RESUMO

Enzyme immunoassay (EIA) has been applied to measurements of human histocompatibility (HLA) antigens (antibodies). The optimal concentration of the antigen immobilized on solid phase and the optimal dilutions of HLA sera have been found. The assays have been performed with the peripheral blood lymphocytes and platelets from donors with a known HLA phenotype. Horseradish peroxidase (Sigma, Rz-2.75) has been employed as the enzymic label, polystyrene tubes manufactured by LKB (Sweden) have been used as solid-phase carriers. The results evidence the possibility of applying EIA to the determination of HLA antigens.

RESUMO

In experimental alloxan diabetes marked hypocalcemia and hyperglycemia are observed in the blood of rats during the acute period of the disease (from 1.5 to 2 months). After 2.5-3.5 months of diabetes calcium level reaches the upper normal limit. Simultaneously, antibodies to calcitonin appear. It is quite likely that excessive calcitonin secretion, leading to hypocalcemia, decreased insulin secretion and hyperglycemia take place in the acute stage of diabetes. When diabetes persists the antibodies to calcitonin which reveal a homeostatic reaction blocking the excess of calcitonin appear. This normalizes the level of calcium in the blood and prevents the further increase in the blood sugar level.

RESUMO

Antibodies to calcitonin, parathyroid hormone and cortisol are detected in acute stage of infection-caused bronchial asthma. The appearance of antibodies is paralleled by marked hypercalcaemia. The antibodies may bind excessive hormones in the blood, preventing further hormonal imbalance. Ten-day treatment with glucocorticoids decreased the amount of antibodies possibly due to normalization of hormonal secretion and restoration of their balance. As a result, calcium blood levels returned to normal.

RESUMO

Antisera to lysozymes of O. maubata, O. paillipes, A. lahorensis and H. asiaticum were prepared for the first time with a purpose of investigating the immunological identity of the lysozymes. Partial antigenic identity of the egg lysozyme and the lysozymes of the above ticks was shown with the methods of precipitation in agar and the immunoenzymatic assay.

The optimum parameters of the immunoenzyme assay system for the identification of antibodies to M. arthritidis and M. fermentans in the sera of patients with rheumatoid arthritis have been established. The investigation has shown that the products obtained by the ultrasonic disintegration of the biomass of M. arthritidis and M. fermentans can be used as soluble antigens for adsorption on the polystyrene surface of plates. The use of the immunonenzyme assay, specially modified, has made it possible to establish that antibodies to M. arthritidis can be detected in 6.5% of cases, antibodies to M. fermentans, in 41.9% of cases and the association of antibodies to M. arthritidis and M. fermentans, in 41.9% of cases. At the same time antibodies to M. arthritidis have been found to belong mainly to IgM and antibodies to M. fermentans, to IgG or to IgG and IgM simultaneously.

RESUMO

Antibodies to calcitonin appear in blood of rats with experimental alloxan diabetes. This phenomenon is observed only under high blood sugar. At the stage of latent diabetes, i.e. during alloxan administration to the body and low blood sugar no antibodies to calcitonin are detected. It is possible that appearance of autoantibodies to calcitonin is one of pathogenetic factors of hyperglycemia development in rats with alloxan diabetes.

RESUMO

Lately it appeared to be necessary to develop-alternative methods for control of antibiotic therapy. Detection of low molecular substances based on the immunoenzymatic assay (IEA) is one of the most promising approaches to the development of such methods. The authors developed a quantitative method for determination of benzylpenicillin (BP) according to the procedure described earlier by Van Weemen. The BP antisera were prepared by immunization of rabbits with a conjugate of bovine serum albumin. The gamma-globulin fraction isolated from the antisera was conjugated with peroxidase. The working dilution of the conjugate was determined by the chess titration method. The test system of competitive binding developed by the authors provides detection of BP in the biological fluids in concentrations of 0.1 to 0.0001 microgram/ml (by BP dry weight).

RESUMO

The optimum conditions for the detection of specific humoral antibodies in patients with meningococcal infection by the immunoenzymatic method have been determined: the sensitizing dose of the antigen is 20 microgram of protein per 1 ml for group A meningococcal antigen and 25 microgram of protein per 1 ml for group C meningococcal antigen, the pH of the buffer solutions is 5.0-6.0 for group A meningococcal antigen and 12.0 for group C meningococcal antigen, the time and temperature of incubation should be 18-20 hours at 14 degrees C. The diagnostic test system developed in this research is capable of detecting antibodies with the accuracy of several ng.
