Despite data showing that nutritional interventions high in antioxidant/anti-inflammatory properties (anthocyanin-rich foods, such as blueberries/elderberries) may decrease risk of memory loss and cognitive decline, evidence for such effects in mild cognitive impairment (MCI) is limited. This study examined preliminary effects of American elderberry (Sambucus nigra subsp. canadensis) juice on cognition and inflammatory markers in patients with MCI. In a randomized, double-blind, placebo-controlled trial, patients with MCI (n = 24, Mage = 76.33 ± 6.95) received American elderberry (n = 11) or placebo (n = 13) juice (5 mL orally 3 times a day) for 6 months. At baseline, 3 months, and 6 months, patients completed tasks measuring global cognition, verbal memory, language, visuospatial cognitive flexibility/problem solving, and memory. A subsample (n = 12, 7 elderberry/5 placebo) provided blood samples to measure serum inflammatory markers. Multilevel models examined effects of the condition (elderberry/placebo), time (baseline/3 months/6 months), and condition by time interactions on cognition/inflammation outcomes. Attrition rates for elderberry (18%) and placebo (15%) conditions were fairly low. The dosage compliance (elderberry-97%; placebo-97%) and completion of cognitive (elderberry-88%; placebo-87%) and blood-based (elderberry-100%; placebo-100%) assessments was high. Elderberry (not placebo) trended (p = 0.09) towards faster visuospatial problem solving performance from baseline to 6 months. For the elderberry condition, there were significant or significantly trending decreases over time across several markers of low-grade peripheral inflammation, including vasorin, prenylcysteine oxidase 1, and complement Factor D. Only one inflammatory marker showed an increase over time (alpha-2-macroglobin). In contrast, for the placebo, several inflammatory marker levels increased across time (L-lactate dehydrogenase B chain, complement Factor D), with one showing deceased levels over time (L-lactate dehydrogenase A chain). Daily elderberry juice consumption in patients with MCI is feasible and well tolerated and may provide some benefit to visuospatial cognitive flexibility. Preliminary findings suggest elderberry juice may reduce low-grade inflammation compared to a placebo-control. These promising findings support the need for larger, more definitive prospective studies with longer follow-ups to better understand mechanisms of action and the clinical utility of elderberries for potentially mitigating cognitive decline.

Elderberry (Sambucus nigra L.) is a widespread deciduous shrub, of which the fruits (elderberries) are used in the food industry to produce different types of dietary supplement products. These berries have been found to show multiple bioactivities, including antidiabetic, anti-infective, antineoplastic, anti-obesity, and antioxidant activities. An elderberry extract product, Sambucol®, has also been used clinically for the treatment of viral respiratory infections. As the major components, phenolic compounds, such as simple phenolic acids, anthocyanins and other flavonoids, and tannins, show promising pharmacological effects that could account for the bioactivities observed for elderberries. Based on these components, salicylic acid and its acetate derivative, aspirin, have long been used for the treatment of different disorders. Dapagliflozin, an FDA-approved antidiabetic drug, has been developed based on the conclusions obtained from a structure-activity relationship study for a simple hydrolyzable tannin, ß-pentagalloylglucoside (ß-PGG). Thus, the present review focuses on the development of therapeutic agents from elderberries and their small-molecule secondary metabolites. It is hoped that this contribution will support future investigations on elderberries.

Polycyclic aromatic hydrocarbons (PAHs) are among the most studied organic compounds in urban environments, due to their known threat to human health. This study extends the current knowledge regarding the ability of different vegetative parts of different tree species to accumulate PAHs. Moreover, exposure intensity to PAHs in areas frequented by population susceptible to adverse health effects of air pollution is evaluated. For this, leaves and barks of Sambucus nigra (S. nigra) and Acacia melanoxylon (A. melanoxylon) were collected at urban areas in the Andean city of Quito, at seven points near hospitals and schools. A methodology, previously developed, for the extraction, purification, and quantification of PAHs associated with the leaves and bark of S. nigra was employed and also validated for leaves and bark of A. melanoxylon. The total PAH level varied from 119.65 ng g-1 DW (dry weight) to 1969.98 ng g-1 DW (dry weight) with naphthalene (Naph), fluoranthene (Flt), pyrene (Pyr), chrysene (Chry), and benzo[a]pyrene (BaP) predominating in all samples. The results indicate that the leaves and bark of tree species studied have certain abilities to bio-accumulate PAH according to their molecular weight. The leaves of S. nigra and bark of A. melanoxylon showed the highest ability to accumulate PAHs, mainly those with high and medium molecular weight, respectively. The highest incidence of light molecular weight PAHs was found in the leaves of A. melanoxylon. Furthermore, coal combustion, biomass burning, and vehicle emissions were identified as the main PAHs sources. Concentrations of PAHs associated with tree species suggest an affectation in areas frequented by populations susceptible to air pollution. This fact shows the importance of regulatory scheme to significantly improve the air quality in the city integrating a knowledge-based decision-making.

The names of plants convey information on their appearance (shape, structure, colour), taste or smell, their uses (practical, ceremonial, magical and medicinal) as well as the beliefs and convictions associated with them. Assuming that the particular features of plants, entrenched in their names, must have been important to language users for some reason, the analysis of plant names can help reconstruct traditional knowledge about plants. The author analyses the standard and dialectal names for the black elder (Sambucus nigra) in Polish, juxtaposing the plant's features revealed in its names (linguistic data) with the cultural accounts associated with the plant ("with-linguistic" data). This allows for the reconstruction of the following features of the plant: (a) the appearance of the shrub, (b) the properties of its fruit, (c) the smell of the plant, (d) the place where it grows, (e) the time of harvesting, (f) its use in folk medicine and (g) the association of the plant with impure powers and diseases. The conducted analyses show that reaching for hard "linguistic evidence" (standard and folk names) makes it possible to compile hierarchies of the characteristics of the plants described. Situating these names against the background of "with-linguistic" data leads to the conclusion that folk nomenclature and folk knowledge enrich and complement each other. The vast number of names for the black elder with different onomasiological bases, presenting different points of view, also demonstrate the relationship between the degree of lexical differentiation and the cultural meaning of the plant. The ethnolinguistic analysis of the names for the black elder (Sambucus nigra), similarly to ethnobotanical studies of folk plant names, provides insights into past and contemporary uses of the plant. Thus, it can provide a starting point for further ethnobotanical research.

The objective of this study was to determine the mineral content in the leaves and flowers of wild-grown Sambucus nigra collected from eleven different locations in Kosovo. The samples were digested in a microwave system using the wet digestion method. The minerals were determined by the application of inductively coupled plasma-atomic emission spectrometry (ICP-AES) and inductively coupled plasma-mass spectrometry (ICP-MS). A total of 31 elements were determined, 15 elements by the ICP-AES method (Al, B, Ba, Ca, Cr, Cu, Fe, K, Mg, Mn, Na, P, Sr, V, and Zn) and 16 elements by the ICP-MS method (Ag, As, Be, Bi, Cd, Co, Cs, Ga, Hg, In, Li, Ni, Pb, Rb, Tl, and U). The leaves of S. nigra show a higher content of minerals compared to the flowers, except for the flower of the sample SN-FL10, which is characterized by a high concentration of Fe, Al, Pb, Be, and Tl. The concentration of heavy metals and toxic elements (Pb, Cd, and Hg) was within the permissible concentrations according to Eur. Ph.

Elderberry, the fruit of Sambucus nigra, has become a popular inclusion in foods, beverages, supplements, and more in recent years. Although the European subspecies, S. nigra ssp. nigra, has been widely studied for its composition, particularly for phenolic and volatile profiles, other subspecies, such as the American elderberry S. nigra ssp. canadensis and the blue elderberry S. nigra ssp. cerulea, have also become contenders in the elderberry supply chain. For the first time, the composition (including micronutrients, macronutrients, organic acids, titratable acid, soluble solids, phenolic compounds, and cyanogenic glycosides) of these three subspecies of elderberry is compared, highlighting the unique qualities of each subspecies and identifying gaps in the available data on the three subspecies.

Pap smear screening is a widespread technique used to detect premalignant lesions of cervical cancer (CC); however, it lacks sensitivity, leading to identifying biomarkers that improve early diagnosis sensitivity. A characteristic of cancer is the aberrant sialylation that involves the abnormal expression of α2,6 sialic acid, a specific carbohydrate linked to glycoproteins and glycolipids on the cell surface, which has been reported in premalignant CC lesions. This work aimed to develop a method to differentiate CC cell lines and primary fibroblasts using a novel lectin-based biosensor to detect α2,6 sialic acid based on attenuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR) and chemometric. The biosensor was developed by conjugating gold nanoparticles (AuNPs) with 5 µg of Sambucus nigra (SNA) lectin as the biorecognition element. Sialic acid detection was associated with the signal amplification in the 1500-1350 cm-1 region observed by the surface-enhanced infrared absorption spectroscopy (SEIRA) effect from ATR-FTIR results. This region was further analyzed for the clustering of samples by applying principal component analysis (PCA) and confidence ellipses at a 95% interval. This work demonstrates the feasibility of employing SNA biosensors to discriminate between tumoral and non-tumoral cells, that have the potential for the early detection of premalignant lesions of CC.

Cholesterol deposition in intimal macrophages leads to foam cell formation and atherosclerosis. Reverse cholesterol transport (RCT), initiated by efflux of excess cholesterol from foam cells, counteracts atherosclerosis. However, targeting RCT by enhancing cholesterol efflux was so far accompanied by adverse hepatic lipogenesis. Here, we aimed to identify novel natural enhancers of macrophage cholesterol efflux suitable for the prevention of atherosclerosis. Plant extracts of an open-access library were screened for their capacity to increase cholesterol efflux in RAW264.7 macrophages trace-labeled with fluorescent BODIPY-cholesterol. Incremental functional validation of hits yielded two final extracts, elder (Sambucus nigra) and bitter orange (Citrus aurantium L.) that induced ATP binding cassette transporter A1 (ABCA1) expression and reduced cholesteryl ester accumulation in aggregated LDL-induced foam cells. Aqueous elder extracts were subsequently prepared in-house and both, flower and leaf extracts increased ABCA1 mRNA and protein expression in human THP-1 macrophages, while lipogenic gene expression in hepatocyte-derived cells was not induced. Chlorogenic acid isomers and the quercetin glycoside rutin were identified as the main polyphenols in elder extracts with putative biological action. In summary, elder flower and leaf extracts increase macrophage ABCA1 expression and reduce foam cell formation without adversely affecting hepatic lipogenesis.

Type 2 diabetes, characterized by insulin resistance and impaired glucose homeostasis, is commonly managed through lifestyle interventions and medications such as metformin. Although metformin is generally well-tolerated, it may cause gastrointestinal adverse effects and, in rare cases, precipitate lactic acidosis, necessitating cautious use in individuals with renal dysfunction. Additionally, concerns regarding its impact on hepatic function have led to its discontinuation in cirrhotic patients. This study explores the potential synergistic benefits of a polyphenol-rich blend containing black currant, chokeberry, and black elderberry extracts alongside metformin in managing type 2 diabetes. In vitro results highlighted distinct effects of AMPK pathway modulation, showcasing reductions in cholesterol and triglyceride levels alongside a notable enhancement in glucose uptake. The blend, when combined with metformin, significantly reduced insulin levels and fasting glucose concentrations in an in vivo model. Furthermore, hepatic analyses unveiled a modulation in cellular pathways, suggesting a potential influence on lipid metabolism, attenuation of inflammatory pathways, a decrease in cellular stress response, and antioxidant defense mechanisms, collectively implying a potential reduction in liver fat accumulation. The findings suggest a potential complementary role of polyphenols in enhancing the efficacy of metformin, possibly allowing for reduced metformin dosage and mitigating its side effects. Further clinical studies are warranted to validate these findings and establish the safety and efficacy of this nutraceutical approach in managing type 2 diabetes.

Fenpropathrin (FNP) is a man-made insecticide of to the pyrethroid class, commonly employed in agricultural and horticultural practices. However, it has a prolonged persistence in the environment. Sambucus nigra, also referred to as SN, is a botanical species recognized for its notable antioxidant characteristics. The objective of this study was to examine if SN extract could mitigate the reproductive toxicity induced by FNP in rats. A total of thirty rats were categorized into six distinct groups: a control group with no treatment, two groups getting SN extract at varying doses, a group receiving FNP, and two groups receiving both FNP and SN extract. The exposure to FNP led to a decline in the number and movement of sperm, lowered levels of testosterone, and reduced the activity of the StAR gene in the FNP group compared to the control group (p < 0.05). In addition, FNP resulted in a significant increase in malondialdehyde levels with a significant drop in GSH content compared to the control group (p < 0.05). Also, a significant increase in the expression of caspase 3. Nevertheless, the administration of SN extract alleviated these effects and reinstated spermatogenesis, thereby bringing the parameters closer to those observed in the control group. The data indicate that FNP can induce testicular harm and infertility, but SN extract can mitigate these detrimental consequences.

Anthocyanin-rich edible berries protect against diet-induced obesity in animal models. Prevention is mediated through the bidirectional relationship with the fecal microbiome, and gut-derived phenolic metabolite absorption increases with physical activity, which may influence bioactivity. The objective of this study was to test elderberry juice powder on the development of diet-induced obesity and its influence on the fecal microbiome alone or in combination with physical activity. Male C57BL/6J mice were assigned to one of four treatments, including (1) high-fat diet without wheel access; (2) high-fat diet with unlimited wheel access; (3) high-fat diet supplemented with 10% elderberry juice powder without wheel access; and (4) high-fat diet supplemented with 10% elderberry juice powder with unlimited wheel access. Body weight gain, fat pads, and whole-body fat content in mice fed elderberry juice were significantly less than in mice fed the control diet independent of wheel access. At the end of the study, active mice fed elderberry juice ate significantly more than active mice fed a control diet. There was no difference in the physical activity between active groups. Elderberry juice increasedBifidobacterium, promotedAkkermansia and Anaeroplasma, and prevented the growth of Desulfovibrio. Elderberry juice is a potent inhibitor of diet-induced obesity with action mediated by the gut microbiota.

To determine the diuretic activity of Sambucus nigra L. ssp. palmensis (Link) R. Bolli (SP). SP was evaluated in adult female Swiss mice. Urinary excretion volume was measured and the concentration of sodium, potassium, chloride, pH and specific conductance of 3 doses of aqueous extract (35.0, 52.2 and 70.0 mg kg-1) were determined. SP (70.0 mg kg-1) produced a higher urinary excretion (6.41 mL) and diuretic index (15%) than hydrochlorothiazide (HCTZ) (6.27 mL and 12%, respectively). The saluretic index indicates a lower sodium excretion than HCTZ (13%) and inversely proportional to the dose (8% -5%). The same is observed for potassium excretion (0.0172-0.0162 mEq.K+/100 g/6 h), which achieves a lower value than the control group (0.0166 mEq.K+/100 g/6 h), suggesting potassium retention. These results support the use of this plant species as a diuretic in Canarian folk medicine.


Determinar la actividad diurética de Sambucus nigra L. ssp. palmensis (Link) R. Bolli (SP). SP fue evaluada en hembras adultas de ratones Swiss. Se midió el volumen de excreción urinaria y se determinó la concentración de sodio, potasio, cloruro, el pH y la conductividad específica de 3 dosis de extracto acuoso (35,0, 52,2 y 70,0 mg kg-1). SP (70,0 mg kg-1) produjo una excreción urinaria (6,41 mL) e índice diurético (15%) superior a hidroclorotiazida (HCTZ) (6,27 mLy 12%, respectivamente). El índice salurético indica una excreción de sodio inferior a HCTZ (13%) e inversamente proporcional a la dosis (8% -5%). Lo mismo ocurre con la excreción de potasio (0,0172-0,0162 mEq.K+/100 g/6 h) que alcanza un dato inferior al del grupo control (0,0166 mEq.K+/100 g/6 h), lo que sugeriría retención de potasio. Estos resultados apoyan el uso de esta especie vegetal como diurético por la medicina popular canaria.

BACKGROUND: In fish farming, the plant extracts containing antioxidant compounds have been added to the diet for enhancing pathogen resistance. In vitro studies evaluating the antioxidant effect of herbal extracts on fish cell models have focused on ROS production and the respiratory burst mechanism. However, the effects on enzymatic antioxidant defense on salmon leukocytes have not been evaluated. This study aims to evaluate the enzymatic antioxidant defense and ROS-induced cell damage in Salmon Head Kidney-1 (SHK-1) cell line exposed to polyphenol-enriched extract from Sambucus nigra flowers. RESULTS: Firstly, the Total Reactive Antioxidant Power (TRAP) assay of elderflower polyphenol (EP) was evaluated, showing 459 and 489 times more active than gallic acid and butyl hydroxy toluene (BHT), respectively. The toxic effect of EP on salmon cells was not significant at concentrations below 120 mg/ mL and no hemolysis activity was observed between 20 and 400 mg/mL. The treatment of SHK-1 cell line with EP decreased both the lipid peroxidation and protein oxidation induced by H2O2, which could be associated with decreasing oxidative stress in the SHK-1 cells since the GSH/GSSG ratio increased when only EP was added. CONCLUSIONS: These results suggest that plant extracts enriched with polyphenols could improve the enzymatic antioxidant defense of salmon leukocytes and protect the cells against ROS-induced cell damage

RESUMEN

A variabilidade estrutural é uma característica das proteínas de venenos de serpentes, e a glicosilação é uma das principais modificações pós-traducionais que contribui para a diversificação de seus proteomas. Recentes estudos de nosso grupo demonstraram que venenos do gênero Bothrops são marcadamente definidos pelo seu conteúdo de glicoproteínas, e que a maioria das estruturas de N-glicanos dos tipos híbrido e complexo identificados em oito venenos deste gênero contêm unidades de ácido siálico. Em paralelo, em glicoproteínas do veneno de B. cotiara foi identificada a presença de uma estrutura de N-acetilglicosamina bissecada. Assim, com o objetivo de investigar a variação do conteúdo de glicoproteínas, assim como os mecanismos envolvidos na geração dos diferentes venenos de Bothrops, neste estudo foram analisados comparativamente os glicoproteomas de nove venenos do gênero Bothrops (B. atrox, B. cotiara, B. erythromelas, B. fonsecai, B. insularis, B. jararaca, B. jararacussu, B. moojeni e B. neuwiedi). As abordagens glicoproteômicas envolveram cromatografia de afinidade e ensaio de pull-down utilizando, respectivamente, as lectinas SNA (aglutinina de Sambucus nigra) e MAL I (lectina de Maackia amurensis), que mostram afinidade por unidades de ácido siálico nas posições, respectivamente, α2,6 e α2,3; e cromatografia de afinidade com a lectina PHA-E (eritroaglutinina de Phaseolus vulgaris), que reconhece N-acetilglicosamina bissecada. Ainda, eletroforese de proteínas, blot de lectina, e identificação de proteínas por espectrometria de massas foram empregadas para caracterizar os glicoproteomas. As lectinas geraram frações dos venenos enriquecidas de diferentes componentes, onde as principais classes de glicoproteínas identificadas foram metaloprotease, serinoprotease, e L-amino ácido oxidase, além de outras enzimas pouco abundantes nos venenos. Os diferentes conteúdos de proteínas reconhecidas por essas lectinas, com especificidades distintas, ressaltaram novos aspectos da variabilidade dos subproteomas de glicoproteínas desses venenos, dependendo da espécie. Ainda, considerando que metaloproteases e serinoproteases são componentes abundantes nesses venenos e fundamentais no quadro de envenenamento botrópico, e que estas enzimas contêm diversos sítios de glicosilação, o papel das unidades de ácido siálico na atividade proteolítica das mesmas foi avaliado. Assim, a remoção enzimática de ácido siálico (i) alterou o padrão de gelatinólise em zimografia da maioria dos venenos, (ii) diminuiu a atividade proteolítica de alguns venenos sobre o fibrinogênio e a atividade coagulante do plasma humano de todos os venenos, e (iii) alterou o perfil de hidrólise de proteínas plasmáticas pelo veneno de B. jararaca, indicando que este carboidrato pode desempenhar um papel na interação das proteases com seus substratos proteicos. Em contraste, o perfil da atividade amidolítica dos venenos não se alterou após a remoção de ácido siálico e incubação com o substrato Bz-Arg-pNA, indicando que ácido siálico não é essencial em N-glicanos de serinoproteases atuando sobre substratos não proteicos. Em conjunto, esses resultados expandem o conhecimento sobre a variabilidade de proteomas de venenos do gênero Bothrops e apontam a importância das cadeias de carboidratos contendo ácido siálico nas atividades enzimáticas das proteases desses venenos


Structural variability is a feature of snake venom proteins, and glycosylation is one of the main post-translational modifications that contributes to the diversification of venom proteomes. Recent studies by our group have shown that Bothrops venoms are markedly defined by their glycoprotein content, and that most hybrid and complex N-glycan structures identified in eight venoms of this genus contain sialic acid units. In parallel, the presence of a bisected N-acetylglucosamine structure was identified in B. cotiara venom glycoproteins. Thus, with the aim of investigating the variation in the content of glycoproteins, as well as the mechanisms involved in the generation of different Bothrops venoms, in this study the glycoproteomes of nine Bothrops venoms (B. atrox, B. cotiara, B. erythromelas, B. fonsecai, B. insularis, B. jararaca, B. jararacussu, B. moojeni e B. neuwiedi) were comparatively analyzed. The glycoproteomic approaches involved affinity chromatography and pulldown using, respectively, the lectins SNA (Sambucus nigra agglutinin) and MAL I (Maackia amurensis lectin), which show affinity for sialic acid units at positions, respectively, α2,6 and α2,3, and affinity chromatography with PHA-E (Phaseolus vulgaris erythroagglutinin), which recognizes bisected N-acetylglucosamine. In addition, protein electrophoresis, lectin blot, and protein identification by mass spectrometry were employed for glycoproteome characterization. The lectins generated venom fractions enriched with different components, where the main classes of glycoproteins identified were metalloprotease, serine protease, and L-amino acid oxidase, in addition to other low abundant enzymes. The different contents of proteins recognized by these lectins of distinct specificities highlighted new aspects of the variability of the glycoprotein subproteomes of these venoms, depending on the species. Furthermore, considering that metalloproteases and serine proteases are abundant components of these venoms and essential in Bothrops envenomation, and that these enzymes contain several glycosylation sites, the role of sialic acid units in their proteolytic activities was evaluated. Thus, enzymatic removal of sialic acid (i) altered the pattern of gelatinolysis in zymography of most venoms, (ii) decreased the proteolytic activity of some venoms on fibrinogen and the clotting activity of human plasma of all venoms, and (iii) altered the hydrolysis profile of plasma proteins by B. jararaca venom, indicating that this carbohydrate may play a role in the interaction of proteases with their protein substrates. In contrast, the profile of amidolytic activity of the venoms did not change after removal of sialic acid and incubation with the substrate Bz-Arg-pNA, indicating that sialic acid is not essential in N-glycans of serine proteases acting on small substrates. Together, these results expand the knowledge about the variability of proteomes of Bothrops venoms and point to the importance of carbohydrate chains containing sialic acid in the enzymatic activities of venom proteases

Introducción: Eucalyptus globulus Labill., (eucalipto), Borago officinalis L. (Borraja) y Sambucus nigra L. (saúco) son especies vegetales de amplio uso popular por sus propiedades antitusígenas y mucolíticas. Objetivo: evaluar los efectos farmacológico y toxicológico de un fitofármaco elaborado con el extracto de la mezcla de las tres plantas. Métodos: a partir de las hojas secas de cada especie, se elaboraron dos extractos fluidos de las tres plantas: E. globulus B. officinalis y S nigra, mezcladas en proporción 25:25:50, respectivamente, se empleó el método de percolación y como disolventes etanol de concentraciones al 30 y 70 por ciento. Para la evaluación del efecto mucolítico se empleó el modelo de Rojo Fenol en secreciones de ratón y se ensayaron los extractos de concentraciones alcohólicas al 30 y 70 por ciento. El estudio toxicológico se realizó al extracto de concentración alcohólica al 70 por ciento. Resultados: en la evaluación farmacológica se encontró que los extractos hidroalcohólicos al 30 y 70 por ciento presentaron efecto mucolítico, con una efectividad ligeramente superior para el extracto en alcohol al 70 por ciento. Cuando se evaluó toxicológicamente, este extracto no presentó efecto tóxico en el ensayo de toxicidad aguda a dosis límite. Conclusiones: las formulaciones elaboradas con los extractos fluidos de las plantas empleando etanol al 30 y 70 por ciento, mostraron efecto mucolítico en el modelo de rojo fenol en secreciones de ratón y con las condiciones experimentales del estudio. El extracto de las plantas en etanol al 70 por ciento no presentó efecto tóxico en el ensayo de toxicidad aguda a dosis límite(AU)


Introduction: Eucalyptus globulus Labill. (eucalyptus), Borago officinalis L. (borage) and Sambucus nigra L. (elder) are plant species commonly used in folk medicine for their antitussive and mucolytic properties. Objective: Evaluate the pharmacological and toxicological effects of a drug made with an extract obtained from a mixture of the three plants. Methods: Two fluid extracts were obtained from dry leaves of the three species, E. globulus, B. officinalis and S. nigra, mixed in a 25:25:50 proportion. The method used was percolation and the solvents were 30 percent and 70 percent ethanol. The mucolytic effect was evaluated with the phenol red model in mouse secretions, and assays were conducted of the 30 percent and 70 percent alcoholic extracts. Toxicological analysis was performed on the 70 percent alcoholic extract. Results: Pharmacological evaluation found that 30 percent and 70 percent hydroalcoholic extracts display a mucolytic effect, effectiveness being slightly higher in the 70 percent alcoholic extract. When evaluated toxicologically, this extract did not show any toxic effect in the dose-limiting acute toxicity test. Conclusions: The formulations developed with fluid plant extracts using 30 percent and 70 percent ethanol were found to have a mucolytic effect in the red phenol model in mouse secretions under experimental test conditions. RESUMO O presente trabalho teve como objetivo realizar a triagem fitoquímica e avaliar a atividade antibacteriana de extratos das flores de Sambucus nigraL. Os extratos; aquoso (10 %), etanólico (5 %) e Acetato de etila (5 %) foram submetidos a testes colorimétricos para triagem fitoquímica e a avaliação da atividade antibacteriana foi realizada pelo método de disco-difusão em ágar. Os resultados mostraram que nas concentrações de 6 e 12 mg o extrato aquoso apresentou halos significativos de inibição para Staphylococcus aureus, Pseudomonas aeruginosa e Streptococcus pyogenes, porém, quando comparado aos medicamentos usados como referência a atividade não foi satisfatória, e, ainda, evidenciou a ausência de inibição para todas as cepas testadas com o aumento da concentração para 18 e 24 mg. A análise da triagem fitoquímica evidenciou a presença de flavonoides com intensa reação de cor no extrato aquoso e etanólico, e de fraca intensidade no extrato acetato de etila. Nos mesmos extratos, pelos testes realizados, não foram detectados taninos, saponinas, antraquinonas e alcaloides. Concluiu-se que o extrato aquoso apresentou melhor efeito inibitório para Staphylococcus aureus, Pseudomonas aeruginosa e Streptococcus pyogenes, porém insuficiente para promover a inativação eficiente quando comparado aos controles.


ABSTRACT This study aimed to perform a phytochemical screening and to evaluate the antibacterial activity of the extracts of Sambucusnigra L. flowers. The aqueous (10%), ethanolic (5%) and ethyl acetate (5%) extracts were subjected to colorimetric tests for phytochemical screening and the antibacterial activity evaluation was performed by the disk-diffusion method in agar. The results showed that in the 6 and 12 mg concentrations the aqueous extract presented significant inhibition halos for Staphylococcus aureus, Pseudomonas aeruginosa and Streptococcus pyogenes, but when compared with the medicines used as a reference, the activity was not satisfactory and, besides, it indicated the absence of inhibition for all the tested strains with the concentration increase of 18 and 24mg. The phytochemical screening analysis showed the presence of flavonoids with intense color reaction in the aqueous and ethanol extracts, and of low intensity in the ethyl acetate sample. In the same extracts, the tests did not detect tannins, saponins, alkaloids and anthraquinones. It was concluded that the aqueous extract showed better inhibitory effect on the Staphylococcus aureus, Pseudomonas aeruginosa and Streptococcus pyogenes, but not enough to promote an effective inactivation when compared with the medicine tests.

Toxoplasma gondii is an obligatory intracellular parasite that infects a wide range of warm-blooded animals and humans. Considering the severity of toxoplasmosis, side effects of current treatments, and the contribution of the ethnopharmacological knowledge for the treatment of parasitic infections, the aim of the present study was to investigate the efficacy of methanolic extracts from the fruits and leaves of Sambucus nigra against tachyzoite of T. gondii. For this, fruits and leaves of S. nigra were collected from Mazandaran province, Iran, were dried under the shade, and powdered using a commercial electrical blender. For extractions, methanol was used as solvent. Virulent RH strain of T. gondii was maintained in mice and macrophages containing tachyzoites were aspirated from the peritoneal cavity. Four concentrations (5, 10, 25 and 50mg/mL) of S. nigra extract were incubated with infected macrophages for 30, 60, 120 and 180 minutes and the viability of the tachyzoites were evaluated by trypan blue staining. Results showed that S. nigra fruit extracts at the concentrations of 5 and 10mg/mL killed 100% of T. gondii tachyzoites after 60 and 120 minutes, respectively; and concentrations of 25 and 50mg/mL killed 100% of the tachyzoites after 30 minutes. Additionally, extract of S. nigra leaves, at the concentrations of 5, 10 and 25mg/mL after 180 minutes, and concentration of 50mg/mL after 60 minutes, resulted with the highest efficacy. Our results showed that S. nigra has acceptable efficacy in vitro and the parasiticidal effect of fruit extract was significantly better than leaf extract. Toxoplasma gondii es un parásito intracelular obligatorio que infecta a una gran variedad de animales y seres humanos. Teniendo en cuenta la gravedad de la toxoplasmosis, los efectos secundarios de los tratamientos actuales, y la contribución de los conocimientos etnofarmacológicos para el tratamiento de infecciones parasitarias, el objetivo del presente estudio fue investigar la eficacia de los extractos metanólicos de los frutos y hojas de Sambucus nigra contra el taquizoito de T. gondii. Para esto, se recogieron frutos y hojas de S. nigra en la provincia de Mazandaran, Irán, se secaron a la sombra, y se pulverizaron con una batidora eléctrica comercial. Para las extracciones, se empleó metanol como disolvente. La cepa virulenta RH de T. gondii se mantuvo en ratones y los macrófagos con taquizoitos se aspiraron de la cavidad peritoneal. Cuatro concentraciones (5, 10, 25 y 50mg/ mL) de extracto de S. nigra se encubaron con los macrófagos infectados durante 30, 60, 120 y 180 minutos y la viabilidad de los taquizoitos se evaluó mediante tinción con azul de tripano. Los resultados mostraron que los extractos de frutos de S. nigra en las concentraciones de 5 y 10mg/mL mataron al 100% de los taquizoitos de T. gondii después de 60 y 120 minutos, respectivamente; y las concentraciones de 25 y 50mg/mL mataron al 100% de los taquizoitos después de 30 minutos. Además, el extracto de hojas de S. nigra, en concentraciones de 5, 10 y 25mg/mL después de 180 minutos, y una concentración de 50mg/mL después de 60 minutos, resultaron más eficientes. Nuestros resultados mostraron que S. nigra tiene una eficacia aceptable in vitro y el efecto parasiticida del extracto de frutos fue significativamente mejor que el del extracto de hoja. Sin embargo, la eficacia in vivo de este extracto necesita más investigación.

Se ha realizado un studio etnobotánico preliminar en la vertiente sur de la sierra de la Albera, situada al noroeste de Cataluña, y noroeste de la comarca del Alto Ampurdán (Gerona). Esta zona cubre 277 km2 y tiene una población de 12.452 habitantes. Para ello, se ha entrevistado a 18 personas, se han identificado las plantas reportadas y se han analizados los resultados en comparación con los de otros territorios. Se ha recogido la información etnobotánica sobre 155 especies de plantas pertenecientes a 67 familias, con 655 informes de uso de los cuales 7 son de elementos inorgánicos o de origen animal. Las especies más veces citadas han sido: Thymus vulgaris, Rosmarinus officinalis, Sambucus nigra y Urtica dioica y las familias más citadas han sido las Compuestas y las Labiadas; las enfermedades con más usos reportados han sido las relacionadas con el aparato digestivo (AU)


Foi efectuado um estudo introductorio etnobotánico no sul da Serra de Albera, localizad no noroeste da Catalunha e regiao nordeste do Alto Ampurdán (Girona). A área abrange 277 km2, com uma populaçao de 15.452 habitantes. Foram entrevistadas 18 pessoas, identificaram-se as plantas utilizadas, e compararam-se os resultados com os de outros territórios. Foram recolhidas informaçoes etnobotánicas sobre 155 espécies pertencentes a 67 famílias, com 655 relatórios de uso dos quais 7 correspondem a productos inorgánicos ou animais. As espécies mais citadas foram Thymus vulgaris, Rosmarinus officinalis, Sambucus nigra e Urtica dioica, sendo as famílias as famílias Compositae e Labiatae as mais citadas; as doenças com mais utilizaçoes relatadas foram as gastrointestinais (AU)


An ethnobotanical introductory study has been conducted in Southern Sierra de la Albera, located in the northwest of Catalonia and northeastern región of Alto Ampurdán (Gerona). The area covers 277 km2 with a population of 12.452 inhabitants. We have interviewed 18 people, identifying the plants reported, and we have compared results with those of other territories. Information on 155 species belonging to 67 botanical families has been collected with 655 use reports of which 7 correspond to inorganic or animal products. The most cited species were Thymus vulgaris, Rosmarinus officinalis, Sambucus nigra and Urtica dioica with Compositae and Labiatae the most cited families. Gastrointestinal diseases were those with more reported uses (AU)

RESUMEN

El objetivo de la monografía consistió en la caracterización fármaco-toxicológica de Sambucus nigra subsp. canadensis (L) R Bolli, planta tradicional cubana con actividad antiinflamatoria, entre otras. Se presentan aspectos farmacológicos como composición fitoquímica, propiedades atribuidas popularmente y comprobadas científicamente, posología, reacciones adversas, advertencias, interacciones, toxicidad, precauciones y contraindicaciones. Se concluye que con la monografía fármaco-toxicológica de esta planta medicinal, se dispone de una completa información, como documentación indispensable para actualizar las bases de datos nacionales e internacionales al respecto, con vistas a desarrollar nuevos fitofármacos, en especial antiinflamatorios con bajo potencial de efectos adversos


The objective of the monograph was the pharmacological and toxicological characterization of Sambucus nigra subspecies canadensis(L) R Bolli, a Cuban traditional plant with anti-inflammatory action, among others. Several pharmacological aspects such as phytochemical composition; scientifically tested properties attributed by the population; posology; adverse reactions; warnings; interactions; toxicity; precautions and counterindications were presented. It was concluded that the pharmaceutical and toxicological monograph of this plant provides full information as indispensable documentation to update national and international databases, with a view to developing new pharmaceuticals, particularly anti-inflammatory drugs with low adverse effect potential

La presente investigación se enfoca en el estudio de la planta o arbusto natural llamada "saúco". Su nombre científico es Sambucus nigra. Su nombre común es saúco, su nombre en varias idiomas, en inglés Eldelberry, en el idioma maya Ixil, de Guatemala, B'ajmam. Es originario del centro de Europa, Asia y África, importada por los colonizadores a América para su cultivo y producción. Esta investigación se basa en un orden sistemático que describe con detalles, pasos que requiere toda investigación científica, además manifiesta la forma de cómo se prepara, los beneficios y bondades que aporta la planta para el hombre a modo de medicina y como alimenticio; es refugio también los mismos beneficios para algunos animales, por ejemplo aves, lombrices y arañas. El propósito de realizarla es registrar y educar a personas interesadas en conocerla a valorarla; beneficios que aporta a la salud de las personas y al medio ambiente a través de la elaboración de una guía didáctica para docentes del nivel básico, para consulta y enseñanza-aprendizaje de su contenido en los establecimientos educativos, especialmente para el Colegio Ixil de Educación Básica Integral Qánil, Nebaj, Quiché. Las personas idóneas que usan esta planta llamada saúco que aprendieron el oficio de manera empírica a través de la observación, la tradición oral, y de sus propias experiencias, para curar enfermos son: terapeutas mayas, sacerdotes mayas, comadronas, curanderos y sopladores ellos tienen los conocimientos previos de cómo prepararlo y aplicarlo en: infusiones, bebido como té para resfriado, tos, bronquitis y catarro; digestivas como cólicos, diarreas, ácido úrico, gota. Otras formas de aplicación son en cataplasmas, compresas, vahos, que sirve para aliviar síntomas de enfermedades respiratorias reumatismo, artritis en el organismo de las personas.
