RESUMO

Nanomaterials that generate reactive oxygen species (ROS) upon light irradiation have significant applications in various fields, including photodynamic therapy (PDT) that is widely recognized as a highly momentous strategy for the eradication of cancer cells. However, the ROS production rate of photosensitizers, as well as the tumor hypoxia environment, are two major challenges that restrict the widespread application of PDT. In this study, a cancer-thylakoid hybrid membrane-camouflaged thulium oxide nanoparticles (Tm2O3) for tumor-homing phototherapy through dual-stage-light-guided ROS generation and oxygen self-supply is developed. Tm2O3 as a type II photosensitizer are viable for NIR-stimulated ROS generation due to the unique energy levels, large absorption cross section, and long lifetime of the 3H4 state of Tm ions. The thylakoid membrane (TK) plays a catalase-like role in converting hydrogen peroxide into oxygen and also acts as a natural photosensitizer that can generate lethal ROS through electron transfer when exposed to light. In addition, fluorescence dye DiR is embedded in the hybrid membrane for in vivo tracing as well as photothermal therapy. Results show that tumors in Tm2O3@TK-M/DiR group are effectively ablated following dual-stage-light irradiation, highlighting the promising potential of rare-earth element-based type II photosensitizers in various applications.

Esculetin (ELT) is one of the best-known and simplest coumarins with powerful natural antioxidant effects but insoluble and difficult to absorb. In order to overcome the problems, cocrystal engineering was first applied to ELT in this paper. Nicotinamide (NAM) was selected as the coformer for its excellent water solubility and potential synergistic antioxidant effect with ELT. The structure of the ELT-NAM cocrystal was successfully prepared and characterized by IR, SCXRD, PXRD, and DSC-TG. Furthermore, the in vitro/vivo properties and antioxidant effects of the cocrystal were adequately studied. The results highlight that the ELT obtained tremendous improvements in water solubility and bioavailability after cocrystal formation. Meanwhile, the synergistic enhancement of ELT with NAM in antioxidant effect was demonstrated by the DPPH assay. Ultimately, the simultaneously optimized in vitro/vivo properties and antioxidant activity of the cocrystal created an improved practical effect of hepatoprotective in rat experiments. The investigation is significant for developing coumarin drugs represented by ELT.

Short peptides that can respond to external stimuli have been considered as the preferred building blocks to construct hydrogels for biomedical applications. In particular, photoresponsive peptides that are capable of triggering the formation of hydrogels upon light irradiation allow the properties of hydrogels to be changed remotely by precise and localized actuation. Here, we used the photochemical reaction of the 2-nitrobenzyl ester group (NB) to develop a facile and versatile strategy for constructing photoactivated peptide hydrogels. The peptides with high aggregation propensity were designed as hydrogelators, which were photocaged by a positively charged dipeptide (KK) to provide strong charge repulsion and prevent self-assembly in water. Light irradiation led to the removal of KK and triggered the self-assembly of peptides and the formation of hydrogel. Light stimulation endows spatial and temporal control, which enables the formation of hydrogel with precisely tunable structure and mechanical properties. Cell culture and behavior study indicated that the optimized photoactivated hydrogel was suitable for 2D and 3D cell culture, and its photocontrollable mechanical strength could regulate the spreading of stem cells on its surface. Therefore, our strategy provides an alternative way to construct photoactivated peptide hydrogels with wide applications in biomedical areas.

WS9326A is a peptide antibiotic containing a highly unusual N-methyl-E-2-3-dehydrotyrosine (NMet-Dht) residue that is incorporated during peptide assembly on a non-ribosomal peptide synthetase (NRPS). The cytochrome P450 encoded by sas16 (P450Sas) has been shown to be essential for the formation of the alkene moiety in NMet-Dht, but the timing and mechanism of the P450Sas-mediated α,β-dehydrogenation of Dht remained unclear. Here, we show that the substrate of P450Sas is the NRPS-associated peptidyl carrier protein (PCP)-bound dipeptide intermediate (Z)-2-pent-1'-enyl-cinnamoyl-Thr-N-Me-Tyr. We demonstrate that P450Sas-mediated incorporation of the double bond follows N-methylation of the Tyr by the N-methyl transferase domain found within the NRPS, and further that P450Sas appears to be specific for substrates containing the (Z)-2-pent-1'-enyl-cinnamoyl group. A crystal structure of P450Sas reveals differences between P450Sas and other P450s involved in the modification of NRPS-associated substrates, including the substitution of the canonical active site alcohol residue with a phenylalanine (F250), which in turn is critical to P450Sas activity and WS9326A biosynthesis. Together, our results suggest that P450Sas catalyses the direct dehydrogenation of the NRPS-bound dipeptide substrate, thus expanding the repertoire of P450 enzymes that can be used to produce biologically active peptides.

This study aims to develop a high-speed and nondestructive mildewed rice grain detection method. First, a set of microscopic images of rice grains contaminated by Aspergillus niger, Penicillium citrinum, and Aspergillus cinerea are acquired to serve as samples, and the mildewed regions are marked. Then, three YOLO-v5 models for identifying regions of rice grain with contamination of Aspergillus niger, Penicillium citrinum, and Aspergillus cinerea in microscopic images are established. Finally, the relationship between the proportion of mildewed regions and the total number of colonies is analyzed. The results show that the proposed YOLO-v5 models achieve accuracy levels of 89.26%, 91.15%, and 90.19% when detecting mildewed regions with contamination of Aspergillus niger, Penicillium citrinum, and Aspergillus cinerea in the microscopic images of the verification set. The proportion of the mildewed region area of rice grain with contamination of Aspergillus niger/Penicillium citrinum/Aspergillus cinerea is logarithmically correlated with the logarithm of the total number of colonies (TVC). The corresponding determination coefficients are 0.7466, 0.7587, and 0.8148, respectively. This study provides a reference for future research on high-speed mildewed rice grain detection methods based on MCV technology.

OBJECTIVE: To observe the effect of electroacupuncture (EA) on vascular endothelial growth factor-C (VEGF-C), vascular endothelial growth factor receptor-3 (VEGFR-3), proinflammatory factors and apoptosis in myocardial tissue in mice with acute myocardial ischemia (AMI), and to explore the mechanism of EA for AMI. METHODS: Fifty male C57BL/6 mice were randomly divided into a sham operation group, a model group, an EA group, an inhibitor group and an inhibitor+EA group, 10 mice in each group. Except for the sham operation group, the mice in the remaining groups were intervented with ligation at the left anterior descending (LAD) coronary artery to establish AMI model. The mice in the sham operation group were intervented without ligation after thoracotomy. The mice in the EA group were intervented with EA at "Shenmen" (HT 7) and "Tongli" (HT 5), disperse-dense wave, 2 Hz/15 Hz in frequency, 1 mA in current intensity, 30 min each time, once a day, for 3 d. The mice in the inhibitor group were treated with intraperitoneal injection of SAR 131675 (12.5 mg•kg-1•d-1, once a day for 3 d). The mice in the inhibitor+EA group were injected intraperitoneally with SAR 131675 30 min before EA. The ECG before modeling, 30 min after modeling and 3 d after intervention was detected, and the ST segment displacement was recorded; after the intervention, the ELISA method was applied to measure the contents of serum creatine kinase isoenzyme (CK-MB), aspartate aminotransferase (AST) as well as tumor necrosis factor-α (TNF-α) and interleukin-23 (IL-23) in myocardial tissue; the HE staining method was used to observe the morphological changes of myocardial tissue; the immunofluorescence double labeling method was applied to measure the number of co-expression positive cells of VEGF-C/VEGFR-3 in myocardial tissue; the TUNEL method was used to detect the level of cardiomyocyte apoptosis; the Western blot method was applied to measure the protein expressions of VEGF-C, VEGFR-3, b-lymphoma-2 (Bcl-2), activated caspase-3 (Cleaved Caspase-3) and activated poly adenosine diphosphate ribose polymerase-1 (Cleaved PARP-1). RESULTS: Compared with the sham operation group, in the model group the ST segment displacement was increased (P<0.01); the contents of CK-MB, AST, TNF-α and IL-23 were increased (P<0.01); the arrangement of myocardial fibers was disordered, and interstitial inflammatory cell infiltration was obvious; the number of co-expression positive cells of VEGF-C/VEGFR-3 was decreased (P<0.01); the number of cardiomyocyte apoptosis was increased (P<0.01); the expressions of VEGF-C, VEGFR-3 and Bcl-2 were decreased (P<0.01); the expressions of Cleaved Caspase-3 and Cleaved PARP-1 were increased (P<0.01). Compared with the model group, in the EA group the ST segment displacement was decreased (P<0.01); the contents of CK-MB, AST, TNF-α, IL-23 were decreased (P<0.01); the severity of myocardial pathological injury was reduced; the number of co-expression positive cells of VEGF-C/VEGFR-3 was increased (P<0.01); the number of cardiomyocyte apoptosis was reduced (P<0.01); the expressions of VEGF-C, VEGFR-3 and Bcl-2 were increased (P<0.01); the expressions of Cleaved Caspase-3 and Cleaved PARP-1 were reduced (P<0.01). There was no significant difference in all the indexes between the model group and the inhibitor group (P>0.05). Compared with the model group, the protein expression of VEGF-C was increased in the inhibitor+EA group (P<0.01). Compared with the inhibitor group, in the EA group the ST segment displacement was decreased (P<0.01); the contents of CK-MB, AST, TNF-α, IL-23 were decreased (P<0.01); the severity of myocardial pathological injury was reduced; the number of co-expression positive cells of VEGF-C/VEGFR-3 was increased (P<0.05); the number of cardiomyocyte apoptosis was reduced (P<0.01); the expressions of VEGF-C, VEGFR-3 and Bcl-2 were increased (P<0.01); the expressions of Cleaved Caspase-3 and Cleaved PARP-1 were reduced (P<0.01). Compared with the inhibitor+EA group, all the indexes in the EA group were improved except the protein expression of VEGF-C (P<0.01). CONCLUSION: EA could relieve the inflammatory reaction and apoptosis in AMI mice, and its mechanism may be related to activating VEGF-C/VEGFR-3 pathway and promoting lymphangion genesis.

The PDGF receptor is mock-coupled with a known active compound, and 14 novel skeleton candidate compounds were designed and synthesized. The structure was confirmed by 1H NMR, 13C NMR and MS. The in vitro cytotoxicity of the two cancer cell lines (SGC-7901 and A549) was evaluated by MTT assay. PDGF receptor protein inhibition assays were performed on I6 and II4 using fluorescence polarization immunoassay (FPIA). [Formula: see text].

Phosphorus, as a limiting and vital macronutrient in water, circulates mainly with sediment through rivers. The construction and operation of large reservoirs can have a profound impact on the characteristics of phosphorus transportation and transformation. Based on measured hydrological and water quality data during 2008-2016 in the Three Gorges Reservoir (TGR), a statistical model of total phosphorus (TP) flux and sediment load in different periods was established. The formula of TP flux was established by analyzing the interpolation of unmeasured TP concentration, tempo-spatial characteristics, flux variation, and the retention effect of TP in the TGR. The results indicated that taking no account of the effects of TP inflows of tributaries, annual TP concentration in the TGR varied between 0.196 mg·L-1 and 0.290 mg·L-1 during 2008-2012, and inner-annual changes in TP concentration presented an "M" type with two obvious peaks. TP concentration decreased slowly from upstream to downstream of the TGR, and increased significantly from Cuntan to Qingxichang in some years. The average annual TP flux and retention rate of the TGR were 82300 t and 49.76% during 2008-2012, respectively. During 2013-2016, the average annual TP flux and retention rate decreased significantly, to about 47900 t and 12.03%, respectively.

Erdafitinib has recently been approved for the treatment of adult patients with locally advanced or metastatic urothelial carcinoma (UC). The current study aimed to evaluate whether erdafitinib inhibits the proliferation in UC via inducing authophagy. Our data showed that erdafitinib demonstrated strong toxicity for the T24 and UMUC6 cell lines. Flow cytometry analysis showed that erdafitinib increased the proportion of G0/G1 phase in both T24 and UMUC6 cells. Additionally, Annexin V staining indicated that erdafitinib enhanced the apoptosis of UC cells. Meanwhile, the expression of apoptosis-related proteins was significantly elevated after treatment with erdafitinib. Transwell assay showed that erdafitinib significantly reduced T24 and UMUC6 cell migration and invasion. More importantly, western blot assay showed that erdafitinib induced the expression of autophagy-related proteins. Besides, GFP-LC3 transfection assay and electron microscopy examination showed that erdafitinib could partially diminish 3-methyladenine (3-MA) induced impairment of autophagy in UC cells. In summary, for the first time we showed novel data that erdafitinib inhibited UC cell malignant proliferation via inducing cell autophagy.

In this study, the alkaline hydrothermal ferric carbon (AHFC), which was prepared by hydrothermal liquefaction method using Fenton iron sludge with NH3·H2O, was used as a skeleton materials for the dewatering of sewage sludge (SS) via thermal hydrolysis. NH2 functional group presented in the AHFC and nano-sized γ-Fe2O3 was anchored on the surface of AHFC. The NH2 functional group notably promoted thermal hydrolysis of SS from the increasing of TOC and TN value. γ-Fe2O3 showed adsorption effect to the water, resulting in decline of the dewatering rate of SS in present condition. When 100% of AHFC was added, dewatering rate of SS was decreased by 19.93% (at 160 °C), 4.50% (at 180 °C) and 8.34% (at 200 °C) respectively. 3D-EEM results showed that the degree of hydrolysis was deepened when AHFC was added for the intensity of soluble microbial products decline. AHFC promoted the decomposition of protein to form heterocycle compounds in the resulting cake according to in situ FTIR results. The nano γ-Fe2O3 catalysis to cake also can be observed for the activation energy was lower than blank in the range of 40˜60%. The study demonstrated concept and the effectiveness the reuse/recycle of the Fenton iron sludge for dewatering of SS.

BACKGROUND: To investigate the diagnostic value of serum miR-324 in patients with prostate cancer (PC). METHODS: Blood samples from 50 patients with prostate cancer, 30 patients with benign prostatic hyperplasia (BPH), and 20 healthy controls were collected and quantified by quantitative reverse transcription polymerase chain reaction (qRT-PCR). The relationship between serum miR-324 level and Gleason classification and TNM staging of prostate cancer was analyzed. ROC curve was used to analyze the value of miR-324 in early diagnosis of prostate cancer. RESULTS: The expression of miR-324 in prostate cancer group was significantly higher than that in the BPH group and normal control group (all p < 0.01). Serum miR-324 was associated with the Gleason score and tumor stage (all p < 0.01). Serum miR-324 was positively correlated with PSA (r = 0.673, p = 0.000); ROC curve analysis showed that the area under the ROC curve of miR-324 (all p < 0.01) (AUC) was 0.911 (95% CI: 0.855 - 0.966, p = 0.000). When the relative expression level of miR-324 was 3.35, the sensitivity, specificity, positive predictive value, and negative predictive values were 86.0%, 82.0%, 82.7%, and 85.4%, respectively. CONCLUSIONS: The increased expression of serum miR-324 in patients with prostate cancer may be a potential new biomarker for the diagnosis of prostate cancer, which is helpful for the differentiation between PC and BPH.

The search for high active, stable, and cost-efficient hydrogen evolution reaction (HER) electrocatalysts for water electrolysis has attracted great interest. The coordinated water molecules in the hydronium ions will obviously reduce the positive charge density of H+ and hamper the ability of H+ to receive electrons from the cathode, leading to large overpotential of HER on nonprecious metal catalysts. Here we realize Pt-like hydrogen evolution electrocatalysis on polyaniline (PANI) nanodots (NDs)-decorated CoP hybrid nanowires (HNWs) supported on carbon fibers (CFs) (PANI/CoP HNWs-CFs) as PANI can effectively capture H+ from hydronium ions to form protonated amine groups that have higher positive charge density than those of hydronium ions and can be electro-reduced easily. The PANI/CoP HNWs-CFs as low-cost electrocatalysts show excellent catalytic performance toward HER in acidic solution, such as super high catalytic activity, small Tafel slope, and superior stability.

Previous studies have established a bovine mammary gland epithelia cells in vitro model by the adenovirus-mediated telomerase (hTERT-bMGEs). The present study was conducted to confirm whether hTERT-bMGEs were effective target cells to improve the efficiency of transgenic expression and somatic cell nuclear transfer (SCNT). To accomplish this, a mammary-specific vector encoding human lysozyme and green fluorescent protein was used to verify the transgenic efficiency of hTERT-bMGEs, and untreated bovine mammary gland epithelial cells (bMGEs) were used as a control group. The results showed that the hTERT-bMGEs group had much higher transgenic efficiency and protein expression than the bMGEs group. Furthermore, the nontransgenic and transgenic hTERT-bMGEs were used as donor cells to evaluate the efficiency of SCNT. There were no significant differences in rates of cleavage or blastocysts or hatched blastocysts of cloned embryos from nontransgenic hTERT-bMGEs at passage 18 and 28 groups (82.8% vs. 81.9%, 28.6% vs. 24.8%, 58.6% vs. 55.3%, respectively) and the transgenic group (80.8%, 26.5% and 53.4%); however, they were significantly higher than the bMGEs group (71.2%, 12.8% and 14.8%), (p < 0.05). We confirmed that hTERT-bMGEs could serve as effective target cells for improving development of somatic cell cloned cattle embryos.

<p><b>OBJECTIVE</b>To investigate the association of miR-199a expression with the clinicopathologic characteristics and the survival of patients with renal cell carcinoma (RCC).</p><p><b>METHODS</b>Real-time quantitative RT-PCR was used to detect the expression of miR-199a in the tumor tissues and paired adjacent normal tissues from 67 patients with RCC. The correlations of miR-199a expression with the clinicopathologic characteristics and survival of RCC patients were analyzed.</p><p><b>RESULTS</b>Positive miR-199a expression was detected in both the RCC and adjacent normal tissues, but the tumor tissues showed a significantly lower expression level by a mean of 13.7 folds (P=0.003). The expression of miR-199a was negatively correlated with tumor recurrence in RCC patients in T stage (P<0.05), but showed no significant correlations with the patients age, histological type of the tumor, lymph node metastasis, distal metastasis, or Fuhrman grade (P>0.05). The patients with lowered miR-199a expression in the tumor tissue had a significantly shorter mean survival time than those without miR-199a down-regulation (P=0.017 by Log-rank test).</p><p><b>CONCLUSION</b>A decreased expression of miR-199a is significantly correlated with a higher tumor stage, a greater likeliness of tumor recurrence, and a poorer prognosis in RCC patients. miR-199a can serve as a promising prognostic factor of renal cell carcinoma.</p>

@#Objective To observe clinical effect of acupuncture combined with rehabilitation training on spastic cerebral palsy. Methods 160 patients with cerebral palsy were divided into acupuncture group(n=80) and control group(n=80). The acupuncture group received acupuncture, including the acupoints such as Baihui, Sishengcong, Yamen, Fengchi, etc., and combined with function training. The control group received Bobath appoach as rehabilitation training. Two groups were treated for 3 months. Results The acupuncture group(n=77) showed powerfully for 26 cases, effective for 46 cases, and invalid for 5 cases, and the efficient rate was 93.5%. The control group (n=78) showed powerfully for 20 cases, effective for 45 cases, invalid for 13 cases, and the efficient rate was 83.3% (P<0.05). Conclusion Acupuncture combined with rehabilitation training can facillitate to reduce muscle tone of spastic cerebral palsy, and improve effect of rehabilitation.

Objective To observe the effects of electrical acupuncture for the treatment of neurogenic detrusor overactivity (DO) caused by spinal cord injury (SCI).Methods Twenty SCI patients with DO were included in this study. There were 18 males and 2 females;age ranged from 17 to 58 years. Patients were given electro-acupuncture treatment at the bilateral S3 foramen at the lateral position during the vidio-urodynamic investigation when the detrusor of the patients began to contract.The different intensities of electrical stimulation were used when the DO appeared during cystometry and the most effective intensity to inhibit DO was determined. Then, the bladder was emptied and the stimulation with selected intensity was used at the beginning of cystometry. The changes of parameters in the urodynamics, the urinary incontinence times per day and the pad used per day were recorded. The acute effects were observed. Ten days were set as one course and after 3 courses and 9 courses the patients underwent urodynamic test again. The long-term effects were observed.Results After treatment, the urinary incontinence times per day and the pad used per day decreased obviously (P<0. 05). The bladder capacity increased significantly and the maximum intravesical pressure decreased significantly. There were 3 patients having the long terms treatment. Conclusions The Chinese electro-acupuncture at S3 foramen in the SCI patients with DO is demonstrated effective. After the treatment bladder capacity could increase and the times of the urinary incontinence per week decrease.

@# Objective To observe the effect of electroacupuncture on migraine. Methods 60 patients were divided into electroacupuncture group (30 cases) who received electroacupuncture, and control group (30 cases) who received local nerve block. Results In the electroacupuncture group, 22 (73.33%) cases were cured, 6 (20.00%) cases were obviously improved, 2 (6.67%) cases were improved. While in control group, 17 (56.67%) cases were cured, 5 (16.67%) cases were obviously improved, 8 (26.67%) cases were improved. There was significant difference between these two groups (P<0.05). The time of treatment was less in the electroacupuncture group than in the control (P<0.05). Conclusion Electroacupuncture is effective on migraine.

@#Objective To observe the effect of traditional Chinese drug combined with training of musculus quadriceps fexoris on knee osteoarthritis(KOA).Methods70 out-patient clinic KOA patients were divided randomly into the Chinese drug group and control group with 35 cases in each group.The Chinese drug group was treated with Shentongzhuyu medicinal broth PO bid;the control group was treated with Sulindac 0.2 g PO bid.Two groups were combined with the training of musculus quadriceps fexoris,having 10 times per course and 3 courses in total with a 3 days interval between two courses.The therapeutic effect was evaluated with footplate pressure gait analysis and modified JOA marks.ResultsAfter treatment,the effect of the Chinese drug group was superior to the control group(P<0.05),especially 12 weeks post treatment.There maximum weight loading,time integral and weight loading intergral of affected limb of the Chinese drug group significantly improved after treatment(P<0.01),but for the control group,only weight loading intergral improved(P<0.05).ConclusionTraditional Chinese drug combined with training of musculus quadriceps fexoris has better curative effect on the pain and functional disturbance of KOA.
