BACKGROUND Non-Hodgkin lymphoma is the most common hematological malignancy in the world. Diffuse large B-cell lymphoma the most common type and the cutaneous involvement due to this neoplasm is rare. Some risk factors, such as exposure to pesticides, alcohol consumption, and tobacco use, are well established. Over the past 10 years, the association between clozapine, which is the criterion standard treatment for refractory schizophrenia, and hematological malignancies has been described. CASE REPORT We report a case of a 44-year-old woman diagnosed with schizophrenia 31 years previously, who had been taking clozapine since 2009, presenting with diffuse cutaneous nodules and subcutaneous masses accompanied by asthenia, dry cough, and a weight loss of 12 kg. Computed tomography revealed multiple enlarged lymph nodes on both sides of the diaphragm, in addition to multiple large subcutaneous masses located on the right flank and on the right upper lateral chest wall (infra-axillary), homogeneous splenomegaly, and heterogeneous nodular areas of hypoenhancement, poorly delimited, in both kidneys. Diffuse large B-cell lymphoma in an advanced stage (IVBX) was diagnosed by skin biopsy, with extranodal involvement. Chemotherapy with R-CHOP (rituximab, cyclophosphamide, doxorubicin, vincristine, and prednisone every 21 days) was performed and the decision to maintain clozapine was made. At 1.5 years after the initial diagnosis the patient presented with relapse of the disease and is in follow-up. CONCLUSIONS This case report suggests the potential association between clozapine and an increased risk of lymphoma, with a few case reports in the literature reinforcing this association. Additional studies are required to either confirm or dismiss this association, and new guidelines are needed to define the safety and monitoring of the long-term usage of clozapine.

The limited options for bone repair have led to an extensive research of the field and the development of alloplastic and xenogeneic grafts. The purpose of this study was to evaluate bone repair with two bone substitutes: deproteinized bovine bone (DBB) and biphasic calcium phosphate ceramic (BCP) in critical-size defect. A total of 8-mm defects were made in the parietal bones of rabbits (n=12). The animals were divided into three experimental groups: sham (defect filled with a blood clot), DBB (defect filled with DBB), and BCP (defect filled with BCP). After the experimental periods of 15 and 45 days, the animals were euthanized and submitted to histomorphometric analysis. The total defect area, mineralized tissue area, biomaterial area, and soft tissue area were evaluated. A greater amount of immature bone tissue and biomaterial particles were observed in the BCP group compared to DBB and sham at 45 days (p<0.05). There was no difference in the qualitative pattern of bone deposition between DBB and BCP. However, the sham group did not show osteoid islands along with the defect, presenting a greater amount of collagen fibers as well in relation to the DBB and BCP groups. There was a greater number of inflammatory cells in the DBB at 45 days compared to BCP and sham groups. In conclusion, BCP and DBB are options for optimizing the use of bone grafts for maxillofacial rehabilitation. Bone defects treated with BCP showed greater deposition of bone tissue at 45 days.

INTRODUCTION: Psoriatic arthritis (PsA) is a multifaceted inflammatory disease that can cause joint destruction and impair quality of life. The Psoriatic Arthritis Quality of Life Questionnaire (PsAQoL) was the first disease-specific tool for determining the impact of the disease on the quality of life of people with PsA. OBJECTIVES: The primary objective was to develop and validate a Brazilian Portuguese version of the PsAQoL. METHODS: The UK PsAQoL was translated into Brazilian Portuguese using two translation panels. This translation then checked for face validity and construct validity with new samples of patients. Finally, a test-retest validation study was conducted with 52 patients with PsA. The survey included the Nottingham Health Profile (NHP) as a comparator instrument. RESULTS: Internal consistency and reproducibility were both excellent for the new adaptation (0.91 and 0.90 respectively Scores on the PsAQoL were found to correlate as expected with the comparator measure and the instrument was able to detect differences in score related to perceived severity of PsA, general health status and presence of a flare. CONCLUSION: The Brazilian PsAQoL was found easy to understand and complete and has excellent reliability and construct validity. The new measure will be a valuable new tool for use in routine PsA practice and clinical trials.

Resumo As opções limitadas para reparo ósseo levaram ao desenvolvimento de abrangente pesquisa na área de enxertos aloplásticos e xenogênicos. O objetivo deste estudo foi avaliar o reparo ósseo com dois substitutos ósseos: osso bovino desproteinizado (DBB) e cerâmica fosfática de cálcio bifásica (BCP) em defeito de tamanho crítico. Material e métodos: defeitos críticos de 8 mm foram feitos nos ossos parietais de coelhos (n=12). Os animais foram divididos em três grupos experimentais: sham (defeito preenchido com coágulo sanguíneo), DBB (defeito preenchido com DBB) e BCP (defeito preenchido com BCP). Após os períodos experimentais de 15 e 45 dias, os animais foram sacrificados e submetidos à análise histomorfométrica. Foram avaliadas a área total de defeitos, área de tecidos mineralizados, área de biomateriais e área de tecidos moles. Resultados: maior quantidade de tecido ósseo imaturo e de partículas de biomaterial foram observados no grupo BCP em comparação aos grupos DBB e sham aos 45 dias (p<0,05). Não houve diferença no padrão qualitativo de deposição óssea entre DBB e BCP. Ainda, o grupo sham não apresentou ilhas osteóides ao longo do defeito, apresentando maior quantidade de fibras colágenas em relação aos grupos DBB e BCP. Houve maior quantidade de células inflamatórias no DBB aos 45 dias em comparação aos grupos BCP e sham. Conclusões: BCP e DBB são opções para otimizar o uso de enxertos ósseos na reabilitação de pacientes. Defeitos ósseos tratados com BCP mostraram maior deposição de tecido ósseo aos 45 dias.

INTRODUCTION: Since the first reports of coronavirus disease 2019 (COVID-19) in December 2019, the disease has spread worldwide. Different social isolation strategies have been adopted to reduce community transmission, but few studies have evaluated the pattern of SARS-CoV-2 (severe acute respiratory syndrome coronavirus 2) infection in a family cluster during periods of isolation. We report an outbreak in 24 members of a family cluster during a period of social distancing. METHODOLOGY: We carried out an observational descriptive study of a family cluster infected with SARS-CoV-2 in Pernambuco, Northeast Brazil. Laboratory confirmation included RT-PCR of nasopharyngeal samples or IgM or IgG serology. RESULTS: The attack rates were 75% (19/24) based on laboratory-confirmed cases and 87.5% (21/24) including probable cases. The time of spread was 17 days from the first case. All patients had mild symptoms, requiring no hospitalization, and none of them died. The frequency of symptomatic, laboratory-confirmed patients was higher among adults (94%) than among children (50%); the paediatric age group also had a higher frequency of exposed individuals who remained negative for infection. Ground-glass opacities on chest computed tomography were present in all patients with reported dyspnoea. CONCLUSION: This study highlights a high risk of intrahousehold transmission from an index case, suggesting the need for (I) specific guidelines during periods of social distancing, (II) minimization of external exposures and, above all, (III) adoption of strict quarantine measures for suspected cases and family members to prevent outbreaks from spreading.

Although primary infection has been shown to prevent reinfection of SARS-CoV-2 (severe acute respiratory syndrome coronavirus 2) in animal models, gaps in the understanding of the immune response to the virus have not been adequately addressed, and some cases of possible reinfection have been reported; however, the frequency, relevance and proof of these events have yet to be determined. We report cases of two doctors who had two episodes of COVID-19 with positive RT-PCR (reverse transcriptase polymerase chain reaction) test results, raising the probability of reinfection. Case 1 was a 40-year-old male physician who presented fever and respiratory symptoms on April 10, with a positive RT-PCR test for SARS-CoV-2, with complete improvement of symptoms in five days. After 44 days, the patient presented the same symptoms of the previous episode, associated with anosmia and dysgeusia. The results of a new RT-PCR test performed two days later were positive for SARS-CoV-2. Case 2 was a 44-year-old female physician who worked in a reference clinic for COVID-19 (coronavirus disease 2019) and had onset of symptoms indicative of the disease on April 30. The RT-PCR test was positive for SARS-CoV-2, with improvement of symptoms in six days. On May 24, the patient presented fever, cough, and sore throat accompanied by headache, asthenia, myalgia, and diarrhea, and in this new episode, anosmia and dysgeusia were also present. A new RT-PCR test from nasopharyngeal swabs was performed with a positive result. Our two patients described here and other patients with possible reinfection who are now being observed in clinical practice reinforce the need to expand the investigation. Then, if the risk of reinfection is confirmed, these findings will be relevant from a clinical-epidemiological perspective to define isolation strategies and develop vaccines.

Os instrumentos de qualidade de vida relacionada à saúde bucal estão sendo usados com crescente frequência em pesquisas. No entanto, esses instrumentos não estão disponíveis em todos os países ou idiomas. A disponibilidade de versões multilíngues de instrumentos, válidas para várias culturas, é importante para a pesquisa epidemiológica. O objetivo deste estudo foi o de traduzir e realizar a adaptação transcultural do questionário desenvolvido por Feldmann em 2007, relativo às percepções de pacientes adolescentes sobre o tratamento ortodôntico, para a língua portuguesa. Inicialmente, a equivalência de conceitos e itens foi discutida por um grupo de especialistas que examinou os itens do questionário. Foram entrevistados quatro adolescentes utilizando a metodologia de grupo focal. O instrumento foi então submetido às seguintes etapas: duas traduções para o português por tradutores independentes, pré-teste dos questionários em 6 pacientes, tradução reversa, revisão por comitê e finalmente um estudo piloto com a versão final. Nessa etapa, foram entrevistados 20 adolescentes com idade entre 12 e 17 anos, de ambos os gêneros, avaliados na Clínica de Ortodontia da Faculdade de Odontologia da Universidade Estadual do Rio de Janeiro. O questionário foi aplicado a dois grupos: adolescentes não tratados e adolescentes em tratamento ortodôntico. A partir dessas avaliações, foi produzida a versão brasileira do questionário de Feldmann, com aplicação e formato apropriados à população alvo. A tradução e adaptação transcultural do questionário para o português foi realizada assegurando sua equivalência semântica e conceitual com o instrumento original e a versão final obteve consistência em estudo piloto.


Introduction: Instruments of quality of life related to oral health are used with increasing frequency in research. However, these instruments are not available in all countries or languages. The availability of multilingual versions of instruments, valid for different cultures, is important for epidemiological research. Objective: To translate and carry out the cross-cultural adaptation of the questionnaire developed by Feldmann in 2007, concerning the perceptions of adolescent patients about orthodontic treatment, into Portuguese. Methods: Initially, the equivalence of concepts and items was discussed by a group of experts who examined the items in the questionnaire. Four adolescents were interviewed using the focus group methodology. The instrument was then submitted to the following steps: two translations into Portuguese by independent translators, pre-testing of the questionnaires in 6 patients, reverse translation, review by committee and finally a pilot study with the final version. In this stage, 20 adolescents aged between 12 and 17 years, of both genders, were interviewed, evaluated at the Orthodontics Clinic of the Faculty of Dentistry of the State University of Rio de Janeiro. The questionnaire was applied to two groups: untreated adolescents and adolescents undergoing orthodontic treatment. Results: From these evaluations, the Brazilian version of the Feldmann questionnaire was produced, with administration and format appropriate to the target population. Conclusions: The translation and cross-cultural adaptation of the questionnaire into Portuguese was performed ensuring its semantic and conceptual equivalence with the original instrument and the final version was consistent in a pilot study

Since the emergence of the chikungunya virus in Brazil in 2014, more than 700,000 cases have been reported throughout the country, corresponding to one-third of all cases reported in the Americas. In addition to its high attack rates, resulting in hundreds of thousands of cases, the disease has high chronicity rates with persistent joint manifestations for more than 3 months, which can spread to more than half of the patients affected in the acute phase. Pain associated with musculoskeletal manifestations, often disabling, has an effect on patients' quality of life at different stages of the disease. Currently, the challenge faced by specialists is identifying the best therapy to be instituted for symptom relief despite the limited number of published intervention studies. In 2016, a multidisciplinary group published pharmacological treatment protocols for pain in patients with chikungunya, which was incorporated into the guidelines for clinical management of the Brazilian Ministry of Health in 2017; in that same year, a consensus was published by the Brazilian Society of Rheumatology about diagnosis and treatment. After 5 years of experience with chikungunya epidemics, in 2019, specialists involved in the protocols of the Brazilian Society of Rheumatology and Brazilian Ministry of Health prepared an update with the main objective of developing flowcharts for the therapeutic approach of musculoskeletal manifestations in adult patients to enable specialists at different levels of healthcare to spread and apply this guideline in a systematic and simplified manner.

Chikungunya fever (CHIK) has caused important epidemic outbreaks in the Americas, with musculoskeletal involvement being the main manifestation, causing chronic symptoms in half of the affected patients. This study was performed to evaluate the clinical course of the infection in 168 patients with autoimmune inflammatory disease using biological disease-modifying antirheumatic drugs (bDMARDs), comparing this group with 56 household controls. Anti-CHIKV IgG serology was positive in 42 (25%) of the patients in the bDMARD group and in 15 (27%) of the controls (p=0.79). Of those with positive serology, 32 (76%) and 14 (93%) were symptomatic among subjects in the bDMARD and control groups, respectively (p=0.87). Persistence of musculoskeletal symptoms for more than three months occurred in 64% of the patients in the control group and only in 28% in the bDMARD group (p=0.021), maintaining a statistically significant difference only for users of anti-TNF. This study found that patients affected by chikungunya fever using bDMARDs did not present severe forms or complications in the acute phase of the disease, and patients using anti-TNF biologicals had a lower frequency of chronic joint symptoms than the household controls. This favorable outcome may be related to the cytokine blockade, with a reduction in the inflammatory response and joint damage.

We demonstrate for the first time that an ultra-broadband 7 femtosecond (fs) few-cycle laser can be used for multicolor nonlinear imaging in a single channel detection geometry, when employing a time-resolved fluorescence detection scheme. On a multi-chromophore-labelled cell sample we show that the few-cycle laser can efficiently excite the multiple chromophores over a >400 nm two-photon absorption range. By combining the few-cycle laser excitation with time-correlated single-photon counting (TCSPC) detection to record two-photon fluorescence lifetime imaging microscopy (FLIM) images, the localization of different chromophores in the cell can be identified based on their fluorescence decay properties. The novel SyncRGB-FLIM multi-color bioimaging technique opens the possibility of real-time protein-protein interaction studies, where its single-scan operation translates into reduced laser exposure of the sample, resulting in more photoprotective conditions for biological specimens.

Abstract Anamnesis: A one-year-old female Yorkshire terrier, weighing 2.2 kg, was referred for evaluation of an abnormal structure in the inguinal region. Clinical and laboratory findings: Upon examination, the structure -similar to a penis- was found protruding from the preputial orifice. Physical, laboratory, ultrasound, and radiographic examinations were performed. The initial suspicion was a case of sexual development disorder, so the dog was referred for exploratory laparotomy and karyotyping. Treatment approach: During the surgical procedure, two macroscopically similar structures were found at the caudal pole of the left and right kidneys. These tissues were attached to two structures similar to the uterine horns, caudally fused to a body in the pelvic cavity. All the tissue was removed and submitted for histopathological examination, revealing the presence of typical seminiferous tubules (testis) and proliferative endometrial glands (uterine tubes). Subsequently, a surgical correction procedure was performed to cover the penile tissue and avoid damage by exposure and contact. Conclusion: This case reports unique information on diagnosis and therapeutic approach in a Yorkshire terrier with persistent Müllerian duct syndrome (PMDS). Because they are unusual, sexual development disorders are challenging in terms of diagnostic and therapy, making this report a valuable tool for veterinarians.


Resumen Anamnesis: Una hembra Yorkshire terrier de un año de edad y 2,2 kg de peso fue remitida para evaluación de una estructura anormal en la región inguinal. Hallazgos clínicos y de laboratorio: Tras el examen, se encontró una estructura similar a un pene sobresaliendo del orificio prepucial. Se realizó examen clínico, paraclínico, ecografía y radiografía abdominal. El diagnóstico presuntivo inicial fue trastorno del desarrollo sexual, por lo que la perra fue remitida para laparotomía exploratoria y cariotipaje. Abordaje terapéutico: Durante el procedimiento quirúrgico se encontraron dos estructuras macroscópicas similares en la región caudal de ambos riñones. Estos tejidos se unían a dos estructuras similares a los cuernos uterinos, caudalmente fusionados a un cuerpo en la cavidad pélvica. Se retiró todo el tejido y se envió para examen histopatológico, revelando la presencia de túbulos seminíferos típicos (testículos) y glándulas endometriales proliferativas (tubos uterinos). Posteriormente se realizó un procedimiento quirúrgico de corrección para cubrir el tejido peneano y evitar daños por exposición y contacto. Conclusión: Este informe aporta información única sobre el diagnóstico y el enfoque terapéutico en un Yorkshire terrier con síndrome de persistencia del conducto Mülleriano (PMDS). Dado que los trastornos del desarrollo sexual son inusuales y plantean un desafío diagnóstico y terapéutico, este reporte puede representar una valiosa herramienta para los veterinarios.


Resumo Anamnese: Uma fêmea Yorkshire terrier de 1 ano de idade, com 2,2 kg de peso e uma estrutura anormal na região inguinal, similar ao orifício prepucial. Achados clínicos e laboratoriais: Após o exame, uma estrutura semelhante a um pênis foi encontrada saindo do orifício prepucial. Foram realizados exames físicos, laboratoriais, ecográficos e radiográficos. A suspeita inicial seria um caso de desordem do desenvolvimento sexual, pelo que a cadela foi encaminhada para laparotomia exploratória e cariotipagem. Abordagem terapêutica: Durante o procedimento cirúrgico macroscopicamente duas estruturas semelhantes foram encontradas: uma na região caudal do rim direito e outro na região caudal do rim esquerdo. Estes tecidos encontravam-se ligados a duas estruturas semelhantes aos cornos uterinos, caudalmente fusionados a um corpo na cavidade pélvica. Todo o tecido foi removido e enviado para exame histopatológico e revelou a presença de túbulos seminíferos típicos (testículos) e glândulas endometriais proliferativas (tubas uterinas). Subsequentemente, foi realizado procedimento cirúrgico de correção para cobrir o tecido peniano e evitar danos por exposição e contato. Conclusão: Este relatório aporta informações únicas sobre diagnóstico e abordagem terapêutica em um Yorkshire terrier com síndrome de persistência do ducto de Müller (PMDS). Distúrbios do desenvolvimento sexual são anomalias incomuns que representam um desafio diagnóstico e terapêutico considerando este relato uma ferramenta valiosa para os veterinários.

The testicles are the primary sexual organs of male and their function is to produce sperm and sexual hormones. Disorders of the testicles are common in domestic cats. Therefore, detailed assessment of the testes is of great importance in veterinary medicine. Considering the recent advances in diagnostic imaging in companion animals, this review aims to describe the applicability elastography (qualitative and quantitative), Doppler, contrast-enhanced ultrasonography and B-mode ultrasonography in testes evaluation in cats. B-mode ultrasonography of the testicles combined with haemodynamic analysis in real time by Doppler and contrast enhanced ultrasonography can assist as diagnostic tool in evaluating testicular abnormalities in sick cats. Furthermore, ARFI elastography is a new ultrasound method that evaluates tissue elasticity by elastogram and shear weave. Ultrasonographic study of the testes is a common diagnostic imaging procedure.


Los testículos son los principales órganos sexuales de los machos y su función es producir espermatozoides y hormonas sexuales. Los trastornos de estos órganos son comunes en gatos domésticos. La ultrasonografía convencional, combinada con el análisis hemodinámico en tiempo real por Doppler y ultrasonido contrastado son herramientas diagnósticas importantes para la evaluación de enfermedades testiculares en gatos. Además, La elastografía-ARFI es un método ecográfico actual que evalúa la elasticidad tisular por medio del elastograma y de la velocidad de propagación de las ondas de cizallamiento. El estudio ecográfico de los testículos es un procedimiento de diagnóstico por imágenes común.

In view of the fact that cancer is considered a chronic disease that can interfere with hormonal homeostasis by means of cytokines, we hypothesized that, even at early stages, mammary carcinoma is able to alter the balance of the hypothalamic-pituitary-thyroid and hypothalamic-pituitary-adrenal axes. To test this hypothesis, the serum concentrations of basal cortisol, thyroxine (T4), free thyroxine (fT4), and thyroid-stimulating hormone (TSH) were evaluated in 20 unspayed bitches that had a histopathological diagnosis of grade 1 mammary carcinoma at clinical stage I according to the World Health Organization (WHO) classification (T1N0M0). The control animals comprised 10 unspayed bitches in perfect health conditions that were matched with those with mammary carcinoma by age. No significant differences regarding the concentrations of basal cortisol, TSH, t4, and fT4 were found between the bitches carrying early stage mammary carcinoma when compared to the control group. This suggests that, even if malignant, early-stage mammary carcinomas do not exhibit the ability to alter the concentrations of hormones produced by the hypothalamic-pituitary-adrenal or hypothalamic-pituitary-thyroid axes.(AU)


Em vista do fato de neoplasias serem consideradas doenças crônicas que por meio de citocinas podem interferir na homeostase hormonal, hipotetizou-se que o carcinoma mamário, mesmo nos seus estádios iniciais, fosse capaz de alterar o equilíbrio dos eixos hipotalâmico-hipofisário-tireóideo e hipotalâmico-hipofisário-adrenal. Para tal, foram avaliadas as concentrações séricas de cortisol basal, tiroxina (T4), tiroxina livre (fT4) e tireotrofina (TSH) de 20 fêmeas caninas, inteiras, com diagnóstico histopatológico de carcinoma mamário grau 1 e estadiamento clínico I segundo a classificação da Organização Mundial da Saúde - OMS (T1N0M0). Os animais controle constituíram-se por 10 fêmeas caninas inteiras, em perfeitas condições de higidez, as quais foram pareadas, por idade, com aquelas portadoras de carcinoma mamário. Não foram encontradas diferenças significativas nas concentrações de cortisol basal, TSH, T4 e fT4 das cadelas portadoras de carcinoma mamário em estádio inicial quando comparadas às controles sugerindo que, mesmo considerados malignos, ainda não apresentam a capacidade de alterar as concentrações dos hormônios produzidos pelos eixos hipotalâmico-hipofisário-adrenal e tireóideo.(AU)

Quantitative analysis of antioxidants in a fast, simple and accurate manner is of great importance in the view of real-time monitoring the health of individuals. Recently, we have developed a UV/vis spectroscopic microfluidic sensor to specifically quantify ascorbic acid based on the immobilization of ascorbate oxidase, a relatively unstable enzyme. In this work, three different strategies for the immobilization of the unstable enzyme, including alumina sol-gel encapsulation, physisorption to PDMS channels with, and without alumina xerogel modification, were compared to build a microsensor. We found that the loading amount of the enzyme is not the determinative factor for the performance of the microfluidic biosensor but the retained activity of the enzyme and diffusion in the microfluidic channel. Taking into account of the two factors, the protocol of adsorbing enzymes to alumina (Al2O3) xerogel modified PDMS surface was demonstrated to be the best for preparing the microfluidic sensor among the utilized protocols. The microsensor prepared under the optimized protocol was further used to quantify ascorbic acid in human blood, where only dozens of microliters of blood (few drops) was required, demonstrating its potential application in clinical diagnosis. The developed strategy is featured with optimized enzymatic activity, simple process of microfluidic platform, low sample consumption, and straightforward spectrophotometry based detection.

Tumores da genitália de machos só não são mais incidentes que ostumores cutâneos. Os tumores testiculares são raros na maioria das espéciesdomésticas, ocorrendo com maior frequência nos cães. O Sertolioma está entre astrês mais frequentes neoplasias testiculares de cães, ao lado de tumores dascélulas de Leydig e seminomas. Vários fatores podem influenciar odesenvolvimento de tumores testiculares no cão, incluindo criptorquidismo, idade,raça e exposição a fatores cancerígenos. Há uma associação significativa entrecriptorquidismo e o desenvolvimento de tumores de células de Sertoli eseminomas. A maioria dos cães com tumores testiculares são assintomáticos, e aneoplasia testicular é geralmente um achado acidental. Os tumores das células deSertoli são comumente localmente invasivos e raramente metastáticos, mas sabeseque os tumores intra-abdominais possuem maior índice metastático. Estetrabalho visa relatar um caso de tumor das células de Sertoli com metástase emrim esquerdo em um cão SRD de 7 anos, criptorquida bilateral, submetido aorquiectomia e nefrectomia esquerda, e posterior tratamento quimioterápico comdoses alternadas de carboplatina e doxorrubicina , e até o presente momento seapresentando em completa remissão.


Tumors of male genitalia are not only more incidents than skin tumors.Testicular tumors are rare in most domestic species, occurring more frequently indogs. The Sertolioma is among the three most frequent testicular neoplasms indogs, besides Leydig cell tumors and seminomas. Several factors may influencethe development of testicular tumors in dogs, including cryptorchidism, age, raceand exposure to carcinogenic factors. There is a significant association betweencryptorchidism and tumor development of Sertoli cells and seminomas. Most dogswith testicular tumors are asymptomatic, and testicular cancer is usually anincidental finding. The Sertoli cell tumors are usually locally invasive and rarelymetastatic, but it is known that the intra-abdominal tumors have a higher metastaticrate. This paper describes a case of Sertoli cell tumor with metastasis in the leftkidney in a seven years old, male, mongrel dog, bilateral cryptorchid , submitted toorchiectomy and left nephrectomy and subsequent chemotherapy with alternatingdoses of doxorubicin and carboplatin every 21 days, and until now performing incomplete remission.

The cAMP/protein kinase A (PKA) signalling cascade is ubiquitous, and each step in this cascade involves enzymes that are expressed in multiple isoforms. We investigated the effects of this diversity on the integration of the pathway in the target cell by comparing prefrontal cortical neurones with striatal neurones which express a very specific set of signalling proteins. The prefrontal cortex and striatum both receive dopaminergic inputs and we analysed the dynamics of the cAMP/PKA signal triggered by dopamine D1 receptors in these two brain structures. Biosensor imaging in mouse brain slice preparations showed profound differences in the D1 response between pyramidal cortical neurones and striatal medium spiny neurones: the cAMP/PKA response was much stronger, faster and longer lasting in striatal neurones than in pyramidal cortical neurones. We identified three molecular determinants underlying these differences: different activities of phosphodiesterases, particularly those of type 4, which strongly damp the cAMP signal in the cortex but not in the striatum; stronger adenylyl cyclase activity in the striatum, generating responses with a faster onset than in the cortex; and DARPP-32, a phosphatase inhibitor which prolongs PKA action in the striatum. Striatal neurones were also highly responsive in terms of gene expression since a single sub-second dopamine stimulation is sufficient to trigger c-Fos expression in the striatum, but not in the cortex. Our data show how specific molecular elements of the cAMP/PKA signalling cascade selectively enable the principal striatal neurones to respond to brief dopamine stimuli, a critical process in incentive learning.
