proportion of forage dry matter intake were considered as covariates. Methane emissions per unit of intake, body weight, and milk yield were significantly related to age. Their development in the cows with age was characterized by an increase to maximum at around 2,000 d of age, followed by a decline. This response was not accompanied by corresponding age-related changes in intake, chewing activity, digesta passage time, and digestibility of organic matter, which would have explained shifts in CH4. However, fiber digestibility showed a similar change with age as methane emissions, resulting in quite stable methane emissions per unit of digestible fiber. As expected, methane emissions intensity per unit of milk produced was greater by 8% without concentrate than with concentrate, but no difference was noted in the response to age when the animals were subjected to different feeding regimens. The efficiency of milk production was only marginally influenced by age and diet, and no different response was observed for age in the 2 dietary regimens. In conclusion, life cycle analyses of milk production systems focusing on longevity should consider changing methane yields with age in addition to the variation in environmental costs for replacements of culled cows.

BACKGROUND: A determination of the risk of lung cancer at low levels of radon exposure is important for occupational radiation protection. METHODS: The risk of death from lung cancer at low radon exposure rates was investigated in the subcohort of 26,766 German uranium miners hired in 1960 or later. RESULTS: A clear association between lung cancer mortality (n=334 deaths) and cumulative exposure to radon in working level months (WLM) was found. The excess relative risk per WLM was 0.013 (95% confidence intervals: 0.007; 0.021). CONCLUSIONS: The present findings provide strong evidence for an increased lung cancer risk after long-term exposure to low radon exposure rates among Wismut miners. The results are compatible to those from residential radon studies and miner studies restricted to low levels.

AIMS: To investigate the relationship between the protonation of medium-chain fatty acids (MCFA) and their inhibitory effect on a ruminal methanogen species. METHODS AND RESULTS: Cell suspensions of Methanobrevibacter ruminantium M1 in 1 mg dry matter (DM) ml(-1) were supplemented with lauric acid (C12 ) and myristic acid (C14 ) at a concentration of 8 µg ml(-1) with different pH levels of the potassium-free buffer, where the calculated degrees of protonation of C12 and C14 varied from 0·3 to 50% and from 1 to 76% respectively. Methane formation, ATP efflux, potassium leakage and cell viability were monitored 15, 30 and 45 min after the reaction started. Declining methane formation rate, increasing ATP efflux and potassium leakage, and decreasing survival of M. ruminantium were observed with increasing degrees of protonation, i.e. with decreasing pH. CONCLUSIONS: The inhibition of methanogenesis by C12 and C14 is more efficient at a pH of 5-6 as compared to pH 7. SIGNIFICANCE AND IMPACT OF THE STUDY: Methane mitigation strategies in ruminants which use supplementation of feed with MCFA such as C12 and C14 may be more effective in a low rumen pH environment. This finding is helpful in designing diets to effectively decrease methane emissions by ruminants.

A meta-analysis was conducted using the results of 82 experiments (78 publications, 266 treatments) to investigate the importance of dietary C18 fatty acids (FA) and feeding regimen for milk C18 FA profile and apparent recovery of selected FA relative to intake of these FA or their precursors. Feeding treatments based on lipid-supplemented diets were excluded. Feeding regimens were defined as grazing [including partial and full-time grazing, at dietary concentrate proportions from 0 to 44% dry matter (DM)], forage-based indoor feeding [>65% forage of total DM intake (DMI)], and concentrate-based indoor feeding (forage DMI ≤65% of DMI). Linoleic acid (LLA), α-linolenic acid (ALA), and total C18 FA proportions in milk fat increased linearly with the respective dietary FA content in all feeding regimens tested. This effect was highest in the forage-based indoor feeding. Slopes were lowest for the grazing regimens, especially regarding ALA and the sum of all C18 FA, whereas the intercepts of the prediction equations of milk ALA and total C18 FA proportions were highest for grazing regimens. This indicates that, in grazing cows, factors other than dietary FA contents determine the C18 FA composition of the milk fat. At equal dietary LLA contents, the type of feeding regimen showed no significant effect on LLA proportion in milk fat. Milk fat proportions of rumenic acid and vaccenic acid were positively related to the sum of dietary ALA and LLA contents. Grazing regimens led to the strongest enrichment of rumenic acid and vaccenic acid in milk fat. The apparent recovery of ALA, LLA, and total C18 FA (secreted, % of intake), an estimate for transfer efficiency, decreased with increasing dietary content. This relationship followed a nonlinear decay function. When the dietary content of these FA exceeded a certain threshold (about 0.2, 0.8, and 2.8% of DM for ALA, LLA, and total C18 FA, respectively) the recovery in milk remained constant at about 5, 10, and 82% of the ingested ALA, LLA, and total C18 FA, respectively. At dietary proportions below 0.01% ALA and 1.5% total C18 FA of DM, their apparent recovery in milk fat exceeded 100%. In conclusion, a general inverse relationship between dietary C18 FA and the corresponding apparent recovery in milk fat seems to exist. Within this frame, the effect of different types of feeding regimens on the eventual milk C18 FA profile varies. Among them, grazing pasture appears to provide the most variable properties.

PURPOSE: To examine exposure-response relationships between ionizing radiation and several mortality outcomes in a subgroup of 4,054 men of the German uranium miner cohort study, who worked between 1946 and 1989 in milling facilities, but never underground or in open pit mines. METHODS: Mortality follow-up was from 1946 to 2008, accumulating 158,383 person-years at risk. Cumulative exposure to radon progeny in working level months (WLM) (mean = 8, max = 127), long-lived radionuclides from uranium ore dust in kBqh/m(3) (mean = 3.9, max = 132), external gamma radiation in mSv (mean = 26, max = 667) and silica dust was estimated by a comprehensive job-exposure matrix. Internal Poisson regression models were applied to estimate the linear excess relative risk (ERR) per unit of cumulative exposure. RESULTS: Overall, a total of 457, 717 and 111 deaths occurred from malignant cancer, cardiovascular diseases and non-malignant respiratory diseases, respectively. Uranium ore dust and silica dust were not associated with mortality from any of these disease groups. A statistically significant relationship between cumulative radon exposure and mortality from all cancers (ERR/100 WLM = 1.71; p = 0.02), primarily due to lung cancer (n = 159; ERR/100 WLM = 3.39; p = 0.05), was found. With respect to cumulative external gamma radiation, an excess of mortality of solid cancers (n = 434; ERR/Sv = 1.86; p = 0.06), primarily due to stomach cancer (n = 49, ERR/Sv = 10.0; p = 0.12), was present. CONCLUSION: The present findings show an excess mortality from lung cancer due to radon exposure and from solid cancers due to external gamma radiation in uranium millers that was not statistically significant. Exposure to uranium was not associated with any cause of death, but absorbed organ doses were estimated to be low.

The Wismut cohort is currently the largest single study on the health risks associated with occupational exposures to ionising radiation and dust accrued during activities related to uranium mining. The cohort has ∼59 000 male workers, first employed between 1946 and 1989, at the Wismut Company in Germany. The main effect is a statistically significant increase in mortality from lung cancer with both increasing cumulative radon exposure and silica dust exposure. Risks for cancers of the extrathoracic airways, all extra-pulmonary cancers and cardiovascular diseases associated with radiation exposures have been evaluated. Cohort mortality rates for some other cancer sites, stomach and liver, are statistically significantly increased in relation to the general population, but not statistically significantly related to occupational exposures. No associations between leukaemia mortality and occupational doses of ionising radiation were found.
