Resumo

Estradiol cypionate (EC) or GnRH have been widely used for ovulation induction in timed embryo transfer (TET). EC administration increases the proportion of cows that show estrus, whereas GnRH promotes more synchronized ovulations. The aim of the present study was to evaluate the potential beneficial effects of combining EC and GnRH in TET. In experiment 1, no difference was observed on serum progesterone concentrations on Day 6 and 13 after GnRH treatment between GnRH and EC+GnRH groups. In experiment 2, pregnancy per embryo transfer (P/ET) did not differ (p = 0.69) between GnRH (62.8%) and EC+GnRH (58.7%) groups. In conclusion, combining EC and GnRH for ovulation induction does not increase progesterone secretion and pregnancy rate after TET in cattle.(AU)

Resumo

In the present study, we investigate the effect of the presence or absence of corpus luteum (CL) at the beginning of a fixed-time artificial insemination (FTAI) protocol and to evaluate the impact of one-time use of intravaginal progesterone device (P4 device) in cows with or without CL. A total of 776 primiparous Nellore cows were subjected to FTAI approximately 45 days postpartum. In Experiment 1, 476 cows were divided into two experimental groups: with (CL-present, n=113) or without (CL-absent, n=363) CL, after ultrasound evaluation. On day 0 (D0), all cows received a new P4 device (1.0 g) and 2.0 mg estradiol benzoate (EB). Eight days later (D8), the P4 devices were withdrawn, and prostaglandin (15 mg), estradiol cypionate (0.5 mg), and eCG (300 IU) were administered i.m. All cows were inseminated 48 h after P4 device withdrawal (D10). In Experiment 2, the cows (n= 300) received (at D0) P4 devices that were previously used once in other cows with (n=109) or without CL (n=191) and 2 mg of EB. The same protocol as that used in Experiment 1 was performed from D8 onwards. In Experiment 1, the overall conception rate after FTAI was 55% (262/476). No difference was found in the conception rate between CL-present and CL-absent cows (52.2 vs. 55.5%). In Experiment 2, the conception rate obtained with P4 devices previously used in cows with CL (58.7%) was greater (P<0.05) than that obtained with P4 devices previously used in cows without CL (42.9%). Thus, this strategy resulted in a 15.8% increase in conception rate. In conclusion, the presence or absence of CL at the beginning of the FTAI protocol did not affect the conception rate in cows synchronized with the new P4 device, but the insertion of P4 devices previously used in cows with CL enhanced the conception rates in cows without CL.


No presente estudo, investigamos o efeito da presença ou ausência de corpo lúteo (CL) no início de um protocolo de inseminação artificial em tempo fixo (IATF) e avaliamos o impacto do uso único de dispositivo intravaginal de progesterona (dispositivo P4) em vacas com ou sem CL. Um total de 776 vacas Nelore primíparas, aproximadamente 45 dias pós-parto foram submetidas à IATF. No Experimento 1, após avaliação ultrassonográfica, 476 vacas foram divididas em dois grupos experimentais: com (CL-presente, n=113) ou sem (CL-ausente, n=363) CL. No dia 0 (D0), todas as vacas receberam um novo dispositivo de P4 (1,0 g) e 2.0 mg de benzoato de estradiol (BE). Após 8 dias (D8), os dispositivos P4 foram retirados e prostaglandina (15 mg), cipionato de estradiol (0,5 mg) e eCG (300 UI) foram administrados i.m. Todas as vacas foram inseminadas 48 horas após a retirada do dispositivo de P4 (D10). No Experimento 2, as vacas (n= 300) receberam (no D0) um dispositivo de P4 previamente utilizado uma única vez em outras vacas com (n=109) ou sem CL (n=191) e 2 mg de BE. O mesmo protocolo utilizado no Experimento 1 foi realizado a partir do D8. No experimento 1, a taxa geral de concepção após IATF foi de 55% (262/476). Não foi encontrada diferença na taxa de concepção entre as vacas com CL presente e CL ausente (52,2 vs. 55,5%). No Experimento 2, a taxa de concepção obtida com dispositivos P4 previamente utilizados em vacas com CL-presente (58,7%) foi maior (P<0,05) quando comparada aos dispositivos P4 previamente utilizados em vacas com CL-ausente (42,9%). Essa estratégia resultou em aumento de 15,8% na taxa de concepção. Em conclusão, a presença ou ausência de CL no início do protocolo de IATF não afetou a taxa de concepção em vacas sincronizadas com dispositivo novo de P4; e a eficácia dos dispositivos de P4 previamente utilizados em vacas com CL é maior durante seu segundo uso em vacas sem CL.

A IATF é uma biotecnologia da reprodução que permite o melhoramento genético dos rebanhos de corte, pois possibilita a sincronização da ovulação e realização da inseminação artificial sem necessidade de detecção do cio. O objetivo deste estudo, foi avaliar a eficiência da pré-sincronização com a utilização prévia de progesterona (P4) injetável, sua influência e efeitos na taxa de prenhez, por meio da realização de um protocolo de IATF em vacas de corte multíparas, em quatro fazendaslocalizadas no Mato Grosso do Sul-SP. Para o estudo foram utilizadas 396 vacas nelores, os animais de cada fazenda foram alocados em grupos distintos experimental e controle. Foram avaliadas 196 vacas pertencentes ao grupo controle, e 200 vacas tratadas com progesterona no total, sendo as fazendas descritas neste estudo como fazenda A, B, C e D. Dez dias antes do início do protocolo (D-10), foi aplicado progesterona injetável, via intramuscular em animais do mesmo lote, no D0 foi aplicado benzoato de estradiol e colocado o dispositivo intravaginal com liberação de progesterona, além da classificação dos animais que apresentavam ou não o corpo lúteo. No D8 o dispositivo foi retirado e aplicado uma dose de gonadotrofina coriônica equina, e cipionato de estradiol e de prostaglandina. A inseminação artificial em tempo fixo foi realizada no D10, com a utilização de sêmen de touros provados. O diagnóstico de gestação foi feito no D40, comparando os resultados das taxas de prenhez das vacas dos diferentes grupos, que foram tratadas ou não com progesterona de forma prévia ao protocolo. Sobre as taxas de prenhez dos animais tratados com progesterona, foram encontrados os resultados da fazenda A= 6,60%, fazenda B=10,80%, fazenda C= 3,90% e fazenda D= 3,70%, assim ficando evidente que essa biotecnologia da reprodução é lucrativa e contribui positivamente para a cadeia de produção de bovinos de corte. Baseado nos resultados, pode-se concluir que o uso da P4 (injetável) obteve êxito no índice de prenhez.(AU)


The IATF is a reproduction biotechnology that allows the genetic improvement of beef herds,as it allows the synchronization of ovulation and artificial insemination without the need for heat detection. The objective of this study was to evaluate the efficiency of pre-synchronization with the previous use of injectable progesterone (P4), its influence and effects on the pregnancy rate, through the performance of an IATF protocol in multiparous beef cows, in four farms located in Mato Grosso do Sul-SP. For the study, 396 Nelore cows were used, the animals from each farm were allocated into different experimental and control groups. A total of 196 cows belonging to the control group were evaluated, and 200 cows treated with progesterone in total, being the farms described in this study as farms A, B, C and D. Ten days before the beginning of the protocol (D-10), injectable progesterone was administered , intramuscularly in animals of the same batch, on D0 estradiol benzoate was applied and the intravaginal device was placed with progesterone release, in addition to the classification of animals that had or did not have the corpus luteum. On D8, the device was removed and a dose of equine chorionic gonadotropin, estradiol and prostaglandin cypionate was applied.Fixed-time artificial insemination was performed on D10, using semen from proven bulls. The pregnancy diagnosis was made on D40, comparing the results of the pregnancy rates of the cows of the different groups, which were treated or not with progesterone prior to the protocol. From the pregnancy rates of animals treated with progesterone, the results were found for farm A= 6.60%, farm B= 10.80%, farm C= 3.90% and farm D= 3.70%, thus becoming evident that this reproductive biotechnology is profitable and contributes positively to the beef cattle production chain. Based on the results, it can be concluded that the use of P4 (injectable) was successful in the pregnancy rate.(AU)


La IATF es una biotecnología de la reproducción que permite la mejora genética de rebaños de carne, ya que permite sincronizar la ovulación y la inseminación artificial sin necesidad de detección de celo. El objetivo de este estudio fue evaluar la eficiencia de la presincronización con el uso previo de progesterona inyectable (P4), su influencia y efectos en la tasa de preñez, mediante la realización de un protocoloIATF en vacas de carne multíparas, en cuatro fincas ubicadas en Mato Grosso do Sul-SP. Para el estudio se utilizaron 396 vacas Nelore, los animales de cada finca se distribuyeron en diferentes grupos experimentales y de control. Se evaluaron un total de 196 vacas pertenecientes al grupo control, y 200 vacas tratadas con progesterona en total, siendo las fincas descritas en este estudio como fincas A, B, C y D. Diez días antes del inicio del protocolo (D-10), se administró progesterona inyectable, por vía intramuscular en animales del mismo lote, sobre D0 se aplicó benzoato de estradiol y se colocó el dispositivo intravaginal con liberación de progesterona, además de la clasificación de animales que tenían o no cuerpo lúteo. El D8 se retiró el dispositivo y se aplicó una dosis de gonadotropina coriónica equina, estradiol y cipionato de prostaglandina. Se realizó inseminación artificial a tiempo fijo en D10, utilizando semen de toros probados. El diagnóstico de preñez se realizó el D40, comparando los resultados de las tasas de preñez de las vacas de los diferentes grupos, que fueron tratadas o no con progesterona previo al protocolo. A partir de las tasas de preñez de los animales tratados con progesterona se encontraron los resultados para la finca A= 6.60%, finca B= 10.80%, finca C= 3.90% y finca D= 3.70%, evidenciándose así que esta biotecnología reproductiva es rentable y contribuye positivamente a la cadena productiva del ganado vacuno de carne. Con base en los resultados, se puede concluir que el uso de P4 (inyectable) fue exitoso en la tasa de embarazo.(AU)

Background: Fixed Time Artificial Insemination (FTAI) has achieved a significant evolution in the last 18 years, however, despite the progress achieved by modern FTAI programs, the conception rates obtained are still low. Therefore, this study aimed to evaluate the interrelation between progesterone levels in the periovulatory period and reproductive parameters of Nellore cows submitted to an FTAI protocol. Materials, Methods & Results: On a random day, called day 0 (D0), 57 cows received a P4 device associated with the intramuscular (IM) application of 2.0 mg of estradiol benzoate. On D9, the P4 devices were removed and then were administered 500 µg of cloprostenol sodium IM; 0.6 mg of estradiol cypionate IM and 300 IUI of Equine Chorionic Gonadotrophin IM. Blood samples were collected for the determination of serum P4 concentrations on D9 and D11 of the protocol. The evaluations of follicular diameter (DFOL), follicular wall area (AFOL) and the vascularization area of the follicle wall (VFOL) were carried out on D11 using B-mode ultrasonography examination and colour Doppler, and then the artificial inseminations were performed. The evaluation of the corpus luteum diameter (CLD), of the total corpus luteum area (CLA), of the area of corpus luteum vascularization (CLV) and blood sampling for determination of postovulatory P4 levels (Post-P4) were performed on D24. For the analysis of the P4 concentration the chemiluminescence method was used, with a sensitivity of 0.1 ng/mL. According to the P4 concentrations on D11, cows were divided into 2 groups, LOW LEVELS OF P4 and HIGH LEVELS OF P4. The diagnosis of pregnancy was performed using transrectal ultrasonography on D45, at this point the cows were divided into 2 groups, PREGNANT and NON-PREGNANT. The correlation between DFOL and P4 dosage on D11 was moderate, negative and significant and between the AFOL and the serum P4 levels on D9, was moderate, negative and significant. As for the other correlations between follicular and luteal parameters and serum P4 levels, these were low to moderate, negative and not significant. Cows in the LOW LEVELS OF P4 group had significantly larger diameter and follicular areas than the cows in the HIGH LEVELS OF P4 group, the other follicular and luteal parameters showed no statistical difference. Of the total 57 cows that were inseminated, 30 cows became pregnant. Cows in the PREGNANT group had serum P4 levels on D9 equivalent to that obtained by the NON-PREGNANT group. However, at D11 the cows that became pregnant presented significantly lower serum P4 levels than cows that did not become pregnant. Discussion: The results of the interrelation between follicular parameters and P4 levels obtained in the present study, pointed out that the lower the levels of P4, the higher the follicular parameters, corroborating with other authors. Thus, larger preovulatory follicles provided high ovulation rates. Periovulatory serum P4 levels did not significantly affect the morphofunctional parameters of the CL. Such findings may be justified by high periovulatory P4 levels resulting from less efficient luteolysis, exert a negative effect on the results of FTAI protocols, because progesterone inhibits the release of LH pulses. It is concluded that lower periovulatory P4 levels established a favourable condition for follicular development and fertility, however, morphofunctional parameters of the corpus luteum were not affected.

This study aimed to determine the efficiency of estradiol cypionate (EC) as an ovulation inducer in a Timed Artificial Insemination protocol. 69 buffalo cows received an intravaginal progesterone device and 2mg of estradiol benzoate (EB) at D0. On D9, the intravaginal device was removed and 0.53mg of prostaglandin (PGF2α) and 400UI of equine chorionic gonadotrophin (eCG) were applied. The cows were distributed into two groups: the first group received 1mg of EC (ECG) in D9, and the second group received 1mg of EB (EBG) in D10. Inseminations occurred on D11. Ovarian activity and pregnancy diagnosis were analyzed by ultrasonography. There was no difference (P>0.05) in follicular diameter (9.6 ± 0.89mm vs. 10.7 ± 1.12mm; P=0.06), in ovulation rate (90.9% vs. 90.9%; P=1) and pregnancy rate (58.8% vs. 62.9%; P=0.79), however, buffalo cows from the ECG treatment have less time between P4 removal and ovulation when compared to EBG buffalos (37.4h vs. 52.8h; P=0.001), respectively. Thus, it was concluded that the implantation of TAI in the floodplain of Amazonas is feasible and the use of EC results in successful rates, similar to EB.(AU)


O objetivo deste trabalho foi determinar a eficiência do cipionato de estradiol (CE) como indutor de ovulação em um protocolo de inseminação artificial de tempo fixo. Para isso, 69 búfalas receberam no D0 um dispositivo intravaginal de progesterona e 2mg de benzoato de estradiol (BE). No D9, o dispositivo intravaginal foi removido e foram aplicados 0,53mg de prostaglandina (PGF2α) e 400UI de gonadotrofina coriônica equina (eCG), para, então, os animais serem divididos em dois grupos: um deles (GCE) recebeu 1mg de CE no D9, e o outro (GBE) recebeu 1mg de BE 24h após. As inseminações ocorreram no D11. A atividade ovariana e o diagnóstico de prenhez foram avaliados por ultrassonografia. Não houve diferença (P>0,05) no diâmetro folicular (9,6 ± 0,89mm vs. 10,7 ± 1,12mm; P=0,06), na taxa de ovulação (90,9% vs. 90,9%; P=1) e na taxa de prenhez (58,8% vs. 62,9%; P=0,79), no entanto búfalas do tratamento GCE apresentaram menor tempo entre a remoção da P4 e a ovulação, quando comparadas com as búfalas do GBE (37,4h vs. 52,8h; P=0,001), respectivamente. A implantação da IATF nas várzeas do Amazonas é viável e a utilização do CE resulta em taxas de sucesso similares ao BE.(AU)

We aimed to compare the effect of three estradiol benzoate (EB) doses on follicular wave emergence (FWE) and dominant follicle growth of suckled Nelore cows submitted to TAI (D0). On a random day of estrous cycle (D−10), multiparous (MULT; n=36) and primiparous (PRIM; n=20) suckled Nelore cows received an intravaginal progesterone (P4) device and were assigned in three groups. Cows in the EB-1 (n=20), EB-1.5 (n=15) or EB-2 (n=21) groups received, respectively, an im treatment with 1, 1.5 or 2 mg EB. A subgroup (n=10-13 cows/group) were subject to daily ovarian evaluations from D−10 to D0. On D−2, P4 devices were removed, and all cows received the same treatment: 1 mg estradiol cypionate, 0.53 mg sodium cloprostenol, and 300 IU eCG. Statistical analyses were performed considering only the main effects of treatment group and parity order. The proportion of cows with a synchronized FWE and the moment of the FWE did not differ (p>0.05) among the treatment groups (overall: 80% [28/35] and 4.1 ± 0.4 days); however, the FWE occurred earlier (p=0.007) in MULT (3.8 ± 0.2 days) than PRIM (5.1 ± 0.4) cows. The proportion of animals detected in estrus was greater (86% [31/36] vs. 70% [14/20]; p=0.02) and the dominant follicle was larger on D−2 (9.7 ± 0.3 mm vs. 7.8 ± 0.7 mm; p=0.006) and D0 (11.9 ± 0.4 mm vs. 10 ± 0.5 mm; p=0.008) in MULT than PRIM cows. In conclusion, the three EB doses presented similar efficiency to synchronize the FWE in suckled Nelore cows. Moreover, a delayed FWE and smaller dominant follicle is observed in PRIM cows, contributing to the reduced reproductive performance in this parity category when using similar TAI protocols of MULT cows.

Background: The indiscriminate use of drugs is an issue in Veterinary Medicine, as it has serious consequences for theanimals. Many drugs are myelotoxic and cause a decrease in the production of blood cells, which may be irreversible insome cases. The present work reports a case of pancytopenia induced by the concomitant use of myelotoxic drugs (estrogen, metamizole and phenobarbital) in a dog and describes findings on myelotoxicity, hematological alterations andtreatment success.Case: A 7-year-old Lhasa Apso bitch was referred to the Veterinary Hospital of Federal University of Paraná, Curitibacampus, with hematuria and a history of treatment with phenobarbital [2 mg/kg twice a day (bis in die, BID)], metamizole[25 mg/kg 3 times a day (ter in die, TID)], and use of estrogen hormone (estradiol cypionate). At physical examination, theanimal was normohydrated and exhibited normal palpable lymph nodes, pale mucous membranes, galactorrhea, and a bodytemperature of 36°C. A complete blood count including reticulocyte count and a total plasma protein (TPP) exam wererequested. The results revealed pancytopenia (18% hematocrit, 1,400 total leucocytes/µL, and 22,000 reticulocytes/µL).An abdominal ultrasound exam did not detect any relevant alterations. In view of the results obtained, medullary aplasiawas suspected. A bone marrow aspiration was performed. A myelogram revealed a decrease in cellularity (erythrocyticand granulocytic hypoplasia), with presence of rare erythroid and granulocytic precursors. The diagnosis was medullaryaplasia. The animal was treated, and the evolution of the hematological alterations was monitored. The treatment consistedof administration of erythropoietin (100UI/kg subcutaneously every 48 h), prednisone (2 mg/kg BID), Leucogen (3 mg/kg BID), interferon (0.2 IU/kg BID) and Eritrós Dog Tabs [1 tablet once a day (semel in die, SID)]. The present work reports a case of pancytopenia induced by the concomitant use of myelotoxic drugs (estrogen, metamizole and phenobarbital) in a dog and describes findings on myelotoxicity, hematological alterations andtreatment success.Case: A 7-year-old Lhasa Apso bitch was referred to the Veterinary Hospital of Federal University of Paraná, Curitibacampus, with hematuria and a history of treatment with phenobarbital [2 mg/kg twice a day (bis in die, BID)], metamizole[25 mg/kg 3 times a day (ter in die, TID)], and use of estrogen hormone (estradiol cypionate). At physical examination, theanimal was normohydrated and exhibited normal palpable lymph nodes, pale mucous membranes, galactorrhea, and a bodytemperature of 36°C. A complete blood count including reticulocyte count and a total plasma protein (TPP) exam wererequested. The results revealed pancytopenia (18% hematocrit, 1,400 total leucocytes/µL, and 22,000 reticulocytes/µL).An abdominal ultrasound exam did not detect any relevant alterations. In view of the results obtained, medullary aplasiawas suspected. A bone marrow aspiration was performed. A myelogram revealed a decrease in cellularity (erythrocyticand granulocytic hypoplasia), with presence of rare erythroid and granulocytic precursors. The diagnosis was medullaryaplasia. The animal was treated, and the evolution of the hematological alterations was monitored. The objective was to determine the effect of some factors on pregnancy rate of fixed-time embryo transfer (FTET), in cows and heifers kept under Mexican tropical conditions. Recipients females (n=405) grazing in pastures were selected according to breed group (Zebu and crosses), parity (nulliparous and multiparous), body condition score (BCS) and the presence of a corpus luteum (CL). The females were synchronized on day 0 using a progesterone vaginal device and 2 mg estradiol benzoate (EB), two groups were established. Group 1 (conventional protocol) were animals in which the progesterone device was removed on day 7. At this time, also received an injection of 50 mg cloprostenol sodium and 1 mg estradiol cypionate. Animals also received 300 IU (heifers) or 360 IU (cows) of eCG. Group 2 (J-Synch protocol) were animals in which the progesterone device was removed on day 6. Cloprotenol and eCG injections were applied as in Group 1. Additionally, on day 9, animals of group 2 received 0.01 mg buserelin acetate. Embryo transfer of in vivo or in vitro was done on day 16 and pregnancy diagnosis was realized by ultrasonography on days 23 and 53 after FTET. Statistical analyses were carried out using Chi-square tests and logistic regression. Pregnancy rate varied between farms (P0.05). However, the logistic regression determined that the only significant factor on pregnancy rate was the type of embryo. In conclusion, pregnancy rate in FTET females was higher for in vivo embryos than for in vitro embryos in cows evaluated under humid tropical conditions in Mexico.

However, the logistic regression determined that the only significant factor on pregnancy rate was the type of embryo. In conclusion, pregnancy rate in FTET females was higher for in vivo embryos than for in vitro embryos in cows evaluated under humid tropical conditions in Mexico.(AU)

The various studies on the manipulation of the estrous cycle culminated in the development of estrous and ovulation synchronization protocols, to the point of defining an optimum moment for artificial insemination. The induction of ovulation is a primordial part for the determination of the moment of the TAI, so to study which hormones and how they behave allows a better understanding of this physiological process and, to manipulate it efficiently. The aim of the present study was to compare the use of estradiol benzoate (EB) and cypionate (ECP) as ovulation inducer in Girolando cows submitted to Timed Artificial Insemination (TAI). Ovulations of 108 Girolando cows were synchronized and the initial day of treatment, recorded as D0, which was when the animals received an intravaginal device containing 1g of progestogen and an intramuscular application (im) of 2mg of EB. After 8 days (D8), the device was removed and 500µg of cloprostenol (im) was administered to all females. Then, the animals were randomly divided into two treatments: BG Group (n = 52) and CG Group (n = 56). The CG animals received 1mg of ECP (im) as the ovulation inducer at the time of device removal, while BG Group cows received 1mg of EB (im) 24 h later (D9). TAI occurred at D10. After device removal, ultrasound evaluations were performed every 12 h up to ovulation. The following reproductive parameters were evaluated: interval from intravaginal device removal to ovulation - IDO (hours); interval from ovulation to TAI-IOT (hours); diameter of the largest follicle at intravaginal device removal (mm); maximum diameter of the DF (mm) at D10; the dominant follicle growth rate (mm/day); synchronization rate - SR (%); ovulation rate - OR (%) and PR - pregnancy rate (%). Only the ovulation rate presented a statistical difference (p<0.05). In conclusion, despite the ovulation rate difference, both estradiol esters administered were effective and presented similar pregnancy rates in Girolando cows submitted to TAI.


Os diversos estudos sobre manipulação do ciclo estral culminaram com o desenvolvimento de protocolos de sincronização do estro e da ovulação, a ponto de se definir um momento ótimo para a inseminação artificial. A indução da ovulação é parte primordial para a determinação do momento da IATF. Dessa forma, estudar quais hormônios e como eles se comportam permite compreender melhor esse processo fisiológico e, ainda, manipulá-lo de forma eficiente. O objetivo do presente estudo foi comparar o uso de benzoato de estradiol (BE) e cipionato (ECP) em vacas Girolando submetidas à inseminação artificial em tempo fixo (IATF). As ovulações de 108 vacas Girolando foram sincronizadas e no início do tratamento, D0, os animais receberam um dispositivo intravaginal contendo 1g de progestágeno e uma aplicação intramuscular (im) de 2mg de BE. No D8, o dispositivo foi removido e 500µg de cloprostenol (im) foram administrados a todas as fêmeas. Depois disso, os animais foram divididos aleatoriamente em dois tratamentos: Grupo GB (n = 52) e Grupo GC (n = 56). Os animais do Grupo GC receberam 1mg de ECP (im) como indutor de ovulação no momento da retirada do dispositivo, enquanto as vacas do Grupo GB receberam 1mg de BE (im) 24 horas depois (D9). IATF ocorreu em D10. Após a remoção do dispositivo, as avaliações ultrassonográficas foram realizadas a cada 12 horas até a ovulação. Os parâmetros reprodutivos avaliados foram: intervalo entre a retirada do dispositivo intravaginal de P4 e a ovulação- IRD (horas); intervalo entre a ovulação e a IATF- IOI (horas); diâmetro do maior folículo na retirada do dispositivo intravaginal de P4 (mm); taxa de crescimento do FD-TCFD (mm/dia); taxa de sincronização - TS (%); taxa de ovulação- TO (%) e taxa de concepção - TC (%). Apenas a taxa de ovulação apresentou diferença estatística (p<0.05). Em conclusão, apesar da diferença na taxa de ovulação, ambos os ésteres de estradiol administrados foram eficazes e apresentaram taxas de prenhez semelhantes em vacas Girolando submetidas a IATF.

Background: Toxicological events in animals may be accidental or intentional and could occur in the home environment.These events could involve different agents such as pesticides, rodenticides, medicines, foods, and plants. Indiscriminateuse of medication in pets by their owners is common. Self-medication of animals with drugs for human use can cause irreversible damage to their health. The emergency contraceptive pill (morning-after pill), comprising concentrated hormonalcompounds, is sold freely in Brazil. The objective of this article was to report a case of pyometra in a bitch possibly causedby simultaneous administration of human emergency contraceptive pill along with a veterinary contraceptive medication.Case: A 6-year-old bitch was referred to a Veterinary Medical Teaching Hospital for abdominal enlargement. Accordingto the guardian, during estrus (30 days before the consultation) the female dog was covered. After intercourse, the owneradministered a human emergency contraceptive pill (morning-after pill - Levonorgestrel) and a veterinary contraceptive(Estradiol Cypionate). Physical examination revealed severe dehydration (10%) and high temperature, besides the abdominalenlargement. The hemogram revealed leukocytosis with a left shift, characteristic of infection. After clinical examination,closed pyometra was suspected, and later confirmed by ultrasonography. Due to the poor clinical condition, fluid therapywas performed, and a systemic antibiotic was administered. After stabilization, the patient underwent ovariohysterectomy.In the postoperative period, the fluid therapy was continued, and antibiotic and anti-inflammatory drugs were administered.Two days after surgery, the patient was discharged for home recovery.Discussion: Most cases of drug poisoning in animals involve female dogs, similar to the patient in this report. There isno theory elucidating the same, but the rate of incidence in female dogs has been reported to be higher...

Tanzania e suplementadas com silagem de milho, receberam o equivalente a 0,5 mg/animal/dia de MGA durante 14 dias. No dia 5, os animais foram divididos aleatoriamente em dois grupos. O grupo 1 (n=21) recebeu uma injeção i.m. de 2,0 mg de ECP enquanto que o grupo 2 (n=19) recebeu 1 ml de solução salina. O cio (detecção visual) foi registrado em todos os animais no período de 120 horas após a retirada do MGA, mas somente os animais do grupo 1 foram inseminados. Os animais do grupo 2 receberam 150 μg do análogo de PGF2α, cloprostenol 17 dias após a retirada do MGA e inseminados no cio induzido. Em ambos grupos, as vacas não detectadas no cio durante o dia foram colocadas com um touro durante a noite para uma eventual cobertura. O diagnóstico de prenhez foi realizado, por ultrassonografia, 40 dias após a IA. Os dados foram analisados pelo teste de Qui-quadrado. Oito (38,1%) animais do grupo 1 e 7 (36,8%) do grupo 2 apresentaram cio após a retirada do MGA (P>0,05). A taxa de concepção dos animais inseminados foi de 37,5% e 50,0% para os grupos 1 e 2, respectivamente (P>0,05). Esse índice aumentou para 58,3% (grupo 1) e 64,3% (grupo 2) quando foram consideradas as vacas cobertas pelo touro durante a noite. Esses resultados sugerem que é possível aproveitar o primeiro cio de fêmeas tratadas com ECP no dia 5 de um tratamento com MGA durante 14 dias. Adicionalmente, considerando o elevado número de animais cobertos pelo touro durante a noite, recomenda-se o uso de ferramentas auxiliares para identificar os animais que apresentam cio noturno ou, ainda, utilizar a monta natural em associação com a IA.

Chi-square analyses were used to detect differences between groups. Eight (38.1%) animals of group 1 and 7 (36.8%) of group 2 showed estrus after withdrawal of MGA (P>0.05). The conception rates in groups 1 and 2 were, respectively, 37.5% and 50.0% for inseminated cows and 58.3% and 64.3% for for inseminated cows in addition to mated by the bull at night (P>0.05). These results suggest that it is possible to use the first estrus of females treated with ECP on day 5 of a 14-day MGA treatment. Furthermore, considering the large number of cows mated by the bull at night, it is recommended to use estrus detection tools as an aid for the identification of such animals or to combine AI and bulls for breeding.

In both groups, cows without estrus detection during the day stayed with a bull at night for possible mating. Pregnancy was diagnosed by ultrasound examination 40 days after AI. Chi-square analyses were used to detect differences between groups. Eight (38.1%) animals of group 1 and 7 (36.8%) of group 2 showed estrus after withdrawal of MGA (P>0.05). The conception rates in groups 1 and 2 were, respectively, 37.5% and 50.0% for inseminated cows and 58.3% and 64.3% for for inseminated cows in addition to mated by the bull at night (P>0.05). These results suggest that it is possible to use the first estrus of females treated with ECP on day 5 of a 14-day MGA treatment. Furthermore, considering the large number of cows mated by the bull at night, it is recommended to use estrus detection tools as an aid for the identification of such animals or to combine AI and bulls for breeding.(AU)

Resumo

The various studies on the manipulation of the estrous cycle culminated in the development of estrous and ovulation synchronization protocols, to the point of defining an optimum moment for artificial insemination. The induction of ovulation is a primordial part for the determination of the moment of the TAI, so to study which hormones and how they behave allows a better understanding of this physiological process and, to manipulate it efficiently. The aim of the present study was to compare the use of estradiol benzoate (EB) and cypionate (ECP) as ovulation inducer in Girolando cows submitted to Timed Artificial Insemination (TAI). Ovulations of 108 Girolando cows were synchronized and the initial day of treatment, recorded as D0, which was when the animals received an intravaginal device containing 1g of progestogen and an intramuscular application (im) of 2mg of EB. After 8 days (D8), the device was removed and 500g of cloprostenol (im) was administered to all females. Then, the animals were randomly divided into two treatments: BG Group (n = 52) and CG Group (n = 56). Os diversos estudos sobre manipulação do ciclo estral culminaram com o desenvolvimento de protocolos de sincronização do estro e da ovulação, a ponto de se definir um momento ótimo para a inseminação artificial. A indução da ovulação é parte primordial para a determinação do momento da IATF. Dessa forma, estudar quais hormônios e como eles se comportam permite compreender melhor esse processo fisiológico e, ainda, manipulá-lo de forma eficiente. O objetivo do presente estudo foi comparar o uso de benzoato de estradiol (BE) e cipionato (ECP) em vacas Girolando submetidas à inseminação artificial em tempo fixo (IATF). As ovulações de 108 vacas Girolando foram sincronizadas e no início do tratamento, D0, os animais receberam um dispositivo intravaginal contendo 1g de progestágeno e uma aplicação intramuscular (im) de 2mg de BE. No D8, o dispositivo foi removido e 500g de cloprostenol (im) foram administrados a todas as fêmeas. Depois disso, os animais foram divididos aleatoriamente em dois tratamentos: Grupo GB (n = 52) e Grupo GC (n = 56). Resumo

No Brasil, cerca de 5% do rebanho bovino é reproduzido através da biotecnologia de inseminação artificial (IA). O sucesso da IA depende de fatores inerentes à fêmea, ao sêmen e a habilidade do inseminador. Dentre os fatores inerentes a fêmea destaca-se: escore corporal, estado sanitário, estado reprodutivo e protocolo hormonal utilizado. A literatura relata diferentes resultados entre os diversos protocolos hormonais. Desta forma, este trabalho teve por objetivo verificar a eficácia do uso do eCG em protocolos de inseminação artificial em tempo fixo (IATF) em bovinos. Foram utilizadas vacas mestiças Girolando (n=4886), com peso médio de 500kg, as quais foram divididas em dois grupos, IATF 1 (n=2,535) e IATF 2 (n=2,351). Os dados das amostras foram coletados dasUnidades Produtoras de Leite (UPLs), as quais fazem parte do complexo leiteiro da Fazenda. As vacas foram divididas em dois grupos, IATF 1 e IATF 2 a cada trimestre. Ambos os protocolos de IATF foram realizados durante um período de dez dias. No dia 0 foi administrado o implante intrauterino impregnado com progesterona (1g), o qual permaneceu durante oito dias e administrado 2 mL de Benzoato de Estradiol pela via intramuscular. Após a remoção do implante no D8, administrou-se por via intramuscular 2 mL de prostaglandina F2α (PGF2α) juntamente com 1 mL de Cipionato de Estradiol. Nas fêmeas IATF 1 com folículos menos desenvolvidos aplicou-se 2 mL de gonadrotofina coriônica equina (eCG) por via intramuscular. Concluiu-se que a utilização de eCG elevou as taxas de fertilização em vacas com folículos menores, porém foi desnecessário para as fêmeas que apresentaram condições uterinas normais.


In Brazil, about 5% of the cattle herd reproduces through artificial insemination (AI) biotechnology. The success of AI depends on factors inherent to the female, inherent to the semen and the inseminator's ability. Among the factors inherent to the female are: body score, health status, reproductive status and hormonal protocol used. The literature reports different results among the various hormonal protocols. Therefore, the objective of this study was to verify the efficacy of eCG in fixed-time artificial insemination (IATF) protocols in cattle. Girolando crossbred cows (n=4886), with an average weight of 500 kg, were divided into two groups, IATF 1 (n=2.535) and IATF 2 (n=2.351). The data of the samples were collected from the Milk Producing Units (UPLs) - Campo Grande, Colmalândia, Ouro Branco and Ouro Verde - that are part of Fazenda dairy complex. Cows were divided into two groups, IATF 1 and IATF 2 each quarter. Both IATF protocols were performed over a ten day period. On day 0, the progesterone-impregnated intrauterine implant (1g) was administered, which remained for eight days and administered 2 mL of Estradiol Benzoate intramuscular. After removal of the implant in the D8, 2 mL of prostaglandin F2α (PGF2α) were administered intramuscularly together with 1 mL of Estradiol Cypionate. It was concluded that the use of eCG increased fertilization rates in cows with smaller follicles, but was unnecessary for females that presented normal uterine conditions.
