RESUMO

The objective of this research was to evaluate circulating concentrations of plasma cortisol and measures of temperament at weaning in calves (steers and heifers) and at transport in steers. Calves ( = 993) were produced from a 3-breed diallel mating design that included calves from 3 consecutive years. Breed types of calves were straightbred Angus (A), Brahman (B), and Romosinuano (R) and all F crossbred combinations (AB, BA, AR, RA, BR, and RB). At weaning (d 0) and at 24 and 72 h after weaning, blood was sampled from calves and the plasma was stored for later cortisol assay. Additionally, at each of these times, temperament was assessed as chute score, exit velocity, and pen score. About 1 mo later, steer calves ( = 471) were sampled before shipment, at arrival, and at 24 h, 72 h, 2 wk, and 4 wk after shipment (2,025 km; Brooksville, FL, to El Reno, OK). At each of these sampling times, blood was collected and plasma was stored for subsequent cortisol assay and temperament was assessed by measurement of exit velocity. At both weaning and transport, plasma concentrations of cortisol did not significantly differ ( > 0.05) among straightbreds or among crossbreds. Significant ( < 0.05) positive genetic effects were observed for plasma concentration of cortisol at weaning (heterosis for BA and direct Romosinuano effect) and transport (heterosis for RA, BR, and BA; direct Romosinuano effect; and maternal Angus effect). Assessment of temperament using the objective measurement of exit velocity or the subjective measures of chute score or pen score (1 [lowest] to 5 [highest excitability] scale, based on behavior in chute and behavior in pen with human observer, respectively) generally provided similar results: Brahman was higher than Brahman crosses, which were higher than Angus, Romosinuano, and their reciprocal crosses. For exit velocity, however, Brahman did not differ from Brahman crosses and Angus did not differ from Romosinuano or Brahman crosses. At transport, sire breed and dam breed affected exit velocity of steers, with higher ( < 0.05) estimates for Brahman than for Romosinuano or Angus. These data suggest that weaned calves and shipped steers of various breed types show a similar response to stressors in cortisol concentration. In contrast, in assessing temperament or behavioral response to humans, Romosinuano and Angus had better temperaments and were less excitable than Brahman.

AIM: Approximately 20% of patients treated with neoadjuvant chemoradiotherapy (nCRT) for locally advanced rectal cancer achieve a pathological complete response (pCR) while the remainder derive the benefit of improved local control and downstaging and a small proportion show a minimal response. The ability to predict which patients will benefit would allow for improved patient stratification directing therapy to those who are likely to achieve a good response, thereby avoiding ineffective treatment in those unlikely to benefit. METHOD: A systematic review of the English language literature was conducted to identify pathological factors, imaging modalities and molecular factors that predict pCR following chemoradiotherapy. PubMed, MEDLINE and Cochrane Database searches were conducted with the following keywords and MeSH search terms: 'rectal neoplasm', 'response', 'neoadjuvant', 'preoperative chemoradiation', 'tumor response'. After review of title and abstracts, 85 articles addressing the prediction of pCR were selected. RESULTS: Clear methods to predict pCR before chemoradiotherapy have not been defined. Clinical and radiological features of the primary cancer have limited ability to predict response. Molecular profiling holds the greatest potential to predict pCR but adoption of this technology will require greater concordance between cohorts for the biomarkers currently under investigation. CONCLUSION: At present no robust markers of the prediction of pCR have been identified and the topic remains an area for future research. This review critically evaluates existing literature providing an overview of the methods currently available to predict pCR to nCRT for locally advanced rectal cancer. The review also provides a comprehensive comparison of the accuracy of each modality.

RESUMO

We summarize evidence that input to the apical tufts of neocortical pyramidal cells modulates their response to basal input. Because this apical amplification and disamplification provide intracortical mechanisms for prioritization, Mather and colleagues' arguments suggest that their effects are enhanced by noradrenergic arousal. Though that is likely, it has not yet been adequately studied. Their article shows that it should be.

Neocortical pyramidal cells can integrate two classes of input separately and use one to modulate response to the other. Their tuft dendrites are electrotonically separated from basal dendrites and soma by the apical dendrite, and apical hyperpolarization-activated currents (Ih) further isolate subthreshold integration of tuft inputs. When apical depolarization exceeds a threshold, however, it can enhance response to the basal inputs that specify the cell's selective sensitivity. This process is referred to as apical amplification (AA). We review evidence suggesting that, by regulating Ih in the apical compartments, adrenergic arousal controls the coupling between apical and somatic integration zones thus modifying cognitive capabilities closely associated with consciousness. Evidence relating AA to schizophrenia, sleep, and anesthesia is reviewed, and we assess theories that emphasize the relevance of AA to consciousness. Implications for theories of neocortical computation that emphasize context-sensitive modulation are summarized. We conclude that the findings concerning AA and its regulation by arousal offer a new perspective on states of consciousness, the function and evolution of neocortex, and psychopathology. Many issues worthy of closer examination arise.

RESUMO

A broad neuron-centric conception of contextual modulation is reviewed and re-assessed in the light of recent neurobiological studies of amplification, suppression, and synchronization. Behavioural and computational studies of perceptual and higher cognitive functions that depend on these processes are outlined, and evidence that those functions and their neuronal mechanisms are impaired in schizophrenia is summarized. Finally, we compare and assess the long-term biological functions of contextual modulation at the level of computational theory as formalized by the theories of coherent infomax and free energy reduction. We conclude that those theories, together with the many empirical findings reviewed, show how contextual modulation at the neuronal level enables the cortex to flexibly adapt the use of its knowledge to current circumstances by amplifying and grouping relevant activities and by suppressing irrelevant activities.

The cyclic-AMP response element binding (CREB) protein has been shown to have a pivotal role in cell survival and cell proliferation. Transgenic rodent models have revealed a role for CREB in higher-order brain functions, such as memory and drug addiction behaviors. CREB overexpression in transgenic animals imparts oncogenic properties on cells in various tissues, and aberrant CREB expression is associated with tumours. It is the central position of CREB, downstream from key developmental and growth signalling pathways, which gives CREB this ability to influence a spectrum of cellular activities, such as cell survival, growth and differentiation, in both normal and cancer cells. We show that CREB is highly expressed and constitutively activated in patient glioma tissue and that this activation closely correlates with tumour grade. The mechanism by which CREB regulates glioblastoma (GBM) tumour cell proliferation involves activities downstream from both the mitogen-activated protein kinase and phosphoinositide 3-kinase (PI3K) pathways that then modulate the expression of three key cell cycle factors, cyclin B, D and proliferating cell nuclear antigen (PCNA). Cyclin D1 is highly CREB-dependent, whereas cyclin B1 and PCNA are co-regulated by both CREB-dependent and -independent mechanisms. The precise regulatory network involved appears to differ depending on the tumour-suppressor phosphatase and tensin homolog status of the GBM cells, which in turn allows CREB to regulate the activity of the PI3K itself. Given that CREB sits at the hub of key cancer cell signalling pathways, understanding the role of glioma-specific CREB function may lead to improved novel combinatorial anti-tumour therapies, which can complement existing PI3K-specific drugs undergoing early phase clinical trials.

Two experiments were conducted to examine the effect of growth rate to similar age or BW on fat deposition in stocker cattle grazing dormant native range (DNR) or winter wheat pasture (WP). In each experiment, fall-weaned Angus steers were randomly allotted to 1 of 4 stocker production programs: 1) control, 1.02 kg/d of a 40% CP cottonseed meal-based supplement during grazing of DNR (CON); 2) corn/soybean meal-based supplement fed at 1% of BW during grazing of DNR (CORN); 3) grazing WP at a high stocking rate to achieve a low rate of BW gain (LGWP); and 4) grazing WP at a low stocking rate to achieve a high rate of BW gain (HGWP). In Exp. 1, a subset of steers (3 steers per treatment) was harvested after winter grazing (138 d) at similar age. The remaining WP steers were transitioned into the finishing phase, whereas DNR steers were allowed to graze the same native range pastures for another 115 d without supplementation before entering the feedyard. In Exp. 2, steers grazed their respective pastures until each treatment reached an estimated HCW of 200 kg (262, 180, 142, and 74 d, respectively, for the CON, CORN, LGWP, and HGWP treatments), at which time a subset of steers (4 steers per treatment) were selected for intermediate harvest before finishing. In both experiments, the remaining steers were fed a finishing diet to a common 12th-rib fat thickness of 1.27 cm. In Exp. 1, winter grazing ADG was 0.19, 0.52, 0.68, and 1.37 ± 0.03 kg/d; and in Exp. 2, winter/summer grazing ADG was 0.46, 0.61, 0.83, and 1.29 ± 0.02 kg/d, respectively for CON, CORN, LGWP, and HGWP treatments. At intermediate harvest in Exp. 1, HGWP steers had greater (P < 0.01) 12th-rib fat thickness and marbling scores, compared with the other treatments. However, in Exp. 2, LGWP steers had greater (P < 0.01) marbling scores compared with HGWP steers, which were greater than DNR steers. At final harvest in Exp. 1, LGWP steers had greater (P < 0.01) 12th-rib fat thickness and smaller LM area, compared with the other treatments; however, there were no differences (P = 0.99) in final marbling scores. In Exp. 2, CON steers had lower (P < 0.05) 12th-rib fat thickness and tended (P = 0.10) to have greater marbling scores, compared with the other treatments. These data suggest that changes in the partitioning of fat among depots during the stocker phase may not be reflected after finishing when steers are fed to a common 12th-rib fat thickness.

The objectives of this work were to estimate heterosis and breed genetic effects for carcass quantity, quality, and palatability traits of steers (Bos spp.) produced from matings of Romosinuano, Brahman, and Angus cattle. Steers (n = 464) were weaned at 7 mo of age and transported to the Southern Great Plains where they grazed winter wheat for 6 mo and were then fed a finishing diet until serial slaughter after different days on feed (average 130 d). Carcass quality and quantity traits were measured; steaks (aged 7 d) were obtained for palatability evaluation. Heterosis was detected for BW, HCW, dressing percentage, LM area, and yield grade for all pairs of breeds. Generally, Romosinuano-Angus heterosis estimates were smallest, Romosinuano-Brahman estimates were intermediate, and Brahman-Angus heterosis estimates were largest. The direct Romosinuano effect was to decrease (P < 0.05) BW (-67 ± 16 kg), HCW (-48 ± 10 kg), dressing percentage (-1.4 ± 0.5 units), 12th rib fat thickness (-5.2 ± 0.8 mm), and yield grade (-0.9 ± 0.1), and to increase LM area per 100 kg HCW (3.6 ± 0.3 cm(2)/100 kg). Significant Brahman direct effects were detected for BW (34 ± 17 kg), HCW (29 ± 10 kg), dressing percentage (1.6 ± 0.6 %), LM area per 100 kg HCW (-3.3 ± 0.4 cm(2)/100 kg), and yield grade (0.6 ± 0.1). Significant Angus direct effects were to increase 12th rib fat thickness (3.8 ± 1 mm). Among sire breed means, Romosinuano had reduced (P = 0.002) marbling score (393 ± 9) than Angus, but greater mean sensory tenderness scores (5.8 ± 0.1), and reduced percentage Standard carcasses (10 ± 2%) than Brahman (P < 0.002). Angus sire breed means for marbling score (475 ± 10), overall tenderness (5.8 ± 0.1), and percentage Choice carcasses (75 ± 5%) were greater (P < 0.05) than Brahman sire breed means (360 ± 11, 5.4 ± 0.1, 31 ± 5%). From consideration only of characteristics of the end product of beef production, Romosinuano did not provide a clearly superior alternative to Brahman for U.S. producers, as they had some quality and palatability advantages relative to Brahman, but at lighter HCW.

Beef cows in the subtropical USA must be adapted to the stressors of the environment, typically supplied by using Brahman (Br) breeding. Calves produced in the region, however, are usually grown and finished in more temperate regions, and have a perceived reputation for poor ADG and feed efficiency during finishing. Compromised fertility and carcass quality often associated with the Br have increased interest in tropically adapted Bos taurus breed types. The objective of this study was to evaluate 3 breeds [An = Angus (Bos taurus, temperate); Br (B. indicus, tropical); and Ro = Romosinuano (B. taurus, tropical)] and all possible crosses during various segments of post-weaning growth, and for feed efficiency during the finishing phase. Steer calves (n = 473) born over 3 yr were weaned in late September, backgrounded for at least 21 d (BKG), shipped 2,025 km to El Reno, OK, in October, fed a preconditioning diet for 28 d (RCV), grazed wheat (Triticum aestivum L.) pasture from November to May (WHT), finished on a conventional feedlot diet (FIN), and serially harvested after approximately 95, 125, and 150 d on feed. Body weight and ADG during each segment were tested using a mixed model that included calf age at weaning, year (Y), breed of sire (SB), breed of dam (DB), and interactions. In addition, winter treatment (continuous wheat or reduced grazing of wheat with supplement) was included for the wheat and feedlot phases. Sire within SB × SB [and pen (barn × year) for feedlot phase] were considered random. The SB × DB interaction was significant for all traits (P < 0.01) except exit velocity taken at weaning and ADG during FIN, but both traits were affected by 3-way interactions with Y or harvest group. Tropically-adapted purebred steers had greater (P < 0.01) ADG than AnAn through weaning and BKG in FL but the reverse was true during the RCV and WHT segments. Similar, but less pronounced results were noted for F(1) steers with 100% tropical influence compared with those with only 50%. Heterosis was numerically greater for most traits for An × Br (11 to 64%) compared with An × Ro and Br × Ro (3 to 42%), which were similar. In a subset of the steers (n = 261), G:F was not influenced by level of tropical breeding, but tropically adapted steers were more efficient (P < 0.05) by residual feed intake. No heterosis was evident. These data show that in temperate zones, winter is the period when productivity of tropically adapted cattle is compromised.

In an effort to determine whether tropical adaptation influences circulating concentrations of the growth-related hormone IGF-I, 3-breed diallel matings were conducted using temperate Bos taurus (Angus), tropical Bos indicus (Brahman), and tropical Bos taurus (Romosinuano). Purebred Angus, Braham, and Romosinuano and crossbred Angus-Braham, Angus-Romosinuano, and Braham-Romosinuano heifers and steers were evaluated in 2 separate calf crops from 2003 and 2004. Blood samples were obtained from 10 heifers of each breed group (n = 90) for each year at weaning and on d 0 and 84 of postweaning trials. Samples were also taken from 10 steers of each breed group (n = 90) at weaning and on d 0 and 60 of individual finishing phase feeding trials for each year. Concentrations of IGF-I were determined by RIA. Analyses included effects of sire breed, dam breed, year of record, the age of the dam of the calf in years, and interactions. Age of calf in days was investigated as a linear and quadratic covariate. Separate analyses were conducted for steers and heifers. The direct effect of Angus was to reduce (P < 0.03) heifer concentrations of IGF-I at d 84 and in the repeated measures analysis. In the repeated measures analysis, the direct effect of Romosinuano was to increase concentrations of IGF-I (P = 0.01). Relative to the temperate Bos taurus breed, plasma concentrations of IGF-I were numerically greater in male and female tropically adapted breed groups.

Signal processing in the cerebral cortex is thought to involve a common multi-purpose algorithm embodied in a canonical cortical micro-circuit that is replicated many times over both within and across cortical regions. Operation of this algorithm produces widely distributed but coherent and relevant patterns of activity. The theory of Coherent Infomax provides a formal specification of the objectives of such an algorithm. It also formally derives specifications for both the short-term processing dynamics and for the learning rules whereby the connection strengths between units in the network can be adapted to the environment in which the system finds itself. A central assumption of the theory is that the local processors can combine reliable signal coding with flexible use of those codes because they have two classes of synaptic connection: driving connections which specify the information content of the neural signals, and contextual connections which modulate that signal processing. Here, we make the biological relevance of this theory more explicit by putting more emphasis upon the contextual guidance of ongoing processing, by showing that Coherent Infomax is consistent with a particular Bayesian interpretation for the contextual guidance of learning and processing, by explicitly specifying rules for on-line learning, and by suggesting approximations by which the learning rules can be made computationally feasible within systems composed of very many local processors.

The objective of this study was to compare the fatty acid (FA) composition of intramuscular fat from the LM of 3 divergent breeds of cattle: Angus (AN, n = 9), Brahman (BR, n = 7), and Romosinuano (RM, n = 11). Cattle were blocked by breed and finished 129 d before slaughter in one year and 157 d in the next year. Longissimus muscle samples were collected from each carcass between the 10th and 13th ribs, trimmed of external fat, frozen in liquid nitrogen, homogenized, and used for fat extraction, using a modified Folch procedure. Extracted fat was analyzed for FA by using a GLC system with an HP-88 capillary column. Fatty acid composition was expressed using both a normalized percentage (%) and gravimetric calculation (mg/g of fresh muscle tissue) in relation to degree of saturation, which was determined using a saturation index (ratio of total SFA to total unsaturated FA). Crude fat determination revealed that LM from AN purebred cattle had the greatest amount of intramuscular fat (7.08%; P = 0.001). Although intramuscular fat of LM from RM contained a reduced percentage of total SFA (P = 0.002) compared with AN, it had the greatest percentage of total PUFA (P < 0.001 and P = 0.020). The percentages of total MUFA were similar among the 3 breeds (P = 0.675). The gravimetric calculation, a measure of actual FA concentration, showed significantly greater concentrations of SFA (26.67 mg/g), MUFA (26.50 mg/g), and PUFA (2.37 mg/g) in LM from AN cattle, as compared with LM from BR and RM cattle (P < 0.001). The objective in this study was to assess breed effects in fecal prevalence of Escherichia coli O157:H7 in heifers on a development program in Florida and in their steer half siblings in stocker and feedlot phases in Oklahoma. A secondary objective was to characterize fecal shedding of Campylobacter and Salmonella in subsets of the same samples. After weaning, heifers (n = 501; purebreds and F1 crosses of Angus, Brahman, and Romosinuano) were preconditioned and placed in a local development program. Steers (n = 481) were transported to Oklahoma, where they grazed wheat for 6 months and then were placed in feedlot pens. Fecal samples were obtained at least every 28 days for 12 months on most animals. None of the 10,982 samples tested positive for E. coli O157:H7. Overall fecal prevalences of Campylobacter and Salmonella in heifers were 1.7 and 0.04%, respectively. Corresponding overall prevalences in steer samples were 27.2 and 0.6%. Campylobacter isolates were mostly C. jejuni and were tetracycline resistant. Eight Salmonella isolates were Salmonella Typhimurium that were either quad or penta resistant, most often to ampicillin, chloramphenicol, sulfamexathole, and tetracycline. Feedlot steers had greater odds of positive detection of Campylobacter (odds ratio, 8.5; confidence interval, 3.7, 19.5) than when grazing winter wheat. No breed effect was detected for fecal prevalence of these pathogens.

Because wheat forage contains high concentrations of N, NPN, digestible DM, and water, beef cattle and sheep require an adaptation period before positive BW are seen. The objective of the present experiment was to determine the impact of length of exposure of lambs and steers to wheat forage on BW gains, N retention, and forage digestibility. Sixteen steer calves (average BW = 210 +/- 12 kg) and 20 wether lambs (average BW = 31.5 +/- 2.0 kg) were randomly assigned to 1 of 2 treatment groups. Group 1 grazed a wheat pasture for 120 d during the winter, whereas group 2 was wintered on dormant warm-season grass pastures plus warm-season grass hay and plant-based protein supplements. In the spring (April 5), all lambs and steers grazed wheat pasture for 14 d and were then housed in metabolism stalls and fed freshly harvested wheat forage to determine forage digestibility and N metabolism. Data were analyzed for lambs and steers separately as a completely randomized design, using the individual animal as the experimental unit. Lambs and steers grazing wheat pasture for the first time in the spring had less ADG during the first 14 d than lambs (80 vs. 270 g, respectively; P = 0.01) and steers (1.06 vs. 1.83 kg, respectively; P = 0.09) that had grazed wheat pastures all winter. Digestibility of DM, NDF, and ADF fractions and N metabolism of freshly harvested wheat forage by lambs and steers were not different (P > 0.10) between the 2 treatment groups. Less ADG during the first 14 d of wheat pasture grazing is most likely the result of less DMI by nonadapted animals and is not due to diet digestibility or N metabolism.

We describe a simple psychophysical paradigm for studying figure-ground segregation by onset asynchrony. Two pseudorandom arrays of Gabor patches are displayed, to left and right of fixation. Within one array, a subset of elements form a figure, such as a randomly curving path, that can only be reliably detected when their onset is not synchronized with that of the background elements. Several findings are reported. First, for most participants, segregation required an onset asynchrony of 20-40 ms. Second, detection was no better when the figure was presented first, and thus by itself, than when the background elements were presented first, even though in the latter case the figure could not be detected in either of the two successive displays alone. Third, asynchrony segregated subsets of randomly oriented elements equally well. Fourth, asynchronous onsets aligned with the path could be discriminated from those lying on the path but not aligned with it. Fifth, both transient and sustained neural activity contribute to detection. We argue that these findings are compatible with neural signaling by synchronized rate codes. Finally, schizophrenic disorganization is associated with reduced sensitivity. Thus, in addition to bearing upon basic theoretical issues, this paradigm may have clinical utility.

The objective herein was to estimate heterosis and breed effects in purebred and crossbred Romosinuano, Brahman, and Angus calves on acute phase protein response to weaning and transportation. Calves (n = 1,032) were weaned in September of 2002, 2003, and 2004 at approximately 7 mo of age. Approximately 28 d after weaning, steer calves (n = 482) were transported 1,800 km (20 h) to Oklahoma. Concentrations of 3 acute phase proteins (ceruloplasmin, fibrinogen, and haptoglobin) were measured in blood samples. Calves (steers and heifers) were sampled at weaning, and 24 and 72 h postweaning. For separate analyses, steers sent to Oklahoma were sampled before shipment, upon arrival, and 24 and 72 h after arrival. Combinations of the following fixed effects were investigated: sire breed, dam breed, sampling time, birth location, calf sex (weaning only), year, cow age, and interactions. Effects of special interest were sire breed x dam breed as an indication of breed group of calf, and the interaction of sire and dam breeds with sampling time. Weaning age and BW were investigated as linear and quadratic covariates. Sire of calf within sire breed was a random term. The correlation structure of repeated measures was determined by comparison of information criterion values for different structures within each analysis. In general, plasma acute phase protein concentrations in weaned calves increased with sampling time. Concentrations in the transported steers increased through sampling at 24 h after arrival, and were lower at 72 h. Significant estimates of heterosis were detected for Brahman-Angus haptoglobin concentrations at weaning (0.38 +/- 0.14 mg/dL x 100; 44%), and for Romosinuano-Angus fibrinogen concentrations at weaning (11.4 +/- 5.5 mg/dL; 10%) and in transported steers (22.5 +/- 8.4 mg/dL; 20%). The direct effect of Romosinuano was to increase (P <0.004) ceruloplasmin concentrations of weaned calves (4.1 +/- 0.9 mg/dL) and of transported steers (3.9 +/- 1.3 mg/dL). The direct effect of Angus was to lower ceruloplasmin concentrations in weaned calves (-3.9 +/- 1.2; P = 0.001). Significant maternal effects were detected at weaning for ceruloplasmin concentrations in Romosinuano (-1.4 +/- 0.5 mg/ dL) and Angus (1.6 +/- 0.7 mg/dL) and fibrinogen concentrations in Brahman calves (-17.7 +/- 8.8 mg/dL). These data imply that acute phase protein concentrations in response to weaning and transportation are impacted by cattle breed.

Multiple lines of evidence have provided compelling evidence for the existence of a tumor suppressor gene (TSG) on chromosome 7q31.1. ST7 may be the target of this genetic instability but its designation as a TSG is controversial. In this study, we show that, functionally, ST7 behaves as a tumor suppressor in human cancer. ST7 suppressed growth of PC-3 prostate cancer cells inoculated subcutaneously into severe combined immunodeficient mice, and increased the latency of tumor detection from 13 days in control tumors to 23 days. Re-expression of ST7 was also associated with suppression of colony formation under anchorage-independent conditions in MDA-MB-231 breast cancer cells and ST7 mRNA expression was downregulated in 44% of primary breast cancers. Expression profiling of PC-3 cells revealed that ST7 predominantly induces changes in genes involved in re-modeling the extracellular matrix such as SPARC, IGFBP5 and several matrix metalloproteinases. These data indicate that ST7 may mediate tumor suppression through modification of the tumor microenvironment.

We report a novel psychophysical paradigm that distinguishes the information present in abrupt stimulus onset from that in the following display. The task is to pick the one odd item from a set added to a pre-existing background of similar items. When all new items are added simultaneously, observers are impaired even at distinguishing one red item amongst several green ones. An asynchrony of about 40 ms between target and distracter items restores performance, with evidence that it is cortical, rather than stimulus timing difference that is significant. The results are consistent with a role for neural synchrony in dynamic grouping.

Two experiments were conducted to examine the effect of previous BW gain during winter grazing on subsequent growth, carcass characteristics, and change in body composition during the feedlot finishing phase. In each experiment, 48 fall-weaned Angus x Angus-Hereford steer calves were assigned randomly to one of three treatments: 1) high rate of BW gain grazing winter wheat (HGW), 2) low rate of BW gain grazing winter wheat (LGW), or 3) grazing dormant tallgrass native range (NR) supplemented with 0.91 kg/d of cottonseed meal. Winter grazing ADG (kg/d) for HGW, LGW, and NR steers were, respectively, 1.31, 0.54, 0.16 (Exp. 1) and 1.10, 0.68, 0.15 (Exp. 2). At the end of winter grazing, four steers were selected randomly from each treatment to measure initial carcass characteristics and chemical composition of carcass, offal, and empty body. All remaining steers were fed a high-concentrate diet to a common backfat end point. Six steers were selected randomly from each treatment for final chemical composition, and carcass characteristics were measured on all steers. Initial fat mass and proportion in carcass, offal, and empty body were greatest (P < 0.001) for HGW, intermediate for LGW, and least for NR steers in both experiments. Live BW ADG and gain efficiency during the finishing phase did not differ (P = 0.24) among treatments, but DMI (% of mean BW) for NR and LGW was greater (P < 0.003) than for HGW steers. Final empty-body composition did not differ (P = 0.25) among treatments in Exp. 1. In Exp. 2, final carcass and empty-body fat proportion (g/kg) was greater (P < 0.03) for LGW and NR than for HGW steers. Accretion of carcass fat-free organic matter was greater (P < 0.004) for LGW than for HGW and NR steers in Exp. 1, but did not differ (P = 0.22) among treatments in Exp. 2. Fat accretion in carcass, offal, and empty body did not differ (P = 0.19) among treatments in Exp. 1, but was greater (P < 0.05) for LGW and NR than for HGW steers in Exp. 2. Heat production by NR steers during finishing was greater (P < 0.02) than by HGW steers in Exp. 1 and 2. Differences in ADG during winter grazing and initial body fat content did not affect rate of live BW gain or gain efficiency during finishing. Feeding steers to a common backfat thickness end point mitigated initial differences in carcass and empty-body fat content. However, maintenance energy requirements during finishing were increased for nutritionally restricted steers that were wintered on dormant native range.

Loss of heterozygosity (LOH) on the short arm of chromosome 8, at 8p12-p23, is one of the most frequent genetic events in both breast and ovarian cancer, suggesting the location of a shared tumour suppressor gene. Microcell-mediated chromosome transfer of chromosome 8 suppresses tumorigenicity and growth of colorectal and prostate cancer cell lines, further supporting the presence of a tumour suppressor gene on 8p. We have taken a candidate gene approach to try to identify this tumour suppressor gene at 8p12-p23. BNIP3L, which has sequence homology to pro-apoptotic proteins and the ability to suppress colony formation in soft agar, is located at 8p21, within a region of ovarian cancer LOH, breast cancer LOH and prostate cancer metastasis suppression. BNIP3L expression was assessed by both RT-PCR and Northern blot analysis in breast and ovarian cancer cell lines and found to be expressed at similar levels relative to expression in their respective normal epithelial cell lines. Genetic analysis of BNIP3L in 40 primary ovarian and 25 primary breast tumours identified one somatic, intronic mutation in one ovarian tumour, as well as several polymorphisms, including one resulting in an amino-acid substitution. These data suggest that BNIP3L is unlikely to be the target of 8p LOH in ovarian or breast cancer.
